Our current understanding of congenital transmission of Toxoplasma gondii from ewe to lamb dictates that infection frequently results in abortion and the death of the developing foetus, that the birth of live infected lambs occurs rarely and that the cat is the predominant source of infection in ewes. Using direct polymerase chain reaction detection of T. gondii, we report high levels of congenital transmission occurring in a commercially managed sheep flock. We sampled foetal-derived placental tissue and tissues from aborted lambs and showed that congenital transmission was detected in these tissues from 61% of all pregnancies. Where pregnancies resulted in the death of one or more lambs, T. gondii was detected in the lamb tissue for all but one of 18 (94%) pregnancies. Of the successful pregnancies resulting in the birth of live lambs we were able to detect T. gondii in foetal-derived placental tissue from 37 of 70 (42%) pregnancies. These results show that congenital transmission is occurring in a high percentage of lambings including normal healthy lambings, at this farm, suggesting that this route of transmission from generation to generation may be much more significant than that reported previously. These results may have implications for sheep husbandry and future epidemiological studies of T. gondii.  相似文献

A study carried out on a sheep farm examined whether Toxoplasma gondii foetal infection and associated abortion occur in successive lambings. We identified 29 ewes that gave birth to lambs on at least 2 successive years over our study period, 2000-2003. Tissue samples from the progeny of these ewes were analysed by PCR to determine infection status with T. gondii. T. gondii-infected lambs were born in 31% of successive pregnancies. T. gondii-positive lambs were aborted in successive pregnancies in 21% of lambings during study period, 2000-2003. The frequency of successive abortions within this flock over the period 1992-2003 was 18%. If a lamb was congenitally infected there was a high risk (69%) that the successive lamb from that ewe would also be congenitally infected. Similarly, if a lamb was aborted there was a high risk (55%) of abortion in the next lamb produced. These data suggest that life-long immunity to T. gondii infections may not always be acquired following an initial infection and raises the question as to whether the mechanisms of T. gondii transmission prior to and during ovine pregnancies are fully understood.  相似文献

Trypanosoma cruzi, the causative agent of Chagas disease, and Toxoplasma gondii, which is responsible for Toxoplasmosis, are two parasites that cause significant protozoan zoonoses and consequently important economic losses in human, companion animals and livestock. For the congenital transmission to occur, both parasites must cross the barrier present in the mammalian placenta, which differs between species. Particularly, hemochorial, endotheliochorial and epitheliochorial placental barriers are present, respectively, in human, dog and sheep. The type of placental barrier has been associated with the probability of transmission of pathogens. In this study, we used experimental placental ex vivo infection models of T. cruzi and T. gondii in the above-mentioned mammals in order to study tissue alterations and to compare infection efficiency. Here, we infected placental term explants from human, dog and sheep and analyzed tissue damage by standard histological and histochemical methods. Comparative infection efficiency was determined by quantitative PCR. Both parasites are able to infect the different placental explants; however, more T. gondii parasites were detected, and T. gondii causes a more severe tissue damage in human and canine explants than T. cruzi. The histopathological changes observed in ovine placenta explants were similar in presence of both parasites. We conclude that the infection efficiency of T. gondii is higher, compared to T. cruzi, during the ex vivo infection of human, canine and ovine placental explants.In addition, the ex vivo infection of mammalian placental explants constitutes an interesting experimental approach to study part of the infection mechanisms as well as host responses during congenital infection of both parasites.  相似文献

