番茄(Solanum lycopersicum)在育种过程中经历了驯化、改良、分化和渐渗等不同阶段, 在这一选择过程中番茄的果重和风味等均发生了显著改变, 但是目前对于番茄育种过程中代谢物的变化及其遗传基础却不是十分清楚。近期, 中国农业科学院深圳农业基因组研究所黄三文研究组与华中农业大学罗杰研究组利用多组学(变异组、转录组及代谢组)手段系统解析了番茄育种过程中代谢物的变化。结果表明, 在番茄驯化过程中有46个甾醇糖基生物碱类物质(SGAs)含量逐渐降低, 并获得了7个与其中44个物质显著相关的遗传位点。因此, 在番茄育种过程中通过优异等位位点的组合可以显著降低SGAs的含量; 同时发现在番茄以果重为目标的选择过程中, 控制果重基因周围其它基因的“搭车效应”是引起许多代谢物变化的重要遗传因素, 及在育种过程中对某一性状的选择会对其它性状产生重要影响。该研究首次利用多组学手段系统解析了选择对作物代谢物的影响, 为番茄品质改良奠定了良好的理论基础。  相似文献

Important agronomic traits such as fruit quality, harvesting efficiency or production largely depend on flowering time. We have analysed the effect of the overexpression of the Arabidopsis APETALA1 MADS-box gene on vegetative and reproductive growth of tomato. Constitutive expression of APETALA1 in tomato plants has major effects on the length of their growth cycle as well as on their growth habit. Transgenic tomato plants initiated flowering after the production of 6 vegetative nodes as compared to 11 nodes for the wild type plants. Most of tomato 35S:AP1 plants also showed determinate growth habit, similar to the phenotype of self pruning tomato mutants, as well as an initial reduction of their axillary growth. Moreover, development and fertility of flowers were not affected in plants expressing AP1. Consequently, fruit formation in transgenic plants grown under greenhouse conditions occurred normally, which permitted a similar fruit yield compared to control plants. Since traits conferred by AP1 expression are dominant, its expression in tomato breeding lines could provide advantages for the development of new hybrid varieties with shorter generation time, determinate growth, and reduced pruning requirements.  相似文献

Genetic modification of crop plants to introduce desirable traits such as nutritional enhancement, disease and pest resistance, and enhanced crop productivity is increasingly seen as a promising technology for sustainable agriculture and boosting food production in the world. Independently, cultural practices that utilize alternative agriculture strategies including organic cultivation subscribe to sustainable agriculture by limiting chemical usage and reduced tillage. How the two together affect fruit metabolism or plant growth in the field or whether they are compatible has not yet been tested. Fruit-specific yeast S-adenosylmethionine decarboxylase (ySAMdc) line 579HO, and a control line 556AZ were grown in leguminous hairy vetch (Vicia villosa Roth) (HV) mulch and conventional black polyethylene (BP) mulch, and their fruit analysed. Significant genotypexmulch-dependent interactions on fruit phenotype were exemplified by differential profiles of 20 fruit metabolites such as amino acids, sugars, and organic acids. Expression patterns of the ySAMdc transgene, and tomato SAMdc, E8, PEPC, and ICDHc genes were compared between the two lines as a function of growth on either BP or HV mulch. HV mulch significantly stimulated the accumulation of asparagine, glutamate, glutamine, choline, and citrate concomitant with a decrease in glucose in the 556AZ fruits during ripening as compared to BP. It enables a metabolic system in tomato somewhat akin to the one in higher polyamine-accumulating transgenic fruit that have higher phytonutrient content. Finally, synergism was found between HV mulch and transgenic tomato in up-regulating N:C indicator genes PEPC and ICDHc in the fruit.  相似文献

