由黄单胞杆菌水稻致病变种Xanthomonas oryzae pv.oryzae(Xoo)引起的白叶枯病是水稻重要细菌性病害之一。迄今,已有7个水稻白叶枯病抗性基因被克隆。Xa21是第一个被克隆的白叶枯病抗性基因,因具有广谱抗性而受到广泛的关注。对Xa21的发现、定位及克隆、表达特征、编码产物XA21的生化特性、作用与调控以及XA21介导的免疫反应模式等方面的研究结果进行综述,并对今后的研究方向进行展望。  相似文献

本文研究了粳稻感病品种沈农1033分别与7个抗白叶枯病的品种(其中汕稻有75-34,辛尼斯,1R20;粳稻有早生爱国3号,邳早15,蚌珠芒,爪哇14)杂交,F花粉培养并观察了花粉株系第二代(H2)的抗性转移及遗传表现和花粉株系第三代,第四代抗性稳定性,572个株系的性状是整齐一致的,占95.5%,其中抗病(高抗和中抗)株系有376个,占62.9%,感病系有196个,占32.7%,表现有分离的杂合系约4.5%,不同组合的H2与F2群体中抗,感反应变异幅度不同,对同一组合不同熟期花粉株系其生育日数与抗病性间存在着极显著相关性,生育期长的株系抗性较强。花粉再生植株后代H2在生育期,株高,穗长,粒重等性状上十分多样,白叶枯病抗性遗传分析结果表明,在4个组合中抗性均由1个显性上位基因控制,抗病性广义遗传力在80%以上。杂种花粉植株第二,第三,第四代的农艺性状以及白叶枯病的抗性持续地稳定,不因世代的增加其性状的整齐度不降,抗性减弱。  相似文献

旨在从含有疣粒野生稻抗白叶枯病基因的新种质SH5、SH76基因组中克隆抗病基因。利用RGAs法得到1个NBS-LRR类同源基因,暂命名为SHNLR(登录号为JF934724)。结果表明,SHNLR的开放阅读框长度为3 105 bp,编码1 034个氨基酸,含有CC、NB-ARC与LRR结构域,具备CC-NBS-LRR类植物抗病基因的结构特征。BLASTn和BLASTp比对显示SHNLR是单拷贝基因,未发现同源性较高且功能已知的基因,仅NBS保守域序列与番茄Prf基因的相似度最高。对SHNLR基因电子定位,发现其位于水稻第11号染色体的长臂末端,但与11号染色体上已定位或克隆的8个白叶枯病抗性基因具有不同序列或处于不同的位置。半定量RT-PCR分析表明,SHNLR在抗病新种质叶片中的表达明显受到白叶枯病菌Zhe173的诱导。因此推测SHNLR可能是1个与抗白叶枯病相关的R基因。  相似文献

利用农杆菌介导的高效遗传转化系统,将白叶枯病抗性基因Xa21转入黄淮稻区主栽品种豫粳6号的胚性愈伤组织,获得转基因植株,GUS染色和PCR分析证明Xa21基因已整合到水稻基因组中,其自交T1代植株经GUS染色和白叶枯病接种鉴定呈现3:1分离,研究为培育抗白叶枯病水稻品种奠定了基础。  相似文献

【目的】水稻白叶枯病是一种严重危害水稻的细菌性病害,培育抗性品种是治理该病害的重要途径。因此,明确云南水稻材料对检疫性病害水稻白叶枯病的抗性,可以为该病害的防治与监测、水稻栽培的合理布局和良好抗性资源的获取提供依据。【方法】采用剪叶接种法测定云南稻区30个品种对7个不同致病型白叶枯病菌的抗性。【结果】在供试的30个云南水稻品种中,2个品种(玉粳16和JS42糯稻)对7个不同致病型菌株均表现为抗性;15个品种对7个致病型菌株均表现感病;对HEN11、SCYC-6、YN7、YN11、FUJ、YN241和PX099等7个致病型菌株表现抗性的水稻品种分别占26.67%、16.67%、23.33%、13.33%、6.67%、10.00%和20.00%。此外,区试材料的抗性比例高于主栽品种,地方稻未发现抗性品种。【结论】现在生产上的大部分水稻品种对优势致病型病原菌入侵的抵抗能力降低甚至丧失。针对云南地区的优势致病小种FUJ筛选得到2个抗性品种:玉粳16和JS42糯稻。  相似文献

