The purpose of the present study was to investigate whether the mechanical properties (i.e. force strain relationship) of the triceps surae tendon and aponeurosis relate to the performed sport activity in an intensity-dependent manner. This was done by comparing sprinters with endurance runners and subjects not active in sports. Sixty-six young male subjects (26+/-5 yr; 183+/-6 cm; 77.6+/-6.7 kg) participated in the study. Ten of these subjects were adults not active in sports, 28 were endurance runners and 28 sprinters. All subjects performed isometric maximal voluntary plantar flexion contractions (MVC) on a dynamometer. The distal aponeuroses of the gastrocnemius medialis (GM) was visualised by ultrasound during the MVC. The results showed that only the sprinters had higher normalised stiffness (relationship between tendon force and tendon strain) of the triceps surae tendon and aponeurosis and maximal calculated tendon forces than the endurance runners and the subjects not active in sports. Furthermore, including the data of all 66 examined participants tendon stiffness correlated significantly (r=0.817, P<0.001) with the maximal tendon force achieved during the MVC. It has been concluded that the mechanical properties of the triceps surae tendon and aponeurosis do not show a graded response to the intensity of the performed sport activity but rather remain at control level in a wide range of applied strains and that strain amplitude and/or frequency should exceed a given threshold in order to trigger additional adaptation effects. The results further indicate that subjects with higher muscle strength possibly increase the margin of tolerated mechanical loading of the tendon due to the greater stiffness of their triceps surae tendon and aponeurosis.  相似文献

The purpose of the present study was to evaluate active muscle stiffness with the stretch reflex according to changes (in 110-ms period after stretching) in torque and fascicle length during slower angular velocity (peak angular velocity of 100 deg·s−1) in comparison with active muscle stiffness without the stretch reflex (in 60-ms period after stretching) during slower and faster (peak angular velocity of 250 deg·s−1) angular velocities. Active muscle stiffness in the medial gastrocnemius muscle was calculated according to changes in estimated muscle force and fascicle length with slower and faster stretching during submaximal isometric contractions (10–90% maximal voluntary contractions). Active muscle stiffness significantly increased for both angular velocities and analyzed periods as torque levels exerted became higher. The effects of angular velocities and the interaction between angular velocities and torque levels were not significantly different between 250 deg·s−1 (in 60-ms period after stretching) and 100 deg·s−1 (in 110-ms period after stretching) conditions. The effects of the analyzed periods and the interaction between analyzed periods and torque levels were not significantly different between the analyzed periods (60-ms and 110-ms periods after stretching) for the 100 deg·s−1 condition. Furthermore, active muscle stiffness measured during the same angular velocity had significant correlations between those calculated in the different analyzed periods, whereas those under 250 deg·s−1 (60-ms period after stretching) did not correlate with those under 100 deg·s−1 (110-ms period after stretching). These results suggest that active muscle stiffness is not influenced by the stretch reflex.  相似文献

It is well known that muscular force production is history-dependent, which results in enhanced (RFE) and depressed (RFD) steady-state forces after stretching and shortening, respectively. However, it remains unclear if force-enhancing mechanisms can contribute to increased performance during in vivo stretch-shortening cycles (SSCs) of human locomotor muscles. The purpose of this study was to investigate whether RFE-related mechanisms contribute to enhanced force and power output during SSCs of the human plantar flexor muscles. Net ankle torques of fourteen participants were measured during and after pure isometric, pure stretch, pure shortening, and SSC contractions when the triceps surae muscles were electrically stimulated at a submaximal level that resulted in 30% of their maximum isometric torque. Dynamic contractions were performed over an amplitude of 15°, from 5° plantar flexion to 10° dorsiflexion, at a speed of 120° s−1. External ankle work during shortening was 11.6% greater during SSCs compared to pure shortening contractions (p = .003). Additionally, RFD after SSCs (8.6%) was reduced compared to RFD after pure shortening contractions (12.0%; p < .05). It is therefore concluded that RFE-related mechanisms contribute to increased performance following SSCs of human locomotor muscles. Since RFD after SSCs decreased although work during shortening was increased, we speculate that the relevant mechanism lies outside actin-myosin interaction. Finally, our data suggests that RFE might be relevant and beneficial for human locomotion whenever a muscle is stretched, but this needs to be confirmed.  相似文献

