Abstract: There is growing evidence that cyclic GMP (cGMP) plays important roles in the brain. In cultured rat astrocytes, we observed that the cGMP-inducing C-type natriuretic peptide (CNP) and cGMP analogues caused a decrease in intracellular pH (pHi). To examine whether this effect was due to inhibition of an Na+/H+ exchanger (NHE), we acidified cells by replacing extracellular Na+ by choline and examined the kinetics of the pHi recovery that occurred on reintroduction of Na+ in the extracellular medium. Both CNP and amiloride analogues inhibited the Na+-dependent pHi recovery, even in the nominal absence of CO2/HCO3?. This indicated that CNP inhibited the activity of an exchanger that was Na+-dependent, HCO3?-independent, and sensitive to known inhibitors of NHE. However, comparison of the potencies of four distinct amiloride analogues revealed a pharmacological profile that was different from that of any other NHE characterized to date. cGMP mimicked the effect of CNP on sodium-dependent pHi recovery, but the native nucleotide was as potent as membrane-permeant analogues. Intracellularly produced cGMP was very rapidly exported out of astrocytes. Probenecid and niflumic acid slowed down the rate of cGMP egression and inhibited the effect of CNP on Na+-dependent recovery, but not that of extracellular cGMP. Altogether, our data indicate that cGMP inhibits a novel type of NHE in astrocytes via an extracellular site of action. If these results with primary cultures transfer to brain, this phenomenon may constitute a mechanism by which natriuretic peptides exert some of their actions in the brain, as pHi transients have been shown to modulate several important astrocytic functions.  相似文献

The contribution of metabolic bicarbonate to cytosolic pH (pHcyto) regulation was studied on isolated perfused rat liver using phosphorus-31 NMR spectroscopy. Removal of external HCO?3 decreased proton efflux from 18.6±5.0 to 1.64±0.29 μmol/min per g liver wet weight (w.w.) and pHcyto from 7.17±0.06 to 6.87±0.06. In the nominal absence of bicarbonate, inhibition of carbonic anhydrase by acetazolamide induced a further decrease of proton efflux of 0.69±0.26 μmol/min per g liver w.w. reflecting a reduction in metabolic CO2 hydration, and hence a decrease of H+ and HCO?3 supplies. Even though 27% of the proton efflux was amiloride-sensitive under bicarbonate-free conditions, amiloride did not change pHcyto, revealing the contribution of additional regulatory processes. Indeed, pH regulation was affected by the combined use of 4-acetamido-4′-isothiocyanostilbene-2,2′-disulfonic acid (SITS) and amiloride since pHcyto decreased by 0.16±0.05 and proton efflux by 0.60±0.14 μmol/min per g liver w.w. The data suggest that amiloride-sensitive or SITS-sensitive transport activities could achieve, by themselves, pHcyto regulation. The involvement of two mechanisms, most likely Na+/H+ antiport and Na+:HCO?3 symport, was confirmed in the whole organ under intracellular and extracellular acidosis. The evidence of Na-dependent transport of HCO?3 in the absence of exogenous bicarbonate implies that the amount of metabolic bicarbonate is sufficient to effectively participate to pHcyto regulation.  相似文献

