Fusarium species can produce fumonisins (FBs), fusaric acid, beauvericin (BEA), fusaproliferin (FUS) and moniliformin. Data on the natural occurrence of FBs have been widely reported, but information on BEA and FUS in maize is limited. The aims of this study were to establish the occurrence of Fusarium species in different maize hybrids in Mexico, to determine the ability of Fusarium spp. isolates to produce BEA, FUS and FBs and their natural occurrence in maize. Twenty-eight samples corresponding to seven different maize hybrids were analyzed for mycobiota and natural mycotoxin contamination by LC. Fusarium verticillioides was the dominant species (44–80%) followed by F. subglutinans (13–37%) and F. proliferatum (2–16%). Beauvericin was detected in three different hybrids with levels ranging from 300 to 400 ng g−1, while only one hybrid was contaminated with FUS (200 ng g−1). All samples were positive for FB1 and FB2 contamination showing levels up to 606 and 277 ng g−1, respectively. All F. verticillioides isolates were able to produce FB1 (13.8–4,860 μg g−1) and some also produced FB2 and FUS. Beauvericin, FUS, FB1 and FB2 were produced by several isolates including F. proliferatum and F. subglutinans and co-production was observed. This is the first report on the co-occurrence of these toxins in maize samples from Mexico. The analysis of the presence of multiple mycotoxins in this substrate is necessary to understand the significance of these compounds in the human and animal food chains.  相似文献

Fusarium wilt of watermelon commonly occurs in locations where the crop has been grown for many seasons. Its occurrence results in a severely decreased watermelon crop. The goal of this study was to assess the capability of a new product (bio-organic fertilizer) to control the wilt in Fusarium-infested soil. Pot experiments were conducted under growth chamber and greenhouse conditions. The results showed that the fertilizer controlled the wilt disease. Compared with control pots, the incidence rates of Fusarium wilt at 27 and 63 days following treatment of the plants with the bio-organic fertilizer at a rate of 0.5% (organic fertilizer + antagonistic microorganisms, including 3 × 109 CFU g−1 Paenibacillus polymyxa and 5 × 107 CFU g−1 Trichoderma harzianum) were reduced by 84.9 and 75.0%, respectively, in both the growth chamber and greenhouse settings. The activities of antioxidases (catalase, superoxide dismutase and peroxidase) in watermelon leaves increased by 38.9, 150 and 250%, respectively. In the roots, stems and leaves, the activity of β-1,3-glucanase (pathogenesis-related proteins) increased by 80, 1140 and 100% and that of chitinase increased by 240, 80, and 20%, respectively, while the contents of malondialdehyde fell by 56.8, 42.1 and 45.9%, respectively. These results indicate that this new fertilizer formula is capable of protecting watermelon from Fusarium oxysporum f.sp. niveum. The elevated levels of defense-related enzymes are consistent with the induction and enhancement of systemic acquired resistance of plant.

Chemical fertilizers have been used in the cultivation of plants due to their high solubility and effect on crops yield. Biofertilizers with phosphate rock (PR) and potash rock (KR) plus sulfur inoculated with Acidithiobacillus may improve plant growth and contribute to addition of available P and K in soil. The effectiveness of biofertilizers from phosphate and potash rocks mixed with sulfur and Acidithiobacillus was studied in a Typic Fragiuldult soil of the Brazilian Northeast Tableland. Cowpea (cv. “IPA 206”) was grown with and without rhizobia inoculation. Treatments were: (a) phosphate rock (1000 kg ha−1); (b) Biofertilizers-BP (250 and 500 kg ha−1); (c) triple superphosphate-TSP (250 kg ha−1); (d) potash rock (1000 kg ha−1); (e) biofertilizer-BK (250; 500 and 750 kg ha−1); (f) potassium chloride-KCl (250 kg K20 ha−1); (g) control without P or K fertilization (P0K0). The soil was maintained under water submersion covered with black plastic (solarization process) for a period of 30 days. Biofertilizers (Bp and BK) and soluble fertilizers increased plant growth and NPK uptake. Biofertilizers reduced soil pH, especially when applied in highest rates. Biofertilizers and TSP+KCl showed the best values of available P and K in soil. Rhizobial inoculation was effective on cowpea, but no nodules were formed by bacteria native from the soil, probably due to the effect of the solarization process. From obtained PK biofertilizers could be used as alternative for cowpea fertilization in Tableland soils.  相似文献

