Summary Phloem tissues ofMicrosorium scolopendria (Polypodiaceae) andPsilotum nudum (Psilotaceae) were examined with light and electron microscopes. The characteristic refractive spherules in the sieve elements ofM. scolopendria apparently develop from endoplasmic reticulum-derived cytoplasmic vesicles. In both taxa they have not been observed to be spatially related to plastids or mitochondria. Refractive spherules contain protein and often occur in the peripheral cytoplasm of mature sieve elements. InM. scolopendria they also occur in pericycle cells. Significant differences in refractive spherule substructure occur between the two taxa studied.  相似文献

A total of 67 bacterial isolates were obtained from apple and pear fruits with signs of soft rot collected from Egyptian markets. Pathogenicity tests showed that 25 isolates (37%) were pathogenic to apple and pear fruits, with considerable variation of virulence. Among these isolates, 16 (64%) were Gram‐positive, motile, spore‐forming long rods and were identified as members of the genus Bacillus based on an API test. In addition, five isolates (20%) were Gram‐negative, non‐spore‐forming, motile, oxidase and catalase‐positive short rods and were identified as members of the genus Pseudomonas. Furthermore, four isolates (16%) were Gram‐negative, non‐spore‐forming, motile, catalase‐positive and oxidase negative short rods and were identified as belonging to the genus Erwinia. All selected isolates showed a wide host range and could cause soft rot of all representative fruits and vegetables tested. The three most virulent isolates, AB4, AB6 and PB6, exhibited the highest soft rot severity on different apple and pear cultivars, and apple cv. Anna (116) was the most susceptible to infection by isolates AB4 and AB6, with soft rot severities of 63.33 and 60.67%, respectively. Also, pear cv. Le‐Conte was most susceptible to infection by isolate AB6, with a soft rot severity of 89.9%. A phylogenetic tree based on 16S rRNA gene sequences indicated that strains AB4 and AB6 were very similar to one another and also showed a similarity of 99% to Bacillus altitudinis, and strain PB6 revealed a similarity of 99% to Bacillus pumilus. To our knowledge, this is the first report of B. altitudinis as a soft rot pathogen for both apple and pear fruits.  相似文献

Survival analysis methodology was used to estimate the shelf life of alfajor (a chocolate‐coated individually wrapped cake) at 20 and 35C by using results obtained from consumers when asked if they would accept or reject samples with different storage times. Sensory acceptability (measured by consumers), off‐flavor (measured by a trained panel) and moisture content were linearly related to time. These correlations were used to estimate values at the shelf‐life times calculated for 25 and 50% rejection probability. Survival analysis provided the following shelf‐life estimation: 74 days at 20C and 33 days at 35C for a 25% of rejection, 87 days at 20C and 39 days at 35C for a 50% of rejection. An alfajor stored at 20C having an acceptability value below 4.9 (1–9 hedonic scale) and off‐flavor intensity above 5.3 (0–10 scale) would be rejected by 25% of the consumers. Chemical data were not good shelf‐life predictors.  相似文献

The new 2,3‐secoaromadendrane 1 , together with the known compounds plagiochilines A and M ( 2 and 3 , resp.), fusicogigantone A ( 4 ), and 1,4‐dimethylazulene ( 5 ) were isolated from an Argentine collection of the liverwort Plagiochila bursata. Structures were elucidated by extensive 1D‐ and 2D‐NMR studies. Compounds 2 and 4 , incorporated to the larval diet at 100 μg per g of diet, reduced the larval growth of Spodoptera frugiperda (Lepidoptera: Noctuidae) by 66±29% and 25±8% and produced 55 and 75% larval mortality at early instars and 20 and 25% pupal mortality, respectively. Treatment with compound 2 also produced abdomen and wing malformation in adults leading to impossibility to mate.  相似文献

