Arteriosclerosis in Oncorhynchur mykiss has been examined histologically in parr, smolts, spawning adults and post-spawning adults captured during downstream migration. All stages of fish from the Clearwater River in Idaho are characterized by the presence of lesions in the main vessel on the bulbus arteriosus. The incidence is lowest in parr, increases significantly in smolts, and is present in 100% of spawning fish. Regression of lesions was not observed in either hatchery or wild fish after spawning. 相似文献

Environmental influences on cardiovascular variables in rainbow trout, Oncorhynchus mykiss (Walbaum)
Two groups of wild (lakedwelling and anadromous), and a group of hatchery-reared Oncorhynchus mykiss (Walbaum) (rainbow trout) were sampled in order to measure cardiac morphometrics, haemoglobin concentration, and the DNA and protein concentration in cardiac muscle. A combination of these variables was used to distinguish wild fish from domestic ones.
The wild fish had significantly higher levels of haemoglobin (for male fish, 10.10 and 10.07 g 100 ml−1 vs. 7.69 g d−1 ) and larger relative ventricle mass (females, 0.091 and 0.099% ofbody mass vs. 0.073%; males, 0.108 and 0.134% vs. 0.102%; immatures, 0.086 and 0.094% vs. 0, 072%, respectively) than the domestic fish. The anadromous and domestic fish had significantly higher amounts of compact tissue when compared with lake fish (females, 43 and 47% of ventricle mass vs. 34%, respectively). Ventricle size distinguished wild fish from domestic fish, except male anadromous and male domestic fish which were distinguished only by haemoglobin and compact tissue values. Immature fishes from all groups had lower total protein levels in the ventricle, lower compact tissue levels, and less haemoglobin. Points regarding the potential environmental influences in determining these cardiovascular trends are discussed. 相似文献

Uptake of vitellogenin into oocytes during early vitellogenic development in the rainbow trout, Oncorhynchus mykiss (Walbaum) 总被引:1,自引:0,他引:1
Uptake of 3 H-vitellogenin (3 H-VTG) into oocytes of various sizes was investigated during early vitellogenic development in the rainbow trout, Oncorhynchus mykiss (Walbaum). Females were injected with 3 H-VTG and uptake into oocytes of different sizes (<0.4,0.4–0.59, 0.6–0.79, 0.8–0.99 and 1.0 1.2 mm in diameter) measured. Oocytes measuring less than 0.6 mm in diameter appeared unable to sequester VTG and were therefore considered pre-vitellogenic. Oocytes measuring 0.6 mm or more all sequestered VTG. The larger the oocyte, the more 3 H-VTG it sequestered, even when uptake was expressed per unit surface area. The latter observation could be due to an increase in the number of VTG receptors per unit surface area, an increase in the rate of turnover of the VTG receptor, greater access of VTG to the receptors as oocytes grow, or a combination of any of these factors. The data suggest that the ability to sequester VTG is developmentally regulated. 相似文献

Blood lysozyme activity was determined in rainbow trout that were stressed by handling and transport, or suffered from acute ammonia poisoning. To optimize the lysoplate assay, the pH dependence of the lysozyme activity was studied. For serum and kidney lysozyme, the pH optimum was 5.5 to 6.0. Ventilation frequency and plasma glucoseconcentration of the fish were used as stress indicators. Less severely stressful situations, such as 30 min of handling, brought about either a stimulation or a suppression of the lysozyme activity. Strong stressors, such as transport lasting 2 h or acute water pollution, reduced the lysozyme level significantly. The decrease of lysozyme activity due to handling and transport persisted for 24 h, but the activity returned to normal within 2 weeks following confrontation with the stressor. 相似文献

Relationship between stress, feeding and plasma ghrelin levels in rainbow trout, Oncorhynchus mykiss

