Abstract: Ca oxalate crystal formation was examined in Pistia stratiotes L. leaves during excess Ca and Ca-deficient conditions. Pistia produces druse crystal idioblasts in the adaxial mesophyll and raphide idioblasts in the abaxial aerenchyma. Raphide crystals were previously found to grow bidirectionally, and here we show that Ca is incorporated along the entire surfaces of developing druse crystals, which are coated with membrane-bound microprojections. Leaves formed on plants grown on 0 Ca medium have fewer and smaller druse crystals than leaves formed under 5 mM Ca ("control") conditions, while raphide crystal formation is completely inhibited. When plants were moved from 0 to 15 mM ("high") Ca, the size and number of crystals in new leaves returned to (druse) or exceeded (raphide) control levels. High Ca also induced formation of druse, but not raphide, crystals in differentiating chlorenchyma cells. When plants were transferred from 15 mM Ca to 0 Ca, young druse crystals were preferentially partially dissolved. Oxalate oxidase, an enzyme that degrades oxalate, increased during Ca deficiency and was localized to the crystal surfaces. The more dynamic nature of druse crystals is not due to hydration form as both crystal types are shown to be monohydrate. Part of the difference may be because raphide idioblasts have developmental constraints that interfere with a more flexible response to changing Ca. These studies demonstrate that excess Ca can be stored as Ca oxalate, the Ca can be remobilized under certain conditions, and different forms of Ca oxalate have different roles in bulk Ca regulation.  相似文献

Summary Lemna minor, which produces many calcium oxalate raphide crystals, was grown on media containing in addition to Ca, 200 M of one of the following divalent cations: Ba, Cd, Co, Mn or Sr. Energy dispersive X-ray analysis showed that only Sr was incorporated into the raphides at levels detectable by the analysis technique. Incorporation of Sr into other insoluble compounds, such as cell wall material, could not be detected. Plant species which form different crystal types in their leaves (Beta vulgaris, crystal sand;Arthrostema ciliatum, druse;Glycine canescens, prismatic) also incorporated Sr into their crystals when grown hydroponically on nutrient medium containing 200 M Sr.Axenic cultures ofL. minor were used to examine further the process of Sr incorporation into plant crystals. When grown on nutrient solution with 5 M Ca, increasing the Sr concentration resulted in increases of the amount of Sr incorporated into the raphide crystals. The ratio of Sr to Ca became greater as the Sr concentration was increased. This ratio change was due to both an increase in the amount of Sr incorporated and a decrease in the Ca incorporated. Analysis of the number of crystal idioblasts formed as a function of Sr concentration shows fewer idioblasts are produced as Sr became high. Competition with Ca and interference of Ca utilization by Sr is indicated.  相似文献

This study was to report and describe the formation of Ca oxalate crystals and to explore whether there is any correlation between their abundant formation and the ability of plant to uptake and accumulate high levels of toxic metals. Soil-grown Corchorus olitorius L. (Tiliaceae) seedlings were further grown in water culture in the presence of Cd, Pb, Cu, or Al (0–10 g/ml) for 20 days. Light and electron microscopic examinations revealed a large number of intracellular prismatic-shaped Ca oxalate crystals in both leaf and callus cells. Crystals were formed in the vacuole, a single large crystal being formed per cell. The crystal-containing cells differed in size and shape from crystal-free cells, they were rich in organelles, membranes, and vesicles and have dense cytoplasm, enlarged nucleus and modified starch-lacking plastids with few grana. These cells look highly active. Corchorus plants treated with Cd, Pb, Cu, and Al accumulated these metals to the levels several times higher than untreated plants. The contents of Pb, Cd, Al, and Cu in leaf tissues of plants grown in the presence of 5 g/ml of these metals were 10, 20, 25, and 40 times higher, respectively, than those in plants grown on media devoid of them. X-ray microanalysis of Ca oxalate crystals in leaves from plants exposed to 5 g/ml Cd, Pb, Al, or Cu indicated the incorporation only of Al into these crystals. Results of this paper suggest a possible contribution for Ca oxalate-crystal formation in sequestering and tolerance of at least some toxic metals.  相似文献

