A Gram-positive, aerobic, non-motile actinobacterial strain, designated YIM C01235T, was isolated from a soil sample collected from the Swallow Cave, Yunnan province, south-west China. The isolate grew at 10–30 °C, pH 6.0–9.0 and 0–8 % (w/v) NaCl. Phylogenetic analysis based on 16S rRNA gene sequences revealed highest similarity to Saccharopolyspora gloriosae YIM 60513T (96.8 %), and lower 16S rRNA gene sequence similarities (95.1–96.7 %) with the other species of the genus Saccharopolyspora. The whole-cell hydrolysates contained meso-diaminopimelic acid (meso-DAP), arabinose and galactose. The polar lipids were diphosphatidylglycerol, phosphatidylglycerol, phosphatidylcholine, phosphatidylethanolamine, phosphatidylmethylethanolamine, two unknown glycolipids, two unknown phospholipids and one polar lipid. MK-9(H4) was the predominant menaquinone. The major fatty acids were iso-C15:0, iso-C16:0, anteiso-C15:0 and anteiso-C17:0. Mycolic acids were not detected. The genomic DNA G+C content was 69.1 mol%. These chemotaxonomic data, together with its morphological properties, were consistent with the assignment of strain YIM C01235T to the genus Saccharopolyspora. The results of physiological and biochemical tests allowed strain YIM C01235T to be differentiated phenotypically from all the recognized Saccharopolyspora species. On the basis of evidence from this polyphasic study, the novel species Saccharopolyspora cavernae sp. nov. is proposed. The type strain is YIM C01235T (=DSM 45825T = CCTCC AA 2012022T).  相似文献

A novel actinomycete, designated strain NEAU-NH11T, was isolated from muddy soil collected from a lake and characterized using a polyphasic approach. The 16S rRNA gene sequence analysis showed that strain NEAU-NH11T belongs to the genus Streptosporangium, and was most closely related to Streptosporangium amethystogenes subsp. amethystogenes DSM 43179T (99.0 %). Phylogenetic analysis based on the 16S rRNA gene sequence indicated that strain NEAU-NH11T formed a monophyletic clade with Streptosporangium purpuratum CY-15110T (98.3 %) and Streptosporangium yunnanense CY-11007T (98.0 %), an association that was supported by a bootstrap value of 80 % in the neighbour-joining tree and also recovered with the maximum-likelihood algorithm. However, the low level of DNA–DNA relatedness allowed the strain to be differentiated from S. amethystogenes subsp. amethystogenes DSM 43179T, S. purpuratum CY-15110T and S. yunnanense CY-11007T. Moreover, strain NEAU-NH11T could also be differentiated from its closest related strains by phenotypic characteristics. Therefore, it is proposed that strain NEAU-NH11T represents a novel Streptosporangium species, Streptosporangium nanhuense sp. nov. The type strain of S. nanhuense is NEAU-NH11T. A Gram-positive, aerobic, non-motile, non-acid-alcohol-fast strain, designated YIM 61095(T), was isolated from the root of Maytenus austroyunnanensis collected from a tropical rainforest of Xishuangbanna in Yunnan Province, south-west China. Strain YIM 61095(T) exhibited chemotaxonomic and morphological characteristics that were consistent with members of the genus Saccharopolyspora. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain YIM 61095(T) was a member of the genus Saccharopolyspora and was most closely related to Saccharopolyspora flava AS 4.1520(T) (97.7% sequence similarity). The major fatty acids were iso-C(15:0), iso-C(16:0), iso-C(17:0) and anteiso-C(17:0). The predominant quinone detected was MK-9(H(4)). The DNA G+C content was 66.2 mol%. The phenotypic characteristics and DNA-DNA hybridization relatedness data indicated that strain YIM 61095(T) should be distinguished from Saccharopolyspora flava AS 4.1520(T). On the basis of the evidence presented in this study, strain YIM 61095(T) represents a novel species of the genus Saccharopolyspora, for which the name Saccharopolyspora endophytica sp. nov. is proposed. A novel actinomycete, designated strain NEAU-GH7T, was isolated from a lake sediment and characterized using a polyphasic approach. Strain NEAU-GH7T was Gram-stain positive, aerobic, non-spore-forming and produced spherical sporangia. Phylogenetic analysis based on the 16S rRNA gene sequence indicated that strain NEAU-GH7T formed a monophyletic clade with the closest relative Streptosporangium longisporum DSM 43180T (99.0 %), an association that was supported by a bootstrap