The dynamics and the level of accumulation of small heat shock proteins (sHSP group 21–27) after a heat exposure were studied in three Drosophila species differing in thermotolerance. The southern species Drosophila virilis, having the highest thermotolerance, surpassed thermosensitive D. lummei and D. melanogaster in the level of sHSPs throughout the temperature range tested. The results suggest an important role of sHSPs in the molecular mechanisms of adaptation to adverse environmental conditions, particularly to hyperthermia.  相似文献

Expression level and kinetics of accumulation of small heat shock proteins (21-27 kDa group) have been investigated in three Drosophila species differing significantly by temperature niche and thermosensitivity. It was shown that low-latitude thermotolerant species D. virilis exceeds the high-latitude thermosensitive closely-related species D. lummei as well as distant thermosensitive species D. melanogaster in terms of small heat shock proteins expression and accumulation after temperature elevation. The data obtained enable to postulate an important role of small heat shock proteins in organism basal thermotolerance and general adaptation to adverse conditions of environment.  相似文献

Waking noncurarized rabbits were subjected to defensive conditioning. Subcutaneous injections of GABA derivative conditioning. Subcutaneous injections of GABA derivative Phenibut (nootropic) (40 mg/kg) were shown to accelerate the acquisition of internal inhibition, to decrease and stabilize the time of intersignal reactions, to increase the heart rate, and to decrease the respiration rate. At the early stage of conditioning, Phenibut facilitated movements in response to a conditioned stimulus. Consequently, when using nootropics for normalization and improvement of the CNS operation, one should take into account their effects not only on excitation but also on inhibition in the CNS, as well not only on cognitive processes but also on the somatic state. The findings confirm the involvement of GBABergic neurotransmitter system in the internal inhibitory conditioning.  相似文献

It has been shown in rat experiments that the action of psychostimulants on the learning ability of rats in a T-shape maze and Dombrovskaya's maze depends to a greater degree on the problem complexity rather than on its type. The learning ability was enhanced in animals which responded to stress by the manifestations of the avoidance conditioned reflex. Sydnocarb and amphetamine were capable to abolish "passive" responding to stress and to enhance the learning ability. Individual level of the animals' investigative effort affected insignificantly the changes in the learning ability induced by psychostimulants (sydnocarb, amphetamine, caffeine, acephen, piracetam, euclidan).  相似文献

The study investigated the effects of internal (DMSO, 1,2-propanediol, glycerol, ethylene glycol, methanol, N,N-dimethylacetamide) and external cryoprotectants (glucose, sucrose) on the viability and on morphometric parameters of zebrafish embryos. From the tested internal cryoprotectants, DMSO had the lowest toxicity, followed by 1,2-propanediol and glycerol. The external cryoprotectants were less toxic then the internal ones. Early ontogenetic stages were more sensible to cryoprotectant exposure than advanced stages. Two-step incubation procedures in increasing concentrations of internal and external cryoprotectants were superior to multiple-step exposure procedures. All tested vitrification solutions exceeded the tolerance limit of embryos. The tolerance of zebrafish embryos to cryoprotectants was highly variable in a concentration range causing approximately 50% embryo mortality. The width of the perivitelline space showed significant morphometrical changes due to cryoprotectant exposure. In the germinative tissue non-significant changes occurred. The yolk did not change morphometrically after exposure to internal cryoprotectants and showed no sign of dehydration after exposure to external cryoprotectants. Based on these results the study comes to the following conclusions: as yolk dehydration was impossible and as vitrification solutions were over the tolerance limit it seems unlikely that successful vitrification of zebrafish embryos can be achieved. Under these considerations slow freezing methods would be a better option as lower cryoprotectant concentrations can be used and embryos can be dehydrated during freezing.  相似文献

The ability of native alpha- and non-coagulating gamma-thrombin to catalyze the hydrolysis of nonspecific high molecular weight substrates was studied using chymotrypsinogen and the oxidized insulin B-chain as substrates. The effect of thrombin on chymotrypsinogen was estimated by the appearance of caseinolytic activity measured by the increase in the number of terminal NH2-groups in the 2,4,6-trinitrobenzol sulfonic acid reaction. The same reaction was used to study the hydrolysis of insulin by thrombin. It was found that the destruction of the additional center necessary for fibrinogen proteolysis during the alpha-thrombin conversion to the gamma-form did not affect the enzyme ability to hydrolyze nonspecific protein substrates. It was assumed that the low efficiency of non-physiological high molecular weight substrate hydrolysis by thrombin is due to the lack of specific remote interactions in the regulatory site outside the enzyme active center.  相似文献

