More than 600 bacterial strains isolated from different horizons of steppe biogeocenoses and zoogenous loci (diplopod intestines and feces) were tested for the ability to lyse yeast cell walls. About half of the strains that were isolated from biotopes with active degradation of plant debris (steppe litters and diplopod intestines and feces) were found to possess yeast-lytic activity. Most of the yeast-lytic strains belonged to the genera Streptomyces, Promicromonospora, Oerskovia, and Agromyces. The yeast-lytic activity of actinobacteria from the genera Agromyces, Mycobacterium, and Micrococcus has not previously been reported.  相似文献

Heat-resistant strains of Clostridium perfringens were isolated from normal feces samples of Japanese people more frequently than from European people. However, the predominant number of the C. perfringens cells in the human intestines were sensitive to the heat at 100 C for 10 min. This proved to be true of the feces samples which were demonstrated to carry the organism resistant to 100 C for 60 min. Further studies indicated that the more the number of the organism in the intestines, the higher was the recovery ratio of the heat-resistant strains. Concomitant estimation for α-toxigenicity of those strains isolated from unheated feces samples revealed that the α-toxigenicity ranged mainly between 0.05 and 0.9 α-antitoxin equivalents.  相似文献

The bacterial communities in the food, intestines, and feces of earthworms were investigated by PCR-denaturing Gradient gel electrophoresis (DGGE). In this study, PCR-DGGE was optimized by testing 6 universal primer sets for microbial 16S rRNA in 6 pure culture strains of intestinal microbes in earthworms. One primer set effectively amplified 16S rRNA from bacterial populations that were found in the food, intestines, and feces of earthworms. Compared with the reference markers from the pure culture strains, the resulting DGGE profiles contained 28 unique DNA fragments. The dominant microorganisms in the food, intestines, and feces of earthworms included Rhodobacterales bacterium, Fusobacteria, Ferrimonas marina, Aeromonas popoffii, and soil bacteria. Other straisn, such as Acinetobacter, Clostridium, and Veillonella, as well as rumen bacteria and uncultured bacteria also were present. These results demonstrated that PCR-DGGE analysis can be used to elucidate bacterial diversity and identify unculturable microorganisms.  相似文献

The assimilation of sodium cyclamate (CHS-Na) by microorganisms was studied. Fifteen strains of cyclamate-assimilating bacteria were isolated from the feces of guinea pig excreting cyclohexylamine (CHA) in urine. The majority of the strain isolated seems to belong to the genera Pseudomonas and Corynebacterium. All strains were able to assimilate CHS-Na as a sole source of carbon and nitrogen, and accumulated clearly CHA in the culture medium. It was confirmed with the cell-free extract that the strains possessed the enzyme system which formed cyclohexanone (CHnone) from CHS-Na via CHA. It seems that the desulfation of CHS-Na to CHA is catalyzed by hydrolase, and that the deamination of CHA to CHnone is catalyzed by amine oxidase depending on oxygen.  相似文献

To isolate actinobacteria from ruminant feces and elucidate their correlations with ruminants, the actinobacterial community in sheep (Ovis aries) and cattle (Bos taurus) feces was determined by cultivation and clone library methods. Most of actinobacteria isolated belonged to Streptomyces, Amycolatopsis, Micromonospora, and Cellulosimicrobium genera. The strains showed above 99 % similarity with the type strains, respectively. All the strains isolated could grow on media containing pectin, cellulose, or xylan as the sole carbon sources. However, most antibacterial and antifungal activities were found in Streptomyces species. Clone library analysis revealed that the genera Mycobacterium, Aeromicrobium, Rhodococcus, Cellulomonas were present in cattle and sheep feces. In contrast, the 16S rRNA genes showed less than 98 % similarity with the type strains. The analysis of actinobacterial community in ruminant feces by clone library and cultivation yielded a total of 10 actinobacterial genera and three uncultured actinobacterial taxa. The ruminant feces harbored diverse actinobacterial community. Ruminants may represent an underexplored reservoir of novel actinomycetes of potential interest for probiotics and drug discovery.  相似文献

