Modern coral taxonomy has begun to resolve many long‐standing problems in traditional systematics stemming from its reliance on skeletal macromorphology. By integrating examinations of colony, corallite, and subcorallite morphology with the molecular sequence data that have proliferated in the last decade, many taxa spread across the scleractinian tree of life have been incorporated into a rigorous classification underpinned by greater phylogenetic understanding. This monograph focuses on one of the most challenging clades recovered to date – its disarray epitomized by the informal name ‘Bigmessidae’. This group of predominantly Indo‐Pacific species previously comprised families Merulinidae, Faviidae, Pectiniidae, and Trachyphylliidae, but in a recent study these have been incorporated within Merulinidae. We studied 84 living merulinid species by examining morphological traits at three different scales of coral skeletal structure ? macromorphology, micromorphology, and microstructure ? to construct a morphological matrix comprising 44 characters. Data were analysed via maximum parsimony and also transformed onto a robust molecular phylogeny under the parsimony and maximum likelihood criteria. Comparisons amongst morphological character types suggest that although many characters at every scale are homoplastic, some to a greater extent than others, several can aid in distinguishing genus‐level clades. Our resulting trees and character analyses form the basis of a revised classification that spans a total of 139 species contained within 24 genera. The tree topologies necessitate the synonymization of Barabattoia as Dipsastraea, and Phymastrea as Favites. Furthermore, Astrea and Coelastrea are resurrected, and one new genus, P aramontastraea Huang & Budd gen. nov. , is described. All the genera in Merulinidae, along with the monotypic Montastraeidae and Diploastraeidae, are diagnosed based on the characters examined. The integrative classification system proposed here will form the framework for more accurate biodiversity estimates and guide the taxonomic placement of extinct species. © 2014 The Linnean Society of London  相似文献

The data on different modes of reproduction in sea anemones are generalized. These animals can reproduce sexually in an ordinary way or by parthenogenesis. Asexual reproduction occurs in various forms, such as transverse and longitudinal fission, pedal laceration, or autotomy of tentacles. The common shallow water species of sea anemones (Actiniaria) and tube anemones (Ceriantharia) of the Azores are listed. Eight species of sea anemones are mentioned, the species Cereus pedunculatus and Sagartia affinis being new records for the archipelago. Both species of Ceriantharia, namely Arachnanthus nocturnus and Pachycerianthus solitarius, are recorded from the Azores for the first time. Arachnanthus nocturnus is also recorded from the Cape Verde Islands and from Madeira for the first time. Communicated by H.-D. Franke  相似文献

Slow mitochondrial DNA sequence evolution in the Anthozoa (Cnidaria)   总被引:33,自引:0,他引:33  
Mitochondrial genes have been used extensively in population genetic and phylogeographical analyses, in part due to a high rate of nucleotide substitution in animal mitochondrial DNA (mtDNA). Nucleotide sequences of anthozoan mitochondrial genes, however, are virtually invariant among conspecifics, even at third codon positions of protein-coding sequences. Hence, mtDNA markers are of limited use for population-level studies in these organisms. Mitochondrial gene sequence divergence among anthozoan species is also low relative to that exhibited in other animals, although higher level relationships can be resolved with these markers. Substitution rates in anthozoan nuclear genes are much higher than in mitochondrial genes, whereas nuclear genes in other metazoans usually evolve more slowly than, or similar to, mitochondrial genes. Although several mechanisms accounting for a slow rate of sequence evolution have been proposed, there is not yet a definitive explanation for this observation. Slow evolution and unique characteristics may be common in primitive metazoans, suggesting that patterns of mtDNA evolution in these organisms differ from that in other animal systems.  相似文献

The cnidom of the sea anemone Sagartiogeton viduatus (Muller, 1776) is described from interference‐contrast light micrographs (LMs) and scanning electron micrographs (SEMs). Special attention is given to nematocyst maturation, including the differentiation of the shaft into proximal and main regions as helical folding of the shaft wall proceeds. Comparisons are made with Metridium senile (Linnaeus, 1761), whose cnidom, with a few exceptions, is closely similar to that of S. viduatus. The two anemones possess b‐ and p‐mastigophores, p‐amastigophores, isorhizas and spirocysts. Although the majority of cnidae in S. viduatus is smaller than corresponding ones in M. senile, they are grouped into the same size classes as those of M. senile, namely small, medium and large. The main differences from M. senile cnidae are the followings: (1) Large acontia p‐amastigophores are the largest nematocysts in S. viduatus. (2) They are noticeably larger than the large acontia b‐mastigophores, and (3) they are separated from the p‐amastigophores of M. senile by the sinusoid pattern of their U‐shaped capsular matrix. (4) The large acontia b‐mastigophores are microbasic and not mesobasic as in M. Senile, and (5) they do not produce darts. (6) Another difference from M. senile is the absence of catch‐tentacle isorhizas.  相似文献

