The effect of somatostatin (GH-RIH) on cholecystokinin octapeptide (OP-CCK) or acetylcholine (ACh) induced contraction of the guinea pig gallbladder was evaluated in vitro. GH-RIH failed to inhibit the muscle contraction induced by OP-CCK or ACh. To correlate with the in vitro results, the effect of GH-RIH on OP-CCK induced contraction of the gallbladder was evaluated in the guinea pig in vivo. GH-RIH did not affect the OP-CCK induced contraction of the gallbladder. Our results suggest that GH-RIH does not have direct inhibitory effect on the contraction of the guinea pig gallbladder induced by OP-CCK or ACh.  相似文献

Summary Tumor immunity induced by bacillus Calmette-Guérin was studied in the line 10 hepatocellular carcinoma (line 10) in the strain-2 guinea pig. Line 10 immunity was investigatedin vitro with a lymphocyte proliferation assay using line 10 tumor protein extracted with 3 M KCl andin vivo by adoptive transfer of line-10-immune spleen cells. Monoclonal antibodies against guinea pig leucocyte markers were used to block functional properties of the immune cells in order to determine which cell types or cell markers are involved in the immune response to the line 10 tumor.In vitro cells from the spleen, peripheral blood and regional lymph node of immune animals reacted with a proliferative response to line 10 protein. This antigen-specific response was caused by T cells and was regulated by major histocompatibility complex (MHC) class II molecules. In blocking experiments it was found that CT5 (anti-PanT), or MSgp4 [anti-(MHC class I antigen)] monoclonal antibodies did not block but some-times stimulated the proliferative response. The effect of H159 (anti-PanT) was irregular, while H155 [anti-(T helper)], and 5C3 [anti-(IL-2 receptor)] monoclonal antibodies blocked the response almost completely. We studied the relevance of the resultsin vitro obtained and found that mAb 5C3 [anti-(IL-2 receptor)] inhibited the adoptive transfer of line 10 immunity, suggesting that the rejection of line 10 cells is caused by a mechanism that is interleukin-2 (IL-2)-dependent. Moreover, complement lysis of MHC-class-II-antigen-positive immune spleen cells inhibited completely the rejection of the line 10 tumor cell challenge in the adoptive-transfer experiments. In conclusion, our data show that MHC class II molecules or cells possessing these molecules are involved in immunity against line 10 tumor cells, as (a) monoclonal antibodies against MHC class II antigens inhibited thein vitro proliferative response of T cells to tumor antigens and (b) removal of MHC-class-II-positive immune spleen cells abrogated the antitumor effect in the adoptive-transfer experiments. Interleukin-2-dependent proliferation of immune T cells is required for the rejection of line 10 tumor cells.  相似文献

Summary Three different immunocytoenzymatic techniques were used to identify and characterize the thyrotropic cells in the pituitary of normal guinea pigs at the ultrastructural level (superimposition technique, immunocytochemical technique using P.A.P. and indirect immunohistoenzymatic method before embedding).These cells are characterized by a dark cytoplasm with granules ranging from 1500 to 2000 Å in diameter. The appearance of these granules is very variable: some display a marked electron density and are homogeneous but some have a less marked electron density with a more electron dense peripherally situated region.The TSH molecules are essentially confined to the granules but when the immunocytochemical reactions are carried out before embedding, positive staining is also seen in the cytoplasm and the outer surface of most of the rough endoplasmic reticulum membranes. 乳酸左氧氟沙星对豚鼠心肌细胞电生理的影响   总被引:1,自引:0,他引:1  
目的了解乳酸左氧氟沙星(LVFX)对豚鼠心室肌细胞电生理的影响.方法经腹腔注射不同剂量的LVFX,记录并分析注药后5~360 min豚鼠Ⅱ导联心电图的QT间期,以及校正的QT间期(QTc).采用全细胞膜片钳技术,记录不同浓度LVFX对体外单个心室肌细胞的延迟整流钾电流(IK)的作用.结果①LVFX给药量为200 mg/kg时,心电图QT间期延长19.38%±3.15%(P<0.05);在50 mg/kg和100 mg/kg等较低剂量时,QT间期延长不明显(P>0.05).②LVFX抑制IK电流,且抑制作用呈现电压依赖性和浓度依赖性.结论LVFX可能通过抑制心肌细胞IK电流引起心脏QT间期延长,临床应谨慎使用.  相似文献

