图像融合技术在现代医学中扮演着极其重要的角色,是现代医学图像技术研究的重点。图像融合技术中,图像的配准又是其中的重点、难点和热点。本文按照图像变换特性对图像配准进行了分类,对每个类别的不同配准方法(特征点的获取、图像配准的变换等)进行介绍。但是,图像配准是一个尚处在发展阶段的学科,实现配准的精确化、快速化、自动化仍需要进一步的努力。  相似文献

Biolistic co-transformation of the nuclear and plastid genomes   总被引:1,自引:0,他引:1  
Particle gun-mediated (so-called 'biolistic') transformation represents a universal genetic transformation technology that is widely applied in nearly all groups of organisms. The mechanism of how accelerated DNA-coated particles, after their entry into the cell, deliver the foreign DNA to the target compartment is not known. Here we have studied this process in plants by performing co-transformation experiments with vectors targeted to two different cellular compartments, the nucleus and the plastids (chloroplasts). We find that coating of particles with both plastid and nuclear transformation vectors can result in co-transformation of chloroplasts and the nucleus. In contrast, mixing of particles coated individually with the vectors does not produce co-transformed plants. Our data suggest that a single DNA-coated particle can transform more than one compartment of the plant cell, opening up the possibility to generate doubly transgenic plants in one step. Importantly, co-transformation can also be obtained in the absence of selection, thus providing a method to produce marker-free transgenic genomes. In addition, our findings raise the possibility of occasional inadvertent co-transformation of two genomes and, therefore, have important implications for the molecular characterization and regulation of transgenic plants.  相似文献

电转化条件对大肠杆菌XL1-Blue菌株转化效率的影响   总被引:10,自引:0,他引:10  
探讨XL1-Blue菌株电转化的最优条件。通过改变电压、质粒DNA浓度、细菌生长周期等影响电转化的重要条件,做出转化率的变化曲线,从中探索电转化的最优条件。实验结果得出在电容25μF、电阻200 Ω、电压2.5 kV、D600nm为0.3~0.4、0.2 cm电转化杯、DNA终浓度0.1μg/ml、感受态细胞终浓度2.5×1012、氨苄青霉素浓度50μg/ml的条件下,电转化效率最高,可达到7.64×108。电转化实验转化效率高,重复性好,为成功的建立抗体库提供了保证。  相似文献

A review of the activity of metal compounds in mammalian cell transformation assays has been completed. Results from these assays appear to correlate well with the known carcinogenic activity displayed by specific metal compounds in vivo. Studies of cell transformation in vitro may provide information pertaining to the mechanism of the induction of carcinogenesis by certain metals.  相似文献

Cereal crops have been the primary targets for improvement by genetic transformation because of their worldwide importance for human consumption. For a long time, many of these important cereals were difficult to genetically engineer, mainly as a result of their inherent limitations associated with the resistance to Agrobacterium infection and their recalcitrance to in vitro regeneration. The delivery of foreign genes to rice plants via Agrobacterium tumefaciens has now become a routine technique. However, there are still serious handicaps with Agrobacterium -mediated transformation of other major cereals. In this paper, we review the pioneering efforts, existing problems and future prospects of Agrobacterium -mediated genetic transformation of major cereal crops, such as rice, maize, wheat, barley, sorghum and sugarcane.  相似文献

The ability to introduce individual molecules of plasmid DNA into cells by transformation has been of central importance to the recent rapid advancement of plasmid biology and to the development of DNA cloning methods. Molecular genetic manipulation of bacteria requires the development of plasmid-mediated transformation systems that include (1) chemical transformation, (2) electro-transformation, (3) biolistic transformation, and (4) sonic transformation, leading to the introduction of exogenous plasmid DNA into bacterial cells. In this review, the manipulation properties and transformation efficiencies of these techniques are described. In addition to these methods, a conceptually novel transformation technique, namely the hydrogel exposure method, was developed. The hydrogel exposure method, based on the Yoshida effect, provides a significant advance over chemical means for transforming many strains of Escherichia coli and a variety of other bacterial species. The new term “tribos transformation” has been proposed for this novel technique. We also determined that, compared to conventional methods, the hydrogel exposure method is a novel and convenient method by which to transform bacteria.  相似文献

建立并优化了农杆菌介导转化轮枝镰孢菌Fusarium verticillioides获得T-DNA插入突变体的体系,在镰孢菌孢子浓度106个/mL、农杆菌OD600=0.15-0.20、乙酰丁香酮浓度为200μmol/mL的条件下共培养36h转化率最高,可达60-120个/106个孢子。共获得转化子1000多个,连续转接5代能够稳定遗传。PCR验证潮霉素B抗性基因已整合进转化子基因组DNA中,部分转化子表现为生长和形态异常。该转化体系的建立为研究该菌的致病机制和功能基因分析奠定了基础。  相似文献