An investigation of stillbirth and early neonatal lamb mortality was conducted in sheep flocks in Norway. Knowledge of actual causes of death are important to aid the interpretation of results obtained during studies assessing the risk factors for lamb mortality, and when tailoring preventive measures at the flock, ewe and individual lamb level. This paper reports on the postmortem findings in 270 liveborn lambs that died during the first 5 days after birth. The lambs were from 17 flocks in six counties. A total of 27% died within 3 h after birth, 41% within 24 h and 80% within 2 days. Most lambs (62%) were from triplet or higher order litters. In 81% of twin and larger litters, only one lamb died. The most frequently identified cause of neonatal death was infectious disease (n=97, 36%); 48% (n=47) of these died from septicaemia, 25% (n=24) from pneumonia, 22% (n=21) from gastrointestinal infections and 5% (n=5) from other infections. Escherichia coli accounted for 65% of the septicaemic cases, and were the most common causal agent obtained from all cases of infection (41%). In total, 14% of neonatal deaths resulted from infection by this bacterium. Traumatic lesions were the primary cause of death in 20% (n=53) of the lambs. A total of 46% of these died within 3 h after birth and 66% within 24 h. Severe congenital malformations were found in 10% (n=27) of the lambs, whereas starvation with no concurrent lesions was the cause of death in 6% (n=17). In 16% (n=43) of the lambs, no specific cause of death was identified, lambs from triplet and higher order litters being overrepresented among these cases. In this study, the main causes of neonatal lamb mortality were infection and traumatic lesions. Most neonatal deaths occurred shortly after birth, suggesting that events related to lambing and the immediate post-lambing period are critical for lamb survival.  相似文献

Two studies, of a natural infection and an experimental infection, were performed in order to study congenital transmission of Toxoplasma gondii in cattle. In the first study, 50 fetuses were harvested from gestating cows that were eutanasied at a municipal slaughterhouse in Jaboticabal, São Paulo state, Brazil. In the second study, 11 gestating cows were divided into four groups for inoculation with T. gondii: GI consisted of three cows inoculated with 1.0 × 105 oocysts during their first trimester of gestation; GII consisted of three cows inoculated with 1.0 × 105 oocysts during their second trimester of gestation; GIII consisted of three cows inoculated with 1.0 × 105 oocysts during their last trimester of gestation; and GIV consisted of two control cows, one during its first and the other during its second trimester of gestation. In both studies, the presence of T. gondii was confirmed both indirectly by immunofluorescence assay (IFAT). In the natural infection experiment, 18% (9/50) of the gestating cows were confirmed to have specific antibodies (IFAT – 1:64) against T. gondii. The bioassay was able to diagnose the presence of T. gondii in the tissue samples from three calves. In the second experiment, the nine cows from groups I, II and III presented with specific antibodies (IFAT) against T. gondii. In contrast, T. gondii could not be detected by IFAT, histopathological examination or the bioassay in any of the nine calves born to cows experimentally infected with T. gondii oocysts. Based on the results from both studies, we conclude that congenital infection of T. gondii in cattle, while infrequent, does occur naturally. The pathogenicity of the strain of T. gondii may influence the likelihood of this route of transmission.  相似文献

Congenital toxoplasmosis can cause fetal damage in humans and domestic animals. This study was focused on the effects of Toxoplasma gondii (Prugniaud strain) infection at different stages of pregnancy on the offspring of mice. Results showed that newborn mice from all infected groups were significantly lower in weight than those from the control group but significant difference was not found among these groups at day 60 after birth. The survival rate of the offspring from the group of mice infected at the earlier stage of pregnancy was significantly lower than those of infected and control groups. The positive offspring (with cysts found in their brain tissues) born from the mice infected at the earlier and intermediate stages of pregnancy showed a shorter latency and greater number of errors in the step-through passive avoidance test than those born from the mice infected at the late stage of pregnancy, the control group and the negative offspring from the infected groups. The number of cysts in the brain tissue was significantly higher in the offspring born from the groups of mice infected at the earlier and intermediate stages of pregnancy than those from the group of mice infected at the late stage of pregnancy. In addition, our results indicated that a high congenital transmission rate (90%) occurred in this NIH mouse model. In conclusion, the earlier and intermediate maternal infection of T. gondii can result in severe congenital toxoplasmosis, exhibiting conditions such as stillbirth or non-viability, and learning or memory capability damage in this mouse model. These results not only provide useful data for better understanding the effects of T. gondii infection on the offspring of mice infected at different stages of pregnancy but also for better consideration of the effect of this infection on other mammalian hosts including humans.  相似文献

