TFAR19促进小鼠肝线粒体膜通透性转运孔的开放   总被引:12,自引:0,他引:12  
TFAR19基因 (TF 1cellapoptosisrelatedgene 19)是北京大学人类疾病基因中心从人白血病细胞株TF 1细胞中克隆到的凋亡相关新基因之一 (GenBank登记号AF0 1495 5 )。初步研究发现 ,该基因在细胞凋亡时高表达 ,并且表达产物具有抑制肿瘤细胞生长和促进凋亡作用。但是其确切的作用机制不明。线粒体膜完整性破坏所导致促凋亡因子 (如细胞色素c等因子 )的释放是细胞凋亡关键性的控制因素。线粒体膜通透性转运孔 (PTP) ,对线粒体膜完整性具有重要的调控作用。研究了重组人TFAR19蛋白在体外条件下 ,对线粒体PTP、跨膜电位 ,以及细胞色素c释放的影响。结果表明 ,TFAR19蛋白使分离的小鼠肝线粒体PTP开放、线粒体跨膜电位下降 ,以及细胞色素c释放。TFAR19对线粒体的上述作用是通过促进PTP开放起作用的。实验结果提示 ,TFAR19对线粒体凋亡信号有正反馈放大作用 ,并进一步揭示了TFAR19促进细胞凋亡的机制  相似文献

By means of cDNA-RDA method, some cDNA fragments were found to have high levels of expression during deprivation of GM-CSF (granulocyte macrophage-colony stimulating factor) in a human myeloid cell line, TF-1 cells. One of these fragments was identified as a novel gene. To get the full length of cDNA, rapid amplification of cDNA ends (RACE) and expressed sequence tags (EST) overlapping fragments assembling strategies were used. The novel gene was named TRAF15 (TF-1 cell apoptosis related gene-15), which consists of 1 218 nucleotides and encodes 212 amino acids. The putative protein product of TFAR15 is partially homologous toC. elegans protein C14A4.11. TFAR15 mRNA is expressed in fetal liver, kidney, spleen and lung, and also in some human myeloid cell lines. Both of the TFAR15 mRNA and protein were highly expressed in TF-1 cells after GM-CSF withdrawal.In vitro analysis showed that the recombinant TFAR15 protein could inhibit the natural cell death of 293 cells, an embryonic kidney cell line.  相似文献

A novel human TF-1 cell apoptosis-related protein, TFAR19, cloned from a human leukemia cell line, TF-1, was first overexpressed in Escherichia coli with the sequence Met-Gly-His(6)-Gly-Thr-Asn-Gly, a hexahistidine sequence followed by a hydroxylamine cleavage site attached to its amino terminus. The resulting protein was soluble and single-step purified to homogeneity by metal chelating affinity chromatography. After cleavage of the purified His(6)-tagged TFAR19 sample with hydroxylamine, highly purified untagged TFAR19 protein was then obtained through an FPLC Resource Q column. The structural characteristics and function of the His(6)-tagged and untagged TFAR19 proteins were studied using circular dichroism, intrinsic fluorescence, and ANS-binding fluorescence spectra and apoptosis activity assay. The results show that alpha-helix is the main secondary structure of the proteins and the two forms of TFAR19 protein fold properly, which correspond well to their apoptosis activity expression. The results also indicate that the extra sequence including the His(6)-tag fused to the N-terminus of TFAR19 protein has a minimal effect on its structure and function, suggesting that the His(6)-tagged TFAR19 protein could be further used as an immobilized target for finding potential proteins which interact with TFAR19 from a cDNA library using in vitro ribosome display technique.  相似文献

构建TF 1细胞凋亡相关基因 19(TF 1cellapoptosisrelatedgene 19,TFAR19)缺失突变体的原核表达载体 ,获取缺失突变体蛋白 ,用于TFAR19促凋亡分子机理的研究 .从真核表达载体pcDI TFAR19扩增出野生型TFAR19和 4个缺失突变体 ,重组到原核表达载体pGEX 4T 2 .经亲和层析方法对缺失体蛋白进行纯化后 ,再利用凝胶过滤的方法进一步纯化 .利用抗GST和抗TFAR19的单克隆抗体对蛋白进行免疫学鉴定 .用白血病细胞株HL 6 0检测蛋白活性 .成功地克隆并重组了野生型TFAR19及缺失突变体 pGEX 4T 2表达载体 ,对融合蛋白的表达条件进行了优化 .SDS PAGE结果显示 ,各个缺失突变体融合蛋白均有较高水平的表达 .免疫学检测证实获得了正确的表达产物 .活性检测证实 ,野生型TFAR19和缺失突变体 4可以明显促进去血清诱导的HL 6 0细胞凋亡 ,第 6外显子可能是一个与TFAR19促凋亡活性密切相关的功能结构域  相似文献

