Sex and strain differences in survival were studied in 7-9 month old germfree mice following transfer to a conventional colony. One outbred and three inbred strains were observed. All outbred CD-1 mice survived transfer and in 4 months increased their weight by 50%. The majority of inbred mice survived 7 months after transfer. Sex differences in survival were evident throughout the experimental period and were most marked 7 months after transfer. An unexpected new finding was the viability of the male sex in germfree mice after transfer. Possible explanations are considered.  相似文献

Members of the genus Ranavirus (family Iridoviridae) can cause catastrophic mortality of pond-breeding amphibians and are associated with an emerging infectious disease that may be contributing to amphibian declines. We conducted three experiments to examine factors that may affect transmission both within and between local breeding populations of the wood frog (Rana sylvatica). In a laboratory study, when exposed to moribund tadpoles collected during a local ranaviral die-off, uninfected tadpoles died as soon as 4 days after exposure. The onset of death was accelerated when tadpoles were allowed to scavenge on carcasses of infected tadpoles. In a mesocosm experiment that was conducted in outdoor wading pools, die-offs of tadpoles began approximately 19 days after infected tadpoles were added to pools containing uninfected tadpoles. Mass die-offs with greater than 98% mortality occurred in all pools, regardless of the initial tadpole density. In a second mesocosm experiment, the addition of water and bottom sediments that were collected from a pond during a ranaviral die-off did not result in lower tadpole survival or growth relative to controls. Only a small percentage of tadpoles appeared to be sick, and most tadpoles survived until the first individuals began metamorphosing within a pool. However, tests for ranavirus using polymerase chain reaction were positive for most pools that received contaminated sediment, suggesting that some infections were sublethal. Our results indicate that transmission within ponds is enhanced by scavenging and that spread between local ponds could occur via the transport of contaminated sediment by animals or humans.  相似文献

Cells were separated on the basis of their buoyant density by equilibrium centrifugation in continuous albumin density gradients. CFUc in the density subpopulations were detected by culture in agar containing three different types of colony stimulating factor (CSF). The sources of the CSF were post-endotoxin mouse serum (CSFES), mouse lung conditioned medium (CSFMLCM) and human urine (CSFHU). Mice were removed from the germfree environment and the buoyant density status of their CFUc was examined 1, 4 and 8 weeks later. No difference was found between germfree and conventional mice in the number of nucleated cells per femur or in their modal density. Neither was the number of CFUc per femur different. The cell cycle status of CFUc, as determined by the thymidine suicide technique was not significantly different. Functional heterogeneity was found among the density subpopulations for both groups of mice. This depended on the type of CSF. The density distribution of CFUc was significantly different in germfree mice. There were proportionately more low density CFUc. The mean modal density of CFUc under CSFES stimulation was less by 0.0045 g/cm3 in germfree mice. The removal of mice from the germfree environment resulted in a shift of the distribution to higher densities. The trend was towards the conventional situation. The significance of the buoyant density status of CFUc is discussed.  相似文献

Four groups of inbred male LEW rats were examined: A, germfree athymic; B, specified pathogen free (SPF) athymic; C, germfree euthymic; D, SPF euthymic. All animals were killed at 18 weeks and compared with respect to body weight, histological appearance and cell density of the lymphoid organs, haematological values and differential counts of bone marrow, peripheral blood and lymph. Athymic rats had a lower body weight, less densely populated lymphoid organs, and fewer lymphocytes in the blood and lymph compared with euthymic animals. No difference was seen between athymic rats under germfree and SPF conditions, and in general the differences between athymic and euthymic animals were less pronounced under germfree conditions.  相似文献

A Plexiglas isolator for rearing germfree vertebrates is described. Plastic components were precut and finished by a Plexiglas supplier. This equipment is relatively inexpensive; moreover, neither special facilities nor skills are required in their construction. Advantages of this type of isolator are: readily dismantled for cleaning, durable, excellent visibility of the interior, and minimal maintenance. In 30 separate groups of animals, 27 of these yielded germfree chicks. Many of these chicks were reared germfree for 4 weeks; and several were maintained for about 2 months, without microbial contamination. Pure cultures of Candida albicans, Streptococcus faecalis, and Clostridium perfringens were established as monocontaminations in the intestinal tracts of chickens. In contrast, Alcaligenes faecalis ATCC 9220 failed to grow in the gut of chicks, following peroral administration of this organism. All chicks described were employed on other experiments, and were killed for various assays at times specified.  相似文献

