3H-ABA was introduced into the xylem stream of maize ( Zea mays}) leaves on intact plants by incubation of a semi-attached flap of the sheath in solutions. The relative contribution of exportation and metabolism to the fate of xylem-delivered ABA was assessed in leaves which were either kept at different water potentials through soil drying treatments or subjected to different xylem pHs (pH 7.4 vs. pH 5.5) through a phosphate buffer in the feeding solutions. Xylem-delivered ABA was rapidly metabolised in well-watered leaves with a half-life of 2.19 h in the relatively mature leaves used in this study. Re-exportation of xylem-delivered ABA from leaves was much slower than metabolism. It took 24 h for half of the fed radioactivity to disappear from the well-watered leaves, and very possibly this radioactivity was in the form of metabolites of fed 3H-ABA. Although soil drying usually increases the output of ABA through phloem as reported in previous studies, it greatly reduced the re-exportation of xylem-fed ABA and/or its metabolites. Metabolism was also significantly reduced by the treatment of soil drying (half-life extended from 2.19 to 3.63 h), although the magnitude of change was much less than that of exportation. Manipulation of the pH in the feeding solution also had its effect on the re-exportation. A shift of pH from 5.5 to 7.4 reduced the rate of disappearance of the total radioactivity fed into the attached leaves, but showed no significant effect on the rate of ABA metabolism. It was concluded that it was the ABA metabolism, rather than a re-exportation from leaves, which was mainly responsible for the disposal of the ABA signal from the xylem and therefore preventing an accumulation in leaves. Water stress and pH increase of xylem sap would increase the time of such ABA's presence in the leaves. Since xylem-imported ABA is unlikely to be re-exported from leaves in its intact form, we believe a recycling of ABA from xylem to phloem through leaves plays only a minor role.  相似文献

When detached maize leaves were fed with an ABA solution viathe xylem, the relationship between the relative stomatal inhibitionand ABA concentrations was similar under different humidityconditions, but the relationship between such inhibition andABA flux was different according to changes of humidity. Tounderstand whether such stomatal behaviour was related to theway through which xylem-delivered ABA was metabolized, detachedleaves of maize and Commelina were fed with tritium-labelled(3H)-ABA at concentrations similar to that found in xylem ofdroughted plants and it was found that xylem-delivered ABA wasmetabolized rapidly in both species. The half-life of ABA metabolism,calculated from the time-related ABA disappearance curve, was42 and 64 min for maize and Commelina, respectively. The veryshort half-life suggests that there is a large capacity in leavesto metabolize xylem-delivered ABA and that metabolism is a majorfactor in the control of ABA accumulation in leaves. When ABAwas fed at different fluxes, either through changing the feedingconcentrations or through manipulating the rates of leaf transpiration(i.e. the volume flux), ABA was metabolized at rates that wereproportional to the amount that was delivered. The absoluterate of ABA metabolism was, therefore, linearly related to theamount of ABA that had arrived. It was found that xylem-deliveredABA reached the epidermis of Commelina, and was metabolizedat the same pattern as that in mesophyll tissues, i.e. at asimilar half-life and at rates constantly related to the amountthat was delivered. The role of the rapid ABA metabolism wasdiscussed in the context of stomatal control by either concentrationor flux of xylem-carried ABA. Metabolism and distribution of xylem-fed ABA were investigated in leaves of maize (Zea mays) and Commelina communis when water stress and xylem pH manipulation were applied. 3H-ABA was fed to excised leaves via the transpiration stream. Water stress was applied through either a previous soil-drying before leaves were excised, or a quick dehydration after leaves were fed with ABA. Xylem-delivered ABA was metabolised rapidly in the leaves (half-life 0.7 h and 1.02 h for maize and Commelina respectively), but a previous soil-drying or a post-feeding dehydration significantly extended the half-life of fed ABA in both species. In the first few hours after ABA was fed into the detached leaves, percentages of applied ABA remaining unmodified were always higher in leaves which received water stress treatments than in control leaves. However the percentage decreased