Both the initial and the terminal 1 hr portions of the subjectiveday fraction, namely the L1- arid L2-phases, of a 24 hr daymust be illuminated in order for the day to be perceived asa long day in the min-LD determination by the long-day plant,Lemna gibba G3 (9). The light requirement of the L1-phase wassatisfied by a 10 min red light pulse given at the beginningof the phase. The red light effect was erased by a subsequent10 min far-red light, indicating phytochrome-mediated processesoccurring in the L1-phase. The light requirement of the L2-phasewas satisfied by blue or far-red light given during the terminal10 min period of the phase; there was no indication of phytochromeinvolvement. The light action on the L1-phase was replaced by10–5 M of cyclic AMP or 10–7 M of DL-isoproterenol.The isoproterenol action was antagonized by 10–7 M ofDL-propranolol. Cyclic AMP (10–5 M) combined with salicylicacid (10–6 M), which can remove the light requirementof the L2-phase (10), rendered a completely dark day physiologicallyequivalent to a long day. Acetylcholine (10–5 M) exertednyctomimetic action on the L1-phase of the second light day.The action of acetylcholine was antagonized by cyclic AMP (10–5M). The L2-phase required no light in the presence of 10–7M of DL-propranolol, and this propranolol action was not affectedby isoproterenol. Spectral response of corn (Zea mays) in root geotropism   总被引:1,自引:0,他引:1  
The primary roots of corn (Zea mays, Wisconsin hybrid 64A ?22R) show positive geotropism following exposure to light. Thisconfirms the works of other investigators. The curvature responsebegins at about 1 hr following irradiation and reached a plateauat 5 hr. A study of wavelengths 350–760 nm, using energiesof 2.24 ? 1014 photon cm–2 and exposure times 60 sec,shows that the most effective light is at 660 nm with lessereffectiveness at 460 and 560 nm. The responses at 660 and 460nm are reversible by a far-red (730 nm) exposure, indicativeof the possible participation of phytochrome. Analyses of freshtips of corn roots with a dual-wavelength difference photometershow the phytochrome content in the root to be about 0.16 (OD) per gram fresh wieght. The requirement of light for thegeotropic growth response of corn roots might be an adaptivephenomenon. The occurrence of photomorphogenic activity in thegreen light should be of concern to those who use green as the"safe" light in "dark" experiments. 1Work supported by U. S. NASA and U. S. ERDA. Although taxol inhibits membrane trafficking, the nature of thisinhibition has not been well defined. In this study, we define theeffects of taxol on endocytosis in CV-1 cells using density gradientcentrifugation of membranes over sorbitol density gradients. Aftertaxol treatment, resident endosomal enzymes and the epidermal growthfactor (EGF) receptor (EGFR) showed significant(P  0.05) enrichment in membraneswith properties of early endosomes(fractions 4 and5); the EGFR andNa+-K+-ATPasewere also significantly (P  0.05)depleted in lysosomal fractions(fractions10 and11). The suggestion that taxolspecifically reduces movement of endosomal constituents to lysosomeswas supported by fluorescence microscopy studies revealing restrictionof EGF to the peripheries of taxol-treated cells, in contrast to the perinuclear lysosomal-like distribution of EGF seen in controls. Kinetic studies with 125I-labeledEGF were also consistent with a taxol-induced block in traffic fromendosomes and lysosomes after 15 min of uptake but also suggested anadditional taxol-sensitive step in trafficking that involvedredistribution of 125I-EGF withinhigh-density compartments after 150 min. Related changes in cytoplasmicdynein distribution were observed within high-density compartments fromtaxol-treated cells, suggesting that this motor might participate inthis later taxol-sensitive trafficking event. Electron microscopicexamination of high-density membranes(fraction12) showed that taxol increased thenumbers of small (<500 nm) dense vesicles, with a relative depletionof the larger (>500 nm) vesicles found in controls. These datademonstrate that disruption of endocytic events by taxol includes theearly accumulation of protein and endocytic markers in endosomes and the later accumulation in a dense compartment that we propose is asubdomain of the lysosomes.

