The whole-body fatty acid balance method was used to investigate the fatty acid metabolism in Murray cod (Maccullochella peelii peelii) fed diets containing canola (CO) or linseed oil (LO). Murray cod were able to elongate and desaturate both 18:2n-6 and 18:3n-3. In fish fed the CO diet, 54.4% of the 18:2n-6 consumed was accumulated, 38.5% oxidized and 6.4% elongated and desaturated to higher homologs. Fish fed the LO diet accumulated 52.9%, oxidized 37% and elongated and desaturated 8.6% of the consumed 18:3n-3. The overall roles of n-6 fatty acids appeared more important in Murray cod compared to other freshwater species. Murray cod also showed a preferential order of utilization of C18 fatty acid for energy production (18:3n-3 > 18:2n-6 > 18:1n-9). Moreover, it is demonstrated that an increase in dietary 18:3n-3 is directly responsible of increased desaturase activity and augmented saturated fatty acid accumulation in the fish body. The present study also suggests that, in the context of the possible maximization of the natural ability of fish to produce long chain polyunsaturated fatty acids, the whole-body approach can be considered well suited and informative and Murray cod is a suited candidate to fish oil replacement for its diets.  相似文献

Juvenile Murray cod Maccullochella peelii exhibited a type II functional response while preying on blackworms Lumbriculus variegatus, and the parameters of the type II model did not differ significantly between clear (0 NTU) and turbid (150 NTU) treatments. Further experiments showed that vision may not be necessary for prey detection and capture by juvenile M. peelii; consumption of inanimate prey was not significantly different between light and dark (<1 × 10(-4) μE m(-2) s(-1)) trials. These results imply that the sensory physiology of M. peelii is well adapted to a turbid visual environment. In addition, habitat complexity increased the food consumption rate of juvenile M. peelii, perhaps by relaxing innate predator avoidance behaviours that depress foraging in more open environments.  相似文献

The temporal dynamics of oocyte growth, plasma sex steroids and somatic energy stores were examined during a 12 month ovarian maturation cycle in captive Murray cod Maccullochella peelii peelii under simulated natural photothermal conditions. Ovarian function was found to be relatively uninhibited in captivity, with the exception that post-vitellogenic follicles failed to undergo final maturation, resulting in widespread pre-ovulatory atresia. Seasonal patterns of oocyte growth were characterised by cortical alveoli accumulation in March, deposition of lipids in April, and vitellogenesis between May and September. Two distinct batches of vitellogenic oocytes were found in Murray cod ovaries, indicating a capacity for multiple spawns. Plasma profiles of 17beta-oestradiol and testosterone were both highly variable during the maturation period suggesting that multiple roles exist for these steroids during different stages of oocyte growth. Condition factor, liver size and visceral fat stores were all found to increase prior to, or during the peak phase of vitellogenic growth. Murray cod appear to strategically utilise episodes of high feeding activity to accrue energy reserves early in the reproductive cycle prior to its deployment during periods of rapid ovarian growth.  相似文献

This study provides information on habitat selection by the threatened Murray cod Maccullochella peelii peelii at two spatial scales in the Ovens and Murray Rivers in south-eastern Australia. Both adult (>450 mm total length, L T) and age 0 year (<150 mm L T) M. p. peelii selected macro and microhabitats based on structural variables. At the macrohabitat scale, adults selected channel habitats in the river, floodplain channels at high floods and within Lake Mulwala, whereas the floodplain proper was avoided. Adult and age 0 year fish selected similar microhabitats regardless of site or hydrologic conditions, and selection was primarily influenced by the presence of higher loadings of structural woody habitat, higher c.v. in depth, more overhanging vegetation, shallower comparative depths and lower water velocities, closer to the bank. Age 0 year M. p. peelii appeared to select shallower habitats with greater amounts of structural woody habitat, closer to the river bank than adult fish.  相似文献

