The contamination of cow’s milk at the farm level with aflatoxin M1 was investigated in South Africa. Samples of feeds, forage, maize and milk were taken at nine dairy farms, and at the same time samples of the processed milk (retail milk) were collected from the respective dairies to which the farms delivered their milk. The feeds were analysed for aflatoxin B1 and the milk samples for aflatoxin M1 using high performance liquid chromatography (HPLC) and fluorescence detection. All milk samples from the dairy farms were positive for aflatoxin M1, ranging from 0.02 μg/l to 1.5 μg/l. Retail milk was also frequently contaminated with AFM1, at levels of 0.01-3.1 μg/l. High AFB1 levels in feed materials on the farms supplying the raw milk indicate that various sources account for this contamination frequency in milk.  相似文献

Aims: The microbial quality of farm bulk‐tank raw milk produced in Estonia during years 2004–2007 was investigated. Methods and Results: Bulk‐tank milk samples were analysed for lactic acid bacteria count (LABC), psychrotrophic bacteria count (PBC), aerobic spore‐forming bacteria count (ASFBC), total bacterial counts using BactoScan and somatic cell count (SCC) using Fossomatic. Randomly selected psychrotrophic isolates were subjected to 16S–23S PCR‐ribotyping. LABC remained below 104 CFU ml?1 in most samples, while psychrotrophic micro‐organisms dominated in 60% of farms. PBC ranged from 4·2 × 102 to 6·4 × 104 CFU ml?1, and ASFBC varied from 5 to 836 CFU ml?1. Conclusions: In general, the microbiological quality of the farm bulk‐tank milk was good – more than 91% of samples contained <50 000 CFU ml?1, and SCC in the majority of samples did not exceed the internationally recommended limits. Genus Pseudomonas spp. was the dominating spoilage flora with Pseudomonas fluorescens as the prevailing species. Significance and Impact of the Study: Specific bacterial groups (LABC, PBC and ASFBC), not analysed routinely by dairies, were determined in bulk‐tank raw milk of numerous dairy farms during 4‐year period. Based on the survey, dairy plants can better control their supply chains and select farms (milk) for the production of specific products, i.e. milk with low PBC and high LABC for cheesemaking.  相似文献

Purpose

The two main functions of dairy farming are to produce raw milk and to generate an economic income for the farmer (Powell et al. in Nutr Cycl Agroecosyst 82:107–115, 2008), both of which drive the downstream value chain. Farm profit is mainly determined by the quantity and quality of milk, and at the same time, dairy farmers have a responsibility for animal welfare and the protection of human health through milk hygiene. When dairy farmers supply milk to a processor, the payment is based on both the quantity and the quality in terms of composition (fat and protein) and hygiene (total bacterial count and somatic cell count). Somatic cell count reflects the health status of the mammary gland and contributes to reduced shelf life and reduced cheese yield and quality, whilst total bacterial count reflects herd health and farm sanitation. The objective of this work was to create a new functional unit for raw milk at the farm gate/ processor gate, which could be used to better capture the economic function of milk, while still reflecting the quality criteria captured by energy corrected milk and fat and protein corrected milk.

Materials and methods

Base price-adjusted milk (BPAM) is proposed as a functional unit to capture more of the functions of the transaction between the farmer and the processor. It expresses the volume of milk delivered as the equivalent volume at base price, such that greater milk solids increase the volume and poor hygiene decreases the volume. The BPAM was compared with energy-corrected milk and was tested using a survey of 54 farms in Ireland and eight scenarios compared with the observed values and an “ideal” scenario with no hygiene penalty imposed.

Results and discussion

It was found that kg BPAM for the sample of farms was strongly correlated with kg ECM because of the overall hygiene standard from the sample of farms available. For specific farms at specific times, the hygiene properties contributed to the economic function of the milk in a way that could not be captured using energy-corrected milk. The scenario analysis indicated that high levels of biological contamination, if captured in the raw milk functional unit, could increase carbon footprint by >?200%.

