The Dwarf Sunflower (Helianthus annuus) is a hyperaccumulator of toxic metals including cadmium (Cd), mercury (Hg), nickel (Ni), and lead (Pb). In order to identify stress response to Pb, plants were exposed to a mixture of 30 mg/l of three ions, Cd, Cr, and Ni, with and without Pb. Soluble proteins were resolved by two-dimensional gel electrophoresis. Four proteins were differentially expressed and very abundant in the leaf samples after plants were exposed to all these four metals. The first protein spot contained two proteins: chitinase and a chloroplast drought-induced stress protein CDSP-34. The second spot contained a thaumatin-like protein. Two proteins in spot 3 were identified as heat-shock cognate 70-1 and the large subunit of ribulose-1,5-bisphosphate carboxylase/oxygenase. Several peptides were identified in spot 4 but none could be matched to any sequence in the NCBI database.  相似文献

Simultaneous Cr(VI) reduction and phenol degradation were investigated in a reactor containing Pseudomonas aeruginosa CCTCC AB91095. Phenol was used as carbon source. P.aeruginosa utilized metabolites formed during phenol degradation as energy source for Cr(VI) reduction. Cr(VI) inhibited both Cr(VI) reduction and phenol degradation when Cr(VI) concentration exceeded the optimum value (20 mg/L), whereas phenol enhanced both Cr(VI) reduction and phenol degradation below the optimum initial concentration of 100 mg/L. Cr(III) was the predominant product of Cr(VI) reduction in cultures after incubation for 24 h. Both Cr(VI) reduction and phenol degradation were influenced by the amount of inocula. The concentration of Cr(VI) and phenol declined quickly from 20, 100 to 3.36, 29.51 mg/L in cultures containing of 5% (v/v) inoculum after incubation for 12 h, respectively. The whole study showed that P. aeruginosa is promising for the reduction of toxic Cr(VI) and degradation of organic pollutants simultaneously in the mineral liquid medium.  相似文献

Degradation of chrysene, a four ring High Molecular Weight (HMW) Polycyclic Aromatic Hydrocarbon (PAH) is of intense environmental interest, being carcinogenic, teratogenic and mutagenic. Multiple PAH degrading halotolerant Achromobacter xylosoxidans was isolated from crude oil polluted saline site. Response Surface Methodology (RSM) using Central Composite Design (CCD) of Bushnell-Haas medium components was successfully employed for optimization resulting 40.79% chrysene degradation on 4th day. The interactions between variables as chrysene and glucose concentrations, pH and inoculum size on degradation were examined by RSM. Under optimum conditions, A. xylosoxidans exhibited 85.96% chrysene degradation on 5th day. The optimum values predicted by RSM were confirmed through confirmatory experiments. It was also noted that pH and glucose as co-substrate play a dynamic role in enhancement of chrysene degradation. Hence, A. xylosoxidans can be further used for subsequent microcosm and in situ experiments for its potential to remediate PAH contaminated saline and non-saline soils.  相似文献

Reductive adsorption of Cr(VI) on coir pith (hereafter CP) was examined as a function of pH, ionic strength, and temperature. The CP contains 1.33 meq g? 1 phenolic, 0.43 meq g? 1 of lactonic, and 0.35 meq g? 1 carboxylic sites. Thus the CP surface is enriched with electron-donating oxygen functionalities. As evidenced by infrared (IR) spectroscopy, the Cr(VI) → Cr(III) conversion is facilitated by CP sites that are enriched with O─ O functional groups. The adsorption of reduced Cr(VI) was found to occur via C─ O─ functional groups first forming innersphere complexes with the CP surface, yielding keto (> C═ O) groups on the CP surface. The reductive adsorption of Cr(VI) was almost completed within 3 to 4 h, and it was dependent on pH and background ionic strength, yielding the highest monolayer coverage (9.56E-7 mol m? 2) at pH 3.7 in 0.1 M NaNO3. The ΓCr(III) followed the order with respect to the ionic strength: Γ0.1 M > Γ0.01 M > Γ0.001 M. The initial rate constant, k i , increased with temperature as k i 313 K > k i 303 K > k i 293 K > k i 283 K.  相似文献

Pseudomonas aeruginosa (P. aeruginosa) was immobilized with polyvinyl alcohol (PVA), sodium alginate and multiwalled carbon nanotubes (MCNTs). After immobilization, the beads were subjected to freeze-thawing to enhance mechanical strength. When exposed to 80 mg/L Cr(VI), the immobilized bacteria were able to reduce 50% of them in 84 h, however the free cells were deactivated at this concentration. The beads were used to reduce 50 mg/L Cr(VI) for nine times, with the reduction efficiency above 90% in the first five times and 65% in the end.  相似文献

