X-ray crystal and other structural analyses indicate that Yb(III) and all four newly synthesized ligands can form a binuclear Yb(III) complex with a 1:1 metal to ligand stoichiometry by octacoordination at the Yb(III) center. Investigations of DNA binding properties show that all the ligands and Yb(III) complexes can bind to Calf thymus DNA through intercalations with the binding constants at the order of magnitude 105–107 M−1, but Yb(III) complexes present stronger affinities to DNA than ligands. All the ligands and Yb(III) complexes may be used as potential anticancer drugs. Investigations of antioxidation properties show that all the ligands and Yb(III) complexes have strong scavenging effects for hydroxyl radicals and superoxide radicals but Yb(III) complexes show stronger scavenging effects for hydroxyl radicals than ligands. Two tetranuclear copper(II) complexes with long rigid Schiff-base ligands [Cu4(bmsbd)2]·2DMF [H2bmsbd = N,N′-bis-(3-methoxy-salicylidene)benzene-1,4-diamine] (1) and [Cu4(besbd)2]·DMF·5H2O [H2besbd = N,N′-bis-(3-methoxy-salicylidene)benzene-1,4-diamine] (2) have been synthesised and characterized. Single X-ray diffraction analysis reveals that both complexes exhibit interesting [2 × 2] molecular grid-like configuration assembled by four organic ligands in a head-to-tail mode to bind four copper(II) ions. Magnetic property investigations reveal weak antiferromagnetic interaction viaσ-bond pathway of long rigid Schiff-base ligand between the neighboring copper(II) ions in both complexes.  相似文献

The new chiral macrocyclic complexes [1,2-bis(1H-benzimidazol-2-yl)-1-(1,8-dihydro-1,3,5,8,10,12-hexaazacyclotetradecane)-2-hydroxyethanolate] copper(II) and -nickel(II) perchlorate, 3 and 4, respectively, were synthesized by the reaction of 1,2-bis(1H-benzimidazol-2-yl)ethane-1,2-diol (L) and (1,8-dihydro-1,3,5,8,10,12-hexaazacyclotetradecane)copper(II) and -nickel(II) diperchlorate complexes, 1 and 2, respectively. All complexes were characterized by various spectroscopic techniques. Molar-conductance measurements showed that all of the complexes are ionic in nature. In complexes 3 and 4, the metal center is encapsulated by the ligand L in a pentacoordinated environment. The optical-rotation values ([alpha](D)) of 3 and 4 at 25 degrees indicate that the complexes are chiral. Absorption- and fluorescence-spectral studies, cyclic voltammetry, and viscosity measurements have been carried out to assess the comparative binding of complexes 1 and 3 with calf thymus (CT)-DNA. Analysis of the results suggests that the new chiral complex 3 binds to CT-DNA through a partial intercalation mode that is different from the binding mode of parent achiral complex 1. The complexes 1 and 3 bind to CT-DNA with binding constants K(b) of 2.7 x 10(4) and 6.6 x 10(4) M(-1), respectively. Circular-dichroism (CD) studies have been further employed to ascertain the binding mode of complex 3, which is consistent with the other spectral studies.  相似文献

A new ligand, N,N′-dibenzylethane-1,2-diamine (L) and its four transition metal(II) complexes, ML2(OAc)2 · 2H2O (M = Cu, Ni, Zn, Co), have been synthesized and characterized by elemental analysis, mass spectra, molar conductivity, NMR and IR. Moreover, the crystals structure of Cu(II) and Ni(II) complexes characterized by single crystal X-ray diffraction showed that the complexes have a similar molecular structure. Ni(II) has an regular octahedral coordination environment complexes, but typical Jahn Teller effect influenced Cu(II) in an elongated octahedral environment. The interaction between complexes and calf thymus DNA were studied by UV and fluorescence spectra measure, which showed that the binding mode of complexes with DNA is intercalation. Under physiological pH condition, the effects of Cu(OAc)2L2 · 2H2O and Ni(OAc)2L2 · 2H2O on human serum albumin were examined by fluorescence. The results of spectroscopic measurements suggested that the hydrophobic interaction is the predominant intermolecular force. The enthalpy change ΔH0 and the entropy change ΔS0 of Cu(OAc)2L2 · 2H2O and Ni(OAc)2L2 · 2H2O were calculated to be −11.533 kJ mol−1 and 46.339 J mol−1 K−1, −11.026 kJ mol−1 and 46.396 J mol−1 K−1, respectively, according to the Scatchard’s equation. The quenching mechanism and the number of binding site (n ≈ 1) were also obtained from fluorescence titration data.  相似文献

