为了明确大鼠背根节(DRG)神经元中存在慢的Ca2+激活K+电流成分,本实验在新鲜分散的DRG神经元胞体上,采用全细胞电压箝技术,给予DRG神经元一定强度的去极化刺激,记录刺激结束后30 ms时的尾电流幅度.结果发现:(1)随着去极化时间从1 ms延长至180 ms时,尾电流幅度由9.3±2.8 pA逐渐增大至64.1±3.4 pA(P<0.001);(2)当去极化结束后的复极化电位降低时,尾电流幅度先逐渐下降到零,然后改变方向,逆转电位约为-63 mV;(3)细胞外施加500μmol/L Cd2+或细胞内液中施加11 mmol/L EGYA时尾电流明显减小甚至完全消失;(4)尾电流中慢成分的幅度在细胞外给与200 nmol/L蜂毒明肽后,减小了约26.32±3.9%(P<0.01);(5)细胞外施加10 mmol/L TEA,可明显降低尾电流中的快成分.结果提示,在DRG神经元后超极化中存在Ca2+激活K+电流的蜂毒明肽敏感成分--ⅠAiHP.  相似文献

本文观察了心肌肥厚对大鼠心肌细胞Na /Ca2 交换电流的影响。我们采用Goldblatt两肾一夹方法诱发大鼠心肌肥厚,应用全细胞膜片钳技术记录电流。结果表明:肥厚心肌细胞的Ni2 -敏感Na /Ca2 交换电流密度大于正常细胞。在钳制电压为+50mV时,正常细胞的外向交换电流密度为1.53±0.31pA/pF,而肥厚细胞则为2.62±0.53pA/pF(P<0.01);钳制电压为-100mV时,正常细胞的内向交换电流密度为0.42±0.14pA/pF,肥厚细胞达1.12±0.33pA/pF(P<0.001)。这些结果提示,肥厚心肌细胞的Na /Ca2 交换电流发生了改变,其意义有待进一步探讨。  相似文献

在大鼠下丘脑薄片标本上对52例视上核神经元进行了全细胞膜片箝记录。膜被动及主动电生理参数测量如下:静息电位,59±8mV;输入阻抗,535±129MΩ;时间常数,32±9ms;动作电位幅度,99±11mV;超射值,37±13mV(n=39)。大多数神经元在接受去极化刺激时出现明显的慢后超极化电位或电流。我们发现,在电压箱状态下几乎所有的视上核神经元均接受兴奋性和/或抑制性突触传λ(n=13)。药理学实验表明,兴奋性突触后电流是由non-NMDA亚型谷氨酸受体介导,而抑制性突触后电流由GABAA受体介导。  相似文献

目的和方法 :采用大鼠海马脑片盲法膜片钳全细胞记录技术研究CA1区锥体神经元电压门控性Ca2 通道的动力学特征。结果 :大鼠海马脑片CA1区锥体神经元电压门控性Ca2 通道电流具有如下特点 :①激活的阈电位偏低 ,为 (- 4 9.3± 8.6 )mV ,范围为 - 6 5~ - 30mV(n =2 3)。②衰减时间常数τ值较大 ,且变化范围大 (10 0~ 70 0ms) (n =12 ) ,并且衰减具有Ca2 电流幅值的依赖性 ,③稳态失活呈现电压依赖性 ,半失活电压为 (- 5 5 .4± 9.7)mV ,斜率因子为 (5 .3± 0 .9)mV(n =10 )。④当细胞外Ca2 浓度为 2 .5mmol/L时 ,Ca2 通道的反转电位为 (5 5±13)mV(n =10 )。⑤尾电流成分较为单一 ,不表现电压依赖性。另外 ,Ca2 电流对戊脉胺及双氢吡啶类化合物硝苯地平均不敏感。结论 :根据上述Ca2 电流特征 ,海马脑片CA1区锥体神经元上的Ca2 通道主要以N型为主  相似文献

