Annexin 1 is secreted by the human prostate   总被引:3,自引:0,他引:3  
The human prostate expresses very high concentrations of annexins 1, 4, and 5 and secretes high concentrations of annexins 1 and 5. Although the biological roles of these proteins in prostate secretions are not known, these studies emphasize the need to consider extracellular sites for physiological functions of annexins. The clear demonstration of secretion of proteins that have blocked N-termini and lack hydrophobic signal sequences raises the possibility that novel cellular secretory pathways exist. Preliminary immunohistochemical experiments in collaboration with Dr. James Fallon indicate that both annexins 1 and 4 are expressed in prostate ductal secretory epithelium. Since annexin 1, but not annexin 4, is secreted, a comparison of the cellular fate of these two related proteins in the prostate may provide a useful model system for determining the structural elements that direct the secretion of proteins which lack conventional signal sequences.  相似文献

Tissue kallikrein activity was monitored in seminal plasma from 3 groups of bovine ejaculates: those with normal total sperm motility (78.43%), with reduced sperm motility (49.29%), and with reduced sperm count (0.68 x 10(9) cells/ml). The tissue kallikrein activity was measured spectrophotometrically by using the specific chromogenic substrate S-2266. It was found that the semen samples with normal sperm motility manifested 1.083 microkat/L, on an average, or 29% higher than the activity recorded in ejaculates with reduced sperm motility (P < 0.05). After storage of a group of ejaculates of normal quality for 5 h at room temperature, sperm motility dropped by approximately 80%, expressed as a percentage of the initial motility, while the tissue kallikrein activity in the respective seminal samples decreased by 23%. No significant differences were found in kallikrein activity between ejaculates with normal and reduced sperm counts. It is concluded that a relationship exists between the level of tissue kallikrein activity in the seminal plasma of bovine ejaculates and sperm motility.  相似文献

Prostate-specific kallikrein, a member of the gene family of serine proteases, was initially discovered in semen and is the most useful serum marker for prostate cancer diagnosis and prognosis. We report the crystal structure at 1.42A resolution of horse prostate kallikrein (HPK). This is the first structure of a serine protease purified from seminal plasma. HPK shares extensive sequence homology with human prostate-specific antigen (PSA), including a predicted chymotrypsin-like specificity, as suggested by the presence of a serine residue at position S1 of the specificity pocket. In contrast to other kallikreins, HPK shows a structurally distinct specificity pocket. Its entrance is blocked by the kallikrein loop, suggesting a possible protective or substrate-selective role for this loop. The HPK structure seems to be in an inactivated state and further processing might be required to allow the binding of substrate molecules. Crystal soaking experiments revealed a binding site for Zn(2+) and Hg(2+), two known PSA inhibitors.  相似文献

A tissue kallikrein from human seminal plasma was isolated by immunoaffinity chromatography and characterized. Its molecular mass was determined by gel filtration to be approximately 40000 Da. The enzyme preparation liberates kinin from human HMW kininogen (specific activity: 0.594 HMW kininogen-U/mg), lowers the blood pressure of dogs after intravenous injection (specific activity: 1740 biol. kallikrein unit/mg) and is strongly inhibited by aprotinin but not by soybean trypsin inhibitor. N alpha-Acetyl-L-phenylalanyl-L-arginine ethyl ester, D-valyl-L-leucyl-L-agrine ethyl ester and N-benzyloxycarbonyl-L-tyrosine p-nitrophenyl ester are cleaved with identical rates by the enzyme from human seminal plasma and human urinary kallikrein.  相似文献

Tissue cultures of hepatocytes were studied for the formation of lipid pigments as related to aging of the cultures and of the addition to them of: 1) colloidal iron; 2) L-ascorbic acid; and 3) tyrosine. The presence of lipid pigments was gauged by their sudanophilia after a light solvent extraction and yellow autofluorescence. It has been found that all the three agents enhanced pigmentogenesis. It has been concluded that both pro-oxidants and antioxidants may enhance lipid pigment formation: the first by enhancing lipid oxidation, the second by enhancing copolymerization which terminates the oxidative chain.  相似文献