Six lignans including two new lignans were obtained as the principal components of the Forsythia koreana flowers via silica gel (SiO2), octadecyl SiO2 (ODS) as well as Sephadex LH‐20 column chromatography. In addition to two new lignans, named koreanaside A ((7R,8S,7′R,8′S)‐7,7‐diepoxy‐5‐hydroxy‐3,3‐dimethoxylignan 4‐Oβ‐d ‐glucopyranoside) and koreanaside B ((7R,8S,7′S,8′R)‐7,9‐epoxy‐9,5,7‐trihydroxy‐3,3‐dimethoxylignan 4‐Oβ‐d ‐glucopyranoside), four known lignans were identified to be (+)‐phylligenin, (?)‐epipinoresinol, pinoresinol, and tinosposide A. The structures and absolute configurations of koreanasides A and B were established by means of analysis of spectroscopic data (NMR, IR, FAB‐MS, and CD), whereas the structures of known lignans were identified by comparison their NMR and MS values with those in the reported literature. Their chemical structures including configuration were established by means of analysis of spectroscopic data (NMR, IR, FAB‐MS, and CD) but also comparison of their NMR and MS values with those in the reported literature. This is the first article for isolation of six lignans of Fkoreana flowers. Koreanasides A and B showed high radical scavenging activity with oxygen radical absorbance capacity (ORAC) values of 0.97 ± 0.01 and 1.02 ± 0.01, respectively. Koreanaside A also prohibited expressing VCAM‐1 in MOVAS cells with 80.5% at 25 mg/mL.  相似文献

Helianthus tuberosus L. (Jerusalem artichoke) is cultivated in Europe and other parts of the world as a food crop and ornamental plant. The volatile oils of the aerial parts of H. tuberosus were investigated more than 30 years ago, but no study could be found to date on the constituents of the tuber essential oil. Herein, the first characterization by GC‐FID, GC/MS, and 13C‐NMR analyses of a hydrodistilled essential oil of Jerusalem artichoke tubers was reported. Fresh plant material collected in Serbia (Sample A) and a commercial sample (Sample B) yielded only small amounts of oil (0.0014 and 0.0021% (w/w), resp.). In total, 195 constituents were identified, representing 88.2 and 93.6% of the oil compositions for Samples A and B, respectively. The main constituents identified were β‐bisabolene ( 1 ; 22.9–30.5%), undecanal (0–12.7%), α‐pinene (7.6–0.8%), kauran‐16‐ol ( 2 ; 6.9–9.8%), 2‐pentylfuran (0.0–5.7%), and (E)‐tetradec‐2‐enal (0.0–4.9%). Several rare compounds characteristic for Helianthus ssp. were also detected: helianthol A ( 6 ; 2.1–1.9%), dihydroeuparin ( 10 ; 0.0–2.3%), euparin ( 9 ; 0.0–0.4%), desmethoxyencecalin ( 7 ; traces – 0.2%), desmethylencecalin ( 8 ; 0.0–0.4%), and an isomer of desmethylencecalin (0.0%‐traces). In vitro.MAO‐A activity was inhibited 16‐25%, and MAO‐B activity was inhibited 20‐50% by SE treatment (12.5, 25 and 50 μg), In vivo.male C57BL/6 mice Received intraperitoneal injection of SE (20 mg/kg/day) for 14 days. The results showed that MAO‐A activity of pre‐SE‐treatment mice brain was inhibited in whole brain, cerebral cortex, substantia nigra. MAO‐B activity of pre‐SE‐treatment mice brain was inhibited in substantia nigra and cerebellum than saline‐treated control group. These results suggest that SE inhibits MAO activity in vivo.which would be expected to results in anti‐depressive and neuroprotective effects.  相似文献

