Callus cultures were initiated from immature cotyledons of Vignaaconitifolia, V. mungo and V. radiata on MS medium supplementedwith NAA, picloram or 2, 4-D. On transfer to L-6 liquid mediumsupplemented with low concentrations of picloram, GA3 and cytokinins,large number of somatic embryos differentiated from the callus.The cells destined to become somatic embryos divided to formspherical or filamentous proembryos. From the filamentous proembryo,the embryo proper developed either at single or multiple sites.Development of somatic embryos from multiple sites resultedin several embryos connected by a common suspensor at the radicleend. Continued divisions of the proembryos led to globular,heart shaped and dicotyledonary stages of somatic embryogenesis.The somatic embryos of V. mungo and V. aconitifolia differentiatedinto tiny plantlets at low frequency (1%) in liquid suspensioncultures supplemented with zeatin, picloram and GA3. The relation of the in vivo nitrate reductase (NR) activityto growth period was studied in the nodules and the leaves ofthe summer moong (Vigna radiata). The maximum NR activity wasobserved 31 days after sowing (DAS) in the leaves and 28 DASin the case of the nodules. In a pot experiment, the effectof the various nitrogen concentrations, namely 0, 3, 6, 9 and12 mg kg–1 was studied on NR activity at three growthstages. The maximum NR activity was observed at 6 mg kg–1N during the pre-flowering stage (26 DAS). Though the noduleshave higher NR activity, its expression was limited by substrateavailability. Piperazine, a chemical used as buffer component, greatly promotedadventitious root formation in cuttings of sunflower (Helianthusannuus L.), pea (Pisum sativum L.), mung bean (Vigna radiataL.) and to a lesser extent in bean (Phaseolus vulgaris L.) seedlings.Piperazine was more effective in acidic pH. The studies withpiperazine analogues showed that any substantial modificationof the structure caused the chemical to be less effective, oreven inhibitory. Histological studies in sunflower hypocotylsdemonstrated that piperazine did not alter the timing of theinitial cell division. In the presence of piperazine, sunflowerhypocotyls failed to develop primary phloem fibres. Piperazineat the concentrations that promote rooting did not kill or damagethe tissue at the base of the hypocotyl. Compared to controls,piperazine treatment did not alter the proportion of primordiathat eventually developed into actively elongating roots. Sixdays after treatment 45% of the control roots in the basal sectionwere actively growing, compared to 51% in the piperazine. The rate of destruction of the far red absorbing form of phytochrome(Pfr) in green or etiolated cuttings of Vigna radiata was slowerin the presence of malformin than in its absence. Malforminhad no effect on the accumulation of total phytochrome in thedark, or on the reaccumulation of phytochrome after destructionin red light. The amount of photoconversion of the red absorbingform of phytochrome (Pr) to Pfr or Pfr to Pr by given dosesof red or far red radiation was slightly but consistently lessin malformin-treated cuttings of V. radiata than in controls.Malformin had no effect on the rate of destruction or photoconversionof phytochrome in etiolated shoots of Avena sativa. The decreasein destruction rate of Pfr by malformin in V. radiata may contributeto the inhibition of dark abscission by malformin after lighttreatment. (Received October 3, 1979; )  相似文献

Prat, R. 1985. A comparative study of some methods for measuringcell growth potentials in Vigna radiata hypocotyl, in situ andafter excision.—J. exp. Bot. 36: 1150–1158. Accurate estimations of growth rates are required to understandthe development of physiological and biochemical parametersinvolved in growth processes. Growth measurements can be madeeither ‘in situ’ on whole seedlings by marking successivelevels or ‘in vitro’, after excision of successivehypocotyl segments. These different methods have been comparedin order to determine which measurements allow one to assessaccurately the normal growth characteristics. In vitro measurementsperformed with excised segments required a prolonged incubationperiod before stable growth rates were achieved. The growthpotentials of the successive hypocotyl levels remain unchangedafter excision. These in vitro measurements can therefore beused to assess the growth characteristics of cells in hypocotyls. By using the Giemsa C-banding technique, chromosome bandingpatterns on the somatic chromosomes of eight important pulsecrops, pea, lentil, guar (cluster bean), chick pea, pigeon pea,mung bean (green gram), urd (black gram) and cowpea have beenstudied. Each species has a characteristic C-banding pattern.The significance of such banding patterns which correlate withthe position of pachytene knobs, in chromosome identification,and in assigning relationships at the cytological level in thepulses of genus Vigna is stressed. A field experiment was conducted to study the effect of nitrogenapplication at 15, 30 and 45 kg ha–1 of urea at pre-flowering(PF) and pod initiation (PI) stages on the activity of nitrogenase(N2ase), nitrate reductase (NR) and other related parametersin the nodules of moong (Vigna radiata). Nitrogen applied atPF or PI stage was found to be inhibitory to N2ase and glutaminesynthetase (GS) activities except at 15 kg N ha–1 whenapplied at PF in the case of N2ase. At both the stages therewas increase in NR and glutamate dehydrogenase (GDH) activitieswith the application of nitrogen. Seed yield increased by 18per cent with the application of 15 kg N ha–1 at PI stagewhereas nitrogen application at PF stage only increased strawyield significantly. Mesophyll and hypocotyl protoplasts were isolated from Vignaradiata using a combination of Cellulase Onozuka RIO (1%) andMacerozyme (0·2%). On a modified V-47 medium, 60% ofthe cultured protoplasts divided and developed into colonies.Protoclones differentiated roots on MS medium supplemented withdifferent auxins and cytokinins; however, shoot differentiationwas not obtained. Co-cultivation of protoplasts with wild andshooter mutants of Agrobacterium tumefaciens did not lead todifferentiation of plantlets. Completely anhydrous fixation with acrolein vapour or osmiumtetroxide vapour was applied to tissues of air-dry seeds: thecoleoptile of wheat (Trilicum aestivum), and plumule and radicleof mung bean (Vigna radiata). Great shrinkage of cells and organelleswas noted, but all the usual organelles could be identified,except for Golgi bodies and (in most cases) ribosomes. The endoplasmicreticulum was very abundant and endoplasmic reticulum tubuleswere closely associated with the storage organelles, namelylipid bodies in the wheat coleoptile, and protein bodies inthe mung bean embryo axis. Callus cultures were initiated from seedling root segments ofmungbean (Vigna radiata (L.) Wilczek var. radiata) cv. K 851on modified PC-L2 basal medium. Growing cells were exposed toincreasing concentrations of NaCl in the medium. A concentrationof 300 mol m–3 NaCl proved completely inhibitory to growthof the calli. On incubation for 25 d, cells which could toleratethis concentration of NaCl grew to form cell clones. Selectedclones were characterized with regard to their growth behaviour,K+, Na+ and free proline content when grown under stress aswell as on normal media and were compared with the normal sensitivecallus. The selected callus was capable of growing on mediumcontaining NaCl at the inhibitory concentration. The K+ contentof the selected callus was lower in the case of the NaCl mediumthan for the normal medium. However, the selected clones maintainedhigher K+ and Na+ levels, with increased salinization comparedwith the wild-type cells. Salt-selected cells accumulated higherlevels of free proline under NaCl stress compared to wild-typecells. Lycoricidinol, a natural growth inhibitor isolated from bulbsof Lycoris radiata Herb. strongly suppressed auxin-induced ethyleneproduction from the hypocotyl segments of etiolated mung bean(Vigna radiata Wilczek) seedlings. The inhibitor did not significantlyinhibit ethylene formation from its immediate precursor, 1-aminocyclopropane-1-carboxylicacid (ACG), during short-term (up to 4 h) incubation. The ACCcontent in tissue treated with IAA was reduced by lycoricidinolin close parallel with the inhibition of ethylene production.Examination of radioactive metabolites in tissues labeled with3,4-14C-methionine indicated that reduction of the ACC contentwas not due to any possible promotive effect of lycoricidinolon conjugation of ACC with malonate. Lycoricidinol showed noinhibitory effect on the activity of ACC synthase if appliedin vitro, but it almost completely abolished the increase inthe enzyme activity when applied in vivo during incubation ofthe tissue with IAA. Lycoricidinol also strongly inhibited incorporationof 14C-leucine into protein in the tissue. The suppression ofthe enzyme induction and, in turn, that, of ethylene productionby lycoricidinol were interpreted as being due to the inhibitionof protein synthesis. When seedlings of Cuscuta japonica were grown with Vigna radiata(the host plant) in a flower pot for 6 d under white light andthen irradiated with far-red or blue light (ca. 6 µmolphotons m–2 s–1), the seedlings parasitized V. radiata.However, no parasitism of the seedlings was observed under redor white light or in darkness. The parasitic behavior of seedlingsof C. japonica was observed even if an acrylic rod was usedas a substitute for the host plant. Upon incubation under far-redlight, the seedling twined tightly around the rod and developedhaustoria towards it. Haustoria also developed when apical andsubapical regions of seedlings were held between two glass platesthat were about 0.7 mm apart and were irradiated with far-redlight. However, no haustoria were induced by either the holdor irradiation alone. These results indicate that parasitismof Cuscuta japonica is controlled by the cooperative effectsof two physical signals, far-red light and appropriate tactilepressure. Multiple components of leghaemoglobins (Lbs) from several legumespecies have been separated by polyacrylamide gel electrophoresis(PAGE), and identified by densitometry at 405 nm, extractionfrom gels and formation of their respective CO-ferrous complexes.Components were numbered consecutively starting from the anodeside, and their relative concentrations were calculated by densitometryin control and -treated nodules. The number of Lb components was five in Pisum sativum, four in Glycinemax, at least two in Lupinus luteus and Trifolium repens, andtwo in Vigna radiata