Summary Evidence is given thatCyclamen persicum callus culture give rise to structures such as shoot-buds, roots, unipolar tubers, bipolar tubers and embryos. Observations made on their structure by means of morphological and histological analyses are reported and discussed in relation to the anatomy of someCyclamen structures such as zygotic embryos and seedling tubers. Such analyses demonstrate that the bipolar structures including somatic embryos and bipolar tubers lack vascular connection with the callus whereas unipolar structures establish vascular strands with the callus. Here, the relationship between these regenerated structures is depicted. The organogenetic pattern inCyclamen persicum callus culture could well be a derived pathway for somatic embryogenesis. A theoretical diagram concerning the morphogenetic program is presented.  相似文献

In reviewing recent research published in Russian on regeneration and asexual reproduction, the following morphogenetic processes in the planarian Dugesia tigrina are considered: 1) regeneration of lost parts of the body; 2) regeneration of the whole worm from fragments of the body, either by normal regeneration when the inital polarity of the fragment is retained or by somatic embryogenesis when one or more new axes of polarity arise; 3) somatic embryogenesis, or development of individuals from somatic cells; 4) hypermorphosis, or the presence of more than the usual number of organs or body parts, a process that can be interpreted in terms of somatic embryogenesis; and 5) asexual reproduction. Some morphological, biochemical, and physiological studies of the division zone in D. tigrina demonstrate peculiarities of a local breakdown of integrative functions, a breakdown which in turn causes division of the individual to take place at this zone; timing of division is controlled by the organism as an integrated whole.  相似文献

We describe a new subfamily of satellite III DNA (pTRS-63), which, by a combination of in situ hybridization to human metaphase chromosomes and analysis of a panel of somatic cell hybrids, is shown to be specific for human chromosome 14. This DNA has a basic 5-bp repeating unit of diverged GGAAT which is tandemly repeated and organized into either one of two distinct higher-order structures of 5 kb (designated the "L" form) or 4.8 kb (designated the "S" form). In addition, a third (Z) form, representing no detectable levels of this satellite III subfamily, is found. Results from five somatic cell hybrid lines and from a number of informative human individuals suggest that, on any one chromosome 14, only one of the three forms may exist. Subchromosomally, this sequence has been mapped to the p11 region and is distal to the domain occupied by another previously described satellite III subfamily (pTRS-47) found on chromosome 14. The pTRS-63 sequence described adds to the understanding of the structural organization of the short arm of human chromosome 14 and should be useful for the investigation of the molecular etiology of the frequently occurring t(13q14q) and t(14q21q) Robertsonian translocations.  相似文献

Somatic embryogenesis is an in vitro morphogenetic route in which isolated cells or a small group of somatic cells give rise to bipolar structures resembling zygotic embryos. Lipids, carbohydrates, and proteins are major compounds in plant and animal metabolism. Comparative analysis along different developmental stages of Acca sellowiana (Myrtaceae) zygotic and somatic embryos, revealed a progressive increase in levels of total lipids. A high degree of similarity could be found in the total lipids composition between A. sellowiana somatic and zygotic embryos. High lipid levels were found in zygotic embryos in the torpedo and cotyledonary stages, and these levels increased according to the progression in the developmental stages. Somatic embryos obtained through direct embryogenesis route showed higher levels of lipids than in indirect somatic embryogenesis. The compounds most frequently were linoleic acid (C18:2), palmitic (C16:0) and oleic (C18:1). These results indicate a high similarity degree of accumulation of total lipids, regardless of zygotic or somatic embryogenesis.  相似文献

Method in establishing mammalian hemodynamic similarity criteria via the classical allometric approach was examined. Metabolic rate (MR) per unit body weight (w), termed the metabolic turn-over rate (MTR), was found to be linearly related to mammalian resting heart rate (fh). The external ventricular work (EW) per unit body weight was found to be constant among mammalian species. The ratio of (EW/w)/(MTR/fh) is dimensionless, and is about 1/70. This latter qualifies as a new similarity principle. It states that the external ventricular work intensity during each cardiac cycle is directly proportional to the metabolic turn-over rate.  相似文献