The majority of information concerning the cardio-metabolic performance of varanids during exercise is limited to a few species at their preferred body temperature (T(b)) even though, being ectotherms, varanids naturally experience rather large changes in T(b). Although it is well established that absolute aerobic scope declines with decreasing T(b), it is not known whether changes in cardiac output (V(b)) and/or tissue oxygen extraction, (Ca(O2) - Cv(O2)), are in proportion to the rate of oxygen consumption (Vo(2)). To test this, we studied six Rosenberg's goannas (Varanus rosenbergi) while at rest and while maximally exercising on a treadmill both at 25 and 36 degrees C. During maximum exercise both at 25 and 36 degrees C, mass-specific rate of oxygen consumption (Vo(2kg)) increased with an absolute scope of 8.5 ml min(-1) kg(-1) and 15.7 ml min(-1) kg(-1), respectively. Interestingly, the factorial aerobic scope was temperature-independent and remained at 7.0 which, at each T(b), was primarily the result of an increase in V(bkg), governed by approximate twofold increases both in heart rate (f(H)) and cardiac stroke volume (V(Skg)). Both at 25 degrees C and 36 degrees C, the increase in V(bkg) alone was not sufficient to provide all of the additional oxygen required to attain maximal Vo(2kg), as indicated by a decrease in the blood convection requirement V(bkg)/Vo(2kg); hence, there was a compensatory twofold increase in (Ca(O2) - Cv(O2)). Although associated with an increase in hemoglobin-oxygen affinity, a decrease in T(b) did not impair unloading of oxygen at the tissues and act to reduce (Ca(O2) - Cv(O2)); both Ca(O2)) and Cv(O2)) were maintained across T(b). The change in Vo(2kg) with T(b), therefore, is solely reliant on the thermal dependence of V(bkg). Maintaining a high factorial aerobic scope across a range of T(b) confers an advantage in that cooler animals can achieve higher absolute aerobic scopes and presumably improved aerobic performance than would otherwise be achievable.  相似文献

Biomanipulation is a method of controlling algal blooms in eutrophic freshwater ecosystems. The most common approach has been to enhance herbivores through a reduction of planktivorous fish and introduction of piscivorous fish. The method was originally intended to reduce grazing pressure on zooplankton, thereby increasing grazing pressure on phytoplankton to increase water clarity and promote the growth of aquatic macrophytes. Biomanipulation has received considerable attention since it was proposed in 1975 where innovative approaches and explanations of the processes have been developed. Although many successful biomanipulation exercises have been conducted internationally, it has received comparatively little attention in the Southern Hemisphere and has not been trialled in the southern temperate climate of South Australia. This is a review to speculate upon the criteria for and against the application of biomanipulation in southern temperate Australia using the native species Murray cod (Maccullochella peelii peelii) and to suggest future research.  相似文献

We examined the effects of increased glucose availability on glucose kinetics and substrate utilization in horses during exercise. Six conditioned horses ran on a treadmill for 90 min at 34 +/- 1% of maximum oxygen uptake. In one trial [glucose (Glu)], glucose was infused at a mean rate of 34.9 +/- 1.1 micromol. kg(-1). min(-1), whereas in the other trial [control (Con)] an equivalent volume of isotonic saline was infused. Plasma glucose increased during exercise in Glu (90 min: 8.3 +/- 1.7 mM) but was largely unchanged in Con (90 min: 5.1 +/- 0.4 mM). In Con, hepatic glucose production (HGP) increased during exercise, reaching a peak of 38.6 +/- 2.7 micromol. kg(-1). min(-1) after 90 min. Glucose infusion partially suppressed (P < 0.05) the rise in HGP (peak value 25.8 +/- 3.3 micromol. kg(-1). min(-1)). In Con, glucose rate of disappearance (R(d)) rose to a peak of 40.4 +/- 2.9 micromol. kg(-1). min(-1) after 90 min; in Glu, augmented glucose utilization was reflected by values for glucose R(d) that were twofold higher (P < 0.001) than in Con between 30 and 90 min. Total carbohydrate oxidation was higher (P < 0.05) in Glu (187.5 +/- 8.5 micromol. kg(-1). min(-1)) than in Con (159.2 +/- 7.3 micromol. kg(-1).min(-1)), but muscle glycogen utilization was similar between trials. We conclude that an increase in glucose availability in horses during low-intensity exercise 1) only partially suppresses HGP, 2) attenuates the decrease in carbohydrate oxidation during such exercise, but 3) does not affect muscle glycogen utilization.  相似文献