Intact ovarian follicles, obtained from untreated and human chorionic gonadotropin (HCG) treated Japanese yellowtail Seriola quinqueradiata during different maturational stages, were incubated with radioactive [3H]pregnenolone, [3H]17‐hydroxyprogesterone or [14C] androstenedione and steroid metabolites identified by thin layer chromatography (TLC) followed by recrystallization to constant specific activity. In untreated late vitellogenic (0 h) follicles, androstenedione was the major product with smaller amounts of testosterone and oestradiol‐17α. In post‐vitellogenic (12 h post‐injection) intact follicles, androstenedione predominated, and although testosterone and oestradiol‐17α were not produced, there were small amounts of 17, 20β‐dihydroxy‐4‐pregnen‐3‐one (17,20β‐P) and 17,21‐dihydroxy‐4‐pregnene‐3, 20‐dione (11‐deoxycortisol). In HCG‐treated fish, a steroidogenic shift resulted in the disappearance of testosterone and oestradiol‐17 coinciding with the appearance of 17, 20β‐P. During early and late final oocyte maturation FOM (24 and 36 h post‐injection), there was a five‐ to seven‐fold increase in the production of 17, 20β‐P, whereas production of 11‐deoxycortisol remained almost the same. During FOM, in addition to 17,20β‐P, its 5β‐reduced metabolite, 17,20β‐dihydroxy‐5β‐pregnan‐3‐one (5β‐17,20β‐P) was synthesized, suggesting a decrease in maturation‐inducing 17,20β‐P activity. 17, 20β,21‐Trihydroxy‐4‐pregnen‐3‐one (20β‐S) was not synthesized by ovarian fragments in Japanese yellowtail at any maturational stage. The metabolites identified on TLC during FOM were tested to evaluate their maturation‐inducing activity in an in vitro bioassay. Of the steroids tested, 17,20β‐P was the most effective inducer of germinal vesicle breakdown (GVBD), followed by 5β‐17,20β‐P. Timely synthesis of 17,20β‐P immediately prior to and during FOM as well as its great potency in inducing GVBD in vitro supports the evidence for a physiological role of 17,20β‐P as a maturation‐inducing hormone in Japanese yellowtail.  相似文献

House flies, Musca domestica L., were collected in copula over two summers from six dairies located in three climatically distinct regions in the U.S.A. southern California, Minnesota and Georgia. Ages of males and females from a total of 511 mating pairs were estimated using pterin analysis. Cuticular hydrocarbon profiles and gonotrophic ages of females also were evaluated. Mean age of mating males ranged from 54 to 102 degree‐days (DD) (4–10 days based on field air temperatures), depending on the farm. Very young males (< 10–20 DD) and old males (> 200 DD) were rare in mating pairs. Mean female age at mating ranged from 20 to 46 DD (2.5–4 days). All mating females had eggs in the early stages of vitellogenesis and 99.2% were nulliparous. However, some older and parous females were collected, demonstrating that re‐mating can occur in the field. Head width measurements of mating pairs suggested that assortative mating by size did not occur. The cuticular hydrocarbon profiles of females were determined, with emphasis on (Z)‐9‐tricosene (muscalure). Overall, only 55% of mating females had detectable amounts (> 4 ηg per fly) of (Z)‐9‐tricosene. Of the females that had detectable (Z)‐9‐tricosene, variation in amount per female was high in all fly populations, and thus was not statistically related to the size or age of the mating female. The proportion of mating females with detectable levels of (Z)‐9‐tricosene varied by geographic region. Seventy‐one, 63, and 27% of females from southern California, Minnesota and Georgia had detectable amounts of (Z)‐9‐tricosene. Principal components analysis of the eight most abundant hydrocarbons from mating females, by state, revealed state‐level distinctiveness of hydrocarbons in house fly populations, which may reflect genetic variation associated with environmental stresses in those geographical zones.  相似文献

The hydrodistilled essential oils obtained from aerial flowering parts of Teucrium stocksianum ssp. stocksianum (TSS) and T. stocksianum ssp. gabrielae (TSG) from Iran were analyzed by capillary GC and GC/MS. The oil analysis of two subspecies led to the identification of 65 compounds that accounted for 93.3 and 95.1% of the total oil compositions, respectively. Sesquiterpenoids (52.9%) constituted the main compounds in the essential oil of TSS represented mainly by cis‐sesquisabinene hydrate (12.0%), followed by epiβ‐bisabolol (6.6%), guaiol (5.4%), and β‐eudesmol (4.4%), whilst monoterpenoids (61.2%) were found to be the major components of the oil of TSG, represented by α‐pinene (23.0%), β‐pinene (13.0%), myrcene (6.3%), and sabinene (6.3%). The principal component in both subspecies, TSS and TSG, was α‐pinene (22.0 and 23.0%, resp.) and β‐pinene (6.5 and 13.0%, resp.). epiα‐Cadinol, myrcene, and sabinene, which were detected as principal compounds of TSG, were characterized in lower amounts (<1.5%) in the oil of TSS. Seven components were identified in the oil of TSS corresponding to 25.9% of total oil, which were totally absent in the oil of TSG, of which cis‐sesquisabinene hydrate (12.0%), guaiol (5.4%), and β‐eudesmol (4.4%) were in considerable amounts. Taxonomic position of the subspecies is discussed on the basis of phytochemical data.  相似文献