TFAR19 is a novel apoptosis-related gene and can accelerate cell apoptosis in the presence of apoptosis inducements. Here, we studied the effects of TFAR19 on some biophysical properties of mouse erythroleukemia (MEL) cells and their molecular and structural basis. After transfected with TFAR19 and apoptosis inducement, MEL revealed a high cell membrane fluidity, a decrease in resynthesis of phospholipids, an increase in the proteins/nucleic acids ratio, a relatively orderly cytoskeleton network, an impaired deformability, a low integrin aM expression, and a decrease in adhesion to endothelial cells. These findings suggest the potential of TFAR19 for antitumor cell migration, and thus for antitumor gene therapy.  相似文献

The expression of TFAR19 was detected by Western blot. Growth curve and flow cytometry analysis showed that after being transfected with TFAR19 gene, the growth of MEL-TF19 is suppressed and its apoptosis is accelerated because of the serum deprivation. Our biorheological study indicated that in the apoptotic process, compared with MEL cells, MEL-TF19 cells exhibit larger osmotic fragility, lower cell surface charge density, increased elastic modulus K1 which is inversely proportional to cells' maximal deformation ability, obviouslydiminished surface viscosity μ, with elastic modulus K2 having no distinct changes. The above results provided some bases for recognizing the function of TFAR19 completely from the viewpoint of biorheology.  相似文献

人程序性细胞死亡分子10(Homo sapiens programmed cell death 10, PDCD10),最初被称为TFAR15(TF-1 cell apoptosis related gene 15),是由撤除粒细胞 巨噬细胞集落刺激因子诱导凋亡的人红白血病细胞系TF-1中克隆得到的1个凋亡相关基因. 后来发现它的突变可引起散发性或家族性颅内海绵状血管瘤(cerebral cavernous malformations,CCMs)的发生,为CCMs的第3个致病基因,所以又被叫做CCM3.近年来研究发现,PDCD10能够和GCKⅢ蛋白、γ-PCDH、CCM2、VEGFR2、ERM等众多蛋白相互作用,并能调控ERK/MAPK通路,增加MST4/VEGFR2的稳定性,增强相应的信号转导,促进细胞的增殖、分化和中枢神经系统的发育,与癌症的发生相关,还能调节细胞的凋亡.以上研究证明了PDCD10 的多种生物学效应,并提示其在血管生成、氧化应激、肿瘤中发挥重要作用.  相似文献

We recently identified TL1A, an endothelium-derived T cell costimulator and a ligand for tumor necrosis factor receptor superfamily members DR3 and decoy receptor 3. To elucidate the signaling events triggered by TL1A-DR3 interaction and to understand the molecular mechanisms regulating DR3-mediated apoptosis, we have studied the effect of TL1A and an agonistic DR3 monoclonal antibody in human erythroleukemic TF-1 cells, which express DR3 endogenously. TL1A induced the formation of a DR3 signaling complex containing TRADD, TRAF2, and RIP and activated the NF-kappaB and the ERK, JNK, and p38 mitogen-activated protein kinase pathways. However, TL1A or an agonistic DR3 monoclonal antibody did not induce apoptosis in these cells nor were there detectable levels of FADD or procaspase-8 seen in the signaling complex. Interestingly, DR3-mediated apoptosis was induced in TF-1 cells in the presence of a NF-kappaB pathway-specific inhibitor but not in the presence of mitogen-activated protein kinase inhibitors, either alone or in combination, suggesting that DR3-induced NF-kappaB activation was responsible for resistance to apoptosis in these cells. Consistent with this, we found that TL1A significantly increased the production of c-IAP2, a known NF-kappaB-dependent anti-apoptotic protein, and that the NF-kappaB inhibitor or cycloheximide prevented its synthesis. Furthermore, inhibition of c-IAP2 production by RNA interference significantly sensitized TF-1 cells to TL1A-induced apoptosis. Our study identifies a molecular mechanism by which TL1A and DR3 regulate cell fate in TF-1 cells.  相似文献

It was recently reported that arsenic trioxide (As_2O_3) can induce complete remission in patients with acute promyelocytic leukemia (APL). In this present article, the biological effect of As_2O_3 on human cervical cancer HeLa cells and HeLa cells overexpressing Bcl-2 is studied. By MTT and colony forming ability assays, morphology alteration, flow cytometric analysis, DNA gel electrephoresis and in situ cell death detection (TUNEL), it was found that As_2O_3 inhibited the growth of HeLa cells and induced G2/M arrest and apoptosis of the cells. RT-PCR, Northern blot, Western blot analysis revealed that As_2O_3 induced HeLa cell apoptosis possibly via decreasing the expression of c-myc and viral genes. HeLa cells overexpressing Bcl-2 partly resist As_2O_3 induced apoptosis, which might be relative to preventing the cells from As_2O_3 caused G2/M block, downregulation of c-myc gene expression and inhibition of viral gene expression was also noted, However, it was found that As_2O_3 at a high concentratio  相似文献

