This study investigated the biomass, lipid production, fatty acid content, and other nutrients present in microorganisms by using four culture methods: (1) photoautotrophic pure Chlorella vulgaris cultures (PP); (2) heterotrophic pure C. vulgaris cultures (PH); (3) mixed cultures of Rhodotorula glutinis and C. vulgaris under photoautotrophic conditions (MP); and (4) heterotrophic mixed cultures (MH). The microorganisms in MP culture showed the optimum growth condition and lipid production. Among the cultures, MP yielded the highest number of cells and biomass (5.9?×?105 cells/mL and 0.523 g/L, respectively). Furthermore, lipid production in MP culture was 114.22 mg/L, which is 136 % higher than that in MH culture (48.22 mg/L). Considering the higher contents of palmitic acid (C16:0) at 24.65 %, oleic acid (C18:1) at 56.34 %, and protein at 42.39 g/100 g in the MP culture than in other cultures, we proposed that MP could be used effectively to support the growth of microorganisms. We attempted to enhance the growth and total lipid production of three microalgal species, Isochrysis galbana LB987, Nannochloropsis oculata CCAP849/1, and Dunaliella salina, which are capable of accumulating high content of lipid in cells. Low nitrogen concentration under photoautotrophic conditions stimulated total lipid production, but a decreasing total lipid content and an increasing biomass were observed with increasing nitrogen concentration. Among the different carbon sources tested for heterotrophic cultivation, glucose improved the growth of all three strains. The optimal glucose concentration for growth of I. galbana LB987 and N. oculata CCAP849/1 was 0.02 M, and that of D. salina was 0.05 M. Enhanced growth occurred when they were cultivated under heterotrophic or mixotrophic conditions compared with photoautotrophic conditions. Meanwhile, high total lipid accumulation in cells occurred when they were cultivated under photoautotrophic or mixotrophic conditions. During mixotrophic cultivation, biomass production was not affected significantly by light intensity; however, both chlorophyll concentration and total lipid content increased dramatically with increasing light intensity up to 150 µmol/m2/s. The amount and composition ratio of saturated and unsaturated fatty acids in cells were different from each other depending on both species and light intensity. The highest accumulation of total fatty acid (C16–C18) among the three strains was found from cells of N. oculata CCAP849/1, which indicates that this species can be used as a source for production of biodiesel.  相似文献

This study demonstrates the production of algal biodiesel from Dunaliella tertiolecta, Nannochloropsis oculata, wild freshwater microalgae, and macroalgae lipids using a highly efficient continuous catalytic process. The heterogeneous catalytic process uses supercritical methanol and porous titania microspheres in a fixed bed reactor to catalyze the simultaneous transesterification and esterification of triacylglycerides and free fatty acids, respectively, to fatty acid methyl esters (biodiesel). Triacylglycerides and free fatty acids were converted to alkyl esters with up to 85% efficiency as measured by 300 MHz 1H NMR spectroscopy. The lipid composition of the different algae was studied gravimetrically and by gas chromatography. The analysis showed that even though total lipids comprised upwards of 19% of algal dry weight the saponifiable lipids, and resulting biodiesel, comprised only 1% of dry weight. Thus highlighting the need to determine the triacylglyceride and free fatty acid content when considering microalgae for biodiesel production.  相似文献

