Physical education students were subjected to electrical stimulation of relatively high frequency and current amplitude for 19 days. A quantitative study of several morphological parameters was performed on biopsy samples from gastrocnemius, using stereological methods at both light and electron microscopic levels. The main results were: muscle fibre size was increased; nuclear volume was also increased, suggesting that a proliferation of nuclei had occurred; this was paralleled by an increased content of nuclear DNA. The size of single myonuclei was increased, and their heterochromatin fraction was decreased, these changes being most pronounced in type II fibres. The increase in the mitochondrial fraction was also greatest in type II fibres. It is concluded that this type of electrical stimulation has predominant effects on type II fibres.  相似文献

The electrical activity of different muscle fibre types during fatigue at varying stimulation frequency and fibre stretch was studied. Extracellular action potentials (ECAPs) were recorded from isolated frog muscle fibres at initial length and stretched by 15%, 25% and 35% and stimulated for 180 s by suprathreshold pulses with frequencies of 5, 6.7 and 10Hz. The changes in ECAP negative phase duration (T(0)), propagation velocity of excitation (PV), potential power spectrum and its median frequency (MDF) were analysed for the period of uninterrupted activity (endurance time, ET). Slow (SMF) and fast (FMF) fatigable muscle fibre types were distinguished by the rate of PV decrease during ET. With the increase of stimulation frequency and fibre stretch, the rate of ECAP parameter changes increased and was larger in FMF, but this proportion was reversed with stretching over 25% and 10Hz stimulation. In both fibre types the power spectrum shift to lower frequencies during continuous activity was more pronounced with higher stimulation frequency. In FMFs the rates of MDF changes were positively and more strongly correlated with the rates of PV changes, whilst in SMFs the inverse correlation between the rates of changes of MDF and T(0) was stronger. The results indicate specific adaptation of slow and fast fatigable muscle fibres to stretch and activation frequency due to the differences in their membrane processes.  相似文献

In anesthetized, artificially ventilated cats with open chest, bilateral stimulation of all afferent vagal fibres (pulse duration: 800 microseconds, 30 Hz, train duration 30 to 40 s) produced marked changes in the spontaneous EEG activities in the primary somatosensory cortex (Sl area). They were characterized by depressed background rhythms, with a tendency to desynchronization, decreased amplitude and number of spindles, with altered pattern, and/or evoked sustained fast rhythmic activities. These effects occurred within 1 to 5 sec during vagal stimulation. On the contrary, the EEG response was weaker or absent when only myelinated vagal afferents were stimulated (100 microseconds). I.v. injection of phenyldiguanide (PDG), used for stimulation of unmyelinated vagal sensory fibres and mainly of pulmonary afferents, induced EEG changes within the first 30 s, similar to those observed during electrical vagal stimulation. These EEG responses were unrelated to the induced hypotension. Cervical bivagotomy produced persistent changes in EEG activity, with enhancement of the magnitude, duration and number of spindles, which resembled the delayed effects induced by PDG. The present results obtained with three test agents (electrical or chemical vagal stimulation and bivagotomy) demonstrated that, in cats, vagal afferent information interacted with the spontaneous EEG rhythms in the Sl area.  相似文献

Acetylcarnitine formation during intense muscular contraction in humans   总被引:9,自引:0,他引:9  
To study the changes in carnitine during intense muscular effort subjects underwent 4 min intermittent electrical stimulation of the quadriceps femoris muscle and on a separate occasion performed 4 min exercise on a bicycle ergometer. Biopsies of the vastus lateralis muscle were taken at rest and after 2 and 4 min of stimulation or exercise. Resting mean muscle total carnitine content was 20.0 mmol/kg dry muscle. Approximately 77% was free carnitine and 19% acetylcarnitine. Four minutes of stimulation or intense exercise did not effect total carnitine but did result in a marked fall in free carnitine and almost equivalent rise in acetylcarnitine. The results indicate that acetylcarnitine is a major metabolite formed during intense muscular effort and that carnitine may function in the regulation of the acetyl-CoA/CoA ratio by buffering excess production of acetyl units.  相似文献

