Genotypic variation in response of barley to boron deficiency   总被引:2,自引:0,他引:2  
Responses of a range of barley (Hordeum vulgare L.) genotypes to boron (B) deficiency were studied in two experiments carried out in sand culture and in the field at Chiang Mai, Thailand. In experiment 1, two barley genotypes, Stirling (two-row) and BRB 2 (six-row) and one wheat (Triticum aestivum L.) genotype, SW 41, were evaluated in sand culture with three levels of applied B (0, 0.1 and 1.0 μM B) to the nutrient solution. It was found that B deficiency depressed flag leaf B concentration at booting, grain number and grain yield of all genotypes. In barley Stirling, B deficiency also depressed number of spikes plant-1, spikelets spike-1 and straw yield. However, no significant difference between genotypes in flag leaf B concentration was found under low B treatments. Flag leaf B concentration below 4 mg kg-1 was associated with grain set reduction and could, therefore, be used as a general indicator for B status in barley. In experiment 2, nine barley and two wheat genotypes were evaluated in the field on a low B soil with three levels of B. Boron levels were varied by applying either 2 t of lime ha-1 (BL), no B (B0) or 10 kg Borax ha-1 (B+) to the soil prior to sowing. Genotypes differed in their B response for grain spike-1, grain spikelet-1 and grain set index (GSI). The GSI of the B efficient wheat, Fang 60, exceeded 90% in all B treatments. The B inefficient wheat SW 41 and most of the barley genotypes set grain normally (GSI >80%) only at the B+. In B0 GSI of the barley genotypes ranged from 23% to 84%, and in BL from 19% to 65%. Three of the barley with severely depressed GSI in B0 and BL also had a decreased number of spikelets spike-1. In experiment 3, 21 advanced barley lines from the Barley Thailand Yield Nursery 1997/98 (BTYN 1997/98) were screened for B response in sand culture with no added B. Grain Set Index of the Fang 60 and SW 41 checks were 98 and 65%, respectively, and GSI of barley lines ranged between 5 and 90%. One advanced line was identified as B efficient and two as moderately B efficient. The remaining lines ranked between moderately inefficient to inefficient. These experiments have established that there is a range of responses to B in barley genotypes. This variation in the B response was observed in vegetative as well as reproductive growth. In 1990 and 1991 grain samples of spring barley were obtained from plots in which fungal diseases were controlled until ear emergence using fungicide sprays, and from comparable plots which had received no fungicide applications. The grain was stored at 10oC, and tested for germination at intervals during storage. Grain from plots treated with fungicide consistently exhibited more dormancy than grain from untreated plots. In 1992, grain development was monitored from anthesis to harvest-ripeness in treated and untreated plots of cvs Blenheim, Camargue and Tyne. In cvs Blenheim and Camargue, grains in fungicide-treated plots had a greater dry weight at harvest-ripeness, and dehydrated later in development than grains in untreated plots. Neither of these differences was observed in cv. Tyne. Possible interpretations of the effects of fungicide treatment of the parent plant on the physiology of the grain are discussed.  相似文献

Abstract  While mites are able to utilize numerous food sources, the suitability of the food strongly influences population growth. The different suitabilities of various stored agricultural products will thus affect the level of infestation. In this study, we compared field mite infestation rates in two stored cereals: wheat and barley. We analyzed mite abundance, frequency and species composition in samples of grain obtained from 79 selected Czech grain stores. Stored barley seemed to be more vulnerable to mite attack than wheat, as we consistently found more infested samples, more species and higher mean and median mite abundance per sample in barley as compared to wheat. The mean mite abundance per sample were 55 and 506 individuals for wheat and barley, respectively. In barley, 10% of samples exceeded allergen risk threshold (i.e., 1 000 individuals per kg of grain). Altogether, 25 species were identified from approximately 35 000 individuals. The most frequently identified species were the same in wheat and barley, that is, Tydeus interruptus Sig Thor, Acarus siro L., Tarsonemus granarius Lindquist, Lepidoglyphus destructor (Schrank) and Tyrophagus putrescentiae (Schrank). Based on principal components analysis, we found a closer association of T. interruptus , T. putrescentiae , L. destructor and Cheyletus eruditus (Schrank) with barley samples, corresponding to the high frequency and abundance values of these mites. The probable reasons for the higher infestation, especially mite abundance in barley, are discussed in relation to the higher proportion of crushed parts, which may release favorable nutrient sources and amplify the abundance values.  相似文献