A low finishing weight and poor carcass characteristics are major causes of lower incomes in extensive sheep flocks; however, the use of terminal sire crossbreeding would improve lamb performance and carcass traits under these conditions. The aim of this study was to evaluate sire breed effects on the performance of lambs born to Corriedale ewes in extensive sheep systems in Western Patagonia. A total of 10 Corriedale, 10 Dorset, nine Suffolk and seven Texel sires, 16 of which were under a genetic recorded scheme and 20 selected from flocks not participating in genetic improvement programmes, were used across six commercial farms for 2 successive years. Data were collected from 685 lambs of the four resulting genotypes. Overall, Corriedale lambs were 0.47 kg lighter at birth than crossbred lambs (P<0.001). Suffolk and Texel sired lambs required more assistance (P<0.01) at birth than Corriedale or Dorset sired lambs, with Suffolk sired lambs requiring the most assistance (8%). Ewes sired with Suffolk rams had larger (P<0.05) litters than ewes sired with Texel or Corriedale rams. Lamb live weight gain from birth to weaning was higher (P<0.001) in crossbred lambs compared with Corriedale lambs, therefore, crossbred lambs averaged 2.9 kg heavier BW (P<0.001) than Corriedale lambs. A significant sire breed x sire source interaction was detected for lamb live weight gain (P<0.05) and lamb live weight at weaning (P<0.01), showing that the heaviest lambs were from recorded sires, except for Suffolk crossbred lambs. Mortality rate to weaning was increased (P<0.05) in Suffolk cross lambs (31%), with Corriedale lambs showing the lowest (17%) mortality. Terminal sire breeds increased (P<0.001) cold carcass weight, with 13.8, 16.0, 15.2 and 14.9 kg for the Corriedale, Dorset, Suffolk and Texel sired lambs, respectively. Carcass length, kidney knob and channel fat, fat grade, grade rule and fat depth measurements were not affected by sire breed (P>0.05). Carcass conformation was higher in Texel sired lambs compared with Corriedale lambs (P<0.05), with Dorset and Suffolk sired lambs being intermediate. Crossbred lambs showed a greater (P<0.001) eye muscle than Corriedale. Commercial cuts were affected by sire breed, as a result of the Corriedale lambs being smaller and having lighter carcass than crossbred lambs. Significant improvement in lamb weights at weaning and carcass traits could be expected when using a terminal sire on Corriedale ewes in Western Patagonia. However, no advantages were detected with the use of recorded sires under these production systems.  相似文献

Lamb survival is impaired in low birth weight lambs, and those that are slow to stand and suck. Many of the factors that influence lamb vigour, such as parity, litter size, and breed, may exert their effects, at least partially, before birth by influencing placenta development. Our hypothesis was that retarded lamb behavioural development was due to differences in placentation in these animals. Data were collected from Blackface and Suffolk lambs in the first 2 h after birth and placentas were collected when delivered. Suffolk lambs, which were behaviourally slower and had lower rectal temperatures than Blackface lambs, were associated with larger but less efficient placentas (placental efficiency defined as foetal weight supported per g placenta) with fewer foetal cotyledons than Blackface placentas. Triplet lambs were significantly slower than twin or single lambs to suck and had lower rectal temperatures. Although placenta efficiency increased with litter size, placenta and cotyledon weight, and cotyledon number increased with twinning but not thereafter. It seemed likely that triplet lambs suffered some placental insufficiency in comparison to other litter sizes. Lambs born to first parity mothers were slower to stand and reach the udder than lambs of more experienced ewes, and first parity ewes also had smaller and less efficient placentas although cotyledon number was not affected. Male lambs tended to be slower than female lambs for most behaviours, although rectal temperatures were not affected. The sire of the lamb also influenced lamb behaviour and rectal temperature. Both lamb sex and lamb sire influenced the average weight of placental cotyledons, thus some of the sire effect on the behaviour and birth weight of his progeny might be mediated through placental development. Lamb neonatal vigour was correlated with placental efficiency suggesting that lamb behaviour immediately after birth is related to placental development and function.  相似文献