This study was designed to examine the time-course of response to inhibition of fatty acid (FA) oxidation in rats rendered mildly diabetic with streptozotocin and fed a high fat diet (50% of energy derived from fat). Etomoxir, a specific carnitine palmitoyltransferase (CPT-1) inhibitor, was administered subcutaneously (12.5 mg/kg) to inhibit long chain fatty acid oxidation. Diabetic and non-diabetic control rats were maintained on the high fat diet. Following an overnight fast, glucose, free fatty acid (FFA) and triglyceride (TG) concentrations were determined after three days, one week and four weeks of treatment. The effect of Etomoxir treatment in reducing fasting glucose concentrations was not evident until after one week, while fasting FFA and TG concentrations were already reduced after three days treatment. All of these changes were maintained over the four week period (P less than 0.001), resulting in reduced levels of fasting plasma glucose (17.6 +/- 2.4 vs 22.3 +/- 1.9 mmol/l), fasting plasma TG (0.32 +/- 0.07 vs 0.98 +/- 0.14 mmol/l) and fasting serum FFA (1.52 +/- 0.26 vs 3.51 +/- 0.69 mEq/l). In addition, the improvements in glucose and lipid levels were accompanied by restored rates of growth towards that of non-diabetic control rats. These results suggest that the short term inhibition of FA oxidation improves fasting glucose, FFA and TG concentrations in diabetic rats fed a high fat diet.  相似文献

The influences of denervation and of direct electrical stimulation of denervated muscle upon the post-hatching differentiation of fibre types in the fast avian muscle posterior latissimus dorsi have been investigated. Denervation inhibits the normal decrease in number of muscle fibres exhibiting acid-stable myofibrillar ATPase activity and leads to weak oxidative activity in all the fibres. Direct stimulation at a low rhythm of denervated muscle induces the normal decrease of fibres exhibiting acid-stable myofibrillar ATPase but does not allow the occurrence of normal oxidative activity pattern. The results emphasize the role of muscular activity upon the differentiation of fibre types in a developing muscle.  相似文献

Diminished sensitivity of hepatic carnitine palmitoyltransferase to inhibition by malonyl-CoA in the fasting and diabetic states is a well-recognized aspect of the regulatory mechanism forhepatic fatty acid oxidation. Inhibition of myocardial carnitine palmitoyltransferase by malonyl-CoA may play an important role in regulation of fatty acid oxidation in the heart, but there has been a discrepancy in data relating to changes in malonyl-CoA sensitivity of the myocardial carnitine palmitoyltransferase during fasting. Analysis of malonyl-CoA inhibition of myocardial carnitine palmitoyltransferase in fasting and fed states under a variety of conditions has indicated that under no condition could any difference be found in malonyl-CoA sensitivity that was attributable to fasting. Proteolysis of the outer carnitine palmitoyltransferase led to artifactual changes in sensitivity due to the appearance of partial inhibition. We have concluded that the sensitivity of myocardial carnitine palmitoyltransferase to malonyl-CoA does not change during fasting. 1-Carnitine was administered to fed rats and the changes in plasma beta-hydroxybutrate concentration and liver acid-insoluble acylcarnitine content were assessed. One hour following injection of carnitine in doses greater than 1 mumol/100 g of body weight there was a dose-dependent increase in liver acid-insoluble acylcarnitine content to levels comparable to those seen in fasting. These increased levels were maintained for a least 2 h following injection. During the period following carnitine administration there was no increase in ketogenesis as evidenced by plasma beta-hydroxybutyrate concentrations. Since acid-insoluble acylcarnitines represent the product of carnitine palmitoyltransferase A, the results are interpreted as contradictory to the theory that this enzyme is rate-limiting and regulatory for ketogenesis.  相似文献

In adult male rats, a pretreatment regimen of serial injections of dexamethasone (1 mg/kg), morphine (20 mg/kg) and pentobarbital (40 mg/kg) was evaluated for use in conjunction with studies on the effects of hypothalamic electrical stimulation on prolactin secretion. Serum prolactin levels were measured before and 15 min after electrical and sham stimulation of the hypothalamic arcuate nucleus in rats subjected to either the pharmacological regimen or to pentobarbital anesthesia alone. Pentobarbital alone caused a transient rise in serum prolactin levels, which obscured any effect of electrical or sham stimulation; this interference was not overcome by the addition of dexamethasone and morphine treatment. Thus, the result indicate that the acute stimulatory affect of pentobarbital anesthesia on prolactin release may interfere with further manipulation of prolactin-controlling mechanisms, by either pharmacological or surgical means. Furthermore, the dexamethasone-morphine-pentobarbital pretreated rat does not provide an adequate preparation for studing the effects of electrical stimulation on prolactin secretion.  相似文献

A differential distribution of carnitine was found in twelve microdissected areas of the frozen canine brain. The cerebellar cortex showed the highest level of carnitine, while the lowest amounts were found in the hippocampus and pons sections of the brain. However, the inability to demonstrate changes in carnitine levels after in vivo stimulation of the major afferent pathways in the caudate nucleus, and failure to demonstrate K+ stimulated release of carnitine in rat brain slices suggest that carnitine does not have a direct neurotransmitter role in the brain.  相似文献

