在小孢子和雄配子形成过程中,中期Ⅱ、小孢子和生殖细胞有丝分裂中期染色体数目增加和减少细胞的百分率分别与前一时期(中期Ⅰ、中期Ⅱ和小孢子有丝分裂中期)染色体数目增加和减少细胞的百分率相近,而中期Ⅰ染色体数目增加和减少细胞的百分率又分别与凝线期2.3→1和1→2.3型染色质胞间转移的百分率相近,统计学上无显著差异。据此认为:(1)染色体数目的改变与染色质的胞间转移活动有关;(2)中期Ⅰ那些染色体数目增加或减少的细胞基本上都能够进一步发育,直至形成雄配子。  相似文献

采用机械破碎和蔗糖梯度离心方法从洋葱根端分生组织中分离出细胞核并制备出核骨架。细胞核SDS-PAGE谱带中135kD处有一多肽,免疫印迹实验结果表明,该多肽可被抗鸡ScⅡ抗体标记,核骨架中没有此多肽。经抗ScⅡ抗体和FITC偶联的二抗标记后,细胞核发出代表ScⅡ的特异性荧光,而核骨架中无荧光发出。经抗ScⅡ抗体和蛋白A胶体金处理后,金颗粒特异性地结合在核内染色质区域。说明ScⅡ类似蛋白是洋葱根端细胞核的组分,且位于核内染色质上,但该蛋白不是核骨架成分。免疫荧光和免疫电镜实验结果还说明ScⅡ类似蛋白是洋葱根端细胞染色体和染色体骨架的组成成分。  相似文献

Somatic embryos were obtained and plants regenerated from immature embryos of onion following culture on embryogenic induction media. Highest rates of somatic embrogenesis resulted from 0.5- to 1.5-mm immature embryos cultured on media containing 5 mg/l of picloram. Somatic embryos formed either directly on the surface of embryos or developed from compact cultures. The production of somatic embryos was significantly affected by the addition of auxin, embryo size and cultivar. The potential of somatic embryogenic cultures for plantlet regeneration has been maintained for over 1 year in some lines. Three types of immature-embryo-derived cultures were characterized by histology. Some cultures were morphologically similar to immature-embryo-derived embryogenic cultures of other monocotyledonous species. Summary. Caged probes offer a novel approach to study plant cell-to-cell communication. Instead of introducing fluorescent molecules into cells by microinjection, their caged counterparts can be preloaded into the tissue by diffusion. Following spatially controlled photoactivation, movement of the uncaged fluorochrome can be followed in time and direction by confocal laser scanning microscopy. In the onion bulb scale epidermis used as a model system, symplasmic transport of the tracer out of a target cell was followed. Transport via the symplasmic pathway was challenged by plasmolysing the tissue. 应用间接免疫荧光标记技术和激光共聚焦扫描显微镜成像技术观察洋葱小孢子母细胞减数分裂过程中微管分布变化。减数分裂之前,小孢子母细胞中的微管较短,呈辐射状,由细胞核表面向四周扩散。减数分裂开始后,细胞质中的一部分微管蛋白聚集成纺锤体微管,控制染色体的分布。进入减数分裂I后期,纺锤体微管变为牵引染色体移向两极的着丝粒微管和连接纺锤体两极的极丝微管。之后,所有微管集中在两个核之间,构成成膜体。然后,微管解聚成微管蛋白弥散在细胞质中。减数分裂I完成后,二分体2个子细胞中的微管蛋白又聚集成2个纺锤体微管,开始减数分裂II过程。经过减数分裂II中期,2个二分体细胞中的微管再次集中在2个细胞核之间形成成膜体,隔离2个细胞核。此后,微管蛋白解聚,弥散分布在小孢子细胞质中。  相似文献

采用焦锑酸钾沉淀钙离子技术,对洋葱(Alliumcepa)花药发育中Ca^2+分布进行了研究。在小孢子母细胞时期,小孢子母细胞中的钙沉淀颗粒很少,但绒毡层细胞的内切向壁已出现明显的钙沉淀颗粒。在四分体时期,四分体小孢子的胼胝质壁中出现较多的钙沉淀颗粒;绒毡层细胞内切向壁的钙沉淀颗粒消失,而在外切向壁和径向壁部位的钙沉淀颗粒增加。在小孢子早期,小孢子中也出现了钙沉淀颗粒,而绒毡层细胞内切向壁表面出现了很多絮状物,其上附有细小钙沉淀颗粒。到小孢子晚期,小孢子中出现一些小液泡,细胞质中的钙沉淀颗粒有所下降。此时绒毡层细胞已明显退化,但在绒毡层膜上仍有一些乌氏体和钙沉淀颗粒。在二胞花粉早期,营养细胞中的液泡收缩、消失,细胞质中又出现了较多的钙沉淀颗粒,在质体和其内部的淀粉粒表面上附有较多的钙沉淀颗粒。到二胞花粉晚期,花粉中的钙沉淀颗粒已明显下降,仅在花粉外壁中还有一地钙沉淀颗粒.  相似文献

