Temperate phages were induced from Streptococcus cremoris R1, BK5, and 134. DNA from the three induced phages was shown to be homologous with prophage DNA in the bacterial chromosomes of their lysogenic hosts by the Southern blot hybridization technique. 32P-labeled DNA from 11 lytic phages which had been isolated on cheese starters was similarly hybridized with DNA from 36 strains of lactic streptococci. No significant homology was detected between the phage and bacterial DNA. Phages and lactic streptococci used included phages isolated in a recently opened cheese plant and all the starter strains used in the plant since it commenced operation. The three temperate phages were compared by DNA-DNA hybridizations with 25 lytic phages isolated on cheese starters. Little or no homology was found between DNA from the temperate and lytic phages. In contrast, temperate phages showed a partial relationship with one another. Temperate phage DNA also showed partial homology with DNA from a number of strains of lactic streptococci, many of which have been shown to be lysogenic. This suggests that many temperate phages in lactic streptococci may be related to one another and therefore may be homoimmune with one another. These findings indicate that the release of temperate phages from starter cells currently in use is unlikely to be the predominant source of lytic phages in cheese plants.  相似文献

Sixty-three strains of lactic streptococci isolated from commercial lactic streptococcal starter cultures were examined for lysogeny by treatment with ultraviolet light or mitomycin C. After treatment with the inducing agent, all strains, whether or not they lysed, were examined for evidence of phage release by electron microscopy. Thirty-eight strains yielded intact phages or phage particles of varying morphology. All the temperate phages had isometric heads and noncontractile tails; some had collars and structurally distinctive baseplates. Indicator host strains were found for phages induced from seven different strains. Three strains that released phages spontaneously yielded titers of 10(3) to 10(4) plaque-forming units per ml. When strains that spontaneously released phages were grown in mixed culture with indicator strains, increased phage titers of 10(6) to 10(7) plaque-forming units per ml were observed. These findings indicate that lysogenic lactic streptococcal strains may serve as a reservoir for phages that attack sensitive strains in mixed- or multiple-strain lactic starter cultures.  相似文献

Lactobacilli are members of the bacterial flora of lactic starter cultures used to generate lactic acid fermentation in a number of animal or plant products used as human or animals foods. They can be affected by phage outbreaks, which can result in faulty and depreciated products. Two groups of phages specific of Lactobacillus casei have been thoroughly studied. 1. The first group is represented by phage PL-1. This phage behaves as lytic in its usual host L. casei ATCC 27092, but can lysogenize another strain, L. casei ATCC 334. Bacterial receptors of this phage are located in a cell-wall polysaccharide and rhamnose is the main component of the receptors. Ca2+ and adenosine triphosphate (ATP) are indispensable to ensure the injection of the phage DNA into the bacterial cell. The phage DNA is double-stranded, mostly linear, but with cohesive ends which enables it to be circularized. The vegetative growth of PL-1 proceeds according to the classical mode. Cell lysis is produced by an N-acetyl-muramidase at the end of vegetative growth. 2. The second group is represented by the temperate phage phi FSW of L. casei ATCC27139. It has been shown how virulent phages originate from this temperate phage in Japanese dairy plants. The lysogenic state of phi FSW can be altered either by point mutations or by the insertion of a mobile genetic element called ISL 1, which comes from the bacterial chromosome. This is the first transposable element that has been described in lactobacilli. Lysogeny appears to be widespread among lactobacilli since one study showed that 27% of 148 strains studied, representing 15 species, produced phage particles after induction by mitomycin C. Similarly, 23 out of 30 strains of Lactobacillus salivarius are lysogenic and produce, after induction by mitomycin C, temperate phages, killer particles, or defective phages. Temperate phages have also been found in 10 out of 105 strains of Lactobacillus bulgaricus or Lactobacillus lactis after induction by mitomycin C. Phages so far studied of the latter 2 and closely related lactobacilli, either temperate or isolated as lytic, may be divided into 4 unrelated groups called a, b, c and d. Most of these phages are found in group a and an unquestionable relationship has already been shown between lytic phages and temperate phages that belong to this group. Lytic phage LL-H of L. lactis LL 23, isolated in Finland, is one of the most representative of those of group a and has been extensively studied on the molecular level.  相似文献