采用全细胞记录膜片钳技术,研究非洲爪蟾脑片视顶盖神经元微突触后电流(miniature inhibitory postsynaptic current,mIPSC)频率和振幅对电压依赖关系。观察到以下结果:(1)当通过改变记录电极内的DC电流,将神经元的膜电位从静息电位逐步(每步10mV的增量)去极化或超极化时,mIPSC的频率和,或振幅分别升高或降低。随着膜电位的去极化,mIPSC的频率逐渐升高;当钳位电压升至+10mV时,mIPSC的频率达到最高值。(2)当神经元去极化时,振幅仅轻微升高。膜电位去极化达到-30mV或-40mV时,mIPSC的振幅最大:进一步去极化,振幅反而下降。另外,在膜电位去极化至-20mV和+10mV之间时,可记录到大的mIPSC。(3)在无Ca^2+浸浴液中,mIPSC的频率和振幅也随膜电位的去极化而逐步增高,但频率的增高幅度远不如在生理盐水浸浴中增高幅度明显。(4)当浸浴液中[K+]0增高时,mIPSC的频率明显降低,而振幅轻微降低。当细胞外[K^+]。浓度升高超过20mmol/L时,神经元产生明显的缓慢内向或外向膜电流。mIPSC频率和振幅与膜电位存在依赖性的可能机制在文中作了简短的讨论。  相似文献

采用微电极技术对分散培养的新生大鼠海马神经元进行细胞内记录,对其被动膜电性质、Ⅰ-V关系曲线、动作电位等进行了观察。测得静息电位为-64±11mV(n=96),膜阻抗为80±20MΩ,时间常数5.6±1.4ms,动作电位幅度69±7mV(n=15),阈电位-48±8mV,超射值7±3mV。利用压力脉冲微量给药技术将乙酰胆碱或谷氨酸钠施加于所观察的细胞表面,发现二者均引起神经元去极化,并伴有强烈的放电反应。证明新生大鼠海马培养神经元细胞内记录是一种稳定可靠的电生理学研究方法。  相似文献

在豚鼠腹腔神经节上对383个神经元作细胞内记录,给予1μmol/L半琥珀酸皮质醇灌流,38个神经元膜电位发生超极化反应,幅度变化为2~12mV(6.3±0.1mV),伴有膜电阻的降低,反应呈剂量效应关系。9个神经元呈去极化反应,其余336个神经元不反应。用单电极间断电压箝方法记录43个神经元在糖皮质激素作用下膜电流的变化,其中5个神经元出现外向电流,膜电导增加;1个神经元为内向电流。用低钙高镁液阻断突触传递和蛋白质合成抑制剂放线菌素D后,超极化反应仍然存在。皮质醇超极化反应的翻转电位为-79.0±4.3mV(n=5)。皮质醇超极化反应和GABA去极化反应可在同一神经元上出现,印防己毒素可拮抗GABA的去极化反应,但不能拮抗皮质醇的超极化反应。钾离子通道阻断剂四乙基铵(TEA)和4-氨基吡啶(4-AP)能拮抗皮质醇的超极化反应。我们推断皮质醇的超极化反应是细胞膜钾离子通道介导的。  相似文献

在低浓度皂素(50μg/ml)制备的浆膜蜕变(通透性增高)而肌浆网(SR)膜无损的蜕膜心肌标本上,以其收缩的幅度作为SRCa(2 )释放的半定量指标;高浓度皂素(500μg/ml)制备的浆膜和SR膜均蜕变的蜕膜心肌标本,以其张力-PCa关系曲线以及产生50%最大张力所需的Ch(2 )浓度(PCa50)分别作为肌钙蛋白(TN)Ca(2 )敏感性的定性和定量指标。结果观察到:(1)在低浓度皂素蜕膜心肌标本上,5和10mmol/L咖啡因分别引起约89.2±12.7和142.5±17mg(n=4,P<0.05)张力的强直收缩,而5mmol/L茶碱未能引起明显的强直收缩;(2)在高浓度皂素蜕膜心肌标本上,5和10mmol/L咖啡因及5mmol/L茶碱均使张力-pCa关系曲线左移;PCa50较对照分别增加了0.261,0.274和0.212PCa单位(P均小于0.001)。上述结果提示:咖啡因和茶碱均能增高TNCa(2 )敏感性;此外,咖啡因尚有促使SR释放Ca(2 )的作用。  相似文献

