Explants from young joints of mature plants of tuna (Opuntia ficus-indica Mill.) were cultured on Murashige and Skoog (MS) medium containing 8.8 M benzyladenine (BA) and 0.5 M naphthaleneacetic acid (NAA). Shoots produced were utilized as secondary explants. Each shoot was cut longitudinally from apex to base into two explants, and some of these explants were cut transversely into proximal and distal explants. The size and number of shoots produced was affected by size and position of the explant within its source. The shoots were rooted in vitro or ex vitro and plants were successfully established in soil from both rooting methods.Abbreviations AC

Cultures of Lavandula latifolia Medicus were established from axillary buds of mature field-grown plants. Explants were initially cultured on media with two different macronutrient combinations and benzyladenine or kinetin added either individually or with naphthaleneacetic acid. Subsequently, explants were subcultured in Murashige and Skoog medium supplemented with 20% coconut milk, 0.57 M indoleacetic acid and 8.88 M benzyladenine. Shoot proliferation from axillary buds was not affected by seasonal fluctuations in the stock plants but depended on the macronutrient composition and on the type and concentration of cytokinin tested. Best results were obtained in explants initially cultured in media with Murashige and Skoog constituents and supplemented with 5 M benzyladenine. In vitro-grown shoots were used to induce multiple shooting by transferring them to subculture medium. Shoots were rooted on Murashige and Skoog medium with macronutrients at half-strength. Shoot induction frequency for the leguminous tree Swartzia madagascariensis Desv. was higher on MS and WP media than on B5. Explants incubated on media solidified with agar produced more shoots with a lower tendency to hyperhydricity than explants on agarose or Gelrite media. Maximum shoot induction was obtained with an agar-solidified MS medium containing 2.2 M benzyladenine (37 shoots/explant). Micropropagation of the Mediterranean species Viburnum tinus 总被引:4,自引:0,他引:4
In vitro propagation of the Mediterranean species Viburnum tinus L. was established from an outdoor-grown shrub. Two standard macrosalt formulations (Margara N30K and Murashige and Skoog), a range of benzyladenine and sucrose concentrations were tested for their effect on shoot multiplication. The cytokinin concentration was the most important factor affecting shoot multiplication. The highest shoot multiplication rate was obtained from single-node explants cultured on Murashige and Skoog medium supplemented with 4.4 M benzyladenine. Cytokinin concentration and an interaction of macrosalts and benzyladenine influenced shoot length on the multiplication stage: best shoot growth was observed on MS medium containing 1.1 M benzyladenine. In addition, sucrose concentrations of 87.6–146.0 mM gave the highest multiplication rates and improved shoot growth. Following a shoot ellongation stage, single shoots were rooted on media containing naphtaleneacetic acid (1.3–5.4 M). Although enhanced in vitro rooting was obtained on media containing 5.4 M naphtaleneacetic acid, reducing the auxin concentration to 1.3 M during the in vitro rooting stage improved acclimatisation frequency and further plant growth in a horticultural substrate. 相似文献

Proliferating cultures ofGarrya elliptica were established from shoot tip and nodal explants grown on woody plant medium containing benzyladenine. Optimum numbers of healthy shoots were induced on media containing 1.25 M benzyladenine; higher concentrations caused more hyperhydricity and less extension. Following growth for 28 days on quarter-strength woody plant medium containing 0.2% activated charcoal, 90% of shoots rooted on the same medium lacking charcoal, but with 2.5 M indolebutyric acid. Survival during weaning was poor, and was strongly correlated with the time of year at which plants were removed from culture.Abbreviations WPM

A procedure for multiple shoot formation from somatic embryo explants of Eastern redbud (Cercis canadensis L.) cultured on DKW medium containing benzyladenine (BA) and thidiazuron (TDZ) was developed. TDZ in combination with BA produced more shoots than either treatment alone. The highest number of shoots (3.3 to 3.4 shoots per explant) was obtained from partially desiccated and wounded explants treated with a combination of 5 or 10 M BA and 0.5 or 1.0 M TDZ for 20 days before being transferred to the same medium without TDZ. The number of shoots formed was increased from 1.5 to 3.2 shoots per explant by cutting through the cotyledonary node prior to culture. In addition, the frequency of explants forming shoots was increased by desiccation of somatic embryo explants to approximately 50% moisture and by using somatic embryos with two well formed cotyledons as explants.Abbreviations ABA