Conclusions

It was concluded that BPAM may not be a necessary functional unit for all studies, but those that focused on the farmer and processor perspectives should consider using BPAM because it captures more of the obligatory properties of raw milk than just quality expressed in terms of milk solids.

Purpose

Life cycle assessment (LCA) studies of carbon footprint (CF) of milk from grass-based farms are usually limited to small numbers of farms (<30) and rarely certified to international standards, e.g. British Standards Institute publicly available specification 2050 (PAS 2050). The goals of this study were to quantify CF of milk from a large sample of grass-based farms using an accredited PAS 2050 method and to assess the relationships between farm characteristics and CF of milk.

Materials and methods

Data was collected annually using on-farm surveys, milk processor records and national livestock databases for 171 grass-based Irish dairy farms with information successfully obtained electronically from 124 farms and fed into a cradle to farm-gate LCA model. Greenhouse gas (GHG) emissions were estimated with the LCA model in CO2 equivalents (CO2-eq) and allocated economically between dairy farm products, except exported crops. Carbon footprint of milk was estimated by expressing GHG emissions attributed to milk per kilogram of fat and protein-corrected milk (FPCM). The Carbon Trust tested the LCA model for non-conformities with PAS 2050. PAS 2050 certification was achieved when non-conformities were fixed or where the effect of all unresolved non-conformities on CF of milk was?<?±5 %.

Results and discussion

The combined effect of LCA model non-conformities with PAS 2050 on CF of milk was <1 %. Consequently, PAS 2050 accreditation was granted. The mean certified CF of milk from grass-based farms was 1.11 kg of CO2-eq/kg of FPCM, but varied from 0.87 to 1.72 kg of CO2-eq/kg of FPCM. Although some farm attributes had stronger relationships with CF of milk than the others, no attribute accounted for the majority of variation between farms. However, CF of milk could be reasonably predicted using N efficiency, the length of the grazing season, milk yield/cow and annual replacement rate (R 2?=?0.75). Management changes can be applied simultaneously to improve each of these traits. Thus, grass-based farmers can potentially significantly reduce CF of milk.

Conclusions

The certification of an LCA model to PAS 2050 standards for grass-based dairy farms provides a verifiable approach to quantify CF of milk at a farm or national level. The application of the certified model highlighted a wide range between the CF of milk of commercial farms. However, differences between farms’ CF of milk were explained by variation in various aspects of farm performance. This implies that improving farm efficiency can mitigate CF of milk.  相似文献

When a Euglena, in a medium containing ATP, is microinjected with 7 × 10−14 l of 0.02 M EDTA, which binds Ca2+ and Mg2+, flagellar motility stops. Flagellar arrest in Chlamydomonas occurs with the injection of 2 × 10−14 l of 0.02 M EDTA. The injection of similar amounts (7 × 10−14 l in Euglena and 3 × 10−14 l in Chlamydomonas) of 0.02 M EGTA, which preferentially binds Ca2+, did not significantly alter flagellar motility. This suggests that a decrease in the internal Ca2+ concentration in Euglena or Chlamydomonas did not stimulate flagellar beating. Further, flagellar motility decreased when internal Mg2+ was chelated. The microinjection of Zn2+ into these cells caused a decrease in flagellar frequency analogous to the decrease in frequency caused by the injection of Ca2+ and EDTA. The microinjection of 7 × 10−14 l of 0.2 M Mn2+ caused an approx. 1.5-fold increase in Euglena flagellar motility. Chlamydomonas flagella, which cease to beat upon impalement in an Mg2+-free medium, resume a flagellar frequency of 18 Hz when injected with 3 × 10−14 l of 0.2 M Mn2+. In the experiments reported here, Mn2+ acts as an analog of Mg2+.  相似文献