由于存在遗传背景狭窄、高度杂合化、育种周期长等特点,巴西橡胶树育种进展非常缓慢。转基因技术为拓展其的遗传范围、加速育种进程提供了契机。在过去20年里,橡胶树转基因研究取得了很大进展:成功应用的转化技术包括基因枪法和农杆菌介导法,受体材料包括花药和内珠被愈伤,育种目标涉及到提高胶乳产量、抗死皮和作为生物反应器等。但同时也存在组培程序不够成熟、转化技术和外植体比较单一、转化效率低下等问题。最后,对以后转化体系的优化、转基因育种方向进行了分析和展望。  相似文献

叶绿体转化系统,独立于传统的核转化,为植物导入外源基因提供了新途径,它有如下优点:超量表达目的的基因;以定点整合方式导入外源基因从而消除了位置效应及基因沉默;具有原核表达方式,能以多 反子的形式表达多个基因;母系遗传方式可防止基因扩散;基因产物区域化并能提供适于某些产物发挥功能的小环境等。随着这项技术在越来越多的领域发挥作用,其优势性逐渐得到认同。本文着重对此技术的特点,发展及应用作一综述。  相似文献

根癌农杆菌介导法(Agrohacterium mediated transformation)和基因枪轰击法( particle bombardment transformation)是植物遗传转化的主要方法。两种方法各有优缺点.农杆菌介导法是一种天然的植物遗传转化系统,外源基因在转基因植物中的拷贝数低,遗传稳定性好;基因枪转化法不受材料基因型的限制。通过结合两种方法的优点,发展了3种农杆菌介导和基因枪轰击法相结合的遗传转化方法,分别为农杆枪法、基因枪轰击/农杆菌感染法、金粉或钨粉包裹菌体细胞作为微弹轰击法。对3种结合转化方法的技术途径、原理、转化受体及研究进展等方面进行了综述。  相似文献

PEG/LiAc法转化酵母适用于酵母双杂交系统的大量筛选,并能获得10^6转化子/μg质粒以上的转化率,但如果没有进口的PEG3350而采用分子量接近的国产PEG4000进行转化的话,转化率仅为10^4,远低于文献报道的水平。我们报道了一个改进的PEG/LiAc方法,采用国产PEG6000,转化率也能达到10^6,且能将的热激时间从20min缩短为2min。  相似文献

非组培转化法获得转基因植株及其Basta耐受性研究   总被引:2,自引:0,他引:2  
以玉米‘昌7-2’、大豆‘晋遗19’和高粱‘晋粱5号’的种子为受体,质粒pCAMBAR.CHI.11为供体,采用非组培转化法导入bar基因。PCR扩增和Southern杂交分析结果证明,‘昌7-2’、‘晋遗19’和‘晋粱5号’的划胚种子都获得了转化植株,转化率均在5%以上,未划胚种子的转化率为0。共培养液中加入浓度为200μmol/L的乙酰丁香酮(AS)能使玉米种子转化率比对照提高8倍,赋予300W强度的超声波处理,转化率可再提高60%。划胚处理对种子的出苗率影响较大,为17%左右。划胚种子于实验室条件下的Basta耐受试验结果表明,不同作物对Basta的耐受性存在差异,玉米、大豆和高粱转bar基因植株的Basta耐受临界值分别为0.15%、0.1%和0.05%,Basta处理后的幼苗成活率在各作物间差异不大。田间喷雾Basta溶液结果证明,按Basta耐受性大小排列顺序为高粱<大豆<玉米,但按对Basta敏感性的排序为大豆<高粱<玉米。不同生育期喷雾除草剂的实验结果(T2)证明,在生育期后期作物植株对除草剂的敏感性比前期有所降低,生长前期喷雾后玉米、大豆和高粱的苗存活率分别为57%、49%和52%,生长后期喷雾后苗存活率分别为77%、70%和73%,但各个群体的PCR扩增阳性结果率均在80%以上,证明存活率差异是由植株对除草剂耐受性和敏感性的差异引起的。多代转基因植株的检测结果证明目的基因可稳定遗传。  相似文献

利用GUS报告基因,建立了农杆菌介导的谷子(Setaria italica)遗传转化体系,并研究了多种影响因素对转化效率的影响,包括受体基因型、外植体类型、菌液浓度、培养基中乙酰丁香酮的浓度、侵染时间和共培养时间.确定了最优的转化条件为:以谷子幼穗诱导的愈伤组织为外植体,用低浓度的农杆菌菌液侵染30~40min,然后在含有0.1mmol/L乙酰丁香酮的LS培养基上共培养2d.  相似文献

甘蓝型油菜的各种外植体经遗传转化、组织培养后可以再生为转基因植株,但再生频率会因外植体的基因型、年龄、培养基添加成分和农杆菌共培养的不同而发生变化。转化方法包括农杆菌介导转化、基因枪法、花粉介导法、PEG介导法等,其应用前景非常广阔。甘蓝型油菜的遗传转化在其品质改良、抗逆性提高、雄性不育系的获得和一些特殊性状方面都取得了很大成就。简要介绍甘蓝型油菜的再生体系建立、转化方法及所取得的部分成就。  相似文献
