The results of a landscape‐scale test of ALEX, a widely used metapopulation model for Population Viability Analysis (PVA), are described. ALEX was used to predict patch occupancy by the laughing kookkaburra and the sacred kingfisher in patches of eucalypt forest in south‐eastern Australia. These predictions were compared to field surveys to determine the accuracy of the model. Predictions also were compared to a “naïve” null model assuming no fragmentation effects.
The naïve null model significantly over‐predicted the number of eucalypt patches occupied by the sacred kingfisher, but the observed patch occupancy was not significantly different from that predicted using ALEX. ALEX produced a better fit to the field data than the naïve null model for the number of patches occupied by the laughing kookaburra. Nevertheless, ALEX still significantly over‐predicted the number of occupied patches, particularly remnants dominated by certain forest types – ribbon gum and narrow‐leaved peppermint. The predictions remained significantly different from observations, even when the habitat quality of these patches was reduced to zero. Changing the rate of dispersal improved overall predicted patch occupancy, but occupancy rates for the different forest types remained significantly different from the field observations. The lack of congruence between field data and model predictions could have arisen because the laughing kookaburra may move between an array of patches to access spatially separated food and nesting resources in response to fragmentation. Alternatively, inter‐specific competition may be heightened in a fragmented habitat. These types of responses to fragmentation are not incorporated as part of traditionally applied metapopulation models. Assessments of predictions from PVA models are rare but important because they can reveal the types of species for which forecasts are accurate and those for which they are not. This can assist the collection of additional empirical data to identify important factors affecting population dynamics.  相似文献

Individual movement patterns and the effects of host plant patch size and isolation on patch occupancy were examined for red milkweed beetles, Tetraopes tetraophthalmus, residing in a heterogeneous landscape. Male beetles were found to move both more often and farther between host plant patches than female beetles, and this difference affected the patterns of patch occupancy observed. Overall, unoccupied milkweed patches were smaller and more isolated than patches occupied by beetles. Patches uninhabited by females tended to be more isolated, but not necessarily smaller, than patches with female beetles, indicating that females may be affected more by patch isolation than patch size. Presence of male beetles on patches showed a stronger response to patch size than to patch isolation. Differences in movement between males and females illustrate the need for demographically based dispersal data. Comparisons of Tetraopes interpatch movement patterns between landscapes composed of patches of different size revealed that landscapes with overall smaller patches may have greater rates of interpatch movement.  相似文献

Past studies with spatially structured herbivore populations have emphasized the primacy of intrinsic factors (e.g., patch quality), patch geometry (e.g., patch size and isolation), and more recently landscape context (e.g., matrix composition) in affecting local population abundance and dispersal rate. However, few studies have examined the relative importance of each factor, or how they might interact to affect herbivore abundance or dispersal. Here, we performed a factorial field experiment to examine the independent and interactive effects of patch quality (plant biomass, leaf protein, leaf phenolics) and matrix composition [mudflat or non-host grass (Bromus inermis)] on planthopper (Prokelisia crocea) emigration from host-plant patches (prairie cordgrass, Spartina pectinata). In addition, a field survey was conducted to examine the relative importance of patch quality, geography, and matrix composition on planthopper occupancy and density. In the experiment, we found that rates of emigration from low and intermediate quality patches were, on average, 21% percent higher for patches embedded in brome than mudflat. In contrast, the emigration rate was unaffected by matrix composition in nutrient-rich patches. Within matrix types, plant quality had little effect on emigration. In the survey, planthopper density and the patch occupancy rate of planthoppers increased nonadditively with increasing patch size and the percentage of the surrounding matrix composed of mudflat. This study suggests that landscape-level factors, such as the matrix, may be more important than factors intrinsic to the patches.  相似文献