While photosynthetic microalgae, such as Chlorella, serve as feedstocks for nutritional oils and biofuels, heterotrophic cultivation can augment growth rates, support high cell densities, and increase triacylglycerol (TAG) lipid content. However, these species differ significantly in their photoautotrophic and heterotrophic characteristics. In this study, the phylogeny of thirty Chlorella strains was determined in order to inform bioprospecting efforts and detailed physiological assessment of three species. The growth kinetics and lipid biochemistry of C. protothecoides UTEX 411, C. vulgaris UTEX 265, and C. sorokiniana UTEX 1230 were quantified during photoautotrophy in Bold''s basal medium (BBM) and heterotrophy in BBM supplemented with glucose (10 g L−1). Heterotrophic growth rates of UTEX 411, 265, and 1230 were found to be 1.5-, 3.7-, and 5-fold higher than their respective autotrophic rates. With a rapid nine-hour heterotrophic doubling time, Chlorella sorokiniana UTEX 1230 maximally accumulated 39% total lipids by dry weight during heterotrophy compared to 18% autotrophically. Furthermore, the discrete fatty acid composition of each strain was examined in order to elucidate lipid accumulation patterns under the two trophic conditions. In both modes of growth, UTEX 411 and 265 produced 18∶1 as the principal fatty acid while UTEX 1230 exhibited a 2.5-fold enrichment in 18∶2 relative to 18∶1. Although the total lipid content was highest in UTEX 411 during heterotrophy, UTEX 1230 demonstrated a two-fold increase in its heterotrophic TAG fraction at a rate of 28.9 mg L−1 d−1 to reach 22% of the biomass, corresponding to as much as 90% of its total lipids. Interestingly, UTEX 1230 growth was restricted during mixotrophy and its TAG production rate was suppressed to 18.2 mg L−1 d−1. This constraint on carbon flow raises intriguing questions about the impact of sugar and light on the metabolic regulation of microalgal lipid biosynthesis.  相似文献

When heterotrophic cell cultures of red goosefoot (Chenopodium rubrum) turned photoautotrophic, their contents of various glycolipids and phospholipids increased. The total lipids and the individual lipid classes, especially monogalactosyldiacylglycerols, became richer in linolenic and poorer in linoleic acids. When photoautotrophic cell cultures were rendered heterotrophic again a reversal of changes occurred; both the composition of lipids and the patterns of their constituent fatty acids became similar to those of the starting heterotrophic cultures.The results indicate that the biosynthesis of linolenic acid in photoautotrophic cell cultures involves mainly desaturation of linoleic acid and that chain extension of hexadecatrienoic acid is possibly another, though minor pathway. Monogalactosyldiacylglycerols are apparently the substrates preferred for linolenic acid biosynthesis, whereas various phospholipids are the substrates preferred for linoleic acid biosynthesis.During a growth period of 6 weeks, the levels of polyunsaturated fatty acids in the lipids from both heterotrophic and photoautotrophic cell cultures decrease with time, whereas the proportions of palmitic acid increase.  相似文献

The present study reports evaluation of an indigenous microalgal isolate Chlorella sp. FC2 IITG as a potential candidate for biodiesel production. Characterization of the strain was performed under photoautotrophic, heterotrophic, and mixotrophic cultivation conditions. Further, an open-pond cultivation of the strain under outdoor conditions was demonstrated to evaluate growth performance and lipid productivity under fluctuating environmental parameters and in the presence of potential contaminants. The key findings were: (1) the difference in cultivation conditions resulted in significant variation in the biomass productivity (73–114 mg l?1 day?1) and total lipid productivity (35.02–50.42 mg l?1 day?1) of the strain; (2) nitrate and phosphate starvation were found to be the triggers for lipid accumulation in the cell mass; (3) open-pond cultivation of the strain under outdoor conditions resulted in biomass productivity of 44 mg l?1 day?1 and total lipid productivity of 10.7 mg l?1 day?1; (4) a maximum detectable bacterial contamination of 7 % of the total number of cells was recorded in an open-pond system; and (5) fatty acid profiling revealed abundance of palmitic acid (C16:0), oleic acid (C18:1) and linoleic acid (C18:2), which are considered to be the key elements for suitable quality biodiesel.  相似文献

