We hypothesized that the polyunsaturated fatty acids of the butterfly were probably derived from the diet and that there might be a great loss of body fat during metamorphosis. To substantiate these hypotheses, we analyzed the fatty acid composition and content of the diet, the larva, and the butterfly Morpho peleides. Both the diet and the tissues of the larva and butterfly had a high concentration of polyunsaturated fatty acids. In the diet, linolenic acid accounted for 19% and linoleic acid for 8% of total fatty acids. In the larva, almost 60% of the total fatty acids were polyunsaturated: linolenic acid predominated at 42% of total fatty acids, and linoleic acid was at 17%. In the butterfly, linolenic acid represented 36% and linoleic acid represented 11% of total fatty acids. The larva had a much higher total fatty acid content than the butterfly (20.2 vs. 6.9 mg). Our data indicate that the transformation from larva to butterfly during metamorphosis drastically decreased the total fatty acid content. There was bioenhancement of polyunsaturated fatty acids from the diet to the larva and butterfly. This polyunsaturation of membranes may have functional importance in providing membrane fluidity useful in flight.  相似文献

A preliminary digestion of total plasma lipid extracts with phospholipase C, which converts the lysolecithins, lecithins, and sphingomyelins into monoglycerides, diglycerides, and ceramides, respectively, has been shown to facilitate subsequent determination of the plasma lipids by gas-liquid chromatography. A further improvement in the chromatographic elution pattern results from acetylation or trimethylsilylation of the liberated alcohol moieties prior to injection into the chromatograph. If tridecanoin is used as internal standard, quantitative estimates can be rapidly obtained for plasma lysolecithins, free cholesterol, lecithins, sphingomyelins, cholesteryl esters, and triglycerides, as well as for free fatty acids. Other plasma lipids do not occur in sufficiently high concentrations to interfere with the analysis. The determination requires 0.1-0.5 ml of plasma and about 6 hr of processing, but many samples can be processed at a time.  相似文献

Substitution of selected saturated fatty acids of the diet of 29 men and 29 women with cis or trans monounsaturated fatty acids did not affect erythrocyte membrane fluidity, insulin binding, and the membrane cholesterol and phospholipid concentrations. Subjects were fed four different controlled diets with a total fatty acid content of 39 to 40 energy percent for four 6-week periods in a Latin square design. The diets were: (1) high oleic acid (16.7 energy percent oleic); (2) moderate trans (3.8 energy percent trans fatty acids); (3) high trans (6.6 energy percent trans fatty acids); and saturated (16.2 energy percent lauric + myristic + palmitic acids). There were no significant diet effects on red cell ghost fluidity determined by fluorescence polarization of the hydrocarbon probe 1,6-diphenyl-1,3,5-hexatriene (DPH) and the polar analog trimethylammonium-DPH (TMA-DPH). There were limited diet effects on fluidity of membranes as determined with DPH-propionic acid (DPH-PA) for the men. Insulin binding was more closely associated with anisotropy of fluorescence of the surface probe, DPH-PA, than with that of the other probes, which is compatible with the localization of the insulin receptor in a domain at the cell membrane surface.  相似文献

The lipid fractions were studied in the testicular tissue of mature bulls, of the lowland black-and-white breed. It was found that the main component of neutral lipids was cholesterol (48%) followed by triglycerides (24%), cholesterol esters (16%) and free fatty acids (12%). In cholesterol esters the main component was palmitic acid (41%) followed by oleic acid (22%), stearic acid (14%) and linoleic acid (14%). In phospholipids the main fraction was composed of lecithins (48%) followed by phosphatidylethanolamine (20%) and phosphatidic acids and phosphatidylglycerol (13%). Palmitic acid was found mainly in the fractions of lecithins and sphingomyelins, stearic acid in fractions of phosphatidylethanolamine, phosphatidylserine and phosphatidylinositol. Linoleic acid was found in the fractions of phosphatidylethanolamine, phosphatidylcholine and sphingomyelin. Arachidonic, docosatetraenoic and docosapentaenoic acids in the fractions of phosphatidylethanolamine, phosphatidylserine, phosphatidylinositol and phosphatidylcholine.  相似文献