Sexually immature rainbow trout were acclimated to small-volume (1 m3) holding tanks and then exposed to short-term stress to examine the relationship between feeding, stress, plasma ghrelin levels and other plasma stress parameters. Plasma ghrelin levels showed an increase 24 h after a single feed, plasma lactate and glucose levels decreased over the same period and plasma cortisol levels were low and constant. One hour of confinement stress resulted in elevations of plasma cortisol, glucose and lactate and depression of plasma ghrelin levels. In a separate experiment, 2 h of confinement stress also depressed feeding immediately after stress, concomitant with increases in plasma cortisol, lactate and glucose; however, in this case there was no change in plasma ghrelin concentrations. A repeat of the 2-h confinement experiment using fish that had not been acclimated to small-volume holding tanks produced a more marked elevation in plasma cortisol and a stronger suppression of feeding post-stress but in this case also, there was no change in plasma ghrelin levels. The results of this study confirm that feeding in rainbow trout is suppressed by confinement stress although the effect is transitory in this domesticated stock. Similar to that in other fishes, plasma ghrelin levels appear to be modulated by feeding status and may be influenced by stress, suggesting an orexigenic role for ghrelin in rainbow trout. 相似文献

Rainbow trout were fitted with latex masks for the measurement of ventilatory flow rate (V?w) and with opercular catheters for sampling expired water from close to the branchial surface. Fish were exposed for 6 h to pH 5.1, 4.7 or 4.1 in the presence (138 μg l 1) or nominal absence (1 μg 1–1) of Al. Inspired and expired Al concentrations and water pH were measured via the opercular catheters. Gills were sampled for accumulated Al at the end of the experiments. V?w increased during Al exposures at all three pHs. However, in the absence of Al, V?w increased only at pH 4.1. Aluminium extraction from the water and Al accumulation on the gills were highest at inspired pH 5.1 and lowest at pH 4.1, and correlated well with expired pH, which was 0.2–0.7 pH units higher than inspired pH. Gill Al accumulations amounted to only about one-tenth of deposition calculated from Al extraction from the water and V?w, and gill Al was tightly bound to the branchial surface. Calculations of Al solubility, oversaturation, and species composition were made using measured expired pH values, and were compared with Al extraction from the water and measured gill Al concentrations. In general, these analyses indicated that reduced Al solubility near the gills is a reasonable explanation of Al extraction from the water, and that Al(OH)2+ and Al(OH)3° are the Al species most likely to interact initially at the gills. It is suggested that mucus sloughing removes most precipitated Al, and that only the charged form persists, bound to structural elements on the gill surface. A model incorporating these results, and pH changes in the fish gill micro-environment in general, is presented to explain previously-reported ionoregulatory and respiratory effects of Al. 相似文献

Trout, Oncorhynchus mykiss , were exposed to water from the Rhine for 24 days and their skin examined by light and electron microscopy. Relative to control fish mitotic figures were common and seen throughout the epidermis. Pavement cells in fish exposed to Rhine water contained significantly more secretory vesicles than control fish. Necrotic pavement cells were apparent after 24 h, and apoptotic cells from day 4 on. Mucous secretion was intense and the differentiation of mucous cells was stimulated. Some of these cells synthesized mucus of high electron density, probably of a serous composition. Leucocytes invaded the dermis and epidermis, and towards the end of the experiment many apoptotic and necrotic lymphocytes were found. In the dermis fibroblasts were abundant and actively producing collagen. Pigment containing cytoplasmic extensions of melanocytes penetrated into the epidermis. After 14 and 24 days of exposure many pigment cells, melanocytes, iridocytes and xanthocytes became apoptotic. Most of these changes are known from fish exposed to heavy metals, acid water or other stressful treatments, indicating that exposure to Rhine water is a stressful experience for trout. 相似文献

Past studies on palatability perception and the chemical requirements of feeding in rainbow trout have focused on the amino acids. The stimulatory effectiveness of other classes of compounds remains largely unknown. In the present study representative members from several chemical classes were tested for their palatability to rainbow trout. Three heretofore untested amino acids (L-norvaline, L-norleucine and c-aminocaproic acid), three amides (acetamide, valeramide and caproic amide), two alcohols (hexanol and octanol) and four sugars (o-ribose, o-glucose, D-fructose and sucrose) were all mildly to highly palatable substances at their highest concentration tested. All other compounds tested within these classes did not evoke a positive behavioural response, demonstrating that rainbow trout are selective in thosecompounds deemed as palatable. Moreover, all of the aldehydes, trimethylamine, pyrrolidine and all of the amines except the alcoholic derivative, ethanolamine, were without effect. A provisional scheme for relating chemical structure versus the perception of palatability in rainbow trout is presented. 相似文献

