Pollen genotype selection for genes expressed in both the haploid and diploid phases of the plant life cycle can lead to correlated responses detectable in the sporophyte. A pollen selection was conducted in two genetic backgrounds of maize (Zea mays L.) for chlorsulfuron resistance, conferred by the XA17 allele. Plants of two backcross (BC) lines segregating 1 (heterozygote, resistant) : 1 (homozygote, susceptible) for chlorsulfuron resistance were used as pollen donor. Selection treatment was applied during microspore development, and tassels were cut about 10 days before anthesis and placed into a liquid medium with or without 40 mg l–1 of chlorsulfuron. Pollen was used to fertilize an unrelated susceptible genotype (tester). The resulting testcrosses (TC) were evaluated in the greenhouse by spraying seedlings with chlorsulfuron at 23 g ha–1. Non-selected TC progenies derived from heterozygous BC plants showed a proportion of resistant and susceptible plants close to the expected 1:1 ratio, while non-selected TC progenies derived from homozygous BC plants showed susceptible plants only. Selected TC progenies derived from heterozygous BC plants showed a frequency of resistant plants ranging from 89% to 100%. BC plants homozygous for the susceptible allele, subjected to selection treatment, gave poor seed set, and no resistant plants were found in their TC progenies. Resistant TC plants obtained through pollen genotype selection were selfed, and the proportion of resistant plants was close to 3:1 in all selfed families, in accordance with the hypothesis that all of them inherited the XA17 allele through selection. Solanum tarnii, a wild diploid, tuber-bearing Mexican species belonging to the series Pinnatisecta is highly resistant to Potato virus Y (PVY) and Colorado potato beetle and shows a strong hypersensitive reaction to Phytophthora infestans. Therefore, it could be a potential source of resistance to pathogens for potato breeders. S. tarnii (2n = 2x = 24) is reproductively isolated from tetraploid Solanum tuberosum and hence difficult to include in potato breeding programmes. In this study, interspecific somatic hybrids were produced for the first time by protoplast electrofusion of the cells of potato cv. Delikat (Solanum tuberosum L.) and Solanum tarnii. The hybrid nature of the regenerants was confirmed by simple sequence repeat (SSR) and amplified fragment length polymorphism (AFLP) markers and by morphological analysis and flow cytometry. Selected somatic hybrids were successfully backcrossed with cv. Delikat. Parental lines, primary somatic hybrids and BC1 progeny were assessed for resistance to PVY by mechanical inoculation, grafting and exposure to viruliferous aphid vectors in the field, and resistance to late blight (P. infestans) by detached leaflet and whole tuber tests. The somatic hybrids showed no symptoms of viral infection and most of them displayed high levels of resistance to foliage blight. The BC1 progenies were highly resistant to PVY and a few were resistant to foliage blight. Selected hybrids and BC1 clones were evaluated in the field for tuber quality and tuber yield. Some BC1 clones produced yields of good quality tubers. The results confirm that both the resistance to PVY and to late blight of S. tarnii is expressed in somatic hybrids, and PVY resistance is transferred to BC1 progeny, whereas blight resistance is harder to transfer. Somatic hybridization again proved to be a valuable tool for producing pre-breeding material with increased genetic diversity.  相似文献

Summary Hybrids between Solanum etuberosum and S. pinnatisectum harboring resistance to titer buildup of potato leafroll virus (PLRV) were reciprocally crossed with tuber-bearing wild species S. acaule and S. verrucosum. A total of 47 hybrids with acaule were obtained with the aid of embryo rescue and sterile culturing of embryos from imbibing seeds. All but two hybrids with acaule had low pollen stainabilities or were pollen sterile. Hybrid seeds from crosses with verrucosum were easily obtained, and the triploid progenies were sterile. Hybrid progeny were screened for resistance to PLRV infection by viruliferous green peach aphid and for resistance to titer buildup. Although hybrids did not exhibit resistance to infection, PLRV was not detectable using ELISA. Virus was detected, however, by graft transmission to Datura stramonium. Crosses of fertile acaule-etuberosum-pinnatisectum hybrids with S. phureja, a cultivated diploid, using the latter as pollen parent, produced berries but seed did not complete development and was aborted. Rescue of immature embryos 25 days after pollination by excision from berries and sterile culture produced vigorous, pot-cultured plants. Segregation of susceptible (virus detected) and resistant (virus undetected) progenies suggests simple inheritance.  相似文献