The algal class Chlorarachniophyceae is comprised of a small group of unicellular eukaryotic algae that are often characterized by an unusual amoeboid morphology. This morphology is hypothesized to be the result of a secondary endosymbiosis in which a green alga was engulfed as prey by a nonphotosynthetic amoeba or amoebaflagellate. Whereas much is known about the phylogenetic relationships of individual chlorarachniophytes to one another, and to possible ancestral host organisms in the genera Cercomonas and Heteromita, little is known about their physiology, particularly that of their lipids. In an initial effort to characterize the lipids of this algal class, seven organisms were examined for their fatty acid and sterol composition. These included Bigelowiella natans, Chlorarachnion globusum, Chlorarachnion reptans, Gymnochlora stellata, Lotharella amoeboformis, Lotharella globosa, and Lotharella sp. Fatty acids associated with chloroplast‐associated glycolipids, cytoplasmic membrane‐associated phospholipids, and storage triglycerides were characterized. Glycolipid fatty acids were found to be of limited composition, containing principally eicosapentaenoic acid [20:5(n‐3)] and hexadecanoic acid (16:0), which ranged in relative percentage from 67–90% and 10–29%, respectively, in these seven organisms. Triglyceride‐associated fatty acids were found to be similar. Phospholipid fatty acid composition was more variable. The principal phospholipid fatty acids, 16:0 (25–32%) and a compound tentatively identified as docosapentaenoic acid [22:5(n‐3)] (26–35%), were found along with a number of C18 and C20 fatty acids. All organisms contained two sterols as free sterols. These were tentatively identified as 24‐ethylcholesta‐5,22E‐dien‐3b‐ol (stigmasterol; 70–95%) and 24‐methylcholesta‐5,22E‐dien‐3b‐ol (brassicasterol; 5–30%).  相似文献

Aims: To investigate the distribution of staphylococcal enterotoxin genes (se) and the molecular features of community‐associated methicillin‐sensitive/resistant Staphylococcus aureus (CA‐MSSA/MRSA) isolates in the nostrils of healthy pets and their owners. Methods and Results: A total of 114 Staph. aureus isolates were identified from 1563 nasal swab samples, and CA‐MRSA accounted for 20·2% (n = 23) of the total identified isolates. CA‐MRSA isolates (91·3%, 21/23) harboured higher percentage of se than did CA‐MSSA isolates (58·2%, 53/91) (P < 0·01), and the two highest se profiles of CA‐MRSA were sebsekseq (42·9%, 9/21) and sebsekseqsep (28·6%, 6/21). Of the MSSAs, 42·8% (39/91) were resistant to at least one antimicrobial drug and 8·8% (8/91) were multidrug resistant (MDR). We identified nine staphylocoagulase (SC) types (I–VIII and X) and three multilocus sequence types (ST59‐MRSA‐IV/V, ST‐239‐MRSA‐V and ST241‐MRSA‐V). SC VII (23·4%, 22/94), a staphylococcal food poisoning isolate found mainly in Japan, and ST‐59‐MRSA‐IV/V (85%, 17/20), a widespread CA‐MRSA clone found mainly in Taiwan, both were the most predominant types. Phylogenetic analysis together with se and molecular characteristics obtained using pulsed‐field gel electrophoresis showed that high levels of antimicrobial resistance and the se‐carrying clone ST59‐MRSA‐IV/V‐SC VII were all clustered in genogroup 5. Conclusions: The CA‐MRSA clone of se‐carrying‐MDR‐ST‐59‐IV/V‐SC VII was identified predominantly in this study, and this clone might play a significant role in staphylococcal food poisoning in community settings. Significance and Impact of the Study: To our knowledge, this is the first study focussing on enterotoxin‐carrying CA‐MRSA/MSSA in pets and their owners, and the results support the future warnings in animal–human bond caused by CA‐staphylococci in the commonwealth and the need to take cautions worldwide.  相似文献

