Studies conducted by the authors and their coworkers on the mechanisms and physiologic significance of radiofrequency hearing effects are reviewed. Results of these studies demonstrate that 1) thermoelastic expansion of fluids and structures within the inner ear is the main mechanism by which auditory stimuli are produced by microwave pulses; 2) the frequency spectra of these stimuli are indistinguishable from the spectra of rectangular pulses with the same durations as the microwave pulses; 3) exposure to continuous-wave (CW) microwave radiation evokes an increase in the metsbolic activities of nuclei in the ascending auditory pathway and also decreases the latency and increases the magnitude of brainstem-evoked responses produced by acoustic clicks; and 4) the mechanism of the effects of CW microwave radiation on the auditory system is intracochlear heating. The significance of these findings is discussed in terras of potential applications of microwave stimuli in basic research on the auditory system and in terms of interpreting the results of past studies that demonstrate behavioral sensitivity to CW microwave fields.  相似文献

The representation of alternative conspecific acoustic signals in the responses of a pair of local interneurons of the bushcricket Tettigonia viridissima was studied with variation in intensity and the direction of sound signals. The results suggest that the auditory world of the bushcricket is rather sharply divided into two azimuthal hemispheres, with signals arriving from any direction within one hemisphere being predominantly represented in the discharge of neurons of this side of the auditory pathway. In addition, each pathway also selects for the most intense of several alternative sounds. A low-intensity signal at 45 dB sound pressure level is quite effective when presented alone, but completely suppressed when given simultaneously with another signal at 60 dB sound pressure level. In a series of intracellular experiments the synaptic nature of the intensity-dependent suppression of competitive signals was investigated in a number of interneurons. The underlying synaptic mechanism is based on a membrane hyperpolarisation with a time-constant in the order of 5–10 s. The significance of this mechanism for hearing in choruses, and for the evolution of acoustic signals and signalling behaviour is discussed. Accepted: 20 November 1999  相似文献

A force-conveying role of the lipid membrane across various mechanoreceptors is now an accepted hypothesis. However, such a mechanism is still not fully understood for mechanotransduction in the hair bundle of auditory sensory hair cells. A major goal of this theoretical assessment was to investigate the role of the lipid membrane in auditory mechanotransduction, especially in generating nonlinear bundle force versus displacement measurements, one of the main features of auditory mechanotransduction. To this end, a hair bundle model that generates lipid membrane tented deformation in the stereocilia was developed. A computational analysis of the model not only reproduced nonlinear bundle force measurements but also generated membrane energy that is potentially sufficient to activate the mechanosensitive ion channel of the hair cell. In addition, the model provides biophysical insight into 1) the likelihood that the channel must be linked in some way to the tip link; 2) how the interplay of the bending and stretching of the lipid bilayer may be responsible for the nonlinear force versus displacement response; 3) how measurements of negative stiffness may be a function of the rotational stiffness of the rootlets; and 4) how the standing tension of the tip link is required to interpret migration of the nonlinear force versus displacement and activation curves. These are all features of hair cell mechanotransduction, but the underlying biophysical mechanism has proved elusive for the last three decades.  相似文献

Mouse myeloma cells were electropermeabilized by single square-wave electric pulses with amplitudes of up to ∼150 kV/cm and durations of 10–100 nsec. The effects of the field intensity, pulse duration and medium conductivity on cell viability and field-induced uptake of molecules were analyzed by quantitative flow cytometry using the membrane-impermeable fluorescent dye propidium iodide as indicator molecule. Despite the extremely large field strengths, the majority of cells survived the exposure to ultra-short field pulses. The electrically induced dye uptake increased markedly with decreasing conductivity of the suspending medium. We assigned this phenomenon to the transient electrodeformation (stretching) force that assumes its maximum value if cells are suspended in low-conductivity media, i.e., if the external conductivity σe is smaller than that of the cytosol σi. The stretching force vanishes when σe is equal to or larger than σi. Due to their capability of delivering extremely large electric fields, the pulse power systems used here appear to be a promising tool for the electropermeabilization of very small cells and vesicles (including intracellular organelles, liposomes, etc.). The disruption force of specific biotin-streptavidin bonds was determined using DNA oligomers as force tags. Forces were generated by an electric field acting on a biotinylated fluorescently labeled DNA oligomer. DNA oligomers were immobilized via biotin-streptavidin bonds on the walls of microfluidic channels. Channel layout and fluid-based deposition process were designed to enable well-defined localized deposition of the oligomers in a narrow gap of the microchannel. Electric fields of up to 400 V/cm were applied and electric field induced desorption of DNA oligomers was observed. At T approximately 30 degrees C, field-induced desorption of both a 12 mer as well as a 48 mer yielded a streptavidin-biotin disruption force of 75 fN. Streptavidin-functionalized surfaces remained intact and could be reloaded with biotinylated oligomers. At approximately 20 degrees C, however, no field-induced unbinding of the oligomers was observed at electric field strength of up to 400 V/cm, indicating a significant temperature dependence of the bond strength.  相似文献