The use of volatile fatty acids (VFAs) for microbial lipid accumulation was investigated in flask cultures of Cryptococcus albidus. The optimum culture temperature and pH were 25 °C and pH 6.0, respectively, and the highest lipid content (27.8%) was obtained with ammonia chloride as a nitrogen source. The lipid yield coefficient on VFAs was 0.167 g/g of C. albidus with a VFAs (acetic, propionic, butyric acids) ratio of 8:1:1, which was in good agreement with a theoretically predicted lipid yield coefficient of the VFAs as a carbon source. The major fatty acids of the lipids accumulated by C. albidus were similar to those of soybean oil and jatropha oil. A preliminary cost analysis shows that VFAs-based biodiesel production is competitive with current palm and soybean based biodiesels. Further process development for lower aeration cost and higher lipid yield will make this process more economical.  相似文献

Effects of nitrate feeding on the cell growth and lipid accumulation of marine microalgae Isochrysis zhangjiangensis were investigated. When nitrate was supplied at interval of 24 h, instead of 72 h, a high lipid content of 40.9% and a biomass density of 3.1 g L−1 were obtained. To confirm whether I. zhangjiangensis accumulates lipid during nitrogen-repletion, a two-stage cultivation method was applied. This algal strain had a high lipid content during sustained nitrate addition and showed a high carbohydrate content under nitrate-depletion conditions. These results revealed that this algal strain can accumulate lipids under nitrogen-repletion conditions and accumulate carbohydrate under nitrogen-depletion conditions. When cultured in an extremely high nitrate concentration, 9 g L−1 at 24 h intervals, the growth of algal cells was suppressed, but the highest lipid content of 53% was attained. This special characteristic of lipid accumulation makes I. zhangjiangensis an ideal candidate for producing biodiesel using N-rich wastewater.  相似文献

Feeding and starvation experiments were carried out with Clione limacina sampled in Kongsfjorden (Svalbard, Arctic) during summer 2002. Dry mass and lipid mass, lipid class and fatty acid compositions were analysed. Specimens of C. limacina used for the feeding study had a mean length of 25 mm, a dry mass (DM) of 13.7 mg, and a moderate lipid content of 12.1%DM. Animals were allowed to ingest only one individual of its exclusive prey, Limacina helicina which had 8.0 mm in diameter, 21.4 mg DM and 8.7% lipid of ash-free DM. Five days after feeding, the dry mass of C. limacina had increased from 13.7 to 25.3 mg which corresponds to an uptake of about 80% of the ash-free DM (14.3 mg) of L. helicina. Lipid mass increased from 1.5 to 3.9 mg which is almost two times more the ingested lipid from L. helicina (1.2 mg lipid). Thus, the major portion of lipids was synthesised de novo by C. limacina from non-lipid compounds. These lipids were triacylglycerols (TAG) and 1-O-alkyldiacylglycerol ethers (DAGE), increasing from low proportions of 6.1% and 5.7% to 42.3% and 25.8%, respectively. Considerable de novo synthesis was observed for the monounsaturated fatty acids 16:1(n − 7), 17:1(n − 8), 18:1(n − 9), and 18:1(n − 7) and the alkyl moiety 16:0. The increase in the polyunsaturated fatty acids 22:6(n − 3), 20:5(n − 3), and 18:4(n − 3) corresponded with the amount available by ingestion of L. helicina, supporting that C. limacina is not able to synthesise polyunsaturates. After 15 days of digestion, dry mass and lipids dropped almost back to the initial values.During the 100-day starvation experiment, two groups of animals were separately considered as storage lipid-rich and lipid-poor animals because of their large differences in the amount and proportion of TAG and DAGE. Storage lipid-rich C. limacina were only found until day 50, whereas lipid-poor animals were present throughout the experiment. In the lipid-rich specimens, the levels of TAG were about twice that of DAGE. The proportions of TAG decreased considerably during the 50 days of starvation (from 48.3% to 25.1% of total lipid). DAGE, varying between 16.5% and 20.5%, showed only a small decrease. The lipid-poor animals survived 100 days of starvation, exhibiting low initial amounts and proportions of storage lipids which were nearly exhausted at the end. In all C. limacina specimens, the total lipid content remained almost constant showing that lipid and non-lipid components were simultaneously utilised. This implies that body shrinkage may be an important adaptation to long-term starvation. Based on these results, it is possible to estimate the potential survival period of lipid-rich C. limacina under food limitation. A model, which considers maturity and reproduction (egg production), reveals that lipid-rich specimens might be able to survive up to 260 days without food.  相似文献