In earlier studies, we found more economical runners having a more compliant quadriceps femoris (QF) tendon at low force levels, and a higher contractile strength and stiffness at the triceps surae (TS). To better understand how these differences influence force generation economy and energy recovery, we simulated contractions using a Hill-type muscle model and the previously determined muscle properties as input parameters. For eight different activation levels, we simulated isovelocity concentric contractions preceded by an isovelocity stretch. The length changes and contraction velocities imposed to the muscle–tendon units (MTU) corresponded to those happening whilst running. The main results of the simulations were: (a) a more compliant tendon at low force levels (QF) led to an advantage in force-generation due to a decrease in shortening velocity of the CE, (b) a higher contractile strength and higher stiffness at the TS led to a disadvantage in force-generation at high activation levels and to an advantage at low activation levels. In addition at the high economy runners both MTUs showed an advantageous energy release during shortening, which at the QF was mainly due to a higher elongation of the SEE and at the TS mainly to the higher contractile strength. Especially at low activation levels both MTUs showed an advantageous force generation per activation and a higher energy release as compared to the low economy runners.  相似文献

The Achilles tendon (AT) moment arm transforms triceps surae muscle forces into a moment about the ankle which is critical for functional activities like walking. Moreover, the AT moment arm changes continuously during walking, as it depends on both ankle joint rotation and triceps surae muscle loading (presumably due to bulging of the muscle belly). Here, we posit that aging negatively effects the architecturally complex AT moment arm during walking, which thereby contributes to well-documented reductions in ankle moment generation during push-off. We used motion capture-guided ultrasound imaging to quantify instantaneous variations in the AT moment arms of young (23.9 ± 4.3 years) and older (69.9 ± 2.6 years) adults during walking, their dependence on triceps surae muscle loading, and their association with ankle moment generation during push-off. Older adults walked with 11% smaller AT moment arms and 11% smaller peak ankle moments during push-off than young adults. Moreover, as hypothesized, these unfavourable changes were significantly and positively correlated (r2 = 0.38, p < 0.01). More surprisingly, aging attenuated load-dependent increases in the AT moment arm (i.e., those between heel-strike and push-off at the same ankle angle); only young adults exhibited a significant increase in their AT moment arm due to triceps surae muscle-loading. Age-associated reductions in triceps surae volume or activation, and thus muscle bulging during force generation, may compromise the mechanical advantage of the AT during the critical push-off phase of walking in older adults. Thus, strategies to restore and/or improve locomotor performance in our aging population should consider these functionally important changes in musculoskeletal behavior.  相似文献

During range of motion (max-ROM) tests performed on an isokinetic dynamometer, the mechanical delay between the button press (by the participant to signal their max-ROM) and the stopping of joint rotation resulting from system inertia induces errors in both max-ROM and maximum passive joint moment. The present study aimed to quantify these errors by comparing data when max-ROM was obtained from the joint position data, as usual (max-ROMPOS), to data where max-ROM was defined as the first point of dynamometer arm deceleration (max-ROMACC). Fifteen participants performed isokinetic ankle joint max-ROM tests at 5, 30 and 60° s−1. Max-ROM, peak passive joint moment, end-range musculo-articular (MAC) stiffness and area under the joint moment-position curve were calculated. Greater max-ROM was observed in max-ROMPOS than max-ROMACC (P < 0.01) at 5 (0.2 ± 0.15%), 30 (1.8 ± 1.0%) and 60° s−1 (5.9 ± 2.3%), with the greatest error at the fastest velocity. Peak passive moment was greater and end-range MAC stiffness lower in max-ROMPOS than in max-ROMACC only at 60° s−1 (P < 0.01), whilst greater elastic energy storage was found at all velocities. Max-ROM and peak passive moment are affected by the delay between button press and eventual stopping of joint rotation in an angular velocity-dependent manner. This affects other variables calculated from the data. When high data accuracy is required, especially at fast joint rotation velocities (≥30° s−1), max-ROM (and associated measures calculated from joint moment data) should be taken at the point of first change in acceleration rather than at the dynamometer’s ultimate joint position.  相似文献