Techniques for the regeneration of Jatropha curcas L. from various explants have been developed. Regeneration from hypocotyl, petiole and leaf explants was evaluated on a range of concentrations of zeatin, kinetin and N6 benzyladenine (BA) either singly or in combination with indole-3-butyric acid (IBA). Higher regeneration from hypocotyl and petiole explants was obtained on BA with IBA than on zeatin- or kinetin-supplemented media. Leaf discs from the third expanding leaf exhibited higher regeneration potential than those from the fourth leaf. Independent of the explant type, direct adventitious shoot bud induction was recorded highest on MS medium with 2.22 M BA and 4.9 M IBA. Although the same BA concentration but with reduced IBA concentration (0.49 M) proved effective in callus mediated regeneration from hypocotyl and leaf explants, the petioles required lower concentrations of the two growth regulators (0.44 M BA and 0.49 M IBA). Regenerated shoots could be rooted on growth regulator-free gelled full-strength MS medium. Following simple hardening procedures, the in vitro-raised plants could be transferred to soil and grown to maturity in the field.Abbreviations BA

Embryos extracted from dried seeds of common ash (Fraxinus excelsior), were germinated on growth regulator-free culture medium. Cotyledonary nodes from these seedlings were placed onto Murashige and Skoog, Woody Plant or Driver and Kuniyuki culture media with 22.2 or 44.4 M benzyladenine, on which they developed into shoot cultures following the outgrowth of axillary buds. With Murashige and Skoog medium, cultures often died. With Woody Plant Medium, survival of the cultures was considerably improved, but large amounts of callus were produced at the cut ends of the explants, and new axillary shoots had long internodes and small leaves. With Driver and Kuniyuki medium, both survival and callus formation were much improved, and the shoots produced were of high quality. Proliferation of axillary shoots was obtained from both shoot tip and nodal explants placed onto Driver and Kuniyuki medium with 22.2 M benzyladenine. Adventitious root formation was best with shoots inserted into half-strength Woody Plant Medium containing 2.45, 4.9 or 9.8 M indolebutyric acid. A protocol was developed for direct differentiation of multiple shoot buds from leaf explants of Cajanus cajan. In a modified Murashige and Skoog's medium supplemented with 2.22 µM benzyladenine (BA), 0.57 µM indole-3-acetic acid (IAA) and 41 µM adenine sulphate (AdS), the segments of basal halves of the first two leaves of a young seedling incubated on filter paper bridges in liquid medium took 20 – 25 d to differentiate shoot buds. The explants after transfer to solidified medium, with lower concentration of BA (0.22 M) resulted in fast growing healthy shoots. The developed shoots (measuring ca. 3 cm) were rooted in a medium supplemented with 1.42 µM IAA. They were subsequently grown in pots with soil with more than 80 % transplantation success. 相似文献

Xanthium strumarium L. was micropropagated by rooting shoots proliferated from shoot-tip explants. The best shoot proliferation was obtained from explants growing on Murashige and Skoog medium supplemented with 4.4 to 8.9 M benzyladenine (BA) and 1.1 to 2.1 M naphthaleneacetic acid (NAA). The micropropagated plants were transferred to potting media and maintained under high humidity conditions in the greenhouse. The media that produced best shoot proliferation from shoot-tip explants also produced the most callus from hypotocotyl, cotyledon and shoot-tip explants, whereas more callus was produced on leaf explants with a lower BA concentration (1.1 M) and 1.1 M NAA.Abbreviations BA