The effect of glucose addition (0 and 500 μg C g−1 soil) and nitrate (NO3) addition (0, 10, 50 and 500 μg NO3–N g−1 soil) on nitric oxide reductase (cnorB) gene abundance and mRNA levels, and cumulative denitrification were quantified over 48 h in anoxic soils inoculated with Pseudomonas mandelii. Addition of glucose-C significantly increased cnorB p (P. mandelii and related species) mRNA levels and abundance compared with soil with no glucose added, averaged over time and NO3 addition treatments. Without glucose addition, cnorB p mRNA levels were higher when 500 μg NO3–N g−1 soil was added compared with other NO3 additions. In treatments with glucose added, addition of 50 μg NO3–N g−1 soil resulted in higher cnorB p mRNA levels than soil without NO3 but was not different from the 10 and 500 μg NO3–N g−1 treatments. cnorB p abundance in soils without glucose addition was significantly higher in soils with 500 μg NO3–N g−1 soil compared to lower N-treated soils. Conversely, addition of 500 μg NO3–N g−1 soil resulted in lower cnorB p abundance compared with soil without N-addition. Over 48 h, cumulative denitrification in soils with 500 μg glucose-C g−1 soil, and 50 or 500 μg NO3–N g−1 was higher than all other treatments. There was a positive correlation between cnorB p abundance and cumulative denitrification, but only in soils without glucose addition. Glucose-treated soils generally had higher cnorB p abundance and mRNA levels than soils without glucose added, however response of cnorB p abundance and mRNA levels to NO3 supply depended on carbon availability.  相似文献

In this work, we investigated the production of transglutaminase (TGase) by an Amazonian isolated strain of Bacillus circulans by solid-state cultivation (SSC). Several agro-industrial residues, such as untreated corn grits, milled brewers rice, industrial fibrous soy residue, soy hull, and malt bagasse, were used as substrates for microbial growth and enzyme production. Growth on industrial fibrous soy residue, which is rich in protein and hemicellulose, produced the highest TGase activity (0.74 U g−1 of dried substrate after 48 h of incubation). A 23 central composite design was applied to determine the optimal conditions of aeration, cultivation temperature and inoculum cell concentration to TGase production. The best culture conditions were determined as being 0.6 L air min−1, 33 °C and 10 log 10 CFU g−1 of dried substrate, respectively. Under the proposed optimized conditions, the model predicted an enzyme production of 1.16 U g−1 of dried substrate, closely matching the experimental activity of 1.25 U g−1. Results presented in this work point to the use of this newly isolated B. circulans strain as a potential alternative of microbial source for TGase production by SSC, using inexpensive culture media.  相似文献

Thirty single-spore isolates of a toxigenic fungus, Fusarium oxysporum, were isolated from asparagus spears and identified by species-specific polymerase chain reaction (PCR) and translation elongation factor 1-α (TEF) sequence analysis. In the examined sets of F. oxysporum isolates, the DNA sequences of mating type genes (MAT) were identified. The distribution of MAT idiomorph may suggest that MAT1-2 is a predominant mating type in the F. oxysporum population. F. oxysporum is mainly recognised as a producer of moniliformin—the highly toxic secondary metabolite. Moniliformin content was determined by high-performance liquid chromatography (HPLC) analysis in the range 0.05–1,007.47 μg g−1 (mean 115.93 μg g−1) but, also, fumonisin B1 was detected, in the concentration range 0.01–0.91 μg g−1 (mean 0.19 μg g−1). There was no association between mating types and the mycotoxins biosynthesis level. Additionally, a significant intra-species genetic diversity was revealed and molecular markers associated with toxins biosynthesis were identified.  相似文献