The kinetics of the light-driven Cl? uptake pump of Synechococcus R-2 (PCC 7942) were investigated. The kinetics of Cl? uptake were measured in BG-11 medium (pHo, 7·5; [K+]o, 0·35 mol m?3; [Na+]o, 18 mol m?3; [Cl?]o, 0·508 mol m?3) or modified media based on the above. Net36Cl? fluxes (?Cl?o,i) followed Michaelis-Menten kinetics and were stimulated by Na+ [18 mol m?3 Na+ BG-11 ?Cl?max= 3·29±0·60 (49) nmol m?2 s?1 versus Na+-free BG-11 ?Cl?max= 1·02±0·13 (54) nmol m?2 s?1] but the Km was not significantly different in the presence or absence of Na+ at pHo 10; the Km was lower, but not affected by the presence or absence of Na+ [Km = 22·3±3·54 (20) mmol m?3]. Na+ is a non-competitive activator of net ?Cl?o,i. High [K+]o (18 mol m?3) did not stimulate net ?Cl?o,i or change the Km in Na+-free medium. High [K+]o (18 mol m?3) added to Na+ BG-11 medium decreased net ?Cl?o,i [18 mol m?3K+ BG-11; ?Cl?max= 2·50±0·32 (20) nmol m?2 s?1 versus BG-11 medium; ?Cl?max= 3·35±0·56 (20) nmol m?2 s?1] but did not affect the Km 55·8±8·100 (40) mmol m?3]. Na+-stimulation of net ?Cl?o,i followed Michaelis-Menten kinetics up to 2–5 mol m?3 [Na+]o but higher concentrations were inhibitory. The Km for Na+-stimulation of net ?Cl?o,i [K1/2(Na+)] was different at 47 mmol m?3 [Cl?]o (K1/2[Na+] = 123±27 (37) mmol m?3]. Li+ was only about one-third as effective as Na+ in stimulating Cl? uptake but the activation constant was similar [K1/2(Li+) = 88±46 (16) mmol m?3]. Br? was a competitive inhibitor of Cl? uptake. The inhibition constant (Ki) was not significantly different in the presence and absence of Na+. The overall Ki was 297±23 (45) mmol m?3. The discrimination ratio of Cl? over Br? (δCl?/δBr?) was 6·38±0·92 (df = 147). Synechococcus has a single Na+-stimulated Cl? pump because the Km of the Cl? transporter and its discrimination between Cl? and Br? are not significantly different in the presence and absence of Na+. (1) The energy state and free intracellular calcium concentration ([Cai) of super-fused cortical slices were measured in moderate hypoxia (~65 μM O2), in mild hypoglycaemia (0.5 mM glucose), and in combinations of the two insults using 19F and 31P NMR spectroscopy. (2) Neither hypoxia nor hypoglycaemia alone caused any significant change in [Cai. Hypoxia caused a 40% fall in phosphocreatine (PCr) content but not in ATP level, and hypoglycaemia produced a slight fall in both (as expected from previous studies). These changes in the energy state recovered on return to control conditions. (3) A combined sequential insult (hypoxia, followed by hypoxia plus hypoglycaemia) produced a 100% increase in [Ca, and a decrease in PCr level to ~25% of control. The reverse combined sequential insult (hypoglycaemia, followed by hypoglycaemia plus hypoxia) had the same effect. On return to control conditions there was some decrease in [Cai and a small increase in PCr content, but neither recovered to control levels. (4) Exposure of the tissue to the combined simultaneous insult (hypoxia plus hypoglycaemia) immediately after the control spectra had been recorded resulted in a fivefold increase in [Cai and a similar decrease in PCr level to 20–25% of control. There was little if any change of [Cai or PCr level on return to control conditions. (5) These results are discussed in terms of metabolic adaptation of some but not all of the cortical cells to the single type of insult, which renders the tissues less vulnerable to the combined insult.  相似文献

In the preceding paper (Bevensee, M.O., R.A. Weed, and W.F. Boron. 1997. J. Gen. Physiol. 110: 453–465.), we showed that a Na+-driven influx of HCO3 causes the increase in intracellular pH (pHi) observed when astrocytes cultured from rat hippocampus are exposed to 5% CO2/17 mM HCO3 . In the present study, we used the pH-sensitive fluorescent indicator 2′,7′-biscarboxyethyl-5,6-carboxyfluorescein (BCECF) and the perforated patch-clamp technique to determine whether this transporter is a Na+-driven Cl-HCO3 exchanger, an electrogenic Na/HCO3 cotransporter, or an electroneutral Na/HCO3 cotransporter. To determine if the transporter is a Na+-driven Cl-HCO3 exchanger, we depleted the cells of intracellular Cl by incubating them in a Cl-free solution for an average of ∼11 min. We verified the depletion with the Cl-sensitive dye N-(6-methoxyquinolyl)acetoethyl ester (MQAE). In Cl-depleted cells, the pHi still increases after one or more exposures to CO2/HCO3 . Furthermore, the pHi decrease elicited by external Na+ removal does not require external Cl. Therefore, the transporter cannot be a Na+-driven Cl-HCO3 exchanger. To determine if the transporter is an electrogenic Na/ HCO3 cotransporter, we measured pHi and plasma membrane voltage (Vm) while removing external Na+, in the presence/absence of CO2/HCO3 and in the presence/absence of 400 μM 4,4′-diisothiocyanatostilbene-2,2′-disulphonic acid (DIDS). The CO2/HCO3 solutions contained 20% CO2 and 68 mM HCO3 , pH 7.3, to maximize the HCO3 flux. In pHi experiments, removing external Na+ in the presence of CO2/HCO3 elicited an equivalent HCO3 efflux of 281 μM s−1. The HCO3 influx elicited by returning external Na+ was inhibited 63% by DIDS, so that the predicted DIDS-sensitive Vm change was 3.3 mV. Indeed, we found that removing external Na+ elicited a DIDS-sensitive depolarization that was 2.6 mV larger in the presence than in the absence of CO2/ HCO3 . Thus, the Na/HCO3 cotransporter is electrogenic. Because a cotransporter with a Na+:HCO3 stoichiometry of 1:3 or higher would predict a net HCO3 efflux, rather than the required influx, we conclude that rat hippocampal astrocytes have an electrogenic Na/HCO3 cotransporter with a stoichiometry of 1:2.  相似文献