An investigation was undertaken to study the effects of germfree (GF) status and mild food restriction on life span in GF and specific pathogen-free (SPF) male ICR mice either full-fed (ad libitum) or on a restricted diet of 4.5 grams per day (equivalent to approximately 80% of full-fed intake) from five-week-old. The mean life span of the full-fed SPF and GF mice was 75.9 and 88.9 weeks respectively, while the mean life span of the food-restricted SPF and GF mice was 117.5 and 109.6 weeks, respectively. Mice in both GF and SPF food-restricted groups were characterized by lower body weight and increased survival. These findings suggest that the cessation of growth may be importantly and perhaps causally related to longevity. The GF mice survived longer than the SPF mice, but the combination of GF status with food restriction did not seem to extend life span more than food restriction alone.  相似文献

Germfree mice housed in isolators under controlled environmental and nutritional conditions were associated with an intestinal microflora. These associated animals and germfree mice drawn from the same population were tested for the rate at which the epithelial cells transited from the crypts of Lieberkuhn to the tips of the villi in their small intestines. The method for estimating the rate of transit of epithelial cells involved the use of liquid scintillation counting to determine the amount of radioactivity entering the cells while the animals were being injected with [3H]thymidine and statistical analysis of th data with a computer program developed for the purpose. As estimated by that method, the cells transited from the crypts to the villous tips in germfree mice in about 115 h and in the associated animals in about 53 h. In monoassociated mice, a strain of a Lactobacillus sp. had no effect on the transit time of the epithelial cells. A strain of Torulopsis pintolopesii stimulated uptake of 3[H]thymidine by the small bowel mucosae in mice monoassociated with the organisms for 5 weeks. In animals monoassociated with the yeast fo 3, 4, and 6 weeks, however, the radioactive compound was incorporated into the bowel mucosae to the same extent as the mucosae of germfree mice. Therefore, similarly to the Lactobacillus strain, T. pintolopesii has no obvious influence on the transit rate of small bowel epithelial cells.  相似文献

Invasive predators can devastate native species and ecosystems. However, native species may be able to coexist with invasive predators through a variety of mechanisms, such as changes in morphology or behavior due to a plastic response or selection on fixed anti-predator traits. We examined whether exposed and naive populations of Pacific tree frog tadpoles (Pseudacris regilla) display divergent morphological and behavioral traits in response to the invasive predatory red swamp crayfish (Procambarus clarkii). Tadpoles were collected from three study streams with and three without crayfish, in the Santa Monica Mountains of Southern California. We analyzed tadpole morphology and tested anti-predator behavior and survival in the laboratory. Tadpoles from streams with crayfish had shallower, narrower tails than tadpoles from streams without crayfish. Tadpoles from streams with and without crayfish were less active after exposure to crayfish chemical cues. The divergent morphology of naive and exposed tadpoles is consistent with tadpoles exhibiting a plastic response to crayfish or undergoing selection from crayfish predation. In laboratory predation experiments, we found no difference in survival between tadpoles from streams with and without crayfish but tadpoles that survived predation had deeper tail muscles than those that were killed or injured. Our results suggest that deeper tails are advantageous in the presence of crayfish, yet tadpoles from crayfish streams had shallower tails than those from crayfish-free streams. Shallower tails may have an alternative unmeasured advantage or there may be a physiological constraint to developing deeper tails in the wild. These results highlight the ability of a native frog to respond to an invasive predatory crayfish, potentially allowing for coexistence.  相似文献

In order to determine the influence of bacterial colonization on amount and composition of colonic mucins, germfree male AS/Ztm rats were colonized with a rat specific intestinal flora for different times (2, 7, 14, 21, 28, 35, 120 days). The amount of colonic mucins was determined by gel filtration on Sepharose CL-4B; the relative amount of acidic mucins was calculated after ion exchange chromatography. In addition, cecal weight and dry matter of feces were monitored. While germfree and SPF rats revealed similar amounts of colonic mucins (7.0 vs. 7.2 mg mucin/300 g body weight), the initial phase of association was characterized by considerably decreasing values. After four weeks of association, the total amount of colonic mucins had almost equalized in the two groups. The amount of acidic mucins, having decreased during the first three weeks of colonization, rendered values comparable to the SPF mucins after four months of adaptation. Cecomegaly in germfree rats disappeared within the first two days, while solidification of the intestinal content occurred within four months. Mucin losses during initial phase of association are attributed 1. to the disappearance of the cecal mucin pool, and 2. to the mucin degrading activity of some bacterial strains known to be present in the intestinal flora. Further development is conducted by a stimulation of mucin secretion, described to follow the colonization. The initially increased secretion of neutral mucins is attributed to a pronounced release of immature mucin glycoproteins, while the shift to more acidic mucins is considered to result from stimulated secretion as well as from a selective bacterial degradation of neutral mucin components.  相似文献