A high oral concentration of 10g Al kg−1 dry weight promoted toxicant accumulation in the muscle, gill, liver, kidney and mucus of trout. One mortality occurred and only a small proportion of the exposure dose was retained by the fish. Plasma levels of cortisone, a steroid hormone of potential physiological significance in fish, have rarely been measured. This study examines the interrelationship between circulating levels of cortisone and the major teleost corticosteroid, cortisol, in the blood of two strains of rainbow trout subject to confinement stress, a condition know to stimulate corticosteroidogenic activity. In unstressed fish from both strains, mean plasma cortisol levels were within the range 0.4–7.5 ng ml−1 . Mean plasma cortisone levels were within the range 7.1–15.9 ng ml−1 . Plasma cortisol levels were elevated within 5 min of the onset of strees and reached peak values within 45 min, although there was a marked difference betweed the maxima observed in the two strains (strain 1:70 ng ml−1 ; strain 2:150 ng ml−1 ). The rate of increase of plasma cortisone levels during strees was more rapid than that of cortisol, maximum values (strain 1:100ng ml−1 ; strain 2:160 ng ml−1 ) being reached within 10 to 20 min of the onset of stress. This rapid stress-induced elevation of plasme cortisone has not previously been reported in fish. We suggest that rapid conversion of cortisol to cortisone during the initial response to stress accounts for the appearance of large amounts of cortisone in the blood, indicating that circulating for the appearance of large amounts of cortisone in the blood, indicating that circulating levels of cortisol alone do not fully reflect the secretory activity of the interregnal during the initial of cortisol alone do not fully the secretory activity of the interregnal during the initial phase of the stress response. The results also indicate that the rate of clearance of cortisone from the circulation may be a major factor in determining stress-stimulated levels of plasma cortisol. 相似文献

Carnobacterium maltaromaticum B26 and Carnobacterium divergens B33, which were isolated from the intestine of healthy rainbow trout (Oncorhynchus mykiss, Walbaum), were selected as being potentially useful as probiotics with effectiveness against Aeromonas salmonicida and Yersinia ruckeri. Thus, rainbow trout administered with feed supplemented with B26 or B33 dosed at >10(7) cells g(-1) feed conferred protection against challenge with virulent cultures of the pathogens. Moreover, both cultures persisted in the gut for up to 3 weeks after administration. The cultures enhanced the cellular and humoral immune responses. Specifically, fish fed with B26 demonstrated significantly increased phagocytic activity of the head kidney macrophages, whereas the use of B33 led to significant increases in respiratory burst and serum lysozyme activity. Also, the gut mucosal lysozyme activity for fish fed with both cultures was statistically higher than the controls. 相似文献

Morphological studies on the effect of low pH on egg shells (chorion) of rainbow trout, Oncorhynrhus mykiss Walbaum (formerly Salmo gairdneri ), and brown trout, Salmo trutta f. fario L., showed that the plugs of the outer layer of the chorion, which block the channels of the underlying layer, developed distinctive vacuolization at low pH exposure. This may lead to loss of chorional semi-permeability. Chorions were incompletely degraded at low pH inducing hatching problems. Partially hatched alevins showed only a small local zone of dissolution of the chorion around the head of the embryo preventing the larvae from being freed. 相似文献

Drinking was measured by the rate of uptake of tritiated dextran in fresh water by the alevins and fry of rainbow trout at various stages of development during a 40-day post-hatch period. Drinking increased almost S-fold during the initial 16 day yolk-sac stage. Drinking rate increased most between 16 and 23 days, the transitional period between yolk-sac absorption and first feeding. The maximum weight-specific drinkingrateof 3.24μlg−1 h−1 recorded for 40-day-old fry was higher than previously recorded for adults. Abrupt transfer from an adaptation temperature of 10 to 19° C increased drinking significantly (Q10 = 1.2), but sudden transfer to 1 1‰ salinity sea water caused a substantial fall in drinking rate in 23-day fry. A 24-h period of adaptation to 1 1‰ and 15‰ salinity restored drinking to a rate similar to that in fresh water. The water drunk by 31-day fry fed to satiation was initially higher than by unfed fry, but drinking rates subsequently fell below the control level. The results are discussed in terms of osmoregulation and the uptake of dissolved or suspended substances by the intestinal tract for the purpose of immunization. 相似文献