Tick-borne fever (TBF) is caused by the rickettsia Anaplasma phagocytophilum (formerly Ehrlichia phagocytophila) and is a common disease in sheep in areas of Norway infested by Ixodes ricinus ticks. TBF can cause both direct and indirect losses to sheep kept on tick-infested pastures. In the present work we studied a sheep flock of 26 ewes and 50 lambs on pasture from May until September. No cases of TBF had earlier been observed on this pasture. Blood samples from lambs with a reduced weekly weight gain were collected and analysed for A. phagocytophilum infection by blood smear examination. In addition, at the end of the study, sera from all lambs were analysed by an indirect fluorescent antibody assay (IFA) to determine the antibody titre to E. equi. No clinical signs of tick-borne infections were observed, except in one lamb. However, 30 (60%) of the lambs grazing on this pasture became infected with A. phagocytophilum, and the infected lambs had a reduced weight gain (mean) of 3.8 kg compared with the uninfected lambs. The present study indicates that A. phagocytophilum infection may be widespread and contribute to considerable productivity losses even on pastures with no apparent tick infestation. Background

The risk of scrapie infection increases with increased duration and proximity of contact between sheep at lambing. Scrapie infectivity has not been detected in milk but cellular prion protein, the precursor of disease-associated prion protein PrPd, has been found in milk from ruminants. To determine whether milk is able to transmit scrapie, 18 lambs with a prion protein genotype associated with high susceptibility to scrapie (VRQ/VRQ) were fed milk from twelve scrapie-affected ewes of the same genotype, and 15 VRQ/VRQ sheep reared on scrapie-free dams served as controls.

Results

Three lambs fed milk from scrapie-affected ewes were culled due to intercurrent diseases at 43, 44 and 105 days of age respectively, and PrPd was detected in the distal ileum of the first two lambs, whilst PrPd was not found in lymphoreticular tissues in the third lamb. A control lamb, housed in a separate pen and culled at 38 days of age, was also negative for PrPd in a range of tissues. Samples of recto-anal mucosa associated lymphoid tissue collected from the remaining 15 live lambs at seven months of age (between five to seven months after mixing) were positive for PrPd in the scrapie milk recipients, whereas PrPd was not detected in the remaining 14 controls at that time. A subsequent sample collected from control lambs revealed PrPd accumulation in two of five lambs eight months after mixing with scrapie milk recipients suggestive of an early stage of infection via lateral transmission. By contrast, the control sheep housed in the same building but not mixed with the scrapie milk recipients were still negative for PrPd.

Conclusion

The presence of PrPd in distal ileum and rectal mucosa indicates transmission of scrapie from ewe to lamb via milk (or colostrum) although it is not yet clear if such cases would go on to develop clinical disease. The high level of infection in scrapie-milk recipients revealed by rectal mucosal testing at approximately seven months of age may be enhanced or supplemented by intra-recipient infection as these lambs were mixed together after feeding with milk from scrapie-affected ewes and we also observed lateral transmission from these animals to lambs weaned from scrapie-free ewes.

Lamb mortality remains a significant welfare and economic issue for sheep production. Lamb survival is to a degree dependent upon an easy delivery and the expression of appropriate behaviours from both mother and offspring, such as rapid standing, udder seeking and sucking by the lamb. Genetic solutions have the potential to improve birth assistance and lamb behaviour but large amounts of data are needed. Therefore, to achieve this objective, simple, proxy methods (scoring systems) were developed to quantify the level of birth difficulties and lamb vigour on farm. In the first study, detailed historical behavioural data from 1156 lambs (Scottish Blackface and Suffolk (S)) were analysed to develop criteria for 3 scores: birth assistance, lamb vigour and sucking assistance. The birth assistance score was developed by analysing the relationships between birth presentation and intervention levels, and intervention level and labour length. Lambs with abnormal birth positions required more assistance than normally presented lambs and lambs with long labours required more and greater assistance than those with short labours. Lamb vigour score was developed by analysing the latencies for the lamb to first perform specific behaviours; more vigorous lambs reach landmark behaviours faster than low vigour lambs. The sucking assistance score was developed from the relationship between the latency to suck successfully and assistance level, where lambs that were slow to suck required more assistance than lambs that were quick to suck. In the second study, the behaviour scoring systems (5-point categorical scales) were validated using a commercial flock of 80 twin-bearing crossbred ewes mated with either Texel (T) or S sires by simultaneously recording scores and the latency to perform specific landmark behaviours (i.e. to stand, seek the udder and suck). The vigour scores (recorded at 5 min of age) were compared with the latency from birth to standing and showed that lambs with lower (better) vigour scores were faster to stand after birth than those with higher scores. The sucking assistance scores were compared with the latency from birth to sucking, and showed that lambs with lower sucking assistance scores are quicker to suck than those with high scores. These results showed that the scoring systems could provide a practical and reliable assessment of birth assistance and lamb behaviour on farm and were sufficiently sensitive to discriminate vigour levels between lambs sired by either S or T rams.  相似文献