A plant growth-promoting rhizobacterium (PGPR) was isolated and identified as Gordonia sp. S2RP-17, which showed ACC deaminase and siderophore synthesizing activities. Its maximum specific growth rate was 0.54 ± 0.12 d−1 at 5,000 mg L−1 of total petroleum hydrocarbon (TPH), and its maximum diesel degradation rate was 2,434.0 ± 124.4 mg L−1 d−1 at 20,000 mg L−1 of TPH. The growth of Zea mays was significantly promoted by the inoculation of Gordonia sp. S2RP-17 in the diesel-contaminated soil. Measured TPH removal efficiencies by various means were 13% by natural attenuation, 84.5% by planting Zea mays, and 95.8% by the combination of Zea mays and Gordonia sp. S2RP-17. The S2RP-17 cell counts were maintained at 1 × 106 CFU g-soil−1 during the remediation period, although they slightly decreased from their initial numbers (2.94 × 107 CFU g-soil−1). These results indicate that rhizoremediation using both Zea mays and Gordonia sp. S2RP-17 is a promising strategy for enhancing remediation efficiency of diesel-contaminated soils.  相似文献

In this study an attempt was made to evaluate the qualitative and quantitative fungal burden (load) in five different working environments of South Assam (India) and the possible risks of indoor fungi to employees and stored products. Fungal concentrations in different working environments were studied using a Burkard personal petriplate sampler. The survey was done in five different working environments for one year. A total of 76 fungal types were recorded in the indoor air of South Assam during the survey period. The maximum fungal concentration (5,437.6 ± 145.3 CFU m−3 air) was recorded in the indoor air of medical wards, followed by the paper-processing industry (3,871.7 ± 93.4 CFU m−3 air). However the lowest concentration was observed in the indoor air of a bakery (1,796.8 ± 54.4 CFU m−3 air). The most dominant fungal genera were Aspergillus (34.2%) followed by Penicillium (17.8%), Geotrichum (7.0%) and the most dominant fungal species were Aspergillus fumigatus (2,650.4 CFU m−3 air) followed by Aspergillus flavus (1,388.2 CFU m−3 air), Geotrichum candidum (1,280.3 CFU m−3 air), Aspergillus niger (783.3 CFU m−3 air), and Penicillium aurantiovirens (774.0 CFU m−3 air). The fungal species viz., Aspergillus fumigatus, Penicillium aurantiovirens, Aspergillus flavus, Aspergillus niger, Geotrichum candidum, and Penicillium thomii, which were recorded well above threshold levels, may lead to adverse health hazards to indoor workers. Setting occupational exposure limits for indoor fungal spores as reference values is obligatory for prevention and control of adverse effects of indoor fungal exposure.  相似文献

Pot trials were carried out under controlled conditions to evaluate the effectiveness against Fusarium wilt of rocket (Fusarium oxysporum f.sp. conglutinans) and basil (Foxysporum f.sp. basilici) of soil amendments based on a patented formulation of Brassica carinata defatted seed meal and compost, combined or not with a simulation of soil solarization. The soil solarization treatment was carried out in a growth chamber by heating the soil for 7 and 14 days at optimal (55–52°C for 6 h, 50–48°C for 8 h and 47–45°C for 10 h/day) and sub‐optimal (50–48°C for 6 h, 45–43°C for 8 h and 40–38°C for 10 h/day) temperatures similar to those observed in summer in solarized soil in greenhouses in Northern Italy. Two subsequent cycles of plant cultivation were carried out in the same soil. Even at sub‐optimal temperature regimes, 7 days of thermal treatment provided very valuable results in terms of disease control on both rocket and basil. In general, the thermal treatment was more effective against F. oxysporum f.sp. basilici than against Foxysporum f.sp. conglutinans. Control of Fusarium wilt of rocket is improved with 14 days of thermal treatment. The combination of organic amendments with a short period of soil solarization (7 or 14 days), although not providing any improvement to the level of disease management, did significantly increase biomass and positively affected yield.  相似文献