Two genetically related wheat lines growing in cabinets were given different temperatures during grain filling, and abscisic acid (ABA) was measured in whole grains by gas chromatography with an electron-capture detector. Three genetically related barley lines grown in the field were assayed for ABA content in endosperm and embryo fractions separately by radiommunoassay.Maximum grain growth rate and final weight per grain of the two wheat lines differed by 50–60% at low temperature and 30–40% at high temperature. During grain development two peaks in ABA level were observed at low temperature but only one at high temperature. At times when differences in grain growth rate between genotypes and between temperature treatments were large, the corresponding differences in ABA concentration were small. In barley, one line (Iabo 14) had 30% heavier grains than the other two (Onice and Opale). Endosperm ABA concentrations showed no clear differences between genotypes until grain filling was nearly complete. Embryo ABA levels were up to 10-times greater than those in the endosperm, with Opale having significantly less ABA in the embryo than the other two cultivars.Our experiments did not provide evidence for a causal relationship between ABA levels during grain filling and grain growth rate or final weight.Abbreviations ABA Abscisic acid - DAA days after anthesis - DW dry weight - FW fresh weight  相似文献

The application of the protein and polysaccharide fractions of barley spent grain as a basis of growth media for probiotic bacteria was studied. High values of biomass yield, cell viability, and organic acid production were observed in the variants of media containing the barley spent grain supplemented with lactose, ascorbic acid, yeast extract, and mineral salts. Colonization of barley grain by Aspergillus flavus and formation of aflatoxin B1 in the presence of Penicillium verrucosum, Fusarium sporotrichioides, and Hyphopichia burtonii were studied over a three-week period in all combinations of 20 or 30 °C and 0.97, 0.95 or 0.90 aw. Grain colonization was assessed initially by observing hyphal extension on the grain surface, using scanning electron microscopy, and then from the proportion of seeds infected and numbers of colony forming units (cfu) formed. Aflatoxin b1 concentrations were determined by enzyme linked immunosorbent assay using a monoclonal antibody. These studies showed that interaction between A. flavus and other fungi in paired culture had different effects on both colonization and aflatoxin formation depending on the species involved and environmental conditions. Germination of A. flavus spores was unaffected by the presence of other species on the grain surface. Subsequently, three principal patterns of A. flavus colonization of barley grain were observed through the incubation period in the presence of other fungal species: (a) colonization unaffected by the presence of other species; (b) colonization initially slower in the presence of other species but later differing little from pure cultures; and (c) colonization adversely affected by the presence of other species. Five main patterns of aflatoxin B1 production were observed relative to pure culture but with no consistent relationship with species, aw, temperature or incubation period; (a) little changed; (b) increased slowly; (c) decreased; (d) enhanced; and (e, f) increased initially but later decreased to (e) the same level as in pure culture or (f) to less than in pure culture. Generally, production of aflatoxin B1 by A. flavus was less than in pure culture but sometimes was changed only slightly by the presence of P. verrucosum, F. sporotrichioides or H. burtonii or was temporarily enhanced.  相似文献

Considerable genetic variation exists in zinc (Zn) efficiency among cereal species and genotypes within the same species. Currently, the mechanisms of Zn efficiency are not understood well; however, the research so far suggests that overall Zn efficiency can be partitioned into uptake, utilisation and translocation or remobilisation efficiency, all or some of which collectively determine the level of Zn efficiency in a particular genotype. In a growth room study, using two barley genotypes differing in Zn efficiency (Zn-efficient Unicorn and Zn-inefficient Amagi Nijo), we attempted to determine which of these components of Zn efficiency contributed to greater Zn efficiency in Unicorn, by examining growth responses to Zn over a wide range of Zn fertilisation rates (0, 0.05, 0.2, 0.8, 3.2 and 12.8 mg Zn/kg soil) during the entire growth period. Zn-efficient Unicorn showed less severe Zn deficiency symptoms, produced more dry matter, and grain yield under Zn deficient conditions compared with Zn-inefficient Amagi Nijo. These responses also varied with the level of Zn deficiency stress and growth stage. Most importantly, the greater Zn efficiency (e.g., ability to grow well under Zn deficiency) at maturity of Unicorn was due to greater translocation of Zn from vegetative to reproductive organs or greater ability to produce higher grain yield with limited Zn rather than Zn uptake from soil which was similar in both genotypes. Zn-efficient Unicorn also had a lower critical deficiency concentration for grain (12 mg Zn/kg DW) than the Zn-inefficient Amagi Nijo (18 mg Zn/kg DW), suggesting a lower requirement for metabolic processes in Zn-efficient Unicorn. The critical deficiency concentration in the grain has the potential to diagnose Zn-deficient soils. The results also show that grain Zn concentration can be increased by Zn fertilisation, with significant increases occurring above the Zn fertilisation rate that is adequate for production of grain. However, genetic variation in grain Zn concentration should be explored and wild relatives of barley may offer potential for crop improvement for this trait.  相似文献