After the frog tongue was adapted for 10 sec to various salts and sugars, the initial phasic component of gustatory neural responses to almost all of quinine hydrochloride (Q-HCl), quinine sulfate (Q-H2SO4). Brucine, caffeine and picric acid was suppressed. 2. Following 10 sec adaptation to acetic acid, the phasic responses to Q-HCl and Q-H2SO4 were unchanged, those to brucine and caffeine were enhanced, and that to picric acid was depressed slightly. 3. The response to any one of Q-HCl, Q-H2SO4. brucine and caffeine was suppressed after adaptation to the other three, while those to picric acid and nicotine were unchanged or enhanced after adaptation to another bitter solution.  相似文献

The objectives of this work were to determine the effect of external (synchronization methods, month, embryo origin and farm effects) and internal factors (age and size of CL, embryo development and quality score, synchronization methods, age of recipient, quality of transfer and reuse of recipients) on a commercial embryo transfer program in a tropical environment. In the program 1466 Holstein-Friesian purchased embryos were implanted to zebu/European crossbred recipients under field conditions. There were 502 pregnancies detected in this large-scale extension programme. Synchronization methods, month, embryo origin, and farm effects were found to have affected the success rate of embryo transfer. Due to the hot climate and large distances between recipient farms, seasonal effects, reused recipient pregnancy results and the effect of embryo development stage differed from previously reported results. Investigation by ultrasonograph showed that embryo loss occurred before 35 days of pregnancy. Under field conditions, routine fetal sexing resulted in <5% misidentification. In conclusion, under tropical conditions external factors have a major influence on the results of pregnancy from embryo transfer.  相似文献

Summary The inner enamel epithelium (IEE) covers the labial tooth aspect as a one cell layer which, when cut sagittally, appears as a longitudinal cell column extending from the tooth origin toward the periphery. Following sudden tooth shortening, the IEE responds by an increased cell production which later declines below normal values. The perturbation affects all cell kinetic parameters; the progenitor compartment, which initially increases, diminishes in size toward end of the experiment. The cell cycle transition times, which initially decline, rise toward the end of the experiment. The mean normal daily cell production rate of 70 cell % (i.e. 70 cells are produced by 100 progenitors) increases to 111 cell % and then declines to a low of 51 cell %. The IEE response typifies the behavior of other cell renewal systems such as intestinal epithelium and epidermis.  相似文献

It has been shown by genetic analysis that the external and internal invertases from Saccharomyces cerevisiae share a common structural gene [Taussig, R., & Carlson, M. (1983) Nucleic Acids Res. 11, 1943-1954]. However, the only amino acid composition of these two forms of invertase reported to date has revealed extensive differences [Gascon, S., Neumann, N.P., & Lampen, J.O. (1968) J. Biol. Chem. 243, 1573-1577]. We have found from amino acid analyses of both enzymes and sodium dodecyl sulfate-polyacrylamide gel analysis of their cyanogen bromide peptides that they are most likely identical in their amino acid sequence. However, the invertases exhibit dramatically different physical properties, particularly in their stability. The most striking difference was in their renaturation following guanidine treatment where it was shown that inactivated external invertase could be renatured completely. Endo-beta-N-acetylglucosaminidase H treated external invertase was restored to 40% of its original activity while internal invertase remained completely inactive. The observed differences may be attributed to the presence and absence of the oligosaccharide moiety in the external and internal invertases, respectively.  相似文献

Two series of experiments were performed in physically untrained subjects. In series A (heat adaptation, HA), seven male subjects were adapted to dry heat (five consecutive days at 55 degrees C ambient air temperature (Ta) for 1 h X day-1) under resting conditions. Before and after HA, the subjects' shivering responses were determined in a cold test (Ta + 10 to 0 degrees C). In series B, eight male subjects underwent mild exercise training (five consecutive days at a heart rate, HR, of 120 b X min-1) under Ta conditions individually adjusted (Ta + 15 to +5 degrees C) to prevent both sweating and cold sensations. Before and after "sweatless training", the subjects were subjected to a combined cold and heat test. During HA the thresholds for shivering, cutaneous vasodilatation (thumb and forearm) and sweating were shifted significantly (p less than 0.05) towards lower mean body temperatures (Tb). The mean decrease in threshold Tb was 0.36 degrees C. "Sweatless training" resulted in a mean increase in work rate (at HR 120 b X min-1) and oxygen pulse of 13 and 8%, respectively. However, "sweatless training" did not change the threshold Tb for shivering or sweating. Neither HA nor "sweatless training" changed the slopes of the relationships of shivering and sweating to Tb. It is concluded that the previously reported lowering of shivering and sweating threshold Tb in long-distance runners is not due to an increased fitness level, but is essentially identical with HA. The decreased shivering threshold following HA is interpreted as "cross adaptation" produced by the stressors cold and heat.  相似文献

A method is given for measuring external cell or vesicle volume and internal vesicle volume using 131I- and 125I-labeled albumin, respectively.