从全国各病区筛选的19个代表菌株,在三套(中国,日本和IRRI)16个鉴别品种上进行测试,获得4个病情指标,即病斑长度,病组,日增速率和日增百分率,采用不同的的病情指标作方差分析,所得结果不完全一致;对4个病情指标进行相关及回归分析,结果表明,病情指标之间是存在显著的线性相关,且日增百分率为日增速率与病叶长度之商,这种相关性在主成分分析中也得到了证实,通过聚类分析发现,病斑长度不能与其它3个指标归  相似文献

体细胞突变体HX-3抗水稻白叶枯病基因的鉴定   总被引:7,自引:0,他引:7  
以感病杂交稻恢复系明恢63的成熟胚为外植体,利用离体筛选技术获得了抗水稻白叶枯病细胞突变体HX-3。连续8年以我国长江流域白叶枯病代表菌析浙173(IV型)对HX-3的R1到R9代进行抗性鉴定,HX-3的抗病性可以稳定遗传。抗性遗传分析表明HX-3的抗性由1对显性核基因控制。1999~2000年连续两年利用我国、菲律宾和日本的32个水稻白叶枯病菌株,测定HX-3及IRBB1等13个具不同显性抗病基因的近等基因系抗性,HX-3抗谱广,且与已知显性抗病基因的抗谱不同。在此基础上,以抗白叶枯病近等基因系IRBB4、IRBB7、CBB12和IRBB21和HX-3杂交,进行等位性分析,4个杂交组合的F2代均出现抗、感分离,说明HX-3与这4个基因不等位。综合以上研究结果,HX-3具有1个新的抗白叶枯病基因,暂命名为Xa-25(t)。  相似文献

Bacterial blight (BB) caused by Xanthomonas oryzae pv. oryzae (Xoo) is a major disease of rice in the tropics for which genetic resistance in the host plants is the only effective solution. This study aimed at identification of resistance gene combinations effective against Xoo isolates and fingerprinting of the Xoo isolates of Andaman Islands (India). Here, we report the reaction of 21 rice BB differentials possessing Xa1 to Xa21 genes individually and in different combinations to various isolates of pathogen collected from Andaman Islands. Pathological screening results of 14 isolates revealed that among individual genes tested across 2 years, Xa4, Xa7 and Xa21 conferred resistance reaction across all isolates, whereas among combinations, IRBB 50 (Xa4 + xa5), IRBB 52 (Xa4 + Xa21) and IRBB 60 (Xa4 + xa5 + xa13 + Xa21) conveyed effective resistance against tested isolates. The nature of genetic diversity among four isolates selected on the basis of geographical isolation in the islands was studied through DNA finger printing. The RAPD primers S111, S119, S1117, S1109, S1103, S109 and S105 were found to be better indicators of molecular diversity among isolates than JEL primers. The diversity analysis grouped 14 isolates into three major clusters based on disease reaction wherein isolate no. 8 was found the most divergent as well as highly virulent. The remaining isolates were classified into two distinct groups. The importance of the study in the context of transfer of resistance gene(s) in the local cultivars specifically for tropical island conditions is presented and discussed.  相似文献

