A Gram stain-negative, aerobic and rod-shaped bacterium, strain DY22T, was isolated from a deep-sea sediment collected from the east Pacific Ocean. The isolate was found to grow in the presence of 0–20.0 % (w/v) NaCl and at pH 4.5–8.5; optimum growth was observed with 0.5–2.0 % (w/v) NaCl and at pH 5.0–7.0. Chemotaxonomic analysis showed the presence of ubiquinone-9 as predominant respiratory quinone and C16:0, C19:0 ω8c cyclo and C12:0 3-OH as major cellular fatty acids. The genomic DNA G+C content was determined to be 59.6 mol%. Comparative 16S rRNA gene sequence analysis revealed that the novel isolate belongs to the genus Salinicola. Strain DY22T exhibited the closest phylogenetic affinity to the type strain of Salinicola salarius with 97.2 % sequence similarity and less than 97 % sequence similarity with respect to other Salinicola species with validly published names. The DNA–DNA reassociation values between strain DY22T and S. salarius DSM 18044T was 52 ± 4 %. On the basis of phenotypic, chemotaxonomic and genotypic data, strain DY22T represents a novel species of the genus Salinicola, for which the name Salinicola peritrichatus sp. nov. (type strain DY22T = CGMCC 1.12381T = JCM 18795T) is proposed.  相似文献

A Gram-stain negative, ovoid-rod shaped, strictly aerobic bacterium, strain Y2T, was isolated from a deep-sea sediment of the Western Pacific. Phylogenetic and phenotypic properties of the organism indicated that it belongs to the genus Altererythrobacter. Strain Y2T shares highest 16S rRNA gene sequence similarity of 96.6 % with Erythrobacter jejuensis CNU001T, followed by the type strains of recognized members of the genus Altererythrobacter (94.8–96.5 %). Strain Y2T forms a clade with E. jejuensis CNU001T in the cluster of species of the genus Altererythrobacter. Growth of strain Y2T was observed at 4–40 °C (optimum, 35–37 °C), at pH 6.0–10.0 (optimum, pH 7.0–8.0) and in the presence of 0–5 % (w/v) NaCl (optimum, 2–3 %). The major cellular fatty acids were found to be C17:1 ω6c (41.5 %), summed feature 8 (C18:1 ω7c and/or C18:1 ω6c; 17.2 %), C17:1 ω8c (11.0 %) and C15:0 2OH (8.1 %). The major respiratory quinone was determine to be ubiquinone 10 (Q-10). The polar lipid analysis indicated the presence of diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylcholine, phosphatidylglycerol, one sphingoglycolipid, three unidentified phospholipids, two unidentified glycolipids, two unidentified aminolipids and three unknown lipids. The DNA G + C content of the type strain is 60.0 mol %. On the basis of the data from the polyphasic characterization, strain Y2T represents a novel species, for which the name Altererythrobacter oceanensis sp. nov. is proposed. Two Gram-positive, rod-shaped bacterial strains, H101(T) and H207, were isolated from deep sea water collected from South-West Indian Ocean. Phylogenetic analysis of 16S rRNA gene sequences showed that the two strains were closely related to one another (100% similarity), and had the closest relationship with Microbacterium hominis NBRC 15708(T) and Microbacterium insulae KCTC 19247(T) (98.2-98.3% similarities). DNA-DNA hybridization value between strains H101(T) and H207 was 87.2 ± 3.7%, and the values between the two strains and the closely related type strains were well below 70%. The two strains also shared a number of physiological and biochemical characteristics that were distinct from the closely related species, and grew at 2-37 ° C, pH 5-11 and 0-8% (w/v) NaCl. Both strains contained MK-12, MK-13 and MK-11 as the detected menaquinones. The peptidoglycan was of type B1γ with an interpeptide bridge D-Glu(Hyg)→ Gly(2)→ l-Lys. The major cellular fatty acids were