to below the control levels several hours later in leaves which received a previous soil-drying treatment prior to excision, but had then been rehydrated by the xylem-feeding process itself. One possible explanation for this could be a changed pattern of compartmentalisation for xylem-carried ABA. A post-feeding dehydration treatment also changed the distribution of xylem-fed ABA within the leaves: more ABA was found in the epidermis of Commelina leaves which had been dehydrated rapidly after ABA had been fed, compared to the controls. The levels of xylem-delivered ABA remaining unmodified increased as the pH of the feeding solution increased from 5 to 8. The results support the hypothesis that water stress and a putative stress-induced xylem pH change may modify stomatal sensitivity to ABA by changing the actual ABA content of the leaf epidermis.  相似文献

Maize (Zea mays L.) was grown in quartz sand culture eitherwith a normal root system (controls) or with seminal roots only(‘single-rooted’). Development of adventitious rootswas prevented by using plants with an etiolated mesocotyl andthe stem base was positioned 5–8 cm above the sand. Eventhough the roots of the single-rooted plants were sufficientlysupplied with water and nutrients, the leaves experienced waterdeficits and showed decreased transpiration as trans plrationalwater flow was restricted by the constant number of xylem vesselspresent in the mesocotyl. As a consequence of this restriction,transpirational water flow velocities in the metaxylem vesselsreached mean values of 270 m h–1 and phloem transportvelocities of 5.2 m h–1. Despite limited xylem transportmineral nutrient concentrations in leaf tissues were not decreasedin single-rooted plants, but shoot and particularly stem developmentwas somewhat inhibited. Due to the lack of adventitious rootsthe shoot:root ratio was strongly increased in the single-rootedplants, but the seminal roots showed compensatory growth comparedto those in control plants. Consistent with decreased leaf conductance,ABA concentrations in leaves of single-rooted plants were elevatedup to 10-fold, but xylem sap ABA concentrations in these plantswere lower than in controls, in good agreement with the well-wateredconditions experienced by the seminal roots. Surprisingly, however,ABA concentrations in tissues of the seminal roots of the single-rooted plants were clearly increased compared to the controls,presumably due to increased ABA import via phloem from the water-stressedleaves. The results are discussed in relation to the role ofABA as a shoot to root signal. (rice) seedlings growing in saline conditionsexhibit pronounced gradients in leaf sodium concentration whichis always higher in the older leaves than the younger ones.Individual leaf transpiration rates have been investigated todiscover whether movement of sodium in the transpiration streamis able to explain these profiles from leaf to leaf. The useof 14C labelled ethane diol to estimate transpiration was evaluatedby direct comparison with values obtained by gas exchange measurements.Ethane diol uptake was linearly related to the transpirationalvolume flow and accurately predicted leaf to leaf gradientsin transpiration rate in saline and non-saline conditions. 14C-ethanediol and 22NaCl were used to compare the fluxes of water andsodium into different leaves. The youngest leaf showed the highesttranspiration rate but the lowest Na accumulation in salineconditions; conversely, the older leaves showed the lower transpirationrates but the greater accumulation of Na. The apparent concentrationof Na in the xylem stream was 44 times lower into the youngerleaf 4 than into the older leaf 1. Exposure to NaCl (50 molm–3) for 24 h elicited an increase in endogenous ABA inthe oldest leaf only, but no significant changes occurred inthe younger leaves. The robustness of a leaf elongation bioassay was evaluated byconducting trials with detached shoots of wheat at several differenttemperatures. Leaf elongation rate (LER) was monitored for shootsfed either an artificial xylem solution or xylem solution plus10–3mol m–3 abscisic acid (ABA). Consistent resultswere obtained when periodic ruler measurements of many shootswere made and compared with simultaneous measurements on a singleshoot made with a linearly variable displacement transducer(LVDT). ABA treatment consistently inhibited leaf growth; however,the magnitude of the inhibition was dependent on the temperatureat which the assay was conducted. Interpretation of resultsfrom such bioas-says in terms of ABA concentration suppliedto the detached