Enzymatic production of methyl esters (biodiesel) by methanolysis of palm oil in presence and absence of organic solvent was investigated using Candida antarctica lipase immobilized on acrylic resin as a biocatalyst. Although, at least molar equivalent of methanol (methanol-palm oil ratio 3:1) is required for the complete conversion of palm oil to methyl esters, lipase catalyzed methanolysis of palm oil in absence of organic solvent was poisoned by adding more than 1/3 molar equivalent of methanol. The use of polar organic solvents prevented the lipase to be poisoned in methanolysis with a molar equivalent of methanol, and tetrahydrofuran (THF) was found to be the most effective. The presence of water in methanolysis of palm oil both in presence and absence of THF inhibited the reaction rate but this inhibition was considerably low in THF containing system. The palm oil-lipase (w/w) ratio significantly influenced the activity of lipase and the optimal ratio in presence and absence of THF was 100 and 50, respectively.  相似文献

A highly active whole cell lipase (WCL) for efficient methanolysis of palm oil (PO) to biodiesel (BD) was prepared by isolation, cultivation and immobilization of lipase producing fungi. Fungi were screened from soil and the best isolate (PDA-6) identified as Aspergillus nomius exhibited maximum WCL methanolysis activity (1.4 g h−1 g−1) when inexpensive waste cooking oil was used as carbon source. The maximum BD yield with PDA-6 WCL reached 95.3% after 40 h at a lipase load 10% (w/w) of PO and methanol to PO molar ratio 5:1. The immobilization of PDA-6 cells within biomass suspended particle (BSP) made of polyurethane foam improved the repeated use of WCL and the remaining activity after 10 cycles was 88.2%. The PDA-6 WCL was more active in methanolysis of PO to BD than most WCLs previously reported. The newly isolated A. nomius is not only potential for producing WCL but also utilizing waste cooking oil.  相似文献

Background, Aims and Scope

The consequential approach to system delimitation in LCA requires that consideration of the technologies and suppliers included are ‘marginal’, i.e. that they are actually affected by a change in demand. Furthermore, coproduct allocation must be avoided by system expansion. Vegetable oils constitute a significant product group included in many LCAs that are intended for use in decision support. This article argues that the vegetable oil market has faced major changes around the turn of the century. The aim of this study is to study the marginal supply of vegetable oil as it has shifted to palm oil and describe the product system of the new supply.

Methods

The methods for identification of marginal technologies and suppliers and for avoiding co-product allocation are based on the work of Weidema (2003). The marginal vegetable oil is identified on the basis of agricultural statistics on production volumes and prices. A co-product from palm oil production is palm kernel meal, which is used for fodder purposes where it has two main properties: protein and energy. When carrying out system expansion, these properties are taken into account.

Results

The major vegetable oils are soy oil, palm oil, rapeseed oil and sun oil. These oils are substitutable within the most common applications. Based on market trends, a shift from rapeseed oil to palm oil as the marginal vegetable oil is identified around the year 2000, when palm oil turns out to be the most competitive oil. It is recommended to regard palm oil and its dependent co-product palm kernel oil as the marginal vegetable oil. The analysis of the product system shows that the demand for 1 kg palm oil requires 4.49 kg FFB (oil palm fruit) and the displacement of 0.035 kg soybeans (marginal source of fodder protein) and 0.066 kg barley (marginal source of fodder energy).

Discussion

The identification of the marginal vegetable oil and the avoidance of co-product allocation by system expansion showed that several commodities may be affected when using the consequential approach. Hence, the product system for vegetable oils is relatively complex compared to traditional LCAs in which average technologies and suppliers are applied and in which co-product allocation is carried out by applying an allocation factor.

Conclusions

This article presents how the marginal vegetable oil can be identified and that co-product allocation between oils and meal can be avoided by system expansion, by considering the energy and protein content in the meal, which displaces a mix of the marginal sources of energy and protein for animal fodder (barley and soy meal, respectively).

Recommendations and Perspectives

The implication of a shift in the marginal vegetable oil is significant. Many LCAs on rapeseed oil have been conducted and are being used as decision support in the bio energy field. Thus, based on consequential LCA methodology, it is argued that these LCAs need to be revised, since they no longer focus on the oil actually affected.