Summary Bundles of fibrils and tubular structures were found to be associated with growing trabeculae ofCaulerpa simpliciuscula. In the rhizome tips, the bundles had an average diameter of 0.1 to 0.3 m, and a length greater than 10 m. The fibrils in the bundles were oriented in a strictly parallel fashion, with an individual thickness of 3–8 nm. The development of trabeculae started with the apposition of material of low electron density onto the bundles, which in this way became the inner skeleton of the trabeculae.Although fibre bundles with the same internal structure also occurred in the frond tip, these rarely contributed to trabecula formation. In the frond tips a different type of bundle with paracrystalline structure was found associated with the trabecular surface, forming a temporary connection between adjacent trabeculae. Permanent connection was achieved by deposition of further layers of trabecular material. These bundles in the frond tip consisted of two layers of tubular elements with a wall thickness of 80 Å and an inner diameter of 20–25 nm.Both fibre bundles and tubular bundles appear to contribute to trabecula formation. The similarity of these structures to the vacuolar inclusions observed in other siphonous algae is discussed.  相似文献

Pueraria lobata hairy roots have faster elongationand more branches than normal roots. The responses of hairy roots and normalroots to treatment with three auxins, indole-3-acetic acid (IAA),indole-3-butyric acid (IBA), and naphthalene acetic acid (NAA) were different.In normal roots, all three auxins strongly stimulated lateral root formation atall tested concentrations. Responses to IAA and IBA in primary root growth andlateral root elongation were similar and depended on concentration; promotionat0.1 M, no effect at 1.0 M, and inhibition at2.5 M. In hairy roots, lateral root formation varied inresponseto the different auxins, i.e. depressed by NAA, unaffected by IAA, and promotedby IBA. Primary root growth was slightly inhibited by IBA and was unaffected byIAA. However, mean lateral root length was reduced in response to IAA and IBA.Only NAA exerted strong inhibition on primary and lateral root elongation inboth root types. The similar free IAA and conjugated IAA content but quitedifferent basal ethylene production and biosynthesis in hairy and normal rootssuggested different mechanisms of response to exogenous auxins in the two roottypes.  相似文献

Beech seedlings (Fagus sylvatica L. provenance Västra Torup) were grown in nutrient solution at low pH (4.2) and exposed to AlCl3 and different concentrations of nitrogen. The effects of AlCl3 and nitrogen on uptake of Ca2+ (45Ca) and H2PO4 - (32P) in roots of intact beech were studied. Crossections of roots were analyzed with respect to element concentrations, by use of Mikro-PIXE (particle induced X-ray emission). The distribution of Al, Ca, P, Mg, K, and S was analysed. The experiments were designed to evaluate the effects of aluminium on localization of elements in plant root tissue.Aluminium reduced the concentration of Ca in plants and increased that of K. High nitrogen levels in the solution further decreased the concentration of Ca in the roots. Aluminium (1.0 mM) effectively reduced the Ca2+ (45Ca) uptake in short time experiments. Aluminium accumulated in high concentration (up to 500 mol/g dry weight in areas of 30×30 m2) on the root surface, epidermis and outer layers of cortex. Corresponding areas had an extremely low Ca concentration (ca 5 mol/g dry weight) which could be harmful for regulation of mineral uptake and development of roots.It is concluded that the calcium concentration in roots was reduced by aluminium. The combination with high nitrogen levels further reduced calcium changing the mineral balance which could result in deficiency conditions of calcium in the root.  相似文献

Cultured hypocotyl explants of tomatillo (Physalis ixocarpa Brot.), were evaluated with regard to their morphogenic responses to combinations of benzyladenine (BA, 0–5 M) with either naphthaleneacetic acid (NAA, 0–50 M) or 2,4-dichlorophenoxyacetic acid (2,4-D, 0–50 M). The induction of shoots or roots was dependent on the cytokinin/auxin combination.Hypocotyl explants failed to form shoots when they were grown on media containing either a cytokinin or an auxin alone. The highest frequency of shoot formation was observed on media containing 12.5–25 M BA and 5 M NAA. Likewise the highest frequency of root formation was observed on media supplemented with 1 M BA and 1 M NAA. Complete plants were regenerated and transferred to soil, where they reached maturity.  相似文献