Pennate diatom Nitzschia palea can be cultured in outdoor vertical-bed photobioreactors to produce biodiesel. To assess the production of biomass and lipids, non-axenic cultures of Nitzschia palea were grown outdoors, and the growth of these cultures was measured biweekly. During the annual cycle of algal culture, the culture temperature ranged from 17.3 °C to 33.5 °C, the dry weight biomass ranged from 0.11 g l−1 to 0.25 g l−1, light energy] ranged from 1.94 Wm−2 to 3.9 Wm−2 and intracellular lipid content ranged from 7.1% to 11.4% of biomass weight after drying at 60 °C. Gas chromatography/mass spectroscopy (GC/MS) analysis of n-hexane extracts showed that the intracellular lipids were primarily C14:0 myristic acid (9.01%), C15:0 pentadecyclic acid (8.26%) and two types of C16:0, palmitic acid (41.13%) and palmitoleic acid (29.25%). Gel permeation analysis showed that carboxylic acids comprised 28.9% of lipids, 16.3% of monoglycerides, 27.3% of diglycerides and 24.3% of triglycerides. Alcoholysis of lipids resulted in the conversion of about 93.9% of fatty acids to equivalent fatty acid methyl esters (FAME) or biodiesel, which, on basis of wt%, consisted primarily of C15:0 methyl myristate (8.3%), C16:0 methyl pentadecanoate] (7.2%), C17:1methyl palmitoleate (28.7%) and methyl palimtate](39.8%).  相似文献

The role of fatty acid synthesis in the acclimation of the photosynthetic machinery to high temperature was investigated in a mutant of the cyanobacterium Synechocystis sp. PCC 6803 that had a lower than wild-type level of enoyl-(acyl-carrier-protein) reductase FabI, a key component of the type-II fatty acid synthase system. The mutant exhibited marked impairment in the tolerance and acclimation of cells to high temperature: photoautotrophic growth of the mutant was severely inhibited at 40 °C. Moreover, mutant cells were unable to achieve wild-type enhancement of the thermal stability of photosystem II (PSII) when the growth temperature was raised from 25 °C to 38 °C. Enhancement of the thermal stability of PSII was abolished when wild-type cells were treated with triclosan, a specific inhibitor of FabI, and the enhancement of thermal stability was also blocked in darkness and in the presence of chloramphenicol. Analysis of fatty acids in thylakoid membranes revealed that levels of unsaturated fatty acids did not differ between mutant and wild-type cells, indicating that the saturation of fatty acids in membrane lipids might not be responsible for the enhancement of thermal stability at elevated temperatures. Our observations suggest that the synthesis de novo of fatty acids, as well as proteins, is required for the enhancement of the thermal stability of PSII during the acclimation of Synechocystis cells to high temperature.  相似文献

The growth and biochemical composition of photoautotrophic and heterotrophic Isochrysis maritima in 50 L of Walne’s medium were compared. Heterotrophic I. maritima fed with 0.02 M glucose had a 4.6-fold higher maximum cell density (38.17 ± 0.23 × 106 cells mL?1) than photoautotrophic cells (8.29 ± 0.70 × 106 cells mL?1). The carbohydrate content was slightly higher in heterotrophic cells at all growth stages (mid-exponential, 40.8%; early stationary, 48.3%; and late stationary, 47.6%), but there was no significant effect on the protein content under either trophic condition. The total saturated fatty acids (SFAs), monounsaturated fatty acids (MUFAs) and polyunsaturated fatty acids (PUFAs) were higher under heterotrophic conditions than those under photoautotrophic conditions. However, because omega-3 PUFAs are the most essential element in feed nutrition, low results for eicosapentaenoic acid (EPA) (0.28 ± 0.06%) and docosahexaenoic acid (DHA) (3.22 ± 0.26%) in the heterotrophic cells compared to the photoautotrophic cells (EPA: 0.44 ± 0.11%; DHA: 8.58 ± 0.73%) plus a low omega-3/6 PUFAs ratio (heterotrophic: 0.16–0.47; photoautotrophic: 2.60–2.88) and high value of (SFA + MUFA)/PUFA (heterotrophic: 5.50–6.81; photoautotrophic: 2.64–3.60) showed that this species is not suitable for aquaculture feed when cultivated under heterotrophic conditions.  相似文献