shoots is complicated by this observation sincethere is no unique relationship between leaf growth inhibitionand ABA concentration. The uptake and transport of salt ions (Na+, Cl-), macronutrients (K+, Ca2+, Mg2+) and abscisic acid (ABA) response to increasing soil salinity were examined in 2-year-old seedlings of Populus euphratica and a hybrid, P. talassica Kom 2 (P. euphratica + Salix alba L.). Leaf burn symptoms appeared in the hybrid after 8 days of exposure to salinity when soil NaCl concentration increased to 206 mM, whereas P. euphratica exhibited leaf damage after day 21 when soil NaCl exceeded 354 mM. Leaf necrosis was the result of excess salt accumulation since the injury followed an abrupt increase of endogenous salt levels. Compared with the hybrid, P. euphratica exhibited a greater capacity to exclude salt ions from leaves under increasing salinity, especially Cl-. Salt treatment altered nutrient balance of the hybrid, leaf K+, Ca2+ and Mg2+ concentrations significantly declined and the same trends were observed in roots with the exception of K+. Although K+ levels decreased in salinised P. euphratica, increasing salinity did not affect the levels of Ca2+ and Mg2+ in leaves, but did increase the uptake of these nutrients when salt stress was initiated. NaCl-induced increase of ABA concentration in xylem sap [ABA] was observed in the two tested genotypes, however xylem [ABA] increased more rapidly in P. euphratica and a fivefold increase of xylem [ABA] was recorded after the first day of exposure to salt stress. Therefore, we conclude that the increase of Ca2+ uptake may be associated with the rise of ABA, and thus contributes to membrane integrity maintenance, which enables P. euphratica to regulate uptake and transport of salt ions under high levels of external salinity in the longer term.  相似文献

The abscisic acid (ABA)-deficient mutant of barley, Az34, exhibiteda much reduced rate of leaf expansion at a bulk density of 1.6g cm–3 as compared to the isogenic wild-type variety,Steptoe. Az34 had a consistently lower xylem sap ABA concentrationat 7 d and 14 d after emergence (DAE). The xylem sap data suggestthat ABA present at Steptoe concentrations may have a directrole in maintaining leaf expansion at the sub-critical bulkdensity (1.6 g cm–3 To test this hypothesis, additionof synthetic ABA either to the rooting environment (100 nM)or directly to the xylem sap (5 pg µl–1 to reproducethe xylem sap ABA concentrations found in Steptoe, increasedleaf expansion in Az34 to the wild-type level. Furthermore,feeding Steptoe xylem sap to Az34 produced similar effects.These experiments provide direct evidence of a positive rolefor ABA as a root-to-shoot signal which assists in maintainingleaf growth in plants experiencing subcritical levels of compactionstress. Wild-type (Steptoe) and abscisic acid (ABA)-deficient mutant(Az34) genotypes of barley were grown in compacted soil to examinethe potential role of ABA as a root-to-shoot signal. Root andshoot growth and leaf conductance were all reduced when plantswere grown in compacted soil with a bulk density of 1.7g cm–3,relative to uncompacted control plants (1.1 g cm–3. Theseeffects occurred in the absence of detectable changes in leafwater status or foliar abscisic acid (ABA) content. Analysisof Steptoe and Az34 xylem sap showed that the ABA concentrationwas greatly increased at 6 d after emergence (6 DAE) when seedlingswere grown in compacted soil (1.7 g cm–3); however, ABAconcentrations were never as high in the mutant as in the wildtype. The increase in xylem sap ABA concentration observed athigh bulk density was closely correlated with reductions inleaf conductance, but not leaf area. These increases were transitory,and xylem sap ABA concentrations subsequently decreased towardsthe control level by 18 DAE in both genotypes. The ABA-deficient mutant, Az34, produced a much lower leaf areathan Steptoe at a bulk density of 1.6 g cm–3. Examinationof epidermal characteristics indicates that this effect resultedmainly from reductions in cell expansion rather than cell division,suggesting that the higher ABA concentrations detected in xylemsap from the wild-type Steptoe may have exerted a positive rolein maintaining leaf expansion in this treatment. The possibleinvolvement of ABA as a root-to-shoot signal mediating the effectsof compaction stress is discussed. Seedlings of Ricinus communis L. were cultivated in quartz sandand supplied with media which contained either different concentrationsof nitrate or ammonium nitrogen and were treated with a lowsalt stress. The concentration