The present study investigates the effect of cannulation and chronic'black-box' confinement, as well as epinephrine administration (4–0 μg kg−1 ), on the degree and time-course of alterations in trout ( Oncorhynchus mykiss ) catecholamine and cortisol concentrations. Plasma cortisol concentrations in seawater trout acclimated to 3–6° C reached 104 ng ml−1 1 day after cannulation/confinement and remained elevated above resting levels (8 ng ml−1 ) until 6 days post-confinement. Although plasma epinephrine and norepinephrine generally declined over the period of confinement (day 1 approx. 12 nM; day 7 approx. 6 nM), norepinephrine titres were usually higher and more variable. Epinephrine injection caused elevations in plasma epinephrine levels but not in norepinephrine levels; epinephrine titres reaching 107 ± 26 nM (range 65–238 nM) at 2 min post-injection and returning to pre-injection levels by 30 min post-injection. Plasma cortisol increased by 20 ng ml−1 following epinephrine administration. Based on the time-course for post-confinement alterations in plasma cortisol, it appears that up to a week may be required before cannulated fish are completely acclimated to 'black-box' confinement. The findings suggest that meaningful results from experiments utilizing epinephrine injection and 'black-box confinement are contingent upon: (1) knowledge of circulating epinephrine levels shortly after injection (i.e. within 2 min post-injection); and (2) an experimental design that takes into account the elevated cortisol titres that are inherent with cannulation/confinement and epinephrine injection. 相似文献

Rainbow trout, Oncorhynchus mykiss, were subjected to either physical stress (twice daily chasing to exhaustion for 5 days) or a period of 2 months of fasting. Following these treatments, the levels of catecholamines, adrenaline and noradrenaline, stored within the kidney and posterior cardinal vein (PCV) were determined. The ability of the catecholamine-storing chromaffin cells to release catecholamines in response to cholinergic stimulation was measured using an in situ saline-perfused PCV preparation. In the physically stressed fish, the concentration (μg catecholamine g?1 tissue) of noradrenaline within the anterior and middle thirds of the kidney increased; the concentration of adrenaline was unchanged in all tissues. The content (μg) of noradrenaline or adrenaline, within the various tissues, was similar in both groups of fish with the exception of a higher noradrenaline content in the middle third of the kidney in the physically stressed fish. The total catecholamine content (μg catecholamine) of these tissues (kidney+PCV) was unaffected by physical stress. With the exception of a lower concentration of adrenaline in the middle third of the kidney, the concentrations of catecholamines were unaffected by fasting. There was a trend towards a greater content (μg) of noradrenaline within all of the tissue regions of the fed fish, however, a significant difference was only observed in the anterior third of the kidney. The content of adrenaline in the fed fish was greater in all regions of the kidney as well as the middle third of the PCV. The total catecholamine content (kidney + PCV) was lower in the fasted fish owing to significantly lower PCV and kidney masses. Prolonged physical stress caused a decrease in the responsiveness of the chromaffin cells to the cholinoceptor agonist carbachol (10?8 to 10?4mol). The ED50 (the dose of carbachol required to elicit a half maximal response) for catecholamine release was 0·96 ± 10?6mol carbachol in the physically stressed fish and 0·84 ± 10?7 in the control fish. Fasting did not alter the pattern of catecholamine release. The ED50 values were 0·96 ± 10?7 and 1·24 ± 10?7 mol for fasted and fed fish, respectively. Thus, a physical stress affected both catecholamine storage and release whereas fasting affected only storage and not the release process. 相似文献

In this study the pronephros of rainbow trout Oncorhynchus mykiss was explanted and cultured. The morphology of cultured cells suggested they were leukocyte derived. To confirm this, cells were incubated with non-opsonized fluorescent beads and human apoptotic polymorphonuclear leukocytes (PMN) to establish that they were capable of phagocytosis and that they became activated following exposure to apoptotic bodies. The cells were studied using light microscopy, transmission electron microscopy and a live cell observer system. Cells in culture were also stained with phalloidin to identify actin reorganization following activation. Cultured cells ingested the inert beads and apoptotic PMNs and c . 80% of cells became activated following exposure to the apoptotic PMN, evidenced by enhanced filopodial extensions. This methodology may play a role in future studies, in particular, the competence of macrophages following exposure to radiation can now be tested. 相似文献