The human kallikrein 5 protein (hK5) is expressed in many normal tissues, most notably in skin, breast, salivary gland and esophagus. It has also been shown to be a potential biomarker for breast, ovarian and testicular cancer. Human kallikrein 3 (hK3; prostate-specific antigen) is the most useful marker for adenocarcinoma of the prostate gland. The aim of this study was to determine whether hK3 and hK5 are expressed in salivary gland tissues and salivary gland tumors (both benign and malignant), in order to compare normal with tumor tissues. Pleomorphic adenomas, adenoid cystic carcinomas, polymorphous low-grade adenocarcinomas, acinic cell carcinomas, mucoepidermoid carcinomas and adenocarcinomas not otherwise specified of both minor and major salivary glands were examined. The results of this study indicate that most salivary gland tumors do not show high levels of expression of hK5. Staining was most prominent in keratinizing epithelia in pleomorphic adenomas. hK3 is not expressed in salivary gland tumors.  相似文献

It has been shown for the first time that the salivary gland secretion of the medicinal leech (Hirudo medicinalis) contains a human blood plasma kallikrein inhibitor which is capable of blocking the amidolytic activity of the enzyme in an irreversible manner (with D-Pro-Phe-Arg-pNA as substrate) and which also suppresses the kininogenase activity of kallikrein. The inhibition of the amidolytic activity of highly purified kallikrein preparations from human blood plasma obeys the pseudo-first order kinetics. The experimental results suggest that in the salivary-gland secretion of H. medicinalis the inhibitor concentration exceeds by one order of magnitude that in whole leech homogenate extracts, which indicates that the inhibitor biosynthesis may be localized in leech salivary glands.  相似文献

Hyperglycemia is associated with greater hematoma expansion and poor clinical outcomes after intracerebral hemorrhage. We show that cerebral hematoma expansion triggered by intracerebral infusion of autologous blood is greater in diabetic rats and mice compared to nondiabetic controls and that this augmented expansion is ameliorated by plasma kallikrein (PK) inhibition or deficiency. Intracerebral injection of purified PK augmented hematoma expansion in both diabetic and acutely hyperglycemic rats, whereas injection of bradykinin, plasmin or tissue plasminogen activator did not elicit such a response. This response, which occurs rapidly, was prevented by co-injection of the glycoprotein VI agonist convulxin and was mimicked by glycoprotein VI inhibition or deficiency, implicating an effect of PK on inhibiting platelet aggregation. We show that PK inhibits collagen-induced platelet aggregation by binding collagen, a response enhanced by elevated glucose concentrations. The effect of hyperglycemia on hematoma expansion and PK-mediated inhibition of platelet aggregation could be mimicked by infusing mannitol. These findings suggest that hyperglycemia augments cerebral hematoma expansion by PK-mediated osmotic-sensitive inhibition of hemostasis.  相似文献

Sperm motility is known to be activated and maintained by kallikrein contained within the seminal plasma. We studied the relationship between the levels of seminal plasma kallikrein activity and in vitro exogenous kallikrein-induced sperm motility enhancement. Semen samples were collected from Holstein-Friesian bulls and grouped on the basis of the initial total sperm motility into Group I with > 60 % (mean 75.3 +/- 1.8 %, n = 25), and Group II with < 60 % (mean 51.2 +/- 1.7%, n = 25). Seminal plasma kallikrein activity was measured with the aid of the specific chromogenic substrate S-2266. In Group I the mean activity was 0.983 +/- 0.042 microkat/L, and in Group II it was 0.805 +/- 0.063 microkat/L (P < 0.05). Then each semen sample was divided into a control and an experimental subgroup treated with 16.7 microkat/L of hog pancreatic kallikrein. Total sperm motility was monitored at 1-h intervals. It was found that the addition of exogenous kallikrein stimulated the sperm motility in both groups but in the 4th h after treatment the difference in sperm motility between the experimental and control subgroups of Group I was 12.4 % whereas in Group II it was 21.7 %. We concluded that adding exogenous kallikrein in vitro to semen samples with lower kallikrein activity in the seminal plasma enhanced total sperm motility more than adding it to ejaculates with higher levels of endogenous kallikrein activity.  相似文献