新疆地区为控制棉花连作引发的棉蚜等害虫规模性爆发,长期大量使用了吡虫啉等农药,为获得适合当地气候土壤环境条件下,降解吡虫啉的微生物资源,以吡虫啉为唯一碳源从新疆棉花长期连作土壤中富集降解菌群,通过高通量测序分析其结构组成,利用多种常规培养基和通过高通量测序结果设计的培养基从吡虫啉降解菌群(BCL)中挖掘细菌资源。结果表明,菌群在门分类水平上主要由变形菌门(Proteobacteria,67.05%);放线菌门(Actinobacteria,10.67%)、厚壁菌门(Firmicutes,9.99%)、绿弯菌门(Chloroflexi,4.1%)、酸杆菌门(Acidobacteria,2.84%)等组成;在属分类水平,占比最多的依次为Pseudomonas(16.33%)、Moraxella(11.14%)、Escherichia(4.57%)、Brochothrix(2.19%)等,未能分类的菌属占58.13%。通过基础无机盐等常规培养基分离出BP2、BP5、BP8、BG5、BJ7、BJ17、BJW8等48株菌株,分别隶属于为Shinella、Nocardioides、Agromyces、Sphingopyxis、Bacillus、Cellulosimicrobium、Bosea等属。经Trace element solution和 Vitamin solution培养基分离获得了BMV3、BMV5、BMV7.1、BMV14.1、BLE1、BLE3.1、BLE4、BLE5、BLE10.1等25株菌株,分别隶属于Paenibacillus、Ammoniphilus、Planococcus、Brevibacillus、 Paenibacillus、Bacillus、Sphingopyxis、Rhodococcus、Nocardioides等属。使用16S rRNA序列对比分析,BP5与Nocardioides nitrophenolicus NSP 41同源性达98.08%,BMV5与Ammoniphilus resinae CC-RT-E同源性达98.5%,其他菌株同源性介于98.29%~100%之间。可见依据高通量测序结果设计培养基,可以有针对性地从源自新疆棉区土壤的吡虫啉降解菌群中分离出一些低丰度菌属。在摸索分离新疆特殊环境微生物资源的同时也能为该地区生物修复农药污染土壤提供参考。  相似文献

The diversity and structure of the intestinal microbial community has a strong influence on life history. To understand how hosts and microbes interact, model organisms with comparatively simple microbial communities, such as the fruit fly (Drosophila melanogaster), offer key advantages. However, studies of the Drosophila microbiome are limited to a single point in time, because flies are typically sacrificed for DNA extraction. In order to test whether noninvasive approaches, such as sampling of fly feces, could be a means to assess fly-associated communities over time on the same cohort of flies, we compared the microbial communities of fly feces, dissected fly intestines, and whole flies across three different Drosophila strains. Bacterial species identified in either whole flies or isolated intestines were reproducibly found in feces samples. Although the bacterial communities of feces and intestinal samples were not identical, they shared similarities and obviously the same origin. In contrast to material from whole flies and intestines, feces samples were not compromised by Wolbachia spp. infections, which are widespread in laboratory and wild strains. In a proof-of-principle experiment, we showed that simple nutritional interventions, such as a high-fat diet or short-term starvation, had drastic and long-lasting effects on the micobiome. Thus, the analysis of feces can supplement the toolbox for microbiome studies in Drosophila, unleashing the full potential of such studies in time course experiments where multiple samples from single populations are obtained during aging, development, or experimental manipulations.  相似文献

产志贺毒素大肠杆菌(Shiga toxin-producing Escherichia coli,STEC)是重要的食源性病原,而STEC往往以正常菌群的形式存在于牛羊等反刍动物肠道。[目的] 本研究对牛羊粪便样品中的STEC分离和鉴定并对分离株进行致病潜力分析。从江苏、云南和河北等地共分离到羊源STEC菌株11株,牛源STEC菌株1株,另新疆农业大学佟盼盼组馈赠牛源菌株10株。[方法] 通过细菌选择培养及特异性基因stx1stx2的检测进行分离鉴定;并通过Vero细胞毒性试验、溶血活性试验和毒力因子的检测分析STEC分离株的致病潜力。[结果] 分离到羊源分离株11株,分离率17.5%(11/63);分离得到牛源分离株1株,分离率0.7%(1/134);11株羊源分离株中有5株对Vero细胞具有强的毒性,3株有溶血活性;11株牛源分离株中有5株对Vero细胞具有强的毒性,3株有溶血活性。11株羊源STEC分离株eae基因携带率为63.6%(7/11),而11株牛源STEC分离株eae基因携带率仅为9.0%(1/11)。[结论] 结果表明羊源STEC菌株的分离率和致病潜力高于牛源菌株,所以,除牛外,羊作为STEC菌株宿主也应该得到更多的重视。  相似文献