Microsatellite markers were utilized to examine the genetic structure of Murray cod Maccullochella peelii throughout its distribution in the Murray--Darling Basin (MDB) of eastern Australia, and to assess the genetic effects of over three decades of stocking hatchery-reared fingerlings. Bayesian analysis using the programme Structure indicated that the species is largely genetically panmictic throughout much of its extensive range, most probably due to the high level of connectivity between catchments. Three catchments with terminal wetlands (the Lachlan, Macquarie and Gwydir), however, contained genetically distinct populations. No stocking effects were detected in the catchments that were genetically panmictic (either because of low genetic power or lack of effects), but the genetically differentiated Gwydir and Macquarie catchment populations were clearly affected by stocking. Conversely, there was no genetic evidence for survival and reproduction of stocked fish in the Lachlan catchment. Therefore, stocking of M. peelii throughout the MDB has resulted in a range of genetic effects ranging from minimal detectable effect, to substantial change in wild population genetic structure.  相似文献

This study determined the effects of elevated plasma epinephrine on fat metabolism during exercise. On four occasions, seven moderately trained subjects cycled at 25% of peak oxygen consumption (VO(2 peak)) for 60 min. After 15 min of exercise, subjects were intravenously infused with low (0.96 +/- 0.10 nM), moderate (1.92 +/- 0.24 nM), or high (3.44 +/- 0.50 nM) levels (all P < 0.05) of epinephrine to increase plasma epinephrine above control (Con; 0.59 +/- 0.10 nM). During the interval between 35 and 55 min of exercise, lipolysis [i.e., rate of appearance of glycerol] increased above Con (4.9 +/- 0.5 micromol. kg(-1). min(-1)) with low, moderate, and high (6.5 +/- 0.5, 7.1 +/- 0.8, and 10.6 +/- 1.2 micromol. kg(-1). min(-1), respectively; all P < 0.05) levels of epinephrine despite simultaneous increases in plasma insulin. The release of fatty acid into plasma also increased progressively with the graded epinephrine infusions. However, fatty acid oxidation was lower than Con (11.1 +/- 0.8 micromol. kg(-1). min(-1)) during moderate and high levels (8.7 +/- 0.7 and 8.1 +/- 0.9 micromol. kg(-1). min(-1), respectively; P < 0.05). In one additional trial, the same subjects exercised at 45% VO(2 peak) without epinephrine infusion, which produced a plasma epinephrine concentration identical to low levels. However, lipolysis was lower (i.e., 5.5 +/- 0.6 vs. 6.5 +/- 0.5 micromol. kg(-1). min(-1); P < 0.05). In conclusion, elevations in plasma epinephrine concentration during exercise at 25% of VO(2 peak) progressively increase whole body lipolysis but decrease fatty acid oxidation. Last, increasing exercise intensity from 25 to 45% VO(2 peak) attenuates the lipolytic actions of epinephrine.  相似文献

Researchers have hypothesised that influxes of pelagic zooplankton to river channels after floods and high flows are necessary for strong recruitment of some native fish species, including Murray cod (Maccullochella peelii peelii) (Mitchell), in the Murray–Darling river system, Australia. This study investigated the composition of the diet and gut fullness of drifting Murray cod larvae weekly during two spawning seasons with contrasting flows, to determine if pelagic zooplankton comprised a greater proportion of the gut contents and guts were fuller in a high flow (2000) than in a low flow (2001) year. Gut fullness and yolk levels of 267 larvae were ranked, and prey identified to family level. Approximately 40 and 70% of individuals had been feeding in 2000 and 2001, respectively. Gut fullness increased with declining yolk reserves. Larvae in both the years had an almost exclusively benthic diet, irrespective of the flow conditions at the time. Substantial inundation of dry ground in 2000, albeit restricted to in-channel benches, anastomosing channels and oxbow lakes, did not lead to an influx of pelagic, floodplain-derived zooplankton subsequently exploited by Murray cod larvae. These results have the implications for the management of regulated temperate lowland rivers: high flows cannot automatically be assumed to be beneficial for the fish larvae of all species and their food resources, and caution should be exercised with the timing of flow releases.  相似文献

Kinetics of intramuscular triglyceride fatty acids in exercising humans.   总被引:6,自引:0,他引:6  
A pulse ([(14)C]palmitate)-chase ([(3)H]palmitate) approach was used to study intramuscular triglyceride (imTG) fatty acid and plasma free fatty acid (FFA) kinetics during exercise at approximately 45% peak O(2) consumption in 12 adults. Vastus lateralis muscle was biopsied before and after 90 min of bicycle exercise; (3)H(2)O production, breath (14)CO(2) excretion and lipid oxidation (indirect calorimetry) rates were measured during exercise. Results: during exercise, 8.2+/-1.2 and 8.4+/-0.7 micromol x kg(-1) x min(-1) of imTG fatty acids and plasma FFA, respectively, were oxidized according to isotopic measurements. The sum of these two values was not different (P = 0.6) from lipid oxidation by indirect calorimetry (15.4 +/-1.6 micromol x kg(-1) x min(-1)); the isotopic and indirect calorimetry values were correlated (r = 0.79, P<0.005). During exercise, imTG turnover rate was 0.32+/-0.07%/min (6.0+/-2.0 micromol of imTG x kg wet muscle(-1) x min(-1)) and plasma FFA were incorporated into imTG at a rate of 0.7+/-0.1 micromol x kg wet muscle(-1) x min(-1). The imTG pool size did not change during exercise. This pulse-chase, dual tracer appears to be a reasonable approach to measure oxidation and synthesis kinetics of imTG.  相似文献