Abstract— The variation with age of the fatty acid composition of the major lipids in human brain myelin was compared with that of cerebral white matter from the same region. The myelin was isolated from the semiovale centre of the cerebrum of 27 subjects neonatal to old aged. The phospholipid, cholesterol and galactolipid concentrations were determined in all the samples, as were the proportions of the major phospholipid classes. The proportions of cholesterol and especially of the galactolipids increased in myelin during the first 6 months, and in cerebral white matter up to 2 years. During this period the individual phospholipids also varied substantially. Serine phosphoglycerides and especially sphingomyelins increased, and choline phosphoglycerides decreased. The fatty acid patterns of ethanolamine phosphoglycerides (EPG) and sphingomyelins underwent the largest changes. The proportions of saturated fatty acids in EPG diminished rapidly, and there was an increase of monoenoic acids. Fatty acids of the linoleic acid series showed a peak between 4 and 12 months, after which time their proportion slowly diminished to old age. The major fatty acid of this series was docosatetraenoic acid, 22:4 (n-6), which constituted more than 25% of total fatty acids at the maximum level. The fatty acid changes were larger in cerebral white matter, but from 2 years of age the EPG fatty acid pattern in myelin was similar to that in white matter. The fatty acid changes in serine and choline phosphoglycerides of myelin with maturation were much less striking than in EPG but of a similar type. In myelin sphingomyelin the proportion of saturated long-chain fatty acids, C16-C22, diminished, while that of monoenoic acids increased and continued to do so up to old age. From 2 years of age the fatty acid patterns in myelin and cerebral white matter were quite similar. Also the fatty acid patterns of cerebrosides and sulphatides in cerebral white matter and myelin were the same except for the first 2 months of life. The same fatty acid changes occurred in cerebrosides and sulphatides as in the sphingomyelins, i.e. increased proportions of unsaturated (monoenoic) acids. The proportions of 24:1 and 24h:1 and of the odd-numbered fatty acids 25:1 and 23h:1 continued to increase to old age. The variations of the individual lipid fatty acid patterns were small except in the youngest age classes, in which the variations were presumably ascribable to the difficulty in determining the gestational age.  相似文献