Slow-tonic anterior latissimus dorsi (ALD) muscle properties were studied following chronic spinal cord stimulation in chick embryo. Stimulation at a fast rhythm was applied from day 7, 8 or 10 of development until the end of embryonic life. When stimulation was applied from day 7 up to day 18 of development, ALD muscle exhibited at day 18 a large decrease in half time to peak of tetanic contraction, a large proportion of fast type II fibres and an increase in fast myosin light chain content as compared to control muscle. When stimulation started at day 8 of development, changes in properties of ALD muscle were reduced when compared to the previous experimental series. Indeed, no fast type II fibres were observed within the muscle, even when stimulation was prolongated until the 20th day of embryonic development. In addition, chronic stimulation at a fast rhythm initiated at day 10 of development did not modify ALD muscle differentiation. The present results indicate that a fast pattern of motor neurone activity can induce some slow-to-fast transformations of ALD muscle fibres. However, after the first week of embryonic life, ALD myotubes appeared refractory to these transformations. The possible mechanisms responsible for the transformation of slow myotubes and for their further loss of plasticity are discussed.  相似文献

The use of ether anesthesia as a technique for obtaining liver tissue and plasma from living rats was compared to decapitation followed by in situ sample freezing. Parameters associated with fatty acid metabolism were examined to demonstrate differences in the two methodologies. Use of ether in fasted rats was associated with lower plasma free fatty acids and acetoacetate concentrations, as well as decreased hepatic total carnitine, carnitine, and acid-soluble acylcarnitine content when compared to decapitated animals. No qualitative differences between the ether and decapitated groups were noted, and the quantitative differences observed were consistent with effects induced by the anesthetic. Thus, with respect to in vivo studies of fatty acid metabolism use of anesthetic agents may induce significant changes without providing advantages over decapitation and in situ freezing.  相似文献

In skeletal muscle from a patient with a mitochondrial myopathy and muscular carnitine deficiency, histochemical analysis demonstrated that mitochondrial ATPase showed activation with loss of latency even before addition of the uncoupler dinitrophenol (DNP). According to combined histochemical and biochemical studies by Meijer and Vloedman (1980), this finding indicates loosely coupled oxidative phosphorylation. After the addition of DNP the reaction intensity was markedly increased, but there were scattered enzyme-deficient fibres in which some residual activity was shown by ultracytochemistry. No defect in mitochondrial enzymes was found in biochemical studies. The enzyme histochemical changes and carnitine deficiency are probably both secondary to an unknown mitochondrial defect. Both the carnitine deficiency and the mitochondrial myopathy remained unchanged following long-term carnitine substitution therapy despite clinical improvement.  相似文献

全身麻醉若操作不当可能造成致命的中枢神经系统损伤,因此其安全性受到广泛关注.为了揭示麻醉不断加深的过程中神经元活动的变化规律,本文研究了大鼠在乌拉坦(urethane)深度麻醉至脑死亡期间海马区神经元兴奋性和信号传导功能的变化.利用微电极阵列记录和电刺激技术,在海马CA1区胞体层分别记录Schaffer侧支上正向刺激和海马白质上反向刺激诱发的群峰电位(population spike,PS).以PS的幅值和潜伏期为指标,分析海马神经元活动的变化.结果表明,随着乌拉坦血药浓度的增加,PS幅值逐渐减小,潜伏期逐渐延长,意味着乌拉坦抑制了神经元的兴奋性以及轴突传导和突触传递.特别是这些变化存在明显的转折点(即突变),将整个衰减过程分成慢变和快变2个阶段.快变期的剧烈衰减迅速导致脑死亡.而且,引起突变的决定性因素可能是乌拉坦的血药浓度,而非麻醉时间的长短.但是,当乌拉坦注射速率较慢时,延长的慢变期仍然会使神经元功能的受损加重.这些研究结果为动物实验的麻醉操作和临床麻醉的安全应用提供了重要的信息.  相似文献

Implantable electronic stimulators were used to subject fast-twitch tibialis anterior and extensor digitorum longus muscles of adult rabbits to a chronically increased level of use. Stimulation was discontinued after 6 weeks and physiological, histochemical and biochemical properties of the muscles were examined at intervals over the ensuing 20 weeks. Previous work had shown that 6 weeks of stimulation was sufficient to bring about a substantial transformation of type in fast-twitch muscles, which then exhibited much of the character of muscles of the slow-twitch type. The present experiments showed that these stimulation-induced changes were completely reversible. The time-course of reversion was such that the muscles had recovered their original fast properties by about 12 weeks after the cessation of stimulation. The contractile characteristics and post-tetanic potentiation typical of fast muscle returned rapidly, in only 3-4 weeks, and over the same period the proportion of histochemical type 1 fibres declined from about 70% to control levels. Changes in fatigue-resistance, capillary density and enzyme activity followed a more prolonged time-course; in particular, the decline in the activity of enzymes of oxidative metabolism corresponded closely to that already established for the mitochondrial volume fraction. Reacquisition of fast properties was not accompanied by any changes in specific force-generating capacity. Observations from these experiments and from a related morphological study fit into a 'first-in, last-out' pattern for the response to stimulation and recovery. The slow-to-fast reversion that takes place during the recovery period provides a further opportunity for testing causal associations within the events underlying type transformation. It has important consequences for therapeutic applications that make use of the fatigue-resistant character of chronically stimulated muscle.  相似文献