异养细胞种子/光自养培养方法是一种可异养培养的能源微藻培养的有效方法,但已有文献尚未从工艺优化角度考察其发展潜力。为了获得较高细胞密度的用于光自养培养的种子和提高光自养培养的细胞密度与油脂产率,对异养细胞种子/光自养培养的培养基和培养条件进行了优化。结果表明,采用优化后的培养基,椭圆小球藻在摇瓶中异养培养的最高藻细胞密度可达11.04 g/L,比在初始培养基条件下提高了28.0%,在5 L发酵罐中异养培养的藻细胞密度达到73.89 g/L;在2 L柱式光生物反应器中光自养培养的藻细胞密度、油脂含量和油脂产率分别达1.62 g/L、36.34%和6.1 mg/(L·h),油脂成分主要为含C16-C18碳链的脂肪酸,是制备生物柴油的理想原料。经过优化,异养细胞种子/光自养培养这一方法能够显著地提高椭圆小球藻产油脂的能力,这进一步表明异养细胞种子/光自养培养方法有望成为可异养的能源微藻的高效培养方式。  相似文献

The effects of supplementing the culture medium with Mg2+ on the growth, lipid production, and fatty acid composition of Monoraphidium sp. FXY-10 were studied under photoautotrophic, heterotrophic, and mixotrophic conditions. Under the photoautotrophic condition, microalgae supplemented with 100 μM Mg2+ grew significantly better than the control group and exhibited a secondary growth state. The final cell density was 1.25-fold higher than that of the control group (2.98 g L?1), and the peak lipid content reached 59.8 % (control group 52.3 %). Culture under the heterotrophic condition did not significantly increase the growth rate, but the experimental group (100 μM Mg2+ supplementation) achieved a 37.03 % lipid content compared to 28.47 % by the control group. The lipid productivity of the experimental group (100 μM Mg2+ supplementation) was higher, reaching 65.93 mg L?1 day?1 compared with 56.10 mg L?1 day?1 for the group without additional Mg2+. Under the mixotrophic condition, the experimental group achieved a final density of 3.10 g L?1, which was higher than that of the control group (2.98 g L?1). There was also no variation in fatty acid composition between the experimental group and the control group. Under the heterotrophic and mixotrophic conditions, the experimental group produced more than 50% saturated fatty and mono-unsaturated fatty acids, and the degree of unsaturation was <137. The present study focused on cost-effective production of microalgal biomass and lipid production on dairy effluent. The novel microalga, Chlorella sp. isolated from the dairy effluent showed high growth and lipid production on the undiluted and two-fold diluted dairy effluent which were four to five times higher than those of Chlorella vulgaris (control). The high growth of Chlorella sp. was thought to be possibly due to its heterotrophic growth capacity, high turbidity, COD, nutrients and trace elements. In contrast, C. vulgaris showed poor heterotrophic and photoautotrophic growth under the highly turbid conditions of dairy effluent. Both Chlorella sp. and C. vulgaris showed similar total FAME (mg FAME/g algal cells). The fatty acid composition analysis revealed that both Chlorella sp. and C. vulgaris possessed major C18 and C20 fatty acids which will be used for biodiesel production. Overall, the novel microalga, Chlorella sp. isolated from the dairy effluent showed high potential for cost-effective algal cultivation and lipid production on dairy effluent without any modification of process.  相似文献

