用高灵敏度的单光子计数系统探测人工老化 (40℃ ,相对湿度 90 % ) 0、7、14和 2 1d的水稻种子吸胀初期的超弱化学发光强度变化 ,首次观测到水稻 (OryzasativaL .)种子吸胀初期超弱化学发光强度与其老化程度有关。加水激活水稻种子的超弱化学发光 ,其强度依赖于种子的老化程度。实验结果表明水稻种子的人工老化时间越短 ,则吸胀初期 (2 0~ 30min)超弱化学发光的强度越强 ,萌发率越高。水稻种子萌发率与超弱化学发光强度呈显著正相关。观测水稻种子吸胀初期超弱化学发光强度的变化可望成为检测种子老化程度的一种快速、简便、无损伤的新方法。  相似文献

紫外A(UVA,320 nm-400 nm)诱发的脂质过氧化反应是通过活性氧(ROS)介导的。在UVA照射之后,单线态氧(1O2)和超氧阴离子(O2-.)是细胞内最初产生的ROS,它们进一步生成过氧化氢(H2O2),羟自由基(.OH)等其它自由基。为了探讨UVA照射后最早生成的1O2和O2-.与细胞氧化损伤后果的关系,我们采用一种特异性检测1O2和O2-.的高灵敏度化学发光探针MCLA(2-methyl-6-(4-methoxyphenyl)-3,7-dihydroimid-azo[1,2-α]pyrazin-3-one hydrochloride)检测人外周血淋巴细胞经UVA照射后的化学发光变化。发现不同剂量UVA照射后,细胞MCLA化学发光变化和MDA浓度变化一致。结果表明UVA照射后1O2和O2-.的水平与由此引发的脂质过氧化损伤存在正相关关系。因此,MCLA化学发光方法可望作为一种检测UVA诱发脂质过氧化水平的简单快速方法。  相似文献

以一种海萤荧光素类似物MCLA〔2 methyl 6 (p methoxyphenyl) 3,7 dihydroimidazo [1,2 a]pyrazin 3 one〕作为高灵敏且有选择性的化学发光探针 ,用化学发光的方法直接观测到了少量Cu2 氧化的低密度脂蛋白 (Ox LDL)中维生素C诱导的单线态氧 (1O2 )的产生。实验中通过叠氮化钠 (NaN3 )对MCLA介导的化学发光的猝灭作用进一步证实了上述体系中1O2的形成。根据实验观察的结果 ,分析了这一体系中1O2 形成的可能途径 ,认为首先是维生素C将Cu2 转变为还原态 ,而自身失去一个电子转变为维生素C自由基 ,从而刺激了过氧自由基和烷氧自由基的形成 ,过氧自由基的双分子反应很可能就是体系内1O2 产生的反应机制  相似文献

Using a highly sensitive single photon counter, a spontaneous chemiluminescence (CL) study on rice (Oryza sativa L.) seeds stored in different years was carried out. We first observed that the degree of ageing in rice seeds was related to the intensity of spontaneous CL during early imbibition (0–30 min). Rice seeds stored for a shorter time had a stronger intensity of CL in early imbibition. The germination rate of rice seeds showed an obvious positive correlation with the intensity of spontaneous CL. Singlet oxygen (1O2) in rice seeds during early imbibition was investigated by a CL method using a cypridina luciferin analogue, 2‐methyl‐6‐(p‐methoxyphenyl)‐3,7‐dihydroimidazo [1,2α] pyrazin‐3‐one (MCLA), as a selective CL probe. Additional experimental evidence for the formation of 1O2 came from the quenching effect of sodium azide (NaN3) on MCLA‐mediated rice seeds' CL. Analysis based on the experimental results demonstrated that spontaneous CL in rice seeds during early imbibition was mainly contributed by singlet oxygen (1O2). 本实验对水稻籽粒灌浆过程中不同时期籽粒进行了超弱发光测定。实验结果表明:在整个灌浆过程中单粒发光量呈单峰曲线,在籽粒全部充实并且颜色由青转黄时,发光量最大;发光强度呈递减趋势;运用该技术有可能进行水稻不同品种间的比较和品种灌浆力强弱的筛选。  相似文献

