里氏木霉内切-β-甘露聚糖酶基因在毕赤酵母中的表达   总被引:4,自引:0,他引:4  
采用PCR方法从里氏木霉(Trichoderma reesei)基因组中获得含有两个内含子的内切-β-甘露聚糖酶全长基因,末端重叠延伸PCR去除内含子后,将其插入到巴斯德毕赤酵母(Picher pastoris)表达载体pPIC9K中,位于α-因子信号肽序列的下游,并与之同框,获得重组质粒pM242。重组质粒线性化后用电击法转化毕赤酵母菌株GS115。经大量筛选,获得高效分泌表达内切甘露聚糖酶的毕赤酵母工程菌株Gpmf25。摇瓶发酵结果表明,培养基中甘露聚糖酶的活力可达12.5IU/mL。重组酶最适pH和最适反应温度分别为5.0和80℃,在pH5.0~6.0时酶活稳定,在pH5.4时70℃保温30min酶活维持50%以上。  相似文献

将短小芽孢杆菌HB030的内切-1,4-木聚糖酶基因克隆到毕赤酵母表达载体pPIC9K,得到重组质粒pH-BM220,将pHBM220经酶切后分别转化三株毕赤酵母KM71、GS115、SMD1168,该木聚糖酶基因在三株毕赤酵母中均实现了分泌表达,将重组毕赤酵母KM71(pHBM220),GS115(pHBM220),GS115(pHBM220),SMD1168(pHBM220)分别诱导产酶,对重组酶进行相关的酶学性质分析表明,三的最适反应pH值约为5.5,最适反应温度约为60℃,在其最适反应条件下测得三粗酶液酶活分别为10.80IU/mL,11.63IU/mL,9.68IU/mL,重组毕赤酵母KM71(pHBM220)所产酶的热稳定性较好,而在pH稳定性方面三没有太大的差异。  相似文献

根据绿色木霉(Trichoderma viride)WL 0422菌株内切葡聚糖酶基因egⅡ的cDNA序列,设计特异引物,以重组质粒pTG19-T-egⅡ为模板,扩增得到全长1 194bp的egⅡ成熟肽cDNA片段.将该片段分别克隆到大肠杆菌(Escherichia coil)表达载体pET-28a( )和毕赤酵母(Pichia pastoris)表达载体pPIC9K中,并在大肠杆菌Rosset(DE3)和毕赤酵母GS115中进行了表达.重组大肠杆菌表达蛋白占菌体总蛋白的15%,表达产物无内切葡聚糖酶活性.重组毕赤酵母经甲醇诱导实现了分泌表达,在摇床水平上酶活性达到2 788U/ml.酶学性质分析表明,该酶作用的最适pH为4.5,pH 3.5~6.0范围内酶活性保留在80%以上;最适温度为50℃,55℃以下酶的稳定性在90%以上.  相似文献

目的:构建高效表达白地霉脂肪酶的毕赤酵母重组菌株,并对筛选得到的菌株进行摇瓶发酵条件优化和分批补料高密度发酵工艺研究。方法:将诱导型表达载体pPIC9K-gcl电转化至毕赤酵母GS115。通过橄榄油-罗丹明B平板和摇瓶发酵筛选高脂肪酶活力的重组菌株,运用基于TaqMan探针的实时荧光定量PCR 法确定其拷贝数,并对菌株进行摇瓶发酵条件优化。在此基础上,研究重组菌在3L 发酵罐中的高密度发酵工艺。结果:筛选得到一株具有3 个白地霉脂肪酶基因拷贝的菌株GS115/pPIC9K-gcl 78#,初始酶活力为220 U/ml。当摇瓶发酵条件为甲醇诱导96 h,每24 h甲醇添加量1 %,接种量2 %,培养基初始pH 7.0,500 ml摇瓶装液量50 ml,甲醇诱导温度25℃ 时酶活力达735 U/ml。3L 发酵罐高密度发酵176.5 h,酶活力达到3360 U/ml,总蛋白含量达到4.30 g/L,且发酵过程中细胞活性一直保持在96 % 以上。结论:基因拷贝数与重组菌株的产酶水平呈正相关,摇瓶优化可显著提高重组菌株的产酶能力,为白地霉脂肪酶的工业化生产奠定了技术基础。  相似文献