The role of cAMP in regulation of intracellular pH in the confluent LLC-PK1 cells was investigated. DibutyrylcAMP and forskolin induce intracellular acidification. This acidification is inhibited by DIDS and ethacrynic acid, inhibitors of Na+-independent Cl?/HCO3? exchange, and by removal of extracellular Cl?. In addition, Bt2 cAMP causes Cl? entry into LLC-PK1 cells. These results suggest that cAMP activates Cl? transport, namely Na+-independent Cl?/HCO3? exchange, which participates in pHi regulation.  相似文献

Pathways for HCO3 transport across the basolateral membrane were investigated using membrane vesicles isolated from rat renal cortex. The presence of Cl---HCO3 exchange was assessed directly by 36Cl tracer flux measurements and indirectly by determinants of acridine orange absorbance changes. Under 10% CO2/90% N2 the imposition of an outwardly directed HCO3 concentration gradient (pHo 6/pHi 7.5) stimulated Cl uptake compared to Cl uptake under 100% N2 in the presence of a pH gradient alone. Mediated exchange of Cl for HCO3 was suggested by the HCO3 gradient-induced concentrative accumulation of intravesicular Cl. Maneuvers designed to offset the development of ion-gradient-induced diffusion potentials had no significant effect on the magnitude of HCO3 gradient-driven Cl uptake further suggesting chemical as opposed to electrical Cl−HCO3 exchange coupling. Although basolateral membrane vesicle Cl uptake was observed to be voltage sensitive, the DIDS insensitivity of the Cl conductive pathway served to distinguish this mode of Cl translocation from HCO3 gradient-driven Cl uptake. No evidence for cotransport was obtained. As determined by acridine orange absorbance measurements in the presence of an imposed pH gradient (pHo 7.5/pHi 6), a HCO3 dependent increase in the rate of intravesicular alkalinization was observed in response to an outwardly directed Cl concentration gradient. The basolateral membrane vesicle origin of the observed Cl−HCO3 exchange activity was verified by experiments performed with purified brush-border membrane vesicles. In contrast to our previous observations of the effect of Cl on HCO3 gradient-driven Na+ uptake suggesting a basolateral membrane Na+−HCO3 for Cl exchange mechanism, no effect of Na+ on Cl−HCO3 exchange was observed in the present study.  相似文献