Temperature is expected to modify the effects of ultraviolet radiation (UVR) on photosynthesis by affecting the rate of repair. We studied the effect of short‐term (1 h) and long‐term (days) acclimation to temperature on UVR photoinhibition in the diatom Thalassiosira pseudonana Hasle et Heimdal. Photosynthesis was measured during 1 h exposures to varying irradiances of PAR and UVR + PAR at 15, 20, and 25°C, the latter corresponding to the upper temperature limit for optimal growth in T. pseudonana. The exposures allowed the estimation of photosynthesis–irradiance (P–E) curves and biological weighting functions (BWFs) for photoinhibition. For the growth conditions used, temperature did not affect photosynthesis under PAR. However, photoinhibition by UVR was highly affected by temperature. For cultures preacclimated to 20°C, the extent of UVR photoinhibition increased with decreasing temperature, from 63% inhibition of PAR‐only photosynthesis at 25°C to 71% at 20°C and 85% at 15°C. These effects were slightly modified after several days of acclimation: UVR photoinhibition increased from 63% to 75% at 25°C and decreased from 85% to 80% at 15°C. Time courses of photochemical efficiency (ΦPSII) under UVR + PAR were also fitted to a model of UVR photoinhibition, allowing the estimation of the rates of damage (k) and repair (r). The r/k values obtained for each temperature treatment verified the responses observed with the BWF (R2 = 0.94). The results demonstrated the relevance of temperature in determining primary productivity under UVR exposures. However, the results suggested that temperature and UVR interact mainly over short (hours) rather than long (days) timescales.  相似文献

Introduction – Naphthoquinones; lawsone ( 1 ), lawsone methyl ether ( 2 ) and methylene‐3,3′‐bilawsone ( 3 ) are the main active compounds of Impatiens balsamina leaves. Objective – To develop and validate an HPLC method for simultaneous quantitative determination of 1 – 3 in I. balsamina leaf extracts. Methodology – The method utilised a Supelco® C18 column (5 µm, 4.6 × 150 mm) at 25°C with the mixture of 2% aqueous acetic acid : methanol (gradient elution as follows: 0–10 min, 25 : 75; 10–20 min, 32 : 68; 20–35 min, 55 : 45) as the mobile phase at a flow‐rate of 1 mL/min, and UV detection at 280 nm. The parameters of linearity, repeatability, reproducibility, accuracy specificity and sensitivity of the method were evaluated. Results – The recovery of the method was 96–101% and linearity (r2 ≥ 0.9995) was obtained for all naphthoquinones. The composition and variability of the terpenes and their derivatives isolated from the needles of a representative pool of 114 adult trees originating from four natural populations of dwarf mountain pine (Pinus mugo Turra ) from the Julian Alps were investigated by GC‐FID and GC/MS analyses. In total, 54 of the 57 detected essential‐oil components were identified. Among the different compound classes present in the essential oils, the chief constituents belonged to the monoterpenes, comprising an average content of 79.67% of the total oil composition (74.80% of monoterpene hydrocarbons and 4.87% of oxygenated monoterpenes). Sesquiterpenes were present in smaller amounts (average content of 19.02%), out of which 16.39% were sesquiterpene hydrocarbons and 2.62% oxygenated sesquiterpenes. The most abundant components in the needle essential oils were the monoterpenes δ‐car‐3‐ene, β‐phellandrene, α‐pinene, β‐myrcene, and β‐pinene and the sesquiterpene β‐caryophyllene. From the total data set of 57 detected compounds, 40 were selected for principal‐component analysis (PCA), discriminant analysis (DA), and cluster analysis (CA). The overlap tendency of the four populations suggested by PCA, was as well observed by DA. CA also demonstrated similarity among the populations, which was the highest between Populations I and II.  相似文献