Charnovs marginal value theorem predicts that female parasitoids should exploit patches of their hosts until their instantaneous rate of fitness gain reaches a marginal value. The consequences of this are that: (1) better patches should be exploited for a longer time; (2) as travel time between patches increases, so does the patch residence time; and (3) all exploited patches should be reduced to the same level of profitability. Patch residence time was measured in an egg parasitoid Anaphes victus (Hymenoptera: Mymaridae) when patch quality and travel time, approximated here as an increased delay between emergence and patch exploitation, varied. As predicted, females stayed longer when patch quality and travel time increased. However, the marginal value of fitness gain when females left the patch increased with patch quality and decreased with travel time. A. victus females appear to base their patch quality estimate on the first patch encountered rather than on a fixed innate estimate, as was shown for another egg parasitoid Trichogramma brassicae. Such a strategy could be optimal when inter-generational variability in patch quality is high and within-generational variability is low.  相似文献

The shoot configuration of each monoclonal patch of phalanx-forming tallgrass, Miscanthus sinensis, is characterized by the formation of a fairy ring, which forms as the result of developing vacant inner areas. One large-sized M.sinensis patch (patch L), observed over a 9-year survey period, underwent lateral expansion in almost all directions as a result of peripheral shoot births. In the year after the shoots in each part of patch L reached a maximum density (Dmax), the number of shoots decreased by approximately 20% per year. However, the overall number of within-patch shoots was stable during the survey because the patch area increased at the periphery. Twelve patches (>900cm2 in area) with orthotropic shoots were selected to observe the distribution pattern of within-patch shoots, and the patch areas were divided into three parts: the exterior, intermediate and interior areas. In 10 of these 12 patches, shoot densities were lowest in the interior areas and highest in the exterior areas, which led to ring formation. The shoot density of each subarea was inversely related to the age of the subarea. This raises the possibility that in any part of these patches, shoot densities decrease annually from Dmax in a similar way.  相似文献

I attempted to characterize spatial units of local dynamics and dispersal in banner-tailed kangaroo rats (Dipodomys spectabilis), to determine if spatial structure influenced population dynamics in the way predicted by current metapopulation models. D. spectabilis exhibited a hierarchical spatial structure. Local populations that appeared as discrete entities on a scale of kilometers were subdivided into clusters of mounds on a scale of meters. This structure, however, cannot be characyerized in terms of the discrete habitat patches envisioned by the metapopulation models. Occupied areas were statistically distinguishable from the surrounding matrix, but this difference was only quantitative. There were no discrete boundaries between occupied areas and the matrix. Habitat within occupied areas was heterogeneous, and occupied areas in different locations were statistically distinguishable from each other. High heterogeneity within occupied areas, and high contrast among them, make it difficult to define what is a suitable habitat patch for D. spectabilis. On a smaller spatial scale, there was significant aggregation of resident mounds within occupied areas. These aggregations, however, do not correspond to discrete habitat patches. Rather, they appear to result from an interaction between fine-scale habitat heterogeneity and limited dispersal due to natal philopatry and low adult vagility. These complications make it difficult to identify habitat patches independent of the species' distribution. For species like D. spectabilis that are patchily distributed but do not occupy discrete habitat patches, a patch occupancy approach does not seem appropriate for describing spatial structure. Hierarchical spatial structure underscores the need for a framework that incorporates multiple scales of spatial structure, rather than one that pre-imposes a single spatial scale as being important for population dynamics. A framework that (i) considers patchiness as a combination of both habitat heterogeneity, and life-history and behavioral characteristics, and (ii) incorporates hierarchical spatial structure, appears to be the most suitable for conceptualizing spatial dynamics of behaviorally complex vertebrates such as D. spectabilis.  相似文献