In the presence of 1–8 mM Ca2+ unilateral 10–15min irradiation with blue light can elicit negative phototropicbending in the tip-growing coenocytic fresh water alga, Vaucheriaterrestris (Xanthophyceae), when it is simultaneously irradiatedwith strong blue or green background light. By changing wavelength,fluence rate, and duration of background light and holding thoseof unilateral light (456 nm, 1.7 Wm–2) negative phototropicresponse was analyzed: the wavelength of the background lighthad to be shorter than 540 nm; red light (660 nm) was ineffectiveeven at very high fluence rates (100W.m–2). The negativebending was strongly and specifically dependent on the externalCa2+ concentration. Other divalent cations, Mg2+ and Ba2+, wereeither toxic or quite ineffective; Sr2+ could partly supportthe growth, but mediated neither positive nor negative phototropicbending. The rate of tip-growth was not significantly alteredbetween 106M and 10-6 mM Ca2+. Pre-irrradiation with the backgroundlight slightly increased the negative curvature; whereas thatwithout subsequent simultaneous irradiation does not cause negativebending, but rather increases the positive curvature. Three-mindelayed start of background light did not cause negative bendingany more. The present results strongly suggest that blue light elicitsan influx of Ca2+ at the apex of Vaucheria and that the increasedcytoplasmic Ca2+ regulates the sensitivity and direction ofphototropic response. Water extracts of the red-tide dinoflagellate Alexandrium excavatumgrown at ‘high’ light intensity (200 µE m–2s–1) show a broad absorbance maximum in the UV regionof the spectrum (310–360 nm). Using TLC and reverse-phaseHPLC a series of mycosporine-like amino acids have been characterized:mycosporine-glycine (max = 310 nm), palythine (max = 320 nm),asterina-330 (max = 330 nm), shinorine (max = 334 nm), porphyra-334(max= 334 nm), palythenic acid (max = 337 nm) and the isomericmixture of usujirene and palythene (max = 359 nm). From theobserved spectral changes during transference from ‘low’(20 µE m–2 s–1) to ‘high’ (200µE m–2 s–1) light intensities and vice versa,the series of compounds are supposed to be biogenically relatedto one another. The presence of these compounds in A.excavatumis discussed in relation to their possible role in the photoprotectionto deleterious UV radiation.  相似文献

The hypothesis that soil water potential (s) is better correlatedto heliotropic leaf orientation, photosaturated photosyntheticCO2 assimilation and stomatal conductance during periods oflimited water availability than is bulk leaf water potential(1) was examined in greenhouse-grown soybean (Glycine max) plants,submitted to a progressive drought. Paired plants were exposedto either 1000 or 100 µmol m–2 s–1 photonflux densities (PFD) for 45–60 mins. The higher irradianceinduced short-term decreases in 1, due to increased transpiration,while l in the plant exposed to low PFD did not decrease. Thesechanges in 1 occurred independently of changes in soil waterstatus. Concurrent to the light treatments, a single attachedleaf from each of the two plants was isolated from the restof the plant by shading, and the pulvinus of its terminal leafletwas exposed to a perpendicular PFD of 500 µmol m–2S–1. Leaf movement of this leaflet was recorded in responseto this light, until a stable leaflet angle was achieved. Valuesof s and l (before and after light treatment), and photosaturatedrates of photosynthesis and stomatal conductance, were thenmeasured on these leaves. Leaflet angle and gas exchange werebetter correlated with s (r2 = 0.50, 0.50 and 0.57 for angle,photosynthesis and conductance, respectively) than with l especiallywhen l was the result of short-term, high-light induced changesin leaf water status (r2 = 0.36, 0.32 and 0.49, for the sameparameters). Leaflet angle was also correlated with stomatalconductance (r2 = 0.61) and photosynthetic rate (r2 = 0.60),suggesting a close association between leaf orientation, leafmetabolism and soil water availability. In vitrorates of gross and net oxygen production were measuredas a function of light intensity in some plankton communitiescollected from Bedford Basin, Nova Scotia, and in a monoclonalculture of Synechococcus. The rate of gross oxygen productionwas measured by a technique in which the stable oxygen isotope,18O, serves as a photosynthetic tracer Net oxygen productionwas measured by automated Winkler technique. The