A simple rapid and reproducible procedure for transferring monoclonal antibodies into mammalian cells by electroporation is described. Two functionally different monoclonal antibodies (Mab 3F3 and Mab 2B4) specific for asparagine synthetase (EC 6.3.1.1) were used for electroporation into HeLa, HT-5, and L5178Y D10/R (L-asparaginase-resistant) cells. The conditions were optimized so that the viability of the electroporated cells was very high (80-90%), and 90% of the viable cells had antibody incorporated. Electropermeabilized cells were structurally intact, and the high voltage electric pulse had no inhibitory effect on overall cellular DNA and protein synthesis. Incorporated immunoglobulins showed unaltered structural integrity and were functionally active. L5178Y D10/R cells incorporated with an antibody (Mab 3F3) known to be a potent inhibitor of tumor asparagine synthetase showed increased dependence on an exogenous source of asparagine in the culture medium, while the growth of cells incorporated with a control (noninhibitory) antibody (Mab 2B4) remained unaffected. These studies demonstrate that electroporation can be employed successfully for large scale transfer of antibodies into cultured mammalian cells for the study of cellular metabolism.  相似文献

以高表达Polo样激酶1(Polo-like kinase 1,PLK1)的宫颈癌细胞系HeLa细胞为模型,观察针对PLK1基因的短发夹状RNA(short hairpin RNA,shRNA)对其凋亡和增殖的影响.设计并合成了针对PLK1的shRNA,将其导入构建的携带增强型绿色荧光蛋白(EGFP)的RNAi(RNA interference)表达载体中.通过 RT-PCR和Western 印迹分别检测HeLa细胞PLK1基因和蛋白水平的表达,以流式细胞仪和PI-Hochest双染法测细胞凋亡,MTT法检测细胞的增殖水平.成功构建了携带EGFP的RNAi表达载体pEGFP-H1.转染shRNA后,HeLa细胞PLK1的表达降低至30%.与对照组和空载体转染组相比,shRNA转染组的HeLa细胞凋亡率明显增加,其增殖活性则明显降低.本课题构建的RNAi表达载体便于观察靶基因的转染情况,且不影响H1启动子的体内转录.PLK1的基因沉默能明显增加HeLa细胞的凋亡,抑制该细胞的增殖,有可能为未来肿瘤的治疗找到新的靶点和有效途径.  相似文献

目的研究小鼠UNCV蛋白质对HeLa细胞凋亡的影响。方法将BALB/c小鼠Uncv基因重组质粒转染HeLa细胞,筛选出稳定过表达UNCV蛋白质的HeLa细胞株。通过细胞计数法和流式细胞术,检测过表达UNCV蛋白质对血清饥饿和阿霉素诱导的HeLa细胞凋亡的影响。结果获得稳定正确过表达UNCV蛋白质的HeLa细胞株。细胞计数和流式细胞术结果显示过表达UNCV蛋白质对血清饥饿诱导的HeLa细胞凋亡有抑制作用,对于阿霉素诱导的HeLa细胞凋亡没有显著影响。结论过表达UNCV蛋白质对血清饥饿诱导的HeLa细胞凋亡有抑制作用。  相似文献

When serum-starved HeLa S3 cells were stimulated to proliferate by addition of fetal calf serum (FCS), (2'-5')oligoadenylate synthetase (2-5A synthetase) activity was induced. Although no interferon (IFN) activity was detectable in the HeLa S3 cell-conditioned culture medium after growth stimulation, addition of anti-IFN-beta monoclonal antibody inhibited both the expression of the 2-5A synthetase gene and the production of the enzyme, suggesting that endogenous IFN-beta was involved in 2-5A synthetase induction. Purified preparations of three growth factors, epidermal growth factor, platelet-derived growth factor, and insulin, also induced 2-5A synthetase through IFN-beta. When serum-starved HeLa S3 cells were treated with FCS, DNA synthesis was initiated synchronously, with peaks after 12 and 32 h, although the level of 2-5A synthetase reached a maximum after the first peak of DNA synthesis. Inhibition of 2-5A synthetase induction by anti-IFN-beta antibody enhanced the second, but not the first cycle of DNA synthesis. These results suggested that in HeLa S3 cells, after stimulation with growth factors the IFN/2-5A synthetase system played a role in cell growth negative regulatory mechanisms.  相似文献