Wool growth in Merino sheep demands a high level of sulphur amino acids, competing with body growth and the immune system, which may play a role in increasing the risk of lamb mortality. The hypothesis that dietary supplementation of methionine (Met) to Merino ewes during the late stages of pregnancy will improve foetal growth and alter immune competency of ewes and lambs was tested in a total of 120 grazing, pregnant Merino ewes. Sixty ewes were group-supplemented with 6.3 g/d rumen-protected Met (Met-Plus) per sheep from day 111 of pregnancy until day 7 after lambing, and the other 60 animals were used as a non-supplemented Control. Lambs from Met-supplemented ewes tended to be 10% heavier than Control lambs (p = 0.10), which did not affected the survival rate at weaning significantly. The supplemented ewes had slightly higher concentrations of total glutathione (GSH) in plasma at lambing (p < 0.06), but there were no differences between 1 and 3 months post-lambing in GSH, glutathione disulphide (GSSG) and the GSSG:GSH ratio. The GSSG:GSH ratio in the blood of ewes was elevated at lambing (p < 0.05), hinting that ewes were undergoing increased oxidative stress. The Met supplementation elevated the total IgG concentration (p < 0.05) in lambs aged 4 and 6 weeks, but did not change the IgG concentrations in colostrum and in plasma of 1-week-old lambs, and white blood cell counts and leukocyte types. The trend towards higher lamb birth weights in the Met-supplemented group requires further investigation as this may influence survival at birth and weaning.  相似文献

During lambing the expression of an appropriate behavioural response from both the ewe and the lamb are extremely important to lamb survival. The aim of this study was to show the effect of length and difficulty of the birth process on the expression of maternal and neonatal behaviour with consequences on homeothermy and survival of the neonate lamb. Data were collected from 61 Finnish Landrace × Rahmani crossbred (second generation) primiparous ewes and their single born lambs. Based on the average length of parturition, the ewes were grouped into short birth (less than 32.5 min) and long birth (equal to or higher than 32.5 min) classes. The data recorded include maternal and neonatal behaviour, lamb body temperature over the first 3 days of life and survival rate of the neonate lamb during the first week after birth. Blood samples were also collected from the lamb, pre-suckling and at 24 and 72 h after birth. The obtained sera were assayed for thyroid hormones (T3 and T4) that are associated with heat production. Ewes that had prolonged and difficult births did not show competent maternal behaviour compared to mothers with short and un-complicated deliveries, as they were slower to begin grooming their lambs after birth, spent less time licking their lambs, made less low-pitched vocalizations and nosing, were more likely to show rejection behaviour (10.34 % vs 5.4 %, P < 0.05), and were more likely to move away when the lamb sought the udder in an attempt to suck (acceptance rate, 55.5 % vs 64.79 %, P < 0.05). Similarly, lambs from a prolonged and difficult birth were significantly less vigorous after birth, as they had taken more time to stand, reach the udder and to suck successfully. These lambs had lower serum concentrations of T3 and T4, and they also had a reduced ability to maintain body temperature after birth. This effect persisted over the first 3 days of life and was associated with higher neonatal mortality in the first week after birth (11.54 %), compared to lambs from short and non-stressful birth processes (2.86 %, P < 0.01). From the present study, it can be concluded that, prolonged deliveries with birthing difficulty were one of the main causes of death of large, single-born lambs, as these complications cause the expression of inappropriate behavioural responses from both the ewe and neonatal lamb. Thus, interventions designed to reduce the incidence of prolonged parturitions are likely to be associated with better welfare for the ewe and the lamb and consequently improved lamb homeothermy and survival.  相似文献