rate of communityrespiration in the light was then determined by the differencebetween gross and net rates of oxygen production. In the naturalpopulations examined, neither gross nor net oxygen productionrates were significantly inhibited at the highest light intensitymeasured (500–800 µE m–2 s–1) In a samplein which the dark respiration rate was small relative to themaximal rate of production [Pmax;sensu Platt et al (1980) JMar. Res., 38, 687–701] the rates of ‘light’respiration were 3 times greater. In two other communities,with high rates of dark respiration relative to Pmaxthe ratesof ‘light’ respiration were closer to rates of darkrespiration. In the Synechococcus clone, both gross and netoxygen production rates were inhibited at high light intensities.Rates of ‘light’ respiration were found to varyas a function of light intensity. The greatest rates of respirationwere measured in samples incubated at light intensities thatwere just saturating (100 µE m–2 s–1). Three aminopeptidases have been separated from cotyledon extractsfrom cowpea, Vigna unguiculata (L.) Walp., and numbered in orderof decreasing affinity for the anion exchange medium DEAE-Sephacel.API showed a wide acceptance of model substrates, with highestactivity under standard conditions against arginyl ß-naphthylamide(NA). AP2 did not act on basic substrates and preferred phenylalanylß-NA. AP3 displayed the narrowest substrate specificity,with strong activity against only alanyl ß-NA andglycyl ß-NA. The chelator 1,10-phenanthroline completelyor almost completely inhibited forms AP1 and AP3, whereas AP2was insensitive to phenanthroline at the same concentration(5 mM). Cell homogenates of dark-grown mung bean hypocotyls were fractionatedinto six fractions (L-0, L-l to L-5) by stepwise sucrose density-gradientcentrifugation. The majority (ca. 84%) of Mg++-activated ATPase activity ofthe 10,000 x g pellet was localized in the L-0 (1.03 d 1.14)and L-l (1.14 d 1.16) fractions. Over 40% of the vesicularmembrane in the L-0 fraction and 60% of the L-l fraction couldbe stained with phosphotungstic acid (PTA)-chromic acid, a selectivestaining for the plant plasma membrane. In vitro binding of 14C-IAA to the fraction components was thegreatest in the L-l fraction among the six. The binding of 14C-IAAto the L-l fraction in vitro was markedly interfered with bythe presence of a high concentration of cold IAA (2 x 10–4M).However, it was not affected by the IAA analogues IPA, IBA andIAN. This indicates that IAA highly specifically binds to theL-l fraction. In vitro specific binding of 14C-IAA to L-l andL-0 was decreased with an increasing acidity from pH 8.0 to5.0. Physiol. 84(2): 544-552, 1998.Myocytes isolated from rat hearts 3 wk aftermyocardial infarction (MI) had decreasedNa+/Ca2+exchange currents(INa/Ca; 3 Na+ out:1Ca2+ in) and sarcoplasmicreticulum (SR)-releasable Ca2+contents. These defects in Ca2+regulation may contribute to abnormal contractility in MI myocytes. Because exercise training elicits positive adaptations in cardiac contractile function and myocardialCa2+ regulation, thepresent study examined whether 6-8 wk ofhigh-intensity sprint training (HIST) would ameliorate some of thecellular maladaptations observed in post-MI rats with limited exerciseactivity (Sed). In MI rats, HIST did not affect citrate synthaseactivities of plantaris muscles but significantly increased thepercentage of cardiac -myosin heavy chain (MHC) isoforms (57.2 ± 1.9 vs. 49.3 ± 3.5 in MI-HIST vs. MI-Sed, respectively;P  0.05). At the single myocytelevel, HIST attenuated cellular hypertrophy observed post-MI, asevidenced by reductions in cell lengths (112 ± 4 vs. 130 ± 5 µm in MI-HIST vs. MI-Sed, respectively;P  0.005) and cell capacitances (212 ± 8 vs. 242 ± 9 pF in MI-HIST vs. MI-Sed, respectively; P  0.015). ReverseINa/Ca wassignificantly lower (P  0.0001) inmyocytes from MI-Sed rats compared with those from rats that were shamoperated and sedentary. HIST significantly increased reverseINa/Ca(P  0.05) without affecting theamount ofNa+/Ca2+exchangers (detected by immunoblotting) in MI myocytes. SR-releasable Ca2+ content, as estimated byintegrating forwardINa/Ca duringcaffeine-induced SR Ca2+ release,was also significantly increased (P  0.02) by HIST in MI myocytes. We conclude that the enhanced cardiacoutput and stroke volume in post-MI rats subjected to HIST aremediated, at least in part, by reversal of cellular maladaptationspost-MI.