Mycelium growth rates, biomass concentration (estimated as glucosamine content) and laccase and endoglucanase secretion were monitored during solid state fermentation (SSF) of wheat straw (WS), reed grass (RG) and bean stalk (BS) residues by Lentinula edodes strains 119, 121, and 122. In a first experiment, these strains were subjected to screening regarding their growth rates and biomass yield, where strain 121 proved to be the fastest colonizer. However, the greater biomass yield at the end of colonization was demonstrated by strain 122 on BS (465.93 mg g−1 d.w.). In a second experiment, growth characters, as well as endoglucanase and laccase production patterns of the selected strains 121 and 122 were monitored at three intervals i.e., at 33, 66, and 100% of substrate colonization. BS furnished the highest endoglucanase production for strain 121, while RG for strain 122. A strain and substrate-dependent behaviour of the enzyme secretion was detected, with strain 122 presenting maximal endoglucanase activity in all substrates at the initial (33%) and final (100%) stages of colonization (0.64–0.90 and 0.79–0.97 U g−1, respectively). However, in strain 121 the peak of endoglucanase production was detected in the early stages of colonization (at 33% on WS and at 66% on RG and BS). Laccase activity showed increased values (maxima on WS, 353.68 and 548.67 U g−1 by strains 121 and 122, respectively) at 66% of colonization. Correlation analysis of growth data demonstrated negative relations between growth rate and biomass yield and between laccase and endoglucanase activities on WS and RG substrates fermented by strain 122. Finally, possible relations of growth parameters with nutritional constituents of the substrates were investigated.  相似文献

Summary Chili pepper (Capsicum annuum L., cv. Tampique?o 74) cell suspensions were employed to study the influence of phenylalanine and phenylpropanoids on the total production of capsaicinoids, the hot taste compounds of chili pepper fruits. The effect of capsaicinoid precursors and intermediates on the accumulation of lignin as an indicator of metabolic diversion was also investigated. Addition of 100 μM of either phenylalanine, cinnamic or caffeic acids to chili pepper cell cultures did not cause significant increases in total capsaicinoids (expressed as capsaicin content, and calculated as averages of the measured values) during the growth cycle. The highest total capsaicinoid content was recorded in cultures grown in the presence of vanillin (142.61 μg g−1 f.wt.), followed by cells treated with 100 μM vanillylamine (104.88 μg g−1 f.wt.), p-coumaric acid (72.36 μg g−1 f.wt.). and ferulic acid (34.67 μg g−1 f.wt.). Capsaicinoid content for control cells was 13.97 μg g−1 f.wt. Chili pepper cell suspensions cultured in the presence of 100 μM of either phenylalanine, or cinnamic, caffeic, or ferulic acids, or the same concentration, of vanillin and vanillylamine, did not exhibit statistically significant differences in the content of lignin as compared with control cells. However, addition of p-coumaric acid (100 μM) to the cultute medium significantly increased thelignin production (c. 10–15 times the contents of control cells).  相似文献

The Fusarium diversity and the mycobiota associated with moldy wheat kernels from Santa Fe province, Argentine, was assessed. The wheat cultivated area in Santa Fe province is divided according to agrometeorological conditions into two zones: Zone I (north-central) and Zone II (south). The natural occurrence of Fusarium toxins BEA, FUP, DON and NIV was also determined. Cladosporium was the most abundant of the 19 genera identified, followed by Fusarium, Phoma and Alternaria. Zone II shows a predominance of F. graminearum and F. culmorum. In Zone I, DON was present in 13/32 samples (range 0.43–3.60 mg kg−1) and NIV in 6/32 samples (range 0.11–0.40 mg kg−1). In zone II, DON was found in 11/21 samples (range 0.57–9.50 mg kg−1) and NIV in 4/21 samples (range 0.10–0.60 mg kg−1). BEA and FP were not detected in both zones.  相似文献