The mixed-ligand complexes [Cu(II)(HisLeu)(phen)](+) (1) and [Cu(II)(HisSer)(phen)](+) (2; phen=1,10-phenanthroline) were synthesized and characterized. The intercalative interaction of the Cu(II) complexes with calf-thymus DNA (CT-DNA) was probed by UV/VIS and fluorescence titration, as well as by thermal-denaturation experiments, and the intrinsic binding constants (K(b)) for the complexes with 1 and 2 were 4.2x10(3) and 4.9x10(3) M(-1), resp. Both complexes were found to be efficient catalysts for the hydrolytic cleavage of plasmid pUC19 DNA, as tested by gel electrophoresis, converting the DNA from the supercoiled to the nicked-circular form at rate constants of 1.32 and 1.40 h(-1) for 1 and 2, resp.  相似文献

The synthesis and characterisation of two dicopper(II) and two dinickel(II) macrocyclic complexes, [CuII2LPr] (10), [CuII2LBu] (11), [NiII2LPr] (12) and [NiII2LBu] (13), are reported. The two new Schiff-base macrocycles (LPr)4− and (LBu)4− are isolated as dimetallic complexes 10-13 by the [2+2] condensation of 5,5-dimethyl-1,9-diformyldipyrromethane (9) and 1,3-diaminopropane or 1,4-diaminobutane, respectively, using Cu2+ or Ni2+ template ions. Single crystal X-ray structure determinations carried out on 10-13 show that each metal atom is in a square planar N4 geometry, being bound to two deprotonated pyrrole nitrogen atoms of one dipyrromethane unit and to the two adjacent imine nitrogen atoms. NMR spectra obtained for the two dinickel(II) complexes 12 and 13 show that in CDCl3 solution they are highly symmetrical and diamagnetic.  相似文献

Cr(III) and Cr(VI) have different binding capacity with sparfloxacin, and have different combination modes with calf thymus DNA. Selecting these two different metal ions, the influence of them on the binding constants between sparfloxacin (SPFX) and calf thymus DNA, as well as the related mechanism has been studied by using absorption and fluorescence spectroscopy. The result shows that Cr(III) has weaker binding capacity to SPFX in the SPFX-Cr(III) binary system, but influences the binding between SPFX and DNA obviously in SPFX-DNA-Cr(III) ternary system. However, although Cr(VI) has a stronger binding capacity to SPFX, it has no effect on the binding between SPFX and DNA. Referring to the different modes of Cr(III) and Cr(VI) binding to DNA, the mechanism of the influence of metal ions on the binding between SPFX and DNA has been proposed. SPFX can directly bind to DNA by chelating DNA base sites. If a metal ion at certain concentration binds mainly to DNA bases, it can decrease the binding constants between SPFX and DNA through competing with SPFX. While if a metal ion at certain concentration mainly binds to phosphate groups of DNA, it can increase the binding constants by building a bridge between SPFX and DNA. If a metal ion at certain concentrations binds neither to bases nor phosphate groups in DNA, it will have no effect on the binding constant between SPFX and DNA. Our result supports Palumbo's conclusion that the binding between SPFX and the phosphata groups is the precondition for the combination between SPFX and DNA, which is stabilized through stacking interactions between the condensed rings of SPFX and DNA bases.  相似文献

Four cobalt(III) complexes containing the polypyridine pentadentate ligands N,N-bis(2-pyridylmethyl)amine-N′-ethyl-2-pyridine-2-carboxamide (PaPy3H), N,N-bis(2-pyridylmethyl)amine-N′-[1-(2-pyridylethyl)acetamide (MePcPy3H), and N,N-bis(2-pyridylmethyl)amine-N′-(2-pyridylmethyl)acetamide (PcPy3H), have been synthesized. All three ligands bind the Co(III) center in the same fashion with the exception of loss of conjugation between the carboxamide moiety and the pyridine ring in the latter two. The structures of [(PaPy3)Co(OH)][(PaPy3)Co(H2O)](ClO4)3 · 3H2O (1), [(PaPy3)Co(NO2)](ClO4) · 2MeCN (2), [(MePcPy3)Co(MeCN)](ClO4)2 · 0.5MeCN (3), and [(PcPy3)Co(Cl)](ClO4) · 2MeCN (4) have been determined. These ligands with strong-field carboxamido N donor stabilize the +3 oxidation state of the Co center as demonstrated by the facile oxidation of the corresponding Co(II) complexes (prepared in situ) by H2O2, [Fe(Cp)2](BF4), or nitric oxide (NO). The Co-Namido bond distances of 1-4 lie in the narrow range of 1.853-1.898 Å. 1H NMR spectra of these complexes confirm the low-spin d6 ground states of the metal centers.  相似文献