将洋葱的胚珠置于酶液中酶解50-110 min后剥去其珠被,可清楚地看到珠心中的胚囊轮廓。用解剖针将珠心从中部横切,然后挤压其珠孔部位,卵器细胞从胚珠的切口处逸出。再用显微操作仪的玻璃针将卵细胞和两个助细胞分开,达到分离洋葱卵细胞的目的。酶对分离卵细胞具有重要作用,在最佳的酶液浓度[0.02%果胶酶Y23、0.08%果胶酶(Serva)、0.05%纤维素酶和0.05%半纤维素酶]下酶解胚珠110 min后,解剖1 h可从24个胚珠中分离出10个卵细胞(41.67%)。随着胚囊的发育,两个助细胞的体积出现明显的二形性。洋葱生活卵细胞的分离为开展洋葱离体受精建立了基础,也为研究洋葱卵器细胞的发育创造了条件。  相似文献

The ultrastructures of male meiocytes in tobacco, onion and lily were studied to elucidate the interaction between cytoplasmic channels (CCs) and plastids. Before meiosis, the male sporogenous cells had identically thickened cell walls (CWs) traversed by typical plasmodesmata (PDs). After entering meiosis, their CWs became uneven in thickness and 80-500nm aperture CCs were formed. Simultaneously, plastids or plastid-like bodies (PLBs) differing in size and morphology assembled at one or both ends of the CCs. These plastids and PLBs commonly orientated their sharper ends to face the CCs and were co-orientated on the axial line crossing the CC. Such pairs of plastids were often interconnected through the CC by thin (50-100nm) threads emanating from their membranes. Sometimes, plastids or PLBs extended directly from one side of a CW to the other, forming a bridge via the CC. In some cases, several plastids formed bridges between cells via one common CC. This is the first report that clearly demonstrates an intercellular continuum of, or communication between, plastids in male plant meiocytes.  相似文献

In Vitro Regeneration of Onion through Repetitive Somatic Embryogenesis   总被引:1,自引:0,他引:1  
A reliable protocol for the regeneration of onion through repetitive somatic embryogenesis was established. Embryogenic callus was derived from mature seeds on Murashige and Skoog (MS) medium supplemented with 2 mg dm-3 2,4-dichlorophenoxyacetic acid (2,4-D). Somatic embryos aroused on the surface of calli cultures and formed plantlets after the removal of 2,4-D or its substitution with 1 mg dm-3 kinetin (Kin). Reculturing the somatic embryos on 2,4-D containing medium led to secondary embryos formation. The embryogenic cultures which were preserved for five months on maintenance medium containing 2 mg dm-3 2,4-D + 0.5 mg dm-3 Kin have retained their ability for regeneration, while those kept on 2,4-D only, failed to form plantlets. Electrophoretic analysis of total soluble proteins revealed that the competence for successful conversion of somatic embryos into plantlets is associated with the expression of new set of proteins (112, 58 and 30 kD). The regenerated plants were successfully transferred to the soil.  相似文献

Iris yellow spot virus (IYSV) was first observed in sweet onions in Georgia (USA) in 2003 in the Vidalia region. The virus had been reported in the onion‐growing regions in western USA several years before being detected in Georgia in the east. Although symptoms were observed on onions in Peru several years earlier, the presence of IYSV was not confirmed in Peru until after the virus was detected in Georgia. We characterized nine isolates of IYSV recovered from sweet onions in both Georgia (four isolates) and Peru (five isolates) by sequencing the nucleocapsid (N) gene and compared those sequences with sequences available in GenBank. Sequence divergence between IYSV isolates from Georgia and Peru was low with 1.1%, and comparisons with IYSV isolates from other regions showed divergence of up to 11.4%. Bootstrap analysis indicated with a high degree of confidence that the Georgia and Peruvian isolates fell into the same clade and were different from known isolates from western USA that fell into sister clades. The high degree of similarity between Georgia and Peruvian isolates suggests that gene flow occurred from Peru into Georgia.  相似文献