Interspecific somatic hybrids between the dihaploid Solanum tuberosum and the wild species S. pinnatisectum Dun. were produced via protoplast fusion. Protoplast isolation, electrofusion, culture of post-fusion products and regeneration of calli/shoots were undertaken following optimized protocols. Regenerants were characterized for hybridity, ploidy and resistance to Phytophthora infestans (Mont.) de Bery, causal fungal pathogen of late blight disease. From a total of 126 regenerated macrocalli, 12 somatic hybrids were confirmed by possessing species-specific diagnostic bands of their corresponding parents as revealed by RAPD, SSRs and cytoplasmic-DNA analyses. Tetraploid status of the 12 hybrids was determined using flow cytometry analysis. Intermediate phenotypes for leaf, flower, and tuber characteristics and high male fertility were observed in field-grown hybrid plants. Hybrids were highly resistant to foliage late blight based on field assessment for two seasons. In contrast, moderate level of resistance to foliage blight was observed in hybrids based on the detached leaf assay under laboratory conditions. Overall, somatic hybrids with moderate levels of resistance to foliage blight were identified, and these will be useful for in situ hybridization in potato breeding efforts.  相似文献

A diallel set of crosses, including selfs and some reciprocal crosses, was made between 15 parents chosen for their male fertility from those included in a tetraploid potato (Solanum tuberosum subsp. tuberosum) breeding programme at the Scottish Crop Research Institute. Seedling progeny tests were used to evaluate the progenies for non-race-specific resistance to late blight (Phytophthora infestans) in both foliage and tubers, quantitative resistance to the white potato cystnematode (PCN) (Globodera pallida) and the commercial worth of their tubers as judged by breeders' visual preference. No reciprocal differences were found. Comparisons of the selfs and crosses revealed inbreeding depression for breeders' preference, which varied among the parents from negligible to severe, whilst there were also statistically significant differences for foliage and tuber blight, but not for PCN. When the selfs were omitted from the combining ability analyses, large differences in general combining ability (GCA) were found for all four traits, and smaller differences in specific combining ability for tuber blight and breeders' preference. The only statistically significant correlation between GCAs for different traits was a favourable one of r = 0.56 between foliage and tuber resistance to late blight. It was concluded that prospects were good for simultaneously improving all four traits by multitrait genotypic recurrent selection.  相似文献

Cupressus lusitanica seedlings from open-pollinated seeds of 18 families were inoculated with day-old first instar Cinara cupressi. Aphid survival was used to determine the genetic basis and inheritance of resistance to the insect. There was marked variation in aphid survival both between and within families. An individual-tree narrow-sense heritability of 0.76 ± 0.30 shows strong additive genetic control which could allow effective selection and breeding for resistance. Resistant parents produced resistant progeny while susceptible parents produced susceptible progeny. There were notable exceptions as some susceptible parents produced highly resistant progeny, indicating that they had acquired pollen from resistant neighbours. Recovery of aphid damaged trees is evident implying that care should be taken in selecting for resistance. The strong additive variance and potentially high heritability indicate that one cycle of selection may yield a resistant population, while intraspecific crossing may give better results. Implications of the results in a C. lusitanica breeding programme are discussed.  相似文献

Seeds of cocoa progenies were inoculated with cocoa swollen-shoot virus manually or by vector transfer techniques to assess the relative resistance of the progenies to infection. Assessment of progeny included in both inoculation tests and in field trials were positively correlated, indicating that the inoculation technique is suitable for the selection of progenies for testing in the field. Pure Upper Amazon progenies were more resistant than selfed Amelonado, with hybrids between Amelonado and Upper Amazons usually intermediate. Among the main Upper Amazon populations, Iquitos and Nanay clones were better sources of resistance than Parinari and Scavina clones. A survey of these populations indicated that, within populations, resistance levels do not vary greatly. A range of progenies based on the Upper Amazon female parents in existing seed gardens was screened for resistance and some were consistently more resistant than the equivalent Amazon x Amelonado hybrid now being distributed to farmers.  相似文献