Marine microalgae have emerged as important feedstock for liquid biofuel production. The identification of lipid-rich native microalgal species with high growth rate and optimal fatty acid profile and biodiesel properties is the most challenging step in microalgae-based biodiesel production. In this study, attempts have been made to bio-prospect the biodiesel production potential of marine and brackish water microalgal isolates from the west coast of India. A total of 14 microalgal species were isolated, identified using specific molecular markers and based on the lipid content; seven species with total lipid content above 20% of dry cell weight were selected for assessing biodiesel production potential in terms of lipid and biomass productivities, nile red fluorescence, fatty acid profile and biodiesel properties. On comparative analysis, the diatoms were proven to be promising based on the overall desirable properties for biodiesel production. The most potential strain Navicula phyllepta MACC8 with a total lipid content of 26.54 % of dry weight of biomass, the highest growth rate (0.58 day?1) and lipid and biomass productivities of 114 and 431 mgL?1 day?1, respectively, was rich in fatty acids mainly of C16:0, C16:1 and C18:0 in the neutral lipid fraction, the most favoured fatty acids for ideal biodiesel properties. The biodiesel properties met the requirements of fuel quality standards based on empirical estimation. The marine diatoms hold a great promise as feedstock for large-scale biodiesel production along with valuable by-products in a biorefinery perspective, after augmenting lipid and biomass production through biochemical and genetic engineering approaches.  相似文献

Microalgal lipids are promising alternative feedstocks for biodiesel production. Scenedesmus sp. NJ-1, an oil-rich freshwater microalga isolated from Antarctica, was identified to be a suitable candidate to produce biodiesel in this study. This strain could grow at temperatures ranging from 4 to 35?°C. With regular decrease in nitrate concentration in the medium, large quantities of triacylglycerols accumulated under batch culture conditions detected by thin layer chromatography and BODIPY 505/515 fluorescent staining. Scenedesmus sp. NJ-1 achieved the average biomass productivity of 0.105?g?l?1?d?1 (dry weight) and nearly the highest lipid content (35?% of dry cell weight) was reached at day 28 in the batch culture. Neutral lipids accounted for 78?% of total lipids, and C18:1 (n-9), C16:0 were the major fatty acids in total lipids, composing 37 and 20?% of total fatty acids of Scenedesmus sp. NJ-1 grown for 36?days, respectively. These results suggested that Scenedesmus sp. NJ-1 was a good source of microalgal oils for biodiesel production.  相似文献

Metabolically-active autotrophic nitrite oxidizers from activated sludge were labeled with 13C-bicarbonate under exposure to different temperatures and nitrite concentrations. The labeled samples were characterized by FAME-SIP (fatty acid methyl ester-stable isotope probing). The compound cis-11-palmitoleic acid, which is the major lipid of the most abundant nitrite oxidizer in activated sludge, Candidatus Nitrospira defluvii, showed 13C-incorporation in all samples exposed to 3 mM nitrite. Subsequently, the lipid cis-7-palmitoleic acid was labeled, and it indicated the activity of a nitrite oxidizer that was different from the known Nitrospira taxa in activated sludge. The highest incorporation of cis-7-palmitoleic acid label was found after incubation with a nitrite concentration of 0.3 mM at 17 and 22 °C. While activity of Nitrobacter populations could not be detected by the FAME-SIP approach, an unknown nitrite oxidizer with the major lipid cis-9 isomer of palmitoleic acid exhibited 13C-incorporation at 28 °C with 30 mM nitrite. These results indicated flexibility of nitrite-oxidizing guilds in a complex community responding to different conditions. Labeled lipids so far not described for activated sludge-associated nitrifiers indicated the presence of unknown nitrite oxidizers in this habitat. The FAME-SIP-based information can be used to define appropriate conditions for the enrichment of nitrite-oxidizing guilds from complex samples.  相似文献