The topics discussed include investigation of microwave effects on the spontaneous action potentials and membrane resistance of isolated snail neurons, effects on the permeability of blood brain barriers in rats, the phenomenon and interaction mechanism for the microwave auditory effect (the hearing of microwave pulses by animals and humans), the development of miniature catheter antennas for microwave interstitial hyperthermia treatment of cancer, the application of transcatheter microwave ablation for treatment of cardiac arrhythmias, and the use of noninvasive wireless technology for sensing of human vital signs and blood pressure pulse waves. The ability of auditory evoked potentials to follow sound pulse (click or pip) rate was studied in bottlenosed dolphins. Sound pulses were presented in 20-ms rhythmic trains separated by 80-ms pauses. Rhythmic click or pip trains evoked a quasi-sustained response consisting of a sequence of auditory brainstem responses. This was designated as the rate-following response. Rate following response peak-to-peak amplitude dependence on sound pulse rate was almost flat up to 200 s−1, then displayed a few peaks and valleys superimposed on a low-pass filtering function with a cut-off frequency of 1700 s−1 at a 0.1-amplitude level. Peaks and valleys of the function corresponded to the pattern of the single auditory brain stem response spectrum; the low-pass cut-off frequency was below the auditory brain stem response spectrum bandwidth. Rate-following response frequency composition (magnitudes of the fundamental and harmonics) corresponded to the auditory brain stem response frequency spectrum except for lower fundamental magnitudes at frequencies above 1700 Hz. These regularities were similar for both click and pip trains. The rate-following response to steady-state rhythmic stimulation was similar to the rate-following response evoked by short trains except for a slight amplitude decrease with the rate increase above 10 s−1. The mechanisms of hearing in the fire-bellied toad Bombina orientalis, an “earless” species of amphibian that lacks a standard tympanic middle ear, were studied using laser Doppler vibrometric and neurophysiological techniques. Laser vibrometry demonstrated that the anterolateral body wall overlying the lung is much more responsive to sound than the lateral head surface overlying the inner ear. Covering the lateral body wall with silicone grease dramatically decreased auditory midbrain sensitivity at all frequencies examined, elevating thresholds by 20–25 dB. Filling the lungs with oxygenated saline produced similar decrements in hearing sensitivity, and both manipulations strongly suggest that the lung is the primary route of sound reception in this species. The precise route of transfer of sound energy from the body wall and lungs to the inner ear remains unclear. The lung-based hearing system of “earless” fire-bellied toads may represent the retention of the first auditory mechanism used by early tetrapod vertebrates for detection of airborne sound. Two potential mechanisms in detection of a 60-Hz electric field by albino rats were examined: field-induced movement of the vibrissae and field-induced vibration of the skin. Specifically, the experiment tested field detection in a moving stream of air designed to mask field-induced movement of the skin, fur, and vibrissae. Rats were trained to detect electric fields and were then tested at field intensities from 0–25 kV/m rms. As previously reported, rats demonstrate unmistakable behavioral evidence of field detection at all intensities above 7.5 kV/m. After establishing detection in still air, field detection was re-examined in moving air (average air velocity approximately 2.8–6.8 m/s). The primary result is that the wind produced no change in detection at field intensities above threshold (> 7.5 kV/m). Indeed, at these intensities detection was virtually identical in still and moving air. A secondary finding is that moving air produced statistically significant (P < .05) but apparently contradictory effects on detection when the field intensity was below threshold. On no-field trials the wind lowered scores (i.e., fewer presses on the field-off lever); however, on subthreshold field trials, the wind actually increased detection scores (i.e., more presses on the field-on lever). While this no-field and subthreshold field result is interesting and deserves further study, we place primary emphasis on the finding that, if the field was detectable in still air, it was also detectable in moving air. This result leads us to believe that movement of the vibrissae, fur, or skin is not likely to be the main mechanism of electric-field detection in our subjects. © 1993 Wiley-Liss. Inc.  相似文献

