Effects of cerium ion (Ce(III)) on water relations of soybean seedlings (Glycine max L.) under ultraviolet B radiation (UV-B, 280–320 nm) stress were investigated under laboratory conditions. UV-B radiation not only affected the contents of two osmolytes (proline, soluble sugar) in soybean seedlings, but also inhibited the transpiration in soybean seedlings by decreasing the stomatal density and conductance. The two effects caused the inhibition in the osmotic and metabolic absorption of water, which decreased the water content and the free water/bound water ratio. Obviously, UV-B radiation led to water stress, causing the decrease in the photosynthesis in soybean seedlings. The pretreatment with 20 mg L−1 Ce(III) could alleviate UV-B-induced water stress by regulating the osmotic and metabolic absorption of water in soybean seedlings. The alleviated effect caused the increase in the photosynthesis and the growth of soybean seedlings. It is one of the protective effect mechanisms of Ce(III) against the UV-B radiation-induced damage to plants.  相似文献

Enhanced ultraviolet-B (UV-B) irradiation is one of the most important abiotic stresses that could influence the growth and physiological traits of plants. In this work, we reported the effects of silicon on the growth and physiological characteristics of wheat seedlings (Triticum aestivum L. cv Hengmai5229) subject to UV-B stress. Treatments with silicon significantly increased total biomass and chlorophyll (a + b) content, and reduced malondialdehyde (MDA) content and the rate of superoxide radical (O2) production in wheat seedlings subjected to UV-B stress. Silicon treatments also induced an increased in soluble sugar, anthocyanins, and flavonoid content. Leaf silicon concentration increased with the increasing of silicon supply to soil. Positive correlations were found in leaf silicon concentration with total biomass, chlorophyll (a + b), proline, and soluble protein content, respectively. MDA content and the rate of O2 production were negatively correlated with leaf silicon concentration in seedlings. The results demonstrated that silicon alleviated the damage caused by UV-B on wheat seedlings to some extent by the increase in antioxidant compounds content and leaf silicon concentration.  相似文献

Selenium (Se) is beneficial for some plants and is able to increase resistance and antioxidant capacity of plants subjected to stressful environment. In this work, the effects of enhanced ultraviolet-B (UV-B) radiation, Se supply, and their combination on growth and physiological traits of wheat (Triticum aestivum L., cv. Han NO.7086) seedlings were studied. The objective was to elucidate whether Se could alleviate the expected adverse effects of UV-B stress on seedlings. UV-B treatment caused a marked decline in growth parameters and total chlorophyll content and changed biomass allocation between aboveground and underground parts, which led to an increase in the root/shoot ratio. UV-B treatment also increased MDA content and the rate of superoxide radical (O2·−) production, although it increased some antioxidant (proline, phenolic compounds, and flavonoids) content and activity of antioxidant enzymes (peroxidase, superoxide dimutase, catalase (CAT)). Se treatment only increased total chlorophyll content and CAT activity. Compared with UV-B treatment alone, the combined treatment with UV-B and Se induced a significant increase in the biomass, total chlorophyll content, antioxidant content, and activity of antioxidant enzymes, and an evident decrease in MDA content and the rate of O2·− production. The results of this study demonstrated that Se alleviated the damage caused by UV-B to wheat seedlings to some extent by increasing antioxidant enzyme activity and antioxidant content.  相似文献

Due to anthropogenic influences, solar UV-B irradiance at the earth’s surface is increasing. To determine the effects of enhanced UV-B radiation on photosynthetic characteristics of Prunus dulcis, two-year-old seedlings of the species were submitted to four levels of UV-B stress, namely 0 (UV-Bc), 4.42 (UV-B1), 7.32 (UV-B2) and 9.36 (UV-B3) kJ m−2 d−1. Effects of UV-B stress on a range of chlorophyll (Chl) fluorescence parameters (FPs), Chl contents and photosynthetic gas-exchange parameters were investigated. UV-B stress promoted an increase in minimal fluorescence of dark-adapted state (F0) and F0/Fm, and a decrease in variable fluorescence (Fv, Fv/Fm, Fv/F0 and F0/Fm) due to its adverse effects on photosystem II (PSII) activity. No significant change was observed for maximal fluorescence of dark-adapted state (Fm). Enhanced UV-B radiation caused a significant inhibition of net photosynthetic rate (P N) at UV-B2 and UV-B3 levels and this was accompanied by a reduction in stomatal conductance (g s) and transpiration rate (E). The contents of Chl a, b, and total Chl content (a+b) were also significantly reduced at increased UV-B stress. In general, adverse UV-B effects became significant at the highest tested radiation dose 9.36 kJ m−2 d−1. The most sensitive indicators for UV-B stress were Fv/F0, Chl a content and P N. Significant P<0.05 alteration in these parameters was found indicating the drastic effect of UV-B radiation on P. dulcis.  相似文献