目的:克隆黑曲霉β-甘露聚糖酶基因,研究该基因在毕赤酵母中的表达情况。方法:运用RT-PCR从黑曲霉AN070902中克隆β-甘露聚糖酶cDNA片段,与载体pPIC9K相连,构建重组载体VMAN-pPIC9K,电转化毕赤酵母GS115,筛选产酶最高菌株进行5 L液体发酵,对该菌株所产重组酶进行酶学性质分析。结果:克隆获得1152 bpcDNA,编码由383个氨基酸残基组成的蛋白质,该蛋白质属于GH5家族,理论pI和相对分子质量分别为4.48和41.6×103;筛选获得的重组菌株VMAN-pPIC9K-GS115在5 L液体发酵中上清酶活达11 785 U/mL;表达的重组酶是一种酸性β-甘露聚糖酶,最适反应pH值为3.0,经pH2.0~9.0处理2 h后剩余酶活保持90%以上;该重组酶最适反应温度为65℃,70℃处理1 h后剩余酶活保持75%以上;该重组酶活性被1 mmol/L的Fe3+和Mn2+显著抑制,被1mmol/L的Co2+显著激活。结论:重组耐酸性β-甘露聚糖酶的特性,决定了其在工业生产中,特别是动物饲料和食品加工中具有应用价值。  相似文献

PFA是来源于Pyrococcus furious的一种耐高温α-淀粉酶,为了使PFA能够在巴斯德毕赤酵母中高效表达,根据巴斯德毕赤酵母密码子的偏好性对PFA的基因序列进行密码子优化,人工合成耐高温淀粉酶PFA基因pfa,并连接到巴斯德毕赤酵母中表达载体pPIC9K上,得到重组质粒pPIC9K-pfa。重组质粒线性化后转化到巴斯德毕赤酵母菌株GS115中,重组菌株在摇瓶中用甲醇诱导表达,分泌表达酶活最高为220U/L。  相似文献

利用PCR技术,从扣囊复膜孢酵母的总DNA中扩增得到β-葡萄糖苷酶(β-Glucosidase)基因(BGL1),长度为2596 bp,连接到pGEM-T载体上,用限制性内切酶切下目的基因,插入到巴斯德毕赤酵母表达载体pPIC9K中,使之位于α-因子信号肽下游,且与之同框,构建成重组质粒pSHL9K.通过电转化将重组质粒pSHL9K插入到Pichia pastoris GS115菌株染色体中,获得高效表达BGL1基因的毕赤酵母重组工程菌株.重组酶的最适温度为50℃,最适pH为5.4.培养基中β-葡萄糖苷酶活性最高可达47U/mL.  相似文献

内切纤维素酶Cel5A缺乏是限制纤维素酶制剂高效酶解天然纤维素的关键因素。本文尝试构建高效表达里氏木霉Cel5A的毕赤酵母重组菌株以弥补目前Cel5A的天然分泌不足,通过基因密码子偏好性优化里氏木霉Cel5A基因和构建表达载体p PIC9K-eg2,并将其电转入毕赤酵母GS115以构建重组子,利用浓度梯度平板和摇瓶发酵筛选获得一株高产毕赤酵母Pichia pastoris菌株GS115-EGⅡ。重组酶的酶学性质分析显示,该酶分子量50 k Da、最适p H(p H 4.5)略有降低及最适反应温度为60℃,专一性地作用于非结晶纤维素,与天然里氏木霉Cel5A并无明显区别。通过摇瓶发酵的初步优化,该菌摇瓶培养条件:培养温度28℃、起始p H 5.0、接种量2%、每24 h添加甲醇1.5%(V/V)、每24 h添加山梨醇4 g/L及吐温80添加4 g/L,发酵192 h重组酶酶活达到24.0 U/m L。进一步上罐(5 L)发酵180 h,该重组酶Cel5A酶活高达270.9 U/m L,蛋白含量达到4.16 g/L。重组毕赤酵母P.pastoris GS115-EGⅡ是一株适合于外源表达Cel5A的工程菌,该重组酶可替代天然分泌Cel5A适用于当前酶基生物炼制模式下木质纤维素基质高效水解中。  相似文献

采用RTPCR技术克隆到一个平菇(Pleurotusostreatus)漆酶基因的全长cDNA,命名为lccPo1,其序列提交GenBank,登录号为AY450404。将其ORF克隆到毕赤酵母表达载体pHBM906,转化3株毕赤酵母GS115、KM71和SMD1168,该漆酶基因在3种毕赤酵母菌株中均实现了分泌表达。3种摇瓶培养条件①25℃,1.0%(VV)甲醇;②20℃,1.0%(VV)甲醇;③20℃,0.5%(VV)甲醇,进行比较研究后发现适当提高甲醇浓度有利于漆酶在低温条件下表达,而降低培养温度到20℃则可以提高漆酶的产量2~6倍。3株重组毕赤酵母在其最适反应条件下测得三者粗酶液最高漆酶酶活分别为3.19UmL[GS115(pHBM565)]、2.56UmL[KM71(pHBM565)]和2.49UmL[SMD1168(pHBM565)]。对重组酶进行相关的酶学性质分析表明,三者的最适反应pH值约为4.2,最适反应温度约为60℃。重组毕赤酵母GS115(pHBM565)所产酶的热稳定性稍好,在pH稳定性方面三者没有太大差异。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. We also discuss the development and practicality of various cell culture and animal model systems to study KSHV replication and pathogenesis.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