of ABA was determined in tissuesand in xylem and phloem saps. Between 41 and 51 day after sowing,abscisic acid (ABA) flows between roots and shoots were modelled.Long-distance transport of ABA was not stimulated under conditionsof nitrate deficiency (0.2 mol m–3). However, when ammoniumwas given as the only N source (1.0 mol m–3), ABA transportin both xylem and phloem was increased significantly. Mild saltstress (40 mol m–3 NaCl) increased ABA transport in nitrate-fedplants, but not in ammonium-fed plants. The leaf conductancewas lowered by salt treatment with both nitrogen sources, butit was always lower in ammonium-fed compared to nitrate-fedplants. A negative correlation of leaf conductance to ABA levelsin leaves or flow in xylem was found only in comparison of ammonium-fedto nitrate-fed plants. The distribution of radioactivity from applied sucrose.14C and32P to various plant parts were studied in relation to the retardationof leaf senescence by applied benzyladenine (BA) in intact beanplants. In short-time experiments sucrose-14C was fed to theplants for 48 h through the second trifoliate leaf at weeklyintervals from the third to the eighth week after planting.In long-term experiments sucrose-14C was fed to all plants for48 h at the third week and changes in distribution examinedat weekly intervals up till the eight week. In both cases, BAapplied to the primary leaves of intact bean plants did notcause a directed mobilization of sucrose-14C. When the plantswere stripped, leaving the primary leaves and the terminal pod,and fed sucrose-14C or 32P through the terminal leaflet of thesecond trifoliate leaf, the BA-treated leaf accumulated relativelymore radioactivity than the opposite water-treated leaf. Itwas concluded that the retardation of senescence by BA in theprimary leaves of intact bean plants does not result directlyfrom the mobilization of metabolites and nutrients from otherplant parts. It is therefore suggested that BA-induced longevityin the primary leaves of the intact plant is accomplished bymetabolic self-sustenance.  相似文献

Drought increased the pH of Commelina communis xylem sap from 6.1 to 6.7. Conductances of transpiring leaves were 50% lower in pH 7.0 than in pH 6.0 buffers, but bulk leaf abscisic acid (ABA) concentration and shoot water status were unaffected by pH. Stomatal apertures of isolated abaxial epidermis incubated on simple buffers increased with external pH, so in vivo this must be overridden by alternative pH effects. Reductions in leaf transpiration rate at pH 7.0 were dependent on the presence of 10-8 mol dm-3 ABA in the xylem stream. We inferred that at pH 7.0 leaf apoplastic ABA concentrations increased: pH did not affect distributions of ABA among leaf tissues, but isolated epidermis and mesophyll tissue took up more 3H-ABA from pH 6.0 than from pH 7.0 buffers. The apoplastic ABA increase at pH 7.0 may result from reduced symplastic sequestration. A portion of 3H-ABA uptake by the epidermis was saturable at pH 6.0 but not at pH 7.0. An ABA uptake carrier may contribute to ABA sequestration by the leaf symplast of well-watered plants, and its inactivity at pH 7.0 may favor apoplastic ABA accumulation in draughted plants. Effects of external pH on stomatal apertures in the isolated epidermis indicate that published data supporting a role for internal guard cell ABA receptors should be reassessed.  相似文献

Plants of Helianthus annuus were grown in soil in pots suchthat approximately 30% of the root system protruded throughthe base of the pot. After 7 d further growth in aerated nutrientsolution, the attached, protruding roots were air-dried for10–15 min and thereafter surrounded with moist still air,in the dark, for 49 h, whilst the soil was kept at field capacity.The roots of the control plants remained in the nutrient solutionthroughout the experiment. This treatment rapidly reduced the water content of protrudingroots from 20.5 to 17.8 g g–1 dry mass (DM), which remainedless than that of the control roots for the rest of the experiment.This treatment also reduced root turgor and water potential.The abscisic acid (ABA) concentrations in the protruding roots,xylem sap and leaves of the treated plants increased significantly,compared to values recorded for control plants. In treated roots, the ABA concentration was significantly increased4 h after treatment, with a maximum of 4.4+0.1 nmol g–1(DM) after 25 h. The ABA concentration in the xylem sap of thetreated plants was significantly greater than in the controls25 h, 30 h, and 49 h after the partial drying of the roots,with a maximum