目的:研究孤啡肽(N/OFQ)对大鼠顶叶皮层神经元瞬时外向钾电流(IA)的影响,初步探讨其作用的通道动力学机制。方法:采用全细胞膜片钳技术,观察N/OFQ对急性分离的大鼠顶叶皮层神经元IA的作用。结果:①0.1μmol/L N/OFQ使IA幅值由给药前的(5356.1±361.6)pA下降为(4113.3±312.7)pA,抑制率为23.20%±2.17%(P〈0.01,n=10)。②0.1μmol/L N/OFQ使IA的电流-电压(I-V)曲线降低(P〈0.01,n=10)。③0.1μmol/L N/OFQ使,IA激活曲线的半数激活电压(V1/2)和斜率因子(κ)分别由给药前的(-9.2±2.5)mV和(20.4±2.3)mV变为给药后的(30.6±3.7)mV(P〈0.01,n=8)和(22.6±2.1)mV(P〉0.05,n=8)。④0.1μmol/L N/OFQ使IA失活曲线的半数失活电压(V1/2)和斜率因子(κ)分别由给药前的(-64.1±3.2)mV和(21.5±2.1)mV变为给药后的(-55.9±1.9)mV(P〈0.05,n=5)和(19.6±2.2)mV(P〉0.05,n=5)。结论:N/OFQ可抑制大鼠顶叶皮层神经元IA,使其激活曲线、失活曲线均右移。  相似文献

Four morphologically indistinguishable isolates of monoecious Gelidium vagum Okamura were crossed reciprocally to obtain hybrids for a study on heterosis in this alga. Approximately 50% outcrossing was achieved by adding a small fragment of the designated female parent to a much larger quantity of designated male thallus in the crossing dish. Hybrids in the mixed population of isomorphic hybrid and inbred sporelings were identified by the presence of male-specific random amplified polymorphic DNA markers. Growth performance of hybrid tetrasporophytes was compared to that of their gametophytic parents and with inbred tetrasporophytes at near-optimum and sub-optimum temperature and density. In general, the hybrids showed growth superiority over inbred lines, particularly under sub-optimum conditions. In these experiments, the hybrid plants from cross 129 × 130 exhibited a 9.5–130% higher growth rate as compared to the mid-value of the related inbred tetrasporophytes, strongly suggesting the presence of heterosis.  相似文献

本工作采用了行为和脑内注射相结合的方法研究了大鼠尾壳核的 GABA 能传递在条件性行为调控中的作用。在分辨学习的基础上训练大鼠完成条件性回避任务,以比较药物对分辨学习和条件性回避的不同效应。实验结果表明,于大鼠双侧尾壳核内分别注入 γ-氨基丁酸(GABA)(每侧100μg/μl)和 GABA 受体激动剂蝇蕈醇(Muscimol)(每侧0.1μg/μl)后可暂时抑制条件性回避反应的出现,但分辨学习无明显影响。作为对照,于尾壳核内注入等量的生理盐水则既不影响条件性回避反应,也不影响分辨学习。在条件性回避反应被 Muscimol抑制后于尾壳核内再注入 GABA 受体阻断剂印防己毒素(PTX)(每侧0.1μg/μl)则可拮抗Muscimol 的行为抑制效应,即条件反应的出现率可恢复到或接近注射前水平。实验结果表明,大鼠尾壳核的 GABA 能传递在条件性行为调控中的重要作用。  相似文献