Summary A technique has been developed for the investigation of the photopigment involved in the photoperiodic control of reproduction in Japanese quail,Coturnix coturnix. When these photoreceptors were exposed to white or monochromatic light a clear relationship was found between light intensity and the extent of photo-induced luteinizing hormone (LH) secretion. A spectroradiometric investigation of the passage of light through the skull and brain enabled us to illuminate the hypothalamic region with equal numbers of photons at a range of wavelengths. Action spectra were then conducted and showed a photopigment with a peak sensitivity at wavelengths near 500 nm. An excellent match was obtained when the standard absorption spectrum for a rhodopsin was fitted to the action spectrum, suggesting a rhodopsin maximally sensitive at 492 nm. The absolute sensitivity of the photoreceptors was calculated at a range of wavelengths: with light at 500 nm, 2.85×10–12 E·cm–2·s–1 triggered the photoperiodic response. Primary production was measured for 7 years, using the in situ14C-method in hypertrophic Hartbeespoort Dam, South Africa,to examine the influence of light and water temperature on theupper limit of Microcystis aeruginosa production. Water temperaturesvaried from 11 to >25°C and chlorophyll concentrationsreached 6500 mg m–3. The maximum volumetric rate of production(Amax) was 12->8800 mg C m–3 h–1 with areal productions(A) of 69->3300 mg C m–2 h–1 for euphotic zonedepths of <0.5–8.4 m. The intrinsic parameters of phytoplanktonproduction (, Amax/B, Ik) indicated that the phytoplankton populationwas adapted to high light levels. Both Amax/B and Ik were correlatedwith temperature. Under optimal conditions, , the theoreticalupper limit of A, was calculated to be 2.8 g Cm–2 h–1,while the measured rate was 2.5 g Cm–2 h–1. Measuredareal rates exceeding were overestimated due to methodologicalproblems when working with Microcystis scums. Light and watertemperature interacted to yield high production rates: watertemperature through its direct effect on photosynthetic ratesand indirectly in the formation of diurnal mixed layers; lightindirectly through water temperature and directly through itsattenuation and induction of light-adapted physiology in Microcystis.  相似文献

The effect of solar radiation and monochromatic UV radiationon the motility of the filamentous cyanobacterium Phormidiumuncinatum was determined. Solar radiation (mid-day, in midsummerat a location near Lisboa, Portugal) was found to impair motilitywithin about 30 min. This effect is neither a result of a temperatureincrease nor of visible light. The spectral sensitivity determinedusing the Okazaki Largé Spectrograph shows the maximaleffectiveness of radiation of 300 nm. The short time requirementfor the response and the lack of any photoreactivation of motilityargues against DNA being the UV target. Using the min-LD method, light requirements of the L1- and L2-phasesof L. gibba G3 were found to be satisfied by only 5 min illuminationgiven respectively from CT 0:00 to 0:05 and from CT 11:55 to12:00. This rigorous time sense was displayed without any alterationeven in the presence of iron reagents, e.g., 10–5 M