Synechococcus R-2 (PCC 7942) actively accumulated Cl? in the light and dark, under control conditions (BG-11 media: pHo, 7·5; [Na+]o, 18 mol m?3; [Cl?]o, 0·508 molm?3). In BG-11 medium [Cl?], was 17·2±0·848 mol m?3 (light), electrochemical potential of Cl? (ΔμCl?i,o) =+211±2mV; [Cl?]i= 1·24±0·11 mol m?3(dark), ΔμCl?i,o=+133±4mV. Cl? fluxes, but not permeabilities, were much higher in the light: ?Cl?i,o= 4·01±5·4 nmol m?2 s?1, PCl?i,o= 47±5pm s?1 (light); ?Cl?i,o= 0·395±0·071 nmol m?2 s?1, PCl?i,o= 69±14 pm s?1 (dark). Chloride fluxes are inhibited by acid pHo (pHo 5; ?Cl?i,o= 0·14±0·04 nmol m?2 s?1); optimal at pHo 7·5 and not strongly inhibited by alkaline pHo (pHo 10; ?Cl?1i,o= 1·7±0·14 nmol m?2 s?1). A Cl?in/2H+in coporter could not account for the accumulation of Cl? alkaline pHo. Permeability of Cl? is very low, below 100pm s?1 under all conditions used, and appears to be maximal at pHo 7·5 (50–70 pm s?1) and minimal in acid pHo (20pm s?1). DCCD (dicyclohexyl-carbodiimide) inhibited ?Cl?i,o in the light about 75% and [Cl?]i fell to 2·2±0·26 (4) mol m?3. Valinomycin had no effect but monensin severely inhibited Cl? uptake ([Cl?]i= 1·02±0·32 mol m?3; ?Cl?i,o= 0·20±0·1 nmol m?2 s?1). Vanadate (200 mmol m?3) accelerated the Cl? flux (?Cl?i,o= 5·28±0·64 nmol m?2 s?1) but slightly decreased accumulation of Cl? ([Cl?], = 13·9±1·3 mol m?3) in BG-11 medium but had no significant effect in Na+-free media. DCMU (dichlorophenyldimethylurea) did not reduce [Cl?], or ?Cl?i,o to that found in the dark ([Cl?]i= 8·41±0·76 mol m?3; ?Cl?i,o= 2·06±0·36 nmol m?2 s?1). Synechococcus also actively accumulated Cl? in Na+-free media, [Cl?]i was lower but ΔΨi,o hyperpolarized in Na+-free media and so the ΔμCl?i,o was little changed ([Cl?]i= 7·98±0·698 mol m?3; ΔμCl?i,o=+203±3 mV). Net Cl? uptake was stimulated by Na+; Li+ acted as a partial analogue for Na+. Synechococcus has a Na+ activated Cl? transporter which is probably a primary 2Cl?/ATP pump. The Cl? pump is voltage sensitive. ΔμCl?i,o is directly proportional to ΔΨi,o(P»0·01%): ΔμCl?i,o= -1·487 (±0·102) ×ΔΨi,o, r= -0·983, n= 31. The ΔμCl?i,o increased (more positive) as the Δμi,o became more negative. The ΔμCl?i,o has no known function, but might provide a driving force for the uptake of micronutrients.  相似文献

Cell pH regulation was investigated in the T84 cell line derived from epithelial colon cancer. Cell pH was measured by ratiometric fluorescence microscopy using the fluorescent probe BCECF. Basal pH was 7.17 ± 0.023 (n= 48) in HEPES Ringer. After acidification by an ammonium pulse, cell pH recovered toward normal at a rate of 0.13 ± 0.011 pH units/min in the presence of Na+, but in the absence of this ion or after treatment with 0.1 mm hexamethylene amiloride (HMA) no significant recovery was observed, indicating absence of Na+ independent H+ transport mechanisms in HEPES Ringer. In CO2/HCO 3 Ringer, basal cell pH was 7.21 ± 0.020 (n= 35). Changing to HEPES Ringer, a marked alkalinization was observed due to loss of CO2, followed by return to the initial pH at a rate of −0.14 ± 0.012 (n= 8) pH/min; this return was retarded or abolished in the absence of Cl or after addition of 0.2 mm DIDS, suggesting extrusion of bicarbonate by Cl/HCO 3 exchange. This exchange was not Na+ dependent. When Na+ was added to cells incubated in 0 Na+ Ringer while blocking Na+/H+ exchange by HMA, cell alkalinization by 0.19 ± 0.04 (n= 11) pH units was observed, suggesting the presence of Na+/HCO 3 cotransport carrying HCO 3 into these cells, which was abolished by DIDS. These experiments, thus, show that Na+/H+ and Cl/HCO 3 exchange and Na+/HCO 3 cotransport participate in cell pH regulation in T84 cells. Received: 3 April 2000/Revised: 22 June 2000  相似文献