Laws of different countries regarding SCC of goat milk are not in agreement with each other and sometimes they fix a threshold for the enhancement of dairy products. The aim of this study was to assess if renneting properties of goat milk are influenced by higher somatic cell count (SCC) measured by an electronic cell counter. Milk samples, taken throughout the lactation of 169 goats from three farms, were analyzed for chemical, physical, hygienic and renneting properties. Samples were divided into three levels on the basis of their SCC: L: low level, samples with SCC lower than 106 cells/ml; M: medium, between 106 and 2 × 106 cells/ml; H: high, higher than 2 × 106 cells/ml. Milk clotting time was between 12.07 and 13.31 min, curd firming time between 1.68 and 2.05 min and curd firmness between 41.66 and 48.97 mm. All the three renneting properties were not affected by the SCC level but they were highly correlated with other factors as protein content and pH. Furthermore, the microbial count showed a high positive correlation with SCC. These results showed that in goat milk, contrarily to other dairy species, higher SCC did not affect renneting properties and that counting of somatic cells by using electronic cell counters might be not suitable for the improvement of dairy products.  相似文献

The social behaviour of Polish Black-and-White dairy cattle (BW) was compared with that of the F1-crossbreds of BW with Jersey (BW × J), Ayrshire (BW × Ay), Holstein-Friesian (BW × HF) and Swedish Black-and-White (BW × SLB). The ethological observations were made under conditions of loose housing.The crossbreds BW × J proved to be most aggressive, excitable and susceptible, as far as milk yield was concerned, to the social tensions connected with moving cows between groups. The BW × Ay were most dominant, least attacked by their group-mates and least susceptible to a group change. The pure-bred BW were calm and subordinate as compared with the crossbreds.The social tensions after a group change caused a mean decrease in milk yield of about 4%. There was a low correlation between the parameters of social behaviour and physical or production traits. The dominance relationships based on spontaneous aggressive interactions differed slightly from those based on interactions motivated by feeding.  相似文献

Sheep herds are a significant capital of dairy industry in Greece, but as dairy farming becomes more complex and intense the need to provide assistance to dairy producers on milk quality is critical. In this study, the quality of raw milk produced in sheep farms of NE Greece was surveyed and the role of various factors was explored. In total, 21 dairy ewe’s farms from the regions of Xanthi and Evros, in the north-eastern Greece were monitored. Milk samples were sampled after the morning milking every 15 days throughout the dairy period (March–June). For the study, a questionnaire was also filled by personal interview with the owners in order to collect information about herd characteristics, health status, handling practices etc. From each farm, air was also sampled for microbiological analysis. Milk samples were examined for chemical components: fat content, protein, lactose, non-fat dry matter (NFDM) and somatic cells count (SCC). Microbiological examination involved the estimation of Total Bacterial Counts (TBC), coliform count (CC), Staphylococcus aureus, Streptococcus sp., and preliminary incubation count (PIC). The possible correlation among different bacterial species and their interaction with SCC and chemical components of milk was also considered. It was examined whether farm management practices could influence the hygiene and the quality of milk. Our results show that as an average TBCs were 5.48 log cfu/ml, SCC: 6.05 log cells/ml, CC: 4.49 log cfu/ml, S. aureus: 3.94 log cfu/ml, Streptococcus sp.: 4.95 log cfu/ml and PIC: 5.7 log cfu/ml. The mean fat, protein, lactose and NFDM were 6.17%, 5.28%, 4.73% and 10.95% respectively. The study revealed significant positive correlation between TBC and PIC (0.825), while SCC was marginally positive correlated with protein and NFDM. No statistically significant correlations observed among SCC with any of the bacterial species. Herd size and farm management practices had considerable influence on SCC and bacterial species.  相似文献