Judgement of exercise performance in birds has been hampered by a paucity of data on maximal aerobic capacity. We measured the maximal rate of oxygen consumption (Vo2,max) in running guinea fowl Numida meleagris, a bird that has been used in several previous studies of avian running. Mean Vo2,max during level treadmill running was 97.5+/-3.7 mL O(2) kg(-1) min(-1) (mean+/-SEM, N=5). Vo2,max was on average 6% higher when the birds ran uphill compared with the value during level running (paired t-test, P=0.041, N=5). The mean basal rate of oxygen consumption (Vo2,bmr) of the same individuals was 7.9+/-0.5 mL O(2) kg(-1) min(-1). Mean factorial aerobic scope based on individually measured values of Vo2,max and Vo2,bmr was 13.2+/-0.6 (mean+/-SEM, N=5). This value was considerably lower than the factorial aerobic scope previously measured during running in Rhea americana, a large flightless ratite. The difference in factorial scope between these two running birds likely reflects the effects of body size as well as size-independent differences in the ability to deliver and use oxygen. These data confirm a previous prediction that birds have a diversity of factorial aerobic scopes similar to that exhibited by mammals.  相似文献

The Australian freshwater cod genus, Maccullochella is represented by three species: Murray cod, M. peelii peelii, eastern freshwater cod, M. ikei, and trout cod, M.macquariensis. Seven novel microsatellite loci from M. ikei and six previously published loci from M. peelii peelii were tested on wild populations of Murray, eastern and trout cod. Levels of polymorphism varied between species with 13 loci polymorphic in Murray cod, 9 in trout cod and 7 in eastern cod. Observed heterozygosities ranged from 0.053 to 0.842. This suite of microsatellite loci will facilitate future studies of the genetic status of wild and hatchery bred populations of Maccullochella.  相似文献

Nine endurance-trained men exercised on a cycle ergometer at approximately 68% peak O2 uptake to the point of volitional fatigue [232 +/- 14 (SE) min] while ingesting an 8% carbohydrate solution to determine how high glucose disposal could increase under physiological conditions. Plasma glucose kinetics were measured using a primed, continuous infusion of [6,6-2H]glucose and the appearance of ingested glucose, assessed from [3-3H]glucose that had been added to the carbohydrate drink. Plasma glucose was increased (P < 0.05) after 30 min of exercise but thereafter remained at the preexercise level. Glucose appearance rate (R(a)) increased throughout exercise, reaching its peak value of 118 +/- 7 micromol. kg(-1). min(-1) at fatigue, whereas gut R(a) increased continuously during exercise, peaking at 105 +/- 10 micromol. kg(-1). min(-1) at the point of fatigue. In contrast, liver glucose output never rose above resting levels at any time during exercise. Glucose disposal (R(d)) increased throughout exercise, reaching a peak value of 118 +/- 7 micromol. kg(-1). min(-1) at fatigue. If we assume 95% oxidation of glucose R(d), estimated exogenous glucose oxidation at fatigue was 1.36 +/- 0.08 g/min. The results of this study demonstrate that glucose uptake increases continuously during prolonged, strenuous exercise when carbohydrate is ingested and does not appear to limit exercise performance.  相似文献