Grain texture in barley is an important quality character as soft-textured cultivars have better malting quality. In wheat, texture is considered to be determined by the puroindolines, a group of basic hydrophobic proteins present on the surface of the starch granule. Hard wheats have been proposed to lack puroindoline a or to have mutant forms of puroindoline b which do not bind to the granule surface. Analysis of six barley cultivars (three soft-textured and three hard) showed that all contained proteins homologous to wheat puroindoline b, but PCR analysis failed to show any differences in amino acid sequences similar to those which have been proposed to determine textural differences in wheat. Southern blot analysis showed two hordoindoline b genes which were isolated and shown to encode proteins with 94% sequence identity. Expression of hordoindoline b mRNA occurred in the starchy endosperm and aleurone layer of the developing seed, but not in the embryo. Analysis of seven soft- and six hard-textured barley varieties showed that all contained hordoindoline a except two hard varieties (Sundance, Hart) which were subsequently shown to both lack hordoindoline a mRNA. It was therefore concluded that there is not a clear relationship between the presence of hordoindoline a and grain texture in barley.  相似文献

Most plants accumulate silicon in their bodies, and this is thought to be important for resistance against biotic and abiotic stresses; however, the molecular mechanisms for Si uptake and accumulation are poorly understood. Here, we describe an Si influx transporter, HvLsi1, in barley. This protein is homologous to rice influx transporter OsLsi1 with 81% identity, and belongs to a Nod26-like major intrinsic protein sub-family of aquaporins. Heterologous expression in both Xenopus laevis oocytes and a rice mutant defective in Si uptake showed that HvLsi1 has transport activity for silicic acid. Expression of HvLsi1 was detected specifically in the basal root, and the expression level was not affected by Si supply. There was a weak correlation between Si uptake and the expression level of HvLsi1 in eight cultivars tested. In the seminal roots, HvLsi1 is localized on the plasma membrane on the distal side of epidermal and cortical cells. HvLsi1 is also located in lateral roots on the plasma membrane of hypodermal cells. These cell-type specificity of localization and expression patterns of HvLsi1 are different from those of OsLsi1. These observations indicate that HvLsi1 is a silicon influx transporter that is involved in radial transport of Si through the epidermal and cortical layers of the basal roots of barley.  相似文献

  • Salinity is one of the major abiotic stresses threatening crop production and yield worldwide. Breeding programmes are therefore needed to improve yield under cultivation in soil. Traits from locally adopted landraces provide a resource to assist breeding of novel elite genotypes. Here, we examine differentially expressed proteins by performing comparative proteomic profiling of the albumin/globulin grain fraction of Tunisian barley genotype landraces with contrasting salinity tolerance.
  • Tunisian barley Boulifa (B, tolerant) and Testour (T, sensitive) mature grains were assessed in 2‐DE profiles. Differentially expressed spots, with an abundance enhanced 1.5‐fold in the grain, were subjected to MALDI TOF/TOF MS for identification.
  • Distinctiveness between tolerant and sensitive genotypes was proved in the albumin/globulin fraction using PCA; 64 spots showed significant differential abundance. Increased accumulation of 40 spots was confirmed in Boulifa with, interestingly, four genotype‐specific spots. Two of these four spots were sHSP. Proteins with highest abundance were serpin Z7, 16.9 KDa Class I HSP and phosphogluconolactonase 2. Proteins such as expansin, kiwellin, kinesin and succinyl‐CoA ligase were identified for the first time in barley grain. Moreover, ß‐amylase, LEA family and others were identified as abundant in Boulifa. On the other hand, proteins more accumulated in Testour are implicated mainly in ROS scavenging and protease inhibition.
  • Our results clearly indicate proteomic contrast between the two selected genotypes. With identification of specific HSP, high abundant stress‐protective and other defined proteins, we provide biochemical traits that will support breeding programmes to address the threat of salinity in agricultural production.