Cytochrome a 1 c 1 was highly purified from Nitrobacter agilis. The cytochrome contained heme a and heme c of equimolar amount, and its reduced form showed absorption peaks at 587, 550, 521, 434 and 416 nm. Molecular weight per heme a of the cytochrome was estimated to be approx. 100,000–130,000 from the amino acid composition. A similar value was obtained by determining the protein content per heme a. The cytochrome molecule was composed of three subunits with molecular weights of 55,000, 29,000 and 19,000, respectively. The 29 kd subunit had heme c.Hemes a and c of cytochrome a 1 c 1 were reduced on addition of nitrite, and the reduced cytochrome was hardly autoxidizable. Exogenously added horse heart cytochrome c was reduced by nitrite in the presence of cytochrome a 1 c 1; K m values of cytochrome a 1 c 1 for nitrite and N. agilis cytochrome c were 0.5 mM and and 6 M, respectively. V max was 1.7 mol ferricytochrome c reduced/min·mol of cytochrome a 1 c 1 The pH optimum of the reaction was about 8. The nitrite-cytochrome c reduction catalyzed by cytochrome a 1 c 1 was 61% and 88% inhibited by 44M azide and cyanide, respectively. In the presence of 4.4 mM nitrate, the reaction was 89% inhibited. The nitrite-cytochrome c reduction catalysed by cytochrome a 1 c 1 was 2.5-fold stimulated by 4.5 mM manganous chloride. An activating factor which was present in the crude enzyme preparation stimulated the reaction by 2.8-fold, and presence of both the factor and manganous ion activated the reaction by 7-fold.Cytochrome a 1 c 1 showed also cytochrome c-nitrate reductase activity. The pH optimum of the reaction was about 6. The nitrate reductase activity was also stimulated by manganous ions and the activating factor.  相似文献

Fitch and Markowitz' theory of concomitantly variable codons (covarions) in evolution predicted the existence of functional correlation in amino acid residue mutations among present-day cytochromes c. Mutational analysis was carried out on yeast iso-2-cytochrome c, where hydrophobic core residues I20, M64, L85, and M98 and surface residue L9 were mutated, in selected combinations, to those found in mammalian and bird cytochromes c. The functionality assay is based upon the ability of yeast cells to grow in YPGE medium. Furthermore, experiments on the single M64L and M98L mutations as well as the double M64L/M98L mutation using NMR showed that the effects of these mutations are to perturb the structural integrity of the protein. We identified functional correlation in two cases of a pair of residue mutations, the I20 V and M98 L pair and the L9 I and L85 I pair. In both cases, only one of the two alternative, putative evolutionary pathways leads to a functional protein and the corresponding pairs of residue mutations are among those found in present-day cytochromes c. Since valine is predicted to be at position 20 in the ancestral form of cytochrome c, the present data provide an explanation for the ancient requirement of leucine rather than methionine in position 98. The present data provide further evidence for the role of those specific atom–atom interactions in directing a pathway in the evolutionary changes of the amino acid sequence that have taken place in cytochrome c, in accordance with Fitch and Markowitz.  相似文献

Trichogramma pretiosum Riley (Hymenoptera: Trichogrammatidae), a generalist parasitoid of lepidopteran eggs, intensifies search in patches contaminated by a kairomone associated with Heliothis zea (Boddie) (Lepidoptera: Noctuidae) oviposition sites. In the laboratory the parasitoid will search any kairomone-treated patch, abandoning standardized (10 cm diam) patches if no hosts are located within 60–180 sec. Total time and searching time on patches in which hosts are discovered are more highly correlated with the number of hosts attacked (Nha) per patch than with actual host density. The parasitoid's perception of host density per patch is often incorrect in this sense. Handling time (Th) and time wasted in re-handling hosts parasitized earlier in the patch visit consume a large proportion of total patch time. Several hypotheses which have been suggested to explain patch time allocation by other parasitoids and predators appear inapplicable to T. pretiosum. The ratio of re-encounters with parasitized hosts to the number of hosts attacked appears to affect the allocation of patch time by this species. The results of previous field experiments are compared to laboratory observations of searching behavior.
Résumé Trichogramma pretiosum Riley, parasitoïde généraliste des oeufs de lépidoptères, intensifie sa prospection sur des emplacements, contaminés par une kairomone associée aux pontes d'Heliothis zea Boddie (Lep. Noctuidae). Au laboratoire T. pretiosum cherchera les emplacements contaminés, abandonnant les morceaux de papier filtre standardisés (de 10 cm de diamètre) si aucun hôte n'est découvert dans les 60 à 180 secondes. Le temps total et le temps de prospection passés sur les emplacements sur lesquels des hôtes ont été découverts présentent une meilleure corrélation avec le nombre d'hôte attaqués (Nha) par morceau de papier qu'avec la densité réelle des hôtes. La perception de la densité d'hôte par morceau de papier est souvent incorrecte dans ce sens. Le temps de palpation (Th) et le temps dépensé à palper à nouveau des hôtes déjà parasités correspondent à une dépense importante du temps passé sur le morceau de papier. Les différentes hypothèses qui ont été utilisées pour expliquer la distribution du temps de prospection par d'autres parasitoïdes et prédateurs semblent inapplicables à T. pretiosum. Le taux de retour sur des hôtes parasités par rapport au nombre d'hôtes attaqués semble affecter les dépenses de temps de cette espèce. Les résultats d'observations antérieures dans les champs sont comparés aux observations de laboratoire du comportement de prospection.