【目的】本试验测定了两个奶牛场健康乳汁和乳房炎乳汁中微生物菌群的变化,以揭示不同奶牛场之乳汁菌群的异同,评估其对乳汁代谢的影响是否相同。【方法】采用16S rRNA高通量测序技术,分别测定两个奶牛场6头健康奶牛和6头乳房炎奶牛乳汁中微生物16S rRNA V4区序列,并对菌群群落结构和多样性进行比较,分析场内及场间的乳汁菌群差异。【结果】四组乳汁样本共获得4013234条原始序列,经过滤后获得2887024条优化序列。Alpha多样性Chao指数、Ace指数、Shannon指数、Simpson指数差异均不显著(P0.05);Beta多样性四组样本均分别聚类;在场1和场2中,引起奶牛乳房炎的优势菌属分别是克雷伯氏菌属和埃希氏菌属;在2个奶牛场的健康乳汁中,场2的埃希氏菌属、葡萄球菌属的丰度显著高于场1;在2个奶牛场的乳房炎乳汁中,场2的埃希氏菌属、乳球菌属的丰度显著高于场1;2个奶牛场健康乳汁中的嗜冷菌总丰度分别为31.87%和38.72%;关联分析及功能预测分析表明,2个奶牛场健康乳汁与乳房炎乳汁优势物种之间的关系差异较大;场1无论是Level 1还是Level 2水平,均发现显著性差异的代谢通路,而场2均未发现显著性差异的代谢通路。【结论】本试验研究了两个奶牛场健康乳汁和乳房炎乳汁微生物菌群之间的异同,为两个奶牛场在乳房炎的预防工作以及原料奶在冷链运输过程中质量控制提供理论依据。  相似文献

The use of compost bedded pack systems (CBS) has increased over the past 5 years in tropical countries, and studies associating production traits with economical outcomes of this system are warranted. Our objectives were to evaluate productive traits, economic outcomes and the risks of losses of dairy farms that switched from a drylot system (DLS) to a CBS and to compare these with similar farms that did not change their system. We collected data from 18 farms over 36 consecutive months (August 2014 to July 2017). All farms started the study as DLS, and six farms switched to CBS in the second year. The other 12 farms kept their DLS during the 36 months of evaluation. Annual technical and economic indexes per farm were collected and calculated. Additionally, a risk analysis was performed based on a 10-year historical series of milk prices. The results were analysed using a regression model including year and herd as categorical variables (fixed), system and herd size as quantitative variables (fixed), and system × herd as a random variable. Furthermore, a non-metric multidimensional scaling plot was used to evaluate producers’ profiles in each year. Milk fat, milk total solids, and somatic cell count did not change when farms switched from DLS to CBS, and averaged 3.80%, 12.04%, and 256 500 cells/ml, respectively. However, milk protein (%) decreased in CBS farms. The majority of milk production variables were not affected. Nevertheless, farms that switched to CBS increased milk production per cow by 13.3% compared with DLS farms. Total operation costs (296 076.83 $/year) were not affected by the system, and neither were the costs of concentrates, roughage, labour or medicines. Net margin per litre (0.09 $/l), operating profit (14.95%), assets per litre (398.68 $/l per day) and return on assets (10.27%) did not change when farms switched from DLS to CBS. Net margin ($/l and $/cow) and asset turnover rate increased in CBS farms. Risk analysis indicated that the risk was reduced by 38% in CBS farms. Furthermore, our analysis showed that producers who switched to CBS had similar technical and economic indexes in the first year before switching their system. In conclusion, this study demonstrates that CBS systems might be promising for producers in tropical countries who are looking for a more productive and less risky system. We did not observe improvements in animal health as previously reported in the literature.  相似文献