Biosurfactant production by Pseudomonas aeruginosa A41, a strain isolated from seawater in the gulf of Thailand, was examined when grown in defined medium containing 2% vegetable oil or fatty acid as a carbon source in the presence of vitamins, trace elements and 0.4% NH(4)NO(3), at pH 7 and 30 degrees C with 200 rpm-shaking for 7 days. The yield of biosurfactant steadily increased even after a stationary phase. Under such conditions the surface tension of the medium was lowered from 55-70 mN/m to 27.8-30 mN/m with every carbon source tested. However, types of carbon sources were found to affect biosurfactant yield. The yields of rhamnolipid biosurfactant were 6.58 g/L, 2.91 g/L and 2.93 g/L determined as rhamnose content when olive oil, palm oil and coconut oil, respectively, were used as a carbon source. Among them, biosurfactant obtained from palm oil was the best in lowering surface tension of the medium. Increase in biosurfactant activities in terms of oil displacement test and rhamnose content were observed to be higher with shorter chain fatty acids than that of the longer chains (C12>C14>C16). In addition, we found that C18:2, highly unsaturated fatty acid, showed higher oil displacement activity and rhamnose content than that of C18:1. The optimal oil displacement activity was found at pH 7-9 and in the presence of 0.5-3% NaCl. The oil displacement activity was stable to temperatures up to 100 degrees C for 15 h. Surface tension reduction activity was relatively stable at pH 2-12 and 0-5% of NaCl. Emusification activity tested with various types of hydrocarbons and vegetable oils showed similarity of up to 60% stability. The partially purified biosurfactant via TLC and silica gel column chromatography gave three main peaks on HPLC with mass spectra of 527, 272, and 661 m/z respectively, corresponding to sodium-monorhamnodecanoate, hydroxyhexadecanoic acid and an unknown compound, respectively.  相似文献

In this paper, whole cell (Rhizopus oryzae IFO 4697) was adopted directly as biocatalyst for biodiesel production. Effects of carbon source on cell growth and whole cell-catalyzed methanolysis of vegetable oils for biodiesel production were studied. The results showed that different oils contained in the cultivation medium had varied effects on the whole cell-catalyzed methanolysis of oils; with some specified oil as the carbon source for cell cultivation, those cells expressed higher catalytic activity in catalyzing the transesterification of the same oil for biodiesel production. The initial reaction rate was increased notably (204%) with oil pretreatment on the cells before catalyzing the reaction, which was possibly due to the improved mass transferring of substrates. A lipase from Candida sp., suitable for transesterification of fats and oils to produce fatty acid methyl ester (FAME), was immobilized on a cheap cotton membrane, in this paper. The conversion ratio of salad oil to biodiesel could reach up to 96% with the optimal reaction conditions. Continuous reaction in a fixed bed reactor was also investigated. A three-step transesterification with methanol (methanolysis) of oil was conducted by using a series of nine columns packed with immobilized Candida sp. 99–125 lipase. As substrate of the first reaction step, plant or waste oil was used together with 1/3 molar equivalent of methanol against total fatty acids in the oil. Mixtures of the first- and second-step eluates and 1/3 molar equivalent of methanol were used for the second- and third-reaction steps. A hydrocyclone was used in order to on-line separate the by-product glycerol after every 1/3 molar equivalent of methanol was added. Petroleum ether was used as solvent (3/2, v/v of oil) and the pump was operated with a flow rate of 15 L/h giving an annual throughput of 100 t. The final conversion ratio of the FAME from plant oil and waste oil under the optimal condition was 90% and 92%, respectively. The life of the immobilized lipase was more than 10 days. This new technique has many strongpoints such as low pollution, environmentally friendly, and low energy costs.  相似文献

Kinetics of production of biodiesel by enzymatic methanolysis of vegetable oils using lipase has been investigated. A mathematical model taking into account the mechanism of the methanolysis reaction starting from the vegetable oil as substrate, rather than the free fatty acids, has been developed. The kinetic parameters were estimated by fitting the experimental data of the enzymatic reaction of sunflower oil by two types of lipases, namely, Rhizomucor miehei lipase (RM) immobilized on ion-exchange resins and Thermomyces lanuginosa lipase (TL) immobilized on silica gel. There was a good agreement between the experimental results of the initial rate of reaction and those predicted by the proposed model equations, for both enzymes. From the proposed model equations, the regions where the effect of alcohol inhibition fades, at different substrate concentrations, were identified. The proposed model equation can be used to predict the rate of methanolysis of vegetable oils in a batch or a continuous reactor and to determine the optimal conditions for biodiesel production.  相似文献