Greenhouse and field evaluations of quantitative resistance to late blight in genotypes of the wild Argentine diploid species Solanum chacoense, Solanum commersonnii, Solanum microdontum and Solanum maglia were performed with a complex race containing the virulence factors 1, 3, 4, 5, 7,10, 11 of Phytophora infestans and using the percentage of leaf area affected by late blight estimated at weekly intervals. From these readings the area under the disease progress curve (AUDPC) was calculated. Highly resistant and susceptible genotypes were identified. Resistance and variability were found in S. chacoense, which together with S. commersonnii showed a significantly higher level of resistance compared to the susceptible checks. Field evaluations of an F1 progeny of 165 individuals, obtained from non-inbred diploid parents segregating for foliage quantitative resistance in S. chacoense, were also done by means of AUDPC. Summary Oryza minuta J. S. Presl ex C. B. Presl is a tetraploid wild rice with resistance to several insects and diseases, including blast (caused by Pyricularia grisea) and bacterial blight (caused by Xanthomonas oryzae pv. oryzae). To transfer resistance from the wild species into the genome of cultivated rice (Oryza sativa L.), backcross progeny (BC1, BC2, and BC3) were produced from interspecific hybrids of O. sativa cv IR31917-45-3-2 (2n=24, AA genome) and O. minuta Acc. 101141 (2n=48, BBCC genomes) by backcrossing to the O. sativa parent followed by embryo rescue. The chromosome numbers ranged from 44 to 47 in the BC1 progeny and from 24 to 37 in the BC2 progeny. All F1 hybrids were resistant to both blast and bacterial blight. One BC1 plant was moderately susceptible to blast while the rest were resistant. Thirteen of the 16 BC2 progeny tested were resistant to blast; 1 blast-resistant BC2, plant 75-1, had 24 chromosomes. A 3 resistant: 1 susceptible segregation ratio, consistent with the action of a major, dominant gene, was observed in the BC2F2 and BC2F3 generations. Five of the BC1 plants tested were resistant to bacterial blight. Ten of the 21 BC2 progeny tested were resistant to Philippine races 2, 3, and 6 of the bacterial blight pathogen. One resistant BC2, plant 78-1, had 24 chromosomes. The segregation of reactions of the BC2F2, BC2F3, and BC2F4 progenies of plant 78-1 suggested that the same or closely linked gene(s) conferred resistance to races 2, 3, 5, and 6 of the bacterial blight pathogen from the Philippines.  相似文献

Sunflower rust, caused by Puccinia helianthi Schw., is a widespread disease of sunflower (Helianthus annuus L.) in China. To study physiological races, sunflower field surveys were undertaken in major sunflower growing areas of China in 2010. Forty‐four rust‐infected sunflower leaf samples were collected from 25 geographical locations. Freshly produced spores were used to study physiological race differentiation on a set of nine differentials. Race 300 was the most prevalent race observed over all locations with a 59% frequency followed by races 735, 310, 500, 724 and 737. To evaluate hybrids and varieties for resistance screening, spores of race 300 were used to inoculate 65 hybrids, and five open‐pollinated varieties selected from breeding programmes and from the seed market. None of the confection hybrids and open‐pollinated varieties was immune to race 300. Conversely, among oilseed hybrids, 3% of them showed immunity, 12% highly resistant, 59% resistant and 26% showed susceptible reactions. Open‐pollinated varieties were the most susceptible to race 300 followed by confection and oilseed sunflower hybrids. Results from this study are projected to assist breeders in selection of hybrids and varieties against prevalent race as our results showed a diversity of resistance levels to race 300.  相似文献

Erwinia soft rot is a destructive disease of Brassica rapa vegetables. Reliable sources of resistance and control methods are limited, so development of highly resistant breeding lines is desirable. Protoplasts from B. rapa and B. oleracea genotypes selected for resistance to soft rot were fused in order to combine different sources of resistance. Twelve somatic hybrids (synthetic B. napus) were obtained and confirmed by morphology, nuclear DNA content, and RAPD analysis. They were normal looking plants that easily set seeds following self-pollination and backcrossing to B. rapa. Assays of detached leaves or seedlings inoculated in a mist-chamber showed that most somatic hybrids had lower disease severity ratings than the B. rapa fusion partner and a commercial variety of B. napus. Some progeny from selfing or backcrossing of somatic hybrids to B. rapa showed much more resistance than either fusion partner. The offspring populations of the somatic hybrids (F1–S1 and F1–BC1) clearly moved to the resistant direction compared to the parents; the percentage of resistant plants increased from 21% (average of parents) to 36% (F1–S1) and 48% (F1–BC1). These results suggest that it may be possible to obtain highly resistant B. rapa lines by further backcrossing and selection. Received: June 1999 / Accepted: 29 July 1999  相似文献