Water scarcity prevailing in the drylands is threatening the sustainability of livestock production systems. The water footprint (WF) indicator was proposed as a metric of water use. This study aimed to determine the WF and the economic water productivity (EWP) of 1 kg of fat and protein-corrected milk (FPCM) in eight dairy farms (n = 8; animals = 117 ± 62; area = 198 ± 127; 95% confidence level) in northern Tunisia. Then, to assess the effects of three simulation scenarios targeting the reduction of the WF of milk production (scenario A: using triticale silage to replace, on DM basis, the silage of maize, sorghum or ray-grass; scenario B: reducing by 56% the wastage of water devoted to milking, cooling, cleaning and servicing; scenario C: using concentrate feeds imported from Brazil and Argentina instead of that imported from France). A year-round monitoring of on-farm practices was performed using water-meters and recording equipment installed in key locations in the target dairy farms: (i) water used for feed production, (ii) cow watering, (iii) servicing water, (v) crop and forage production and (iv) economic and production performance were controlled by water source (green and blue). Over the eight farms evaluated, milk production consumed on average 1.36 ± 0.41 m3/kg FPCM, of which 0.93 ± 0.40 m3/kg FPCM was green water and 0.42 ± 0.30 m3/kg FPCM was blue water. However, virtual water of 1 kg FPCM averaged 43% ± 14.3%. Water used for feed production for lactating cows represents approximately 87% ± 6% of the total WF of milk production. However, drinking and servicing water contributed by 3.75% ± 2% and 9% ± 5% to the total WF of milk, respectively. The EWP assessment revealed that the selected dairy farms had a relatively small gross margin per m3 of water averaging US$ 0.05 ± 0.04. The variation in WF of milk was mainly associated with diets’ ingredients, which affected milk productivity and water consumption. Scenario analysis indicated that using feed with less water requirements or importing feeds from countries where its water consumption is low could reduce consumptive water use for milk production by up to 16%. The efficient use of servicing water could reduce blue WF of milk by up to 4%. The implementation of these measures would lead to potential total water savings in the Tunisian dairy sector of 646 million m3 per year (30%).  相似文献

Growth rates in pre-weaned calves influence their health, age at first calving and lifetime productivity. Many farms restrict milk rations to encourage solid feed intake and facilitate early weaning, but this can compromise growth. This study determined the milk feeding policies and associated growth rates on 11 commercial dairy farms in South East England, each following their normal management regime. Between 26 and 54 heifers were recruited per farm, providing a final cohort of 492, of which 71% were pure Holstein. Information on calf rearing practices (feeding, weaning, housing) and health was collected via questionnaires and weekly observations. Estimates of actual milk fed (kg solids) between 1 and 63 days were calculated for individual calves. Morphometric data (weight, height, length) were taken at weeks 1, 5 and 9 and at a median age of 7.5 months and growth rates were calculated. Most calves were fed milk replacer via automated feeders (four farms), teat feeder (one) or buckets (four) whereas two farms provided drums of acidified waste milk. Farms fed between 4 and 6 l/day of milk at mixing rates of 10% to 15%, providing 400 to 900 g/day of milk solids. Both skeletal growth rates and average daily weight gain (ADG) increased in the second month of life compared with the first: height growth from 0.17±0.14 to 0.25±0.16 cm/day and ADG from 0.48±0.25 to 0.71±0.28 kg/day. Post-weaning heifers up to 7.5 months had height increases of 0.16±0.035 cm/day and ADG of 0.83±0.16 kg/day. From 1 to 63 days 70% of calves had growth rates <0.7 kg/day and of these 19.6% gained <0.5 kg/day. Mean ADG before 9 weeks varied between farms from 0.52±0.30 to 0.75±0.20 kg/day. This was related to the amount of milk fed at both a farm and individual calf level. Increasing the total milk solids fed between 1 and 63 days from 20.4 to 46.3 kg (the 10th to 90th percentile observed) was associated with an increase of 0.11 kg/day ADG. All farms had a wide variation in growth rates despite single feeding policies. Higher circulating immunoglobulin G and IGF1 concentrations were associated with better growth, whereas low temperatures in month of birth, high scores for diarrhoea, respiratory and umbilical disease and large birth size reduced growth. Many commercially grown dairy heifers therefore experienced growth restriction in the pre-weaned period, potentially reducing their health, welfare and productivity.  相似文献