Fruit set and fruit development in tomato is largely affected by changes in environmental conditions, therefore autonomous fruit set independent of fertilization is a highly desirable trait in tomato. Here, we report the production and characterization of male‐sterile transgenic plants that produce parthenocarpic fruits in two tomato cultivars (Micro‐Tom and Moneymaker). We generated male‐sterility using the cytotoxic gene barnase targeted to the anthers with the PsEND1 anther‐specific promoter. The ovaries of these plants grew in the absence of fertilization producing seedless, parthenocarpic fruits. Early anther ablation is essential to trigger the developing of the transgenic ovaries into fruits, in the absence of the signals usually generated during pollination and fertilization. Ovaries are fully functional and can be manually pollinated to obtain seeds. The transgenic plants obtained in the commercial cultivar Moneymaker show that the parthenocarpic development of the fruit does not have negative consequences in fruit quality. Throughout metabolomic analyses of the tomato fruits, we have identified two elite lines which showed increased levels of several health promoting metabolites and volatile compounds. Thus, early anther ablation can be considered a useful tool to promote fruit set and to obtain seedless and good quality fruits in tomato plants. These plants are also useful parental lines to be used in hybrid breeding approaches.  相似文献

This study addresses the physiological response of four tomato genotypes with distinct sensitivity to high NaCl concentrations, with the aim of identifying physiological traits to rank the genotypes’ sensitivity to salt stress. The central hypothesis was that tomato genotypes grown in saline environments show a characteristic salinity-triggered absorption and translocation of ions, leading to a distinct distribution pattern of Na, K, and soluble sugars. Experiments were conducted on two commercial cultivars: one assumed to be sensitive (Solanum lycopersicum L. F1 hybrid Harzfeuer) and one known to be tolerant (S. lycopersicum L. var. edkawi) to high salt concentrations. Furthermore, two wild salt-tolerant relatives (S. pennellii and S. lycopersicum var. cerasiforme) were selected. Based on our results regarding vegetative performance and partitioning of Na, K, glucose, fructose, and sucrose, it is possible to classify the genotype S. lycopersicum F1 hybrid Harzfeuer as moderately sensitive to salt stress and the genotypes S. lycopersicum var. edkawi, S. lycopersicum var. cerasiforme, and S. pennellii as moderately resistant to 210 mM NaCl. Calculations of the percentage of modification revealed non-specific genotype responses for the amount of sodium in roots and leaves, as well as the sucrose concentration and the osmotic potential of leaves. As shown, the salt-induced changes in potassium levels in leaves, and glucose concentration in roots might be used as additional traits to discriminate genotypes regarding their salt-sensitivity. These parameters might be useful when comparisons of fruit production or vegetative performance provide no conclusive indication e.g. due to the distinct growth habits of commercial cultivars selected for fruit productivity and native genotypes having a stronger vegetative development. However, additional studies should be conducted to evaluate a large number of genotypes differing in their NaCl tolerance. Furthermore, plant responses considering the dynamic source-sink relations due to fruit load needs to be considered.  相似文献

The effects of suppressing deoxyhypusine synthase (DHS) have been examined in tomato (Solanum lycopersicum cv UCT5). DHS mediates the first of two sequential enzymatic reactions that activate eukaryotic translation initiation factor-5A (eIF-5A) by converting a conserved Lys to the unusual amino acid, deoxyhypusine. DHS protein levels were suppressed in transgenic plants by expressing the 3'-untranslated region of tomato DHS under regulation of the constitutive cauliflower mosaic virus promoter. Fruit from the transgenic plants ripened normally, but exhibited delayed postharvest softening and senescence that correlated with suppression of DHS protein levels. Northern-blot analysis indicated that all four gene family members of tomato eIF-5A are expressed in fruit, and that three are up-regulated in parallel with enhancement of DHS mRNA as the fruit begin to senesce and soften. Transgenic plants in which DHS was more strongly suppressed were male sterile, did not produce fruit, and had larger, thicker leaves with enhanced levels of chlorophyll. The activity of PSII was 2 to 3 times higher in these transgenic leaves than in corresponding leaves of wild-type plants, and there was also enhanced deposition of starch in the stems. The data collectively indicate that suppression of DHS has pleiotropic effects on growth and development of tomato. This may, in turn, reflect the fact that there is a single DHS gene in tomato and that its cognate protein is involved in the activation of four distinct isoforms of eIF-5A.  相似文献