The proliferative effects of endothelin-1 (ET-1), both alone and in combination with epidermal growth factor (EGF), and the effect of nitric oxide (NO) on the cell proliferation were investigated in cultured guinea pig bronchial smooth muscle cells. ET-1 (10-100 nM) alone augmented cell proliferation, and was additive to the effect of EGF (0.48 nM) in a concentration-dependent manner. An ET(A) antagonist, BQ-123 (10 microM), reduced the cell-proliferative effect of ET-1, whereas an ET(B) antagonist, BQ-788 (10 microM), did not influence the effect. A NO donor, SIN-1 (10 nM-1 microM), reduced the cell-proliferative effect of ET-1 in a concentration-dependent manner. The effect of SIN-1 (1 microM) was partly, but significantly, reversed by a soluble guanylyl cyclase inhibitor, ODQ (1 microM). These results suggest that ET-1 acts not only as a co-mitogen with EGF but also as a mitogen alone, and that its action is mediated through activation of ET(A) receptors. Therefore, ET-1 may contribute to airway remodeling, a pathophysiological hallmark of asthma. In addition, NO, which is produced mainly in the airway epithelium and is partly mediated through cGMP-dependent pathway, may reduce the phenomenon.  相似文献

Inner ear barotrauma (IEB) that is caused by acute pressure changes can often lead to permanent severe sensorineural hearing loss (SNHL). However, the mechanism that causes IEB is still unknown. In the current study, we assessed the involvement of reactive oxygen species (ROS) in IEB and the therapeutic effect of 3-methyl 1-phenyl-2-pyrazolin-5-one (edaravone), which is a free radical scavenger. To create the IEB model, guinea pigs were subjected to quick pressure changes that resulted in acute SNHL. The animals were then divided into two groups, an edaravone-treated IEB group and a non-treated IEB group that only received normal saline. Immunohistochemical analyses for 8-hydroxy-2-deoxyguanosine (8-OHdG) and 4-hydroxy-2-nonenal (4-HNE) were performed to examine the amount of oxidative DNA damage and lipid peroxidation that occurred in guinea pig cochlea. To assess the curative efficacy of edaravone, auditory brainstem response (ABR) testing was performed to evaluate auditory function. Strong immunoreactivities against 8-OHdG and 4-HNE were observed in the inner ear tissues of the non-treated IEB group. Lesser amounts of immunoreactivity were observed in the same region of the edaravone-treated IEB group as compared to the non-treated IEB group. Furthermore, ABR measurement revealed that there was a faster improvement in the threshold shift for the edaravone-treated IEB group as compared to that of the non-treated IEB group. At the final 7-week time point, the threshold shift for the edaravone-treated IEB group was significantly smaller as compared to the non-treated IEB group. These results strongly suggest that ROS is produced in the cochlea in response to acute pressure changes and that ROS plays an important role in the pathophysiology of IEB. Furthermore, edaravone treatment had a therapeutic effect on IEB-induced acute SNHL and thus, edaravone might possibly be able to be used as a therapeutic treatment for IEB.  相似文献

Cell morphology and proliferation was investigated in the atretic follicles during estrous cycles in the guinea pig. Ovarian samples on days 1, 4, 8, 12 and 16 of the estrous cycle in the guinea pig were taken in the morning for histologic staining with hematoxylin and eosin (HE), and immunohistochemical staining of the protein proliferating cell nuclear antigen (PCNA). The results indicated that the granulosa cells degenerated and eliminated first in atretic follicles, while the fibroblast-like cells appeared in the innermost layer of theca interna cells. When the fibroblast-like cells migrated to the antrum, they proliferated and formed a new tissue in peripheral to the zona pellucida of the oocyte. Our results also revealed that the orientation of the theca interna cell arrangement changed twice during the process of atresia, and the loose connective tissue in the antrum was critical for follicular atresia. Therefore, follicular atresia was not a simple process of cell death and elimination, but coexisted with cell proliferation. To our knowledge, we have for the first time confirmed cell proliferation and the presence of new tissue in atretic follicles in guinea pigs.  相似文献