The effect of indole-3-butyric acid (IBA) on the formation of non-transformed and rol gene transformed roots on stem slices of in vitro cultured shoots of Rosa hybrida L. Moneyway was examined. Formation of adventitious roots on this rootstock was dependent on the IBA dose; it was not affected by the presence of other root primordia on the same explant. Application of 0.32 to 1 M IBA during 5 days, followed by transfer to medium without hormones resulted in maximum root formation (90%) after three weeks. The formation of such untransformed roots was completely inhibited by transfer to medium with 5 mg 1–1 kanamycin two days after excision. Ri roots were formed upon inoculation with A. rhizogenes LBA9402 harbouring two plasmids: pRi1855, comprising the rol genes and the binary plasmid p 35Sgusintron with the nptII gene for kanamycin resistance and the CaMV 35Sgusintron gene. The formation of these Ri roots on kanamycin-containing medium was independent of the presence of IBA. Stem slices inoculated with a disarmed A. tumefaciens GV3101, harbouring only the nptII gene, formed callus and subsequently roots in the presence of kanamycin exclusively on medium with high IBA concentrations (10 or 100 M). Root formation at 100 M IBA was considerably improved by transformation with the rolB gene under the influence of the strong CaMV 35S promoter. In addition, low IBA (0.1 and 1 M) stimulated the formation of roots only on stem slices transformed with A. tumefaciens harbouring the rolA+rolB+rolC genes; the rooting response at 10 M IBA was much improved. During treatment of isolated, peeled leaflets of Gleditsia triacanthos with 0.5–2 mM [45Ca]acetate, saturation of the cell-wall free space with Ca2+ occurred within 10 min and was followed by a period of 6–10 h during which there was no significant Ca-uptake into the protoplast, but apoplastic Ca2+ was periodically released into the medium. Later, Ca2+ was absorbed for 3–4 d at rates of up to 2.2 mol Ca2+·h-1·(g FW)-1 to final concentrations of 350 mol Ca2+· (g FW)-1. The distribution of absorbed Ca2+ between cell wall, vacuole and Ca-oxalate crystals was determined during Ca-uptake. Wheras intact, cut leaflets deposited absorbed Ca2+ as Ca-oxalate in the crystal cells, peeled leaflets lacking crystal cells accumulated at least 40–50 mol·(g FW)-1 soluble Ca2+ before the absorbed Ca2+ was precipitated as Ca-oxalate. These observations indicate that the mechanisms for the continuous uptake of Ca2+, the synthesis of oxalate and the precipitation of Ca2+ as Ca-oxalate are operational in the crystal cells of intact leaflets, but not in the mesophyll cells of peeled leaflets where they must be induced by exposure to Ca2+. The precipitation of absorbed Ca2+ as Ca-oxalate by the crystal cells of isolated Gleditsia leaflets illustrates the role of these cells in the excretion of surplus Ca2+ which enters normal, attached leaves with the transpiration stream.In addition to acetate, only Ca-lactate and Ca-carbonate lead to Ca-uptake, but at rates well below those observed with Ca-acetate. Other small organic anions (citrate, glycolate, glyoxalate, malate) and inorganic anions (chloride, nitrate, sulfate) did not permit Ca-uptake. Acetate-14C was rapidly absorbed during Ca-uptake, but less than 20% was incorporated into Ca-oxalate; the rest remained mostly in the soluble fraction or was metabolized to CO2. Acetate, as a permeable weak acid, may enable rapid Ca-uptake by stimulating proton extrusion at the plasmalemma and by serving as a counterion during Ca-accumulation in the vacuole, but is unlikely to function as the principal substrate for oxalate synthesis.  相似文献