A colorimetric assay for antibacterial susceptibility testing of clinical isolates (Escherichia coli, Pseudomonas aeruginosa, Shigella dysenteriae, Staphylococcus aureus, Bacillus cereus, and Streptococcus pneumoniae) is described based on the reduction of a novel tetrazolium salt, 3‐(4,5‐dimethylthiazol‐2‐yl)‐5‐(3‐carboxymethoxyphenyl)‐2‐(4‐sulfophenyl)‐2H‐tetrazolium (MTS), in the presence of phenazine methosulfate (PMS) as an electron‐coupling agent. The combination of 200 μg/mL MTS with 25 μmol/L PMS resulted in production of large amounts of formazan within 1 h of exposure. In this setting, fractions extracted from Chinese Masson pine (Pinus massoniana Lamb.) needles damaged by the pine caterpillar Dendrolimus punctatus Walker were found to have enhanced levels of antibacterial activity. These fractions, which were designated “Master”, “Technique”, and “Strength”, were isolated and identified by reverse‐phase C18 cartridge concentration, gel filtration, and affinity chromatography. Two fractions purified from healthy and undamaged needles were designated H1 and H2, respectively. For all test bacteria species. Technique produced the lowest minimal inhibitory concentrations (MICs), ranging from 2 to 32 μg/mL, and H2 produced the highest values, with four of the six MICs being higher than 128 μg/mL We found that the Rmax model fitted the data well in that the r2 ranged between 0.87 and 0.96 (median, 0.92) and no statistically significant deviations from the model were found (P= 0.23). The median coefficient of variation of the log RC50 values and the slope m of the fitted model for all six strains among the replicates were 38 and 41%, respectively. In the course of the investigation, the physiological and functional factors involved in pest damage to plants were also explored. In summary, the MTS‐PMS colorimetric assay has advantages over existing methods for the examination of antibacterial activity, and could be developed further such that it would be suitable for screening new antibiotic molecules. ‘Beauty bush’ and ‘twin flower’ are common names attributed to two well‐recognizable species belonging to the genus Linnaea (16 spp.) – L. amabilis and L. borealis – long admired by botanists and gardeners for their perfumed paired bell‐shaped flowers. In the present study, we investigated their floral scent compositions through gas chromatography – mass spectrometry (GC‐MS) analysis of dynamic headspace samples. Because the flowers of L. borealis in wild populations are fragrant both during the day and in the evening, circadian variation of scent emission was also assessed for this species. In total, 26 chemical compounds comprise the floral scent bouquets of L. amabilis and L. borealis, identified as monoterpenes (14), benzenoids and phenylpropanoids (5), aliphatics (3), sesquiterpenes (3) and irregular terpenes (1). Whereas monoterpenes, notably (‐)‐α‐ and β‐pinene, dominated the scent of L. amabilis (over 82% relative abundance), benzene derivates: 1,4 dimethoxybenzene, anisaldehyde, 2‐phenylethanol, benzaldehyde and nicotinaldehyde were exclusive to analysed headspace samples of L. borealis, accounting for 52% to 100% of their relative compositions, in three Swedish populations. A southwestern Finnish population was characterized by the four first mentioned benzenoid compounds and large amounts of (‐)‐α‐ and β‐pinenes plus two aliphatic substances. The scent compounds identified for both species are ubiquitous and may serve as generalist attractants/stimulants for a broad assortment of anthophilous insects. The basic work on the flower scent of L. amabilis and L. borealis should inspire studies of their pollination biology, primarily the behaviour‐guiding roles of the characteristic emitted volatiles.  相似文献

The chemical composition of the essential oils isolated from the aerial parts of Anthemis pignattiorum Guarino, Raimondo & Domina and A. ismelia Lojac . and the aerial parts and flowers of Anthemis cupaniana Tod . ex Nyman , three endemic Sicilian species belonging to the section Hiorthia, was determined by GC‐FID and GC/MS analyses. (Z)‐Muurola‐4(14),5‐diene (27.3%) was recognized as the main constituent of the A. pignattiorum essential oil, together with isospathulenol (10.6%), sabinene (7.7%), and artemisyl acetate (6.8%), while in the oil obtained from the aerial parts of A. ismelia, geranyl propionate (8.8%), bornyl acetate (7.9%), β‐thujone (7.8%), neryl propionate (6.5%), and τ‐muurolol (6.5%) prevailed. α‐Pinene was the main compound of both the aerial part and flower oils of A. cupaniana (18.4 and 13.2%, resp.). Also noteworthy are the considerable amounts of artemisyl acetate (12.7%) and β‐thujone (11.8%) found in the oil from the aerial parts and those of tricosane (9.8%) and sabinene (7.6%) evidenced in the flower oil. Furthermore, an update on the main compounds identified in the essential oils of all the Anthemis taxa studied so far was presented, and cluster analyses were carried out, to compare the essential oils of these taxa.  相似文献