The presence of actin in eukaryotic nuclei and chromosomes,and especially in higher plant nuclei and chromosomes,has not been well established.We detected actin in meristematic cells of Allium cepa with indirect immunofluorescence technique and observed bright fluorescence in the intact nuclei and chromosomes,indicating that actin is present in the nuclei and chromosomes of the higher plant.We labeld sections of the meristematic cells of A.cepa with immunogold technique,gold parti cles were concentrated in condensed chromatin and nucleoli,confirming the results of the immunofluoresence observations.We traeated the nuclei and chromosomes of A.cepa with DNase I and 2M NaCl and obtained DNA-and histone-depleted nuclei and chromosomes.Indirect immunofluorescence tests showed that the DNA-and histonedepleted nuclei and chromosomes reacted positively with the anti-actin antibodies.These results demonstrate that the anti-actin antibodies.These results demonstrate that actin exists not only in intact nuclei and chromosomes,but also in DNA-and histone-depleted nuclei and chrmosomes of the plant.In addition,our immuno-fluorescence tests indicate that tropomyosin is present in the nuclei and chromosomes of A.cepa.  相似文献

Summary Microtubules tightly cross-linked into bundles are described in the sperm cells ofBrassica oleracea pollen tubes. The sperm cells are lobed and tailed and the microtubule bundles are often located in these parts of the cells. 以杉木(Cunninghamialanceolata(Lamb.)Hook.)传粉后的胚珠为材料,分离获得了生活的雌配子和雄配子。当花粉管进入雌配子体组织后,在颈卵器上方形成膨大的精原细胞或成对的精细胞。解剖出的精细胞呈FDA正反应,荧光增白剂染色表明精细胞具有细胞壁。雌配子体在酶液作用下可同时释放出多个卵细胞和精细胞。游离出的卵细胞呈FDA正反应,近圆形,内含1~2个大液泡。用PEG方法诱导卵细胞与卵细胞,卵细胞与精细胞的融合,只获得了相互粘连的两个原生质体。  相似文献

Viable male and female gametes were isolated from pollinated ovules of Cunninghamia lanceolata (Lamb.) Hook. Prior to their penetration into the female gametophyte, the pollen tubes were drawn out from the nucetlus. The isolated pollen tubes were branched and one of them became swollen. An enlarged spermatogenous cell and subsquently a pair of sperm cells were formed as the pollen tube reached the regions over and against the archegonia. The sperm cells were released from the pollen tubes manually with the use of a stereomicroscope. The positive FDA reaction gave evidence of the sperm cells viability, and the Fluorescent Brightener 28 positive demonstrated the presence of cell wall. The egg cells were enzymatically isolated from the female gametophytes. The isolated egg cells were spherical, contained 1 to 2 large and many small vacuoles. FDA test showed the egg cells were viable, and the viability sustained for 8 days in 2 to 4 ℃ without any protectants. Fusion between single pair of male and female gametic protoplasts was attempted with PEG method, but only adhesion of the two was obtained.  相似文献

Transgenic onion plants (Allium cepa) tolerant to herbicides containing active ingredients glyphosate and phosphinothricin were recovered from immature embryos of open pollinated and hybrid parent onion lines at a maximum transformation frequency of 0.9%. Transformants of different onion cultivars, grown on different selective agents and confirmed by Southern analysis, thrived with no apparent ill effects when sprayed with the respective herbicides at double the recommended field dosage for weed eradication. This study demonstrates that the transformation process described previously can be used with different selective agents and is cultivar independent.  相似文献

The efficacy of antibiotics, kanamycin, hygromycin, and geneticin, and the herbicide phosphinothricin as selective agents in onion (Allium cepa L.) tissue culture was investigated. Immature embryos and immature-embryo-derived cultures were grown on media containing a range of concentrations of the four selective agents. Kanamycin was shown to be unsuitable while phosphinothricin, geneticin or hygromycin significantly reduced further growth of the onion cultures. The level of casein affected growth on medium containing phosphinothricin. The effectiveness of phosphinothricin was reduced when high levels (200 mg/l) of casein were present in the medium. Omission of casein reduced the shoot regeneration frequency from immature embryo cultures. Low levels of hygromycin (10–30 mg/l) reduced callus growth but significantly increased the regeneration frequency from immature-embryo-derived cultures.