We evaluated the effect of lipopolysaccharides from the plant-growth-promoting associative bacterium Azospirillum brasilense Sp245 and from the enteric bacterium Escherichia coli K12 on the morphogenic potential of in vitro-growing somatic calluses of soft spring wheat (Triticum aestivum L. cv. Saratovskaya 29). A genetic model was used that included two near-isogenic lines of T. aestivum L. cv. Saratovskaya 29 with different embryogenic capacities; one of these lines carries the Rht-B1 dwarfing gene, whereas the other lacks it. When added to the nutrient medium, the lipopolysaccharide of A. brasilense Sp245 promoted the formation of calluses with meristematic centers and stimulated the regeneration ability of the cultured tissues in both lines. By contrast, the lipopolysaccharide of the enteric bacterium E. coli K12 barely affected the morphogenetic activity of callus cells and the yield of morphogenic calluses and regenerated plants. These findings indicate that the lipopolysaccharide of the plant-growth-promoting associative bacterium A. brasilense Sp245 specifically enhances the morphogenetic activity of wheat somatic tissues, which increases the efficacy of culturing of genotypes with a relatively low morphogenic potential. The results of the study may contribute to the improvement of the efficacy of plant cell selection and gene engineering and to a better understanding of the mechanisms responsible for plant recognition of lipopolysaccharides of associative bacteria.  相似文献

The morphology, morphogenesis, and phylogeny of an undescribed oligotrich sensu lato (s. lat.) ciliate, Lynnella semiglobulosa n. g., n. sp., found in Daya Bay, southern China, were investigated. This species shares some features with both oligotrichs sensu stricto and choreotrichs, but most morphological and morphogenetic characters as well as the phylogenetic analysis suggest that it should be assigned into subclass Choreotrichia temporarily. Lynnella semiglobulosa is distinguished from members of all known genera and families of the subclass Choreotrichia by a unique combination of characteristics of the buccal and somatic ciliatures. Thus, a new family Lynnellidae n. fam. and a new genus Lynnella n. g. are proposed for it. The new family is distinguished by an open adoral zone of membranelles (AZM) in which, however, there are no ventral membranelles; the distal and proximal portions of the new adoral zone lie close to each other forming an open circle in stomatogenesis. The new genus Lynnella is characterized by possessing two longitudinally oriented somatic kineties, one dorsal and one ventral, several proximal membranelles progressively lengthened toward the proximal end of adoral zone, and two macronuclear nodules. In phylogenetic analyses based on small subunit rRNA gene sequences, L. semiglobulosa clustered basally to all choreotrichs, but with relatively weak support; nevertheless, the possibility of a relationship with the subclass Oligotrichia was not rejected by the approximately unbiased nor Shimodaira-Hasegawa test. Based on morphological, morphogenetic, and molecular evidence of L. semiglobulosa, it is confirmed that the open AZM should be a plesiomorphic character of oligotrichs s. lat. as suggested previously.  相似文献

Light and electron microscopical observations on the stomatogenesis of Coleps amphacanthus Ehrenberg, 1833, show that this "gymno"-stome ciliate has a well developed oral ciliature made of 19–23 "paroral dikinetids" and three "adoral organelles." These structures were previously known as "circumoral ciliature" and "dorsal brosse," and it was thought that they originated from the distal ends of all the 22–26 somatic kineties. Contrary to this view, only four stomatogenic kineties (K1, Kn, Kn-1, and Kn-2) are involved in stomatogenesis of the opisthe. All paroral dikinetids arise from one single kinetofragment (KF1) to the right of the oral anlage while the adoral organelles originate from the three left kinetofragments (KFn, KFn-1, and KFn-2). In particular, the future paroral dikinetids perform a complex morphogenetic movement that leads to a situation where the postciliary microtubules of the once posterior kinetosome of each oral dikinetid give rise to the cytopharyngeal microtubular ribbons. The postciliary origin of the cytopharyngeal ribbons which could only be detected by an EM study of stomatogenesis shows that the basket of Coleps belongs to the cyrtos-type and not to the rhabdos-type basket, where transverse microtubules accompany the basket-forming nematodesmata. The taxonomic implications of these observations, which may lead to a revision of the systematic position of the genus Coleps , are discussed.  相似文献