Some characteristics of Bacillus subtilis phage SPO2 which show that it is a temperate phage are presented. Wild-type SPO2 forms turbid plaques, similar to those of other temperate phages. SPO2 lysogenic strains which are resistant to SPO2 can be isolated; these strains remain stable lysogens despite the fact that they can no longer adsorb SPO2. SPO2 lysogenic strains can be grown for many generations in SPO2 antiserum and remain lysogenic. Phage SPO2 plates on phi105 lysogens and phage phi105 plates on SPO2 lysogens; this indicates that SPO2 and phi105 are heteroimmune. Phage phi105 plates on an SPO2-resistant strain; this indicates that SPO2 and phi105 adsorb to different receptor sites on the bacterial surface.  相似文献

A large number of strains of Oenococcus oeni (formerly Leuconostoc oenos) that had been isolated from wines were checked for lysogeny with mitomycin C as inducer. As a result of this test, 45% of the strains proved to be lysogenic, suggesting that lysogeny is widespread among bacteria isolated from wines during malolactic fermentation. The sensitivity of bacteria to phages was very different, depending on the strain. All the lysogenic strains were resistant to infection by the temperate phage they released. Some phages infected none of the strains. Phages of Oenoc. oeni had a classical morphology, an isometric head, and a long striated tail. Seven bacteriophages active on Yersinia enterocolitica (YE) were isolated from surface water samples collected in Granada, Spain. A comparison of the respective host ranges of these new phages and of reference phages used for YE phage typing showed that YE strains belonging to various phage types, grown at either 37 or 25 degrees C, expressed susceptibility to reference sewage water phages whereas susceptibility to new waterborne phages, as well as to reference phages from lysogenic YE, was only demonstrated in YE strains grown at 25 degrees C. A YE strain isolated by stool culture from a pig was lysogenic for a bacteriophage which behaved like waterborne phages and reference phages from lysogenic YE strains. The possibility that the isolation of waterborne bacteriophages might, in certain circumstances, reflect the presence of lysogenic YE was raised.  相似文献

Five temperate phages were isolated from strain 4042B of Bacillus thuringiensis subsp. aizawai. The phages, which were heteroimmune, could also be distinguished by their host ranges, plaque and particle morphologies, serological specificities, and locations of restriction endonuclease cleavage sites on their chromosomes. Besides maintaining a stable lysogenic relationship with the 4042B host strain, each phage formed a stable lysogen with Bacillus cereus.  相似文献

All of the 1070 strains of Salmonella weltevreden tested were lysogenic, temperate phage being demonstrable in 83.9% of the strains in broth culture, 6.4% in mixed culture and the remaining 9.7% after induction by UV-irradiation and MC treatment. A phage-typing scheme for Salmonella weltevreden based on the host range of temperate phages was developed. The strains could be classified into eight groups by means of the host range of their temperate phages on six indicator strains and a ninth group, the symbiotic phages of which failed to lyse any of the indicator strains.  相似文献

Aims: Frequency of lysogeny in Lactobacillus delbrueckii strains (from commercial and natural starters) and preliminary characterization of temperate bacteriophages isolated from them. Methods and Results: Induction of strains (a total of 16) was made using mitomycin C (MC) (0·5 μg ml−1). For 37% of the MC-treated supernatants, it was possible to detect phage particles or presence of killing activity, but only two active bacteriophages were isolated. The two temperate phages isolated were prolate-headed phages which belonged to group c of Lact. delbrueckii bacteriophages classification. Different DNA restriction patterns were obtained for each phage, while the structural protein profiles and packaging sites were identical. Distinctive one-step growth curves were exhibited by each phage. An influence of calcium ions was observed for their lysis in broth but not on the adsorption levels. Conclusions: Our study showed that lysogeny is also present in Lact. delbrueckii strains, including commercial strains. Significance and Impact of the Study: Commercial strains could be lysogenic and this fact has a great practical importance since they could contribute to the dissemination of active-phage particles in industrial environments.  相似文献

A total of 17 of 21 Lactobacillus strains of the subgenus Streptobacterium were lysogenic. Two different temperate phages isolated from such lysogens are very similar to Lactobacillus casei phage PL-1. The narrow host range of bacteriophage PL-1 appears to be caused by homoimmunity.  相似文献