The following Schiff bases were employed as ligands in synthesizing copper(II) and zinc(II) complexes: N-[(2-pyridyl)-methyl]-salicylimine (Hsalampy), N-[2-(N,N-dimethyl-amino)-ethyl]-salicylimine (Hsaldmen), and N-[(2-pyridyl)-methyl]-3-methoxy-salicylimine (Hvalampy). The first two ligands were obtained by reacting salicylaldehyde with 2-aminomethyl-pyridyne and N,N-dimethylethylene diamine, respectively, while the third one results from the condensation of 3-methoxysalicylaldehyde with 2-aminomethyl-pyridine. Four new coordination compounds were synthesized and structurally characterized: [Cu(salampy)(H2O)(ClO4)] 1, [Cu2(salampy)2(H2trim)2] 2 (H2trim? = the monoanion of the trimescic acid), [Cu4(valampy)4](ClO4)4 · 2CH3CN 3, and [Zn3(saldmen)3(OH)](ClO4)2 · 0.25H2O 4. The crystal structure of 1 consists of supramolecular dimers resulted from hydrogen bond interactions established between mononuclear [Cu(salampy)(H2O)(ClO4)] complexes. Compound 2 is a binuclear complex with the copper ions connected by two monoatomic carboxylato bridges arising from two molecules of monodeprotonated trimesic acid. The crystal structure of 3 consists of tetranuclear cations with a heterocubane {Cu4O4} core, and perchlorate ions. Compound 4 is a trinuclear complex with a defective heterocubane structure. The magnetic properties of complexes 13 have been investigated. Compound 4 exhibits solid-state photoluminescence at room temperature.  相似文献

The reactions of Re(CO)5Cl with the chelating ligands 2-(2-pyridyl)-N-methylbenzimidazole, 2-(2-pyridyl)benzoxazole and 2-(2-pyridyl)benzothiazole afforded neutral fac-Re(CO)3(L)Cl and ionic complexes with structures confirmed by means of X-ray measurements. UV-vis absorption and emission properties have been studied at room and 77 K temperatures in order to determine the nature of the lowest electronically excited states. Electrochemical behaviour of the investigated fac-Re(CO)3(L)Cl and complexes has been studied using cyclic and square-wave voltammetry. Preliminary results from the electrogenerated chemiluminescence studies of the ionic and the neutral fac-Re(CO)3(MPBI)Cl complexes are briefly presented.  相似文献

Epichlorohydrin (a probable human carcinogen) was allowed to react with adenosine and the adducts were characterized by NMR and UV spectroscopy, and mass spectrometry. The adduct initially formed was 1-(3-chloro-2-hydroxypropyl)-adenosine, which subsequently ring closures to 1,N(6)-(2-hydroxypropyl)-adenosine at neutral and basic conditions. At acid conditions, the N-1 adduct undergoes a slow deamination to yield 1-(3-chloro-2-hydroxypropyl)-inosine. Minor adducts identified were 7-(3-chloro-2-hydroxypropyl)-adenosine and 3-(3-chloro-2-hydroxypropyl)-adenosine which are easily deglycosylated, and an adduct where the epichlorohydrin residue was attached to the sugar moiety of adenosine. A diadduct, 1,N(6)-(2-hydroxypropyl)-N(6)-(3-chloro-2-hydroxypropyl)-adenosine was also identified. The reaction of epichlorohydrin with calf thymus DNA gave 1,N(6)-(2-hydroxypropyl)-deoxyadenosine and 3-(3-chloro-2-hydroxypropyl)-adenine (major adduct).  相似文献

Optically active Schiff base ligands have been formed by the condensation of various salicylaldehydes with a series of chiral amino alcohols. The Eu(III) derivatives of these ligands were obtained as oligomeric materials, exhibiting an empirical 1:1 metal:ligand stoichiometry. The complexes were found to be somewhat soluble in chloroform, where they existed primarily as trimeric species. The optical activity experienced by the Eu(III) ion was observed to be dominated by the presence of configurational effects, corresponding to a dissymmetric arrangement of Schiff base ligands about the Eu(III) ion.  相似文献