and V. unguiculata. Only one componentwas detected in fresh nodules of Phaseolus vulgaris. Nitratesignificantly reduced the total content of Lb in all legumes,except Lupinus, the extent of reduction varying with the species.It also had a major effect on the proportions of components,causing an inversion in the relative abundance of Lb I and LbII of V. radiata: the concentration of Lb I increased from 8%to 81% with application, while total Lb only decreased by 13%. Drastic changes in proportions of Lbcomponents were not artifactual and cannot be attributed exclusivelyto different turnover rates. Structural, biochemical, and immunological comparisons of nodulesfrom ten species of plants were made to determine if a correlationexists between nodule structure, ureide production, urate oxidaseactivity, and antigenic similarity in urate oxidase. In specieswith high urate oxidase activity and cross-reaction with soybeananti-urate oxidase [soybean (Glycine max), green bean (Phaseolusvulgaris), mung bean (Vigna radiata), cowpea (Vigna unguiculata)],the nodules were determinate and contained numerous interstitialcells, interspersed among the infected cells. Within the interstitialcells of the ureide producing nodules numerous peroxisomes werenoted and these peroxisomes appear to be structurally similar,viz. a large electron opaque core surrounded by a less electronopaque rim. Although hemp sesbania (Sesbania exaltata) noduleswere similar in ultrastructure to other ureide producers withdetectable urate oxidase activity, no cross-reactivity was observedwith anti-soybean urate oxidase. Amide producing nodules eithercontained no interstitial cells [e.g. Indian jointvetch (Aeschynomeneindica), showy crotalaria (Crotalaria spectabilis)} or interstitialcells with few peroxisomes [e.g. alfalfa (Medicago saliva),broad bean (Vicia faba), pea (Pisum sativum)] with little urateoxidase activity, exhibiting no cross-reaction with soybeananti-urate oxidase. These data indicate that the urate oxidasein most ureide producing nodules is very similar and, structurally,ureide producing nodules are organized in a specialized wayto carry out ureide assimilation in the uninfected interstitialcells. The rates of increase in dry weight, leaf area, and length ofhypocotyl plus radicle of mung bean (Vigna radiata L.) and morningglory (Pharbitis nil Chois) all show marked responses to decreasingtemperature, although when drawn as Arrhenius plots these responsesresemble smooth curves rather than sets of intersecting straightlines. Chilling sensitivity was shown to be dependent on temperature,light intensity, and the previous history of the plant. It isargued on theoretical grounds that changes in membrane lipidmobility would not be expected to cause abrupt changes in theslope of such Arrhenius plots at either the enzyme or wholeplant level.  相似文献

Mung bean (Vigna radiata L. Wilcz. cv. Berken) seedlings wereraised hydroponically in 0, 5 and 15 mol m–3 choline.Fourteen-day-old plants were chilled at 5°C (under lightconditions) for 24 h and then returned to warm conditions fora further 24 h. Primary leaf lamina tissue was used for thedetermination of phospholipid, mole ratio of sterol to phospholipid(ST/PL), sterol composition, and ethylene-forming enzyme (EFE)activity for the various choline-temperature treatments employed.Chilling caused an irreversible loss of lipid in the absence,of choline. Differential loss of lipid resulted in an increasein ST/PL and a decline in the mole ratio of sitosterol to stigmasterol(S/S). There was no recovery of EFE activity following chillingin the absence of choline. Choline (5 and 15 mol m–3)enhanced phospholipid and sterol levels prior to chilling andmaintained lipid levels throughout the chill-warm cycle, withoutany significant change in ST/PL. At 5.0 mol m–3 choline,the chilling-induced decline in S/S was reduced, while at 15mol m–3 S/S increased following chilling. Choline treatment,though reducing EFE activity prior to chilling, allowed recoveryof EFE activity following transfer of plants from chilling towarm conditions. Seeds of barley (Hordeum vulgare L.) and mung bean (Vigna radiata(L.) Wilczek), with orthodox seed storage behaviour, were imbibedfor between 8 h and 96 h at 15 °C and 25 °C, respectively,while barley seeds were also maintained in moist aerated storageat 15 °C for 14 d. These seeds and seedlings, together withcontrols, were then dried to various moisture contents between3% and 16% (wet basis) and hermetically stored for six monthsat —20°C, 0°C or 15°C. In both species, neitherdesiccation nor subsequent hermetic storage of the control lotsresulted in loss in viability. The results for barley seedsimbibed for 24 h were similar to the control, but desiccationsensitivity increased progressively with duration of imbibitionbeyond 24 h in barley or 8 h in mung bean; these treatmentsalso reduced the longevity of the surviving seeds in air-drystorage. Loss in viability in barley imbibed for 48 h was mostrapid at the two extreme seed storage moisture contents of 3·6%and 14·3%, and in both these cases was more rapid at15 °C than at cooler temperatures. Similarly, for mung beanimbibed for 8 h, loss in viability was most rapid at the lowest(4·3%) moisture content, but in this case it was morerapid at –20 °C than at warmer temperatures.