The Muzaffarnagari sheep, a mutton-producing breed of India, is generally known for its fast growth rate and high feed conversion efficiency. The study identified the causes of perinatal and postnatal deaths and factors contributing to the cause of mortalities in lambs. Data were recorded for 4628 lambs born to Muzaffarnagari ewes in single-sire matings from 165 rams, between 1978 and 2002. The least-squares means for overall lamb mortality from birth to 1 year of age in the flock was 12.6% (582/4628), while the overall pre- and post-weaning mortality rate averaged 6.6% and 6.0%, respectively. The mortality rate of the lambs declined as the lambs grew older. The major causes associated with lamb losses were pneumonia (31.4%), digestive disorders (14.6%), starvation (9.6%), endoparasitism (5.0%), septicaemia and toxaemia (10.1%), accidental (2.1%) and undetermined causes (27.2%), respectively. The main causes of early lamb mortality (within 15 days of birth) were pneumonia followed by starvation. The highest lamb mortalities occurred within the first 15 days of life. The year of birth of the lamb significantly affected (P < 0.01) the cumulative lamb mortality rate at all ages. The sire of the lamb also had a significant (P < 0.01) effect on lamb mortality rate at all ages. Lambs reared under an intensive feeding management system had a remarkably (P < 0.01) lower mortality rate than lambs maintained under a semi-intensive management system, at 3 months of age and later. The lamb losses were significantly (P < 0.01) higher in ram lambs than in females during the post-weaning stages. The linear regressions of lamb birth weight and dam weight at lambing were highly significant (P < 0.01) with lamb mortality rate during pre- and post-weaning stages. Logistic regression analyses revealed lambs maintained under a semi-intensive system, to be at a greater risk of dying from all ailments than lambs maintained under intensive feeding systems. Male lambs had greater risk of death from respiratory, digestive and other causes, compared to their female counterparts. The findings of this study revealed causes of pre-weaning lamb mortalities to be mainly due to a low birth weight and non-parasitic diseases (predominantly pneumonia). The aim of this study was to evaluate the effects of age and breed on the reproductive performance and lamb output of crossbred hill ewes relative to purebred Scottish Blackface (BF). BF ewes were compared alongside Swaledale (SW) × BF, North Country Cheviot (CH) × BF, Lleyn (LL) × BF and Texel (T) × BF ewes on six commercial hill farms across Northern Ireland, on which all the ewes were born and reared. Ewes were mated to a range of sire breeds, balanced across breeds, for up to five successive breeding seasons. Mature live weight of adult BF, SW × BF, CH × BF, LL × BF and T × BF ewes was 52.8, 54.9, 60.3, 55.6 and 58.6 kg (P < 0.001), respectively. Compared with the pure BF, the number of lambs born per ewe lambed was higher with LL × BF and SW × BF (P < 0.05), whereas the number of lambs weaned per ewe lambed was greater for LL × BF and T × BF (P < 0.01). Total litter weight at birth of all the crossbred ewes was heavier (P < 0.01) than the pure BF, except in primiparous 2-year-old ewes. Lambs born to CH × BF and T × BF dams were 0.24 to 0.35 kg heavier at birth (P < 0.01) than the other ewe breeds, whereas lambs born to CH × BF, LL × BF and T × BF dams were, on average, 1.7, 1.3 and 1.5 kg, respectively, heavier (P < 0.01) at weaning than those from BF dams due to their higher (P < 0.05) average daily gain. Compared with the pure BF, total weaned lamb output per ewe lambed was 3.7, 4.8, 6.7 and 5.4 kg heavier (P < 0.05) for SW × BF, CH × BF, LL × BF and T × BF, respectively. However, as a result of the heavier live weight of the crossbred ewes, production efficiency (lamb output per kilogram live weight (W) and lamb output per kilogram metabolic live weight (W0.75)) was higher (P < 0.001) for LL × BF ewes only. For all ewe breeds, litter size at birth per ewe lambed, total lamb birth weight per ewe lambed and litter size at weaning increased (P < 0.001) with age up to 5 years, but decreased in 6-year-old ewes. Average lamb weaning weight and total weaned lamb output per ewe lambed increased (P < 0.001) with age up to 4 years . Production efficiency of the 6-year-old ewes was lower (P < 0.01) than the younger ewes. This study shows that adopting a flock replacement policy based on crossing BF ewes with LL, SW, T and CH sires can lead to significant improvements in the productivity of hill flocks.  相似文献