Two series of complexes of the type [CoIII{(naph)2dien}(amine)]BPh4 {(naph)2dien = bis-(2-hydroxy-1-naphthaldimine)-N-diethylenetriamine dianion, and amine = piperidine (pprdn) (1), pyrrolidine (prldn) (2), pyridine (py) (3), N-methylimidazole (N-MeIm) (4)}, and [CoIII{(naph)2dpt}(amine)]BPh4 {(naph)2dpt = bis-(2-hydroxy-1-naphthaldimine)-N-dipropylenetriamine dianion, and amine = piperidine (pprdn) (5), 3-methylpyridine (3-Mepy) (6)} have been synthesized and characterized by elemental analyses, IR, UV-Vis, and 1H NMR spectroscopy. The crystal structures of (2) and (6) have been determined by X-ray diffraction. The redox potentials of the central cobalt ion show a relatively good correlation with the σ-donor ability of the axial ligands. The spectroscopic and electrochemical properties of these complexes are also influenced by the mutual steric hindrance between the pentadentate Schiff base and the ancillary ligands.  相似文献

Four gold(III) complexes of terpyridine derivatives 14 have been synthesized and characterized by spectroscopic methods. In vitro data demonstrated that all of them showed higher cytotoxicity than cisplatin against the human non-small-cell lung cancer cell line (A-549), the human stomach carcinoma cell line (SGC-7901), the human cervix carcinoma cell line (HELA), the human colon carcinoma cell line (HCT-116), the human liver carcinoma cell line (BEL-7402), the murine leukemia cell line (P-388) and the human acute promyelocytic leukemia cell line (HL-60). Complex 3 exhibits the highest activity, with growth inhibition rates of over 80% at 10−8 mol L−1 against the A-549, HCT-116 and HELA tumor cell lines. Interestingly, ligands L1–L4 are also very cytotoxic against the cell lines tested. Complexes 14 are stable in aqueous solution for 2 days in the presence of the biological reducing agent glutathione. The inductively coupled plasma mass spectrometry data showed that DNA isolated from cells treated with complexes 1 and 3 contained gold with gold-to-nucleotide ratios of approximately 1:6,400 and 1:4,900, respectively. Fluorescence titration, UV and circular dichroism analyses proved that the steric and electrostatic effects of the ligand remarkably influence the interactions of their gold(III) complexes with DNA. The DNA binding ability of the complexes has been correlated with their cytotoxicity, which could potentially provide a new rationale for the future design of terpyridine-based metal complexes with antitumor potential.Electronic Supplementary Material Supplementary material is available to authorized users in the online version of this article at .  相似文献

Three new C3-symmetric tritopic ligands with a central phloroglucinol bridging unit have been synthesized and characterized. The ligands are accessible through Schiff-base condensation of 2,4,6-triformylphloroglucinol with 2-aminomethylpyridine (H3tfpg-ampy), N,N-bis(pyridin-2-ylmethyl)-ethylenediamine (H3tfpg-unspenp), and benzhydrazide (H6tfpg-bhy). These ligands differ in nature and number of the donor atoms within the resulting binding pockets. Based on these ligands the synthesis of the first trinuclear phloroglucinol-bridged nickel(II) complexes with three octahedrally coordinated nickel centers is reported. The ligands H3tfpg-ampy and H6tfpg-bhy, which provide tridentate binding pockets, react with nickel(II) perchlorate in the presence of bis(pyridin-2-ylethyl)-amine (bpea) as an additional tridentate capping ligand leading to the formation of the trinuclear complexes [Ni3(tfpg-ampy)(bpea)3](ClO4)3 and [Ni3(tfpg-bhy)(bpea)3](ClO4)3, respectively. Due to the pentadentate binding pocket in ligand H3tfpg-unspenp, no additional coligand is needed and a water molecule occupies the sixth coordination site at the nickel(II) ion resulting in the complex [Ni3(tfpg-unspenp)(H2O)3](ClO4)3. Temperature-dependent magnetic measurements reveal overall weak antiferromagnetic exchange interactions within the trinuclear complex together with a rather strong zero-field splitting (ZFS) for the nickel(II) ions. The observed isotropic coupling constants for the three complexes are in the range of 0.14 < − J < 0.37 cm−1, whereas for the zero-field splitting parameter ∣D∣ values between 1.8 and 5.5 cm−1 are found. This is indicative for competitive spin-polarization and superexchange mechanisms, with the latter prevailing the interaction between the nickel(II) ions through the meta-phenylene-linkage for the complexes reported.  相似文献

