This study evaluated mitochondrial DNA (mtDNA) sequence variation in a 552-bp fragment of the control region of Arctic charr (Salvelinus alpinus) by analyzing 159 individuals from 83 populations throughout the entire range of the complex. A total of 89 (16.1%) nucleotide positions were polymorphic, and these defined 63 haplotypes. Phylogenetic analyses supported the monophyly of the complex and assigned the observed haplotypes to five geographic regions that may be associated with different glacial refugia. Most notably, a formerly defined major evolutionary lineage (S. a. erythrinus) ranging from North America across the Arctic archipelago to the Eurasian continent has now been partitioned into the Arctic group and the newly identified Siberian group. The Beringian group, formed entirely by specimens assigned to S. malma (Dolly Varden), encompassed the area formerly assigned to S. a. taranetzi. The latter, due to a unique haplotype, became the basal member of the Arctic group. Overall, the S. alpinus complex reflects divergent evolutionary groups coupled with shallow intergroup differentiation, also indicated by an analysis of molecular variance that attributed 73.7% (P < 0.001) of the total genetic variance among groups. Time estimates, based on sequence divergence, suggest a separation of the major phylogeographic groups during early to mid-Pleistocene. In contrast, colonization of most of today's range started relatively recently, most likely late Pleistocene during the last retreat of ice sheets some 10,000-20,000 years ago. This time scale obviously is too shallow for detecting significant variation on a smaller scale using mtDNA markers. However, other studies using nuclear microsatellite DNA variation strongly suggested ongoing evolution within groups by revealing strong population-genetic substructuring and restricted gene flow among populations. Thus, Arctic charr could serve as a model organism to investigate the linkage between historical and contemporary components of phylogeographic structuring in fish, and, with a global perspective of the distribution of genetic variation as a framework, meaningful comparisons of charr studies at a smaller geographic scale will now be possible.  相似文献

A study of body and head development in three sympatric reproductively isolated Arctic charr (Salvelinus alpinus (L.)) morphs from a subarctic lake (Skogsfjordvatn, northern Norway) revealed allometric trajectories that resulted in morphological differences. The three morphs were ecologically assigned to a littoral omnivore, a profundal benthivore and a profundal piscivore, and this was confirmed by genetic analyses (microsatellites). Principal component analysis was used to identify the variables responsible for most of the morphological variation of the body and head shape. The littoral omnivore and the profundal piscivore morph had convergent allometric trajectories for the most important head shape variables, developing bigger mouths and relatively smaller eyes with increasing head size. The two profundal morphs shared common trajectories for the variables explaining most of the body and head shape variation, namely head size relative to body size, placement of the dorsal and pelvic fins, eye size and mouth size. In contrast, the littoral omnivore and the profundal benthivore morphs were not on common allometric trajectories for any of the examined variables. The findings suggest that different selective pressures could have been working on traits related to their trophic niche such as habitat and diet utilization of the three morphs, with the two profundal morphs experiencing almost identical environmental conditions.  相似文献

Synopsis The degree of genetic differentiation among four morphs of Arctic charr (small benthivorous, large benthivorous, piscivorous, and planktivorous) from Thingvallavatn, Iceland, was determined electrophoretically. Five of 36 enzyme loci were found to be polymorphic (Est2, Gpi3, Ldh4, Mdh4, 5 and Pgm2). However, only Est2 and Mdh4,5 showed enough variability to permit statistical analysis of divergence among morphs. All four morphs are very closely related; the values of Nei's (D) range from 0.00004 to 0.00126. These morphs are conspecific and do not represent different evolutionary lineages. There is significant genetic differentiation between the small benthivorous charr and the other three morphs. The relative relatedness of morphs based on gene frequency data is only partially concordant with that based on morphology and ecological specialization. The biological significance of this result is unclear because of the limited number of polymorphic loci upon which the genetic analysis is based and the high degree of relatedness among morphs.  相似文献

Mitochondrial DNA analysis of a total of 82 individuals from seven Scottish populations of Arctic charr ( Salvelinus alpinus L.) using restriction fragment length polymorphism and direct nucleotide sequencing of the cytochrome b gene revealed little variation within or between populations. One of 19 restriction enzymes showed a polymorphism that was present in three of the seven populations. Only one nucleotide substitution was detected in a 288 base pair segment of the cytochrome b gene examined in 26 fish. Approximately 4% of the mitochondrial genome was surveyed with these complementary procedures.  相似文献