Using nematophagous fungi for the biological control of animal parasitic nematodes will become one of the most promising strategies in the search for alternative chemical drugs. The purpose of this study was to check the in vitro activity of four anthelmintics, four chemical fungicides and two antifungal drugs on the spore germination of nematophagous fungi: Duddingtonia flagrans (SF170), Arthrobotrys oligospora (447), Arthrobotrys superba (435) and Arthrobotrys sp. (PS011). A modified 24-well cell culture plate assay was conducted to evaluate the susceptibility of nematophagous fungi against drugs tested by calculating the effective middle concentrations (EC50) of each tested drug to inhibit the germination of fungal spores. EC50 ranged between 0·7 and 47·2 μg ml−1 for fenbendazole, thiabendazole and ivermectin, except levamisole (546·5–4057·8 μg ml−1). EC50 of tested fungicides was 0·6–2·3 μg ml−1 for carbendazim, 55·9–247·4 μg ml−1 for metalaxyl, 24·4–45·2 μg ml−1 for difenoconazole, and 555·9–1438·3 μg ml−1 for pentachloronitrobenzene (PCNB). EC50 of two antifungal drugs was 0·03–3·4 μg ml−1 for amphotericin B and 0·3–10·9 μg ml−1 for ketoconazole. The results showed that 10 tested drugs, except for levamisole and PCNB, had in vitro inhibitory effects on nematophagous fungi. The chlamydospores of Dflagrans had the highest sensitivity to nine tested drugs, except for ketoconazole.  相似文献

Nutrient regeneration and respiration rates of natural zooplankton from a tropical reservoir were experimentally measured. Excretion rates of ammonia (Ea), orthophosphate (Ep) and community respiration rates (R) were estimated considering the variations in the concentrations of ammonia, orthophosphate and dissolved oxygen between control and experimental units. The ranges obtained for these rates from the 2 h assays were Ea = 1.95–4.95 μg N-NH4 · mg · DW−1 · h−1; Ep = 0.12–0.76 μg P-PO4 mg DW−1 · h−1. Respiratory rates were quite constant (R = 0.01–0.02 mg O2 · mg DW−1 · h−1). The uptake of nutrients due to bacteria can affect the experimental determination of excretion rates of zooplankton. Orthophosphate release increased from 0.28 to 0.82 μg P-PO4 · mg DW−1 · h−1 when bacterial activity was depleted by antibiotic addition in experimental vessels (Exp IV). This demonstrates that free living bacteria are able to consume promptly most phosphorus excreted by zooplankton. Ammonia excretion rates were lower in experimental units containing antibiotics. Lower excretion rates were also obtained with longer exposure times and higher biomass levels in the experimental units. Finally, this study also showed that zooplankton excretion can affect significantly turn over rates of total phosphorus in Pampulha Reservoir. In some periods, specially during the dry season when zooplankton biomass was very high, phosphorus release by zooplankton, during one single day, can be as high as 40% of the total phosphorus content in lake water (Turn over time = 2.5 days).  相似文献