Two binuclear 3N-chelated monofunctional PtII complexes, [Pt2L1Cl2]Cl2 (complex III) and [Pt2L2Cl2]Cl2 (complex IV) [L1 = 3,6,9,16,19,22-hexaazatricyclo[22.2.2.211,14]-triaconta-11,13,24,26(1),27,29-hexaene, L2 = 3,6,9,17,20,23-hexaazatricyclo[23.3.1.111,15]-triaconta-1(29),11(30),12,14,25,27-hexaene] have been synthesized and structurally characterized. Structural determination revealed that each PtII center was coordinated by one chloride anion and three N atoms from each diethylenediamine motif. The Pt-Cl bonds in complex III are shorter than those found in complex IV. The rigid para- and meta-xylylene groups make the two complexes adopt a rigid boat-like conformation and a flexible twisted chair-like conformation, respectively. Moreover, complex III has higher tendency to bind with each other than complex IV. DNA binding studies demonstrated that complex IV could bind effectively with calf thymus DNA, possibly via platination of N7 of guanine residue, while no obvious DNA binding was observed for complex III. However, complex III displays a comparable cytotoxicity to cisplatin against HeLa cell line, while compound IV does not show any effective cell inhibition at low concentration. Therefore, the rigid spacers in complexes III and IV play a determining role in their anti-cancer activity and DNA binding ability.  相似文献

The new macrocyclic complex 2-amino-2-deoxy-N-[2-(1,3,5,8,11-pentaazacyclotridecan-3-yl)ethyl]-beta-D-glucopyranosylamine copper(II) dichloride (1a) was prepared and thoroughly characterized by various techniques. Molar-conductance measurements showed that 1a (and its Ni analogue 1b) are ionic in nature. On the basis of spectroscopic data, both complexes were assigned a square-planar geometry, and found to be highly stabile and hydrolytically robust in H2O over a wide range of pH, as confirmed by cyclic voltammetry (CV). The Cu(II) complex 1a was found to bind to CT-DNA, with a binding constant Kb of 2.4x10(3) M(-1), as derived by UV/VIS titration, and confirmed by CV, circular dichroism (CD), and viscosity measurements. DNA binding seems to occur mostly via H-bonding. In an in vitro antitumor MTT assay, 1a exhibited significant anticancer activity against the SY5Y and PC-12 cell lines, with an estimated IC50 value in the micromolar range for SYSY, similar to the standard drug 5-fluorouracil.  相似文献

The acyclic Schiff-base ligands (2-(OH)-5-(R3)C6H2-1,3-(HCNC(R1)(R2)CO2H), derived from the dialdehyde 2-hydroxy-5-R-1,3-benzenedicarboxaldehyde (R = Me or t-Bu) and two equivalents of the amino acids glycine, 2,2-diphenylglycine or phenylalanine, have been reacted with the metal acetates M(OAc)2 (M = Cu, Zn) in the presence of triethylamine, affording the complexes [HNEt3][M2(CH3CO2)2(2-(O)-5-(t-Bu)C6H2-1,3-(HCNC(R1)(R2)CO2)2] (M = Cu, R1 = R2 = C6H5, R3 = Me (1); M = Zn, R1 = R2 = H, R3 = t-Bu (2); M = Zn, R1 = R2 = C6H5, R3 = t-Bu (3); M = Zn, R1 = H, R2 = CH2C6H5, R3 = t-Bu (4)) in good yields. The crystal structures of 1·MeCN, 2·, 3·2MeOH, and 4·3MeOH have been determined.  相似文献

The aim of this study was to synthesize and evaluate plasmid DNA interaction of new platinum(II) complexes with some 2-substituted benzimidazole derivatives as carrier ligands which may have potent anticancer activity and low toxicity. Twelve benzimidazole derivatives carrying indole, 2-/or 3-/or 4-methoxyphenyl, 4-methylphenyl, 3,4-dimethoxyphenyl, 3,4,5-trimethoxyphenyl, 4-methoxybenzyl, 3,4,5-trimethoxybenzyl, 3,4,5-trimethoxystyryl, 3,4,5-trimethoxybenzylthio or dimethylamino ethyl groups in their position 2 and twelve platinum(II) complexes with these carrier ligands were synthesized. The chemical structure of the platinum complexes have been characterized by their elemental analysis and FIR, 1H NMR and mass spectra and their 1H NMR and FIR spectra were interpreted by comparison with those of the ligands. The interaction of all the ligands and their complexes with plasmid DNA and their restriction endonuclease reactions by BamHI and HindIII enzymes were studied by agarose gel electrophoresis. It was determined that complex 1 [dichloro-di(2-(1H-indole-3-yl)benzimidazole)platinum(II)·2H2O] has stronger interaction than carboplatin and complex 10 [dichloro-di(2-(3,4,5-trimethoxystyryl)benzimidazole)platinum(II)·2H2O] has stronger interaction than both carboplatin and cisplatin with plasmid DNA.  相似文献