The objective of this study was to evaluate the nutritional and ecological aspects of feeding systems practiced under semi-arid environments in Jordan. Nine dairy farms representing the different dairy farming systems were selected for this study. Feed samples (n = 58), fecal samples (n = 108), and milk samples (n = 78) were collected from the farms and analysed for chemical composition. Feed samples were also analysed for metabolisable energy (ME) contents and in vitro organic matter digestibility according to Hohenheim-Feed-Test. Furthermore, fecal nitrogen concentration was determined to estimate in vivo organic matter digestibility. ME and nutrient intakes were calculated based on the farmer’s estimate of dry matter intake and the analysed composition of the feed ingredients. ME and nutrient intakes were compared to recommended standard values for adequate supply of ME, utilizable crude protein, rumen undegradable crude protein (RUCP), phosphorus (P), and calcium (Ca). Technology Impact Policy Impact Calculation model complemented with a partial life cycle assessment model was used to estimate greenhouse gas emissions of milk production at farm gate. The model predicts CH4, N2O and CO2 gases emitted either directly or indirectly. Average daily energy corrected milk yield (ECM) was 19 kg and ranged between 11 and 27 kg. The mean of ME intake of all farms was 184 MJ/d with a range between 115 and 225 MJ/d. Intake of RUCP was lower than the standard requirements in six farms ranging between 19 and 137 g/d, was higher (32 and 93 g/d) in two farms, and matched the requirements in one farm. P intake was higher than the requirements in all farms (mean oversupply = 19 g/d) and ranged between 3 and 30 g/d. Ca intake was significantly below the requirements in small scale farms. Milk nitrogen efficiency N-eff (milk N/intake N) varied between 19% and 28% and was mainly driven by the level of milk yield. Total CO2 equivalent (CO2 equ) emission ranged between 0.90 and 1.88 kg CO2/kg ECM milk, where the enteric and manure CH4 contributed to 52% of the total CO2 equ emissions, followed by the indirect emissions of N2O and the direct emissions of CO2 gases which comprises 17% and 15%, respectively, from total CO2 equ emissions. Emissions per kg of milk were significantly driven by the level of milk production (r2 = 0.93) and of eDMI (r2 = 0.88), while the total emissions were not influenced by diet composition. A difference of 16 kg ECM/d in milk yield, 9% in N-eff and of 0.9 kg CO2 equ/kg in ECM milk observed between low and high yielding animals. To improve the nutritional status of the animals, protein requirements have to be met. Furthermore, low price by-products with a low carbon credit should be included in the diets to replace the high proportion of imported concentrate feeds and consequently improve the economic situation of dairy farms and mitigate CO2 equ emissions.  相似文献

Real-time analysis of milk coagulation properties as performed by the AfiLab™ milk spectrometer introduces new opportunities for the dairy industry. The study evaluated the performance of the AfiLab™ in a milking parlor of a commercial farm to provide real-time analysis of milk-clotting parameters –Afi-CF for cheese manufacture and determine its repeatability in time for individual cows. The AfiLab™ in a parlor, equipped with two parallel milk lines, enables to divert the milk on-line into two bulk milk tanks (A and B). Three commercial dairy herds of 220 to 320 Israeli Holstein cows producing ∼11 500 l during 305 days were selected for the study. The Afi-CF repeatability during time was found significant (P < 0.001) for cows. The statistic model succeeded in explaining 83.5% of the variance between Afi-CF and cows, and no significant variance was found between the mean weekly repeated recordings. Days in milk and log somatic cell count (SCC) had no significant effect. Fat, protein and lactose significantly affected Afi-CF and the empirical van Slyke equation. Real-time simulations were performed for different cutoff levels of coagulation properties where the milk of high Afi-CF cutoff value was channeled to tank A and the lower into tank B. The simulations showed that milk coagulation properties of an individual cow are not uniform, as most cows contributed milk to both tanks. Proportions of the individual cow's milk in each tank depended on the selected Afi-CF cutoff. The assessment of the major causative factors of a cow producing low-quality milk for cheese production was evaluated for the group that produced the low 10% quality milk. The largest number of cows in those groups at the three farms was found to be cows with post-intramammary infection with Escherichia coli and subclinical infections with streptococci or coagulase-negative staphylococci (∼30%), although the SCC of these cows was not significantly different. Early time in lactation together with high milk yield >50 l/day, and late in lactation together with low milk yield<15 l/day and estrous (0 to 5 days) were also important influencing factors for low-quality milk. However, ∼50% of the tested variables did not explain any of the factors responsible for the cow producing milk in the low – 10% Afi-CF.  相似文献