We investigated the composition of benthic microbial mats in permanently ice-covered Lake Hoare, Antarctica, and their irradiance vs. photosynthetic oxygen exchange relationships. Mats could be subdivided into three distinct depth zones: a seasonally ice-free “moat” zone and two under-ice zones. The upper under-ice zone extended from below the 3.5 m thick ice to approximately 13 m and the lower from below 13 m to 22 m. Moat mats were acclimated to the high irradiance they experienced during summer. They contained photoprotective pigments, predominantly those characteristic of cyanobacteria, and had high compensation and saturating irradiances (Ec and Ek) of 75 and 130 μmol photons·m−2·s−1, respectively. The moat mats used light inefficiently. The upper under-ice community contained both cyanobacteria and diatoms. Within this zone, biomass (as pigments) increased with increasing depth, reaching a maximum at 10 m. Phycoerythrin was abundant in this zone, with shade acclimation and efficiency of utilization of incident light increasing with depth to a maximum of 0.06 mol C fixed·mol−1 incident photons under light-limiting conditions. Precipitation of inorganic carbon as calcite was associated with this community, representing up to 50% of the carbon sequestered into the sediment. The lower under-ice zone was characterized by a decline in pigment concentrations with depth and an increasing prevalence of diatoms. Photosynthesis in this community was highly shade acclimated and efficient, with Ec and Ek below 0.5 μmol·m−2·s−1 and 2 μmol·m−2·s−1, respectively, and maximum yields of 0.04 mol C fixed·mol−1 incident quanta. Carbon uptake in situ by both under-ice and moat mats was estimated at up to 100 and 140 mg·m−2·day−1, based on the photosynthesis–irradiance curves, incident irradiance, and light attenuation by ice and the water column.  相似文献

The enthalpies of the hexokinase-catalyzed phosphorylation or glucose, mannose, and fructose by ATP to the respective hexose 6-phosphates have been measured calorimetrically in TRIS/TRIS HCl buffer at 25.0, 28.5, and 32.0°C. The effects on the measured enthalpy of the glucose/hexokinase reaction due to variation of pH (over the range 6.7 to 9.0) and ionic strength (over the range 0.02 to 0.25) have been examined. Correction for enthalpy of buffer protonation leads to δHo and δCpo values for the processes: eq-D-hexose + ATP4− = eq-D-hexose 6-phosphate2− + ADP3−+ H+. Results are δHo = −23.8 ± 0.7 kJ · mol−1 and δCpo = −156 ± 280 J·mol−1·K−1 for glucose. δHo = −21.9 ± 0.7 kJ·mol−1 and δCpo = 10 ± 140 J·mol−1·K−1 for mannose, and δHo = −15.0 ± 0.9 kJ·mol−1 and δCpo = −41 ± 160 J·mol−1·K−1 for fructose. Combination of these measured enthalpies with Gibbs energy data for hydrolysis of ATP4− and that for the hexose 6-phosphates lead to δSo values for the above hexokinase-catalyzed reactions.  相似文献

To determine the most favorable conditions for the production of ethanol by Pachysolen tannophilus, this yeast was grown in batch cultures with various initial concentrations of two of the constituents of the culture medium: d-xylose (so), ranging from 1 g·l−1 to 200 g·l−1, and yeast extract (lo), ranging from 0 g·l−1 to 8 g·l−1. The most favorable conditions proved to be initial concentrations of So=25 g·l−1 and lo=4 g·l−1, which gave a maximum specific growth rate of 0.26 h−1, biomass productivity of 0.023 g·l−1·h−1, overall biomass yield of 0.094 g·g−1, specific xylose-uptake rate (qs) of 0.3 g·g−1·h−1 (for t=50 h), specific ethanol-production rate (qE) of 0.065 g·g−1·h−1 and overall ethanol yield of 0.34 g·g−1; qs values decreased after the exponential growth phase while qE remained practically constant.  相似文献

