Host plant-mediated orientation and oviposition by diamondback moth (DBM) Plutella xylostella (L.) (Lepidoptera: Yponomeutidae) and its predator Chrysoperla carnea Stephens (Neuroptera: Chrysopidae) were studied in response to four different brassica host plants: cabbage, (Brassica oleracea L. subsp. capitata), cauliflower (B. oleracea L. subsp. botrytis), kohlrabi (B. oleracea L. subsp. gongylodes), and broccoli (B. oleracea L. subsp. italica). Results from laboratory wind tunnel studies indicated that orientation of female DBM and C. carnea females towards cabbage and cauliflower was significantly greater than towards either broccoli or kohlrabi plants. However, DBM and C. carnea males did not orient towards any of the host plants. In no-choice tests, oviposition by DBM did not differ significantly among the test plants, while C. carnea layed significantly more eggs on cabbage, cauliflower, and broccoli than on kohlrabi. However, in free-choice tests, oviposition by DBM was significantly greater on cabbage, followed by cauliflower, broccoli, and kohlrabi, while C. carnea preferred to oviposit on cabbage and cauliflower, followed by broccoli and kohlrabi. The predation rates of DBM by C. carnea on kohlrabi and broccoli were not significantly different from one another, but were significantly higher than that on cabbage and cauliflower. When two types of plant, intact and injured, were available to adult DBM, female oviposition was significantly greater on injured plant leaves than on intact plants leaves. Similarly, C. carnea oviposition was significantly greater on injured plant leaves than on intact leaves. Implications regarding the possible role of green leaf volatiles in host selection/preference, as well as in tritropic interactions, are discussed.  相似文献

A procedure for the production of fertile transgenic brassicas via Ri-mediated transformation is reported in this paper. Transgenic hairy root lines were selected for 12 vegetable brassica cultivars and lines representing six varieties: broccoli, Brussels sprouts, cabbage, cauliflower, rapid-cycling (allBrassica oleracea) and Chinese cabbage (B. campestris). Leaf explants or petioles of intact cotyledons were co-cultivated withAgrobacterium strain A4T harbouring various binary vectors. The T-DNA region of all binary vectors contained a neomycin phosphotransferase II gene for kanamycin resistance, in addition to other genes. Hairy root lines grew prolifically on hormone-free medium containing kanamycin. Transgenic shoots were regenerated from all cultivars either spontaneously or after transfer of hairy roots to a hormone-containing medium. Southern analysis confirmed that the plants were transgenic. Plants from all brassica types were successfully transferred to greenhouse conditions. Identification of broccoli and cauliflower cultivars with RAPD markers   总被引:43,自引:0,他引:43  
Summary RAPD (Random Amplified Polymorphic DNA) markers generated by 4 arbitrary 10-mer primers, discriminated 14 broccoli and 12 cauliflower cultivars (Brassica oleracea L.) by banding profiles. The size of the amplified DNA fragments ranged from 300 to 2600 base pairs. Twenty-eight percent of the markers were fixed in both broccoli and cauliflower, whereas 12.5% were specific to either crop. The rest were polymorphic in either or both crops. The markers generated by two and three primers were sufficient to distinguish each of the broccoli and cauliflower cultivars, respectively. The average difference in markers was 14.5 between broccoli and cauliflower markers, 5.8 between two broccoli cultivars and 7.9 between two cauliflower cultivars. Larger differences for each crop were found between cultivars from different seed companies than within the same company. RAPD markers provide a quick and reliable alternative to identify broccoli and cauliflower cultivars.  相似文献

The production of transgenic roots was scored for eight Brassica oleracea cultivars from broccoli, cabbage, cauliflower and kale following inoculation with an Agrobacterium rhizogenes cell line carrying a binary plasmid bearing the green fluorescence protein (gfp) gene in the T-DNA. Significant differences in the numbers of explants producing transgenic roots were observed between cultivars, ranging from 1.4% for Marathon F1 to 57.8% for the Green Duke F1. Three F1 cultivars were subjected to anther culture, and doubled-haploid (DH) lines were used for transformation. The DH lines produced showed considerable variation for transgenic root production with some lines showing increased efficiency compared to the parental F1 cultivar. Grouping of the DH lines into response classes with respect to transgenic root production allowed the development of potential genetic models to explain the variation in performance released from each F1 cultivar. No apparent segregation distortion for transgenic root production was observed in the DH lines following anther culture.  相似文献