As the world population grows, the demand for food increases. Although vegetable oils provide an affordable and rich source of energy, the supply of vegetable oils available for human consumption is limited by the "fuel vs food" debate. To increase the nutritional value of vegetable oil, metabolic engineering may be used to produce oil crops of desirable fatty acid composition. We have isolated and characterized β-ketoacyl ACP-synthase II (KASII) cDNA from a high-oleic acid palm, Jessenia bataua. Jessenia KASII (JbKASII) encodes a 488-amino acid polypeptide that possesses conserved domains that are necessary for condensing activities. When overexpressed in E. coli, recombinant His-tagged JbKASII was insoluble and non-functional. However, Arabidopsis plants expressing GFP-JbKASII fusions had elevated levels of arachidic acid (C20:0) and erucic acid (C22:1) at the expense of stearic acid (C18:0) and oleic acid (C18:1). Furthermore, JbKASII failed to complement the Arabidopsis KASII mutant, fab1-2. This suggests that the substrate specificity of JbKASII is similar to that of ketoacyl-CoA synthase (KCS), which preferentially elongates stearic and oleic acids, and not palmitic acid. Our results suggest that the KCS-like JbKASII may elongate C18:0 and C18:1 to yield C20:0 and C22:1, respectively. JbKASII may, therefore, be an interesting candidate gene for promoting the production of very long chain fatty acids in transgenic oil crops.  相似文献

Studies conducted with various inexpensive carbon sources such as whey, vegetable oils (palm, mustard, soybean and coconut), a low-cost source of glucose-D, rice and wheat bran, and mustard and palm oil cakes demonstrated palm oil as the best substrate for accumulation of a novel short-chain-length–long-chain-length polyhydroxyalkanoate (SCL–LCL-PHA) co-polymer containing SCL 3HAs [3-hydroxybutyric acid (3HB) and 3-hydroxyvaleric acid (3HV)] and LCL 3HAs of 3-hydroxyhexadecanoic acid (3HHD) and 3-hydroxyoctadecanoic acid (3HOD) units as constituents by a sludge-isolated Pseudomonas aeruginosa MTCC 7925. The co-polymer content reached up to 60% of dry cell weight (dcw) at 48 h of incubation in 0.5% (v/v) palm oil and the extract of 0.5% (v/v) palm oil cake supplemented vessels. The PHAs pool was further enhanced up to 69 and 75% (dcw), when the above culture was subjected to P- and N-limitation, respectively. The mol fraction of 3HB:3HV:3HHD:3HOD units were, respectively, 83.1:7.7:3.8:5.4 and 87.3:5.1:3.6:4.0 in P- and N-limited cultures. Consequently, a co-polymer yield of 5 g l−1 (approx.) was achieved, which was about 80-fold higher as compared to 69 mg l−1 of the control culture. On substrate basis, the accumulation reached up to 0.62 g PHAs per g substrate, which was significantly higher as compared to the yield obtained from starch by Haloferax mediterranei and Azotobacter chroococum, from molasses by A. vinelandii UWD, and from lactose and xylose by Pseudomonas cepacia. This novel P(3HB-co-3HV-co-3HHD-co-3HOD) co-polymer exhibited better thermal and mechanical properties as revealed from the differential scanning calorimetry and mechanical property studies, thus opens up new possibilities for various industrial applications.  相似文献

The enzymatic production of biodiesel by methanolysis of cottonseed oil was studied using immobilized Candida antarctica lipase as catalyst in t-butanol solvent. Methyl ester production and triacylglycerol disappearance were followed by HPLC chromatography. It was found, using a batch system, that enzyme inhibition caused by undissolved methanol was eliminated by adding t-butanol to the reaction medium, which also gave a noticeable increase of reaction rate and ester yield. The effect of t-butanol, methanol concentration and temperature on this system was determined. A methanolysis yield of 97% was observed after 24h at 50 degrees C with a reaction mixture containing 32.5% t-butanol, 13.5% methanol, 54% oil and 0.017 g enzyme (g oil)(-1). With the same mixture, a 95% ester yield was obtained using a one step fixed bed continuous reactor with a flow rate of 9.6 mlh(-1) (g enzyme)(-1). Experiments with the continuous reactor over 500 h did not show any appreciable decrease in ester yields.  相似文献

A strain of Aspergillus niger isolated from atmospherically exposed bread and Jatropha curcas seed was utilized as a whole‐cell biocatalyst for palm oil methanolysis to produce fatty acid methyl esters (FAME), or biodiesel. The A. niger strain had a lipase activity of 212.58 mU mL?1 after 144 h incubation at 25 °C with an initial pH value of 6.5, using 7% polypeptone (w/w on basal medium) as the nitrogen source and 3% olive oil (w/w on basal medium) as a carbon source. The A. niger cells spontaneously immobilized within polyurethane biomass support particles (BSPs) during submerged fermentation. Thereafter, the methanolysis of palm oil was achieved via a three‐step addition of methanol in the presence of BSPs‐immobilized with A. niger cells. The influence of water content, reaction temperature and enzyme concentration on reaction rate was investigated. An 8% water content and a temperature of 40 °C in the presence of 30 immobilized BSPs, resulted in an 87% FAME yield after 72 h.  相似文献