Catalytic combustion of hydrocarbons is an important technology to produce energy. Compared to conventional flame combustion, the catalyst enables this process to operate at lower temperatures; hence, reducing the energy required for efficient combustion. The reaction and activation energies of direct combustion of hydrocarbons (CH?→?C?+?H) on a series of metal surfaces were investigated using density functional theory (DFT). The data obtained for the Ag, Au, Al, Cu, Rh, Pt, and Pd surfaces were used to investigate the validity of the Brønsted-Evans-Polanyi (BEP) and transition state scaling (TSS) relations for this reaction on these surfaces. These relations were found to be valid (R2?=?0.94 for the BEP correlation and R2?=?1.0 for the TSS correlation) and were therefore used to estimate the energetics of the combustion reaction on Ni, Co, and Fe surfaces. It was found that the estimated transition state and activation energies (ETS?=??69.70 eV and Ea?=?1.20 eV for Ni, ETS?=??87.93 eV and Ea?=?1.08 eV for Co and ETS?=??92.45 eV and Ea?=?0.83 eV for Fe) are in agreement with those obtained by DFT calculations (ETS?=??69.98 eV and Ea?=?1.23 eV for Ni, ETS?=??87.88 eV and Ea?=?1.08 eV for Co and ETS?=??92.57 eV and Ea?=?0.79 eV for Fe). Therefore, these relations can be used to predict energetics of this reaction on these surfaces without doing the time consuming transition state calculations. Also, the calculations show that the activation barrier for CH dissociation decreases in the order Ag ? Au ? Al ? Cu ? Pt ? Pd ? Ni?>?Co?>?Rh?>?Fe.  相似文献

The purpose of this study was to provide evidence on the fact that the observed decrease in EMG activity of the gastrocnemius medialis (GM) at pronounced knee flexed positions is not only due to GM insufficiency, by examining muscle fascicle lengths during maximal voluntary contractions at different positions. Twenty-two male long distance runners (body mass: 78.5+/-6.7 kg, height: 183+/-6 cm) participated in the study. The subjects performed isometric maximal voluntary plantar flexion contractions (MVC) of their left leg at six ankle-knee angle combinations. To examine the resultant ankle joint moments the kinematics of the left leg were recorded using a Vicon 624 system with 8 cameras operating at 120 Hz. The EMG activity of GM, gastrocnemius lateralis (GL), soleus (SOL) and tibialis anterior (TA) were measured using surface electromyography. Synchronously, fascicle length and pennation angle values of the GM were obtained at rest and at the plateau of the maximal plantar flexion using ultrasonography. The main findings were: (a) identifiable differences in fascicle length of the GM at rest do not necessarily imply that these differences would also exist during a maximal isometric plantar flexion contraction and (b) the EMG activity of the biarticular GM during the MVC decreased at a pronounced flexed knee-joint position (up to 110 degrees ) despite of no differences in GM fascicle length. It is suggested that the decrease in EMG activity of the GM at pronounced knee flexed positions is due to a critical force-length potential of all three muscles of the triceps surae.  相似文献

The purpose of this study was to investigate the effect of the differences between the actual fascicle length during a voluntary contraction and the fascicle length at rest of the triceps surae muscle on the determination of the voluntary activation (VA) by using the interpolated twitch technique. Twelve participants performed isometric voluntary maximal (MVC) and submaximal (20%, 40%, 60% and 80% MVC) contractions at two different ankle angles (75 degrees and 90 degrees ) under application of the interpolated twitch technique. Two ultrasound probes were used to determine the fascicle length of soleus, gastrocnemius medialis and gastrocnemius lateralis muscles. Further, the MVCs and the twitches were repeated for six more ankle angles (85 degrees , 95 degrees , 100 degrees , 105 degrees , 110 degrees and 115 degrees ). The VA of the triceps surae muscle were calculated (a) using the rest twitch force (RTF) measured during the same trial as the interpolated twitch force (ITF; traditional method) and (b) using the RTF at an ankle angle where the fascicle length showed similar values between ITF and RTF (fascicle length consideration method). The continuous changes in fascicle length from rest to MVC affect the accuracy of the assessment of the VA. The traditional method overestimates the assessment of the VA on average 4% to 12%, especially at 90 degrees ankle angle (i.e. short muscle length). The reason for this influence is the unequal force-length potential of the muscle at twitch application by the measure of ITF and RTF. These findings provide evidence that the fascicle length consideration method permits a more precise prediction (an improvement of 4-12%) of the voluntary contraction compared to the traditional method.  相似文献