In order to study E. coli aerosol spreading from chicken houses to their surrounding air, air samples, including indoor and outdoor air (upwind 10 and 50 m as well as downwind 10, 50, 100, 200 and 400 m away) of 5 chicken houses were collected using six-stage Andersen microbial samplers and Reuter-Centrifugal samplers (RCS). E. coli concentrations (CFU/m3 air) collected from different sampling sites were calculated. E. coli strains from chicken feces samples were also isolated. Furthermore, the enterobacterial repetitive intergenic consensus (ERIC)-PCR method was applied to amplify the isolated E. coli strain DNA samples. Through the genetic similarity analyses of the E. coli obtained from different sampling sites, the spreading of bioaerosol from animal houses to the ambient air was characterized. The results showed that the isolated E. coli concentrations in indoor air (9―63 CFU/m3) in 5 chicken houses were higher than those in upwind and downwind air, but there were no significant differences between the indoor and downwind sites 10 m away from all the 5 houses (P>0.05). The phylogenetic tree indicated that a part of the E. coli (34.1%) isolated from indoor air had 100% similarity with those isolated from feces, and that most of E. coli isolated (54.5%) from downwind at 10, 50, 100 or even 200 m had 100% similarity with those isolated from indoor air or feces too. But those isolated from upwind air had a lower similarity (73%―92%) with corresponding strains isolated from indoor air or feces. Our results suggested that some strains isolated from downwind air and indoor air originated in the chicken feces, but most of isolates obtained from upwind air samples did not come from the chicken feces or indoor air. Effective hygienic measures should be taken in animal farms to prevent or minimize downwind spreading of microorganism aerosol.  相似文献

为了解喀斯特典型物种-小蓬竹根际土壤微生物及不同部位内生真菌多样性,采用沿等高线等距离取样法采集小蓬竹根际土壤及健康植株,通过可培养对根际土微生物及内生菌进行分离,利用分子技术对其进行鉴定,根据鉴定结果构建系统发育树,并计算小蓬竹根际土壤微生物和根茎叶内生真菌多样性。结果如下:(1)共从根际土壤、根、茎、叶分离得到139个真菌菌株,隶属于27属,其中根际土壤分离得到34个真菌菌株隶属于12属,根部分离得到的63个内生真菌菌株隶属于17个属,茎部分离得到的14个内生真菌菌株隶属于8个属,叶部分离得到28个内生真菌菌株隶属于9个属;(2)根际土壤共分离得到41株细菌菌株,隶属于7个属26个种,20株放线菌菌株,隶属于1属15种;从Shannon-Wiener多样性指数、均匀度指数、Simpson指数排序来看,真菌主要表现为根 > 根际土壤 > 茎 > 叶,细菌和放线菌多样性均较低。(3)按层次聚类分析可分别将真菌、细菌、放线菌聚为3支。小蓬竹根际土壤、根、茎和叶具有丰富的微生物多样性,不同部位菌群组成存在差异性(P<0.05),且存在以假单胞菌属、芽孢杆菌属等为优势属的抗盐耐旱菌群,这有助于揭示小蓬竹对喀斯特生境的适应性,以及为微生物-植物群落之间相互关系提供一定基础数据,为后期寻找小蓬竹相关耐性功能菌奠定基础。  相似文献

By using fosmidomycin and mevinolin (inhibitors of the synthesis of isoprenoid pigments), spectrophotometry, and mass spectrometry, the presence of isoprenoid pigments is shown in 71 of the 78 strains under study. All of these strains belong to 11 genera of the family Microbacteriaceae. Yellow, orange, and red pigments are found to have absorption spectra typical of C40-carotenoids. Eight out of the sixteen strains of the genus Microbacterium are able to synthesize neurosporene, a precursor of lycopene and β-carotene. The biosynthesis of carotenoids in some representatives of the genera Agromyces, Leifsonia, and Microbacterium is induced by light. Inhibition of the biosynthesis of isoprenoid pigments by fosmidomycin suggests that they are synthesized via the nonmevalonate pathway. Twelve strains are found to exhibit both the nonmevalonate and mevalonate pathways of isoprenoid synthesis. About 2700 strains of various kinds of microorganisms isolated from soil were tested for phospholipase D activity. All of the 200 selected strains were actinomycetes which were divided into two groups on the basis of the isomeric form of diaminopimelic acid (A2pm) present in the cell wall. Among 18 strains containing meso-A2pm, two strains were selected on the basis of taxonomical characteristics and productivity of phospholipase D. These two strains were examined for microbiological characteristics and found to belong to the genera Actinomadura and Nocardiopsis.  相似文献