We have studied the effects of six high-lysine barley mutations (Risø mutants 1508 and 56, Notch 1 and 2, lys 95 and 449) on the protein fractions of the grain. All mutants had a decreased relative and total amount of the lysine-poor hordein fraction, but only in Risø 1508 and 56 was the polypeptide composition of this fraction greatly affected. In all the mutants there were increases in the lysine-rich glutelin proteins and in nonprotein N while in Risø 1508 and the Notch mutants the total amount of salt-soluble proteins was also increased and their relative polypeptide composition substantially altered.This work was supported by EEC Contract No. 473 under the common program on plant protein improvement.  相似文献

Two contrasting barley (Hordeum vulgare L.) cultivars: Kepin No.7 (salt sensitive), and Jian 4 (salt tolerant) were grown in a hydroponics system containing 120 mol m-3 NaCl only and 120 mol m-3 NaCl with 1.0 mol m-3 Si (as potassium silicate). Compared with the plants treated with salt alone, superoxide dismutase (SOD) activity in plant leaves and H+-ATPase activity in plant roots increased, and malondialdehyde (MDA) concentration in plant leaves decreased significantly for both cultivars when treated with salt and Si. The addition of Si was also found to reduce sodium but increase potassium concentrations in shoots and roots of salt-stressed barley. Sodium uptake and transport into shoots from roots was greatly inhibited by added Si under salt stress conditions. However, Si addition exhibited little effect on calcium concentrations in shoots of salt-stressed barley. Thus, Si-enhanced salt tolerance is attributed to selective uptake and transport of potassium and sodium by plants. The results of the present study suggest that Si is involved in the metabolic or physiological changes in plants. Aluminium/silicon interactions in barley (Hordeum vulgare L.) seedlings   总被引:3,自引:0,他引:3  
The response of seedlings of the monocot Hordeum vulgare L. cv. Bronze to 0,25 and 50 M aluminium in factorial combination with 0, 1.4, 2.0 and 2.8 mM Si was tested in hydroponic culture at pH 4.5. Nutrient solution (500 M calcium nitrate) and Al/Si treatments were designed to avoid the precipitation of Al from solution. Silicon treatments gave significant amelioration of the toxic effects of Al on root and shoot growth and restored calcium levels in roots and shoots at harvest to levels approaching those of control plants. Aluminium uptake by roots was also significantly diminished in the presence of Si. Silicon alone gave a slight stimulation of growth, insufficient to explain its ameliorative effect on Al toxicity. 选用耐旱性不同的两个大麦品种作为研究对象,分析其叶片结构的异同。结果表明:两个大麦品种的叶片发育可以分为幼叶萌发期、幼叶抽出期、幼叶生长期和叶片成熟期四个阶段,其中在幼叶萌发期,叶片结构无明显差异。经PAS染色,从幼叶生长期开始,耐旱性弱的Moroc 9-75,含淀粉粒的叶肉细胞少,淀粉粒颗粒小; 耐旱性强的HS 41-1,含淀粉粒的叶肉细胞多,淀粉粒颗粒大。遭受干旱胁迫后,两个品种的植株长势明显较弱,叶片短而窄; 表皮细胞角质层变厚,叶片中叶肉细胞变小,叶肉细胞胞间隙变大,叶肉细胞破裂现象增多; PAS染色反应显示,含淀粉粒的叶肉细胞减少,淀粉粒颗粒变小或基本没有; HS 41-1解体的细胞不如Moroc 9-75多。因此,在光镜下,叶片结构的差异,特别是细胞含有的淀粉粒大小与数量的区别,是植物对水分胁迫的一种适应; 同时叶脉对植物刚性的影响较大。  相似文献