Cd~(2+)胁迫对小桐子幼苗叶片抗氧化系统的影响   总被引:1,自引:0,他引:1  
以小桐子幼苗为材料,设置不同浓度CdCl_2处理,测定Cd~(2+)胁迫对小桐子幼苗叶片中可溶性蛋白、丙二醛(MDA)含量,以及5种抗氧化酶活性和2种抗氧化剂含量的变化,探讨镉胁迫对小桐子幼苗抗氧化系统的影响。结果表明:(1)Cd~(2+)胁迫导致小桐子幼苗叶片中可溶性蛋白含量降低、MDA含量增加;(2)随着镉胁迫时间的延长,幼苗叶片中愈创木酚过氧化物酶(POD)、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、抗坏血酸专一性过氧化酶(APX)、谷胱甘肽还原酶(GR)等抗氧化酶活性表现出先升高然后降低的变化趋势;(3)幼苗叶片中还原型抗坏血酸(ASA)和还原型谷胱甘肽(GSH)含量随着胁迫时间延长而降低,但其中氧化型抗坏血酸(DHA)和氧化型谷胱甘肽(GSSG)含量则升高。研究表明,镉胁迫初期能诱导小桐子幼苗抗氧化系统活性显著增强,提高其抗氧化能力,但随着胁迫时间的延长,致使其抗氧化酶的活性和抗氧物质含量下降,植株遭受明显氧化胁迫,幼苗生长受到镉的严重毒害。  相似文献

Physiological and biochemical responses of wheat seedlings to drought, UV-B radiation, and combined stress were investigated. Drought, UV-B, and combined stresses retarded seedling growth by 26.5, 29.1, and 55.9%, respectively. One reason for growth retardation may be the oxidative damage indicated by an increase in the H2O2 content and lipid peroxidation degree. Furthermore, there was negative correlation between shoot fresh weight and H2O2 content, fresh weight and the content of thiobarbituric acid-reacting substances (TBARS), and the positive correlation between H2O2 content and TBARS (R 2 = 0.9251, 0.9005, and 0.9007, respectively). The activities of superoxide dismutase, guaiacol peroxidase, and ascorbate peroxidase increased under drought, UV-B, and the combination of stresses, while catalase activity decreased under the combined stress as compared to the control. The combination of drought and UV-B caused more severe damage to wheat seedlings than stress factors applied separately. Thus, the combined application of drought and UV-B had more strong adverse effects on wheat seedlings. The addition of 0.2 mM sodium nitroprusside (SNP) enhanced wheat seedling growth under drought, UV-B, and combined stress, likely, due to decreasing the accumulation of H2O2 and lipid peroxidation as well as activating the antioxidant enzymes. Bean (Phaseolus vulgaris L. cv. Berbukskaya) seedlings were pre-treated with choline compounds, 19 mM 2-ethyltrimethylammonium chloride (Ch) or 1.6 mM 2-chloroethyltrimethylammonium chloride (CCh), during 24 h, then after 6 d the excised primary leaves were exposed to UV-B and high temperature stress. Chlorophyll (Chl) fluorescence, delayed light emission, accumulation of photosynthetic pigments, contents of thiobarbituric acid reactive substances, and activities of the active oxygen detoxifying enzymes (superoxide dismutase, ascorbate peroxidase, and glutathione reductase) were examined. Pre-treatment of plants with Ch or CCh enhanced the resistance of photosystem 2 (PS2) photochemistry to UV-B and heat injuries. The higher stress resistance can be explained by the increased activity of the detoxifying enzymes. The increased content of UV-B-absorbing pigments may also contribute to the enhanced resistance of choline-treated plants to UV-B radiation.  相似文献