The present study investigated potential sex-related differences in the metabolic response to carbohydrate (CHO) ingestion during exercise. Moderately endurance-trained men and women (n = 8 for each sex) performed 2 h of cycling at approximately 67% Vo(2 max) with water (WAT) or CHO ingestion (1.5 g of glucose/min). Substrate oxidation and kinetics were quantified during exercise using indirect calorimetry and stable isotope techniques ([(13)C]glucose ingestion, [6,6-(2)H(2)]glucose, and [(2)H(5)]glycerol infusion). In both sexes, CHO ingestion significantly increased the rates of appearance (R(a)) and disappearance (R(d)) of glucose during exercise compared with WAT ingestion [males: WAT, approximately 28-29 micromol x kg lean body mass (LBM)(-1) x min(-1); CHO, approximately 53 micromol x kg LBM(-1) x min(-1); females: WAT, approximately 28-29 micromol x kg LBM(-1) x min(-1); CHO, approximately 61 micromol x kg LBM(-1) x min(-1); main effect of trial, P < 0.05]. The contribution of plasma glucose oxidation to the energy yield was significantly increased with CHO ingestion in both sexes (from approximately 10% to approximately 20% of energy expenditure; main effect of trial, P < 0.05). Liver-derived glucose oxidation was reduced, although the rate of muscle glycogen oxidation was unaffected with CHO ingestion (males: WAT, 108 +/- 12 micromol x kg LBM(-1) x min(-1); CHO, 108 +/- 11 micromol x kg LBM(-1) x min(-1); females: WAT, 89 +/- 10 micromol x kg LBM(-1) x min(-1); CHO, 93 +/- 11 micromol x kg LBM(-1) x min(-1)). CHO ingestion reduced fat oxidation and lipolytic rate (R(a) glycerol) to a similar extent in both sexes. Finally, ingested CHO was oxidized at similar rates in men and women during exercise (peak rates of 0.70 +/- 0.08 and 0.65 +/- 0.06 g/min, respectively). The present investigation suggests that the metabolic response to CHO ingestion during exercise is largely similar in men and women.  相似文献

To examine the role of beta-adrenergic mechanisms in the regulation of endogenous glucose (Glu) production [rate of appearance (R(a))] and utilization [rate of disappearance (R(d))] and carbohydrate (CHO) metabolism, six horses completed consecutive 30-min bouts of exercise at approximately 30% (Lo) and approximately 60% (Hi) of estimated maximum O(2) uptake with (P) and without (C) prior administration of the beta-blocker propranolol (0.22 mg/kg iv). All horses completed exercise in C; exercise duration in P was 49.9 +/- 1.2 (SE) min. Plasma Glu was unchanged in C during Lo but increased progressively in Hi. In P, plasma Glu rose steadily during Lo and Hi and was higher (P < 0.05) than in C throughout exercise. Plasma insulin declined during exercise in P but not in C; beta-blockade attenuated (P < 0.05) the rise in plasma glucagon and free fatty acids and exaggerated the increases in epinephrine and norepinephrine. Glu R(a) was 8.1 +/- 0.8 and 8.4 +/- 1.0 micromol. kg(-1). min(-1) at rest and 30.5 +/- 3.6 and 42.8 +/- 4.1 micromol. kg(-1). min(-1) at the end of Lo in C and P, respectively. During Hi, Glu R(a) increased to 54.4 +/- 4.4 and 73.8 +/- 4.7 micromol. kg(-1). min(-1) in C and P, respectively. Similarly, Glu R(d) was approximately 40% higher in P than in C during Lo (27.3 +/- 2.0 and 39.5 +/- 3.3 micromol. kg(-1). min(-1) in C and P, respectively) and Hi (37.4 +/- 2.6 and 61.5 +/- 5.3 micromol. kg(-1). min(-1) in C and P, respectively). beta-Blockade augmented CHO oxidation (CHO(ox)) with a concomitant reduction in fat oxidation. Inasmuch as estimated muscle glycogen utilization was similar between trials, the increase in CHO(ox) in P was due to increased use of plasma Glu. We conclude that beta-blockade increases Glu R(a) and R(d) and CHO(ox) in horses during exercise. The increase in Glu R(d) under beta-blockade suggests that beta-adrenergic mechanisms restrain Glu R(d) during exercise.  相似文献

Cytochemistry has proven effective in differentiating specific cell lineages and elucidating their functional properties. This study utilised a range of cytochemical techniques to further investigate the leucocyte populations from Murray cod, an iconic Australian teleost fish species. This analysis provided clear insight into the structure and function of the leucocytes from this fish, which were found to be broadly similar to those of other fish species. However, some important differences were identified in Murray cod, such as the presence of naphthol AS chloroacetate esterase activity in the heterophil population, positive staining for periodic acid Schiff's, alkaline phosphatase and Sudan black B in the lymphocyte population, and a prevalent population of myeloid precursor cells.  相似文献