The lipids from the electric organ of the ray, Torpedo marmorata, have been isolated and characterized. The major lipids were cholesterol, choline phospholipids, ethanolamine phospholipids, and sphingomyelins. The major fatty acids of ethanolamine phospholipids were 18:1, 18:0, 22:6, and 20:4. More than 50% of the acids in choline phospholipids were 16:0. The sphingomyelins consisted of five major ceramide species, all with sphingosine and the fatty acids 14:0, 15:0, 16:0, 22:1, and 24:1. The fatty acid 15:0 was mostly branched (n-2), a fatty acid earlier identified in sphingomyelins of the rectal gland of spiny dogfish. All long-chain bases were dihydroxy bases with a small percentage of branched chains. Sulfatides (cerebroside sulfate) made up the largest glycolipid fraction. The polar moiety wase galactose-3-sulfate. The fatty acids were normal and 2-hydroxy; the homologue 24:1 was the most abundant in both types of fatty acids. Most fatty acids were higher homologues of mono-unsaturated acids, but normal 18:0 fatty acid was also found. The long-chain bases were both dihydroxy and trihydroxy, with very small amounts of branched chains. The two major ceramide species of sulfatides were sphingosine combined with normal and hydroxy 24:1 fatty acids, respectively. Smaller amounts of trihydroxy base (18:0) were found linked to hydroxy 24:1 fatty acid, but not to its normal homologue. The cerebrosides contained the two major species mentioned above but lacked the trihydroxy base-hydroxy fatty acid species. The ratio of the activity of Na+-K+-dependent ATPase (EC 3.6.1.3) and the concentration of sulfatides was similar to ratios found for other tissues with normal and increased Na+ and K+ transporting capacity. The metabolism of sphingomyelins and ceramides with defined labeled fatty acids was compared after injection in vivo or incubation with cultured cells. The liver was the major site of uptake of sphingomyelins and ceramides with 18:2 or 16:0 fatty acids, but with both sphingolipids a higher recovery of radioactivity was found with 16:0 species. The distribution of radioactivity among liver lipids showed that 1.5 h after injection of 18:2 sphingomyelin, only 21% of the label was found as sphingomyelin, and this value was 37% in the case of 16:0 sphingomyelin. There was a very marked difference in the metabolism of 18:2 and 16:0 ceramides. After injection of 18:2 ceramide only 14% of the radioactivity was recovered as sphingomyelin, and this value was more than 50% with 16:0 ceramide. [14C]18:2 ceramide was converted also to glucoceramide and hydrolyzed more extensively than 16:0 ceramide. These observations were extended to sphingomyelins and ceramides with other fatty acids, using Hep-G2 cells in culture. Significantly more radioactivity was recovered as labeled sphingomyelin after incubation with 16:0, 18:0, 20:0 and 24:0 sphingomyelins than with 18:1 and 18:2 sphingomyelins, while more labeled phosphatidylcholine and phosphatidylethanolamine were found with the unsaturated sphingomyelins. In analogy to the findings in vivo, in the Hep-G2 cells more 16:0, 18:0 and 24:0 ceramides were converted to sphingomyelin than 18:1 or 18:2 ceramides. These differences were also seen with cultured macrophages, in which a more marked reutilization for sphingomyelin formation was found with the saturated ceramide series. The sphingomyelin liposomes were tested also for their capacity to mobilize cholesterol, and a rise in plasma unesterified cholesterol occurred after injection of 18:2 sphingomyelin. Marked enhancement of cholesterol efflux from cholesterol ester-loaded macrophages was also seen with 18:1 and 18:2, 20:0 sphingomyelin in the presence of delipidated high-density lipoprotein. The present results demonstrate that the metabolic fate of sphingolipids is related to their fatty acid composition. While ceramides with saturated fatty acids are predominantly reutilized for sphingomyelin formation, those with unsaturated fatty acids undergo probably more rapid hydrolysis with liberation of fatty acids and channeling into glycerolipids.  相似文献

Serum sphingomyelins were analyzed by argentation chromatography of the corresponding ceramide diacetates. Six subfractions were obtained. Three of them contained 4-sphingenines in combination with saturated, trans-, or cis-monoenoic fatty acids; the remaining three contained sphingadienine, also in combination with saturated, trans-, or cis-monoenoic fatty acids. Palmitic acid was the principal fatty acid combined with 4-sphingenines, while nervonic acid was the principal fatty acid combined with sphingadienine. About 4% of the total fatty acids of sphingomyelin were trans-monoenoic. They were comprised of many positional isomers of straight-chain C(22-24) compounds. The cis-monoenoic acids made up 33% of the total acids and consisted of almost pure nervonic acid. The rest of the acids were saturated. The 4-sphingenines contained small amounts of iso-C(18) and anteiso-C(19) compounds in addition to the straight-chain C(16-18) bases.  相似文献

Abstract— The metabolism of rat brain sphingomyelins containing short-chain (C16-C18) and long-chain (C20- C24) fatty acids has been studied by determination of the content of radioactivity in the sphingo-sinc. fatty acids and phosphate of the sphingomyelins over a period of 60 days following the intracisternal injection of [14C]acetate and [32P]phosphate. From the rate of decrease of the specific radioactivities of the different constituents of short-chain fatty acid sphingomyelins, we have calculated a half-life of 65 days for sphingosine. 41 days for fatty acids and 62 days for phosphate. For the long-chain fatty acid sphingomyelins the half-life of sphingosine was approximately 465 days. The fatty acids and phosphate from these sphingomyelins had fast and slow turnover pools. The half-life for the fast pool was 7 days for the two constituents and the estimated half-lives for the slow pool were 220 days for fatty acids and 480 days for phosphate. These results suggest that one can distinguish at least three metabolic pools of brain sphingomyelins: (a) sphingomyelins with long-chain fatty acids situated in myelin whose half-lives are 465 days for sphingosine, 220 days for fatty acids and 480 days for phosphate; (b) sphingomyelins with long-chain fatty acids located mainly in non-myelin structures having half-lives of 465 days for sphingosine. 7 days for fatty acids and 7 days for phosphate; (c) sphingomyeiins with short-chain fatty acids with half-lives of 65 days for sphingosine. 41 days for fatty acids and 62 days for phosphate. The differences between the half-lives of the three metabolic pools of sphingomyelin, together with the subcellular localizations of the two molecular species of these compounds, suggest that the metabolism of the different molecular species of sphingomyelin are independent and that in various subcellular fractions the long-chain fatty acid and short-chain fatty acid sphingomyelins have different turnover rates.  相似文献