A new genus and species, Thrinascotrema brisbanica, is proposed to accommodate a plagiorchiidan trematode parasitic in the stomach of the freshwater turtle Elseya latisternum. The distinctive taxonomic features of the parasite are the shape and extent of the excretory bladder, and the stenostomate arrangement of the excretory collecting ducts in the adult, cercaria and metacercaria together with a cercarial protonephridial formula of 2(12+12+12)+(12+12+12). The life-cycle is three- host and aquatic. The pulmonate snail Glyptophysa gibbosa served as both a first and second intermediate host and tadpoles of Limnodynastes peronii, Adelotus brevis and Bufo marinus, and the snail Austropeplea lessoni also served as second intermediate hosts. Eggs were fully embryonated and infective when laid, but did not hatch until eaten by the snail. Cercariae first emerged 55 days after infection at 24-28 °C. They were sluggish swimmers and survived for about 48 hr. They attached firmly to the skin of snails and tadpoles on contact and began to penetrate the skin after a short exploratory migration. Metacercariae survived in snails and tadpoles for at least 3 months. It is concluded that Thrinascotrema is best placed within a new family, the Thrinascotrematidae (Digenea: Plagiorchiida), based on the unusual morphology of the excretory system.  相似文献

Growth and development rates often differ among populations of the same species, yet the factors maintaining this differentiation are not well understood. We investigated the antipredator defences and their efficiency in two moor frog Rana arvalis populations differing in growth and development rates by raising tadpoles in outdoor containers in the nonlethal presence and absence of three different predators (newt, fish, dragonfly larva), and by estimating tadpole survival in the presence of free-ranging predators in a laboratory experiment. Young tadpoles in both populations reduced activity in the presence of predators and increased hiding behaviour in the presence of newt and fish. Older tadpoles from the slow-growing Gotland population (G) had stronger hiding behaviour and lower activity in all treatments than tadpoles from the fast-growing Uppland population (U). However, both populations showed a plastic behavioural response in terms of reduced activity. The populations differed in induced morphological defences especially in response to fish. G tadpoles responded with relatively long and deep body, short tail and shallow tail muscle, whereas the responses in U tadpoles were often the opposite and closer to the responses induced by the other predators. U tadpoles metamorphosed earlier, but at a similar size to G tadpoles. There was no evidence that growth rate was affected by predator treatments, but tadpoles metamorphosed later and at larger size in the predator treatments. G tadpoles survived better in the presence of free-ranging predators than U tadpoles. These results suggest that in these two populations, low growth rate was linked with low activity and increased hiding, whereas high growth rate was linked with high activity and less hiding. The differences in behaviour may explain the difference in survival between the populations, but other mechanisms (i.e. differences in swimming speed) may also be involved. There appears to be considerable differentiation in antipredator responses between these two R. arvalis populations, as well as with respect to different predators.  相似文献

Several attempts were made to colonize the alimentary tract and infect germfree BALB/c mice and germfree Sprague-Dawley rats with two human isolates of Helicobacter pylori. The alimentary tracts of mice, sacrificed at intervals between 1 day and 20 weeks after oral challenge, were culture negative for H. pylori. The alimentary tract, kidney, liver, and mesenteric lymph nodes were culture negative for H. pylori 5 h after intravenous challenge. Growth of H. pylori was inhibited by homogenates of murine stomach, small intestine, liver, and mesenteric lymph nodes. Germfree rats and mice do not appear to be readily colonized or infected by human strains of H. pylori.  相似文献

1. The role of non-consumptive predator effects in structuring ecological communities has become an important area of study for ecologists. Numerous studies have shown that adaptive changes in prey in response to a predator can improve survival in subsequent encounters with that predator. 2. Prey-mediated changes in the shapes of predators' functional response surfaces determine the qualitative predictions of theoretical models. However, few studies have quantified the effects of adaptive prey responses on the shape of predator functional responses. 3. This study explores how prey density, size and previous predator experience interact to change the functional response curves of different-sized predators. 4. We use a response surface design to determine how previous exposure to small or large odonate predators affected the short-term survival of squirrel tree frog (Hyla squirella) tadpoles across a range of sizes and densities (i.e. the shape of odonate functional response curves). 5. Predator-induced tadpoles in a given size class did not differ in shape, although induction changed tadpole behaviour significantly. Induced tadpoles survived better in lethal encounters with either predator than did similar-sized predator-naive tadpoles. 6. Induction by either predator resulted in increased survival with both predators at a given size. However, different mechanisms led to increased survival for induced tadpoles. Attack rate for the small predators, whereas handling time increased for the large predators.  相似文献