anteiso-C(15:0), anteiso-C(17:0), and iso-C(16:0). Based on the genetic and phenotypic properties, it is proposed that strains H101(T) and H207 be classified as representatives of a novel species of the genus Microbacterium, with the name Microbacterium marinum sp. nov. A taxonomic study was carried out on strain D104T, which was isolated from deep-sea subsurface sediment sample from the Arctic Ocean. The bacterium was found to be Gram-negative, oxidase negative and catalase weakly positive, rod shaped, motile by means of polar flagellum. The organism grows between 4 and 37 °C (optimum 25–28 °C) and 0.5–6 % NaCl (optimum 3 %). Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain D104T belongs to the genus Marinomonas, with highest sequence similarities of 97.7 % to Marinomonas ushuaiensis DSM 15871T, followed by M. dokdonensis DSW10-10T (96.9 %), M. arenicola KMM 3893T (96.7 %), M. arctica 328T (96.6 %) and other 18 species of the genus Marinomonas (94.4–96.5 %). The average nucleotide identity and estimated DNA–DNA hybridization values between strain D104T and M. ushuaiensis DSM 15871T were 84.24 % and 20.80 ± 2.33 % respectively. The principal fatty acids were C16:0, sum in feature 3 (C16:1 ω7c/C16:1 ω6c), sum in feature 8 (C18:1 ω7c/C18:1 ω6c) and C12:1 3OH. The G + C content of the chromosomal DNA was determined to be 44.8 mol%. The respiratory quinone was found to be Q8 (100 %). Polar lipids include phosphatidylglycerol and phosphatidylethanolamine as major phospholipids and aminolipid and phospholipid as minor components. The results of the genotypic and phenotypic analyses indicate that strain D104T represents a novel species of the genus Marinomonas, for which the name Marinomonas profundimaris sp. nov. is proposed, with the type strain D104T (=MCCC 1A07573T = LMG 27696T).  相似文献

A Gram-negative, nonmotile, aerobic and oxidase- and catalase-positive bacterium, designated D25(T), was isolated from the deep-sea sediments of the southern Okinawa Trough area. Phylogenetic analyses of 16S rRNA gene sequences showed that strain D25(T) fell within the genus Myroides, with 99.2%, 96.0% and 93.4% sequence similarities to the only three recognized species of Myroides. However, the DNA-DNA similarity value between strain D25(T) and its nearest neighbour Myroides odoratimimus JCM 7460(T) was only 49.9% (<70%). Several phenotypic properties could be used to distinguish strain D25(T) from other Myroides species. The main cellular fatty acids of strain D25(T) were iso-C(15:0), iso-C(17:1)omega9c, iso-C(17:0)3-OH and Summed Feature 3 (comprising C(16:1)omega7c and/or iso-C(15:0)2-OH). The major respiratory quinone was MK-6. The DNA G+C content was 33.0 mol%. The results of the polyphasic taxonomy analysis suggested that strain D25(T) represents a novel species of the genus Myroides, for which the name Myroides profundi sp. nov. is proposed. A novel Gram-stain-negative, rod-to-spiral-shaped, oxidase- and catalase- positive and facultatively aerobic bacterium, designated HS6T, was isolated from marine sediment of Yellow Sea, China. It can reduce nitrate to nitrite and grow well in marine broth 2216 (MB, Hope Biol-Technology Co., Ltd) with an optimal temperature for growth of 30–33 °C (range 12–45 °C) and in the presence of 2–3 % (w/v) NaCl (range 0.5–7 %, w/v). The pH range for growth was pH 6.2–9.0, with an optimum at 6.5–7.0. Phylogenetic analysis based on 16S rRNA gene sequences demonstrated that the novel isolate was 93.3 % similar to the type strain of Neptunomonas antarctica, 93.2 % to Neptunomonas japonicum and 93.1 % to Marinobacterium rhizophilum, the closest cultivated relatives. The polar lipid profile of the novel strain consisted of