The epitaxial growth of a diamond single-crystal film in a torch microwave discharge excited by a magnetron of a domestic microwave oven with the power of ≤1 kW in an argon-hydrogen-methane mixture with a high concentration of methane (up to 25% with respect to hydrogen) at atmospheric pressure on a sub-strate of a synthetic diamond single crystal (HPHP) with the orientation (100) and 4 × 4 mm in size is obtained. A discharge with the torch diameter of ∼2 mm and the concentration of the microwave power absorbed in the torch volume of >103 W/cm3 is shown to be effective for epitaxial enlargement of a single crystal of synthetic diamond. The structure of the deposited film with the thickness up to 10 μm with high-quality morphology is investigated with an optical microscope as well as using the methods of the Raman scattering and scanning electron microscopy.  相似文献

Absolute thresholds and critical masking ratios were determined behaviorally for the European barn owl (Tyto alba guttata). It shows an excellent sensitivity throughout its hearing range with a minimum threshold of −14.2 dB sound pressure level at 6.3 kHz, which is similar to the sensitivity found in the American barn owl (Tyto alba pratincola) and some other owls. Both the European and the American barn owl have a high upper-frequency limit of hearing exceeding that in other bird species. Critical masking ratios, that can provide an estimate for the frequency selectivity in the barn owl's hearing system, were determined with a noise of about 0 dB spectrum level. They increased from 19.1 dB at 2 kHz to 29.2 dB at 8 kHz at a rate of 5.1 dB per octave. The corresponding critical ratio bandwidths were 81, 218, 562 and 831 Hz for test-tone frequencies of 2, 4, 6.3 and 8 kHz, respectively. These values indicate, contrary to expectations based on the spatial representation of frequencies on the basilar papilla, increasing bandwidths of auditory filters in the region of the barn owl's auditory fovea. This increase, however, correlates with the increase in the bandwidths of tuning curves in the barn owl's auditory fovea. Accepted: 27 November 1997  相似文献

Hepatic lysosomes were exposed in vitro to microwave radiation (2450 MHz) either prior to or simultaneously with treatment with retinol (vitamin A), and the release of the lysosomal enzymes, β-glucuronidase, acid phosphatase, and cathepsin D, determined. A 60-min microwave exposure (10 or 100 mW/g) of retinol-treated lysosomes had no effect on the amount of release of β-glucuroni-dase, cathepsin D, or acid phosphatase. In addition, 10 and 100 mW/g irradiation of lysosome fractions for 40 min prior to a 20-min retinol and microwave treatment, had no influence on the release of these enzymes. Finally, the effect of microwave radiation on the loss of latency of acid phosphatase and β-glucuronidase from retinol-treated lysosomes was determined. Microwave radiation had no influence on the rate of appearance of these enzymes in the suspending medium. The results indicate that microwave radiation had no effect on the retinol-induced lysosomal enzyme release.  相似文献