Sixteen crosses between eight winter wheat cultivars were screened for resistance to Septoria nodorum leaf and glume blotch in the F1 and F4 generations using artificial inoculation in the field. The F1 of most crosses showed dominance for susceptibility on both ear and leaf. The effects of general combining ability were of similar magnitude as the effects for specific combining ability. On the basis of the phenotypic difference of the parents, no prediction was possible about the amount and the direction of genetic variance in the segregating populations. The variation observed in this study both within and among the segregating populations suggests a quantitative inheritance pattern influencing the expression of the two traits. The components of variance between F2 families within a population were as high as (for S. nodorum blotch on the ear) or higher (for S. nodorum blotch on the leaf) than those between populations. Therefore, strong selection within a few populations may be as effective to obtain new resistant genotypes as selection in a large number of populations. In almost all crosses, progenies were found that were more resistant than the better parent. Thus transgression breeding may be a tool to breed for higher levels of resistance to S. nodorum blotch. Highly resistant genotypes were found even in combination with two susceptible parents. The genetic source for Septoria resistance is probably broader than is generally assumed and could be used to improve S. nodorum resistance by combination breeding followed by strong selection in large populations. Marker assisted selection of bacterial blight resistance genes in rice   总被引:4,自引:0,他引:4  
Bacterial leaf blight caused by Xanthomonas oryzae pv. oryzae is one of the most important diseases affecting rice production in Asia. We were interested in surveying rice genotypes that are popularly used in the Indian breeding program for conferring resistance to bacterial blight, using 11 STMS and 6 STS markers. The basis of selection of these DNA markers was their close linkage to xa5, xa13, and Xa21 genes and their positions on the rice genetic map relative to bacterial blight resistance genes. Eight lines were found to contain the xa5 gene while two lines contained Xa21 gene and none of the lines contained the xa13 gene with the exception of its near-isogenic line. Using the polymorphic markers obtained in the initial survey, marker-assisted selection was performed in the F3 population of a cross between IR-64 and IET-14444 to detect lines containing multiple resistance genes. Of the 59 progeny lines analyzed, eight lines contained both the resistance genes, xa5 and Xa4.  相似文献

Summary Studies on the genetics of leaf blight caused byAlternaria triticina using generation mean analysis revealed that additive components played a major role, but that dominance components also contributed significantly in controlling the variability for leaf blight resistance in wheat crosses. Furthermore, the additive x additive type of epistasis was predominant in the first three crosses, whereas in the fourth cross additive x dominance (j) and dominance x dominance (1) components of epistasis were most significant. Because of this it may be desirable to follow a simple recurrent selection scheme for higher tolerance, to isolate resistant plants from the segregating populations derived from crosses of parents of diverse origin following the pedigree method of breeding. CPAN-1887 was very tolerant to leaf blight in the present study and should be utilized in hybridization programs to develop leaf-blight-resistant varieties.  相似文献

Hexaploid somatic hybrids resulting from mesophyll protoplast fusions between Solanum brevidens Phil., PI 218228, and Solanum tuberosum L., PI 203900 were tested for late blight resistance using two races of Phytophthora infestans Monte., de Bary. The S. tuberosum parent was a late blight differential possessing the R4 gene which confers resistance to race 0. The S. brevidens parent is resistant to potato leaf roll virus. Inoculations with both compatible (race 1.3.4.5) and incompatible (race 0) races of P. infestans clearly demonstrated the expression of the late blight resistance gene in all of the hybrid progeny tested. Most of the hybrids tested were also resistant to potato leaf roll virus (PLRV), indicating that the S. brevidens genes for PLRV resistance were present and expressed.  相似文献

To evaluate the effects of pollen contamination from outside of Cryptomeria japonica seed orchard on the growth performance (height and diameter at breast height, DBH) and morphological traits (stem straightness and basal stem straightness), paternity testing using seven microsatellite markers was performed in a progeny test. In the studied progeny test, high rates of inconsistency were found between the observed and expected genotypes. The average rates of pollen contamination from outside the orchard and self-fertilization were 58.47% and 0.65%, respectively. We divided the individuals of the studied progeny test into two groups based on their genotype data, for which: (1) both parents were elite trees and (2) only the mother trees were elite trees, and then compared them with respect to the growth performance and morphological traits of progenies using data at 20 and 30 years old. Significant adverse effects of contaminating pollen were detected in relation to straightness, but not tree height and DBH. The results suggest that the genetic gains for straightness generally show higher narrow-sense heritability than growth traits, which should be increased by reduction of pollen contamination. Breeding with paternal analysis (BWPA) is an effective approach for evaluating breeding materials based on maternal and paternal information revealed by DNA markers. The use of BWPA in progeny test allows effective forward- and backward selection without laborious and time-consuming tasks. In this study, we also suggest that the significant pollen contamination and paternal deviation found in the open-pollinated progeny test are serious impediments for BWPA.  相似文献