分子生物学技术在微生物生态学研究中具有灵敏、精确和快速的优势,但不能提供微生物的形态学、数量性状、空间分布等信息。荧光原位杂交技术结合了分子生物学的精确性和显微镜的可视性信息,可以在自然生境中监测和鉴定不同的微生物个体,尤其是对难培养和未被培养的微生物进行检测。荧光原位杂交技术被广泛用于微生物群落结构诊断和评价,现已成为微生物分子生态学研究中的热点技术。对荧光原位杂交技术的发展和在微生物分子生态学中的应用进行了综述,探讨了该技术应用中存在的问题和发展前景。  相似文献

Observations of meiotic and/or mitotic chromosomes for 16 specific, infraspecific, and hybrid taxa from 56 populations are presented; chromosome numbers for 11 taxa are reported for the first time. All samples reported have 2n = 22, with the exception that 3 of 12 collections of C. × intermedia have at least some individuals that are triploid. North American diploid collections of this hybrid are heterozygous for one reciprocal translocation; only 1 of 5 European diploid collections also exhibited a translocation. Meiosis in four naturally occurring Asian hybrids is also analyzed: C. × ovata (C. cordata × C. mollis), C. × decipiens (C. erubescens × C. lutetiana) and C.dubia (C. cordata × C. eurbescens) possess a single translocation; C. × mentiens (C. alpina × C. eurbescens) is structurally homozygous. With the exception of the translocation, chromosome pairing in these hybrids is normal. Early diakinesis chromosomes possess densely staining centric regions and diffusely staining arms that are subequal in length.  相似文献

Osmophores of the myophilous genus Restrepia (Orchidaceae) were studied developmentally at the ultrastructural level. They are located at petal apices and on the adaxial surface of the dorsal sepal apex. Up to and through anthesis a dense, osmiophilic exudate is synthesized probably in endoplasmic reticulum or in plastids of papillose epidermal cells, and then appears to be transported through the plasmalemma by granulocrine elimination. As the exudate is amassed, the cuticle ruptures to form numerous pores that extend from the cell wall. Mitochondria and amyloplasts are especially abundant at anthesis. From anthesis to post-anthesis, lipid droplets appear in the cytoplasm and vacuoles of epidermal cells, and frequency of cell organelles drops markedly with increasing vacuolation.  相似文献

Sarcandra is the only genus of Chloranthaceae hitherto thought to be vesselless. Study of liquid-preserved material of S. glabra revealed that in root secondary xylem some tracheary elements are wider in diameter and have markedly scalariform end walls combined with circular pits on lateral walls. Examination of these wider tracheary elements with scanning electron microscope (SEM) demonstrated various degrees of pit membrane absence in the end walls. Commonly a few threadlike fibrils traverse the pits (perforations); these as well as intact nature of pit membranes in pits at ends of some perforation plates are evidence that lack of pit membranes does not result from damage during processing. Some perforations lack any remnants of pit membranes. Although perforation plates and therefore vessels are present in Sarcandra roots, no perforations were observed in tracheary elements of stems or lignotubers. Further, stem tracheids do not have the prominently scalariform end walls that the vessel elements in roots do. Presence of vessels in Sarcandra removes at least one (probably several) hypothetical events of vessel origin that must be postulated to account for known patterns of vessel distribution in angiosperms, assuming that they are primitively vesselless. Seven (perhaps fewer) vessel origin events in angiosperms could account for these patterns; two of those events (Nelumbo and monocotyledons) are different from the others in nature. Widely accepted data on trends of vessel specialization in woody dicotyledons yield an unappreciated implication: vessel specialization has happened in a highly polyphyletic manner in dicotyledons, and therefore multiple vessel origins represent a logical extension backward in time. If a group of vesselless dictyoledons ancestral to other angiosperms existed, they can be hypothesized to have had a relatively homogeneous floral plan now that Sarcandra-like plants no longer need be imagined within that group. Sarcandra and other Chloranthaceae show that the borderline between vessel absence and presence is less sharp than generally appreciated.  相似文献