Summary Phytoplankton abundance in tropical lakes is more often judged to be limited by nitrogen than phosphorus, but seldom does the evidence include controlled enrichments of natural populations. In January 1980 we performed the first experimental fertilization in an equatorial African soda lake, Lake Sonachi, a small, meromictic volcanic crater lake in Kenya. During our study the natural phytoplankton abundance was ca. 80 g chl a/l, and the euphotic zone PO4 and NH4 concentrations were less than 0.5 M. In the monimolimnion PO4 reached 180 M and NH4 reached 4,600 M. Replicate polyethylene cylinders (5 m long, 1.2 m3) were enriched to attain 10 M PO4 and 100 M NH4. Phytoplankton responses were measured as chlorophyll, cell counts and particulate N, P and C. After two days, the chlorophyll increase in the P treatment was significantly higher than the control (P<0.01) while the N treatment was not. After five days the molar N/P ratio of seston was the same in the N treatment and control (23) but only 6 in the P treatment. The molar N/P ratio of seston in an unenriched Lake Sonachi sample was 21 and in samples from Lakes Bogoria and Elmenteita, two shallow soda lakes in Kenya, the ratios were 12 and 70 respectively. We conclude that limitation of phytoplankton abundance by phosphorus can occur even in some tropical African soda lakes.  相似文献

Adventitious shoot primordia were formed on horseradish hairy root cultured in dark. Plantlet formation frequency from the primordia was higher than that from root fragments. Culture for 26 days provided the adventitious shoot primordia, which had the highest potential for plantlet formation (53% explants at 40 days). Benzyladenine supplementation in the dark caused primordium enlargement, but did not increase the number of primordia formed. After adventitious shoot primordia were encapsulated with calcium alginate, kinetin supplementation (2.0–4.0 M) increased the shoot formation frequency (65–80% explants at 20 days) in the light, but also promoted the undesirable formattion of multiple shoots. Supplementation with naphthaleneacetic acid (0.27–5.4 M) in the calcium alginate beads in light enhanced the root emergence from primordia without inhibition of plantlet formation when the encapsulated beads were put on the agar-medium without naphthaleneacetic acid.  相似文献

Summary Precipitation of Ca oxalate in the sarcoplasmic reticulum of chemically skinned rabbit psoas fibers caused an increase in light scattering which was proportional to the amount of Ca accumulated per unit fiber volume. The increase in scattering was used to measure net accumulation rates and steady-state Ca capacities of the sarcoplasmic reticulum in single fibers. The data obtained were qualitatively and quantitatively similar to those reported for isolated vesicle preparations.Under conditions in which Ca was not depleted from the medium, Ca accumulation was linear with time over much of its course. Steady-state capacities were independent of the Ca concentration; uptake rates were half-maximal at 0.5 m Ca++ and saturated above about 1.0 m. Both rate and capacity varied with the oxalate concentration, being maximal at oxalate concentrations >=5mm and decreasing in proportion to one another at lower concentrations, with a threshold near 0.25mm. At the lower loads, electron micrographs showed many sarcoplasmic reticulum elements empty of precipitate alongside others that were full, whereas virtually all were filled in maximally loaded fibers. These data indicate that the Ca oxalate capacity of each fiber varies with the number and volume of elements in which Ca oxalate crystals can form at a given oxalate concentration, and that individual regions of the sarcoplasmic reticulum within each sarcomere differ in their ability to support Ca oxalate precipitation. Our working hypothesis is that this range in ability to form Ca oxalate crystals involves differences in ability to accumulate and retain ionized Ca inside the sarcoplasmic reticulum.  相似文献

Summary Apical application of silver nitrate (AgNO3; 50 and 100 g per plant) and silver thiosulphate anionic complex (Ag(S2O3) 2 3– ; STS; 25, 50 and 100 g per plant) to female plants of Cannabis sativa induced the formation of reduced male, intersexual and fully altered male flowers on the newly formed primary lateral branches (PLBs); 10 g per plant of AgNO3 was ineffective and 150 g treatment proved inhibitory. A maximum number of fully altered male flowers were formed in response to 100 g STS. The induced male flowers produced pollen grains that germinated on stigmas and effected seed set. Silver ion applied as STS was more effective than AgNO3 in inducing flowers of altered sex. The induction of male flowers on female plants demonstrated in this work is useful for producing seeds that give rise to only female plants. This technique is also useful for maintaining gynoecious lines.  相似文献