Fruits of Euterpe oleracea (FEO) are currently known as elements that present a very high antioxidant activity (AAO), as measured by the Oxygen Radical Absorbance Capacity. They are particularly rich in total phenolics (TP) and total anthocyanins (TA). Response Surface Methodology was used to optimise the solvent extraction of phenolic antioxidants from FEO, using a second-order polynomial equation to describe the experimental data for TP, TA, and AAO. In order to determine the best solid-to-liquid ratio and time of extraction, some preliminary studies were conducted. A rotatable central composite design with three variables (ethanol proportion, hydrochloric acid concentration and temperature) was then used. The results showed a good fit to the proposed model (R2 > 0.89). TP and TA, as well as TA and AAO, showed significant correlations (P < 0.05). The optimised conditions that maximized the yields of phenolic compounds (TP and TA) and AAO from FEO were: ethanol proportion between 70% and 80%, hydrochloric acid concentration between 0.065 and 0.074 mol/L and a temperature of 58 °C.  相似文献

This report attempts to elucidate the potential of plant seed extract assisted synthesis of graphite-based zinc oxide nanoparticles (C-ZnO NPs) towards removal of chromium(VI) ions from water samples. The graphite-based zinc oxide (C-ZnO) composites were characterized using thermogravimetric analysis (TGA), X-ray diffraction (XRD), Fourier-transform infrared (FTIR) spectroscopy and scanning electron microscopy (SEM). The C-ZnO nanocomposites have found to remove chromium from the sample through an adsorption process. The sensitivity of chromium removal through adsorption is found to be in the range of 40 to 240 mg. The adsorption behaviour was found to be fitting with Langmuir isotherm model and the adsorption reaction follows pseudo second-order kinetics.  相似文献

The study explores utilization of waste cyanobacterial biomass of Nostoc linckia from a lab-scale hydrogen fermentor for the biosorption of Cr(VI) from aqueous solution. The biomass immobilized in alginate beads was used for removal of the metal in batch mode optimizing the process conditions adopting response surface methodology (RSM). Kinetic studies were done to get useful information on the rate of chromium adsorption onto the cyanobacterial biomass, which was found to follow pseudo second-order model. Four important process parameters including initial metal concentration (10-100 mg/L), pH (2-6), temperature (25-45 °C) and cyanobacterial dose (0.1-2.0 g) were optimized to obtain the best response of Cr(VI) removal using the statistical Box-Behnken design. The response surface data indicated maximum Cr(VI) biosorption at pH 2-4 with different initial concentrations of the metal in the aqueous solution. The biosorbent could remove 80-90% chromium from solutions with initial metal concentration of 10-55 mg/L. Involvement of the surface characteristics of the biomass was studied through its scanning electron micrographs and Fourier transform infrared (FTIR) analysis.  相似文献

To assess whether foliar application of K+S as potassium sulfate (K2SO4) could alleviate the adverse effects of salt on sunflower (Helianthus annuus L. cv. SF-187) plants, a greenhouse experiment was conducted. There were two NaCl levels (0 and 150 mM) applied to the growth medium and six levels of K+S as K2SO4 (NS (no spray), WS (spray of water+0.1% Tween 20 solution), 0.5% K+0.21% S, 1.0% K+0.41% S, 1.5% K+0.62% S, and 2.0% K+0.82% S in 0.1% Tween-20 solution) applied two times foliarly to non-stressed and salt-stressed sunflower plants. Salt stress markedly repressed the growth, yield, photosynthetic pigments, water relations and photosynthetic attributes, quantum yield (Fv/Fm), leaf and root K+, Mg2+, P, Ca2+, N as well as K+/Na+ ratios, while it enhanced the cell membrane permeability, and leaf and root Na+ and Cl concentrations. Foliar application of potassium sulfate significantly improved growth, achene yield, photosynthetic and transpiration rates, stomatal conductance, water use efficiency, leaf turgor and enhanced shoot and leaf K+ of the salt-stressed sunflower plants, but it did not improve leaf and root Na+, Cl, Mg2+, P, Ca2+, N as well as K+/Na+ ratios. The most effective dose of K+S for improving growth and achene yield was found to be 1.5% K+0.62% S and 1% K+0.41% S, respectively. Improvement in growth of sunflower plants due to exogenously applied K2SO4 was found to be linked to enhanced photosynthetic capacity, water use efficiency, leaf turgor and relative water content.  相似文献