Shoot regeneration was achieved from in vitro-produced leaves of Elaeagnus angustifolia L. Half-leaf explants from the terminal part of the shoot produced more shoots than explants from the basal part of the in vitro-derived shoots on agar-solidified WPM medium supplemented with 1 M benzyladenine (BA). In liquid medium of the same formulation, compact shoots that did not elongate were formed on the explants. Leaf cross-section explants (1 mm thick) produced shoots both on solid and liquid medium with 1 M BA, whereas again compact shoots were formed with 10 M BA. A simple, rapid and reproducible protocol for direct shoot regeneration from different explants of Jatropha integerrima was developed. Prolific adventitious shoot bud initiation was obtained using a combination of 2.2 or 4.4 M benzyladenine and 4.9 M indole-3-butyric acid (IBA). Reduction of IBA concentration (2.5 M) promoted further development of shoots. Regenerated shoots rooted readily on Murashige and Skoog (MS) medium lacking growth regulators. A system for in vitro clonal propagation has been developed in tea plants. Shoots obtained from primary explants were induced from terminal buds and axillary buds of mature field-grown plants. Cultures were initiated from both types of explants on Murashige and Skoog (MS) medium supplemented with 10% coconut milk (CM), 200 mg l-1 of yeast extract (YE), 1.4 M indoleacetic acid (IAA) and 17.8 M benzyladenine (BA). The shoot tips were multiplied on 1/2 strength MS medium containing 10% CM, 2.9 M IAA and 17.8 M BA. The larger shoots were separated after multiplication and rooted on 1/2 MS medium supplemented with 11.4 M ascorbic acid and 34.5 M indolebutyric acid (IBA). A pretreatment of the plants with an aqueous solution of 493 M IBA greatly increased the frequency of rooting. Regeneration of Aeschynomene sensitiva Sw. after callogenesis was obtained from small (2–5 mm long) root explants of 30-day-old seedlings aseptically cultivated on Murashige and Skoog medium supplemented with various concentrations of growth regulators. After 4 weeks, the best results were observed with 0.54 M -naphthaleneacetic acid and 2.22 M benzyladenine. On this medium, the rate of regeneration depended on seedling age and agar concentration. The highest number of shoots per explant was obtained with small cuttings from 30-day-old seedlings grown on a medium containing 8 g l–1 of agar. Regeneration success was also dependent on explant size. When longer explants (7–20 mm) were cut from the main root, direct regeneration was obtained in two weeks. These cuttings also generated shoots through callogenesis in four weeks but always in lower quantities than with direct regeneration, whatever the seedling age. here also, the best regeneration was obtained with cuttings from 30-day-old seedlings maintained on a medium with 8 g l–1 of agar. Regenerants were rooted on growth-regulator-free Murashige and Skoog medium and then acclimatized in a greenhouse. A better survival to transplantation was observed when plantlets were inoculated with the photosynthetic Bradyrhizobium strain ORS 278. Immature and mature nonstratified seeds of white ash (Fraxinus americana L.) were dissected transversely and 2/3 of each seed was placed onto agar-solidified Murashige and Skoog medium. Adventitious buds, shoots, and somatic embryos formed on callus, cotyledons, and hypocotyls of the resulting seedlings. Shoot organogenesis was induced on explants cultured on medium with 10 M thidiazuron but not on explants on media with benzyladenine (BA) or isopentenyladenine. Not all seed sources were equally capable of shoot organogenesis and embryogenesis. Atypical of adventitious regeneration of other woody plants, mature seed explants of white ash were more organogenic with shoots that elongated better than explants from immature seeds. Somatic embryogenesis was observed in cultures where mature seeds were first cultured for 4 weeks on a medium containing 10 M adenine 2,4-dichlorophenoxyacetic acid in combination with 0.1 and 1.0 M thidiazuron, followed by transfer to a medium containing 0.05 M 6-benzyladenine and 0.5 M naphthaleneacetic acid. An efficient in vitro plant regeneration system via hypocotyl segments of tetraploid Isatis indigotica Fort. was established. Murashige and Skoog's (MS) and Gamborg's (GB5) media were found to be superior to White medium for promoting shoot regeneration. The highest shoot regeneration (92 %) was achieved from hypocotyls cultured on MS medium containing 8.9 M benzyladenine (BA) and 2.7 M naphthaleneacetic acid (NAA), with an average of 4.2 shoots developed per explant. Plant regeneration was also improved when the explants were cultured in MS basal medium containing 3 % (m/v) sucrose and grown under a 12-h photoperiod. The developed shoots were well rooted in a half-strength MS medium supplemented with 0.5 M indole-3-butyric acid (IBA) and were morphologically normal after transfer to soil. 相似文献

Culture conditions have been established for callus induction and growth from different explants in L. angustissimus L. Calli were obtained from hypocotyls, leaves, stems, cotyledons and roots cultured on media containing 2,4-dichlorophenoxyacetic acid or -naphthaleneacetic acid with kinetin, N6 – 2 or benzyladenine in different combinations and concentrations. Only those calli induced in presence of -naphthaleneacetic acid with benzyladenine or kinetin produced shoots. Calli induced from hypocotyl explants were the most efficient in regeneration of shoots. Transformation with an Agrobacterium rhizogenes binary vector carrying the plasmid pBI 121.1 is reported. The percentage of cotransformation was estimated by testing GUS activity in hairy roots. The integration of Ri T-DNA and the NPTII gene in transformed plants was confirmed by molecular analyses and in vitro culture of transgenic tissues in the presence of kanamycin.Abbreviations BA

Protocols have been developed for the in vitro production of plants from callus derived from root explants of Lathyrus sativus cv. P-24. Callus and shoot regeneration were achieved only in MS medium supplemented with 10.7 M naphthaleneacetic acid and an increased concentration of kinetin (0.9 M for 14 days to 1.4 M for 18 days) during callusing. The shoots obtained rooted in 1/2 MS supplemented with 0.5 M indolebutyric acid. During the year plants have been regenerated several times.