concentration of approximately 970 pmol ABA cm-3at 49 h. Initially, the ABA concentration in the leaves was0.45 nmol g–1 (DM) which increased significantly to 1.1±0.1 nmol g–1 at 25 h, to 1.7±0.3 nmol g–1at 49 h. Leaf conductance was significantly less in plants with air-driedroots than in the controls 8 h after the start of the treatmentand thereafter. The water relations of the leaves of the treatedplants did not differ from those of the control plants. These results confirm previous reports that ABA is rapidly generatedin partially-dried and attached root systems and demonstratesa concomitant large increase in the ABA content of the xylemsap. It is suggested that partial dehydration of some of theroots of Helianthus annuus, increases ABA concentration in thetranspiration stream and decreases leaf conductance in the absenceof changes in leaf water status. As these responses were initiatedin free-growing roots the stimulus is independent of any increasesin soil shear strength that are associated with soil drying. The characteristics of ABA-induced changes in the fluxes ofCO2 and water vapour from whole leaves of spring wheat (Triticumaestivum cv. Wembley) were examined. Aqueous solutions of ABAwere supplied via the transpiration stream to intact leavesof different ages mounted within a gas exchange cuvette. ABA caused a reduction in stomatal conductance (g) that wasproportional to the concentration in the solution fed to theleaf. For the maintenance of a reduction in g there was a requirementfor a continual supply of ABA. At concentrations greater than10–2 mol m–3 ABA reduced g by at least 50% of thecontrol value, while 1.0 mol m–3 closed stomata within2 h. Concentrations as low as 10–3 mol m–3 produceda 20% reduction in g. As leaves aged they became less responsiveto applied ABA. The possibility that the stomatal response may change aftera leaf has previously experienced a pulse of ABA was exploredby repeating the exposure of a leaf to 10–2 mol m–3ABA. The first pulse of ABA produced a greater reduction ing than a subsequent exposure the following day. This declinein response of g to ABA on repeated exposure was maintainedwith leaves of different ages. The characteristics of the stomatal response to ABA are discussedin the context of what is known about the location of receptorsfor the hormone. It seems likely that a failure to respond toABA that has previously accumulated in the guard cells shouldbe viewed by means of maximizing the sensitivity to the currentsupply of ABA. It is suggested that the smaller response ofthe stomata of older leaves to ABA makes them more susceptibleto water stress, so that they can act as sensors for decliningwater potentials to give early protection to younger, metabolicallyactive leaves. ABA Levels and Effects in Chilled and Hardened Phaseolus vulgaris   总被引:3,自引:0,他引:3  
Leaf abscisic acid (ABA) levels of chilled P. vulgaris weremeasured after 18 h chilling at 5°C, at a saturation deficitof 1.24 g m–3 (SD), and after chilling in a water-saturatedatmosphere. Changes were also followed during a chill hardeningperiod of 4 d at 12°C, 2.1 g m–3 SD. It was foundthat hardening resulted in an almost 5. fold increase in ABAlevels after 3 d at 12°C, and this decreased to approximatelycontrol levels on the fourth day. Subsequent chilling of hardenedplants produced no change in ABA levels from that of controlplants (22° C). In contrast, non-hardened plants chilledat 1.24 g m–3 SD had ABA levels almost 3 times the levelof control plants. However, chilling in a water-saturated atmosphereresulted in a decrease in ABA levels. In addition, the response of leaf diffusion resistance (LDR)to exogenous ABA fed via the transpiration stream was measuredat 5 ° C and 22° C in hardened and non-hardened plants.Use of tritium-labelled ABA was made to calculate the stomatalsensitivity to ABA. It was found that exogenous ABA caused anincreased in LDR at 22°C in both hardened and non-hardenedplants. However, the sensitivity of the hardened plants to ABAwas greater in terms of rate of closure and amount of ABA requiredto close the stomata. At 5°C, however, ABA caused stomatalopening and the maintainance of open stomata in non-hardenedplants. In hardened plants, ABA caused stomatal closure at 5°C.These results are discussed in relation to the locking-openresponse of chilled P. vulgaris stomata. Using modified compartmental analysis the unidirectional fluxesof abscisic acid (ABA) and their cytoplasmic and vacuolar contentsin 3H-ABA preloaded barley root segments (Hordeum distichoncv. Aura) have been studied. When root