The main aim of this study was to investigate the influence of the sulfate ion on the tolerance to Cr(VI) and the Cr(VI) reduction in a yeast strain isolated from tannery wastewater and identified as Candida sp. FGSFEP by the D1/D2 domain sequence of the 26S rRNA gene. The Candida sp. FGSFEP strain was grown in culture media with sulfate concentrations ranging from 0 to 23.92 mM, in absence and presence of Cr(VI) [1.7 and 3.3 mM]. In absence of Cr(VI), the yeast specific growth rate was practically the same in every sulfate concentration tested, which suggests that sulfate had no stimulating or inhibiting effect on the yeast cell growth. In contrast, at the two initial Cr(VI) concentrations assayed, the specific growth rate of Candida sp. FGSFEP rose when sulfate concentration increased. Likewise, the greater efficiencies and volumetric rates of Cr(VI) reduction exhibited by Candida sp. FGSFEP were obtained at high sulfate concentrations. Yeast was capable of reducing 100% of 1.7 mM Cr(VI) and 84% of 3.3 mM Cr(VI), with rates of 0.98 and 0.44 mg Cr(VI)/L h, with 10 and 23.92 mM sulfate concentrations, respectively. These results indicate that sulfate plays an important role in the tolerance to Cr(VI) and Cr(VI) reduction in Candida sp. FGSFEP. These findings may have significant implications in the biological treatment of Cr(VI)-laden wastewaters.  相似文献

A Bacillus sp. RE was resistant to chromium and reduced Cr(VI) without accumulating chromium inside the cell. When Cr(VI) was 10 and 40 μg ml−1, >95% of the total Cr(VI) was reduced in 24 and 72 h of growth, respectively, whereas at 80 μg Cr(VI) ml−1 only 50% of Cr(VI) was reduced. However growth was not affected; the cell mass was 0.7–0.8 mg ml−1 in all cases. The cell-free extract showed Cr(VI) reducing enzyme activity which was enhanced (>5 fold) by NADH and NADPH. Like whole cells the enzyme also reduced Cr(VI) with decreasing efficiency on increasing Cr(VI) concentration. The enzyme activity was optimal at pH 6.0 and 30 °C. The enzyme was stable up to 30 °C and from pH 5.5 to 8, but from pH 4 to 5 the enzyme was severely destabilized. Its Km and Vmax were 14 μm and 3.8 nmol min−1 mg−1 respectively. The enzyme activity was enhanced by Cu2+ and Ni2+ and inhibited by Hg2+. The effect of extracellular polymeric substances (EPS) of Gram-positive Bacillus subtilis and Gram-negative Pseudomonas putida on Cu(II) adsorption was investigated using a combination of batch adsorption, potentiometric titrations, Fourier transform infrared spectroscopy. Both the potentiometric titrations and the Cu(II) adsorption experiments indicated that the presence of EPS in a biomass sample significantly enhance Cu(II) adsorption capacity. Surface complexation modeling showed that the pKa values for the three functional groups (carboxyl, phosphate and hydroxyl) were very similar for untreated and EPS-free cells, indicating no qualitative difference in composition. However, site concentrations on the untreated cell surface were found to be significantly higher than those on the EPS-free cell surface. Infrared analysis provided supporting evidence and demonstrated that carboxyl and phosphate groups are responsible for Cu(II) adsorption on the native and EPS-free cells.  相似文献

Promastigotes of Leishmania (Viannia) panamensis were successfully transfected with p6.5-egfp to express green fluorescent protein. The transfectants remained infective to macrophages, providing an in vitro model for screening antileishmanial drugs. This was demonstrated by flow cytometry of macrophage-associated GFP after exposure of infected cultures to known antileishmanial drugs, i.e. amphotericin B and glucantime®. Fluorescence of GFP diminished progressively from infected cells with increasing drug concentrations used in both cases. The availability of this fluorescent assay for infection of macrophages by L. (V.) panamensis facilitates drug discovery program for the Viannia species, which differ significantly from those of the Leishmania subgenus.  相似文献

Detoxification of Cr(VI) under alkaline pH requires attention due to the alkaline nature of many effluents. An alkaliphilic gram-positive Bacillus subtilis isolated from tannery effluent contaminated soil was found to grow and reduce Cr(VI) up to 100% at an alkaline pH 9. Decrease in pH to acidic range with growth of the bacterium signified the role played by metabolites (organic acids) in chromium resistance and reduction mechanism. The XPS and FT-IR spectra confirmed the reduction of Cr(VI) by bacteria into +3 oxidation state. Chromate reductase assay indicated that the reduction was mediated by constitutive membrane bound enzymes. The kinetics of Cr(VI) reduction activity derived using the monod equation proved (Ks = 0.00032) high affinity of the organism to the metal. This study thus helped to localize the reduction activity at subcellular level in a chromium resistant alkaliphilic Bacillus sp.  相似文献