Calcitonin (CT) is a 32 amino acidic polypeptide hormone which has been found in almost all species and whose effects are mainly concerned with calcium and phosphorous homeostasis. Three preparations are employed for therapeutic uses: salmon (sCT), porcine (pCT) and human CT (hCT). The sCT is the most powerful one and in human volunteers a strong relaxing effect has been shown on gallbladder (GB) basal volume and emptying in response to a meal, intraduodenal instillation of a liquid meal and i.v. cholecystokinin (CCK) infusion. Our study was aimed at investigating if a direct sCT effect could be demonstrated on smooth muscle strips from guinea pig GBs "in vitro" (organ bath). Isometric contractions were measured in response to maximal doses of acetylcholine (ACh: 10(-4) M), KCl (80 mM) and cholecystokinin octapeptide (CCK-OP: 10(-6) M), in absence and in presence of four doses of sCT (1 x 10(-9), 1 x 10(-8), 1 x 10(-7) and 1 x 10(-6) M). sCT did not affect the initial strip basal tone. ACh, CCK-OP and KCl caused, as expected, a powerful contraction of the strips, but no effect was shown when each of the sCT doses was administered before ACh (1.28+ 0.69 SEM without sCT vs 1.28g+ 0.69 with sCT; n = 6) and CCK-OP (1.46g+ 0.19 without sCT vs 1.46g+ 0.19 with sCT; n = 8) or 5 min after the induced KCl contraction. On the basis of these preliminary results, we conclude that no evidence of a direct sCT effect was found on guinea pig GBs when considering either basal smooth muscle tone or isometric contraction in response to ACh, KCl and CCK-OP. Further studies are therefore required to clarify the influence of CT on GB dynamics in vivo and to elucidate its the physiological significance.  相似文献

目的:观察Cajal样间质细胞(ICCs)在成年豚鼠膀胱的分布情况.方法:取5只成年豚鼠膀胱,制作全层冰冻切片,行ICCs特异性标志物c-kit免疫荧光染色,按粘膜层、粘膜下层和肌层进行统计分析.结果:豚鼠膀胱内可见c-kit免疫阳性细胞,胞体呈梭形,两端伸出长的突起,其形态与肠壁肌层Cajal细胞相似.且肌层和粘膜下层多于粘膜层.结论:豚鼠膀胱存在Cajal样间质细胞,并在肌层高表达,可能参与膀胱自主节律性运动的调控,调节逼尿肌的运动,为"神经-Cajal样间质细胞-肌肉"单元的形成提供组织学基础.  相似文献

Summary Electron-microscopical studies on the organ of Corti in kanamycin-intoxicated guinea pigs showed that progressive changes occurred in the cochlear hair cells. The changes appeared first in the basal turn of the organ of Corti, and affected primarily the outer hair-cells. During the later stages of intoxication, degeneration spread from the basal turn upwards, in the direction of the apex of the cochlea and, finally, affected all the external hair-cells.The early changes caused clumping of the chromatin in the nucleus, swelling of the nuclear membrane, damage in the mitochondria, with vesiculation of the mitochondrial cristae, and the formation of lamellated structures. The lysosomes were converted into dense bodies exhibiting various degeneration patterns. The ribosomes disappeared at an early stage from the cytoplasm.Even in cells where severe changes had occurred the hairs and cuticle seemed to be intact.During the final stages the plasma membrane, as well as the hairs and cuticle, was destroyed, and only remnants of these structures remained.Even after the disappearance of the sensory cells and the afferent nerve endings, some efferent nerve endings were still present in the organ of Corti.The action of kanamycin seems to differ slightly from that of streptomycin, since the changes caused by kanamycin in the plasma membrane are a fairly late effect, whereas the earliest changes occur apparently in the RNA protein-synthesis system and in the mitochondria of the external hair-cells.
Zusammenfassung Elektronenmikroskopische Untersuchungen am Cortischen Organ von Meerschweinchen mit Kanamycinintoxikation ergaben eine fortschreitende Schädigung der cochlearen Haarzellen. Die Veränderungen begannen in der basalen Schneckenwindung und betrafen vorwiegend die äußeren Haarzellen. In späteren Intoxikationsstadien breitete sich die Degeneration von der Basalwindung nach aufwärts bis in die Schneckenspitze aus und wurde schließlich an allen äußeren Haarzellen gesehen.Die früheren Veränderungen manifestierten sich am Kern mit Chromatinverklumpung, Schwellen der Kernmembran, Schädigung der Mitochondrion mit vesikulären Cristae mitochondriales und Bildung von Lamellenstrukturen. Die Lysosomen waren in dichte Körper verwandelt und wiesen verschiedene Degenerationsmuster auf. Die Ribosomen verschwanden früh aus dem Cytoplasma. Sogar in schwer geschädigten Zellen erschienen Haare und Cuticula intakt.In den Endstadien waren Plasmamembran, Haare und Cuticula bis auf Reste zerstört. Einige efferente Nervenendigungen waren selbst nach dem Verschwinden der Haarzellen und der afferenten Endigungen noch anzutreffen.Die Wirkung von Kanamycin ist offenbar etwas anders als die von Streptomycin, da die Veränderungen an der Plasmamembran ziemlich spät auftreten, während sich die frühesten Schäden im System der RNA-Proteinsynthese und in den Mitochondrien der äußeren Haarzellen zu manifestieren scheinen.