Six strains of thermophilic hydrocarbon-oxidizing Actinomyces griseomycini were tested for lysogeny. The lysogenic state was detected in the four strains and their phages were isolated. The phages isolated from the three strains were virulent and cause lysis of the host culture. All isolated phages were specific and did not cause lysis of other actinomycetes species. However, the phages had different activity towards the six studied strains of Actinomyces griseomycini. One phage induced lysis only of its indicator culture, other phages caused lysis of four strains, including those two from which no phage was isolated. All phages produced negative colonies of identical morphology. The morphology of their particles was also the same. The phages induced cross-resistance and were characterized by thermoresistance.  相似文献

Our investigations to detect naturally lysogenic strains of mycobacteria were limited to 1 strain ofMycobacterium smegmatis, 4 strains ofMycobacterium borstelense var.niacinogenes, and to 5 strains ofMycobacterium marinum (Syn:Mycobacterium balnei), all together 10 strains. They were chosen because as a sign of lysis they secrete a large quantity of cytoplasmatic components (nucleic acids proteins, amino acids etc.) into the fluid medium (for instance phosphate buffer), in which they are suspended. In a first series of experiments culture filtrates were tested on 84 strains of slowly and rapidly growingMycobacterium species as indicator strains. Using this method free phage particles were only found in the culture filtrate of 1 strain,Mycobacterium smegmatis SN 46, isolated from a patient with achalasia. Phage particles could not be found in the filtrates of the other 9 probably lysogenic strains. In a second series of experiments more closely related indicator strains were used. The 10 probably lysogenic strains were cultured in bovine serum or antiphage-antiserum containing medium and single selected colony cultures a small part of which showed sensitivity to the filtrates. The released and adapted phages, designated as B24, B30, B32, B33, B34 and B35 have a very narrow host range. The plaques are very small and turbid. On electron micrographs the temperate phages B24, B30 and B35 exhibit the typical head-tail morphology. The head of the temperateborstelense var.niacinogenes phage B30 is 45 nm in diameter, the tength of tail is about, 120nm. The average dimensions of the long head ofsmegmatis phage B24 are 40 × 80 nm, the tail is about 160 nm long. The balnei phage B35 is very similar morphologically to phage B30. The head is about 50 nm in diameter, the length of tail about 160 nm. The phage sensitive variants are not “carrier” strains. Their phage sensitivity is not a stable property. After several culture passages in serum-free medium the variants regain their phage immunity completely and release phages like the lysogenic parent strains. The sensitive variants must therefore be considered to be also lysogenic. TheMycobacterium borstelense var.niacinogenes phages are serologically very related. A comparison was made between the results of phage and lysogenic typing of S. aureus strains isolated during several outbreaks of staphylococcal infection and S. aureus cultures isolated from the same carriers at different periods. The study of the groups of strains having the same origin showed that the differences in the number of reactions were more pronounced in lysogenic typing than in phage typing. For this reason lysogenic typing can be recommended only for the identification of those strains which cannot be identified with the use of the phages of the International Basic Set. The results of the experiments with induced phages proliferating in a restriction-defective strain indicated that restriction and modification were mainly responsible for the specificity of lytic reactions.  相似文献

A series of mutants derived from the temperate corynebacteriophages beta(tox+), gamma(tox-), and L(tox+) was isolated and characterized. In three-factor crosses between mutant beta phages the relative map order of the genetic markers determining extended host ranges (h and h') and loss of ability to lysogenize (c) was found to be h--c--h'. Recombination between markers was observed in matings between phage beta and the heteroimmune corynebacteriophages gamma and L. In such matings between heteroimmune phages the c markers of phages beta and gamma failed to segregate from the imm markers which determine the specificity of lysogenic immunity in these phages. The factor which directs the synthesis of diphtherial toxin during infection of appropriate corynebacterial hosts by toxinogenic corynebacteriophages is designated tox(+). It was possible to show that the tox(+) determinant of phage beta behaves as a single genetic element which occupies a position between the loci h and imm on the genetic map of this phage. Genetic recombination between mutants of phage beta occurred at very low frequencies in biparental matings performed by mixed infection of Corynebacterium diphtheriae C7(s)(-)(tox-). Considerably higher recombination frequencies were observed when lysogenic bacterial strains carrying one parental phage as prophage were induced by ultraviolet irradiation and then superinfected by the second parental phage. Maximal stimulation of genetic recombination between mutant beta phages was detected when superinfection followed ultraviolet irradiation of the lysogenic cells within a limited period of time. In matings between phages with incomplete genetic homology, the stimulation of recombination by ultraviolet radiation was much less effective.  相似文献