Shearing during the latter half of pregnancy is a common practice to improve flock health and productivity. Previous studies have demonstrated that shearing pregnant ewes at mid or late pregnancy is associated with an increase in lamb birth weight. In the present study, we used singleton Polypay × Dorset pregnant sheep, to investigate the potential roles of placental function and changes in maternal metabolism in underlying this increased birth weight response. Two groups were randomly established and blocked at enrollment by animal BW, body condition score and subcutaneous adipose tissue depth. The groups were shorn (SH; n = 18) or not (C; n = 20) at gestational day (GD) 107 ± 1 (mean ± SEM). Weekly maternal plasma samples were collected between shearing and birth, but only six samples were assayed for progesterone, pregnancy-associated glycoproteins (PAG1), glucose and non-esterified fatty acids (NEFAs). At birth, sex, birth weight, and newborn body mass index (BMI) were recorded. Maternal BW during mid- to late-pregnancy was similar between groups. Shearing resulted in increased lamb birth weight and BMI (P < 0.05) regardless of fetal sex but did not affect the maternal concentration of PAG1 or progesterone from GDs 100 to 142. After shearing (GD100) and up to lambing, shorn females had higher circulating glucose concentrations (P < 0.05), but not NEFA, compared to the control group. Maternal circulating PAG1, progesterone, glucose or NEFA concentration across pregnancy did not differ according to lamb sex. Across pregnancy, birth weight was positively associated with PAG1 (P < 0.001), but not with progesterone concentrations. In conclusion, weight and BMI at birth were higher in both sexes upon shearing in singleton pregnancies. Despite PAG1 being associated with birth weight, late-pregnancy shearing did not alter the placental endocrine response. Whether other placental factors are altered upon shearing and may influence the increase in birth weight and BMI remain to be investigated.  相似文献

Understanding how species respond to environmental conditions can assist with conservation strategies and harvest management, especially in arctic and boreal regions that are experiencing rapid climate change. Although climatic influences on species distributions have been studied, broad-scale effects of climate on survival are less well known. We examined the interactive effects of meteorological and remotely sensed environmental variables on survival of Dall's sheep (Ovis dalli dalli) lambs and adults by synthesizing radio-telemetry data across their range. We used data from 9 studies of adult sheep and 2 studies of lambs that were conducted between 1997 and 2012 at sites spanning the species' range in Alaska, USA, and northwestern Canada. We obtained environmental variables throughout the range of Dall's sheep, including the normalized difference vegetation index (NDVI) from optical remote sensing, freeze-thaw frequency (FTF) from passive microwave remote sensing, and gridded climate variables such as snow water equivalent, temperature, and precipitation. We used Cox proportional hazard regression to investigate the effects of environmental variables recorded during summer, winter, and the previous winter on annual survival rates of Dall's sheep lambs and adults. Summer NDVI was the most influential environmental factor affecting lamb survival, with improved lamb survival occurring in years with a high maximum NDVI. Also, lamb predation by coyotes (Canis latrans) and golden eagles (Aquila chrysaetos) decreased substantially with increasing NDVI. The previous winter FTF had the strongest effect on adult survival, with decreased survival occurring after winters with high FTF. In addition, these remotely sensed environmental factors interacted with meteorological factors to affect survival, such that effects of winter temperature depended on summer NDVI and winter FTF. Warm winters increased lamb survival only when preceded by summers with high NDVI, and warm winters increased adult survival only when winter FTF was low. Thus, potential benefits of climate warming may be counteracted if wintertime freeze-thaw events markedly increase. Correlations among environmental variables across sites were low, and regional climate cycles such as the Pacific Decadal Oscillation (PDO) had weak effects, indicating substantial local variability in climatic conditions experienced by Dall's sheep across their range. These findings can help managers anticipate how Dall's sheep populations will respond to changes in local environmental conditions. Our results also highlight the utility of multiple remotely sensed environmental conditions for ungulate management, especially passive microwave products that provide valuable information on winter icing events. © 2020 The Wildlife Society.  相似文献