In this paper, a number of strategies to overcome the deleterious effects of salinity on plants will be reviewed; these strategies include using molecular markers and genetic transformation as tools to develop salinity-tolerant genotypes, and some cultural techniques. For more than 12 years, QTL analysis has been attempted in order to understand the genetics of salt tolerance and to deal with component traits in breeding programmes. Despite innovations like better marker systems and improved genetic mapping strategies, the success of marker-assisted selection has been very limited because, in part, of inadequate experimental design. Since salinity is variable in time and space, experimental design must allow the study of genotype x environment interaction. Genetic transformation could become a powerful tool in plant breeding, but the growing knowledge from plant physiology must be integrated with molecular breeding techniques. It has been shown that the expression of several transgenes promotes a higher level of salt tolerance in some species. Despite this promising result, the development of a salt-tolerant cultivar by way of transgenesis has still not been achieved. Future directions in order to overcome the present limitations are proposed. Three cultural techniques have proved useful in tomato to overcome, in part, the effects of salinity: treatment of seedlings with drought or NaCl ameliorates the adaptation of adult plants to salinity; mist applied to tomato plants grown in Mediterranean conditions improves vegetative growth and yield in saline conditions; and grafting tomato cultivars onto appropriate rootstocks could reduce the effects of salinity.  相似文献

Harnessing natural variation is an important aspect of modern marker assisted breeding. Traditionally breeding programmes have focused on increased yield and resistance to biotic and abiotic pressures. However, consumer demands for improved quality have lead to increased effort into the breeding of nutritional quality traits in crop plants. In the present study, health-related phytochemicals (carotenoids, tocopherols and phenolics) present in green, yellow and red wild relatives of tomato have been analyzed during fruit development and ripening. This study shows that the differences in the final colour of the fruits were due to a distinct accumulation of carotenoids mainly related to the expression of the phytoene synthase-1 gene (Psy-1). In ripe red-fruited tomatoes, the different deposition of pigments gave rise in some cases to colour differences visually discernible by the consumer. Important quantitative differences between and across taxa were noticed for the in vitro antioxidant activity (AA) of the samples.  相似文献

Tomato represents an important source of fiber and nutrients in the human diet and is a central model for the study of fruit biology. To identify components of fruit metabolic composition, here we have phenotyped tomato introgression lines (ILs) containing chromosome segments of a wild species in the genetic background of a cultivated variety. Using this high-diversity population, we identify 889 quantitative fruit metabolic loci and 326 loci that modify yield-associated traits. The mapping analysis indicates that at least 50% of the metabolic loci are associated with quantitative trait loci (QTLs) that modify whole-plant yield-associated traits. We generate a cartographic network based on correlation analysis that reveals whole-plant phenotype associated and independent metabolic associations, including links with metabolites of nutritional and organoleptic importance. The results of our genomic survey illustrate the power of genome-wide metabolic profiling and detailed morphological analysis for uncovering traits with potential for crop breeding.  相似文献

The salt-tolerant capability of the candidate bioenergy crop prairie cordgrass greatly surpasses that of previously characterized prairie grass species and most other plants. To understand the mechanism of inherited salt tolerance, we compared phenotypic and genetic qualities in half-sib families of prairie cordgrass after salt treatment. Each family was treated with a 400 mM NaCl solution or a water control and then measured for various health phenotypes. Phenotypes associated with salt tolerance were shown to be moderately heritable between parent and offspring. RNA-seq analysis revealed differential regulation in unique pathways including metabolism, signaling, photosynthesis, and the circadian rhythm. The studies herein suggest that alternative regulation of the photosynthetic pathway could confer increased salt resistance in halophytes and can be monitored phenotypically or genetically in breeding programs. The improvement of salt-tolerant traits in prairie cordgrass would increase its potential to be grown as a bioenergy crop on lands that are not suitable for the growth of food crops.  相似文献