The partition and purification of α-amylase from a culture supernatant of Aspergillus oryzae CBS 819.72 was made in aqueous two-phase system (ATPS). According to bibliography and preliminary studies, the factors polyethylene glycol (PEG) molecular weight (MPEG) and concentration (CPEG), buffer type (BU) and concentration (CBU), temperature (T), salt nature (SALT) and concentration (CSALT), bioligand (BL) and concentration (CBL) and pH were investigated using a Plackett–Burman design to identify the factors affecting separation. Taking into consideration a simultaneous increase in enzyme recovery (RY) and purification factor (PF), the best performance of the system was obtained at 4 °C and pH 6 using PEG 8000 g/mol, citrate buffer, KCl and sucrose. Experimental Box–Behnken design together with the Response Surface Methodology (RSM) have been used to find optimum CPEG, CCitrate and CSALT. Quadratic models were predicted for PF and RY in the top phase and a better compromise between these two parameters can be found by superimposing the contour plots of PF and RY for 8% citrate. A region in the experimental space can be defined where the purification factor is always higher than 3 with yields exceeding 65%.  相似文献

Several studies have shown that the initial surface wettability, is of importance in the settlement of macrofouling larvae such as barnacles, bryozoans and hydroids in the field as well as in laboratory assays. In this study we present results from laboratory assays using hydrophilic and hydrophobic polystyrene (PS) and cyprid larvae of Balanus improvisus (Darwin). The results obtained differ markedly from those reported for the barnacle Balanus amphitrite (Darwin), where a high surface wettability seemed to be preferred for settlement. Our results show that a surface with intermediary wettability (hydrophilic PS) reduced settlement by 38% as compared to surfaces of low wettability (hydrophobic PS) during an 8-day period. During the experiment, the wettability in the hydrophilic PS dishes was not significantly changed as measured by advancing contact angle with mQ water. Over an 8-day period wettability of the hydrophobic PS dishes approached that of the hydrophilic PS surfaces. We further conducted experiments with highly hydrophilic and highly hydrophobic methylsilane-treated glass surfaces with known chemistry. In this experiment, the settlement of cyprid larvae was completely inhibited by the high wettability surfaces. Contact angle measurements revealed that the wettability during the length of the experiment of the hydrophilic glass surfaces was not significantly altered. We conclude by these experiments that even an intermediate wettability can significantly affect the overall settlement success of the barnacle B. improvisus. The mechanism by which the settlement is impeded might be biologically mediated through the recognition by cyprid larvae of the molecular composition of the surface when the cyprid reverts to the settlement phase, i.e. when swimming behaviour is abandoned in favour of surface exploration, or it is mediated by physicochemical forces acting between the surface and the larval body or the larval antennules.  相似文献

The optimal medium for butyric acid production by Clostridium thermobutyricum in a shake flask culture was studied using statistical experimental design and analysis. The optimal composition of the fermentation medium for maximum butyric acid yield, as determined on the basis of a three-level four-factor Box-Behnken design (BBD), was obtained by response surface methodology (RSM). The high correlation between the predicted and observed values indicated the validity of the model. A maximum butyric acid yield of 12.05 g/l was obtained at K2HPO4 7.2 g/l, 34.9 g/l glucose, 20 g/l yeast extract, and 15 g/l acetate, which compared well to the predicated production of 12.13 g/l.  相似文献

We isolated four cultures of chromate resistant, unicellular, non-motile green algae from disposal sites of the paper-pulp and electroplating industries. These algae were maintained in Tris-acetate-glycerophosphate medium containing 30 μM K2Cr2O7. The morphological features as well as analysis of the 500-bp fragment of 18S rDNA (NS 12 region) showed that these isolates belong to Chlorella spp. These isolates showed EC50 values for chromate ranging from 60 to 125 μM. Uptake studies with radioactive 51Cr(VI) showed that 10–19% of total radioactivity was intracellular, and 1–2% was bound to the cell wall. The rest of the activity remained in the medium, suggesting that resistance was not related to accumulation of Cr(VI) in the cells. Interestingly, when these isolates were grown in the presence of 30 μM of K2Cr2O7, a decrease in the Cr(VI) concentration in the medium was observed. Only live cells could deplete Cr(VI) from the supernatant, suggesting the presence of chromium reduction activity in these Chlorella isolates. Cr(VI) reduction activity of the cells of Chlorella was stimulated by light as well as by acetate and glycerophosphate. Treatment of Chlorella cells with 3-(3,4 dichlorophenyl),1,1dimethyl urea (DCMU) did not affect the Cr(VI) reduction. However, if the cells were treated with sodium azide, Cr(VI) reduction was severely affected. Though chromate resistance has been well documented in algae, the information on chromate reduction by algae is scant. This paper discusses the Cr(VI) reduction by Cr(VI) resistant Chlorella, which may find a use in the effective bioremediation of Cr(VI).  相似文献