By means of differential centrifugation three fractions: large granules (L2), microsomes (M1), and supernatant fluid (MS), have been isolated from the cytoplasm of mouse liver cells and the contained lipides have been extracted and characterized.The particulate fractions, large granules and microsomes, make up approximately 38% of the total solids of the cytoplasm, but they contain 62% of the total lipide and 85% of the phospholipide. The phospholipide of the particulates has an N:P ratio of 1 while the supernatant phospholipide has an N:P ratio of 1.3, suggesting that the supernatant fluid contains lipides of higher nitrogen content than that of either lecithins or cephalins.The fatty acids of the large granule fraction are highly unsaturated and contain 20% of fatty acids with four double bonds. The unsaturation of the fatty acids of microsomes and the supernatant fluid is comparable, as indicated by the iodine values, but the supernatant fluid contains much more of the fatty acids with one double bond and less of the fatty acids with four double bonds than do the microsomes.  相似文献

NMR and electrospray ionization tandem mass spectrometry were used to show for the first time the presence of sphingomyelins in extracts of the tobacco hornworm Manduca sexta (Lepidoptera). The sphingosine in the ceramide was identified as tetradecasphing-4-enine, and the fatty acids were C18:0, C20:0, C22:0, and C24:0 (compound 1). Heterogeneity in the ceramide was observed in sphingomyelins from M. sexta. All of the sphingomyelins were associated with their doubly unsaturated sphingosine, tetradecasphing-4,6-dienine (compound 2), which contained the same set of fatty acids as compound 1 and represents a novel set of sphingomyelins not previously reported in Lepidoptera. Lipid rafts were isolated from brains of M. sexta, and the association of these novel sphingomyelins with rafts was confirmed. The existence of the additional double bond was also observed in ceramide and ceramide phosphoethanolamine isolated from M. sexta. The levels of the doubly unsaturated ceramide showed modest changes during metamorphosis of M. sexta. These results suggest that Manduca sphingomyelins may participate in the formation of lipid rafts, in keeping with their function in vertebrates.  相似文献

1. The predominant lipids of nerve cords, ganglion and brain from horseshoe crabs were cholesterol (11% of lipid) and phospholipid (81% of lipid). 2. Major phospholipids were phosphatidyl ethanolamine and phosphatidyl choline with lesser amounts of phosphatidyl serine and phosphatidyl inositol and sphingomyelin. 3. The phospholipid fraction was characterized by a high content of plasmalogen, i.e. alk-1-enyl acyl phosphatides, so that 42% of the ethanolamine phosphatides were the plasmalogen, phosphatidal ethanolamine. 4. Phosphatidyl choline and phosphatidyl ethanolamine were high in polyunsaturation with 20:4 and 20:5 major fatty acids. Sphingomyelin had predominantly long chain saturated fatty acids. 5. 1. Lecithin was prepared from the livers of rats deficient in essential fatty acids and analysed by means of countercurrent distribution. Thin-layer chromatography showed that only lecithin was present. 2. The distributions of phosphorus and the fatty acids at the 3 and 2 positions were determined. 3. It has been shown that 26% of the fatty acids in the 3 position were unsaturated and that most of the Delta(5,8,11)-eicosatrienoic acid and the arachidonic acids occur as the stearoyl or oleoyl lecithins.  相似文献