Epidemiological and clinical studies indicate that a steady dietary intake of bioavailable lycopene, a C40 carotenoid and potent natural antioxidant, may be associated with a decreased incidence of prostate cancer in humans. Since fresh tomatoes and processed tomato products represent approximately 85% of the average human??s dietary lycopene intake, the identification of novel genetic factors which regulate high fruit lycopene content in tomato is imperative for the improvement of nutritional quality in this commercially valuable specialty crop. To understand the genetic control of the extraordinarily high fruit lycopene content in an accession (LA2093) of the tomato wild species Solanum pimpinellifolium, a quantitative trait locus (QTL) mapping study was conducted using a recombinant inbred line (RIL) population of a cross between LA2093 and a cultivated tomato (S. lycopersicum) breeding line, NCEBR-1. The parental lines, F1 progeny, and F7-F10 RIL populations were grown in replicated field trials in four successive years and evaluated for lycopene content as well as several other traits, including fruit fresh weight, soluble solids content, pH of puree, and plant maturity. The lycopene content of ripe fruit was estimated using three methods: high-performance liquid chromatography (HPLC), spectrophotometry, and colorimetric assays. Based on these measurements, QTL were identified and compared across generations. Among the QTL identified for lycopene, two QTL, located on chromosomes 7 and 12, had very large effects and were consistent across generations. The genomic intervals in which these two QTL reside do not correspond to known map positions of carotenoid biosynthetic genes, indicating that these QTL may represent novel alleles with potentially important implications for tomato breeding as well as increased understanding of carotenoid accumulation in tomato. Several QTL were also identified for fruit weight, soluble solids content and plant maturity. The potential implications of these results for tomato crop improvement are discussed.  相似文献

Fruit size and shape are two major factors determining yield, quality and consumer acceptability for many crops. Like most traits important to agriculture, both are quantitatively inherited. Despite their economic importance none of the genes controlling either of these traits have been cloned, and little is known about the control of the size and shape of domesticated fruit. Tomato represents a model fruit-bearing domesticated species characterized by a wide morphological diversity of fruits. The many genetic and genomic tools available for this crop can be used to unraveal the molecular bases of the developmental stages which presumably influence fruit architecture, size and shape. The goal of this review is to summarize data from the tomato QTL studies conducted over the past 15 years, which together allow the identification of the major QTLs responsible for fruit domestication in tomato. These results provide the starting point for the isolation of the genes involved in fruit-size/shape determination in tomato and potentially other fruit-bearing plants. Tomato (Solanum lycopersicum), which is used for both processing and fresh markets, is a major crop species that is the top ranked vegetable produced over the world. Tomato is also a model species for research in genetics, fruit development and disease resistance. Genetic resources available in public repositories comprise the 12 wild related species and thousands of landraces, modern cultivars and mutants. In addition, high quality genome sequences are available for cultivated tomato and for several wild relatives, hundreds of accessions have been sequenced, and databases gathering sequence data together with genetic and phenotypic data are accessible to the tomato community. Major breeding goals are productivity, resistance to biotic and abiotic stresses, and fruit sensorial and nutritional quality. New traits, including resistance to various biotic and abiotic stresses and root architecture, are increasingly being studied. Several major mutations and quantitative trait loci (QTLs) underlying traits of interest in tomato have been uncovered to date and, thanks to new populations and advances in sequencing technologies, the pace of trait discovery has considerably accelerated. In recent years, clustered regularly interspaced short palindromic repeats (CRISPR)/Cas9 gene editing (GE) already proved its remarkable efficiency in tomato for engineering favorable alleles and for creating new genetic diversity by gene disruption, gene replacement, and precise base editing. Here, we provide insight into the major tomato traits and underlying causal genetic variations discovered so far and review the existing genetic resources and most recent strategies for trait discovery in tomato. Furthermore, we explore the opportunities offered by CRISPR/Cas9 and their exploitation for trait editing in tomato.  相似文献