Parathyroid hormone (PTH) has previously been shown to enhance the transepithelial secretion of Cl? and HCO3? across the intestinal epithelia including Caco-2 monolayer, but the underlying cellular mechanisms are not completely understood. Herein, we identified the major signaling pathways that possibly mediated the PTH action to its known target anion channel, i.e., cystic fibrosis transmembrane conductance regulator anion channel (CFTR). Specifically, PTH was able to induce phosphorylation of protein kinase A and phosphoinositide 3-kinase. Since the apical HCO3? efflux through CFTR often required the intracellular H+/HCO3? production and/or the Na+-dependent basolateral HCO3? uptake, the intracellular pH (pHi) balance might be disturbed, especially as a consequence of increased endogenous H+ and HCO3? production. However, measurement of pHi by a pH-sensitive dye suggested that the PTH-exposed Caco-2 cells were able to maintain normal pH despite robust HCO3? transport. In addition, although the plasma membrane Na+/K+-ATPase (NKA) is normally essential for basolateral HCO3? uptake and other transporters (e.g., NHE1), PTH did not induce insertion of new NKA molecules into the basolateral membrane as determined by membrane protein biotinylation technique. Thus, together with our previous data, we concluded that the PTH action on Caco-2 cells is dependent on PKA and PI3K with no detectable change in pHi or NKA abundance on cell membrane.  相似文献

We reported previously that poliovirus infection induces alkalinization in HeLa cells and that an alkaline intracellular pH (pHi) promoted viral replication. Additional experiments were carried out to understand the underlying mechanism. Virus-infected or control monolayer cultures were incubated with nominally bicarbonate-free Eagle's minimal essential medium (MEM) buffered with N-2-hydroxyethylpiperazine-N-3-ethanesulfonic acid (HEPES), and immediately following preincubations, changes in pHi were monitored via benzoic acid uptake around 2 h postinfection. The absence of pH increase in cells infected with ultraviolet light-inactivated virus (UV-virus) indicated that viral gene expression was required for this effect. On the other hand, lack of effect of 3 mM guanidine, an inhibitor of poliovirus-specific RNA but not protein synthesis, suggested that translation of input viral genome RNA is sufficient for the pH increase. Activation of Na+/H+ exchange, Cl?HCO?3 exchange, or H+-ATPase was considered as possible mechanisms by which alkalinization occurs in virus-infected cells. Na+/H+ exchange was excluded because the pH effect occurred in a Na+/H+ exchange deficient HeLa cell mutant. Similarly, Cl?/HCO?3 exchange was excluded because virus-specific alkalinization was evident in the presence of Cl? or bicarbonate deficient medium and was not associated with an increase in HCO?3 uptake or a decrease in Cl? uptake. Lack of dependence on Na+, abrogation by 10 μM 7-chloro-4-nitrobenz-2-oxa-1,3-diazole (NBD-Cl), and resistance to 1 mM vandate suggested that this effect was due to the activation of a vacuolar-type (V) proton ATPase. Studies using protein kinase inhibitors indicated that activation of the ATPase in virus-infected cells probably involved protein kinase C-mediated phosphorylation. © 1993 Wiley-Liss, Inc.  相似文献