The effect of pre-treatment with 200 mM NaCl on the response of four barley cultivars (Hordeum vulgare L. cv. Bülbül-89, Kalaycı-97, Tarm-92 and Tokak-157/37) to UV-B radiation was investigated. Salt stress as well as UV-B irradiation led to a decrease of the total chlorophyll (chl) content in all cultivars, except in Kalaycı-97. While carotenoids are almost not affected by NaCl treatment, UV-B irradiation caused an increase by 5–20% of carotenoid content of all cultivars. UV-B induced damages of photosynthetic apparatus were estimated by the rate of photosynthetic electron transport measured by chl fluorescence and the rate of oxygen evolution, the latter being more affected. Pre-treatment with NaCl alleviated harmful effect of UV-B irradiation on F v/F m and ETR, but not on oxygen evolution. UV-B-induced and UV-B-absorbing compounds with absorption at 300 and 438 nm increased as a result of UV-B treatment. The level of stress marker proline increased considerably as a result of NaCl treatment, while UV-B irradiation resulted in a pronounced increase of the level of H2O2. MDA enhanced in the seedlings subjected to salt and UV-B stress. Established cross-acclimation to UV-B as a result of salt treatment could be due to the increased free proline and the level of UV-B absorbing compounds in barley seedlings subjected to NaCl.  相似文献

Enhanced ultraviolet-B radiation (UV-B, 280?C320?nm) is recognized as one of the environmental stress factors that cannot be neglected. Jasmonic acid (JA) is an important signaling molecule in a plant??s defense against biotic and abiotic stresses. To determine the role of exogenous JA in the resistance of wheat to stress from UV-B radiation, wheat seedlings were exposed to 0.9?kJ?m?2?h?1 UV-B radiation for 12?h after pretreatment with 1 and 2.5?mM JA, and the activity of antioxidant enzymes, the level of malondialdehyde (MDA), the content of UV-B absorbing compounds, photosynthetic pigments, and proline and chlorophyll fluorescence parameters were measured. The results of two-way ANOVA illustrated that the activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT), MDA level, anthocyanin and carotenoid (Car) content, and almost all chlorophyll fluorescence parameters were significantly affected by UV-B, JA, and UV-B?×?JA (P?<?0.05) [the maximal efficiency of photosystem II photochemistry (F v/F m) was not affected significantly by UV-B radiation]. Duncan??s multiple-range tests demonstrated that UV-B stress induced a significant reduction in plant photosystem II (PSII) function and SOD activity and an increased level of membrane lipid peroxidation, indicative of the deleterious effect of UV-B radiation on wheat. JA pretreatment obviously mitigated the detrimental effect of UV-B on PSII function by increasing F v/F m, reaction centers?? excitation energy capture efficiency (F v??/F m??), effective photosystem II quantum yield (??PSII), and photosynthetic electron transport rate (ETR), and by decreasing nonphotochemical quenching (NPQ) of wheat seedlings. Moreover, the activity of SOD and the content of proline and anthocyanin were provoked by exogenous JA. However, the MDA level was increased and Car content was decreased by exogenous JA with or without the presence of supplementary UV-B, whereas the contents of chlorophyll and flavonoids and related phenolics were not affected by exogenous JA. Meanwhile, exogenous JA resulted in a decrease of CAT and POD activities under UV-B radiation stress. These results partly confirm the hypothesis that exogenous JA could counteract the negative effects of UV-B stress on wheat seedlings to some extent.  相似文献

