Extracellular polysaccharide (EPS) antigens produced by different mould species were purified and partially characterized. Purification included (NH4)2SO4 treatment, Sepharose CL-4B column chromatography and Con A-sepharose chromatography. The EPS of Penicillium digitatum, Mucor racemosus and Cladosporium cladosporioides showed high antigenic capacities. Immunologically the EPS were partially genus-specific, but cross-reactivity was observed. The EPS antigens produced by species of Penicillium, Aspergillus repens and Geotrichum candidum lost their immunological activity upon heating (100C) at pH 18, while the EPS antigen of M. racemosus, Rhizopus oligosporus and C. cladosporioides were stable under the same conditions. The dominant monosaccharides present in the EPS antigen were mannose, galactose and glucose. The EPS obtained from cultures of M. racemosus and R. oligosporus also contained rhamnose. In the EPS produced by Penicillium spp. and A. repens the galactose residues were determined to be immunodominant.  相似文献

Moulds are able to produce extracellular polysaccharide antigens which are heat-stable and almost genus specific. Of 44 different strains of Penicillium 41 (93%) and all 12 strains of Aspergillus tested produced detectable quantities of an immunologically related antigen. Additionally 10 of these 56 strains produced an antigen immunologically related to the antigen produced by the genera Mucor and Fusarium. Immunologically different, but genus-specific antigens were produced by each of the species belonging to the genus Geotrichum, Fusarium, Cladosporium, Mucor and Rhizopus. The antigens produced by Mucor and Rhizopus, however, were immunologically related.  相似文献

The antigenic profiles of 10 mucoraceous fungi--Absidia corymbifera, Mortierella wolfii, Mucor miehei, M. pusillus, M. racemosus, Rhizopus arrhizus, R. microsporus, R. oryzae, R. rhizopodiformis, R. stolonifer,--Candida albicans and Aspergillus fumigatus were compared by crossed immunoelectrophoresis (XIE). Antigen-rich material was obtained from homogenized hyphae (or yeasts in the case of C. albicans), and antisera by multiple subcutaneous innoculation of rabbits with macerated but viable hyphal fragments of Ab. corymbifera, M. pusillus, R. oryzae or Asp. fumigatus. Unique and common antigens were demonstrable amongst the mucoraceous species although Mort. wolfii revealed little antigenic similarity with the others. Considerable sharing of antigens between Ab. corymbifera and M. pusillus was evident. Little or no cross reactivity was seen between extracts of C. albicans and Asp. fumigatus and the mucoraeceous antisera. R. oryzae and R. arrhizus, now regarded as synonymous, revealed close antigenic similarity. On the other hand, the distinction between both M. pusillus and M. miehei--which are regarded by some as belonging to a separate genus Rhizomucor--and less thermotolerant M. racemosus was reflected in their antigenic dissimilarity. Partial separation and characterization of antigens from the crude Absidia extract was achieved by concanavalin A-Sepharose chromatography. Antigens with and without affinity for concanavalin A could be demonstrated. Cross reactivity between Absidia antigens and M. pusillus antiserum appeared to be contained predominantly in material (possibly carbohydrate) which bound to concanavalin A and could be eluted with alpha-methyl-D-mannoside.  相似文献

Mycelial dry weights of Mucor pusillus and Humicola lanuginosa reached maxima after two and eight days of incubation, respectively, in starch-yeast media. Maximum levels of extracellular amylase activities measured in the growth media were recorded after three to 11 days of incubation of M. pusillus, and after 9–25 days of incubation of H. lanuginosa, periods corresponding to observed reductions in mycelial dry weights. In both cases, abnormally high concentrations of reducing sugars were measured in the growth media prior to the attainment of maximum mycelial dry weight. It is suggested that a membrane-bound form of amylase might be principally responsible for providing reducing sugars necessary for growth, and extracellular amylase might be due principally to autolysis.  相似文献

Extracts with immunological activity were prepared from Aspergillus fumigatus, A. flavus, A. terreus, A. niger and A. nidulans. In each case crude mycelial wall was extracted with an aqueous solution of Triton X-100 giving detergent-soluble material. Further fractionation was achieved by removing the detergent from this solution; the resultant precipitate was removed by centrifugation, and the aqueous supernatant was used as a source of soluble antigens. The sensitivity of these preparations was compared with that of water-soluble antigenic material, prepared from whole macerated mycelium, by double diffusion and counterimmunoelectrophoresis using homologous antisera and sera from patients suffering from aspergilloma and allergic bronchopulmonary aspergillosis. The selectivity of these antigenic preparations was monitored with heterologous antisera raised in rabbits. Batch variability was analysed for one strain of A. fumigatus using chemical and immunological methods. The nature of the antigenic sites involved in these reactions was investigated by studying the susceptibility of the preparations to proteolytic hydrolysis, periodate oxidation and concanavalin A treatment. The total protein and carbohydrate content of each fraction was determined and the constituent sugars analysed in an attempt to correlate chemical composition with antigenic activity.  相似文献