The purpose of the present research is to study the production of thermophilic alkaline protease by a local isolate, Streptomyces sp. CN902, under solid state fermentation (SSF). Optimum SSF parameters for enzyme production have been determined. Various locally available agro-industrial residues have been screened individually or as mixtures for alkaline protease production in SSF. The combination of wheat bran (WB) with chopped date stones (CDS) (5:5) proved to be an efficient mixture for protease production as it gave the highest enzyme activity (90.50 U g−1) when compared to individual WB (74.50 U g−1) or CDS (69.50 U g−1) substrates. This mixed solid substrate was used for the production of protease from Streptomyces sp. CN902 under SSF. Maximal protease production (220.50 U g−1) was obtained with an initial moisture content of 60%, an inoculum level of 1 × 108 (spore g−1 substrate) when incubated at 45°C for 5 days. Supplementation of WB and CDS mixtures with yeast extract as a nitrogen source further increased protease production to 245.50 U g−1 under SSF. Our data demonstrated the usefulness of solid-state fermentation in the production of alkaline protease using WB and CDS mixtures as substrate. Moreover, this approach offered significant benefits due to abundant agro-industrial substrate availability and cheaper cost.  相似文献

Field trials were performed in Florida to evaluate tomato and pepper transplants amended with formulations of several plant growth-promoting rhizobacteria (PGPR) in a production system that included soil solarization. Transplants grown in five different formulations of PGPR were planted into plots treated by soil solarization, MeBr fumigation, or untreated soil. Treatments were assessed for incidence of several naturally occurring tomato and pepper pathogens including root-knot nematode (Meloidogyne incognita) and species of Pythium, Phytophthora, and Fusarium. Highly significant increases in tomato and pepper transplant growth occurred in response to most formulations of PGPR tested. Transplant vigor and survival in the field were improved by PGPR treatments in both tomato and pepper. Diseases of tomato caused by root-knot nematodes, Fusarium, Phytophthora, and Pythium were not affected by PGPR treatments. PGPR formulation LS261 reduced numbers of root-knot nematode galls on pepper while pepper root condition was improved with formulations LS213, LS256 and LS261. Individual PGPR strains affected the number of Pythium colonies isolated from pepper roots, but did not affect isolation of Pythium from tomato roots. Greater numbers of colonies of Pythium were isolated from pepper roots in the MeBr treatment and fewest in the solarization treatment. Numbers of colony forming units of Fusarium were significantly higher in the untreated soil than in MeBr fumigated or solarized soil with no effect of PGPR on isolation of Fusarium from either crop. Incidence of wilt symptoms on tomato was significantly lower in MeBr treated plots and highest in the untreated plots. Yield of extra large tomato fruit and total yield increased with PGPR formulation LS256. Yield of pepper was increased with formulations LS255 and LS256. Solarization combined with LS256 on pepper produced yields comparable to MeBr.  相似文献

Lindane or γ- hexachlorocyclohexane (γ-HCH) is a chlorinated pesticide and its toxic effects on biota necessitate its removal. Microbial degradation is an important process for pesticide bioremediation and the role of soil fungi in recycling of organic matter prompted us to study the biodegradation of lindane using fungi. This study aims at enrichment, isolation and screening of soil fungi capable of metabolizing lindane. Two Fusarium species (F. poae and F. solani) isolated from the pesticide contaminated soil showed better growth on the plates supplemented with lindane as a sole carbon source, when compared with the growth performance of other fungal isolates from the same contaminated soil. However, ANOVA revealed a significant difference in fungal biomass production in both F. poae (F = 22.02; N = 15; P < 0.001) and F. solani (F = 268.75; N = 15; P < 0.001) across different lindane concentrations (0–600 μg ml−1). Growth of both Fusarium sp. was maximum at a lindane concentration of 100 μg ml−1, while minimum at 600 μg ml−1 concentrations. Results on the time dependent release of chlorine by the Fusarium strains in the presence of various concentration of lindane showed the highest mineralization of the pesticide on 10th day of incubation. Time dependent variations in the release of chlorine from 1st to 10th day by both the selected fungal strains were found to be statistically significant. A significant positive relationship exists between fungal biomass increase and chlorine release existed for both F. solani (R2 = 0.960) and F. poae (R2 = 0.628). The results of gas chromatograph analysis of γ- HCH confirmed the biodegradation and utilization of γ- HCH by F. poae and F. solani. The data on lindane degradation by the two fungal strains demonstrated that the biodegradation of lindane by F. solani (59.4%) was slightly higher than that by the F. poae (56.7%).  相似文献