The surface antigen 2 (SAG2) gene of the protozoan parasite, Toxoplasma gondii, was cloned and extracellularly expressed in the yeast Pichia pastoris. The effectiveness of the secreted recombinant SAG2 (rSAG2-S) as a serodiagnosis reagent was assessed by western blots and ELISA. In the western blot assay, rSAG2-S reacted with all Toxoplasma-antibody positive human serum samples but not with Toxoplasma-negative samples. In the ELISA, rSAG2-S yielded sensitivity rates ranging from 80% (IgG negative, IgM positive) to 100% (IgG positive, IgM negative). In vivo experiments showed that serum from mice immunized with rSAG2-S reacted specifically with the native SAG2 of T. gondii. These mice were protected when challenged with live cells of T. gondii.  相似文献

Undernutrition restricted to the time around conception in sheep alters endocrine and metabolic responses in the offspring. Studies in rats suggest that such an early insult can also alter the behaviour of the offspring. We studied the effects of mild maternal periconceptional undernutrition (10–15% body weight reduction) on the lamb's response to separation from and reunion with the mother, and on the ewe–lamb bond, evaluated as the preference for each other over an alien ewe/lamb in a test enclosure, at 24 h, 1 and 4 weeks of age. Lamb birth weight was not affected by maternal nutrition. Maternal periconceptional undernutrition did not affect the lambs’ responses to separation and reunion (number of vocalisations, times to leave pen and achieve proximity with ewes) or the bond between ewes and lambs (percentage of time spent near their own dam/lamb). However, there were effects of sex, litter size and time on lambs’ responses to separation and reunion and on the ewe–lamb bond. Female lambs vocalised more during separation (P ≤ 0.01) and left the holding pen faster than males (P ≤ 0.05), and twins vocalised more than singletons (P ≤ 0.05). The time to leave the holding pen, achieve proximity with the ewes and number of vocalisations decreased with increasing postnatal age of the lamb (all P ≤ 0.001). Ewes showed a stronger preference for the lamb at 24 h and 1 week than at 4 weeks postpartum (P ≤ 0.001), whereas lambs followed an opposite trend, showing a lower preference for the mother at 24 h and 1 week and increased preference at 4 weeks of age (P ≤ 0.001). We conclude that periconceptional undernutrition does not affect offspring response to separation from the mother or the bond between ewes and lambs, but that these behaviours are affected by sex and litter size for up to 4 weeks after birth.  相似文献

The objective of this study was to refine the rat model of congenital toxoplasmosis. In Fischer rats we found that visualization of spermatozoa in vaginal exudates and the detection of at least 6 g body weight increase between days 9 and 12 of pregnancy, allowed the diagnosis and timing of pregnancy with 60% specificity and 84% sensitivity. A dose of 104Toxoplasma gondii bradyzoites or 102T. gondii oocysts of the Prugniaud strain resulted in more than 50% of congenital infection of the rat litters. Transmission of T. gondii via lactation was not detected in rats inoculated with either bradyzoites or oocysts. Bioassays of 51 neonates born from mothers inoculated with bradyzoites (in tissue cysts) and 29 neonates from mothers inoculated with oocysts demonstrated that both liver and lungs can be used for the diagnosis of congenital transmission in this model.  相似文献