The systematica of coexisting morphotypes of Arctic charr, Salvelinus alpinus , has been a matter of dispute ever since the days of Linnaeus. Widespread allelic variation at an esterase locus has led some investigators to propose that the morphotypes reflect a complex of at least three sibling species. We tested this hypothesis by examining 42 electrophoretically detectable loci in natural and transplanted charr populations from 15 localities in S Norway. The absolute values of Nei's genetic distance between morphotypes and populations are small (typically in the order of 0.001), and morphotype changes may occur without accompanying changes in frequencies of esterase alleles. Differentiation among localities explains far more of the total gene diversity than differences between morphotypes in the four cases of naturally occurring sympatric morphotypes examined. The data are consistent with an intraspecific population structuring based on locality, and the multiple species hypothesis is rejected.  相似文献

Morphological differences, haematocrit value and chloride cells were examined in downstream migrating Arctic charr, Salvelinus alpinus (L.) from the Hals River, North Norway, and resident charr from Lake Storvann. Fish were classified as visual parr, silvery parr or smolt based on the degree of silvering and the Occurrence of lateral parr marks. On average, 47% of downstream migrating Arctic charr were classified as visual molts, but only 14% of the resident charr. Charr longer than 20 cm fork length were mainly classified as visual molts. Morphometrical analyses of body size and shape revealed that most of the variations could be explained in terms of variations in fork length. Length adjusted ratios of post-anal distances were significantly higher in migrating charr than in resident charr. The number of developed chloride cells, the cell nucleus diameter in the basin of secondary gill lamellae and blood haematocrit values were significantly higher among downstream migrating charr than in resident charr. However, morphological differences between anadromous and resident Arctic charr were not necessarily synchronized with the development of chloride cells.  相似文献

We studied the salmonid fish Arctic charr, Salvelinus alpinus , in a small and shallow landlocked lake in NW Iceland. The lake is productive but die only fish present is Arctic charr. Despite the apparent absence of discrete benthic and limnetic habitats for fish, two forms of Arctic charr are found in the lake. They show subde differences in morphology related to swimming performance and manoeuvrability, but differences in life history such as growth, and age and size at sexual maturation are more pronounced. Both forms have benthic feeding habits with one form consuming greater number of species than the other. We suggest that the segregation of these forms is based on the evolution of a specialist from a local generalist and that this has been made possible by the absence of a common fish competitor in similar lakes, the threespined stickleback Gasterosteous aculeatus.  相似文献

Restriction fragment length polymorphism analysis of mitochondrial DNA (mtDNA) was used to reconstruct postglacial dispersal routes of arctic charr Salvelinus alpinus in North America. Twelve of 35 restriction enzymes detected polymorphisms among representative populations, revealing two distinct lineages with an estimated nucleotide divergence of 1.32%. Subsequent screening of 869 fish from 54 populations with four diagnostic restriction enzymes showed that these lineages have largely allopatric distributions, suggesting their dispersal from separate northern and eastern glacial refugia. In addition, geographical and genetic structure among eastern populations suggested the existence of a second eastern refuge. Among the three lineages, the most divergent (Arctic) lineage occurred from Alaska east to northern Labrador. Quebec, New Brunswick, and New England were colonized by a second (Laurentian) lineage, and Labrador by a third group. Contact between refugial groups was only detected in two Labrador populations. The Arctic lineage was highly differentiated from eastern North American and European haplotypes, and probably diverged during the early Pleistocene. By contrast, the Laurentian and Labrador groups were similar to Old World charr, suggesting a shared ancestry during the mid-Pleistocene. In addition, the close relationship between Labrador and Laurentian charr indicates their probable divergence during the Wisconsinan glaciation.  相似文献