Rhinocladiella similis biodegraded volatile organic compounds (VOCs) of different polarity in gas-phase biofilters. Elimination capacities, (EC) of 74 ghexane m−3 h−1, 230 gethanol m−3 h−1, 85 gtoluene m−3 h−1 and 30 gphenol m−3 h−1 were obtained. EC values correlated with the solubility of the VOCs. R. similis grown with n-hexane or ethanol in biofilters packed with Perlite showed that the surface hydrophobicity was higher with n-hexane than ethanol. The hydrophobin-like proteins extracted from the mycelium produced with n-hexane (15 kDa) were different from those in the ethanol biofilter (8.5 kDa and 7 kDa).  相似文献

The extent of soil microbial diversity in agricultural soils is critical to the maintenance of soil health and quality. The aim of this study was to investigate the influence of land use intensification on soil microbial diversity and thus the level of soil suppressiveness of cucumber Fusarium wilt. We examined three typical microbial populations, Bacillus spp., Pseudomonas spp. and Fuasarium oxysporum, and bacterial functional diversity in soils from three different land use types in China’s Yangtze River Delta, and related those to suppressiveness of cucumber Fusarium wilt. The land use types were a traditional rice wheat (or rape) rotation land, an open field vegetable land, and a polytunnel greenhouse vegetable land that had been transformed from the above two land use types since 1995. Results generated from the field soils showed similar counts for Bacillus spp. (log 5.87–6.01 CFU g−1 dw soil) among the three soils of different land use types, significantly lower counts for Pseudomonas spp. (log 5.44 CFU g−1 dw soil) in the polytunnel greenhouse vegetable land whilst significantly lower counts for Fusarium oxysporum (log 3.21 CFU g−1 dw soil) in the traditional rice wheat (or rape) rotation land. A significant lower dehydrogenase activity (33.56 mg TPF kg−1 dw day−1) was observed in the polytunnel greenhouse vegetable land. Community level physiological profiles (CLPP) of the bacterial communities in soils showed that the average well color development (AWCD) and three functional diversity indices of Shannon index (H′), Simpson index (D) and McIntosh index (U) at 96 h incubation in BIOLOG Eco Micro plates were significantly lower in the polytunnel greenhouse vegetable land than in both the traditional rice wheat (or rape) rotation land and the open field vegetable land. A further greenhouse experiment with the air-dried and sieved soils displayed significantly lower plant growth parameters of 10-old cucumber seedlings as well as significantly lower biomass and total fresh fruit yield at the end of harvesting at day 70 in the polytunnel greenhouse vegetable soil sources. The percentages of Fusarium wilt plant death were greatly increased in the polytunnel greenhouse vegetable plants, irrespective of being inoculated with or without Fusarium oxysporum f. sp. cucumerinum. Our results could provide a better understanding of the effects of land use intensification on soil microbial population and functional diversity as well as the level of soil suppressiveness of cucumber Fusarium wilt.  相似文献