pH i recovery in acid-loaded Ehrlich ascites tumor cells and pH i maintenance at steady-state were studied using the fluorescent probe BCECF.Both in nominally HCO 3 -free media and at 25 mm HCO 3 , the measured pH i (7.26 and 7.82, respectively) was significantly more alkaline than the pH i . value calculated assuming the transmembrane HCO 3 gradient to be equal to the Cl gradient. Thus, pH i in these cells is not determined by the Cl gradient and by Cl/HCO 3 exchange.pH i recovery following acid loading by propionate exposure, NH 4 + withdrawal, or CO2 exposure is mediated by amiloride-sensitive Na+/H+ exchange in HCO3 free media, and in the presence of HCO 3 (25 mm) by DIDS-sensitive, Na+-dependent Cl/HCO 3 exchange. A significant residual pH i recovery in the presence of both amiloride and DIDS suggests an additional role for a primary active H+ pump in pH i regulation. pH i maintenance at steady-state involves both Na+/H+ exchange and Na+-dependent Cl/HCO 3 exchange.Acute removal of external Cl induces a DIDS-sensitive, Na+-dependent alkalinization, taken to represent HCO 3 influx in exchange for cellular Cl. Measurements of 36Cl efflux into Cl-free gluconate media with and without Na+ and/or HCO 3 (10 mm) directly demonstrate a DIDS-sensitive, Na+ dependent Cl/HCO 3 exchange operating at slightly acidic pH i (pHo 6.8), and a DIDS-sensitive, Na+-independent Cl/HCO 3 exchange operating at alkaline pH i (pH o 8.2).The excellent technical assistance of Marianne Schiødt and Birgit B. Epileptic foci are associated with locally reduced taurine (2-aminoethanesulfonic acid) concentration and Na+, K+-ATPase (EC 3.6.1.3) specific activity. Topically applied and intraperitoneally administered taurine can prevent the development and/or spread of foci in many animal models. Taurine has been implicated as a possible cytosolic modulator of monovalent ion distribution, cytosolic “free” calcium activity, and neuronal excitability. Taurine may act in part by modulating Na+, K+-ATPase activity of neuronal and glial cells. We characterized the requirements for in vitro modulation of Na+, K+-ATPase by taurine. Normal whole brain homogenate Na+, K+-ATPase activity is 5.1 ± 0.4 (4) μmol Pi± h?1± mg?1 Lowry protein. Partial purification of the plasma membrane fraction to remove cytosolic proteins and extrinsic proteins and to uncouple cholinergic receptors yields a membrane-bound Na+, K+-ATPase activity of 204.6 ± 5.8 (4) mol Pi± h?1± mg?1 Lowry protein. Taurine activates the Na+, K+-ATPase at all levels of purification. The concentration dependence of activation follows normal saturation kinetics (K1/2= 39 mM taurine, activation maximum =+87%). The activation exhibits chemical specificity among the taurine analogues and metabolites: taurine = isethionic acid > hypotaurine > no activation =β-alanine = methionine = choline = leucine. Taurine can act as an endogenous activator/modulator of Na+, K+-ATPase. Its action is mediated by a membrane-bound protein.  相似文献

Internal pH (pH i ) is in Vero cells regulated mainly by three antiports. Na+/H+ antiport and Na+-dependent Cl+/HCO 3 + antiport increase pH i in acidified cells, and Na+-independent Cl+/HCO 3 + antiport lowers pH i in cells after alkalinization. The activities of the antiporters were altered in cells after exposure to 41–45°C. Under such conditions the Na+/H+ antiport and the Na+-dependent Cl+/HCO 3 + antiport were both stimulated, whereas the Na+-independent Cl+/HCO 3 + antiport was inhibited in such a way that a higher pH value was required to activate it. This alteration was also induced by some other forms of cellular stress, but did most likely not involve stress proteins as protein synthesis was not required. Abstract: The role of voltage-sensitive Ca2+ channels in mediating Ca2+ influx during ischemia was investigated in NG108-15 cells, a neuronal cell line that does not express glutamate-sensitive receptor-mediated Ca2+ channels. Concurrent 31P/19F and 23Na double-quantum filtered (DQF) NMR spectra were used to monitor cellular energy status, intracellular [Ca2+] ([Ca2+]i), and intracellular Na+ content in cells loaded with the calcium indicator 1,2-bis-(2-amino-5-fluorophenoxy)ethane-N,N,N′,N′-tetraacetic acid (5FBAPTA) during ischemia and reperfusion. Cells loaded with 5FBAPTA were indistinguishable from unloaded cells except for small immediate decreases in levels of phosphocreatine (PCr) and ATP. Ischemia induced a steady decrease in intracellular pH and PCr and ATP levels, and a steady increase in intracellular Na+ content; however, a substantial increase in [Ca2+]i (about threefold) was seen only following marked impairment of cellular energy status, when PCr was undetectable and ATP content was reduced to 55% of control levels. A depolarization-induced increase in [Ca2+]i could be completely blocked by 1 µM nifedipine, whereas up to 20 µM nifedipine had no effect on the increase in [Ca2+]i seen during ischemia. These data demonstrate that voltage-gated Ca2+ channels do not mediate significant Ca2+ flux during ischemia in this cell line and suggest an important role for Ca2+i stores, the Na+/Ca2+ antiporter, or other processes linked to cellular energy status in the increase in cytosolic Ca2+ level during ischemia.  相似文献