o-phenanthroline,10–5M,'-dipyridyl and 10–6 M kinetin, which completely eliminatedcircadian rhythmicity in reproductive (flower production) aswell as vegetative (frond production) response to a light pulsescanning a continuous dark period. Circadian rhythms of metabolicactivities, e.g., active K+ ion uptake and respiratory CO2 output,were not changed at all by the iron reagents. These and relevantresults suggested that in this long-day duckweed, the circadianoscillator, probably located in the meristem and sensitive toiron deficiency, only modulates the frond and flower productionin the meristem and is not related to the critical daylengthmeasurement. Effects of light flux density (LFD) during growth and uptakeassay on induction of transport system and kinetics of transport were studied using the Azolla pinnata-Anabaena azollae association (Azolla). Theinduction and uptake kinetics of the transport system were determined using an automated system that measuredthe NO3 concentration in the growth medium as a function oftime, using an on-line high performance liquid chromatograph(HPLC) with a UV-VIS detector. Full induction of the transport system required about 1.5 to 2.0 h and occurred without any apparent lag phase,regardless of the LFD provided. The level of induction of transport of Azolla grown at 600 µmol m–2s–1 LFD was higher than for that grown at 100 µmolm–2 s–1. Similarly, 600 µmol m–1 s–1LFD during the assay resulted in a higher level of inductionthan did 100 umol m–2 s–1. An increase in the LFDeither during the growth or the assay period increased the uptake rate; however, an increase in LFD duringthe latter period had greater effect. Azolla grown and assayedat 600 umol m–2 s–1 had the highest uptake rate. The uptake rate at 50 cm3 m–3ambient CO2 concentration was initially higher than at 305 cm3m–3, but the uptake rate decreased rapidly with time andeventually dropped below that at 305 cm3 m–3 CO2. Thesedata suggest that the energy required for transport in Azolla may bypass the photosynthetic CO2 fixationand carbon-cycling. Acclimatization to altitude involves an increase in the acutehypoxic ventilatory response (AHVR). Because low-dose dopamine decreases AHVR and domperidone increases AHVR, the increase in AHVR ataltitude may be generated by a decrease in peripheral dopaminergicactivity. The AHVR of nine subjects was determined with and without aprior period of 8 h of isocapnic hypoxia under each of threepharmacological conditions: 1)control, with no drug administered;2) dopamine (3 µg · min1 · kg1);and 3) domperidone (Motilin, 40 mg).AHVR increased after hypoxia (P  0.001). Dopaminedecreased (P  0.01), and domperidone increased (P  0.005) AHVR. The effect of both drugs on AHVR appearedlarger after hypoxia, an observation supported by a significantinteraction between prior hypoxia and drug in the analysis of variance(P  0.05). Although the increasedeffect of domperidone after hypoxia of 0.40 l · min1 · %saturation1[95% confidence interval (CI) 0.11 to 0.92 l · min1 · %1]did not reach significance, the lower limit for this confidence interval suggests that little of the increase in AHVR after sustained hypoxia was brought about by a decrease in peripheral dopaminergic inhibition.