【背景】由于滥用抗生素导致细菌耐药性日益严重。对于双歧杆菌,人们往往注重其益生功能的挖掘而忽视了对其耐药性的研究,存在一定的安全隐患。【目的】检测母婴肠道中假小链双歧杆菌的耐药性,探究婴儿肠道中假小链双歧杆菌耐药性的来源。【方法】利用微量肉汤稀释法测定48株分离自母婴肠道的假小链双歧杆菌对14种抗生素的耐药性,比较分离自不同家庭母婴肠道中假小链双歧杆菌的耐药性。【结果】48株母婴肠道分离株对四环素、氯霉素、新霉素、环丙沙星100%耐药,对其余10种抗生素耐药率依次为:卡那霉素98%、利福平80%、克林霉素78%、甲氧苄啶63%、红霉素59%、庆大霉素43%、链霉素16%、万古霉素14%、氨苄西林6%、利奈唑胺2%。母婴肠道分离株的耐药性无显著差异,分离自同一家庭母婴肠道的菌株具有相似的耐药表型。【结论】分离自母婴肠道的假小链双歧杆菌对多种抗生素具有耐药性,婴儿肠道中假小链双歧杆菌的耐药性可能是由母亲肠道垂直传递而来。  相似文献

Phylogenetic diversity of culturable bacteria from genus Bacillus and related genera, isolated from 18 Bulgarian hot springs was investigated in association with their functional diversity. Sixty-seven thermophilic and facultative thermophilic strains were isolated under aerobic conditions at 60°C. Sixty-six of them belonged to eight species in four genera from Bacillus group: Anoxybacillus, Geobacillus, Brevibacillus and Bacillus. Representatives of the genus Anoxybacillus predominated. Based on phylogenetic analysis (<97% sequence similarity) four strains belonged to groups representing potentially novel species. Producers of carbohydrases, degrading 12 from the tested 13 substrates were isolated. About half of the isolates degraded amylose by exo- or endo-mechanism of action of their enzymes. The isolates degrading hemicellulose carbohydrates like arabinan, arabinoxylan, β-glucan, galactan, galactomannan and xyloglucan were reached to. Some of the microorganisms were able to uptake microbial polysaccharides like curdlan and gellan and their enzymes were between first reported thermostable enzymes in their groups, like gellan lyase and curdlan lyase A relation between species affiliation and their functional activity was observed—all A. gonensis strains were producer of amylolytic enzymes, most of Brevibacillus ruber strains were able to grow in a minimal medium with xanthan.  相似文献

A nitrogen-fixing strain identified as Klebsiella pneumonia 402-2 and two endoglucanase-synthesizing Bacillus strains were isolated from the intestines of phytophagous animals. One of the Bacillus strains was identified as Bacillus subtilis GL. Klebsiella pneumoniae 402-2 increased the endoglucanase activity of both Bacillusstrains in mixed cultures.The data on the taxonomic position of strains 402-2 and GL and on the nitrogen-fixing capacity of strain 402-2 were confirmed by sequencing and analyzing their 16S rRNA genes and by amplifying the nitrogenase gene nifH.  相似文献

2005年9月15日—30日,在内蒙古锡林郭勒盟西乌珠穆沁旗的典型草原上研究了狭颅田鼠(Microtus gregalis)冬季集群与来自艾虎(Mustela eversmanni)的捕食风险。采用鼠洞口数量作为狭颅田鼠集群大小的指标,分析了艾虎对不同大小狭颅田鼠集群的捕食优先选择偏好。对实验样地内狭颅田鼠的洞群洞口数进行了调查统计(样地内总共涉及102个狭颅田鼠有效洞群),并计数此期间的艾虎粪便堆数以及攻击掘开狭颅田鼠洞群的情况。运用非参数的Mann-Whitney U检验法进行数据分析,结果表明:从艾虎遗留的粪便痕迹来看,没有被艾虎访问过的田鼠洞群与被艾虎访问过的洞群之间差异达到极显著水平;另外,从狭颅田鼠洞口被艾虎掘开的痕迹来分析,没有被艾虎挖掘的洞群与被艾虎挖掘的洞群之间的差异也达到极显著的水平;另外,随着田鼠洞群洞口数量的增加,出现艾虎粪便和掘洞的频次和概率就越高。表明艾虎对狭颅田鼠集群洞口数的选择差异性非常显著,明显倾向于选择在洞口数量多的狭颅田鼠洞群停留,同时也更倾向于掘开洞口数量较高的狭颅田鼠洞群作为捕食对象。  相似文献