The leakage of various inorganic carbon species from air-grown cells of Synechococcus UTEX 625 was investigated after a light to dark transition or during a light period using a mass spectrometer under a wide variety of experimental conditions. Total inorganic carbon efflux and CO2 efflux during the initial period of darkness were measured with or without carbonic anhydrase in the reaction medium respectively. The HCO3? efflux after a light to dark transition was estimated by difference. Carbon dioxide efflux in the light was measured by inhibiting CO2 transport with either Na2S or COS3 or quenching the 13C inorganic carbon transport by the addition of 12C inorganic carbon in excess. In cells in which CO2 fixation was inhibited, when only the HCO3? transport system was fully operative, CO2 effluxed continuously during the light period at a rate equal to about 25% of that in darkness. When only the CO2 transport system was operative, HCO3? effluxed during the light period. The difference between the light and dark efflux rates was consistent with a 0.6 unit decrease in the intracellular pH upon darkening the cells. The permeabilities of the cell for CO2 (2.94 ± 0.14 ± 10?8ms?1; mean ± SE, n=137) and HCO3? (1.4–1.7 ± 10?9 ms?1) were calculated.  相似文献

The formation of competent spermatozoa is a complex event that depends on the establishment of adequate environments throughout the male reproductive tract. This includes the control of bicarbonate (HCO3 ?) concentration, which plays an essential role in the maintenance of extracellular and intracellular pH (pHi) values. Diabetes mellitus alters pHi regulation in mammalian cells, mainly by altering the activity of ion transporters, particularly HCO3 ?-dependent mechanisms. Yet, little is known about the effects of this pathology and its prodromal stage, prediabetes, on the membrane transport mechanisms of male reproductive tract cells. Herein, we analyzed protein and mRNA levels of the most relevant HCO3 ? transporters of the SLC4 family [anion exchanger 2 (AE2), Na+-driven Cl?/HCO3 ? exchanger (NDCBE), electrogenic Na+/HCO3 ? cotransporter 1 (NBCe1), electroneutral Na+/HCO3 ? cotransporter 1 (NBCn1)] in the testis and epididymis of a prediabetic animal model. Firstly, we identified the HCO3 ? transporters of the SLC4 family, in both testicular and epididymal tissue. Secondly, although no alterations were detected in protein expression, mRNA levels of NBCe1, NBCn1 and NDCBE were significantly increased in the testis of prediabetic rats. On the other hand, in the epididymis, prediabetes caused an increase of AE2 and a decrease of NDCBE protein levels. These alterations may be translated into changes of HCO3 ? transepithelial epididymal fluxes in vivo, which may represent a threat for sperm survival. Moreover, these results provide evidence of the molecular mechanism that may be responsible for the significant increase in abnormal sperm morphology already reported in prediabetic rats.  相似文献