Two contrasting sea buckthorn (Hippophae rhamnoides L.) populations from the low (LA) and high (HA) altitudinal regions were employed to evaluate the plant physiological responses to solar UV-A radiation and near-ambient UV-B radiation (UV-B+A) under the sheltered frames with different solar ultraviolet radiation transmittance. LA-population was more responsive to solar UV-A. Some modification caused by UV-A only existed in LA-population, such as significant reduction of leaf size, relative water content, and chlorophyll (Chl) b content as well as δ13C elevation, coupled with larger increase of contents of total carotenoids (Cars). This higher responsiveness might be an effective pre-acclimation strategy adapting for concomitant solar UV-B stress. Near-ambient UV-B+A radiation caused significant reduction of leaf size and Chl content as well as slight down-regulation of photosystem 2 activity that paralleled with higher heat dissipation, while photosynthetic rate was modestly but significantly increased. The higher photosynthesis under near-ambient UV-B+A radiation could be related to pronounced increase of leaf thickness and effective physiological modification, like the increase of leaf protective pigments (Cars and UV-absorbing compound), constant high photochemical capacity, and improved water economy.  相似文献

Abstract

UV-B radiation (280–320 nm) as an environmental stress has damaging effect on plants and its overexposure can potentially interfere with growth and development. The effect of thiourea (TU) on UV-B stress tolerance of 10-day-old Brassica juncea seedlings subjected to supplementary UV-B for 5 days was investigated. An increase in the UV-B absorbing compounds anthocyanin, flavonoids and phenolic compounds was observed in UV-B + TU treated seedlings as compared to that of control. The enhanced synthesis of UV-B screening compounds resulted in lesser damage to chlorophyll and also gain in fresh weight and dry weight in UV-B + TU as compared to UV-B alone treatment. The enzymatic activities of guaiacol peroxidase (GPX) and superoxide dismutase (SOD) also increased in UV-B + TU. The expression profiling of phenylalanine ammonia lyase (pal) and chalcone synthase (chs) indicated an upregulation under UV-B + TU treatment, compared to that of control, suggesting that TU treatment ensured an early and efficient induction of flavonoid biosynthetic pathway. The results indicate that TU helps in ameliorating the damaging effects of UV-B stress by efficiently maintaining the antioxidant status and attenuating the penetration of the UV-B.  相似文献

The effects of exogenous salicylic acid (SA), sodium nitropusside (SNP, a nitric oxide donor), or their combination on dwarf polish wheat (Triticum polonicum L.) seedlings under UV-B stress were studied. The UV-B stress significantly decreased plant height, shoot dry mass, pigment content, net photosynthetic rate, intercellular CO2 concentration, stomatal conductance, transpiration rate, and variable to maximum chlorophyll fluorescence ratio (Fv/Fm) in all plants, but less in the presence of SA, SNP, and their combination. On the other hand, there were considerable increases in malondialdehyde (MDA), proline, O2 ?-, and H2O2 content under the UV-B stress. When SA, SNP, and their combination were applied, content of MDA, proline, H2O2, and O2 ?- were less increased. Moreover, there were considerable increases in activities of superoxide dismutase, peroxidase, ascorbate peroxidase, and glutathione reductase under the UV-B stress and more in the presence of SA, SNP, and their combination. Therefore, it is considered that SA, SNP, and especially their combination could alleviate UV-B stress in dwarf polish wheat.  相似文献

Pea (Pisum sativum L., cv. Scinado) seedlings were exposed to low doses of ultraviolet-B (UV-B; 4.4 and 13.3 kJ m−2 d−1) or UV-C (0.1 and 0.3 kJ m−2 d−1) radiation for 14 d. Aminocyclopropane carboxylic acid (ACC), indoleacetic acid (IAA) and abscisic acid (ABA) contents were quantified by gas chromatography coupled to mass spectrometry (GC-MS). The accumulation of ACC upon irradiation was dose-dependent. ABA content was reduced and IAA content increased upon UV-C treatment whereas the UV-B doses used did not cause significant changes in ABA and IAA contents.  相似文献

Cucumber plants (Cucumis sativus L., cv. Jingchun 3) were grown in a greenhouse under PAR illumination of 400–600 mol/(m2 s) at 30/15°C (day/night) temperature. Two enhanced biologically effective UV-B radiation levels per day were applied: 8.82 kJ/m2 (T1) and 12.6 kJ/m2 (T2). Cucumber seedlings were irradiated 7 h per day for 25 days under T1 and T2. A comparative study of growth, membrane permeability, and polyamine content in cucumber leaves under T1 and T2 treatments was conducted. UV-B radiation resulted in the dose-dependent decrease in leaf area, dry weight of foliage, and plant height. The T1 and T2 treatments caused an increase in the contents of putrescine, spermine, and spermidine. However, the total polyamine content declined slightly when electrolyte leakage increased dramatically on the 18th day of treatment, especially after T2 treatment. It can be concluded that polyamine accumulation in the cucumber leaves is an adaptive mechanism to the stress caused by UV-B radiation.  相似文献