为了解甘蔗(Saccharum)与斑茅(Erianthus arundinaceus)杂交后代作为抗病亲本的利用价值,通过特异引物PCR鉴定出78份甘蔗与斑茅杂交BC_1真实杂种;通过人工接种花叶病毒和黑穗病菌,初步评价了甘蔗和斑茅杂交BC_1的抗病表现。结果表明,甘蔗和斑茅杂交BC_1的抗花叶病具有普遍性,而黑穗病抗性则出现分离。初步筛选出BC_1无性系YCE01-48、YCE01-71、YCE01-105、YCE01-125、YCE02-184和YCE01-118可同时抗花叶病和黑穗病,有望成为甘蔗杂交利用的高抗病源亲本。  相似文献

Populations of Arisaema triphyllum commonly harbor a systemic fungal pathogen, Uromyces ari-triphylli, which reduces leaf area and leaf longevity. Infected plants produced 42% fewer asexual progeny (cormlets). Furthermore, all asexual progeny from infected parents were themselves infected with Uromyces. Mean seed production of infected plants was only 21% of that among healthy plants. A hand pollination experiment demonstrated that 1) infected male plants can produce viable pollen, 2) the low seed production of naturally pollinated infected females was not due to inadequate pollination, and 3) seed production among healthy female plants was strongly pollinator limited. Seeds from infected and healthy parents had similar viability, and all seeds germinating from both types of parents were uninfected. The contrast in disease transmission to sexual vs. asexual progeny suggests that pathogen attack may be one selective factor favoring sexual reproduction in A. triphyllum populations.  相似文献

Interspecific somatic hybrids between commercial cultivars of potato Solanum tuberosum L. Agave and Delikat and the wild diploid species Solanum cardiophyllum Lindl. (cph) were produced by protoplast electrofusion. The hybrid nature of the regenerated plants was confirmed by flow cytometry, simple sequence repeat (SSR), amplified fragment length polymorphism (AFLP), microsatellite-anchored fragment length polymorphism (MFLP) markers and morphological analysis. Somatic hybrids were assessed for their resistance to Colorado potato beetle (CPB) using a laboratory bioassay, to Potato virus Y (PVY) by mechanical inoculation and field trials, and foliage blight in a greenhouse and by field trials. Twenty-four and 26 somatic hybrids of cph + cv. Agave or cph + cv. Delikat, respectively, showed no symptoms of infection with PVY, of which 3 and 12, respectively, were also resistant to foliage blight. One hybrid of cph + Agave performed best in CPB and PVY resistance tests. Of the somatic hybrids that were evaluated for their morphology and tuber yield in the field for 3 years, four did not differ significantly in tuber yield from the parental and standard cultivars. Progeny of hybrids was obtained by pollinating them with pollen from a cultivar, selfing or cross-pollination. The results confirm that protoplast electrofusion can be used to transfer the CPB, PVY and late blight resistance of cph into somatic hybrids. These resistant somatic hybrids can be used in pre-breeding studies, molecular characterization and for increasing the genetic diversity available for potato breeding by marker-assisted combinatorial introgression into the potato gene pool.  相似文献

From interspecific hybridisation in the genus Zantedeschia, we have previously produced albino, variegated and virescent hybrids. The inhibition of chloroplast development in these hybrids is due to plastome-genome incompatibility. The albino hybrids did not develop prolamellar bodies in their etioplasts in darkness and formed only distended membranes, but no grana in light. This indicates that the block to chloroplast development occurs before or during the development of etioplasts. In albino leaf sectors of variegated hybrids, chlorophyll and carotenoid contents were only 2–4% of those in green plants. The mRNA levels for five plastid-development-related genes were also severely reduced, but they were still readily detectable using Northern hybridisation techniques. In the green sectors of variegated hybrids, chlorophyll content and mRNA levels of the fives genes were slightly reduced. We have earlier shown that F1 hybrids between Z. aethiopica and Z. odorata were albino when Z. odorata plastids were present but virescent when Z. aethiopica plastids were present. When the F1 hybrids with Z. aethiopica plastids were backcrossed to Z. odorata, the progeny were albino, virescent or green. Z. odorata plastids were inherited only from maternal parents in the F1 progenies but inherited from either maternal or paternal parents in the backcross. The increased compatibility with, and inheritance of Z. odorata plastids in the backcross suggests that multiple genes play a major role in the plastome-genome incompatibility. An increase in the number of genes from the parent contributing the plastids improves chloroplast development in the hybrids of the genus Zantedeschia.