Abstract We studied oviposition preference of Plutella xylostella for four cultivars of cabbage, broccoli and cauliflower in the field. There were no differences in the number of eggs found on the various cultivars of broccoli or cauliflower. Significantly more eggs were laid on cultivar Savoy King than any of the other cabbage cultivars tested. We then compared the development time and feeding damage of larvae reared on Savoy King and Green Coronet cabbages. Larvae developed more rapidly and fed more and for longer on Green Coronet than Savoy King. Thus, while Savoy King is more attractive to oviposition in the field, net impacts on the crop may be lessened to some degree through lower feeding proficiency of the larvae on that cultivar.  相似文献

Brassica oleracea is an important agricultural species encompassing many vegetable crops including cabbage, cauliflower, broccoli and kale; however, it can be susceptible to a variety of fungal diseases such as clubroot, blackleg, leaf spot and downy mildew. Resistance to these diseases is meditated by specific disease resistance genes analogs (RGAs) which are differently distributed across B. oleracea lines. The sequenced reference cultivar does not contain all B. oleracea genes due to gene presence/absence variation between individuals, which makes it necessary to search for RGA candidates in the B. oleracea pangenome. Here we present a comparative analysis of RGA candidates in the pangenome of B. oleracea. We show that the presence of RGA candidates differs between lines and suggests that in B. oleracea, SNPs and presence/absence variation drive RGA diversity using separate mechanisms. We identified 59 RGA candidates linked to Sclerotinia, clubroot, and Fusarium wilt resistance QTL, and these findings have implications for crop breeding in B. oleracea, which may also be applicable in other crops species.  相似文献

Plutella xylostella is an important pest of cruciferous crops worldwide. However, information regarding the age‐stage, two‐sex life parameters of P. xylostella, which is vital for designing more effective control methods, is currently lacking. The present study reports age‐stage, two‐sex life table parameters for P. xylostella on napa cabbage (Brassica oleracea var. napa), white cabbage (B. oleracea var. capitata), and cauliflower (B. oleracea var. botrytis) under laboratory conditions at 25 ± 2°C, 50–60% relative humidity, and a 16‐h light : 8‐h dark photoperiod. The time for development from an egg to a male or female adult P. xylostella on white cabbage (mean [± SE] 41.15 ± 0.54 and 39.50 ± 0.54 days, respectively) was significantly longer than that on cauliflower and napa cabbage. Furthermore, P. xylostella fecundity on cauliflower (261.90 ± 4.53 eggs female) was significantly highest than on napa cabbage and white cabbage. Intrinsic rate of increase (r) and finite rate of increase (λ) were highest on cauliflower 0.182 day?1 and 1.199 day?1 respectively as comparison to napa cabbage and white cabbage. The highest gross reproductive rate (GRR) and net reproductive rates (R0) of P. xylostella 65.87 and 52.58 respectively on cauliflower then those of other hosts. The findings of the present study indicate that cauliflower is the most suitable cultivar (host) for the development of P. xylostella. Based on these findings, crops like cauliflower can be used as trap crops when napa cabbage and white cabbage are the main crops.  相似文献