利用“Lemont/特青”重组自交系(RI)群体研究了水稻对白叶枯病致病菌株CR6的水平抗性。双亲和F1均为感病,重组自交系(RILs)的病斑长度(LL)为带有明显双向超亲的连续变异,显示出典型的多基因遗传特征。部分重组自交系(约占总数90%)对CR6表现高水平抗性(LL≤3cm)。利用由178个良好分离的RFLP标记构建的饱和连锁图,鉴定出11个数量形状位点(QTLs)和3对互作位点解释了RI群体的大部分病斑变异。抗性QTLs定位于水稻第2、3、4、8、9、10、11、12等8条染色体。在来自特青的Xa-4位点上检测到一个有很大加性效应的QTL。其余10个QTLs的抗性等位基因有7个来自特青,3个来自Lemont。研究结果表明多个数量性状位点和失效主基因(Xa-4)残效的累加效应构成了对白叶枯病水平抗性的遗传基础,是重要的抗性组成部分。可以预期在DNA标记的辅助下,这些数量性状位点与主效抗性基因的组合将使水稻品种具有持久抗病性。  相似文献

Two hundred and eighty‐five isolates of Xanthomonas oryzae pv. oryzae were randomly collected from 22 rice‐growing provinces in China. Ninety‐one representative isolates were chosen to assess the differential characteristics of 24 near‐isogenic rice lines containing a single resistance gene or two to four genes. Most isolates were avirulent on pyramided lines, except IRBB51, and hence, the pyramided lines cannot be used as differentials for the virulence analysis of X. oryzae pv. oryzae in China. The 13 rice lines with a single gene were used further to establish a system of races classification of X. oryzae pv. oryzae in China. IR24 and IRBB10 were susceptible to the isolates with several exceptions, whereas IRBB5, IRBB7 and IRBB21 were resistant. Based on the interactions between the isolates of X. oryzae pv. oryzae and the 13 near‐isogenic rice lines, six single‐gene rice cultivars (IRBB5, IRBB13, IRBB3, IRBB14, IRBB2 and IR24) were chosen as differentials, and the 285 tested isolates were classified into nine races. The reaction patterns of the nine races in order were: RRRRRR, RRRRRS, RRRRSS, RRRSSS, RRSSSS, RSRRRS, RSSRRS, RSSSSS and SSSSSS. The race frequencies were 10.18%, 10.53%, 4.91%, 10.18%, 24.21%, 5.96%, 11.23%, 22.46% and 0.35% respectively. The virulence of representative strains of eight Philippine races on 13 rice lines with a single gene was determined and compared with the Chinese races. The frequency distributions of X. oryzae pv. oryzae races were primarily described for the different regions in China.  相似文献

类Tubby蛋白质(Tubby-like protein,TLP)在动植物中广泛存在,暗示其在生命过程中发挥重要的作用。水稻(Oryza sativa)基因组中有14个TLP家族成员,首先制备了这些蛋白质的抗体,用免疫印迹方法检测了它们在水稻叶片不同生长时期的表达情况,揭示其表达模式;然后对Xa21介导的水稻白叶枯病抗性反应不同时间点进行检测,发现OsTLP2、OsTLP7、OsTLP8和OsTLP9等4个蛋白质的表达发生了变化;进一步比较它们在抗病、感病反应和对照处理中的表达情况,发现不同反应间的表达也有区别。该研究结果为阐释水稻TLP在叶片生长过程中的功能,尤其是在水稻-白叶枯病菌互作过程中的作用提供了重要线索。  相似文献

对植物病原菌的分型是生物技术应用的新领域,水稻白叶枯病菌(Xanthomonas oryzae pv. Oryzae)是亚洲地区水稻的主要病害。前人曾根据不同菌株在5个鉴别品种上的反应型将中国白叶枯病菌分为7个致病型⑴。近来,Leach等⑵已开始利用基因组特异的重复顺序对白叶枯病菌株进行DNA指纹分析,章琦等⑶也利用相同的探针对我国部分菌株进行了RELP分型,其结果与传统病理学的分类结果有相符之处.但又有相当的不同,说明传统分类结果尚有许多值得探索之处。与RFLP相比,RAPD(Random Amplified Polymorphic DNA)具有简便和快速的优点,而且相当灵敏,在动植物基因组分型(gennome typing)上已得到较为广泛的应用〔4.5〕。我们利用RAPD标记对我国28个菌株进行分型,发现随机引物A-14可用以区分所研究的28个菌株。进而对扩增带型进行了相似百分率分析,并利用另一随机引物A一1 3作了进一步分析。本文是用RAPD方法尝试对部分水稻白叶枯病菌分型的首次报道。  相似文献