We aimed to synthesize hydroxyethyl starch (HES) 200/0.5-loaded bovine serum albumin nanoparticles (HBNs) and investigate the compatibility and binding mechanism in simulated physiological environments. Here, to elucidate the morphology, biocompatibility, and formation mechanism of HBNs, techniques such as scanning electron microscopy, haemolysis test, fluorescence, and circular dichroism spectroscopy were applied. The thermodynamic parameters at body temperature (ΔS° = −26.7 J·mol−1·K−1, ΔH° = −3.20 × 104 J·mol−1, and ΔG = −2.35 × 104 J·mol−1) showed a 1:1 binding stoichiometry, which was formed by hydrogen bonds and van der Waals interactions. In addition, the conformational analysis showed that the microenvironment of fluorophores was altered with the adaptational protein secondary structural changes. Energy transfer occurred from the fluorophores to HES with a high possibility. All these results provided accurate and complete primary data for demonstrating the interaction mechanisms of HES with BSA, which helps to understand its pharmaceutical effects in blood.  相似文献

Binding of [3H]aflatoxin B1 to rat plasma was investigated in vivo and vn vitro. Column chromatographic and polyacrylamide gel electrophoretic analysis clearly demonstrated that aflatoxin B1 bound primarily plasma albumin. Very little binding activity was shown by other plasma proteins. Spectrofluorimetric studies were undertaken to gain some insight into the nature of the aflatoxin-albumin interaction. Quenching of the lone tryptophan fluorescence intensity upon aflatoxin binding was due, at least in part, to a ligand-induced conformational change in the albumin molecule. Aflatoxin B1 binds an apolar site with an association constant of 30 mM−1 at pH 7.4 and 20°C. Neither charcoal treatment of rat albumin nor the presence of 0.15 M NaCl had many significant effect on the interaction. The association constant was pH-dependent, increasing about 1.7-fold as the pH increased from 6.1 to 8.4. This pH dependence is ascribed to a pH-induced conformational change in the albumin molecule. Thermodynamic studies indicated that the aflatoxin-albumin interaction was exothermic (ΔH = −29.3 kJ·mol), with a ΔS value of −13.8 J·mol−1·K−1.  相似文献

PurposeWe assessed fascicle behaviors of the upper extremities during isometric contractions at different joint angles in this study.MethodsThirteen healthy men and women performed isometric elbow extension tasks at 50% and 75% of maximal voluntary contraction (MVC) at 60°, 90°, and 120° of elbow extension (full extension = 180°). Extended field-of-view B-mode ultrasonography was used to obtain sagittal plane panoramic images of the long head (TB-Long) and medial head (TB-Med) of the triceps brachii at rest and during contraction; fascicle length and pennation angle were measured.ResultsIn the TB-Long, significant fascicle shortening from rest was found during 50% and 75%MVC at 60° and during 75%MVC at 90° of extension. There was no significant fascicle shortening in the TB-Med muscle under any conditions. There was no significant pennation angle change from rest in either muscle. The pennation angle of the TB-Long was significantly greater than that of the TB-Med under all conditions.ConclusionsThese results suggest that fascicle shortening in the TB-Long muscle occurs in flexion; however, no change was found in the TB-Med. In the upper extremity muscle–tendon complex, the superficial and deeper muscles may have different force-transmission efficiency at flexed joint angles.  相似文献