 Several simple methods of DNA preparation from plant tissues were evaluated for PCR-RFLP analyses of SLG and SRK alleles, which can be used for the identification of S haplotypes of breeding lines in broccoli and cabbage (Brassica oleracea L.) and in purity tests of F1 hybrid seeds. On the five methods tested, the NaI method was found to be the most suitable for the amplification of the SLG and SRK alleles. This method enables the use of a single seed as testing material. Using this method, we identified S haplotypes of 31 broccoli and 31 cabbage cultivars. Ninety-four percent of the cultivars of broccoli and 97% of those of cabbage were-single cross F1 hybrids. Nine and 15 S haplotypes were found in broccoli and cabbage, respectively. The small number of S haplotypes in broccoli suggests the importance of incorporating new S haplotypes in the breeding program. The structure of gastropod communities was examined from January to June 1999 in four sites of the streams of Mont Saint-Michel Bay along a gradient of salinity, and the occurrence of larval trematodes infecting snails was studied. Abundance and species richness of gastropods increased from polyhaline (95 snails, 1 species) to oligohaline waters (6672 snails, 6 species). Whatever the salinity, the most abundant species was Potamopyrgus antipodarum, an invasive non-indigenous species that represented 80% of the gastropods. Only one male was found in P. antipodarum populations suggesting a predominantly parthenogenetic mode of reproduction. Among 7218 gastropods collected, 1.2% were infected by larval trematodes: 5 species in Lymnaea peregra (4.4%), 4 species in Planorbis planorbis (12.0%), one echinostome in Physa acuta (0.2%), and a new species of Sanguinicola in P. antipodarum (0.5%). This is the first record of infected P. antipodarum in Europe. No parasites were found in polyhaline waters. The prevalence per host population varied from 0 to 100% depending on time of collection, salinity and host species. In the lowest-salinity site, abundance of gastropods and prevalence of trematodes were negatively correlated. The dominance of P. antipodarum in the gastropod communities is discussed in relation with euryhalinity, parthenogenesis and weak rate of parasitism.  相似文献

  1. The wheat bug Nysius huttoni is a major pest of brassica seedlings. Management of this insect currently relies on seed treatment with neonicotinoids and spraying with chlorpyrifos and pyrethroid insecticides. These practices can generate severe external costs, including human health, the environment and biodiversity. Trap cropping is one alternative option to protect brassica seedlings from N. huttoni.
  2. Trap crop species evaluated in field cage experiments were: alyssum (Lobularia maritima L. Desvauxcv. Benthamii White), wheat (Triticum aestivum L. cv. Morph), coriander (Coriandrum sativum L. cv. Santo) and clover (Trifolium repens L. cv. Nomad). These were compared with kale (Brassica oleracea L. cv. Kestrel). In open‐field experiments, alyssum (L maritima), wheat (T. aestivum) and a mixture of alyssum (L. maritima) and wheat (T. aestivum) were used. All of these were compared to kale (B. oleracea).
  3. Alyssum and wheat were the most favoured potential trap plants for N. huttoni. Results indicated that two treatments: alyssum (used as a single trap crop) or ‘alyssum plus wheat’ (a multiple trap crop), may be useful in brassica fields to protect the seedlings from N. huttoni damage.
  4. Such a trap cropping protocol potentially reduces pesticide use in forage brassicas and can also deliver multiple ecosystem services such as biological control of insect pests.

  • 1 The performance of the second generation (G2) of alates and apterae of a generalist, Myzus persicae, and a specialist, Brevicoryne brassicae, aphid species reared on Chinese cabbage or cabbage was evaluated on five cultivars of Brussels sprout.
  • 2 Aphid performance was influenced both by the type of host on which the parent aphid had been reared and by the host on which it was feeding when reproducing.
  • 3 The fecundity of the G2 of alates of both aphid species reared on Chinese cabbage differed significantly between all the cultivars of Brussels sprout and, on average, was 25% higher than those reared on cabbage. These differences were also apparent for the intrinsic rate of increase of B. brassicae but not for M. persicae.
  • 4 There was a trend for the G2 of alates from Chinese cabbage to have greater fecundity compared with aphids from cabbage. These differences were significant for the fecundity of the G2 of alates of both aphid species on Brussels sprout cultivars Fillbasket (30% higher), Red Delicious (35% higher) and Winter Harvest (25% higher) than those reared on cabbage.
  • 5 The intrinsic rate of increase for the G2 of alates of B. brassicae from Chinese cabbage was significantly different on all Brussels sprout cultivars tested. The intrinsic rate of increase differed significantly between aphids reared on either Chinese cabbage or cabbage on cultivars Oliver and Darkmar‐21 (M. persicae) and Red Delicious and Winter Harvest (B. brassicae). The cv. Oliver appeared to be the most consistently good host; Red Delicious was the poorest host overall.