Size and frequency of occurrence of prey of brown trout Salmo trutta L. and Arctic charr Salvelinus alpinus (L.) were recorded in 13 Norwegian lakes during 1973–1990. Piscivores usually comprised less than 5% of the total population. Arctic charr were less piscivorous than brown trout. Trout and charr became piscivorous at 13 and 16 cm length, respectively. These size thresholds were similar to those of other facultative piscivorous freshwater fish species. When present, three-spined sticklebacks, Gasterosteus aculeatus (L.), were preferred by all length groups of piscivorous brown trout and Arctic charr. Length of prey increased with increasing predator length, and the mean body length of prey was about 33 and 25% of predator length for trout and charr, respectively. Recent studies have demonstrated that Arctic charr ( Salvelinus alpinus ) exhibit higher growth rates when held at high stocking densities. It has been argued that these increased growth rates are a result of decreased social interactions at the increased densities. The current study was designed to test this hypothesis by examining: (1) the frequency of agonistic interactions, (2) the proportion of time spent swimming and (3) growth rates among groups of young of the year Arctic charr held at different stocking densities. It was found that charr reared at higher densities exhibited significantly higher mean weights and lengths than those at the lower densities. The behavioural observations revealed that charr initiated significantly fewer agonistic interactions and spent a significantly greater amount of time shoaling at high densities than charr held at the medium or low densities. It is argued that these behavioural responses may serve to lower energy expenditures, resulting in the increased growth rates.  相似文献

The effects of food quality on maturation rate were followed in progeny from wild Arctic charr, caught in Lake Fattjaure, northern Sweden. The fish were reared at five different food quality levels. In the first summer, the fry were given feed of three qualities: diluted with cellulose in three proportions (0, 15 and 25%). During the second winter the impact of changed food quality level was studied by transferring half the fish from the high (control) to the low food quality level and vice versa.
Maturation rate was lower in males reared at the high and improved food quality levels than in males reared at the medium, low and reduced food quality levels. The maturation rate in females was similar at all levels, though the rate tended to increase at the reduced food quality level.
Fish reared at the high and medium levels had similar growth rates, whereas fish at the low food quality level experienced slower growth. Reduced food quality did not arrest the growth offish, whereas improved food quality enhanced their growth.  相似文献

The role of environmental factors in the regulation of sub-annual increment formation in fish otoliths appears to differ markedly between species. To examine the periodicity of primary increment formation in the otoliths of O + Arctic charr, Salvelinus alpinus (L.), and the effects of temperature, and photoperiod on their formation, fish were held under controlled environmental conditions. Primary growth increments were found in the otoliths of fish held at constant temperature (18° C) and at ambient temperature [fluctuating with a circadian and circannual rhythm (4–18° C)]. Consistent and significant disruptions in increment formation occurred however, in experimental groups subjected to rapid change from ambient photoperiod to a 6L: 6D photo-period for 96 h. Disruptions in increment formation were also observed immediately following transportation of fish between holding facilities and following disease treatment. The number of otolith increments formed in fish held on an ambient photoperiod regime, correlated closely with time elapsed in days since checkmark formation ( r = 0.989, P ≤0.001) in fish sampled sequentially over a period of 10 to 105 days. Thus we demonstrate that under conditions of ambient photoperiod, primary increments are formed daily.  相似文献

Two morphologically and ecologically distinct forms of Arctic charr, Salvelinus alpinus , are found in Loch Rannoch, Scotland. The differences in morphology are adaptations to different modes of life, one being pelagic, the other benthic. Both forms have been the subjects of extensive genetic studies including cytogenetics, nuclear and mitochondrial DNA analysis, and protein electrophoresis. Significant differences between the two forms are revealed by some techniques but not others and provide evidence for the reproductive isolation of these two morphs. The findings are discussed in relation to the derivation of sympatry and the phylogenetics of Arctic charr.  相似文献

Growth and digestive enzymes were studied in alevins of Arctic charr raised on six commercial diets that differed in protein source and level. There were links between diet composition (protein quality), fish performance and proteolytic and amylase enzyme activities. Trypsin and amylase activities were up to 10-fold higher in charr fed high protein (65–70%) diets than in fish fed live feed. A depression of pancreaticenzyme activities was noticeable when charr were fed starter diets in which fish meal was partially replaced by soybean meal. Specific growth rates (SGR) showed a cyclic pattern during early ontogeny of charr and SGR was highly correlated with food conversion efficiency. Activities of pancreatic enzymes also demonstrated a cyclic pattern but the periodicities of the growth cycle (50.6 days) and those of the enzymes differed. We hypothesize that both photostimulation and feedback responses might be involved in pancreatic responses in fish, which result in cyclic growth patterns.  相似文献

