Acidic extracellular pH is characteristic of the cell microenvironment in several important physiological and pathological contexts. Although it is well established that acidic extracellular pH can have profound effects on processes such as cell adhesion and migration, the underlying molecular mechanisms are largely unknown. Integrin receptors physically connect cells to the extracellular matrix, and are thus likely to modulate cell responses to extracellular conditions. Here, we examine the role of acidic extracellular pH in regulating activation of integrin α(v)β(3). Through computational molecular dynamics simulations, we find that acidic extracellular pH promotes opening of the α(v)β(3) headpiece, indicating that acidic pH can thereby facilitate integrin activation. This prediction is consistent with our flow cytometry and atomic force microscope-mediated force spectroscopy assays of integrin α(v)β(3) on live cells, which both demonstrate that acidic pH promotes activation at the intact cell surface. Finally, quantification of cell morphology and migration measurements shows that acidic extracellular pH affects cell behavior in a manner that is consistent with increased integrin activation. Taken together, these computational and experimental results suggest a new and complementary mechanism of integrin activation regulation, with associated implications for cell adhesion and migration in regions of altered pH that are relevant to wound healing and cancer.  相似文献

The effect of a low pH (6, 5 and 4) and different COD/SO42− ratios (9 and 3.5) on thermophilic (55 °C) sulfate reduction and acidification of sucrose was investigated using three upflow anaerobic sludge bed reactors fed with sucrose at an organic loading rate of 3.5 gCOD (lreactor d)−1. The three reactors showed nearly 100% acidification of sucrose for all pH values and COD/SO42− ratios investigated. Sulfate reduction was complete at pH 6 and a COD/SO42− ratio of 9. At pH 5, sulfate reduction efficiencies were 80–95% for both COD/SO42− ratios (9 and 3.5). At pH 4, sulfate reduction efficiencies further dropped to 55–65% at a COD/SO42− ratio of 9 and 30–40% at a COD/SO42− ratio of 3.5. The pH decrease from 6 to 5 or 4 caused a shift in the acidification products from mainly acetate to butyrate, as well as a higher production of ethanol, especially at pH 4. At pH 4, propionate and methane were not formed and hydrogen concentrations in the biogas reached 50%, equivalent to a hydrogen yield of 1.3 mol H2 (mol glucose)−1. This study shows that sulfate reduction is possible in the acidification phase of anaerobic wastewater treatment at pH values as low as 6 till 4 and that the pH strongly affects both the acidification pathways and the sulfate reduction efficiencies.  相似文献

Sinularia leptoclados (Ehrenberg, 1834) is re-described. Sinularia leptoclados var. gonatodes Kolonko, 1926 is synonymized with Sinularia maxima Verseveldt, 1977. Two new species of Sinularia with digitiform lobules, leptoclados-type surface clubs and unbranched interior spindles, are described. An updated maximum likelihood tree of Sinularia species with leptoclados-type clubs (clade 5C) based on two mitochondrial genes (mtMutS, COI) and a nuclear gene (28S rDNA) is presented.  相似文献

Oxidative stimulation induced by hydrogen peroxide (H2O2) on human lens epithelial cells (HLECs) was performed to observe the effects on cell viability and caveolin expression, and cholesterol depletion in HLECs caused by methyl-β-cyclodextrin (MβCD) was also studied. SRA01/04 HLECs were exposed to H2O2 or MβDC of various concentrations and durations. We used a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay to measure the effect of H2O2 on the viability of SRA01/04 HLECs. The distributions of caveolins after oxidative stimulation were probed by fluorescence microscopy and laser scanning confocal microscopy. Immunoblotting was performed to analyze alterations of caveolins expression. We observed that the viability of SRA01/04 HLECs under 0.1 mM H2O2 for 10 min or longer was significantly reduced (*p < 0.05, F = 11.63). Laser scanning microscopy showed immunofluorescent caveolins in SRA01/04 HLECs under 0.1 mM H2O2 for 10 min or longer; caveolins were largely confined to intracellular domains. Western blots showing both membrane and total caveolin protein (22 kDa) levels in SRA01/04 HLECs treated with 0.1, 0.2, 0.5, or 1.0 mM H2O2 for 30 min were significantly reduced, compared with the untreated (*p < 0.05, F = 6.149, or *p < 0.05, F = 14.489, respectively). In addition, the membrane and total caveolin protein level after being treated with 0.1 mM H2O2 (*p < 0.05, F = 6.843, or *p < 0.05, F = 7.944, respectively) for different durations also down regulated. Fluorescence microscopy also showed that phosphorylated caveolin-1 was distributed near the focal adhesions of the cells. This study concludes that the responses of HLECs to oxidative stress may include down regulation of caveolin and phosphorylation of caveolin-1 on tyrosine 14 and that MβCD also down regulates caveolin while depleting cholesterol in HLECs. 为了提高黑曲霉(Aspergillus niger)D08生产木聚糖酶的能力,考察了不同p H条件对菌株生长及产酶的影响,结果发现木聚糖酶发酵过程中菌体生长的最适p H与木聚糖酶合成的最适p H不同,分别为3.5和4.5。由此针对性地提出了两阶段p H控制策略:在30 h内控制p H为3.5;30 h后p H控制为4.5。结果表明:分阶段p H调控策略的实施进一步提高了菌体合成木聚糖酶的能力,木聚糖酶比酶活(2 223.38 U/m L)和生产强度(26.47U/(m L·h))分别比恒定p H 4.5时提高了14.3%和30.6%。该工艺对提高木聚糖酶活力具有一定的指导意义,为后续研究和生产奠定了良好的基础。  相似文献