phosphatidylethanolamine, phosphatidylglycerol and some other unknown lipids. Major cellular fatty acids were summed feature 3 (C16:1 ω7c/iso-C15:0 2-OH), C18:1 ω7c and C16:0 and the main respiratory quinone was Q-8. The DNA G+C content of strain HS6T was 61.2 mol %. Based on the phylogenetic, physiological and biochemical characteristics, strain HS6T represents a novel genus and species and the name Motiliproteus sediminis gen. nov., sp. nov., is proposed. A novel bacterial strain, designated SGD-1123T was isolated from Chorao Island, in Goa Province, India. The strain was found to be able to grow at 15–42 °C, pH 5–12 and 0–12 % (w/v) NaCl. The whole cell hydrolysates were found to contain meso-diaminopimelic acid, galactose and arabinose. The major fatty acids were identified as iso-C15:0 and anteiso-C15:0, MK-7 was identified as the predominant menaquinone and the predominant polar lipids were identified as diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine and an unidentified aminolipid. The genomic DNA G+C content was determined to be 44.6 mol%. Phylogenetic analysis based on 16S rRNA gene sequences placed the isolate within the genus Bacillus and further revealed that strain SGD-1123T had highest sequence similarity with Bacillus aquimaris, and forms a separate clade with its closest relatives i.e. B. aquimaris, Bacillus vietnamensis and Bacillus marisflavi, with which it shares 94.5, 94.1 and 94.1 % similarity respectively. The phylogenetic, chemotaxonomic and phenotypic analyses indicated that strain SGD-1123T represents a novel species within the genus Bacillus, for which the name Bacillus enclensis is proposed. A novel Gram-positive strain, designated NIO-1003T, was isolated from a marine sediment sample collected from the Chorao Island, Goa Provence, India. Strain NIO-1003T was found to be strictly aerobic, motile, endospore-forming rods. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain NIO-1003T belongs to the genus Fictibacillus and to be most closely related to Fictibacillus rigui KCTC 13278T, Fictibacillus solisalsi KCTC 13181T and Fictibacillus barbaricus DSM 14730T with 98.2, 98.0 and 97.2 % similarity and 25, 28, 39 nucleotide differences respectively. Strain NIO-1003T was characterized by having cell-wall peptidoglycan based on meso-diaminopimelic acid and MK-7 as the predominant menaquinone. The polar lipid profile exhibited the major compounds diphosphatidylglycerol, phosphatidylglycerol and phosphatidylethanolamine. In addition, minor amounts of an aminophospholipid were detected. The major fatty acids were identified as ai-C15:0, iso-C15:0, ai-C17:0 and C16:0, supporting the grouping of strain NIO-1003T into the family Bacillaceae. The DNA G+C content of strain NIO-1003T was determined to be 42.6 mol%. On the basis of phenotypic properties, phylogeny and DNA–DNA hybridisation analysis, strain NIO-1003T is considered to represent a novel species of the genus Fictibacillus for which the name Fictibacillus enclensis sp. nov. is proposed. A novel bacterial strain designated as NIO-1008T was isolated from marine sediments sample in Chorao Island India. Cells of the strains were gram positive and non-motile, displayed a rod–coccus life cycle and formed cream to light grey colonies on nutrient agar. Strain NIO-1008T had the chemotaxonomic markers that were consistent for classification in the genus Arthrobacter, i.e. MK-9(H2) (50.3 %), as the major menaquinone, and the minor amount of MK-7 (H2-27.5 %), MK-8 (H4-11.6 %) and MK-8 (H2-10.4 %). anteiso-C15:0, iso-C15:0, iso-C16:0 and C15:0 were the predominant fatty acids. Galactose, glucose and rhamnose are the cell-wall sugars, and DNA G+C content was 61.3 mol%. Phylogenetic