Whereas the transfer of Listeria from surfaces to foods and vice versa has been well documented, little is known about the mechanism of bacterial transfer. The objective of this work is to gain a better understanding of the forces involved in listerial biofilms adhesion using atomic force microscopy (AFM). L. monocytogenes Scott A was grown as biofilms on stainless steel surfaces by inoculating stainless steel coupons with Listeria and incubating the coupons for 48 h at 32 °C with a diluted 1:20 tryptic soy broth. After growth, biofilms were equilibrated over saturated salt solutions at a constant relative humidity (%RH) before measurement of adhesion forces using AFM. The effects of contact time, loading force, and biofilm relative humidity (%RH) suggested that neither contact time, loading force nor biofilm %RH had a significant effect on biofilm adhesiveness at a cellular level (P > 0.05). In a second set of experiments, the influence of material type on biofilm adhesiveness was evaluated using two different colloidal probes (SiO2 and polyethylene). Results showed that the maximum pull-off force and retraction work needed to retract the cantilever for glass (−85.42 nN and 1.610−15 J, respectively) were significantly lower than those of polyethylene (−113.38 nN and 2.7 × 10–15 J, respectively; P < 0.001). The results of this study suggest that Listeria biofilms adhere more strongly to hydrophobic surfaces than hydrophilic surfaces when measured at a cellular level. These results provide important insights that could lead to new ways to remediate and avoid listerial biofilm formation in the food industry.  相似文献

The aim of this study is to explain the mechanism of the effect of low-level modulated microwave radiation on brain bioelectrical oscillations. The proposed model of excitation by low-level microwave radiation bases on the influence of water polarization on hydrogen bonding forces between water molecules, caused by this the enhancement of diffusion and consequences on neurotransmitters transit time and neuron resting potential. Modulated microwave radiation causes periodic alteration of the neurophysiologic parameters and parametric excitation of brain bioelectric oscillations. The experiments to detect logical outcome of the mechanism on physiological level were carried out on 15 human volunteers. The 450-MHz microwave radiation modulated at 7, 40 and 1000 Hz frequencies was applied at the field power density of 0.16 mW/cm2. A relative change in the EEG power with and without radiation during 10 cycles was used as a quantitative measure. Experimental data demonstrated that modulated at 40 Hz microwave radiation enhanced EEG power in EEG alpha and beta frequency bands. No significant alterations were detected at 7 and 1000 Hz modulation frequencies. These results are in good agreement with the theory of parametric excitation of the brain bioelectric oscillations caused by the periodic alteration of neurophysiologic parameters and support the proposed mechanism. The proposed theoretical framework has been shown to predict the results of experimental study. The suggested mechanism, free of the restrictions related to field strength or time constant, is the first one providing explanation of low-level microwave radiation effects.  相似文献

Thresholds for evoked vocal responses and thresholds of multiunit midbrain auditory responses to pure tones and synthetic calls were investigated in males of Pleurodema thaul, as behavioral thresholds well above auditory sensitivity have been reported for other anurans. Thresholds for evoked vocal responses to synthetic advertisement calls played back at increasing intensity averaged 43 dB RMS SPL (range 31–52 dB RMS SPL), measured at the subjects’ position. Number of pulses increased with stimulus intensities, reaching a plateau at about 18–39 dB above threshold and decreased at higher intensities. Latency to call followed inverse trends relative to number of pulses. Neural audiograms yielded an average best threshold in the high frequency range of 46.6 dB RMS SPL (range 41–51 dB RMS SPL) and a center frequency of 1.9 kHz (range 1.7–2.6 kHz). Auditory thresholds for a synthetic call having a carrier frequency of 2.1 kHz averaged 44 dB RMS SPL (range 39–47 dB RMS SPL). The similarity between thresholds for advertisement calling and auditory thresholds for the advertisement call indicates that male P. thaul use the full extent of their auditory sensitivity in acoustic interactions, likely an evolutionary adaptation allowing chorusing activity in low-density aggregations.  相似文献

A biological membrane undergoes a reversible permeability increase through structural changes in the lipid domain when exposed to high external electric fields. The present study shows the occurrence of electric field-induced changes in the conductance of the proton channel of the H(+)-ATPase as well as electric field-induced structural changes in the lipid-protein domain of photosystem (PS) II in the photosynthetic membrane. The study was carried out by analyzing the electric field-stimulated delayed luminescence (EPL), which originates from charge recombination in the protein complexes of PS I and II of photosynthetic vesicles. We established that a small fraction of the total electric field-induced conductance change was abolished by N,N'-dicyclohexylcarbodiimide (DCCD), an inhibitor of the H(+)-ATPase. This reversible electric field-induced conductance change has characteristics of a small channel and possesses a lifetime < or = 1 ms. To detect electric field-induced changes in the lipid-protein domains of PS II, we examined the effects of phospholipase A2 (PLA2) on EPL. Higher values of EPL were observed from vesicles that were exposed in the presence of PLA2 to an electroporating electric field than to a nonelectroporating electric field. The effect of the electroporating field was a long-lived one, lasting for a period > or = 2 min. This effect was attributed to long-lived electric field-induced structural changes in the lipid-protein domains of PS II.  相似文献