Five isoforms of tropomyosin, designated as TMa, TMb, TMc, TMd, and TMe, were detected in the sea anemone, Anthopleura japonica. The apparent molecular weights of these isoforms were estimated to be approximately 30 kD to 37.5 kD, and their pI values were approximately 4.55 (TMa and TMb) and 4.65 (TMc, TMd, and TMe). Although sea anemone tropomyosin isoforms have the ability to bind to rabbit skeletal muscle actin, they preferably bind to actin at higher concentrations of Mg(2+) (10-20 mM) and slightly lower pH (6.2-7.2) than those used in conventional conditions. Antigenic properties of sea anemone tropomyosin seemed to be considerably specific to each isoform. Distribution of tropomyosin isoforms in the sea anemone body was somewhat portion-specific. TMa, TMb, and TMe were detected similarly in the extracts from tentacle, oral disc, column, mouth, and pedal disc. Although TMc and TMd were detected abundantly in the tentacle extract and moderately in the column and mouth extracts, these components were not contained in the pedal disc extract and detected only faintly in the oral disc extract.  相似文献

Sea anemones (order Actiniaria) are among the most diverse and successful members of the anthozoan subclass Hexacorallia, being found at all depths and latitudes and in all marine habitats. Members of this group exhibit the greatest variation in anatomy, biology, and life history in Hexacorallia, and lack any morphological synapomorphy. Nonetheless, previous molecular phylogenetic studies have found that Actiniaria is monophyletic with respect to other extant hexacorallians. However, relationships within Actiniaria have remained unresolved, as none of these earlier works have included sufficient taxon sampling to estimate relationships within Actiniaria. We have analyzed sequences from two mitochondrial and two nuclear markers for representatives of approximately half of the family-level diversity within the order, and present the first phylogenetic tree for Actiniaria. We concur with previous studies that have suggested that molecular evolution is unusually slow in this group. We determine that taxonomic groups based on the absence of features tend not to be recovered as monophyletic, but that at least some classical anatomical features define monophyletic groups.  相似文献

Sphaeralcyon weddellensis gen. nov. et sp. nov. is described and illustrated from three colonies collected in the eastern Weddell Sea on the Polarstern cruises ANT V/3 and ANT VI/3. The new genus is compared with three closely related alcyoniid genera, all having capitate or mushroom-shaped colonies and dimorphic polyps. Sphaeralcyon is readily distinguishable from Malacacanthus by the presence of sclerites (absent in the latter genus), and from Anthomastus by sclerite shape, mainly in the form of tuberculate spheroids, and by the horny periderm on the stalk (absent in Anthomastus). Sphaeralcyon also differs from Verseveldtia in the set of sclerites, which are elongate spiny rods and a different type of tuberculate spheroid, and in the presence of eight-points as anthocodial armature (absent in Verseveldtia). The discovery of this new genus lends further support to the notion that diversity in benthic communities in Antarctica is higher than suggested by previous studies. Body wall strips and oral rings of Bunodosoma caissarum respond to the application of at least four categories of drugs with sustained contractions. Propionylcholine, butyrylcholine and nicotine never failed to evoke a response; acetylcholine eventually was inactive. Occasionally, a correlation between dose and response was possible. Adrenaline and noradrenaline also elicited responses, although, occasionally, both were ineffective. Tryptamine and serotonin (less) were also effective. L-Glutamate was particularly effective and its action could be depressed by GABA. Histamine, betaine, taurine and aspartate were ineffective. The results are discussed in terms of depolarization due to specific membrane receptors or to unspecific causes.  相似文献

Acontia—nematocyst‐dense, thread‐like extensions of the mesenterial filaments―are the characteristic feature of the actiniarian group Acontiaria. Phylogenetic analyses have shown that acontiate taxa form a clade that also includes some taxa without acontia. We analyse five molecular markers from 85 actiniarians to explore the phylogenetic relationships among families in Acontiaria, including acontiate species assigned to other higher taxa and species without acontia that have been allied to Acontiaria. Based on our results, we redefine the group to accommodate those lineages that have lost acontia, and formalize it as superfamily Metridioidea. Based on stable and well supported clades, we resurrect Phelliidae and Amphianthidae, redefine Kadosactinidae and Actinoscyphiidae, and move two species to new genera: that previously termed Sagartiogeton erythraios belongs in Jasonactis gen. nov.; and that previously termed Anthosactis pearseae belongs in Ostiactis gen. nov., type genus of Ostiactinidae fam. nov. We also synonymized Halcampoididae and Halcampidae (as Halcampidae) and Andvakiidae and Isophelliidae (as Andvakiidae). The results of our phylogenetic analyses indicate that the diagnostic morphological characters used in the family‐level taxonomy of acontiate actiniarians such as the nematocysts of the acontia, the marginal sphincter muscle, and mesenteries divisible into macro‐ and micro‐cnemes, have to be revisited, as these features are highly homoplasious.  相似文献