value of 74 % in the neighbour-joining tree and also recovered with the maximum-likelihood algorithm. However, the low level of DNA–DNA relatedness allowed the strain to be differentiated from its closest relative. Moreover, strain NEAU-GH7T could also be differentiated from S. longisporum DSM 43180T and other Streptosporangium species showing high 16S rRNA gene sequence similarity (>98.0 %) by morphological and physiological characteristics. On the basis of phylogenetic analysis, DNA–DNA hybridization and phenotypic characteristics, strain NEAU-GH7T should be classified as a new species of the genus Streptosporangium, for which the name Streptosporangium shengliensis sp. nov. is proposed. A novel actinomycete, strain NEAU-st1T, was isolated from a soil sample collected in Shaanxi province, Northwest China and characterized using a polyphasic approach. 16S rRNA gene sequence similarity studies showed that strain NEAU-st1T belongs to the genus Nonomuraea, being most closely related to Nonomuraea rosea GW12687T (98.91 %), Nonomuraea solani NEAU-Z6T (98.44 %), Nonomuraea rhizophila YIM67092T(98.24 %) and Nonomuraea monospora PT708T (98.02 %); similarities to sequences of other type strains of the genus Nonomuraea were lower than 98 %. Both tree-making algorithms used also supported the position that strain NEAU-st1T formed a distinct clade with its most closely related species. Morphological and physiological characteristics confirmed that the strain belongs to the genus Nonomuraea and distinguished it from its most closely related species. DNA–DNA hybridization further differentiated strain NEAU-st1T from its nearest phylogenetic neighbours. These results suggested that strain NEAU-st1T represents a novel species of the genus Nonomuraea, for which the name Nonomuraea shaanxiensis sp. nov. is proposed. A strain of Nonomuraea was isolated from Maheshkhali, Cox's Bazar, an unexplored region of Bangladesh. Strain 16-5-14(T) is a Gram-positive, aerobic, non-motile actinomycete that formed branched substrate and aerial mycelia. On the basis of 16S rRNA gene sequence similarity studies, strain 16-5-14(T) was shown to belong to the genus Nonomuraea, being most closely related to Nonomuraea kuesteri. Chemotaxonomic data supported allocation of the strain as a member of the genus Nonomuraea. The strain 16-5-14(T) contained MK-9(H(4)) as the major menaquinone, the polar lipid was phosphatidylethanolamine and major cellular fatty acids were observed as C(16 : 0 )(15.5%), iso-C(16 : 0) (13.8%) and 10-methyl C(17 : 0) (9.6%). Results of DNA-DNA hybridization and physiological tests allowed genotypic and phenotypic differentiation of strain 16-5-14(T) from closely related species N. kuesteri. Thus 16-5-14(T) represents a novel species of the genus Nonomuraea. On the basis of evaluation of the morphological, physiological and chemotaxonomic characteristics, 16S rRNA gene sequence comparisons and DNA-DNA hybridization, Nonomuraea maheshkhaliensis sp. nov. (type strain, 16-5-14(T)=JCM 13929(T)=MTCC 8545(T)) is proposed.  相似文献

A novel halophilic, filamentous actinomycete, designated TRM 4064T, was isolated from a hypersaline habitat in Sichuan Province, China. Phylogenetic analysis based on an almost-complete 16S rRNA gene sequence of strain TRM 4064T showed that it was most closely related to Actinopolyspora mortivallis (99.1 % sequence similarity). The sequence similarities between strain TRM 4064T and other Actinopolyspora species with validly-published names were <97.0 %. However, it had relatively low mean values for DNA–DNA relatedness with the A. mortivallis DSM 44261T (23.2 %). Optimal growth occurred at 37 °C, pH 7.0 and in the presence of 13 % (w/v) NaCl. The whole-cell sugar pattern consists of xylose, glucose, ribose and arabinose. The predominant menaquinones are MK-10(H4) (38.2 %), MK-9(H4) (25.1 %), MK-9(H2) (28.6 %) and MK-8(H4) (7.3 %). The major fatty acids are anteiso-C17:0 (36.9 %) and iso-C17:0 (19.3 %). The diagnostic phospholipids detected were diphosphatidylglycerol (DPG), phosphatidylglycerol (PG), phosphatidylcholine (PC), phosphatidylinositol (PI) and two unknown phospholipids. The G+C content of the genomic DNA of the type strain is 66.3 mol%. Strain TRM 4064T therefore represents a novel species of the genus Actinopolyspora, for which the name Actinopolyspora dayingensis sp. nov. is proposed. A halophilic actinomycete strain designated H19(T), was isolated from a Saharan soil in the Bamendil region (Ouargla province, South Algeria) and was characterized taxonomically by using a polyphasic approach. The morphological and chemotaxonomic characteristics of the strain were consistent with those of members of the genus Actinopolyspora, and 16S rRNA gene sequence analysis confirmed that strain H19(T) was a novel species of the genus Actinopolyspora. DNA-DNA hybridization value between strain H19(T) and the nearest Actinopolyspora species, A. halophila, was clearly below the 70?