To study phage-mediated gene transfer in Yersinia, the ability of Yersinia phages to transduce naturally occurring plasmids was investigated. The transduction experiments were performed with a temperate phage isolated from a pathogenic Yersinia enterocolitica strain and phage mixtures isolated from sewage. Small plasmids (4.3 and 5.8 kb) were transduced at a frequency of 10−5 to 10−7/PFU. However, we could not detect the transduction of any indigenous virulence plasmid (ca. 72 kb) in pathogenic Yersinia strains. Transductants obtained by infection with the temperate phage were lysogenic and harbored the phage genome in their chromosomes.  相似文献

Wild-type bacteriophage phie and IS (interference-sensitive) mutants of the related phage SP82G did not productively infect strains of Bacillus subtilis that were lysogenic for temperate phage SPO2. In these abortive infections, the sensitive phages adsorbed to and penetrated the nonpermissive host, phage-directed macromolecular syntheses were initiated, but both viral and bacterial nucleic acid production abruptly stopped about 15 min after addition of the phages. The cessation of RNA and DNA synthesis was preceded or coincident with a reduction in oxygen utilization by the infected cultures. Genetic studies of both phie and SP82G suggest sensitivity to SPO2-mediated abortive infection was controlled by a single gene. A mutant of SPO2, SPO2ehp4-, lysogens of which no longer interfere with the development of SP82GIs, was also isolated. The discovery of this ehp- variant suggests the normal SPO2 prophage synthesized a substance that alters cell physiology in some manner detrimental to SP82GIs development. Since SPO2ehp4- grew on and lysogenized bacteria sensitive to wild-type SPO2, the product of the eph gene was apparently not an essential function of this temperate phage.Overall, these observations exhibit remarkable similarities to the inhibition of T4rII mutants by the product of the rex gene of phage lambda.  相似文献

The possibility of using phages, isolated from the lysogenic cultures of methicillin-resistant staphylococci and modified in methicillin-resistant cultures of phage 85, for the differentiation of nontypable staphylococcal strains has been studied. The variants of phage 85 cannot be used for the determination of differences between the strains of methicillin-resistant cultures; they are not suited for typing. For this purpose the collection of phages isolated from lysogenic methicillin-resistant cultures should be used.  相似文献

Forty-eight strains of lactobacilli used as starter strains in the dairy industry were examined for lysogeny after treatment with mitomycin C. Two strains of L. delbrueckii subsp. bulgaricus were able to produce active phages. These temperate phages as well as 4 virulent phages isolated during abnormal fermentations were compared to a previously characterized phage mv4 which is temperate. All these phages were shown to be partially homologous by DNA-DNA hybridization. Genes that code for viral proteins seem to be well conserved since 2 major virion polypeptides of 18 (or 19) kD and 34 kD could be detected in the protein composition of each phage. Immunoblotting studies of the 7 phages using serum raised against phage mv4 confirmed that the proteins of the different phages were related. All these phages can be classified in the previously constituted group a, which now comprises 4 temperate and 15 virulent phages. These results show that some virulent phages appearing during abnormal fermentations and some temperate phages isolated by appearing during abnormal fermentations and some temperate phages isolated by induction of starter strains can be closely related genetically. Five virulent phages of L. helveticus were also compared according to their restriction pattern and their DNA homology. They were shown to be related to one another, but unrelated to phages of other lactic acid bacteria species.  相似文献

One virulent phage from soil lysed all tested strains ofFaenia rectivirgula, whereas six others—three of them from lysogenic cultures—lysed only nonlysogenic strains. Restriction analysis showed that these six phages are closely related, although one of them is virulent and the others are temperate. Another member of this group is aStreptomyces erythraeus phage, which also lysed only the four nonlysogenic strains ofF. rectivirgula.  相似文献