analysis, based on 16S rRNA gene sequencing, showed that the strains were most similar to Arthrobacter equi IMMIB L-1606T, Arthrobacter chlorophenolicus DSM 12829T, Arthrobacter defluvii KCTC 19209T and Arthrobacter niigatensis CCTCC AB 206012T with 98.5, 98.4, 98.0 and 97.8 %, respectively, and formed a separate lineage. Combined phenotypic data and DNA–DNA hybridization data supported the conclusion that strains NIO-1008T represent a novel species within the genus Arthrobacter, for which the name Arthrobacter enclensis sp. nov., is proposed. A Gram-staining-negative, aerobic and pear-shaped bacterial strain, designated WL0036T, was isolated from coastal sediment sample collected in Nantong city, Jiangsu province of China (120° 51′ 13″ E, 32° 6′ 26″ N) in October 2020. Strain WL0036T was found to grow at 20–37 °C (optimum, 28 °C) with 0–9.0% NaCl (optimum, 2.5–4.0%) and displayed alkaliphilic growth with the pH range of pH 6.0–10.0 (optimum, pH 7.0–8.0). The polar lipids profile of strain WL0036T included phosphatidylcholine, phosphatidylethanolamine, glycolipid and an unidentified lipid. The major isoprenoid quinone was determined to be Q-11 and the major fatty acids were C16:0, 11-methyl-C18:1ω7c, and summed features 8 (C18:1ω6c and/or C18:1ω7c). The G?+?C content of genomic DNA was 61.8%. Phylogenetic trees constructed based on 16S rRNA gene sequence and bac120 gene set (a collection of 120 single-copy protein sequences prevalent in bacteria) indicted that strain WL0036T clustered with strains Hyphomonas neptunium ATCC 15444T and H. polymorpha PS728T. The average nucleotide identities between strain WL0036T and strains H. neptunium ATCC 15444T and H. polymorpha PS728T were 80.7% and 81.2%, respectively. Strain WL0036T showed 22.8% and 23.2% of digital DNA-DNA hybridization identities with H. neptunium ATCC 15444T and H. polymorpha PS728T, respectively. As inferred from the phenotypic and genotypic characteristics and the phylogenetic trees, strain WL0036T ought to be recognized as a novel species in genus Hyphomonas, for which the name Hyphomonas sediminis sp. nov. is proposed. The type strain is WL0036T (=?MCCC 1K05843T?=?JCM 34658T?=?GDMCC 1.2413T).

A novel facultatively anaerobic strain DH1T was isolated from deep sub-seafloor sediment at a depth of 900 m below the seafloor off Seo-do (the west part of Dokdo Island) in the East Sea of the Republic of Korea. The new strain was characterized using polyphasic approaches. The isolate was Gram-stain-negative, motile by gliding, non-spore-forming rods, oxidase-negative, and catalase-positive; and formed colonies of orange-red color. The NaCl range for growth was 0.5–7.0% (w/v) and no growth was observed in the absence of NaCl. The isolate grew optimally at 30°C, with 2% (w/v) NaCl and at pH 7. The cell-wall hydrolysates contained ribose as a major sugar. The DNA G+C content was 40.8 mol%. The closest related strains are Sunxiuqinia faeciviva JAM-BA0302T and Sunxiuqinia elliptica DQHS-4T (97.9 and 96.3% sequence similarity, respectively). The level of DNA-DNA relatedness between strain DH1T and S. faeciviva JAM-BA0302T was around 41% (but only 6% between DH1T and S. elliptica DQHS-4T). The major cellular fatty acids of the isolate were contained iso-C15:0 (25.9%), anteiso-C15:0 (16.7%), and summed feature 9 (comprising C16:0 3-OH and/or unknown fatty acid of dimethylacetal ECL 17.157; 13.2%). The predominant menaquinone was MK-7. On the basis of polyphasic evidence from this study, the isolate was considered to represent a novel species of the genus Sunxiuqinia, for which the name Sunxiuqinia dokdonensis sp. nov. is proposed; the type strain is DH1T (=KCTC 32503T =CGMCC 1.12676T =JCM 19380T).  相似文献