The morphogenetic events that give rise to a specific body pattern have to date avoided extensive elucidation. Extant models of pattern formation have dealt almost exclusively with the "primary patterning" of structures in the embryo. These "universal" models fail to explain many morphological conditions, such as the simultaneous change in position of the limbs, celom, mesonephros, and umbilical artery relative to the somites as a result of a single mutation. In the present paper, we propose that the relation between two non-periodic waves may function to determine the position of "secondary structures", such as the limbs, in the embryo, relative to primary structures such as the somites. We propose that if two morphogenetic events are initiated at different times from the same region of the embryo, and are progressing in a cranio-caudal sequence at different rates, then the location of a given structure along the body axis can be described as a function of the two events. Applications and predictions based on the proposal are presented. Evidence from observations of morphogenetic events in the chick embryo, which tend to support the model, are also presented.  相似文献

Three features contribute to the complexity of an entity: numberof parts, their order, and their iteration. Many functionalbiological entities are complex when measured by those attributes,and although they are produced in tree-like architectures, theorganizational structures that permit them to function are inthe form of hierarchies. Natural hierarchies can be thoughtof as organizing structures that are emergent properties ofcomplex functional entities, and which are transformed fromtrees by process networks. For example, hierarchies are observedin the architecture of metazoan bodies (the somatic hierarchy)and in the biotic structure of ecogeographic units (the ecologicalhierarchy). As the metazoan developmental genome is quite complexand has been evolved through tree-like processes, it must harborat least one hierarchy, which is most clearly indicated in thedevelopmental processes that create the somatic hierarchy. Formulticellular organisms, the processes that serve to transformtrees of gene expression events into a somatic hierarchy haveproduced complicated signaling networks whose histories canprobably be recovered in general outline.  相似文献

Insect societies are hallmarks of cooperation because one or a few queens monopolize reproduction and several non-reproductive workers cooperatively raise brood. However, the loss of the queen exposes a colony to potential reproductive conflict, which is resolved only after a new queen takes over. We studied queen succession in natural and experimental colonies of the primitively eusocial wasp Ropalidia marginata to understand the proximate behavioral strategies involved in the resolution of this conflict. Previous work has shown that in this species, experimental queen removal always results in only one worker becoming hyper-aggressive and taking over the colony as its next queen, without ever being challenged. Here we show that even during natural queen turnover, one and only one worker becomes hyper-aggressive and takes over as the next queen, without being challenged. During natural queen turn-over, aggression of the successor may sometimes begin before the loss of the old queen and may sometimes decline more rapidly, unlike in the case of experimental queen removal. The successor begins to lay eggs sooner after a natural queen turn-over as compared to experimental queen removal. This is expected because workers might detect the gradual decline of the queen preceding her disappearance. Because queen succession is expected to be more prevalent in tropical perennial species, we expect natural selection to have favored such an orderly queen succession so that a route to direct fitness is available without significant reduction in cooperation.  相似文献

We report the identification, via their cDNAs, of genes which are temporarily transcribed during the initiation of somatic embryogenesis in carrot (Daucus carota L.) cells cultured in an auxin-free medium. Their expression is roughly associated with the first morphogenetic, or globular, stage. A cDNA library ( gt 10) was established using poly(A)+ -rich RNAs from cells deprived of auxin for 8 d. By differential screening a number of clones corresponding to early-induced embryogenic genes were identified. For several a temporary accumulation of the specific mRNA between 6 and 16 d after induction was observed. Somatic embryo-like structures were produced from the hypocotyls of aseptic plants of Capsicum chinense. Different concentrations of 2,4-dichlorophenoxyacetic acid (0, 4.5, 9.05 μM), several exposure times of the explant to this auxin (15, 30, 45, 60 days) and the development of somatic embryos cultured in a solid and/or liquid medium were evaluated. As a result, a novel system of regeneration via direct somatic embryogenesis in liquid medium was established, with an efficiency of 1.77 × 104 somatic embryos per liter of medium. Critical stages of embryogenesis, including cellular acquisition of morphogenetic competence, suspensor formation, and development and maturation of somatic embryos, were identified by histological analysis and scanning electron microscopy. Our results show a promising new outlook on the in vitro regeneration of this species. Contrary to what has been reported to date for the Capsicum genus, it is a species of plants with higher embryogenic potential in vitro.  相似文献

Background and Aims

Secondary somatic embryogenesis has been postulated to occur during induction of peach palm somatic embryogenesis. In the present study this morphogenetic pathway is described and a protocol for the establishment of cycling cultures using a temporary immersion system (TIS) is presented.