Biodiesel is an alternative to petroleum-based conventional diesel fuel and is defined as the mono-alkyl esters of vegetable oils and animal fats. Biodiesel has been prepared from numerous vegetable oils, such as canola (rapeseed), cottonseed, palm, peanut, soybean and sunflower oils as well as a variety of less common oils. In this work, Moringa oleifera oil is evaluated for the first time as potential feedstock for biodiesel. After acid pre-treatment to reduce the acid value of the M. oleifera oil, biodiesel was obtained by a standard transesterification procedure with methanol and an alkali catalyst at 60 degrees C and alcohol/oil ratio of 6:1. M. oleifera oil has a high content of oleic acid (>70%) with saturated fatty acids comprising most of the remaining fatty acid profile. As a result, the methyl esters (biodiesel) obtained from this oil exhibit a high cetane number of approximately 67, one of the highest found for a biodiesel fuel. Other fuel properties of biodiesel derived from M. oleifera such as cloud point, kinematic viscosity and oxidative stability were also determined and are discussed in light of biodiesel standards such as ASTM D6751 and EN 14214. The (1)H NMR spectrum of M. oleifera methyl esters is reported. Overall, M. oleifera oil appears to be an acceptable feedstock for biodiesel.  相似文献

【目的】探讨复合酶协同催化体系在含水量较高的体系中催化油脂制备生物柴油的工艺条件。【方法】通过基因工程手段在毕赤酵母中分别高效分泌表达南极假丝酵母脂肪酶(CALB)和米根霉脂肪酶(ROL),构建CALB和ROL复合酶协同催化体系制备生物柴油,利用单因素实验优化工艺条件,以甲酯化得率作为复合酶协同催化体系效能的评价标准。【结果】优化工艺条件为:CALB?ROL最佳复合酶配比为7?3,每克大豆油中加入16 U的复合脂肪酶,甲醇与大豆油摩尔比为4?1,并按0 h时2?1醇油摩尔比,12 h和24 h时以1?1醇油摩尔比分批加入甲醇,含水量为30%-60%之间,40°C反应29-34 h,甲酯得率达到93%。【结论】该复合酶协同催化体系对环境友好,与常规酶法制备生物柴油工艺相比对酶的使用量和催化时间减少幅度都在50%以上,本复合酶协同催化体系能有效降低生物柴油制备成本,具有较好的工业化应用前景。  相似文献

Optimal conditions for enzymatic synthesis of biodiesel from palm oil and ethanol were determined with lipase from Pseudomonas fluorescens immobilized on epoxy polysiloxane–polyvinyl alcohol hybrid composite under a microwave heating system. The main goal was to reduce the reaction time preliminarily established by a process of conventional heating. A full factorial design assessed the influence of ethanol-to-palm oil (8:1–16:1) molar ratio and temperature (43–57 °C) on the transesterification yield. Microwave irradiations varying from 8 to 15 W were set up according to reaction temperature. Under optimal conditions (8:1 ethanol-to-oil molar ratio at 43 °C), 97.56 % of the fatty acids present in the palm oil were converted into ethyl esters in a 12-h reaction, corresponding to a productivity of 64.2 mg ethyl esters g?1 h?1. This represents a sixfold increase from the process carried out under conventional heating, thus proving to be a potential tool for enhancing biochemical modification of oils and fats. In general, advantages of the new process include: (1) microwaves speed up the enzyme-catalyzed reactions; (2) there are no destructive effects on the enzyme properties, such as stability and substrate specificity, and (3) the microwave assistance allows the entire reaction volume to be heated uniformly. These bring benefits of a low energy demand and a faster conversion of palm oil into biodiesel.  相似文献

Fatty acids esters were produced from two Nigerian lauric oils, palm kernel oil and coconut oil, by transesterification of the oils with different alcohols using PS30 lipase as a catalyst. In the conversion of palm kernel oil to alkyl esters (biodiesel), ethanol gave the highest conversion of 72%, t-butanol 62%, 1-butanol 42%, n-propanol 42% and iso-propanol 24%, while only 15% methyl ester was observed with methanol. With coconut oil, 1-butanol and iso-butanol achieved 40% conversion, 1-propanol 16% and ethanol 35%, while only traces of methyl esters were observed using methanol. Studies on some fuel properties of palm kernel oil and its biodiesel showed that palm kernel oil had a viscosity of 32.40 mm2/s, a cloud point of 28 degrees C and a pour point of 22 degrees C, while its biodiesel fuel had a viscosity of 9.33 mm2/s, a cloud point of 12 degrees C and a pour point of 8 degrees C. Coconut oil had a viscosity of 28.58 mm(2)/s, a cloud point of 27 degrees C and a pour point of 20 degrees C, while its biodiesel fuel had a viscosity of 7.34 mm2/s, a cloud point of 5 degrees C and a pour point of -8 degrees C. Some of the fuel properties compared favourably with international biodiesel specifications.  相似文献