水稻白叶枯病抗性基因Xa4已被定位于第11染色体长臂末端的分子标记VG181和L1044之间,并与抗性基因同源序列片段RS13共分离。利用这3个标记筛选IRBB56的BAC文库,共得到128个阳性BAC克隆,其中RS13获得18个阳性克隆,这18个克隆中有4个和6个我隆分别同时为G181和L1044的阳性克隆,选其中的12克隆进行分析,构建了一个从G181到L1044区间的BAC跨叠克隆,全长420kb,并且56M22、106P13和104B153个BAC克隆可覆盖整个跨叠克隆群。这一研究结果为进一步分离Xa4基因打下基础。  相似文献

Virulence analysis and two polymerase chain reaction–based assays were used to evaluate the population structure of Xanthomonas oryzae pv. oryzae (Xoo) from different elevations ranging from 150 to 2600 m in south‐west China. Among the 218 isolates of Xoo, 18 pathotypes were identified using six near‐isogenic rice lines, each containing a single resistance gene. Among them, pathotype 9 predominated in low and mid‐elevations was virulent to all resistance genes, including Xa2, Xa3, xa5, xa13, Xa14 and Xa18. However, pathotype 2 was predominant at high elevation and was virulent to Xa18 only. The 18 pathotypes were grouped into four clusters. Isolates belonging to cluster 1 were mainly found at high and mid‐elevations, while those of cluster 4 were mainly found at low elevations. There were significant trends of virulence of isolates from low to high with the elevation from high to low. The ERIC and J3 primers were used to screen the genomes of 218 isolates, and 56 molecular haplotypes were found. Multiple correspondence analyses revealed that 56 haplotypes were divided into four putative genetic lineages. Lineage 2 was the most frequently detected from 150 to 2600 m; it was clearly shown that isolates from high elevation with 80% is much more than from low and mid‐elevation in the lineage. It is intriguing that genetic variation of Xoo is restricted by physical geographical barriers of elevations. This is the first report on the relationship of pathotypic and genotypic diversity of Xoo at different elevations.  相似文献

The agronomically important Indica (group 1) rice varieties IR64, IR72, hybrid restorer line Minghui 63, and BG90-2 were co-transformed by microbombardment of embryogenic suspensions with plasmids that contain the Xa21 gene which confers resistance to Xanthomonas oryzae pv. oryzae and the hph gene for resistance to hygromycin B. Six of the 55 transgenic R0 plant lines containing the Xa21 gene displayed high levels of resistance to the pathogen, and no partial resistance was observed. The trait was stably inherited in subsequent generations, and transgenic plants are currently in field tests. The ability to transfer agronomically important genes into elite Indica rice varieties demonstrates the applicability of genetic engineering for the agronomic improvement of rice.  相似文献

为了探讨水稻白叶枯病成株抗性是否与防卫基因相关 ,通过RT PCR研究了几种防卫基因PAL、LOX、PBZ1、Cht 1和PR1a的表达 ,同时也分析了PAL和LOX的酶活性变化 .结果表明 ,苗期PAL受病原菌诱导上调表达 ,而成株期病原菌和伤害均可诱导PAL上调表达 ,且成株期诱导表达强于苗期 .LOX在苗期和成株期都可受病原菌诱导表达 ,但成株期表达强于苗期且更持久 .PBZ1在苗期和成株期均受病原菌和伤害诱导表达 ,但成株期表达较苗期早且更强 .PR1a和Cht1没检测到期望的扩增片段 .PAL和LOX的酶活性分析结果与基因表达分析结果一致 .因此 ,PAL、LOX和PBZ1在苗期和成株期的表达差异可能与水稻白叶枯病成株抗性相关 .  相似文献