The uptake of three anthracycline derivatives: doxorubicin, daunorubicin and pirarubicin, into large unilamellar vesicles (LUV) in response to a driving force provided by DNA encapsulated inside the LUV has been investigated as a function of the temperature and of the bilayers lipid composition. The kinetics of the decay of the anthracycline fluorescence in the presence of DNA-containing liposome was used to follow the diffusion of the drug through the membrane. For the three drugs, the permeability coefficient of the neutral form of the drug (P0) decreases as the amount of negatively charged phospholipid in the bilayers increases. This can be explained by the fact that the kinetics of passive diffusion of the drugs depends on the amount of neutral form embedded in the polar head group region, which decreases as the quantity of negatively charged phospholipids increases. P0 also decreases as the amount of cholesterol, that makes the bilayer more rigid, increases. The activation energies, Ea, for the passage of the neutral form of these anthracyclines through the bilayers lie within 100±15 kJ·mol−1, except for pirarubicin and doxorubicin through anionic phospholipid-rich membranes (Ea=57 kJ·mol−1) and cholesterol-rich membranes (Ea=167 kJ·mol−1).  相似文献

Nuclear magnetic resonance line-widths data have been used to determine the rate of solvent exchange from the first coordination sphere of ferro-and ferriprotoporphyrin(IX) dimethylester (Fe-PPD) in pyridine/chloroform. The average values of kinetic parameters for pyridine (PY) exchange indicate an SN2 mechanism tor Fe(III)-PPD(ΔH&;#; = 36 kJ · mol−1 ; ΔS&;#; = −53 J·mol−1K−1; TM(298 K) = 0.07 msec) and an SNI mechanism for Fe(II)-PPD (ΔH&;#; = 67 kJ·mol−1; ΔS&;#; = 42 J · mol−1K−1; TM(298 K) = 0.06 msec). Parallel to the accelerated ligand exchange rate at rising temperatures a redistribution of the electrons causing a transition of the metal porphyrin from the low-spin state to the high-spin state is observed. Enthalpy and entropy of the thermodynamic equilibrium between low- and high-spin Fe-PPD have been determined from experimental values of the average magnetic moment. A mean lifetime of low-spin Fe(III)-PPD was estimated from line. widths changes (TL→H(298 K)≈ 20 msec) and the corresponding activation parameters have been obtained (ΔH&;#;L→H(298 K) = 26 kJ · mol−1; ΔS&;#;L→H(298K) = −125 J · mol−1K−1).  相似文献

An experiment was conducted in the growth chamber to quantify the biomass production, N removal and N2 fixation from a synthetic medium by Chlamydomonas reinhardtii and Anabaena flos-aquae. Nitrogen was supplied at a concentration of 100 mg liter−1 of NO315N and NH4+15 (3·5 atom %), respectively. After 21 days Chlamydomonas reinhardtii removed an average of 83·8 and 78·7 mg N liter−1 as NO3 and NH4+, respectively. Averages of 0·89 and 0·71 g liter−1 (first batch), 1·63 and 0·95 g liter (second batch) algal biomass were collected from NO3 and NH4+ media, respectively. Uptake rates of 0·11 mg 15N g−1 algae day−1 from NO3 medium and 0·10 mg 15N g−1 algae day−1 from NH4+ medium were calculated. Algal cells grown in NO3 and NH4+ medium contained 71 and 65 g N kg−1 (first batch), 39 and 58 g N kg−1 (second batch), respectively. Anabaena flos-aquae produced averages of 0·58 and 0·46 g liter−1 (first batch), 0·55 and 0·48 g liter−1 (second batch) after 14 days of growth from NO3 and NH4+ media, respectively. Blue-green algal biomass contained higher N (81–98 g kg−1) than green algae. Isotope dilution method for determining N2 fixation indicated that 55% and 77% of total N of blue-green algae grown in NO3 and NH4+ media, respectively, was derived from the atmosphere.  相似文献