Angiostatin, a potent endogenous inhibitor of angiogenesis, is generated by cancer-mediated proteolysis of plasminogen. The culture medium of human prostate carcinoma cells, when incubated with plasminogen at a variety of pH values, generated angiostatic peptides and miniplasminogen. The enzyme(s) responsible for this reaction was purified and identified as procathepsin D. The purified procathepsin D, as well as cathepsin D, generated two angiostatic peptides having the same NH(2)-terminal amino acid sequences and comprising kringles 1-4 of plasminogen in the pH range of 3.0-6.8, most strongly at pH 4.0 in vitro. This reaction required the concomitant conversion of procathepsin D to catalytically active pseudocathepsin D. The conversion of pseudocathepsin D to the mature cathepsin D was not observed by the prolonged incubation. The affinity-purified angiostatic peptides inhibited angiogenesis both in vitro and in vivo. Importantly, procathepsin D secreted by human breast carcinoma cells showed a significantly lower angiostatin-generating activity than that by human prostate carcinoma cells. Since deglycosylated procathepsin D from both prostate and breast carcinoma cells exhibited a similar low angiostatin-generating activity, this discrepancy appeared to be attributed to the difference in carbohydrate structures of procathepsin D molecules between the two cell types. The seminal vesicle fluid from patients with prostate carcinoma contained the mature cathepsin D and procathepsin D, but not pseudocathepsin D, suggesting that pseudocathepsin D is not a normal intermediate of procathepsin D processing in vivo. The present study provides evidence for the first time that cathepsin D secreted by human prostate carcinoma cells is responsible for angiostatin generation, thereby causing the prevention of tumor growth and angiogenesis-dependent growth of metastases.  相似文献

The accessory reproductive glands of male mammals contribute the bulk of the secretions in which spermatozoa are transported to the female tract during copulation. Despite their morphological diversity,and the chemical complexity of their products,little is known about the possible effects of sexual selection upon these glands in mammals. Here we consider the seminal vesicles and prostate glands in a sample of 89 species and 60 genera representing 8 Orders of mammals. The sizes of the accessory glands are analysed in relation to body weight and testes weight. Both the seminal vesicles size and prostate size (corrected for body weight) correlate positively with relative testes size in this sample; this finding remains highly significant after application of procedures to correct for possible phylogenetic biases in the data set. The accessory reproductive glands are also significantly larger in those mammals which have large relative testes sizes,and in which the likelihood of sperm competition is greatest. These results support the hypothesis that sexual selection has played an important role in the evolution of the mammalian prostate gland and seminal vesicles.  相似文献

Interferon tau (IFNtau) is the pregnancy recognition signal produced by the conceptus trophectoderm and acts in a paracine manner on the ovine endometrium to increase expression of IFN-stimulated genes primarily in the stroma and deep glandular epithelium, including IFN regulatory factor-1 (IRF-1). The roles of Stat1, Stat2, and IRF-9 in IFNtau regulation of IRF-1 expression were determined using human stromal fibroblasts lacking specific IFN signaling components or complemented with specific Stat1 mutants. In parental (2fTGH) cells treated with IFNtau, Stat1alpha/beta was tyrosine phosphorylated by 15 min, and IRF-1 mRNA and protein increased from 0 to 6 h, was maximal at 6 h, and decreased to 24 h. In contrast, IFNtau did not affect IRF-1 expression in Stat1- and Stat2-deficient cells or in Stat1-deficient cells complemented with Stat1 Y701Q or Stat1 R602L mutants. In Stat1-deficient cells complemented with the Stat1 S727A mutant, Stat1alpha, or Stat1beta and treated with IFNtau, IRF-1 increased from 0 to 6 h, was maximal at 6 h, and decreased thereafter. In IRF-9-deficient cells stimulated with IFNtau, IRF-1 increased from 0 to 6 h but did not exhibit the sharp decline from 6 to 12 h observed in other cells. Collectively, results indicate that IFNtau effect on IRF-1 expression is primarily regulated by tyrosine-phosphorylated Stat1alpha or Stat1beta dimers, whereas the decline of IRF-1 after 6 h of IFNtau treatment is regulated by IRF-9.  相似文献