% threshold. The genotypic and phenotypic data showed that the organism represents a novel species of the genus Actinopolyspora for which the name Actinopolyspora algeriensis sp. nov. is proposed, with the type strain H19(T) (=?DSM 45476(T)?=?CCUG 62415(T)).  相似文献

An alkaliphilic actinobacterium, designated EGI 80050T, was isolated from a desert soil sample of Xinjiang, north-west China, and characterized by a polyphasic approach. The isolate was observed to produce purple orange-yellow aerial mycelium and dark orange-yellow substrate mycelium on yeast extract-malt extract agar medium. Whole-cell hydrolysates of strain EGI 80050T were found to contain ll-diaminopimelic acid as the diagnostic diamino acid, and galactose, glucose, rhamnose and mannose as the main sugars. The major fatty acids identified were C16:0-iso (36.8 %), C15:0-anteiso (17.3 %), 15:0-iso (13.2 %) and 14:0-iso (10.5 %). The predominant menaquinones detected were MK-9(H6) and MK-9(H8), while the characteristic polar lipids were identified as diphosphatidylglycerol, phosphatidylglycerol, phosphatidylinositol, phosphatidylinositol mannosides, phosphatidylmethylethanolamine and three unknown phospholipids. The G+C content of the genomic DNA was determined to be 67.9 mol%. Phylogenetic analysis based on 16S rRNA gene sequences affiliated the strain EGI 80050T to the genus Streptomyces. Levels of 16 rRNA gene sequence similarities between strain EGI 80050T and Streptomyces candidus NRRL ISP-5141T, Streptomyces cremeus NBRC 12760T, Streptomyces spiroverticillatus NBRC 12821T, Streptomyces violaceorectus NBRC 13102T, Streptomyces cinereoruber subsp. cinereoruber NBRC 12756T were 96.7, 96.6, 96.6, 96.6 and 96.6 %, respectively. Based on the phenotypic, chemotaxonomic and phylogenetic data, strain EGI 80050T is considered to represent a novel species of the genus Streptomyces, for which the name Streptomyces fukangensis sp. nov. (type strain EGI 80050T = BCRC 16945T = JCM 19127T) is proposed.  相似文献

Morphological and chemotaxonomic characterization of actinomycete strain S582 isolated from the gut of a termite (Speculitermes sp.) in Pathum Thani Province, Thailand, clearly demonstrated that this strain is a member of the genus Saccharopolyspora. 16S rDNA sequence analysis for the strain supported the assignment of the strain to the genus Saccharopolyspora. The similarity value of sequences between this strain and the closely related species Saccharopolyspora endophytica was 99.5%. The DNA G+C content was 70.2 mol%. DNA-DNA hybridization results (53.3%) and some physiological and biochemical properties indicated that strain S582(T) was distinguished from the phylogenetically closest relatives. Based on these genotypic and phenotypic data, strain S582(T) should be a new species in the genus Saccharopolyspora and the name Saccharopolyspora pathumthaniensis sp. nov. is proposed for the strain. The type strain is S582(T) (=NBRC 104112(T) =BCC 28624(T)).  相似文献

A novel actinomycete strain YIM 33378T was isolated from a soil sample collected from Lijiang, Yunnan Province, China. Based on the results of phenotypic and genotypic characteristics, strain YIM 33378T should be assigned to a new species of the genus Nocardia, for which the name Nocardia lijiangensis sp. nov. is proposed. A novel actinomycete, designated PA3T, was isolated from an oil refinery wastewater treatment plant, located in Palos de la Frontera, Huelva, Spain, and characterized taxonomically by using a polyphasic approach. Phylogenetic analysis based on 16S rRNA gene sequences showed that the isolate formed a distinct subclade in the Pseudonocardia tree together with Pseudonocardia asaccharolytica DSM 44247T. The chemotaxonomic properties of the isolate, for example, the presence of MK-8 (H4) as the predominant menaquinone and