The basis for the outcome of competition between sulfidogens and methanogens for H2 was examined by comparing the kinetic parameters of representatives of each group separately and in co-culture. Michaelis-Menten parameters (V max and K m) for four methanogens and five sulfate-reducing bacteria were determined from H2-depletion data. Further, Monod growth parameters (max, K s, Y H2) for Desulfovibrio sp. G11 and Methanospirillum hungatei JF-1 were similarly estimated. H2 K m values for the methanogenic bacteria ranged from 2.5 M (Methanospirillum PM1) to 13 M for Methanosarcina barkeri MS; Methanospirillum hungatei JF-1 and Methanobacterium PM2 had intermediate H2 K m estimates of 5 M. Average H2 K m estimates for the five sulfidogens was 1.2 M. No consistent difference among the V max estimates for the above sulfidogens (mean=100 nmol H2 min-1 mg-1 protein) and methanogens (mean=110 nmol H2 min-1 mg-1 protein) was found. A two-term Michaelis-Menten equation accurately predicted the apparent H2 K m values and the fate of H2 by resting co-cultures of sulfate-reducers and methanogens. Half-saturation coefficients (K s) for H2-limited growth of Desulfovibrio sp. G11 (2–4 M) and Methanospirillum JF-1 (6–7 M) were comparable to H2 K m estimates obtained for these organisms. Maximum specific growth rates for Desulfovibrio sp. G11 (0.05 h-1) were similar to those of Methanospirillum JF-1 (0.05–0.06 h-1); whereas G11 had an average yield coefficient 4 x that of JF-1. Calculated max and V max/K m values for the methanogens and sulfidogens studied predict that the latter bacterial group will process more H2 whether these organisms are in a growing or resting state, when the H2 concentration is in the first-order region.  相似文献

In order to assess the ability of Porites corals to accurately record environmental variations, high-resolution (weekly/biweekly) coral 18O records were obtained from four coral colonies from the northern Gulf of Aqaba, which grew at depths of 7, 19, 29, and 42 m along one transect. Adjacent to each colony, hourly temperatures, biweekly salinities, and monthly 18O of seawater were continuously recorded over a period of 14 months (April 1999 to June 2000). Contrary to water temperature, which shows a regular and strong seasonal variation and change with depth, seawater 18O exhibits a weak seasonality and little change with depth. Positive correlations between seawater 18O and salinity were observed. The two parameters were related to each other by the equation 18O Seawater (, VSMOW) = 0.281 × Salinity – 9.14. The high-resolution coral 18O records from this study show a regular pattern of seasonality and are able to capture fine details of the weekly average temperature records. They resolve more than 95% of the weekly average temperature range. On the other hand, attenuation and amplification of coral seasonal amplitudes were recorded in deep, slow-growing corals, which were not related to environmental effects (temperature and/or seawater 18O) or sampling resolution. We propose that these result from a combined effect of subannual variations in extension rate and variable rates of spine thickening of skeletal structures within the tissue layer. However, no smoothing or distortion of the isotopic signals was observed due to calcification within the tissue layer in shallow-water, fast-growing corals. The calculations from coral 18O calibrations against the in situ measurements show that temperature (T) is related to coral 18O ( c ) and seawater 18O ( w ) by the equation T (°C) = –5.38 ( c w ) –1.08. Our results demonstrate that coral 18O from the northern Gulf of Aqaba is a reliable recorder of temperature variations, and that there is a minor contribution of seawater 18O to this proxy, which could be ignored.  相似文献