Summary The cytoarchitecture of the medial layer of rat thoracic aorta was examined by scanning electron microscopy after removal of the connective tissue. The outermost lamella showed a lattice-like structure of muscle bundles of closely apposed smooth muscle cells (SMCs), whereas the inner lamellae consisted of more-or-less continuous muscle sheets of vaguely defined subgroups of parallel SMCs. Longitudinal rows of ridges ran along the adventitial surface of these muscle bundles and sheets. The SMCs of the outermost lamella, were 5.1 m wide, and varied in shape, whereas those of the inner lamellae, were 52.7 m long, 2.6 m wide and 4.1 m thick, and were elongated, spindle-shaped cells with serrated outlines. These latter SMCs extended obliquely, and partially overlapped each other. The surface of the SMCs in the outermost lamella exhibited a rugged texture, with nodular protrusions and oblique and longitudinal laminar folds, while the inner lamellar cells showed longitudinal laminar folds and finger-like processes on both sides of the ridges, pointing in opposite directions to the ridges. The angle of deviation from the transverse axis of the vessel, of the muscle bundles and subgroups in the outermost lamella, was 33.6°, in the second and third lamellae, 22.5°, and in the innermost lamellae, 12.8°. The mean angle of the muscle bundle and subgroup arrangement, with respect to the long axis of the vessel, however, was basically 90° in all lamellae.  相似文献

The growth and differentiation of callus tissues derived from cotyledons of ten cultivars ofCucumis sativus L. were investigated. Cotyledonary explants from all ten cultivars formed callus tissue on Murashige and Skoog (MS) medium supplemented with 0.5 M 2,4-dichlorophenoxyacetic acid and 5 M 6-benzylaminopurine. Fresh weight of the callus tissues averaged 1 to 8 g per flask after five weeks of culture. Shoot development was achieved in three cultivars, Hukchinju, Manchoonchoungjang and Seoul, on MS medium supplemented with 0.5 M -naphthaleneacetic acid and 5 M 6-benzylaminopurine. Reducing the 6-benzylaminopurine concentration to 0.01 M resulted in root formation on callus tissues and on shoots transferred to this medium. All cultivars gave the same response in tests of root formation, but shoot regeneration from callus culture of cucumber cotyledons was dependent on genotype with cultivar Manchoonchoungjang exhibiting the best shoot differentiation capability among the genotypes examined. Examination of mitotic metaphase from the regenerants revealed that all were tetraploid.  相似文献

This study revealed strong evidence that nymphs of the silverleaf whitefly, Bemisia argentifolii Bellows and Perring, are obligate feeders on vascular bundles and that there are large differences between different host plants as to the availability of vascular bundles to silverleaf whitefly nymphs. The relationship between nymphs and leaf vascular bundles was studied using 1) leaf sectioning and 2) techniques of leaf clearing of intact leaves. A geometric model is presented of the feeding relationship of vascular bundle-using homopterans. The relative abundance of vascular bundles was examined in six species of host plants that varied from highly preferred to tolerably acceptable. Included in order of acceptance were cantaloupe, cotton, hibiscus, broccoli, lantana and lettuce. The length of vascular bundle per 1.0 mm2 of leaf surface ranged from about 10 mm in cantaloupe to 2.8 mm in lettuce. Salivary sheaths were found to connect with vascular bundles in 100% of the intact nymphs examined by the staining and clearing technique. However only 64% of those examined by the sectioning technique appeared to be connected to vascular bundles. This indicates that the sectioning technique leads to a high rate of error, causing an underestimation of the importance of direct contact with vascular bundles. About 50% of epidermal stylet penetrations were through epidermal cells; the remaining 50% went through intercellular junctions. On cotton leaves, the distance between the point of labial contact with the leaf surface and the nearest point of the vascular bundle rarely exceeded 60 m. Our studies show that while 50% of lettuce leaf-surface was beyond 60 m of a vascular bundle, only 10% of cantaloupe leaf surface area was outside of the 60 m range. In cotton, mean distance from labium to the nearest point of the vascular bundle was 40.9 m (SEM=2.66, N=50, range 0–80 m). Over 98% of all salivary sheaths went to minor veins (78% to single-filament vascular bundles, nearly 20% to double filament bundles). Fewer than 2% went to bundles with 3 or more filaments.  相似文献