A total of 96 lactic acid bacteria (LAB) were isolated from African indigenous fermented products and cow’s intestines to study their inhibitory capability against multi-drug-resistant uropathogens. Escherichia coli accounted for approximately 45% of isolated uropathogens, followed by Staphylococcus spp. (20%). The Gram negative uropathogens were highly resistant to quinolones, co-trimoxazole, teicoplanin and some β-lactams, while the Staphylococcus spp. showed high resistance to aminoglycosides, β-lactams and macrolides. Twenty-four LAB isolates were selected based on their antimicrobial activity against two uropathogenic Staphylococcus aureus strains and bacteriocin production. LAB strains showing antimicrobial activity were grouped into smaller groups through amplified ribosomal DNA restriction analysis (ARDRA). Representative strains were identified as Weissella spp., Enterococcus faecium, Lactococcus lactis and Lactobacillus brevis through sequencing of 16S rDNA. The Weissella spp. and L. brevis strains demonstrated remarkable inhibitory activity against seven strains of Gram negative uropathogens. Two strains of L. lactis produced a bacteriocin-like inhibitory substance active against Lactobacillus sakei. In this study, an unusual high rate of co-trimoxazole, quinolones and macrolides resistance among uropathogens from south west Nigeria was discovered. Based on their sensitivity to Weissella spp., there is a potential for using these LAB as a natural approach for the protection against the uropathogens assayed.  相似文献

【目的】研究深圳大鹏半岛海域石磺海牛中可培养的共附生细菌的数量和种类,并对分离获得菌株的代谢产物进行活性筛选。【方法】通过R2A平板培养、分离纯化和16SrRNA测序,分析鉴定石磺海牛中5个部位可培养细菌;使用分离菌株的菌液及发酵液上清,测定对群体感应信号分子降解的活性和抗生物膜活性。【结果】从石磺海牛中共分离到215株细菌,归属于87个种,54个属,有16株菌疑似为新菌。分离获得的菌株分布于变形菌门(Proteobacteria,126株),拟杆菌门(Bacteroidetes,44株),厚壁菌门(Firmicutes,34株),放线菌门(Actinobacteria,10株)和浮霉菌门(Planctomycetes,1株)。石磺海牛卵中的细菌数量和种类最为多样。从分离的菌株中筛选出28株菌能够降解群体感应信号分子,8株菌具有抗生物膜活性。【结论】首次报道了海洋无脊椎动物石磺海牛中具有丰富多样的可培养细菌,包含潜在的新微生物和天然产物资源,为今后研究石磺海牛共附生微生物提供了研究基础和参考。  相似文献

Thirty different fungal strains were isolated from A. tequilana leaves showing disease symptoms such as wilt and curled leaves, black, red and chlorotic spots. Ten genera were identified and confirmed by using the LSU D1/D2 rDNA and ITS1‐5.8S‐ITS2 regions, mainly of the Ascomycota phylum, where the Lasiodiploidia and Neoscytalidium genera were the more (46.6%) abundant. The other genera identified were Cladosporium, Cytospora, Epicoccum, Flavodon, Lasiodiplodia, Myrmaecium, Neoscytalidium, Penicillium, Peniophora, Purpureocillium, Trametes and Fusarium. Five strains of Lasiodiplodia and one of Fusarium were selected based on their representativeness and pathogenic potential on Agaves. Pathogenic potential was analysed by both, an infection assay, evidenced as necrosis, and by pectinolytic activity. Specifically, necrosis infection assay was conducted by puncture (wounded) infection and by direct mycelium contact. In general, Lasiodiplodia strains exhibited different pathogenic profiles according to their necrosis percentages, regardless of the infection method used. Fusarium strain analysed also showed a high necrosis infection (> 99%). Pectinolytic activity used as an indirect measurement of pathogenesis presented a high Fusarium extract activity (peaking at 23.9 U). Lasiodiplodia strains exhibited up 6 times more enzymatic activity (peaking at 143.5) than Fusarium strain analysed. In addition, Agave leaf extracts used totally or partially as carbon source during fungal induction culture may induce different pathogenic activities in these strains. In general, the two pathogenicity assays implemented evidenced differences in the pathogenicity profile of these analysed strains.  相似文献