While there is agreement that both habitat quality and habitat network characteristics (such as patch size and isolation) contribute to the occupancy of patches by any given species, the relative importance of these factors is under debate. This issue is of fundamental ecological importance, and moreover of special concern for conservation biologists aiming at preserving endangered species. Against this background we investigated patch occupancy in the violet copper Lycaena helle, one of the rarest butterfly species in Central Europe, in the Westerwald area (Rhineland-Palatinate, Western Germany). Occupied (n = 102) differed from vacant (n = 128) patches in altitude, size, connectivity, availability of wind shelter, in the abundance of the larval host-plant, in the abundance of a grass species indicating favorable habitat conditions and in the abundance of nitrophilous plants. Overall, patch occupancy was primarily determined by patch size, connectivity and the abundance of the larval host plant, while all other parameters of habitat quality were of subordinate importance. Therefore, our findings suggest that even for extremely sedentary species such as L. helle habitat networks are decisive and—next to the preservation of habitat quality—need to be an integral part of any conservation management for this species.  相似文献

Little information exists on the pattern of nitrogen (N) uptake, remobilization and N use efficiency (NUE) in Leafy and stay-green (SG) maize (Zea mays L.) genotypes. A pot experiment was conducted under controlled nutrition and growing conditions to determine the response of Leafy and SG maize genotypes to different levels of N-deficiency and timing of N supply. Three contrasting maize hybrids, Pioneer 3905 (a conventional hybrid with moderate SG characteristics), Pioneer 39F06 Bt (with a high score of SG trait) and Maizex LF850-RR (with a Leafy trait) were grown in 6 L plastic pots. Five different N treatments [no supply of N until V8 (N1), no supply of N after V8 (N2), no supply of N after silking (N3), no supply of N beyond 3 weeks after silking (N4), and continuous N supply from emergence to physiological maturity (N5; standard check)] were imposed through modified Hoagland solution applied manually. Labeled 15N of 5% 15N2–NH4NO3fertilizer was applied at 3 g per pot at the start of each schedule N treatment. Total amounts of N applied in different treatments were 3.13, 1.32, 1.90, 2.63 and 3.40 g, respectively in N1, N2, N3, N4 and N5. Dry matter, N concentration, 15N (atom% enrichment) and NUE were determined in roots, stalk, leaves and grains at crop maturity. The three contrasting hybrids did not differ in grain yield, total N acquisition, partitioning of 15N and NUE. Restriction of N supply until V8, and from V8 to physiological maturity significantly reduced grain yield and N-uptake in all hybrids. Irrespective of the level of N-deficiency in plant and timing when the labeled fertilizer was applied, the amount of 15N recovered in the matured plant was the same in all N treatments. It has been evident that maize continued to take up N beyond 3 weeks after silking and the later N was applied during the development, the higher proportion of it was partitioned to grains. Of the total 15N uptake, 78% was partitioned to kernels in the N4 treatment compared to only 61% in the control. Our data showed no evidence of differential N uptake, remobilization and NUE in the SG or Leafy hybrids tested, but the timing of N application and level of N-deficiency in plant significantly influenced N uptake, remobilization and N-dynamics in maize.  相似文献

Habitat fragmentation is a major cause of species rarity and decline because it increases local population extinctions and reduces recolonisation rates of remnant patches. Although two major patch characteristics (area and connectivity) have been used to predict distribution patterns in fragmented landscapes, other factors can affect the occurrence of a species as well as the probability of it becoming extinct. In this paper, we study the spatial structure and dynamics of the butterfly Iolana iolas in a 75-patch network of its host plant (Colutea hispanica) to determine the relative importance of patch area, connectivity and habitat quality characteristics on occupancy, extinction and density over the period 2003–2006. Occupancy in 2003, incidence (proportion of years occupied) and probability of extinction were mostly affected by patch area. Smaller patches were less likely to be occupied because they had a higher probability of extinction, partly due to environmental stochasticity. The density of I. iolas was negatively related to patch area in all study years. Only in 2004 was the density of I. iolas positively influenced by fruit production per plant. Our results suggest that for I. iolas, and probably for other specialist butterflies with clearly delimited resource requirements, metapopulation dynamics can be satisfactorily predicted using only geometric variables because most habitat characteristics are subsumed in patch area. However, this hypothesis should be subject to further testing under diverse environmental conditions to evaluate the extent of its generalisation.  相似文献