We report electric-dichroism and electron-microscopic studies of chromatin fibers fixed by protein–protein crosslinking at salt concentrations ranging from 10 to 100 mM. The results confirm a progressive disorganization of the fiber as the salt concentration is lowered. The positive dichroism and large polarizability anisotropy characteristic of the 300-Å diameter fiber found in 100 mM salt are replaced by negative dichroism and smaller effective polarizability anisotropy or dipole moment for samples fixed at lower salt concentration. We interpret the results in terms of segmental, field-induced orientation of the disorganized structure which is present in low salt concentrations. We also observed a field-induced absorbance decrease in chromatin fibers fixed at salt concentration at and below 100 mM. All three optical effects, namely overall orientation of the high-salt fixed fiber, segmental orientation of the low-salt fixed fiber, and field-induced absorbance decrease, occur on roughly the same time scale, 20–100 μs for 50 nucleosome polynucleosomes. The polarizability anisotropy of fibers fixed in 100 mM salt was found to be proportional to the length of the fragment and to the reciprocal square root of the conductivity of the solution used for electric-dichroism measurements. Addition of Mg2+ to the measurement buffer affected the dichroism amplitude of samples fixed below 100 mM salt but not those fixed at 100 mM salt. The results reinforce the need for caution in interpreting electric-dichroism measurements on chromatin fibers because of possible field-induced distortion effects.  相似文献

Little is known on antimicrobial peptide permeation through outer membrane channels in Gram-negative bacteria. Herein, we probed at a single-molecule level the interaction of two different peptides, magainin 2 and HPA3P with OmpF from E. coli. HPA3P is an analogue of the antimicrobial peptide HP(2–20) isolated from the N-terminal region of the Helicobacter pylori ribosomal protein. Our data show that the shorter and more charged HPA3P peptide is more accessible to the inner volume of the OmpF than magainin 2. We demonstrate the ability of HPA3P peptides to interact with OmpF in a voltage- and concentration-dependent manner, which does not rule out a novel mechanism by which such peptides could reach the periplasmic space of Gram-negative bacteria. Unexpectedly, we found that increasing the applied voltage led to an increase of the residence time of HPA3P peptide inside the pore, possibly reflecting electric field-induced changes in pore and peptide geometry.  相似文献

Oriented purple-membrane preparations from Halobacterium halobium were obtained by electrophoretic sedimentation of a purple-membrane suspension on a transparent current-conducting surface. Light exposure of orderly oriented purple-membrane films causes the generation of a photopotential amounting to several volts. The effects of external electric field on the dark state and photocycle of bacteriorhodopsin is studied in dry orderly oriented purple-membrane films. In contrast to nonuniformly oriented preparations (Borisevich, G.P., Lakashev, E.P., Kononenko, A.A. and Rubin, A.B. (1979) Biochim. Biophys. Acta 546, 171–174 and Lukashev, E.P., Vozary, E., Kononenko, A.A. and Rubin, A.B. (1980) Biochim. Biophys. Acta 590, 258–266), a specific feature of the field-induced phenomena observed in orderly oriented films is their vectorial character. The field-induced bathochromic shift of the maximum absorbance of bacteriorhodopsin is observed in an electric field, directed from the periplasmatic to cytoplasmatic side of the purple membrane and the field-induced rise of the photo-stationary M412 concentration in a field of opposite sign. This field-induced rise is a result of slowering of M412 decay. The observed effects seem likely to reflect the existence of the potential-dependent regulation of the bacteriorhodopsin photocycle in intact purple membranes.  相似文献