The 3‐aminophthalic acid anion is a light emitter in luminol chemiluminescence. In the present study, the chemiluminescence of the 3‐aminophthalic acid anion itself in the presence of hydrogen peroxide–cobalt (II) was studied. The results indicated that 3‐aminophthalic acid anion is highly chemiluminescent in the typical hydrogen peroxide–cobalt (II) system. The peak wavelength of this chemiluminescence and the kinetic profile of the 3‐aminophthalic acid anion–hydrogen peroxide–cobalt (II) reaction showed similarity with that of luminol, but the chemiluminescence of 3‐aminophthalic acid anion had a much lower background signal. In addition, the chemiluminescence mechanism of 3‐aminophthalic acid anion–hydrogen peroxide–cobalt (II) was also discussed and speculated as the interaction between 3‐aminophthalic acid anion and singlet oxygen.  相似文献

A weak chemiluminescence (CL) emission was observed due to the production of singlet oxygen ((1)O(2)) during the decomposition of peroxomonosulphate (HSO(5)(-)) catalysed by cobalt(II). Low molecular mass aliphatic monocarboxylic acids, such as formic, acetic, propionic, butyric and valeric acids, influenced the CL emission, and the reaction of aliphatic monocarboxylic acids with HSO(5)(-)/Co(2+) solution was further investigated using a flow injection analysis (FIA) CL method. The results indicated that the CL intensities of aliphatic monocarboxylic acids were improved with increase in the carbon chain length in the potassium peroxomonosulphate-cobalt(II) sulphate system. Generation of singlet oxygen was confirmed by the fact that the CL emission of aliphatic monocarboxylic acids with the HSO(5)(-)/Co(2+) solution was quenched by NaN(3), and from the CL spectrum of the reaction system. Additionally, a possible mechanism of aliphatic monocarboxylic acids CL emission enhancement was proposed.  相似文献

Electron microscope observation revealed that mitochondria in plumular root cells of soy bean seed were invisible at the beginning of seed germination. They were sequentially formed re-differentiated and developed from proplastids during the process of germination. The proplastids in the control plumular root markedly expanded at low temperature imbibition process and did not develop into mitochondria when normal temperature was restored. Even the structure of reticulata membrane was further destroyed and vacuolized. In contrast, mitochondria of soybean seed pretreated with polyvinyl alcohol (PVA) and polythylene glycol-6000 (PEG) continused to develop well under the same conditions, as characterized by a clear structure of bilayered membrane and tube cristae. The results suggest that continuous re-differentiation and development of proplastids into mitochondria at low temperature imbibition process play an important role in the increase of soybean seed vigour and its resistance to chilling injury.  相似文献

The feasibility of a luminol-dependent chemiluminescence analysis of hydrogen peroxide production by intact wheat seedlings using a KhL-003 chemiluminometer was determined. It was shown that the minimal H2O2 concentration that can be detected in a 0.5-ml sample with this instrument is 0.125 µM. Analysis of biological activity of a mixture of chitooligosaccharides with molecular masses from 5 to 10 kD and acetylation degree of 65% demonstrated that, at a concentration of 1 µg/ml, they induce rapid overproduction of H2O2 in roots of 3-day-old wheat seedlings.  相似文献

A polymorphism in rice amylases at an early stage of seed germination is analyzed by zymogram. In nonglutinous cultivars of rice, α-amylase isozymes are mainly confirmed in germinating seeds. However, in glutinous cultivars, β-amylase isozymes, which are not confirmed in nonglutinous cultivars, make up the major part of the total amylase activity and the expression of α-amylases are repressed.  相似文献