It has been established that Kalanchoe blossfeldiana and Xanthiumpensylvanicum require CO2 during the light period of short daysfor successful photoperiodic induction of flowering, even ifall but the induced leaf are held in normal air. In X. pensylvanicumfloral induction in normal air was independent of the starchstatus of the leaves but when reserves were reduced, lack ofCO2 in the light suppressed floral induction to an even greaterextent. Injection into the induced leaf (Kalanchoe) or leaftip feeding (Xanthium) of carbohydrates, organic and amino acidsor several other metabolites failed to substitute for the CO2requirement for induction. A small response was produced by10 mg ml–1 sucrose in X. pensylvanicum while in normalair 25 parts 10–6 ATP reduced the time to flowering inK. blossfeldiana and 10–4 M proline was inhibitory. Anexperiment on the light requirement established a need for redlight ( max 660 nm) during photoperiods but red light alonedid not facilitate maximal induction. It is concluded that someearly, possibly labile, product of photosynthetic CO2 fixationis essential to floral induction in these species.  相似文献

Smith, J. R., Smith, F. A. and Walker, N. A. 1987. Potassiumtransport across the membrane of Chara. I. The relationshipbetween radioactive tracer influx and electrical conductance.—J.exp. Bot. 38:731–751. The 42K influx () and the electrical conductance (Gm) were measured simultaneously for the ‘membrane’of internodal cells of Chara australis as a function of theexternal [KCl] (K?. In bathing solutions of pH = 5?0, progressively increased from 20?5to 430?60 nmol m–2 s–1 and Gm increased from 0?36?0?02to 3?8?0?8 S m–2 when K? was increased from 0?1 to 10mol m–3. The resting membrane potential difference (p.d.)was approximately -135 mV for low K? and approached the expectedNernst equilibrium p.d. for K+ ions when K? > 1?0 mol m–3.Measurements of 36Cl influx suggested that the 42K influx waspredominantly electrogenic. The equivalent Goldman permeabilityto K+ ions (Pk) was approximately 20–30 nm s–1 anddid not vary significantly with increasing K?. The equivalentconductance attributable to the electrogenic transport of K+ ions was calculated from assuming passive, independent diffusionof K+ ions and the ratio was found to be typically close to one. It was also found that themagnitudes of and Gm measuredsimultaneously for each individual cell were also well correlatedfor K? 1?0 mol m–3, and that the slope of the line ofbest fit was close to one. For each K? it was found that theconductance not attributable to K+ translocation and presumablyassociated primarily with the transport of protons or theirequivalents was typically 0?2–0?5 Sm–2. For K? >1?0 mol m–3 the results indicated that the transport ofK+ ions was essentially independent, i.e. there was no evidencefor flux interactions. The results also indicated that the equivalentconductance derived from the measured 42K influx could usefullyindicate the fraction of the electrical conductance attributableto the translocation of K+ ions. The luciferin-luciferase method was used to determine ATP extractedfrom darkmaintained and light-exposed samples of the green algaChlorella pyrenoidosa and of the blue-green alga Anacystis nidulans.A few measurements on Synechococcus lividus (a bluegreen thermophile,clone 65?C) are also reported.
  1. The light-minus-dark ATP levels (ATP) from aerobic cells ofChlorella and Anacystis were negative; however, ATP from Synechococcuswas positive. Large positive ATP was obtained in regularly grown(RG: moderate light) Chlorella treated with oligomycin; darklevels were reduced, light levels remained essentially unaffected.In high-light exposed (HLE) Chlorella, oligomycin reduced bothlight and dark ATP levels, but positive ATP was still obtained.However, in Anacystis, which has a different organization ofthylakoid membrane, oligomycin severely reduced both the lightand the dark ATP levels and the ATP remained negative.
  2. Theoligomycin (12 µM) treated Chlorella and the untreatedAnacystis and Synechococcus show the presence of cyclic photophosphorylationunder conditions in which the non-cyclic electron flow fromphotosystem II to photosystem I is blocked by 10 µM 3-(3,4-dichlorophenyl)-l,l-dimethylurea(DCMU), or not allowed to operate by the absence of CO2. Cyclicphotophosphorylation ranged from 10–30% of the maximumATP in RG, to 40–50% in HLE Chlorella. In RG Chlorella,cyclic and non-cyclic (in the absence of DCMU) photophosphorylation(ATP) saturate at about 103 ergs cm–2 sec–1 and104 ergs cm–2 sec–1 and 104 ergs cm–2 sec–1red (>640 nm) light, respectively; a lag was observed inthe light curve.
  3. In Chlorella, the addition of the photosystemI electron acceptormethyl viologen (MV; 1 mM) increased ATPby twofold. Furtheraddition of DCMU (25 µm) reduced thisto the level observedwith DCMU alone. If 1 mM reduced dichlorophenolindophenol orphenazine methosulphate (DCPIPH2 or PMSH2, respectively)wasadded along with DCMU, the ATP level was 30–40% ofthecontrol. Further addition of MV increased the JATP to be70–80%of that of the control. These and other resultsconfirm thepresence of both non-cyclic and cyclic photophosphorylationin vivo, the former predominating in Chlorella, and the latterin Anacystis and Synechococcus.