Acidic mining lakes (ML) in Lusatia (Germany) are characterised by their geogenically determined chemistry. The present study describes the structure, main components and relationships within the food webs of three acidic mining lakes with different pH values (ML 111: pH 2.6; ML 117: pH 2.8; ML Felix: pH 3.6) in order to show their typical characteristics. The investigation covered the period 1995–1997. The number of species and the biomass are both low, but increase with increasing pH. Planktonic components in the most acidic ML 111 (pH 2.6–2.9) comprise bacteria, Ochromonas spp. and Chlamydomonas spp. and a few rotifers (E. worallii, C. hoodi). Heliozoans are the top-predators. In ML 117 (pH 2.8–3) Gymnodinium sp., ciliates, the rotifer B. sericus and the pioneer crustacean Chydorus sphaericus join the pelagial community. Heliozoans were not found in ML 117 or ML Felix (pH 3.4–3.8). ML Felix had the most taxa. The benthic food chain of all three lakes includes phytobenthic algae as producers, chironomids as primary consumers and corixids as top predators in the profundal. Corixids predate on small cladocerans inhabiting the pelagial in lakes with a pH above 2.8 such as ML Felix. They invade the pelagial and act as a connecting link between the benthic and the pelagic food chains, which are isolated in lakes with a lower pH. Occasionally primary producers and consumers were abundant in all three lakes. These organisms do not depend on the degree of acidity, but on the availability of essential ressources. Mass variations covered up any seasonal variation in the extremely acidic ML 111 (0.9 mm3 l–1), while in the other two lakes seasonal patterns of biomass were found.  相似文献

Background

One of the central debates in membrane bioenergetics is whether proton-dependent energy coupling mechanisms are mediated exclusively by protonic transmembrane electrochemical potentials, as delocalized pmf, ΔµH+, or by more localized membrane surface proton pathways, as interfacial pmf, ΔµHS.

Methods

We measure ?pHS in rat liver mitoplasts energized by respiration or ATP hydrolysis by inserting pH sensitive fluorescein-phosphatidyl-ethanolamine(F-PE) into mitoplast surface.

Results

In the presence of rotenone and Ap5A, succinate oxidation induces a bi-phasic interfacial protonation on the mitoplast membranes, a fast phase followed by a slow one, and an interfacial pH decrease of 0.5 to 0.9 pH units of mitoplast with no simultaneous pH changes in the bulk. Antimycin A, other inhibitors or uncouplers of mitochondrial respiration prevent the decrease of mitoplast ?pHS, supporting that ΔµHS is dependent and controlled by energization of mitoplast membranes. A quantitative assay of ATP synthesis coupled with ?pHS of mitoplasts oxidizing succinate with malonate titration shows a parallel correlation between ATP synthesis, State 4 respiration and ?pHS, but not with ?ΨE.

General Significance

Our data substantiate ?pHS as the primary energy source of pmf for mitochondrial ATP synthesis. Evidence and discussion concerning the relative importance and interplay of ?pHS and ?ΨE in mitochondrial bioenergetics are also presented.  相似文献

Searching for materials and technologies of efficient CO2 capture is of the utmost importance to reduce the CO2 impact on the environment. Therefore, the (AlN)n clusters (n = 3–5) are researched using density functional theoretical calculations. The results of the optimization show that the most stable structures of (AlN)n clusters all display planar configurations at B3LYP and G3B3 methods, which are consistent with the reported results. For these planar clusters, we further systematically studied their interactions with carbon dioxide molecules to understand their adsorption behavior at the B3LYP/6–311+G(d,p) level, including geometric optimization, binding energy, bond index, and electrostatic. We found that the planar structures of (AlN)n (n = 3–5) can capture 3–5 CO2 molecules. The result indicates that (AlN)n (n = 3–5) clusters binding with CO2 is an exothermic process (the capture of every CO2 molecule on (AlN)n clusters releases at least 30 kcal mol-1 in relative free energy values). These analysis results are expected to further motivate the applications of clusters to be efficient CO2 capture materials.  相似文献