Somatic cell count (SCC) can be used as a proxy for the prevalence of mastitis in a herd, reflecting the hygiene conditions and management practices on dairy farms, and thus an indicator of milk quality. In this study, we investigated how the adoption of management practices in milking systems can contribute to the reduction of SCC levels and improve milk quality. We collected data regarding management practices from 91 dairy farms in three municipalities of southeastern Pará: Parauapebas, Curionópolis, and Eldorado dos Carajás. Fifty milliliters of milk from each farm were collected in bottles containing bronopol, to preserve SCC. An exploratory factorial analysis (EFA) was performed to reduce the number of variables (management techniques) on dairy farms to some latent factors. We then used the selected factors to estimate the bovine mastitis management index to classify farms according to their use of technology and management techniques. Our results showed that most of the farmers (65.9%) used management techniques inefficiently in their systems, resulting in a significant loss of product quality, while only 3.3% had adopted the full set of techniques. The EFA results demonstrated that simple management practices including regular cleaning of the milking lines, a strip cup test, the California mastitis test, and washing teats with water before milking could be adopted to improve milk quality. However, in scenarios where the regulations become more rigorous, most farmers are unable to meet the maximum allowable SCC requirements, necessitating management innovations to reduce SCC. The potential association between hygienic conditions in the environment of lactating cows and the presence of gliotoxinogenic Aspergillus fumigatus strains was studied. Milk samples (individual cow’s milk [ICM], bulk tank milk [BTM]) from 44 dairy farms were sampled. In ICM samples, eight different species of Aspergillus were identified. A. flavus and A. fumigatus were predominant, with 37.8 % and 26.1 % relative densities, respectively. A. fumigatus strains were isolated from 61.4 % of the BTM samples, and 34 % of these strains were able to produce gliotoxin. Principal component analysis was used to associate the presence of A. fumigatus with some hygienic and sanitary practices. A significant and positive correlation was observed between dry cow therapy and forestripping. The presence of A. fumigatus gliotoxin producers in milk was associated with high somatic cells count (SCC) samples. Good hygienic and sanitary practices were associated with absence of A. fumigatus and relatively low SCCs of <250,000 cells/ml. In general, a high percentage of dairy farms were positive for A. fumigatus in BTM samples. This is the first work that indicates the positive effects of adequate hygienic and sanitary practices in dairy herds on the control of A. fumigatus and related species. By reducing the frequency of Aspergillus spp. in the dairy environment, the risk of farm handlers’ exposure and the risk of intramammary fungal infections would also be reduced.  相似文献

Gossypol is a putative BH3 mimetic proposed to inhibit BCL2 and BCLXL based on cell-free assays. We demonstrated previously that gossypol failed to directly inhibit BCL2 in cells or induce apoptosis in chronic lymphocytic leukemia (CLL) cells or platelets, which require BCL2 or BCLXL, respectively, for survival. Here, we demonstrate that gossypol rapidly increased activity of phospholipase A2 (PLA2), which led to an increase in cytoplasmic calcium, endoplasmic reticulum (ER) stress, and up-regulation of the BH3-only protein NOXA. Pretreatment with the PLA2 inhibitor, aristolochic acid, abrogated the increase in calcium, ER stress, and NOXA. Calcium chelation also abrogated the gossypol-induced increase in calcium, ER stress, and NOXA, but not the increase in PLA2 activity, indicating that PLA2 is upstream of these events. In addition, incubating cells with the two products of PLA2 (lysophosphatidic acid and arachidonic acid) mimicked treatment with gossypol. NOXA is a pro-apoptotic protein that functions by binding the BCL2 family proteins MCL1 and BFL1. The BCL2 inhibitor ABT-199 is currently in clinical trials for CLL. Resistance to ABT-199 can occur from up-regulation of other BCL2 family proteins, and this resistance can be mimicked by culturing CLL cells on CD154+ stroma cells. We report here that AT-101, a derivative of gossypol in clinical trials, overcomes stroma-mediated resistance to ABT-199 in primary CLL cells, suggesting that a combination of these drugs may be efficacious in the clinic.  相似文献