Cell suspensions of Methanobrevibacter arboriphilus catalyzed the reduction of O2 with H2 at a maximal specific rate of 0.4 U (mol/min) per mg protein with an apparent K m for O2 of 30 M. The reaction was not inhibited by cyanide. The oxidase activity was traced back to a coenzyme F420-dependent enzyme that was purified to apparent homogeneity and that catalyzed the oxidation of 2 F420H2 with 1 O2 to 2 F420 and 2 H2O. The apparent K m for F420 was 30 M and that for O2 was 2 M with a V max of 240 U/mg at 37°C and pH 7.6, the pH optimum of the oxidase. The enzyme did not use NADH or NADPH as electron donor or H2O2 as electron acceptor and was not inhibited by cyanide. The 45-kDa protein, whose gene was cloned and sequenced, contained 1 FMN per mol and harbored a binuclear iron center as indicated by the sequence motif H–X–E–X–D–X62H–X18D–X60H. Sequence comparisons revealed that the F420H2 oxidase from M. arboriphilus is phylogenetically closely related to FprA from Methanothermobacter marburgensis (71% sequence identity), a 45-kDa flavoprotein of hitherto unknown function, and to A-type flavoproteins from bacteria (30–40%), which all have dioxygen reductase activity. With heterologously produced FprA from M. marburgensis it is shown that this protein is also a highly efficient F420H2 oxidase and that it contains 1 FMN and 2 iron atoms. The presence of F420H2 oxidase in methanogenic archaea may explain why some methanogens, e.g., the Methanobrevibacter species in the termite hindgut, cannot only tolerate but thrive under microoxic conditions.Dedicated to Hans Schlegel on the occasion of his 80th birthday.  相似文献

A callus induction and in vitro plantlet regeneration system for the endangered state flower of Uttaranchal (Saussurea obvallata) was optimized by studying the influence of explant type (root, hypocotyl, cotyledon and leaf), age and different concentrations of plant growth regulators. Explants from 10 to 15-day-old seedlings showed maximum callus induction. Callus formation and shoot differentiation was initiated on Murashige-Skoog (MS) medium containing 6-benzyladenine (BA) and -naphthalene acetic acid (NAA) in all explant types. The best results were obtained using leaf explants: 100% callusing was achieved in MS medium supplemented with 2.5 M BA and 1.0 M NAA, and 100% differentiation along with a multiplication rate of 12 shoots per explant with a combination of 5.0 M BA and 1.0 M NAA. However, the results reflected the existence of high inter-explant variability in response to growth regulators. In vitro rooting of shoots was achieved at an efficiency of 100% in one-half strength MS medium supplemented with 2.5 M indole-3-butyric acid. Application of this protocol has potential for mass multiplication of the target species in a limited time period.  相似文献