iso-C16:0 as the major fatty acid, are consistent with its classification in the genus Pseudonocardia. DNA:DNA pairing experiments between the isolate and the type strain of P. asaccharolytica DSM 44247T showed that they belonged to separate genomic species. The two strains were readily distinguished using a combination of phenotypic properties. Consequently, it is proposed that isolate PA3T represents a novel species for which the name Pseudonocardia hispaniensis sp. nov. is proposed. Exploration of unexplored habitats for novel actinobacteria with high bioactivity potential holds great promise in the search for novel entities. During the course of isolation of actinobacteria from desert soils, four actinobacteria, designated as 5K548T, 7K502T, 16K309T and 16K404T, were isolated from the Karakum Desert and their bioactivity potential as well as taxonomic provenances were revealed by comprehensive genome analyses. Pairwise sequence analyses of the 16S rRNA genes indicated that the four strains are representatives of putatively novel taxa within the prolific actinobacterial genus Saccharopolyspora. The strains have typical chemotaxonomic characteristics of the genus Saccharopolyspora by having meso-diaminopimelic acid as diagnostic diaminoacid, arabinose, galactose and ribose as whole-cell sugars. Consistent with this assignment, all of the isolates contained phosphatidylcholine in their polar lipid profiles and MK-9(H4) as the predominant menaquinone. The sizes of the genomes of the isolates ranged from 6.0 to 10.2 Mb and the associated G + C contents from 69.6 to 69.7 %. Polyphasic characterizations including determination of overall genome relatedness indices revealed that the strains are representatives of four novel species in the genus Saccharopolyspora. Consequently, isolates 5K548T, 7K502T, 16K404T and 16K309T are proposed as novel Saccharopolyspora species for which the names of Saccharopolyspora karakumensis sp. nov., Saccharopolyspora elongata sp. nov., Saccharopolyspora aridisoli sp. nov. and Saccharopolyspora terrae sp. nov. are proposed, respectively. Comprehensive genome analysis for biosynthetic gene clusters showed that the strains have high potential for novel secondary metabolites. Moreover, the strains harbour many antimicrobial resistance genes providing more evidence for their potentiality for bioactive metabolites.  相似文献

During the investigation of exploring potential sources of novel species and natural bioactives, a novel actinomycete, designated strain HIT-DPA4T, was isolated from a soil sample, which was collected from Nanjing, Jiangsu Province, PR China and characterized using a polyphasic approach. On the basis of 16S rRNA gene sequence similarities and the result of phylogenetic analysis, strain HIT-DPA4T was most closely related to Streptomyces cyaneus CGMCC 4.1671 T, and shared the highest sequence similarity of 98.76%. In addition, the cell walls of the species HIT-DPA4T contained LL-diaminopimelic acid as the diagnostic diamino acid and the whole-cell hydrolysates were identified as glucose and ribose, and the principal phospholipids were found to be diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylinositol mannoside and phosphatidylmonomethylethanolamine. MK-9(H6) and MK-9(H4) were predominant menaquinones; and C16:0, anteiso-C15:0 and C15:0 as major cellular fatty acids of the organism HIT-DPA4T. Gene Ontology database analysis and antiSMASH server predicted results displayed that strain HIT-DPA4T was a promising classification units, which has various types of functions and contains multiple biosynthetic gene clusters with the similarity more than 80%. Multilocus sequence analysis (MLSA) of five housekeeping genes (atpD, gyrB, recA, rpoB and trpB) illustrated that Streptomyces luteolifulvus formed a separate branch in the genus Streptomyces. However, a combination of low level of DNA-DNA relatedness and physiological properties indicated that strain HIT-DPA4T can be distinguished from its phylogenetically related species Streptomyces cyaneus CGMCC 4.1671 T. Moreover, gene synteny research could be further differed organism HIT-DPA4T from similarity species. Therefore, the strain is concluded to represent a novel species of the genus Streptomyces, for which the name Streptomyces luteolifulvus sp. nov. is proposed. The type strain is HIT-DPA4T (=?CGMCC 4.7558 T?=?TISTR 2751 T).