Emissions of N2O and CO2 were measured following combined applications of 15N-labelled fertiliser (100 μg N g−1; 10 atom % excess 15N) and organic olive crop weed residues (Avena sativa, Ononis viscosa, Ridolfia segetum and Olea europea; 100 μg N g−1) to a silt loam soil under controlled environment conditions. The objective was to determine the effect of varying combinations of inorganic fertiliser and plant residues on these emissions and soil mineral N dynamics. Emissions were generally increased following application of residues alone, with 23 ng N2O–N g−1 soil (2 ng N2O–N g−1 soil mg−1 biomass) and 389 μg CO2–C g−1 soil (39 μg CO2–C g−1 soil mg−1 biomass) emitted over 28 days after addition of the Ridolfia residues in the absence of fertiliser-N. N2O emissions from these residue-only treatments were strongly negatively correlated with residue lignin content (r = −0.91; P < 0.05), total carbon content (r = −0.90; P < 0.05) and (lignin + polyphenol)-to-N ratio (r = −0.70; P < 0.1). However, changes in the net input of these compounds through application of 25:75, 50:50 and 75:25 proportional mixtures of Avena and Ononis residues had no effect on emissions compared to their single (0:100 or 100:0) applications. Addition of fertiliser-N increased emissions (by up to 30 ng N2O–N g−1 28 days−1; 123%), particularly from the low residue-N treatments (Avena and Ridolfia) where a greater quantity of biomass was applied, resulting in emissions above that of the sum from the unfertilised residue and fertilised control treatments. In contrast, fertiliser application had no impact on emissions from the Olea treatment with the highest polyphenol (2%) and lignin (11%) contents due to strong immobilisation of soil N, and the 15N–N2O data indicated that residue quality had no effect on the denitrification of applied fertiliser-N. Such apparent inconsistencies mean that before the potential for manipulating N input (organic + inorganic) to lower gaseous N losses can be realised, first the nature and extent of interactions between the different N sources and any interactions with other compounds released from the residues need to be better understood.  相似文献

Impacts of sustainable soil-borne pest management strategies on the soil ecosystem were compared to that of methyl bromide fumigation using nematode community analysis. A field experiment was conducted in 2003 and repeated in 2004. Soil treatments carried out in summer months included methyl bromide (MB) fumigation, solarization (S) for 6 weeks, cowpea (Vigna unguiculata) cover cropping for 3 months (CP), combination of solarization and cowpea cover cropping (S + CP), and a weedy fallow throughout the summer used as a control (C). All treated plots were planted to pepper (Capsicum annuum) after the application of the treatments at the end of the summer. In general, responses of nematode communities to soil treatments were more obvious at pepper planting than at 4 months after planting. In 2003, initial population densities of bacterivores and fungivores at pepper planting followed a hypothesized pattern of MB > S > S + CP > CP > C. However, this perturbation did not persist after a cycle of vigorous growth of a pepper crop. Omnivorous nematodes were the most sensitive nematode trophic group, with impact from soil treatment lasting until the end of the pepper crop. Nematode community indices such as ratio of fungivores to fungivores plus bacterivores, richness, and structure index were especially useful in detecting impacts by the various soil treatments. While disturbance from MB on the nematode communities lasted at least until the end of the pepper crop, that from the solarization often reduced or disappeared at the end of the experiment. The CP treatment enhanced many of the beneficial nematodes but failed to suppress the final population densities of herbivorous nematodes at pepper harvest (Pf). However, CP + S consistently reduced the Pf of herbivores to levels equivalent to MB in both years, whereas, this level of suppression could not be achieved by either CP or solarization alone.  相似文献

Streptomyces rimosus CN08 isolated from Tunisian soil produced 8.6 mg l−1 of oxytetracycline (OTC) under submerged fermentation (SmF). Attempts were made for enhancing OTC production after irradiation-induced mutagenesis of Streptomyces rimosus CN08 with Co60-γ rays. 125 OTC-producing colonies were obtained after screening on kanamycin containing medium. One mutant called Streptomyces rimosus γ-45 whose OTC production increased 19-fold (165 mg l−1) versus wild-type strain was selected. γ-45 mutant was used for OTC production under solid-state fermentation (SSF). Wheat bran (WB) was used as solid substrate and process parameters influencing OTC production were optimized. Solid-state fermentation increased the yield of antibiotic production (257 mg g−1) when compared with submerged fermentation. Ammonium sulphate as additional nitrogen source enhanced OTC level to 298 mg g−1. Interestingly, OTC production by γ-45 mutant was insensitive to phosphate which opens the way to high OTC production even in medium containing phosphate necessary for optimal mycelia growth.  相似文献