A glycoprotein (GP72) has been isolated from Trypanosoma cruzi and found to contain 41% protein, 49% carbohydrate and 10% phosphate. All phosphate was covalently attached to the carbohydrate which contained the following sugars: ribose, xylose, fucose, galactose, mannose, glucose and glucosamine. The carbohydrate side chains were linked to protein by fucose, xylose and N-acetylglucosamine; 50% of the total N-acetylglucosamine was involved in glycoprotein linkages. Two classes of carbohydrate side chains were detected. One class comprised 15% of the total carbohydrate and contained glucosamine, mannose and galactose; some of these chains were phosphorylated. The other class comprised 85% of the total carbohydrate and contained xylose, ribose, fucose, galactose, mannose, glucosamine and phosphate; these chains were antigenic and reacted with a monoclonal antibody with specificity for the whole glycoprotein.  相似文献

Populations of aerobic heterotrophic bacteria, mycelial fungi and yeasts occurring in malting barley were estimated by a plate technique and scanning electron microscopy. There was an increase in the total number of micro-organisms during germination, although populations declined after kilning. Bacteria dominated on all samples, with progressively lower populations of yeasts and filamentous fungi. There was no obvious spatial distribution of micro-organisms on the samples although there appeared to be high populations of bacteria and fungal hyphae on the inner surface of the kernels. The dominant groups of aerobic heterotrophic bacteria were presumptively identified as Alcaligenes sp., Arthrobacter globiformis, Clavibacter iranicum, Erwinia herbicola, Lactobacillus spp. and Pseudomonas fluorescens. The principal filamentous fungi were identified as Aiternaria alternata, Aspergillus glaucus (group), Cladosporium macrocarpum, Epicoccum purpurascens, Fusarium avenaceum, Geotrichum candidum and Penicillium spp. The yeasts isolated most frequently were Candida catenulata, C. vini, Debaryomyces hansenii, Hansenula polymorpha, Kloeckera apiculata, Rhodotorula mucilaginosa, Sporobolomyces roseus and Trichosporon beigelii.  相似文献

Almost 300 common fungi were screened for synthesis of extracellular keratinases. About 54% of the fungi grew on agar plates with soluble keratin and excreted the enzymes. Some representatives of Fusarium , Acremonium and Geotrichum were the most active. However, when cultivating the promising strains in submerged conditions in a medium with porcine nail as the sole source of carbon and nitrogen, other fungi proved to be potent. Aspergillus flavus was the most powerful producer of extracellular keratinases, followed by Alternaria radicina , Trichurus spiralis and Stachybotrys atra.  相似文献

对学院植物组织培养实验室部分受污染的组培苗进行了污染物的分类与鉴定 ,发现棒杆菌属 (Corynebac terium)、肠杆菌属 (Enterobacter)、葡萄球菌属 (Staphylococus)、地霉属 (Geotrichum)、曲霉属 (Aspergillus)、毛霉属(Mucor)、根霉属 (Rhizopus)等常见的细菌和真菌是引起组培苗严重污染的主要菌类。取样测试鉴定表明 ,大部分的微生物来源于无菌接种室内的空气、超净工作台鼓风机排出的气体。抑菌试验发现 ,2mg/L的庆大霉素和 0 .3%质量分数的丙酸钠对细菌均有明显的抑制作用。  相似文献

The nature of the antigenic specificities of several antigens associated with the T/t complex in the mouse were analyzed by means of glycosidase and haptene inhibition studies. Results indicate that on testicular cells sugar residues are involved in at least six different T/t antigenic determinants. The immunodominant sugar appears to be different for each of the specificities. The specificity for the following T/t antigens resides predominantly in the sugars indicated: T:sialic acid; t12:beta-D-galactose; tw32:beta-D-galactose; t0:L-fucose; tw1:N-acetyl-D-galactosamine; tw18:L-fucose. It seems probable that these sugars are found at the terminal reducing ends of the carbohydrate portion of T/t-bearing moleculse. These studies imply that at least some of the genes in the T locus code for glycosyltransferases or regulators of glycosyltransferases which modigy oligosaccharide structures and impart specificity to the T/t antigens by alteration of their terminal sugar residues.  相似文献

Forty-nine powdered herbal drugs were analyzed for their mold profile and for the potential presence of three mycotoxins (aflatoxin, sterigmatocystin, ochratoxin A). Aspergillus and Penicillium species were predominant, but Rhizopus, Mucor, Cladosporium, and Aureobasidium spp. were also found in a few samples. Summary Weekly samplings of airborne spores have been carried out with an S.A.S. apparatus in an urban and a rural site in Franche-Comté (France) during two years. The amount of air spora is higher in the rural environment than in the urban atmosphere. Many allergenic genera have been identified. Quantitative seasonal variations of many genera have been observed. The most frequent genera are:Cladosporium, Geotrichum, Epicoccum, Alternaria, Aspergillus, Aureobasidium andPenicillium appear only sporadically.  相似文献