Summary This report describes a quantitative histochemical study of myoglobin in skeletal muscle fibres. The muscle fibres were classified as fast or slow on the basis of their quantitative myofibrillar ATPase histochemistry. A large range of myoglobin absorbance values was found among fast skeletal muscle fibres. This range was relatively small among slow fibres. The concentrations of myoglobin and the activities of succinate dehydrogenase in individual muscle fibres in serial sections are weakly correlated in both the mouse soleus and plantaris muscle. The myoglobin concentration is higher in fast and slow oxidative soleus muscle fibres and the succinate dehydrogenase activity in these fibres is lower than in oxidative plantaris muscle fibres in the same range of cross-sectional area.  相似文献

The rodent whisker sensory system is a commonly used model of cortical processing; however, anesthetics cause profound differences in the shape and timing of evoked responses. Evoked response studies, especially those that use spatial mapping techniques, such as fMRI or optical imaging, will thus show significantly different results depending on the anesthesia used. To describe the effect of behavioral states and commonly used anesthetics, we characterized the early surface-evoked response potentials (ERPs) components (first ERP peak: gamma band 25-45 Hz; fast oscillation: 200-400 Hz; and very fast oscillation: 400-600 Hz) using a 25-channel electrode array on the somatosensory cortex during whisker stimulation. We found significant differences in the ERP shape when ketamine/xylazine, urethane, propofol, isoflurane, and pentobarbital sodium were administered and during sleep and wake states. The highest ERP amplitudes were observed under propofol anesthesia and during quiet sleep. Under isoflurane, the ERP was nearly absent, except for a very late component, which was concombinant with burst synchronization. The slowest responses were seen under urethane and propofol anesthesia. Spatial mapping experiments that use electrical, NMR, or optical techniques must consider the anesthetic dependency of these signals, especially when stimulation protocols or electrical and metabolic responses are compared.  相似文献

The effects of a single oral dose of carnitine on fasting-induced ketosis was investigated in four normal individuals, five patients with muscular dystrophy, and one patient with a generalized cytochrome c oxidase deficiency. Plasma carnitine, free fatty acids, glucose, insulin, and glucagon were also measured. Normal individuals showed an average 0.09 mM increase in blood beta-hydroxybutyrate concentration during a 12- to 18-hr period of fasting and carnitine administration did not affect this response (average: 0.12 mM). Muscular dystrophy patients showed a greater fasting-induced elevation in beta-hydroxybutyrate (average 0.29 mM) and carnitine administration greatly enhanced this ketogenic response (average 0.84 mM). The cytochrome c oxidase deficient patient showed an even larger increase in beta-hydroxybutyrate with fasting (1.67 mM) and carnitine further augmented this ketotic effect (3.78 mM). Plasma free fatty acids were also elevated in patients that showed enhanced ketosis. Plasma glucagon concentration did not change, but insulin levels decreased during the 12- to 18-hr period of fasting; no major differences were found between controls and patients. These results indicate that some patients with muscular dystrophy and cytochrome c oxidase deficiency are more prone to develop ketosis than normal individuals and that carnitine administration enhances this response. Since both muscular dystrophy patients and the patient with cytochrome c oxidase deficiency had similar ketogenic responses, the data suggest that ketone body utilization may be impaired in these patients. The ability of L-carnitine to be ketogenic should be considered in the treatment of these patients.  相似文献

In fasted rodents hepatic carnitine concentration increases considerably which is not observed in PPAR alpha-/- mice, indicating that PPAR alpha is involved in carnitine homeostasis. To investigate the mechanisms underlying the PPAR alpha-dependent hepatic carnitine accumulation we measured carnitine biosynthesis enzyme activities, levels of carnitine biosynthesis intermediates, acyl-carnitines and OCTN2 mRNA levels in tissues of untreated, fasted or Wy-14643-treated wild type and PPAR alpha-/- mice. Here we show that both enhancement of carnitine biosynthesis (due to increased gamma-butyrobetaine dioxygenase activity), extra-hepatic gamma-butyrobetaine synthesis and increased hepatic carnitine import (OCTN2 expression) contributes to the increased hepatic carnitine levels after fasting and that these processes are PPAR alpha-dependent.  相似文献