为了揭示不同环境因子对青藏高原栽培大麦籽粒淀粉含量(GSC)积累的影响程度,完善大麦GSC空间分异与环境因子的关系,明确青藏高原不同地区大麦品种GSC的环境效应,利用83个样点的地理、气候、土壤、农艺因子数据,研究了青藏高原栽培大麦GSC的分布特征,结果表明:(1)在地理水平方向上,青藏高原栽培大麦GSC的水平分布总体呈现出斑块状交错分布和南高北低的格局,并形成了以西藏拉孜、隆子、堆龙德庆、曲水、尼木、定日、萨迦、达孜、扎囊、日喀则、墨竹工卡、贡嘎、琼杰为中心的青藏高原中南部和以青海共和、贵德、海晏、同德为中心的青藏高原东北部等2个栽培大麦GSC高值区;(2)在地理垂直方向上,栽培大麦GSC的变化呈现出"S"型分布格局,即在海拔3300.0—3600.0 m以上,随着海拔的升高,栽培大麦GSC逐渐增加,在海拔4200.0 m与4500.0 m之间达到最高值,此后随着海拔的升高略有下降;(3)影响栽培大麦GSC的因子从大到小的顺序是穗密度﹥6月平均气温日较差芒长﹥9月平均气温1月平均气温年日照时数﹥≥0℃积温5月平均气温﹥8月平均气温日较差8月平均气温6月平均气温≥10℃积温6月平均月降水量5月平均月降水量7月平均相对湿度8月平均相对湿度7月平均气温。这一研究结果显示,对栽培大麦GSC影响最大的是基因型,其次是气候因素,土壤因素对GSC的影响不明显。影响栽培大麦GSC的农艺因子主要是穗密度和芒长,气候因子主要是拔节抽穗期的气温日较差和籽粒灌浆成熟期的平均气温,日照和降水的影响相对较小。  相似文献

Aims: To evaluate mycobiota and aflatoxins B1 (AFB1), B2 (AFB2), G1 (AFG1), G2 (AFG2) and fumonisin B1 (FB1) contamination in different malted barley types and brands and brewer’s grain collected from a major Argentinean brewery. Methods and Results: Total fungal counts were performed using the plate count method. Aflatoxin B1, AFB2, AFG1, AFG2 and Zearalenone (ZEA) analyses were performed by thin‐layer chromatography (TLC). Fumonisin B1 was determined by HPLC. Eighty‐three percentage of the malted barley (100% M1, 50% M2 and 100% M3) and 61% of brewer’s grain samples had a count >1 × 104 CFU g?1. Yeasts were isolated from all malt and brewer’s grain samples. Genera containing some of the most important mycotoxin producer species –Fusarium ssp., Aspergillus ssp., Penicillium ssp. and Alternaria ssp. – were isolated from the analysed samples, along with other environmental saprophytic fungi such as Geotrichum ssp., Mucorales and Cladosporium ssp. All samples were contaminated with 104–145 μg kg?1 FB1. Eighteen per cent of brewer’s grain samples were contaminated with 19–44·52 μg kg?1 AFB1. Aflatoxin B2, AFG1, AFG2 and ZEA were not detected in any of the analysed samples. Conclusions: Fungal and mycotoxin contamination in malt and brewer’s grain is an actual risk for animal and human health. Significance and Impact of the Study: This study may be useful for assessing the risk of mycotoxins in Argentinean beers and especially in animal feeds.  相似文献

The aim of these experiments was to evaluate how thresholds for phytotoxic substances obtained in seedling bioassays relate to yield losses or changes in yield components of mature barley crops after a short-term exposure to p-hydroxybenzoic acid. Under laboratory conditions a treatment with 1.81 mM p-hydroxybenzoic acid significantly reduced the radicle length of barley, whereas coleoptile elongation was less sensitive. The inhibition of the radicle length and coleoptile elongation was greater if the pH of the test solution was not buffered at pH 5.5. In a glasshouse trial the effect of p-hydroxybenzoic acid on the radicle and coleoptile elongation of spring barley was compared with the yield response after a three day exposure either during germination or at the double ridge stage of apex development. Applications of 0.72 mM, 1.44 mM and 3.62 mM p-hydroxybenzoic acid averaged over the treatments during germination or at the double ridge stage of development caused a yield reduction in the single ear weight of 5%, 13% and 19% in comparison with the control, respectively. The higher tiller categories in general showed a greater sensitivity towards an application of p-hydroxybenzoic acid and, therefore, could not compensate for the yield decrease of the main stem tiller. A single application of p-hydroxybenzoic acid either at germination or at the double ridge stage may cause yield losses, as reported from no-till systems or cereal monocultures. The data have implications for the interpretation of seedlings bioassays in allelopathic research and their applicability in estimating yield losses caused by phytotoxic substances. ei]Section editor: R Merckx  相似文献