Methods

Zygotic embryos were used as explants, and induction of somatic embryogenesis and plantlet growth were compared in TIS and solid culture medium. Light microscopy, scanning electron microscopy (SEM) and transmission electron microscopy (TEM) were used to describe in vitro morphogenesis and accompany morpho-histological alterations during culture.

Key Results

The development of secondary somatic embryos occurs early during the induction of primary somatic embryos. Secondary somatic embryos were observed to develop continually in culture, resulting in non-synchronized development of these somatic embryos. Using these somatic embryos as explants allowed development of cycling cultures. Somatic embryos had high embryogenic potential (65·8 ± 3·0 to 86·2 ± 5·0 %) over the period tested. The use of a TIS greatly improved the number of somatic embryos obtained, as well as subsequent plantlet growth. Histological analyses showed that starch accumulation precedes the development of somatic embryos, and that these cells presented high nucleus/cytoplasm ratios and high mitotic indices, as evidenced by DAPI staining. Morphological and SEM observations revealed clusters of somatic embryos on one part of the explants, while other parts grew further, resulting in callus tissue. A multicellular origin of the secondary somatic embryos is hypothesized. Cells in the vicinity of callus accumulated large amounts of phenolic substances in their vacuoles. TEM revealed that these cells are metabolically very active, with the presence of numerous mitochondria and Golgi apparatuses. Light microscopy and TEM of the embryogenic sector revealed cells with numerous amyloplasts, large nuclei and nucleoli, and numerous plasmodesmata. Plantlets were obtained and after 3 months in culture their growth was significantly better in TIS than on solid culture medium. However, during acclimatization the survival rate of TIS-grown plantlets was lower.

Conclusions

The present study confirms the occurrence of secondary somatic embryos in peach palm and describes a feasible protocol for regeneration of peach palm in vitro. Further optimizations include the use of explants obtained from adult palms and improvement of somatic embryo conversion rates.  相似文献

The centrosome (centriole) and the cytoskeleton produced by it are structures, which probably determine differentiation, morphogenesis, and switching on the mechanism of replicative aging in all somatic cells of multicellular animals. The mechanism of such programming of the events seems to include cytoskeleton influences and small RNAs related to the centrosome. 1) If these functions are really related with centrioles, the multicellular organism's cells which: a) initially lack centrioles (e.g., higher plant cells and also zygote and early blastomeres of some animals) or cytoskeleton (e.g., embryonic stem cells); or b) generate centrioles de novo (e.g., zygote and early blastomeres of some animals), will be totipotent and lack replicative aging. Consequently, the absence (constant or temporary) of the structure determining the counting of divisions also means the absence of counting of differentiation processes. 2) Although a particular damage to centrioles or cytoskeleton (e.g., in tumor cells) fails to make the cells totipotent (because the morphogenetic status of these cells, as differentiated from that of totipotent ones, is not zero), but such a transformation can suppress the initiation of the aging mechanism induced by these structures and, thus, make such cells replicatively "immortal".  相似文献

The clinostate rotation causes a wide range of gastrulation abnormalities in the Anura. A special investigation has shown that all gastrulation variants may be represented as various spatial-temporal compositions of the same morphogenetic processes isolated by means of formal experimental procedure. These processes are structurally stable rearrangements of tissue morphology and are spread in the tissue as a wave embracing more and more cells. The change of one local morphology for another may proceed either in the form of "shock wave", a moving distinct border between the cells already switched and not yet switched to the new automorphous movement, or in the form of "deployment" when the transition from one local morphology to another is continuously deployed in space.  相似文献