A hydroponic experiment was carried out to study genotypic differences in effect of Al and Cd on growth and antioxidant enzyme activities by using 2 two-row winter barley genotypes (Hordeum vulgare L.) with different Al resistance, the relatively resistant Gebeina and the sensitive Shang 70–119. The seedling growth, presented as shoot height, root length and dry weight of root and shoot, and tillers per plant were inhibited by all stress treatments, including low pH, 100 M Al (pH 4.0) and 1.0 M Cd+100 M Al (pH 4.0), while 1.0 M Cd showed a slight stimulation of growth. The inhibition was more severe in 1.0 M Cd +100 M Al (pH 4.0) than in 100 M Al (pH 4.0), indicating that the effect of Cd and Al is synergistic. Al-sensitive genotype Shang 70–119 was more inhibited than Al-resistant genotype Gebeina. Proline concentration in leaves was significantly increased when plants were exposed to all stress treatments, being more pronounced in Shang 70–119 than in Gebeina. A highly significant increase in malonaldehyde (MDA) concentration, and a stimulation of superoxide dismutase (SOD) and peroxidase (POD) activities were recorded in the plants subjected to low pH, 100 M Al (pH 4.0) and 1.0 M Cd +100 M Al(pH 4.0) treatments, and the extent of the increase varied greatly depending on concentration and time of exposure. Shang 70–119 had a higher MDA concentration, and less increase in SOD activity when first exposed than Gebeina had.  相似文献

Root segments (1 cm long) were excised from 15–20 day old seedlings of silktree (Albizzia julibrissin) grown on B5 medium. About 50% of the control (no growth regulators added) root explants formed shoot buds within 15 days after placement on the culture medium. After 30 days, there were about 4 shoots per control explant. Addition of low levels of various auxins (0.5 M) did not influence the formation of shoot buds from the explants. Higher concentrations (5M), however, decreased shoot regeneration. Kinetin and 2iP did not influence shoot regeneration at the concentrations tested (1 & 10 M). Addition of benzyladenine, Zeatin, or thidiazuron to the culture medium increased both the percentage of explants that formed shoots and the number of shoots per explant. Thidiazuron was highly effective in stimulating shoot formation at low concentrations (<1 M). At 0.05 M thidiazuron, 95% of the explants produced shoots and about 10 shoots were formed per explant. Compared to TDZ, higher concentrations (10 M) of benzyladenine and Zeatin were required to enhance shoot formation. The relationship between root Al concentrations and Al fractions in the soil solution was examined in a mature Abies amabilis ecosystem in the Cascade Range of Washington State. The naturally acidic soils in these ecosystems lead to high concentrations of aqueous Al in soil solutions and contribute to the biocycling of Al by the A. amabilis/T. mertensiana stand. Root concentrations of Al were very closely related to aqueous Al3+ activities, but poorly correlated with total aqueous Al concentrations. The solution Al/Ca molar ratios followed a seasonal cycle with low values during the fall and high values during the spring. Ratios remained <1 throughout the year in the Oa horizon while they varied between 2 and 14 in the E and Bhs horizons. The vertical distribution of roots and the mortality of fine roots may be linked to the soil solution Al/Ca ratio. Root cation exchange capacity ranged between 180 and 225 mol g-1 and the exchangeable Al fraction represented from 12–17% of the total Al content in the root. Evidence for solid-phase co-precipitates of Al with PO4 and oxalate was indicated from selective dissolution of the root tissue. Sufficient quantities of PO4 and oxalate exist in the roots to tie up 20–40% of the Al present in the roots of the Oa and E horizons, but only 9% of that present in the Bhs horizon. Species differences in the distribution of Al between the above-ground and below-ground components may be dictated by these retention processes in the fine roots.  相似文献