Abstract: The impact of UV-B irradiation on unialgal cultures of the temperate marine diatom Odontella sinensis and of the Antarctic Odontella weissflogii was tested under controlled laboratory conditions. Uptake rates of inorganic nitrogen by Odontella sinensis were more affected by UV-B radiation than those of Odontella weissflogii. Utilization of 15N-ammonium was reduced after 3 h of UV-B exposure. Values of Km from several marine phytoplankton species were estimated from algae not exposed to UV-B and from those after 3 h of UV-B radiation. Calculation of Lineweaver-Burk plot showed, under UV-B stress, a noncompetitive inhibition for 15NH+4 uptake and a competitive inhibitory effect for 15NO?3 by Odontella weissflogii. The damage to 15NH+4 uptake by UV-B was more pronounced under red than blue light; a contradictory result was obtained for 15NO?3 utilization by Odontella weissflogii. Reduction of the pigments by UV-B under white light was dependent on the exposure time; a strong depression of the contents of chlorophyllide a, chlorophyll c, diatoxanthin and the fucoxanthins was found. Contents of the chlorophylls are markedly affected by UV-B under red light whereas the chlorophyll a concentration is enhanced in blue and green light. UV-B exposure in conjunction with blue and green light led to a reduction of the protein content and an increase in the amino acid contents. The pattern of pool sizes of free amino acids varied after UV-B exposure. UV-A irradiance had no effect. The possible targets of UV-B irradiation on the uptake system of inorganic nitrogen are discussed in detail. Adaptation to the environmental conditions, e.g. via synthesis of UV stress proteins or mycosporine-like amino acids, were also considered.  相似文献

In true leaves of common bean (Phaseolus vulgaris L., cv. Berbukskaya) 11-day-old seedlings, pretreatment with 1.6 mM chlorocholine chloride (CCC) resulted in the improved photosystem II (PSII) tolerance to UV-B radiation, an increase in the contents of cytokinins, ABA, and H2O2, but a decrease in the content of gibberellins. It is suggested that development of increased PSII tolerance to UV-B under the influence of CCC is partially related to the increase in the contents of ABA, cytokinins, and UV-absorbing pigments in the leaves of pretreated plants.  相似文献

The dynamic state of antioxidant capacity of flavonoid was investigated for a further demonstration of alleviating the damage of the ultraviolet (UV)-B radiation in the La-treated soybean seedlings under UV-B stress. Using hydroponics culture, the effects of lanthanum on the contents of flavonoid and its ability of antioxidant under elevated ultraviolet-B radiation (280–320 nm) was studied. The results showed flavonoid content in soybean seedlings with UV-B treatment during the stress and convalescent period was increased initially and then decreased, compared with control. Membrane permeability and MDA contents increase at first (first to fifth day) and then decrease (6th–11th day). A similar change of flavonoid content and clearance of flavonoid scavenging and ·OH in soybean seedlings occurred. Flavonoid content and ability of flavonoid scavenging and ·OH in soybean seedlings with La(III) + UV-B treatment were higher than those of UV-B treatment. Meanwhile, membrane permeability and MDA contents in soybean seedlings were lower than those of UV-B treatment. Compared with control, phenylalanine content in soybean seedlings with UV-B treatment is depressed, phenylalanine content in soybean seedlings with La(III) treatment was enhanced. However, phenylalanine content in La(III) + UV-B treatment is not decreased but slightly increased, compared with UV-B treatment. It suggested that the regulative effect of La(III) of the optimum concentration on flavonoid improved the metabolism of ROS, diminished the concentration of MDA and maintained normal plasma membrane permeability, and that its protective effect against low UV-B radiation is superior to that of high UV-B radiation. The defensive effect of La(III) on soybean seedlings under UV-B stress is carried out on the layer of defense system.  相似文献