Mutation is the source of all genetic variation, but rate of input and effects of new mutations for phenotypic traits related to fitness and the role they play in the maintenance of genetic variation are still subject to controversy. These parameters are important in models of the evolution of sex and recombination, the persistence of asexual populations, and the extinction of small populations. Most estimates have come from a few model organisms. Here, mutation accumulation experiments were conducted with three clones of grape phylloxera, Daktulosphaira vitifoliae Fitch, a gall forming herbivore and pest of grapes, to estimate the rate of input and effects of spontaneous mutation on life history traits. This is perhaps the first such experiment using a non-model organism of economic importance. Significant heritable genetic variation accrued in one of three sets of lines for one of four traits measured, and deleterious effects of mutation were found for two of four traits in two of the three sets of lines. Estimates of the parameters by the Bateman–Mukai (BM) method were within the range found in previous studies but at the lower end for genomic mutation rate, U 0.023 and mutational variance, V M 0.0003, the upper end for average heterozygous effect, , of –0.11, and on the order of previous estimates for mutational heritability, h M 0.007. Under a model of equal effects of mutations, maximum likelihood (ML) estimates of U were slightly higher, and of lower, than the BM estimates. Summary The Bacillus subtilis cdd gene encoding cytidine/2-deoxycytidine deaminase has been located by transduction at approximately 225 degrees on the chromosome, and the gene order rpC-lys-cdd-aroD was established. The gene was isolated from a library of B. subtilis DNA cloned in D69 by complementation of an Escherichia coli cdd mutation. Minicell experiments revealed a molecular mass of 14000 dalton for the cytidine deaminase subunit encoded by the cloned DNA fragment. The molecular weight of the native enzyme was determined to be 58000, suggesting that it consists of four identical subunits. The nucleotide sequence of 1170 bp, including the cdd gene, was determined. An open reading frame encoding a polypeptide with a calculated molecular mass of 14800 dalton was deduced to be the coding region for cdd. The deduced amino acid composition of the 136-amino acid-long subunit shows that it contains six cysteine residues. A computer search in the GenBank DNA sequence library revealed that the 476 bp HindIII fragment containing the putative promoter region and the first ten codons of cdd is identical to the P43 promoter-containing fragment previously isolated by Wang and Doi (1984). Dispersal in myrmecochorous herbs is frequently short-distance and this might limit patch occupancy. Here we ask whether recruitment and patch occupancy in the myrmecochere Helleborus foetidus are seed- or microsite-limited and what role dispersal plays in recruitment limitation. In pine forests from two widely separated mountain systems in the Iberian Peninsula, we performed two seed addition experiments to explore the nature of recruitment limitation in this perennial herb. In an introduction experiment, we added seeds in patches far from reproductive individuals, which rarely receive any seed. In an augmentation experiment, we added seeds in patches beneath reproductive plants. Each experiment considered three main factors: seed addition (added or not added), rodent exclusion (excluded or not excluded), and microhabitat (scrub-covered or not scrub-covered). The augmentation experiment tests whether recruitment is limited by seed availability (exploring also the possible role of rodent granivory in such limitation) while the introduction experiment tests whether the empty patches occur either because they are unsuitable for the species (microsite limitation) or due to a low probability of seed arrival (dispersal limitation). Adding seeds almost always increased seedling emergence and recruitment, demonstrating that recruitment in this species is seed-limited and refuting the possibility of microsite limitation. The comparison of the introduction and augmentation experiments indicated that Helleborus recruitment dynamics and patch occupancy are constrained by additive effects of fecundity and dispersal limitation.  相似文献

We have applied enzyme kinetic analysis to electrophysiological steady-state data of Zhou et al. (Zhou, J.J., Trueman, L.J., Boorer, K.J., Theodoulou, F.L., Forde, B.G., Miller, A.J. 2000. A high-affinity fungal nitrate carrier with two transport mechanisms. J. Biol. Chem. 275:39894–9) and to new current-voltage-time records from Xenopus oocytes with functionally expressed NrtA (crnA) 2H+-NO 3 symporter from Emericella (Aspergillus) nidulans. Zhou et al. stressed two Michaelis-Menten (MM) mechanisms to mediate the observed nitrate-induced currents, I NO 3 . We show that a single straightforward reaction cycle describes the data well, pointing out that during exposure to external substrate, S = (2H++NO 3 )o, the product concentration inside, [P] = [H+] i 2 · [NO 3 i, may rise substantially near the plasma membrane, violating the condition [P] [S] for MM kinetics. Here, [P] and its changes during experimentation are treated explicitly. K 1/2 20 µM for I NO 3 at pHo from Zhou et al. is confirmed. According to our analysis, NrtA operates between about 0.2 and 0.6 of the electrical distance in the membrane (outside 0, inside 1). In absence of thermodynamic gradients, the predominant orientation of the binding site(s) is probably inwards. The activity of the enzyme is sensitive to the transmembrane voltage, V, with an apparent gating charge of +1.0 ± 0.5 for inactivation, and transition probabilities of 0.3–1.3 s–1 at V = 0. This gating mode impedes loss of cellular NO 3 during depolarization.  相似文献