F ST and RST estimates for Arctic charr from six microsatelite markers collected from two neighbouring Scottish lakes, Loch Maree and Loch Stack, confirm the presence of two distinct genetic groupings representing separate populations within each lake. In both lakes, there was also a clear body size dimorphism, with large and small body size forms that segregated according to genetic grouping. There was evidence of only subtle foraging ecology differences between morphs, with the small body size morph in both lakes being more generalist in its foraging in the summer (consuming mostly plankton but also some macrobenthos) than the large body size morph, which specialized on planktonic prey. Trophic morphology (head and mouth shape) did not differ significantly between morphs (although the small sample size for Maree makes this a preliminary finding). Cluster analysis of the microsatelite data and the presence of private alleles showed that morphologically similar forms in different lakes were not genetically similar, as would be expected under a multiple invasion hypothesis. Thus, the data do not support a hypothesis of a dual invasion of both lakes by two common ancestors but instead suggest an independent origin of the two forms in each lake. The mortality of Arctic charr, Salvelinus alpinus , previously grown-on in salt water during the summer months, was studied during the winter 1980–81. Mortality did not appear to be related to a' critical size' but rather to rates of growth shown by the fish during the summer months. Mortality was high amongst the slow-growing fish.
The results are discussed in relation to published data on the osmoregulatory capabilities of Salvelinus spp.  相似文献

A specific sex steroid-binding protein (SBP) is believed to be involved in regulation of circulating sex steroids, steroid delivery to target cells and intracellular signalling in sex steroid-sensitive tissues. In the present work, interactions between xenoestrogens and the plasma SBP in Arctic charr (Salvelinus alpinus L.) were determined using ligand-protein binding studies. The test compounds were all able to displace tritiated 17 beta-estradiol (E2) from the Arctic charr SBP (acSBP) in a competitive and dose-dependent manner. The acSBP affinities for the xenoestrogens ranged over several orders of magnitude (17 beta-estradiol>ethynylestradiol (EE2)>zearalenone (ZEA)>diethylstilbestrol (DES)>genistein (GEN)>bisphenol A (BPA), 4-t-octylphenol (OP)>o,p'-DDT, and dieldrin (DIN)), but were consistently lower than that of 17 beta-estradiol (about 4 x 10(2) -10(6)-fold less potent). The relative binding affinity (RBA) for selected chemicals were independent of both gender, age and maturation status, as well as variations of acSBP binding affinity. The affinity of endogenous steroids and estrogen mimics for the acSBP shows a high correlation to the affinity for the rainbow trout SBP, thus suggesting a phylogenetically conserved ligand-binding site between closely related species. Furthermore, it is argued that interaction with the acSBP- and SBP-mediated processes may introduce novel pathways for endocrine disruption, which may work in concert with the classical receptor-mediated effects.  相似文献

An atypical strain of Carnobacterium divergens, strain 6251, was isolated from the small intestine of Arctic charr (Salvelinus alpinus L.), fed high dietary carbohydrate. This strain showed marked growth inhibitory effects in vitro against the fish pathogens Aeromonas salmonicida subsp. salmonicida (furunculosis), Vibrio anguillarum (vibriosis) and Vibrio viscocus (winter ulcer). The strain is a non-motile Gram-positive psychrotrophic rod that lacks both catalase and oxidase, grows at pH 9.1 (CTAS agar), but not on acetate containing media (pH < or = 5.4), on TCBS or at < or =6% sodium chloride content. Strain 6251 is facultatively anaerobic and utilises tryptone as a sole source of nutrient. Further characterisation showed the most abundant cellular fatty acid of strain 6251 to be oleic acid (18:1) (n-9) (36.0%). Sequencing of a 16S rDNA region of 578 nucleotides and AFLP microbial fingerprinting suggested that strain 6251 is not closely related to any carnobacteria known, however, DNA-DNA similarity determinations showed high similarity (96.2%) with the type strain of Carnobacterium divergens. The unique phenotypic attributes of this strain represent new information on the biodiversity and ecology of carnobacteria and especially of the species C. divergens.  相似文献

The chromosomes, together with their C and Q banding patterns, of Arctic charr from Loch Rannoch, Scotland are described. Q banding revealed fewer bands than C banding and variation between individuals for the number of C bands was found. These results suggest that subsets of heterochromatin exist and that there may be an individual variation in DNA content.  相似文献