A crop legume Vigna unguiculata L. (Walp.) and a wild legume Crotalaria juncea L. were evaluated for their relative responses to the oxidative stress injury induced by various doses of UV-B radiation (UV-B, 280–315 nm; 0, 1.0, 1.4, 4.7, and 6.0 kJ m−2 d−1). A dose-dependent damage in lipid peroxidation was determined as an index of membrane injury caused by UV-B. The impact was significantly higher in V. unguiculata than in C. juncea. The specific activities of superoxide dismutase, ascorbate peroxidase, monodehydroascorbate reductase, and dehydroascorbate reductase increased directly proportional to UV-B doses. However, the activities of these enzymes were significantly higher in V. unguiculata than in C. juncea indicating that V. unguiculata was inflicted with more severe oxidative stress injury under UV-B. In C. juncea the glutathione reductase and ascorbate oxidase activities were 35 and 40 % greater than in V. unguiculata, respectively. Further, the non-enzymatic antioxidants ascorbate and glutathione, and their reduced/oxidizes ratios in C. juncea were much greater than V. unguiculata indicating C. juncea has an inherently greater antioxidative potential than V. unguiculata. Thus C. juncea is better adapted to oxidative stress than V. unguiculata by means of efficient cellular antioxidant mechanisms helping to combat the photooxidative stress injury elicited by UV-B.  相似文献

The impact of elevated ultraviolet-B radiation (UV-B, 280–320 nm) on membrane systems and lipid peroxidation, and possible involvement of active oxygen radicals was investigated in leaves of two UV-B susceptible rice cultivars (Oryza sativa L. cvs IR74 and Dular). Rice seedlings were grown in a greenhouse for 10 days and then treated with biologically effective UV-B (UV-BBE) radiation for 28 days. Oxidative stress effects were evaluated by measuring superoxide anion (O2) generation rate, hydrogen peroxide (H2O2) content, malondialdehyde (MDA) concentration and relative electrolyte conductivity (EC) for IR74 and Dular at 0 (control), 6 or 13 kJ m?2 day?1 UV-BBE. Significant increases in these parameters were found in rice plants grown at 13 vs 0 kJ m?2 day?1 UV-BBE after 28 days; indicating that disruption of membrane systems may be an eventual reason for UV-B-induced injury in rice plants. There was a positive correlation between O2? generation and increases in EC or MDA in leaves. Activities of enzymatic and nonenzymatic free radical scavengers were measured for IR74 after 7, 14, 21 and 28 days of exposure to 13 or 0 UV-BBE to evaluate dynamics of these responses over time. Activities of catalase and superoxide dismutase (but not ascorbate peroxidase) and concentrations of ascorbic acid and glutathione were enhanced by 13 vs 0 UV-BBE after 14 days of UV-B exposure. Further exposure to 28 days of UV-B was associated with a decline in enzyme activities and ascorbic acid, but not glutathione. It is suggested that UV-B-induced injury may be associated with disturbance of active oxygen metabolism through the destruction and alteration of both enzymatic and nonenzymatic defense systems in rice.  相似文献

7-d-old etiolated and green barley seedlings (Hordeum vulgare L. cv. Alfa) were irradiated with UV-B for 30 min and then kept for 24 h in light or darkness. Chlorophyll (Chl) synthesis was inhibited by about 30 % as a result of UV-B irradiation, but there were no significant changes in photochemical activity measured by variable to maximum fluorescence ratio (Fv/Fm), quantum yield (ΦPS2) and oxygen evolution rate. Electron transport of etiolated seedlings was similar to that of green ones, nevertheless, the Chl content was more then 2-fold lower. Ribulose-1,5-bisphosphate carboxylase/oxygenase large and small subunits were diminished as a result of UV-B irradiation in etiolated and green plants, especially in those kept in the darkness. Catalase activity decreased and total superoxide dismutase activity increased in green and etiolated plants following UV-B treatment. When benzidine was used as a substrate, an isoform located between guaiacol peroxidases 2 and 3 (guaiacol peroxidase X) appeared, which was specific for UV-B treatment. As a result of irradiation, the contents of UV-B absorbing and UV-B induced compounds increased in green seedlings but not in etiolated seedlings.  相似文献