Laboratory and greenhouse studies were conducted to investigate the suitability of 2 hymenopterous parasites,Diadegma eucerophaga Horstmann andApanteles plutellae Kurdjumov for introduction to control diamondback moth (DBM),Plutella xylostella (L.), a destructive pest of crucifers in tropical to subtropical Southeast Asia. Parasitism byD. eucerophaga was high at temperature range of 15°C to 25°C and that ofA. plutellae, at 20°C to 35°C. Both parasites were active in searching for host and oviposited only during photophase. No parasitism was observed during darkness. WhereasA. plutellae could parasitize all instars of DBM larvae,D. eucerophaga parasitized only the first 3 instars and failed to parasitize the 4th. Parasitism byD. eucerophaga was greater when DBM larvae were feeding on common cabbage (Brassica oleracea var.capitata L.), than on cauliflower (B. oleracea var.italica L.), broccoli (B. oleracea var.botrytis L.) or Chinese cabbage [Brassica campestris L. ssp.pekinensis (Lour) Olsson].A. plutellae parasitism was greater when DBM larvac were feeding on Chinese cabbage than on common cabbage, cauliflower or broccoli. Storage of pupae at 0°C and 4°C to 6°C for up to 2 weeks reduced emergence ofD. eucerophaga adults more than that ofA. plutellae. A non-selective insecticide, deltamethrin, was toxic to adults of both parasites but selective ones such asBacillus thuringiensis, teflubenzuron, and pirimicarb were not. Pupae were more tolerant than adults to insecticides. The insecticide-resistant Luchu strain and susceptible laboratory strain of DBM suffered an equal level of parasitism by both parasites.   相似文献

Four accessions of the wild species Brassica fruticulosa Cyrillo (Brassicaceae) were studied in order to identify its tolerance and antibiosis resistance to the cabbage root fly, Delia radicum L. (Diptera: Anthomyiidae), in comparison to a widely cultivated cauliflower cultivar and a rapid cycling Brassica oleracea L. line. Antibiosis was prominent, as the insects reared on resistant accessions showed reduced individual pupal weight, total pupal weight, adult dry weight, and the longest average fly eclosion time. Host plant resistance, however, did not affect the sex ratio of adult flies. A study of the root architecture of plants with and without root fly inoculation revealed differences in the structure within B. oleracea accessions. A long main root and a high number of lateral roots appeared to be important characteristics for a Brassica type, with a higher tolerance level to cabbage root fly attack.  相似文献

Brassica oleracea L. vars.italica (broccoli) andbotrytis (cauliflower) both undergo developmental arrests which result in heading phenotypes. We characterized these arrested tissues at the morphological and molecular levels, and defined the developmental changes that ensue after arrest has been broken. We found that the order of floral organ initiation and the positions of resulting floral organ primordia in this species differed in some respects from that ofArabidopsis, which is a member of the same family, Brassicaceae. We also cloned homologs of theArabidopsis floral homeotic genesAPETALA1 (AP1) andAPETALA3 (AP3) fromB. oleracea and characterized their expression patterns. We found that theAP1 homolog was expressed in some of the meristems of arrest-stage cauliflower, providing evidence that this tissue is florally determined. In broccoli, both the API andAP3 homologs were expressed. However, the spatial pattern of expression of the broccoliAP1 homolog differed from that ofArabidopsis. In addition, we identified a homolog of theCAULIFLOWER (CAL) gene,BoiCAL, from broccoli. We studied the effect of different host plants and rearing atmosphere on life cycle of cabbage butterfly, Pieris brassicae. Insects were reared in the field (fluctuating weather) as well as in the laboratory (constant rearing conditions) on four host plants, viz. cabbage, cauliflower, knol-khol and broccoli. Significant differences were not found in the incubation and pupal period of butterfly reared on different host plants. However, larval period was found to be significantly lower on cabbage followed by knol-khol and highest on broccoli. Therefore, the developmental period (from eggs to adult) was reasonably lower on cabbage. Furthermore, inverse relationship was found in the body weight of butterfly and developmental period, where weight of full grown caterpillar and pupae was significantly higher on cauliflower and lowest on cabbage. Besides, significant differences were not found in the body weight of P. brassicae caterpillar reared under field and in the laboratory. Nevertheless, pupal weight of butterfly was significantly higher under field conditions than the laboratory conditions. Overall, the development of P. brassicae was much faster on cabbage than other hosts; but its body weight was considerably higher on cauliflower.  相似文献