In order to test whether monounsaturated fatty acids (MUFA, M) would lower plasma cholesterol and/or triglycerides (TG), 14 young men were studied in a metabolism ward. They were given two experimental diets with the same P/S ratio (P: polyunsaturated; S:saturated fatty acids) but different P + M/S ratios in composition of dietary fatty acids. Six men were in experiment I. In two groups of three, each group was given either the high or the low P + M/S ratio diet for 6 weeks. In experiment II, two groups of four men were subjected to a 3-week cross-over feeding trial (total of 6 weeks). We found in the crossover study that when the dietary P/S ratio was fixed at 1.0, those subjects given a diet with the higher P + M/S ratio of 4.5 showed significantly higher plasma total TG, very low density lipoprotein (VLDL)-TG, low density lipoprotein (LDL)-TG, and LDL-cholesterol than when given a diet with a lower P + M/S ratio of 1.5. Plasma total cholesterol, apoprotein A-I, and apoprotein B-100 were not significantly different between the two dietary periods. Plasma fatty acid patterns showed a significantly higher plasma MUFA level during the period of a higher P + M/S diet. In contrast to some recent reports, our studies indicate that a large amount of dietary MUFA may raise some fractions of plasma lipids in humans.  相似文献

Fourteen patients with established maturity-onset diabetes were treated as outpatients with a high-carbohydrate-(about 60% of total daily energy requirements)-modified fat diet (ratio of polyunsaturated fatty acids to other fatty acids greater than or equal to 1:1) for six weeks. Commercially available and acceptable cereal foods and tuberous vegetables high in both digestible and non-digestible carbohydrates were used. Simple sugars were restricted. Compared with their usual, low-carbohydrate diabetic diet this diet resulted in a fall in basal plasma glucose concentration (average of values measured at 0300, 0500, and 0700), mean preprandial plasma glucose concentration (average of values measured at 0800, 1230, and 1730), and percentage of glycosylated haemoglobin. Modifying dietary fat also decreased the fasting plasma cholesterol concentration. The findings suggest that it is no longer justifiable to prescribe a low-carbohydrate diet for maturity-onset diabetes.  相似文献

Background  

The effects of an omega-3 (n-3) fatty acid enriched diet alone and in combination with gamma irradiation (IR) therapy in nude mice bearing a human MDA-MB231 breast cancer xenograft were tested. The cancer cells were injected into the mammary fat pad of young female mice. Six weeks later, mice were randomly divided into two diet groups: 1) mice with 10% corn oil (rich in omega 6 fatty acids) in their food, 2) mice consuming a 10% fat diet that was enriched in n-3 fatty acids. After two weeks on the diet, treatment with 200 cGy of IR every second day for four treatments (total 800 cGy) was initiated on half of the mice from each diet group. Some mice in each of the 4 groups were euthanized 24 hours after the end of IR while the remaining mice were followed for 3 additional weeks. Tumor sections were stained for endothelial cells with CD31 and PAS and for hypoxia inducible factor 1α (HIF-α).  相似文献

We evaluated lipid metabolism during 90 min of moderate-intensity (50% VO(2) peak) cycle ergometer exercise in five men and five women who were matched on adiposity (24 +/- 2 and 25 +/- 1% body fat, respectively) and aerobic fitness (VO(2) peak: 49 +/- 2 and 47 +/- 1 ml x kg fat-free mass(-1) x min(-1), respectively). Substrate oxidation and lipid kinetics were measured by using indirect calorimetry and [(13)C]palmitate and [(2)H(5)]glycerol tracer infusion. The total increase in glycerol and free fatty acid (FFA) rate of appearance (R(a)) in plasma during exercise (area under the curve above baseline) was approximately 65% greater in women than in men (glycerol R(a): 317 +/- 40 and 195 +/- 33 micromol/kg, respectively; FFA R(a): 652 +/- 46 and 453 +/- 70 micromol/kg, respectively; both P < 0.05). Total fatty acid oxidation was similar in men and women, but the relative contribution of plasma FFA to total fatty acid oxidation was higher in women (76 +/- 5%) than in men (46 +/- 5%; P < 0.05). We conclude that lipolysis of adipose tissue triglycerides during moderate-intensity exercise is greater in women than in men, who are matched on adiposity and fitness. The increase in plasma fatty acid availability leads to a greater rate of plasma FFA tissue uptake and oxidation in women than in men. However, total fat oxidation is the same in both groups because of a reciprocal decrease in the oxidation rate of fatty acids derived from nonplasma sources, presumably intramuscular and possibly plasma triglycerides, in women.  相似文献