Glycerolysis of palm and palm kernel oils were conducted using a commercial 1,3-specific lipase from Humicola lanuginosa (trade name: SP 398) as catalyst (500 units lipase g–1 oil) at 40°C and oil:glycerol (1:2 mol mol–1) in a solvent-free system. After 24 h, the glycerolysis products of palm and palm kernel oils consisted of 23% triacylglycerols, 18% monoacylglycerols, 38% diacylglycerols and 18% triacylglycerols, 31% monoacylglycerols, 42% diacylglycerols, respectively. The monoacylglycerol fraction of the glycerolysis product of palm oil was enriched in oleic acid. Palmitic acid content of the monoacylglycerol fraction of the same product was less than that of the original oil. Under the same conditions, monacylglycerol fraction of the palm kernel oil glycerolysis product was enriched in palmitic, stearic and oleic acids.  相似文献

Biocatalysis by immobilized lipase is an efficient alternative process for conversion of crude vegetable oil with high free fatty acid content to biodiesel, which is the limit of the conventional alkaline-catalyzed reaction. In this study, influences of solid-state organic and inorganic buffer core matrices with different pKa on catalytic performance of cross-linked protein coated microcrystalline biocatalysts prepared from Thermomyces lanuginosus lipase (CL-PCMC-LIP) toward esterification of palmitic acid (PA), transesterification of refined palm oil (RPO), and co-ester/transesterification of crude palm oil (CPO) to fatty acid methyl ester (FAME) was studied. Glycine, CAPSO (3-(cyclohexylamino)-2-hydroxy-1-propanesulfonic acid), and TAPS ([(2-hydroxy-1,1-bis(hydroxymethyl)ethyl)amino]-1-propanesulfonic acid) were shown to be potent core matrices for these reactions. The optimal reaction contained 4:1 [methanol]/[fatty acid] molar equivalence ratio with 20% (w/w) CL-PCMC-LIP on glycine in the presence of tert-butanol as a co-solvent. Deactivation effect of glycerol on the biocatalyst reactive surface was shown by FTIR, which could be alleviated by increasing co-solvent content. The maximal FAME yields from PA, RPO, and CPO reached 97.6, 94.9, and 95.5%, respectively on a molar basis under the optimum conditions after incubation at 50 °C for 6 h. The biocatalyst retained >80% activity after recycling in five consecutive batches. The work demonstrates the potential of CL-PCMC-LIP on one-step conversion of inexpensive crude fatty acid-rich feedstock to biodiesel.  相似文献

This paper selects biofuel scenarios to substitute diesel in Brazil based on oil reserves increase, diesel imports, CO2 emissions, crops agronomic yields, byproducts marketing and social impacts. This hard task still considers that agricultural practices in developing countries have large social impacts. Brazil presents high consumption of diesel oil in transport; low agronomic yield of traditional vegetable oil crops, which demand large cultivation areas contrasting with microalgae and palm oils which present high productivity. Concerning technologies, thermal cracking and transesterification of vegetable oils present a difficult economic situation related to vegetable oils price, food competition and glycerin market; BTL technology, meaning thermal gasification of biomass to liquids, faces problems related to low density of biomaterials and low viscosity of synthetic biodiesel produced. Biorefinery algal integrated systems and co-solvent technology to introduce up to 8% of ethanol into diesel seem to be feasible routes to reduce diesel consumption.  相似文献

SC_9-4是以植物油为原料研制成功的一种高速冷轧工艺润滑油。经武钢高速冷轧机轧制性能鉴定,它的高速轧制性能优于棕榈油。我们使用高效液相色谱比较分析鉴定了SC_9-4和棕榈油的甘油三酯组成。分析结果:SC_9-4:的主要甘油三酯是POP(42.8%)和PPP(14.6%),而棕榈油的主要甘油三酯是POP(31.3%)、POO(20.1%)、PLP(12.2%)和PLO(11.6%)。我们初步探讨甘油三酯组成与轧制性能关系认为,SC_9-4的高速轧制性能优于棕榈油,可能是和SC_9-4含PPP、PPS和PSS的含量(18.8%)高于棕榈油(9.9%)有关。  相似文献