The olfactory response of the parasitoid Aphidius colemani (Viereck) (Hymenoptera: Braconidae) to odours in a tritrophic system involving three cultivars of common cabbage, Brassica oleracea var capitata, characterized by different levels of susceptibility to Myzus persicae (Sulzer) (Hemiptera: Aphididae) was studied in a four‐way olfactometer. Odours influenced A. colemani response in the olfactometer to varying degrees. The magnitude of parasitoid response to odours of uninfested cabbage depended on cultivar, with Derby Day [green‐leaved, susceptible to M. persicae and the crucifer specialist, Brevicoryne brassicae (Linnaeus) (Hemiptera: Aphididae)] and Minicole (green‐leaved, partially resistant with known antibiosis factors for B. brassicae) preferred over Ruby Ball (red‐leaved with antixenosis factors for M. persicae and B. brassicae). The odour of the cabbage cultivar on which the parasitoid had been reared was preferred over the other cultivars. However, when provided with a choice between odours of infested plants, parasitoids did not show a significant preference for the cultivar on which they were reared. Results from the study show that parasitioids differentiated between odour of the three cultivars in dependence of their rearing history when the plant is uninfested.  相似文献

Fire has long been recognised as a natural force in structuring Northern Hemisphere salt marshes, yet little is known about the impact of fire on molluscs and native vegetation dynamics of Southern Hemisphere coastal salt marshes. Following a fire at Ash Island, Hunter River New South Wales, Australia in the summer 2012, we assessed patterns of recovery through time of gastropod populations and resident salt marsh vegetation including biomass for three keystone native plant species, Native Rush (Juncus kraussii Hochst.), a chenopod (Sarcocornia quinqueflora Bunge ex Ungen‐Sternberg A.J. Scott), Salt Couch (Sporobolus virginicus, L. Kunth) and the invasive Spiny Rush (Juncus acutus). In temperate east‐coast Australian salt marshes, Spiny Rush is displacing native salt marsh vegetation. After twelve months, the biomass of Native Rush recovered to similar pre‐burn levels. While fire affected the abundance, richness and composition of the gastropod assemblage differences were also largely driven by spatial variability. Gastropod assemblages associated with two of the higher elevation native species (Native Rush and Salt Couch) were impacted the most by fire. Greater abundance (between 1 and 5 orders of magnitude difference in abundance) and richness of gastropods were found in unburnt compared with burnt Native Rush and Salt Couch vegetation, while more gastropods were found in Spiny Rush in one site. Species prevalent in burnt vegetation included larger species of gastropods Ophicardelus ornatus (Ferussac, 1821) and Phallomedusa solida (Martens, 1878) with an unexpected spike in number of the smaller gastropod Tatea huonensis (Tenison‐Woods, 1876) in the spiny rush at one site only. In salt marsh habitats, many gastropods have planktonic larval dispersal stages which are dependent on the tidal height for transport and the structural complexity provided by vegetation at settlement. Since fire appears to negatively affect salt marsh gastropod populations within structurally complex Native Rush and Salt Couch, due consideration of the importance of these refuges for gastropods is recommended when fire or other disturbances occur in ecologically endangered salt marsh in the Southern Hemisphere. Managers need to consider spatial heterogeneity of molluscs and their recovery in the event of fire in Southern Hemisphere salt marshes.  相似文献

  • 2 We grew brassica plants in several types of fertilizer, including those commonly used in conventional and sustainable farming systems, and an unfertilized control. The effects of fertilizer type on the performance of two aphid species and foliar glucosinolate content were investigated.
  • 3 Both aphid species performed poorly (with reduced fecundity) on the unfertilized treatment compared with those feeding on fertilized host plants.
  • 4 Brevicoryne brassicae, the brassica specialist, performed best on Brassica oleracea plants fertilized with an organic animal manure, with a 72% increase in fecundity and an 18% increase in intrinsic rate of increase compared with plants fertilized with ammonium nitrate.
  • 5 By contrast, the generalist Myzus persicae had an intrinsic rate of increase that was reduced by 15% on plants growing in the animal manure compared with those growing in ammonium nitrate.
  • 6 These results may explain earlier findings on the effects of fertilizer type on aphid populations in the field, and are discussed in the context of pest species' responses to sustainable and conventional agricultural systems.