Resting or basal intracellular pH (pHi) measured in cultured human syncytiotrophoblast cells was 7.26 ± 0.04 (without HCO3) or 7.24 ± 0.03 (with HCO3). Ion substitution and inhibitor experiments were performed to determine whether common H+-transporting species were operating to maintain basal pHi. Removal of extracellular Na+ or Cl or addition of amiloride or dihydro-4,4'-diisothiocyanatostilbene-2,2'-disulfonate (H2DIDS) had no effect. Acidification with the K+/H+ exchanger nigericin reduced pHi to 6.25 ± 0.15 (without HCO3) or 6.53 ± 0.10 (with HCO3). In the presence of extracellular Na+, recovery to basal pHi was prompt and occurred at similar rates in the absence and presence of HCO3. Ion substitution and inhibition experiments were also used to identify the species mediating the return to basal pHi after acidification. Recovery was inhibited by removal of Na+ or addition of amiloride, whereas removal of Cl and addition of H2DIDS were ineffective. Addition of the Na+/H+ exchanger monensin to cells that had returned to basal pHi elicited a further increase in pHi to 7.48 ± 0.07. Analysis of recovery data showed that there was a progressive decrease in pH per minute as pHi approached the basal level, despite the continued presence of a driving force for H+ extrusion. These data show that in cultured syncytial cells, in the absence of perturbation, basal pHi is preserved despite the absence of active, mediated pH maintenance. We studied the regulation of intracellular pH (pHi) in single cultured astrocytes passaged once from the hippocampus of the rat, using the dye 2′,7′-biscarboxyethyl-5,6-carboxyfluorescein (BCECF) to monitor pHi. Intrinsic buffering power (βI) was 10.5 mM (pH unit)−1 at pHi 7.0, and decreased linearly with pHi; the best-fit line to the data had a slope of −10.0 mM (pH unit)−2. In the absence of HCO3 , pHi recovery from an acid load was mediated predominantly by a Na-H exchanger because the recovery was inhibited 88% by amiloride and 79% by ethylisopropylamiloride (EIPA) at pHi 6.05. The ethylisopropylamiloride-sensitive component of acid extrusion fell linearly with pHi. Acid extrusion was inhibited 68% (pHi 6.23) by substituting Li+ for Na+ in the bath solution. Switching from a CO2/HCO3 -free to a CO2/HCO3 -containing bath solution caused mean steady state pHi to increase from 6.82 to 6.90, due to a Na+-driven HCO3 transporter. The HCO3 -induced pHi increase was unaffected by amiloride, but was inhibited 75% (pHi 6.85) by 400 μM 4,4′-diisothiocyanatostilbene-2,2′-disulfonic acid (DIDS), and 65% (pHi 6.55–6.75) by pretreating astrocytes for up to ∼6.3 h with 400 μM 4-acetamide-4′-isothiocyanatostilbene-2,2′-disulfonic acid (SITS). The CO2/HCO3 -induced pHi increase was blocked when external Na+ was replaced with N-methyl-d-glucammonium (NMDG+). In the presence of HCO3 , the Na+-driven HCO3 transporter contributed to the pHi recovery from an acid load. For example, HCO3 shifted the plot of acid-extrusion rate vs. pHi by 0.15–0.3 pH units in the alkaline direction. Also, with Na-H exchange inhibited by amiloride, HCO3 increased acid extrusion 3.8-fold (pHi 6.20). When astrocytes were acid loaded in amiloride, with Li+ as the major cation, HCO3 failed to elicit a substantial increase in pHi. Thus, Li+ does not appear to substitute well for Na+ on the HCO3 transporter. We conclude that an amiloride-sensitive Na-H exchanger and a Na+-driven HCO3 transporter are the predominant acid extruders in astrocytes.  相似文献

Sertoli cells are responsible for regulating a wide range of processes that lead to the differentiation of male germ cells into spermatozoa. Cytoplasmic pH (pH i ) has been shown to be an important parameter in cell physiology, regulating namely cell metabolism and differentiation. However, membrane transport mechanisms involved in pH i regulation mechanisms of Sertoli cells have not yet been elucidated. In this work, pH i was determined using the pH-sensitive fluorescent probe 2′,7′-bis-(2-carboxyethyl)-5-(and-6)-carboxyfluorescein (BCECF). Addition of weak acids resulted in rapid acidification of the intracellular milieu. Sertoli cells then recovered pH i by a mechanism that was shown to be sensitive to external Na+. pH i recovery was also greatly reduced in the presence of 4,4′-diisothiocyanatostilbene-2,2′-disulfonic acid (DIDS) and amiloride. These results point toward the action of an Na+-driven HCO3/Cl exchanger and/or an Na+/HCO3 cotransporter and the action of the Na+/H+ exchanger on pH i regulation in the experimental conditions used. pH i recovery was only slightly affected by ouabain, suggesting that the inhibition of Na+/K+-ATPase affects recovery indirectly, possibly via the shift on the Na+ gradient. On the other hand, recovery from the acid load was independent of the presence of concanamycin A, a specific inhibitor of the V-type ATPases, suggesting that these pumps do not have a relevant action on pH i regulation in bovine Sertoli cells.  相似文献