Data from 113 Dutch organic farms were analysed to determine the effect of cross-breeding on production and functional traits. In total, data on 33 788 lactations between January 2003 and February 2009 from 15 015 cows were available. Holstein–Friesian pure-bred cows produced most kg of milk in 305 days, but with the lowest percentages of fat and protein of all pure-bred cows in the data set. Cross-breeding Holstein dairy cows with other breeds (Brown Swiss, Dutch Friesian, Groningen White Headed, Jersey, Meuse Rhine Yssel, Montbéliarde or Fleckvieh) decreased milk production, but improved fertility and udder health in most cross-bred animals. In most breeds, heterosis had a significant effect (P < 0.05) on milk (kg in 305 days), fat and protein-corrected milk production (kg in 305 days) and calving interval (CI) in the favourable direction (i.e. more milk, shorter CI), but unfavourably for somatic cell count (higher cell count). Recombination was unfavourable for the milk production traits, but favourable for the functional traits (fertility and udder health). Farm characteristics, like soil type or housing system, affected the regression coefficients on breed components significantly. The effect of the Holstein breed on milk yield was twice as large in cubicle housing as in other housing systems. Jerseys had a negative effect on fertility only on farms on sandy soils. Hence, breed effects differ across farming systems in the organic farming and farmers can use such information to dovetail their farming system with the type of cow they use.  相似文献

Gastrointestinal nematode (GIN) infection is one of the main constraints to sheep production both in temperate and tropical countries. Economic losses caused by GIN are related to decreased production, treatment costs and even animal death. The present paper was aimed at assessing the anthelmintic efficacy (based on faecal egg count reduction) of moxidectin and ivermectin both admistered per os at dose rate of 0.2 mg/Kg body weight and the benefit of anthelmintic treatments on milk production in a commercial dairy sheep farms in central Italy whose animals were naturally infected by GIN.  相似文献

Pathogenic Escherichia coli or its associated virulence factors have been frequently detected in dairy cow manure, milk, and dairy farm environments. However, it is unclear what the long-term dynamics of E. coli virulence factors are and which farm compartments act as reservoirs. This study assessed the occurrence and dynamics of four E. coli virulence factors (eae, stx1, stx2, and the gamma allele of the tir gene [γ-tir]) on three U.S. dairy farms. Fecal, manure, water, feed, milk, and milk filter samples were collected from 2004 to 2012. Virulence factors were measured by postenrichment quantitative PCR (qPCR). All factors were detected in most compartments on all farms. Fecal and manure samples showed the highest prevalence, up to 53% for stx and 21% for γ-tir in fecal samples and up to 84% for stx and 44% for γ-tir in manure. Prevalence was low in milk (up to 1.9% for stx and 0.7% for γ-tir). However, 35% of milk filters were positive for stx and 20% were positive for γ-tir. All factors were detected in feed and water. Factor prevalence and levels, expressed as qPCR cycle threshold categories, fluctuated significantly over time, with no clear seasonal signal independent from year-to-year variability. Levels were correlated between fecal and manure samples, and in some cases autocorrelated, but not between manure and milk filters. Shiga toxins were nearly ubiquitous, and 10 to 18% of the lactating cows were potential shedders of E. coli O157 at least once during their time in the herds. E. coli virulence factors appear to persist in many areas of the farms and therefore contribute to transmission dynamics.  相似文献