Tomato (Solanum lycopersicum) is a highly valuable fruit crop, and yield is one of the most important agronomic traits. However, the genetic architecture underlying tomato yield-related traits has not been fully addressed. Based on ∼4.4 million single nucleotide polymorphisms obtained from 605 diverse accessions, we performed a comprehensive genome-wide association study for 27 agronomic traits in tomato. A total of 239 significant associations corresponding to 129 loci, harboring many previously reported and additional genes related to vegetative and reproductive development, were identified, and these loci explained an average of ∼8.8% of the phenotypic variance. A total of 51 loci associated with 25 traits have been under selection during tomato domestication and improvement. Furthermore, a candidate gene, Sl-ACTIVATED MALATE TRANSPORTER15, that encodes an aluminum-activated malate transporter was functionally characterized and shown to act as a pivotal regulator of leaf stomata formation, thereby affecting photosynthesis and drought resistance. This study provides valuable information for tomato genetic research and breeding.

Until recently breeding efforts centred on high-yield production while sacrificing flavour and taste quality traits of mass produced food products, such as tomatoes. The recent publication of Davidovich-Rikanati et al. demonstrates the technical feasibility of the genetical engineering of pathways in tomato plants to modify their fruit flavour profile in a proof-of-concept approach. The reported work ranks among an increasing number of reported successful modifications of edible plants with a focus on the benefits to end-consumers.  相似文献

Morphological, physiological, fruit yield and quality related traits were compared between the seed-grown and tissue-cultured plants of tomato (Lycopersicon esculentum Mill.) cv. Red Coat in a greenhouse. No significant differences were observed for any of the traits studied except for the number of leaves and branches, which were higher in the seed-grown plants than in tissue-cultured plants at the later stages of growth. No phenotypic abnormality of the tissue-cultured plants was observed suggesting that genetic fidelity of tissue cultured plants can be maintained if appropriate plant growth regulators are used with fewer member of subcultures in the multiplication medium.  相似文献

Saline soils constitute a serious production problem for vegetable crops as they are known to suppress plant growth. One of the possible measures to improve crop health in such conditions is to use salt-tolerant bacterial inoculants which can control diseases and promote plant growth. In the present work the ability of Pseudomonas chlororaphis isolate TSAU13 to promote cucumber and tomato plant growth and to improve fruit yield by protecting these plants against soil-borne pathogens in salinated soil were investigated. The bacterial strain stimulated shoot growth (up to 32%), dry matter (up to 43%), and the fruit yield of tomato and cucumber (up to 16%) compared to the uninoculated control plants under saline conditions. The strain was able to survive on the root of 2-month-old plants. 29% of the cucumber and 27% of the tomato plants which had grown in soil to which no Fusarium solani spores had been added were diseased, whereas in the presence of the pathogenic fungus 58% of the cucumber and 52% tomato plants had disease symptoms. P. chlororaphis TSAU13 showed statistically significant disease reduction in comparison to the Fusarium-uninfected and infected control plants. Those results showed that P. chlororaphis TSAU13 has a great biotechnological potential in improvement of vegetable production in commercial greenhouses under saline conditions.  相似文献

Abstract

Tomato quality is a multi-variant attribute directly depending on fruit chemical composition, which in turn determines the benefits of tomato consumption for human health. Commercially available tomato varieties possess limited variability in fruit quality traits. Wild species, such as Solanum pimpinellifolium, could provide different nutritional advantages and can be used for tomato breeding to improve overall fruit quality. Determining the genetic basis of the inheritance of all the traits that contribute to tomato fruit quality will increase the efficiency of the breeding program necessary to take advantage of the wild species variability. A high-density linkage map has been constructed from a recombinant inbred line (RIL) population derived from a cross between tomato Solanum lycopersicum and the wild-relative species S. pimpinellifolium. The RIL population was evaluated for fruit mineral contents during three consecutive growing seasons. The data obtained allowed for the identification of main QTL and novel epistatic interaction among QTL controlling fruit mineral contents on the basis of a multiple-environment analysis. Most of the QTL were flanked by candidate genes providing valuable information for both tomato breeding for new varieties with novel nutritional properties and the starting point to identify the genes underlying these QTL, which will help to reveal the genetic basis of tomato fruit nutritional properties.