Scanning and transmission electron microscopy of the pharynx of the sea anemone Aiptasia pallida revealed a heavily ciliated epidermis and two types of gland cells not known previously to be innervated. By tracing serial cross sections of the pharynx, we located and characterized two types of neuroglandular synapses (i.e., those having clear vesicles and those with dense-cored vesicles). The diameters of the vesicles at each synapse were averaged; clear vesicles ranged from 70 to 103 nm in diameter and were observed at synapses to both mucous and zymogenic gland cells. Dense-cored vesicles ranged from 53 to 85 nm in diameter and were observed at synapses to two mucous gland cells. One mucous gland cell had three neuroglandular synapses, one with clear vesicles and two with dense-cored vesicles. The occurrence of either clear or dense-cored vesicles at neuroglandular synapses suggests that at least two types of neurotransmitter substances control the secretion of mucus in the sea anemone pharynx. To date, only clear vesicles have been observed at a neurozymogenic gland cell synapse in the pharynx. No evidence of immunoreactivity to phenylethanolamine-N-methyl transferase was observed at neuroglandular synapses, suggesting that adrenaline is not a transmitter in the pharynx of A. pallida.  相似文献

The toxicity of Cnidaria is a subject of high concern owing to its influence on human activities and public health and to the implications in ecological relationships between organisms. In order to simulate as much as possible the natural conditions and the consequences of stinging against injured cells, as well considering that Cnidarian venoms are located not only in nematocysts but also in tissues, the cytotoxicity and genotoxicity of crude extracts from nematocyst and surrounding tissues of the sea-anemone Actinia equina (Cnidaria: Anthozoa) were assessed on cultured fibroblasts (V79). The cytotoxicity was remarkable and cell survival was highly reduced at highest tested concentration (150,000 nematocysts/ml); using the Trypan blue dye exclusion test, only 40% of treated cells survived after the first hour of treatment and viable cells were not counted after the second hour. Also the results obtained utilizing the Neutral Red assay showed serious effects of crude toxin (only 31.5% and 12.6% cell survival after one and three hours respectively). Genotoxic effects were not observed at tested doses.  相似文献

Sea anemones feed by discharging nematocysts into their prey, but the pathway for control of nematocyst discharge is unknown. The purpose of this study was to investigate the ultrastructural evidence of neuro-nematocyte synapses and to determine the types of synaptic vesicles present at different kinds of nematocyst-containing cells. The tip and middle of tentacles from small specimens of Aiptasia pallida were prepared for electron microscopy and serial micrographs were examined. We found clear vesicles in synapses on mastigophore-containing nematocytes and dense-cored vesicles in synapses on basitrich-containing nematocytes and on one cnidoblast with a developing nematocyst. In addition, we found reciprocal neuro-neuronal and sequential neuro-neuro-nematocyte synapses in which dense-cored vesicles were present. It was concluded that : (1) neuro-nematocyte synapses are present in sea anemones, (2) different kinds of synaptic vesicles are present at cells containing different types of nematocysts, (3) synapses are present on cnidoblasts before the developing nematocyst can be identified and these synapses may have a trophic influence on nematocyst differentiation, and (4) both reciprocal and sequential synapses are present at the nematocyte, suggesting a complex pathway for neural control of nematocyst discharge. beta-N-Acetylhexosaminidases (EC 3.2.1.52) belong to an enzyme family that hydrolyzes terminal beta-d-N-glucosamine and beta-d-N-galactosamine residues from oligosaccharides. In this report, we purified a novel beta-N-acetylhexosaminidase (Pcb-NAHA1) from the marine zoanthid Palythoa caribaeorum by applying ammonium sulfate fractionation, affinity chromatography on a chitin column, followed by two rounds of size exclusion chromatography. SDS-PAGE analysis indicated a single band protein of apparent homogeneity with a molecular mass of 25kDa. The purified enzyme preferentially hydrolyzed p-nitrophenyl-2-acetoamide-2-deoxyamide-2-deoxy-beta-d-N-acetylglucosamide (pNP-GlcNAc) and to a lesser extent p-nitrophenyl-2-acetoamide-2-deoxyamide-2-deoxy-beta-d-N-acetylgalactosamide (pNP-GalNAc). Detailed kinetic analysis using pNP-GlcNAc resulted in a specific activity of 57.9 U/mg, a K(m) value of 0.53 mM and a V(max) value of 88.1 micromol/h/mg and k(cat) value of 0.61s(-1). Furthermore, purified Pcb-NAHA1 enzyme activity was decreased by Hg Cl(2) or maltose and stimulated in the presence of Na(2)SeO(4,) BaCl(2), MgCl(2,) chondroitin 6-sulfate, and phenylmethylsulfonylfluoride. The optimum activity of Pcb-NAHA1 was observed at pH 5.0 and elevated temperatures (45-60 degrees C). Direct sequencing of proteolytic fragments generated from Pcb-NAHA1 revealed remarkable similarities to plant chitinases, which belong to family 18, although no chitinase activity was detected with Pcb-NAHA1. We conclude that beta-N-acetylhexosaminidases, representing a type of exochitinolytic activity, and endo-chitinases share common functional domains and/or may have evolved from a common ancestor.  相似文献