The cell wall protein antigen was solubilized from the isolated cell walls of Clostridium botulinum type A by autolysis and purified by diethylaminoethyl-cellulose column chromatography followed by gel filtration on Sephadex G-150. The two fractions showed a high degree of the serological activity and produced a main fused precipitin line in immunodiffusion tests against the homologous antiserum. The fact that antigenic fractions contained various kinds of amino acids but no detectable amounts of amino sugars or carbohydrates suggests that the antigens were principally composed of proteins. The protein antigen possessed multiple antigenic components on immunoelectrophoresis. As serological activity, the antigen was heat-stable and resistant to tryptic digestion but sensitive to the actions of pronase, nagarse or pepsin. The protein antigen appeared to be responsible for the common antigenicity among the proteolytic strains of C. botulinum.  相似文献

The possibility of using experimental culture medium K-4 prepared on the basis of casein hydrolysate peptides with the isoelectric point 4.1 for obtaining antigens from P. aeruginosa strains was evaluated. Two antigenic fractions were isolated from the culture fluid containing extracellular slime. The study of the toxicity of the antigenic preparations revealed that one of these fractions had low toxicity for mice (the second antigenic fraction was highly toxic). The former P. aeruginosa antigenic fraction was used for obtaining pyocyanic vaccine. One vaccination dose of this vaccine contained 0.2 mg of the antigen adsorbed on aluminum hydroxide. Pyocyanic vaccine ensured the active protection of mice challenged with P. aeruginosa homologous and heterologous strains.  相似文献

从昆明,石林,西双版纳,思茅等地区的菊科植物根系上群,昆明,石林地区市售洋姜表土,洋姜提取液中污染菌及洋姜表面霉烂处,分离纯化产菊粉酶菌株,共获得105株霉菌,对以上菌株进行了初步鉴定,对产酶情况进行了初步的研究。发现分属于拟青霉属(Paecilomyces sp.),头孢霉属(Cephalosporium sp.),阜孢霉属(Papularia sp.),丝衣霉属(Byssochlamys sp.),指轮枝孢霉属(Stachylidium sp.)。刚毛菌属(Lacellina sp.)交链孢属(Alternaria sp.),地霉属(Geotrichun sp.)毛霉属(Mucor sp.),枝孢霉属(Claddosporium sp.),金孢霉属(Chrysosporium sp.),镰刀菌属(Fusarium sp.),根霉属(Rhizopus sp.)等13个避的霉菌,是国内未见报道的新的菊粉酶产生菌。  相似文献

Serum antibodies were analyzed in rabbits immunized with live and formalin-killed Francisella (F. tularensis, F. novicida, F. novicida-like, and F. philomiragia). Passive hemagglutination test with erythrocytes sensitized by these bacteria' LPS showed much higher titers of species-specific antibodies in all sera to live microorganisms than sera to killed bacteria. The results of immunoblotting with purified LPS and bacterial lysates indicate that sera to live bacteria contained mainly immunoglobulins to species-specific antigenic epitopes of LPS O-polysaccharide chain and few antibodies to the protein component of the cell. By contrast, killed bacterial cells induced weak production of antibodies to S-LPS and a pronounced antibody response to protein antigens. Besides the quantitative differences, live and killed bacteria differed by the qualitative spectrum of immunodominant proteins. Serum to live F. tularensis 15/10 contained antibodies to at least 3 immunodominant antigens of the cell, while serum to killed bacteria contained antibodies to only two of these. Immunoglobulins to protein antigens, absent in homologous sera to live bacteria, were detected in the sera to killed F. novicida and F. novicida-like bacteria. Both sera to F. philomiragia had antibodies reacting with LPS epitopes and immunodominant complex containing protein. In contrast to other Francisella, F. philomiragia was found to synthesize an uncommon LPS representing two major lipooligosaccharides with different molecular weights and antigenic specificity. Therefore, immune response of the host to live and killed Francisella is different: live cells more effectively induce the production of antibodies to S-LPS epitopes, while killed ones to protein antigens.  相似文献

采用涂布平板法从甘蔗渣泥中共分离得到5株优势真菌,分别为头孢霉属(Cephalosporium)、曲霉属(Asper-gillus)、木霉属(Trichoderma)、毛霉属(Mucor)和地霉属(Geotrichum)。对分离得到的真菌进行产酶特性的研究。采用两种不同的产酶培养基(培养基Ⅴ,培养基Ⅵ),这两种培养基的无机盐成分、含量相同,它们的主要差别为碳源不同。培养基Ⅴ的主要碳源为葡萄溏,培养基Ⅵ的主要碳源为甘蔗渣(过60目筛)。结果表明:分离得到的真菌在这两种培养基中的产酶酶活力相差很大,最优菌株为木霉属Q-3,在培养基Ⅵ中产酶酶活最高,赖锰过氧化物酶和漆酶最高活性分别为4.2245U/mL、1.2525U/mL,相对应的纤维素酶活性较低。  相似文献