Summary Tadpoles ofBufo calamita, a species that spawns in little temporary pools, were tested on intra- and interspecific crowding-effects. Water was conditioned by tadpoles of the following species:Bufo calamita (tadpoles of different age),Alytes obstetricans, Bombina variegata, Bufo bufo, Hyla arborea, Rana lessonae, R. ridibunda andR. temporaria. Tadpoles ofBufo calamita are most sensitive to water conditioned by older tadpoles of the own species during the first 2–3 weeks of development. They are also strongly inhibited by tadpoles ofRana temporaria andBufo bufo (that, as a rule, do not occur in the same habitat). Relative little effects are observed in water conditioned by tadpoles of the two species that occur very often in ecological sympatry withBufo calamita: Hyla arborea andBombina variegata. Young tadpoles ofBufo calamita are most tolerant of tadpoles of the same age of the own species. The crowding-effect increases with the number and weight of tadpoles/volume conditioning the water. In the age of 2–3 weeks the sensibility to intra- and interspecific crowding-effects fades. Thus inBufo calamita the age specific sensibility to crowding-effects in tadpoles may act as a density regulation mechanism in temporary pools.

Neuroventilation is highly plastic and exposure to either of two distinct teratogens, nicotine or ethanol, during development results in a similar loss of the neuroventilatory response to hypercapnia in bullfrog tadpoles. Whether this functional deficit is permanent or transient following nicotine or ethanol exposure was unknown. Here, we tested the persistence of hypercapnic neuroventilatory response impairments in tadpoles exposed to either 30 μg/L nicotine or 0.12–0.06 g/dL ethanol for 10 weeks. Brainstem breathing‐related neural activity was assessed in tadpoles allowed to develop teratogen‐free after either nicotine or ethanol exposure. Nicotine‐exposed animals responded normally to hypercapnia after a 3‐week teratogen‐free period but the hypercapnic response in ethanol‐exposed tadpoles remained impaired. Tadpoles allowed to develop for only 1 week nicotine free after chronic exposure were unable to respond to hypercapnia. The hypercapnic response of ethanol‐exposed tadpoles returned by 6 weeks following chronic ethanol exposure. These findings suggest that some nicotine‐ and ethanol‐induced impairments can be resolved during early development. Understanding both the disruptive effects of nicotine and ethanol exposure and how impaired responses return when teratogen exposure stops may offer insight on the function and plasticity of respiratory control. © 2010 Wiley Periodicals, Inc. We investigated the effects of pectin with different degrees of methylation (34.5, 70.8, and 92.6%, respectively) on the composition and concentration of intestinal and fecal bile acids and neutral sterols in conventional and germfree rats. Diets containing 6.5% pectin (galacturonan) were given for 3 weeks. High concentrations of free and secondary bile acids appeared in cecum and colon of conventional rats. With increasing degree of methylation, more bile acids were transported into lower parts of intestinal tract and excreted whereas the proportion of secondary bile acids decreased. In contrast, the composition of bile acids in intestinal contents and feces was relatively unchanged in germfree rats. Exclusively cholesterol was found as a neutral sterol in germfree rats. Coprostanol appeared in cecum of conventional rats and additionally coprostanone in colon. Amounts of neutral sterols increased with increasing degree of methylation of pectin. Additionally, concentrations of bile acids in plasma decreased if the pectin-containing diets were given. Besides the degree of methylation, the molecular weight of pectin used in the diets influenced concentration and composition of intestinal and fecal steroids in rats.  相似文献

The effect of Amphenone B, an inhibitor of corticoid synthesis, on thyroxine (T4)-induced metamorphosis was studied in toad tadpoles kept in thiourea. Amphenone injections retarded T4-induced tail resorption markedly. The effect of Amphenone was nullified by aldosterone and corticosterone added to the water in which tadpoles were kept. Steroidogenic cells of adrenals in Amphenone-injected animals were enlarged markedly as compared with those in the saline-injected tadpoles or the Amphenone-injected tadpoles which were supplemented with corticoids. The results strongly suggest that endogenous corticoids act together with thyroid hormone to accelerate metamorphosis.  相似文献

[3H8]Leukotriene C4 was administered to germfree rats and to conventional rats having a bile duct cannula. Several radioactive metabolites were isolated. Two polar biliary metabolites from conventional rats were identified as N-acetyl-omega-carboxy-leukotriene E4 and N-acetyl-omega-hydroxy-leukotriene E4. A polar fecal metabolite from germfree rats was found to be N-acetyl-omega-carboxy-leukotriene E4. Chemical identities were established using UV spectroscopy and cochromatographies with authentic compounds in several HPLC systems. The fecal metabolite was further characterized by reductive desulfurization followed by gas-liquid-radiochromatography. The yield of the two biliary metabolites was 5% of the administered tritium after three hours and the yield of fecal N-acetyl-omega-carboxy-leukotriene E4 was 3.5% after three days.  相似文献