The effect of additional dietary trans fatty acids (7% energy) on plasma lipids was assessed in a double-blind comparison of four separate diets: 1, enriched with butter fat (lauric-myristic-palmitic); 2, oleic acid-rich; 3, elaidic acid-rich; 4, palmitic acid-rich. The total dietary period was 11 weeks and comprised normal foods plus specific fat supplements. In 27 mildly hypercholesterolemic men, total and LDL cholesterol were significantly lower during the 3-week oleic acid-rich diet, and were similar during the other three diets. For the four diets LDL cholesterol levels were in mg/dl: 1, 163; 2, 151; 3, 165; 4, 161. HDL cholesterol was significantly higher with the palmitic acid-rich diet, 42 mg/dl, compared with elaidic acid, 38 mg/dl, which in turn was not lower than with oleic acid, 38 mg/dl. Plasma elaidic acid concentration rose seven-fold with the trans fatty acid diet but did not increase the vulnerability of LDL to oxidative change. The elaidic acid-rich diet led to significant elevations in the level of Lp[a] compared to all the other test diets. The Lp[a] level increased to 296 +/- 220 U/l in the elaidic acid-rich period from 235 +/- 182 (mean +/- SD) in the first ("butter") period (P less than 0.001) compared with 249 +/- 204 in the palmitic acid period (P less than 0.001) and 236 +/- 201 in the oleic acid period (NS).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Serum lipoprotein[a] (Lp[a]) is a strong risk factor for coronary heart disease. We therefore examined the effect of dietary fatty acid composition on serum Lp[a] levels in three strictly controlled experiments with healthy normocholesterolemic men and women. In Expt. I, 58 subjects consumed a control diet high in saturated fatty acids for 17 days. For the next 36 days, 6.5% of total energy intake from saturated fatty acids was replaced by monounsaturates plus polyunsaturates (monounsaturated fatty acid diet; n = 29) or by polyunsaturates alone (polyunsaturated fatty acid diet; n = 29). Both diets caused a slight, nonsignificant, increase in median Lp[a] levels, with no difference between diets. In Expt. II, 10% of energy from the cholesterol-raising saturated fatty acids (lauric, myristic, and palmitic acid) was replaced by oleic acid or by trans-monounsaturated fatty acids. Each of the 59 participants received each diet for 3 weeks in random order. The median level of Lp[a] was 26 mg/l on the saturated fatty acid diet; it increased to 32 mg/l (P less than 0.020) on the oleic acid diet and to 45 mg/l (P less than 0.001) on the trans-fatty acid diet. The difference in Lp[a] between the trans-fatty acid and the oleic acid diets was also highly significant (P less than 0.001). Expt. III involved 56 subjects; all received 8% of energy from stearic acid, from linoleic acid, or from trans-monounsaturates, for 3 weeks each. Previous studies suggested that women synthesise docosahexaenoic acid (DHA) more efficiently from their precursors than men. This study investigated the relationship between diet, platelet phospholipids fatty acids and gender. Dietary intake and platelet phosphatidyl-choline (PC) and phosphatidylethanolamine (PE) fatty acids were determined in Caucasian 40 men and 34 women. Absolute and %energy intakes of arachidonic acid (AA), eicosapentaenoic acid (EPA), and DHA, and the ratios of total n-6/n-3 PUFA and linoleic/alpha-linolenic acids did not differ between the sexes. However, women had higher DHA in PC (1.19 vs 1.05 wt%, p<0.05) and PE (3.62 vs 3.21 wt%, p<0.05) than men. Also EPA (1.10 vs 0.93 wt%, p<0.05) was higher in women's PE. Conversely, men had elevated AA and total n-6 fatty acids in PC. The higher platelet DHA levels and lower platelet AA/EPA and AA/DHA ratios in women of child-bearing age compared with men, may lead to less platelet aggregation and vaso-occlusion.  相似文献