segments were stressedosmotically with sorbitol (osmotic potential of the media 0= 0.2 MPa) cytoplasmic and vacuolar contents of ABA were enhanced.Under increased stress cytoplasmic and vacuolar contents weremuch lower than in the unstressed controls. ABA fluxes werevery sensitive to osmotic stress and ABA transport from thecytoplasm of the xylem parenchyma to the xylem vessels (cx)was rapidly inhibited. The cultivar Aura has higher cytoplasmicand vacuolar ABA contents than the barley cultivar Kocherperle.This correlates well with the higher stress tolerance of theAura cultivar. A model of maize stomatal behaviour has been developed, in which stomatal conductance is linked to the concentration of abscisic acid ([ABA]) in the xylem sap, with a sensitivity dependent upon the leaf water potential (Ψ1). It was tested against two alternative hypotheses, namely that stomatal sensitivity to xylem [ABA] would be linked to the leaf-to-air vapour pressure difference (VPD), or to the flux of ABA into the leaf. Stomatal conductance (gs) was studied: (1) in field-grown plants whose xylem [ABA] and Ψ1 depended on soil water status and evaporative demand; (2) in field-grown plants fed with ABA solutions such that xylem [ABA] was artificially raised, thereby decreasing gs and increasing Ψ1 and leaf-to-air VPD; and (3) in ABA-fed detached leaves exposed to varying evaporative demands, but with a constant and high Ψ1. The same relationships between gs, xylem [ABA] and Ψ1, showing lower stomatal sensitivity to [ABA] at high Ψ1, applied whether variations in xylem [ABA] were due to natural increase or to feeding, and whether variations in Ψ1, were due to changes in evaporative demand or to the increased Ψ1 observed in ABA-fed plants. Conversely, neither the leaf-to-air VPD nor the ABA flux into the leaf accounted for the observed changes in stomatal sensitivity to xylem [ABA]. The model, using parameters calculated from previous field data and the detached-leaf data, was tested against the observations of both ABA-fed and droughted plants in the field. It accounted with reasonable accuracy for changes in gs (r2 ranging from 0.77 to 0.81). These results support the view that modelling of stomatal behaviour requires consideration of both chemical and hydraulic aspects of root-to-shoot communication.  相似文献

Differences in maximum leaf conductance in grapevine plants growing in soils with contrasting water availabilities during mid-summer in Portugal could be accounted for by differences in the concentration of ABA in xylem sap. This conclusion is reinforced by the observation that the relationship between leaf conductance and endogenous ABA concentration can be mimicked by the application of exogenous ABA to leaves detached from irrigated plants. During the day, leaf conductance decreased after a morning peak, even when the leaves remained in a constant environment at a moderate temperature and leaf-to-air vapour pressure difference. This decline in leaf conductance was not a consequence of an increase in the xylem ABA concentration or the rate of delivery of this compound by the transpiratory stream. The afternoon depression in leaf conductance was associated with an apparent limitation in stomatal opening potential, which persisted even when detached leaves were fed with water and rehydrated. Rooting volume restriction (RVR) reduces shoot growth of plantsprovided with sufficient water or nutrients. The effects ofRVR on water status, abscisic acid (ABA) levels in leaves, roots,or xylem sap from detopped plants of watermelon [Citrullus lanatus(Thunb.) Matsum. and Nakai ‘StarBrite’] seedlingswere evaluated with five rooting volumes (18, 26, 36, 46, or80 cm3). Shoot water potential increased with increasing rootingvolume, with no difference between plants from 18 and 26 cm3cells or between plants from 36 and 46 cm3 cells. Stomatal conductancewas not consistently affected by RVR; at 10 and 20 DAE, stomatalconductance in plants grown in 36 cm3 cells was higher thanthat of plants grown in any other cell volume. Severe RVR (18and/or 26 cm3) tended to produce plants with higher ABA levelsin roots (15 DAE only), xylem sap (all dates), and leaves (5and 10 DAE). Plants grown in 18 and 26 cm3 cells had higherroot ABA levels than those from 46 and 80 cm3 cells at 15 DAE.Plants grown in 18 cm3 cells had the highest xylem sap ABA levelat all dates, but ABA levels did not differ among plants grownin the other cell volumes. Plants grown in 18 cm3 cells at 5DAE and 18 and 26 cm3 cells at 10 DAE also had higher leaf ABAlevels than those from other rooting volumes.
