Field bean (Vicia faba L.) cv. Maris Bead seeds were inoculated with Rhizobium Catalogue No. 1001, supplied by Rothamsted Experimental Station and grown in sand culture supplied with 15N-labelled nitrate at two concentrations. Plants were sampled at intervals throughout their growth for 15N and total N analysis. The rate of nitrate uptake was almost uniform up to pod-fill and was proportional to the nitrate concentration. Nodule weight was slightly depressed by the larger nitrate concentration at all samplings, and there was a corresponding reduction in the amount of atmospheric nitrogen fixed. However, at harvest the bean seeds from plants given most nitrate contained slightly more total N, as the enhanced nitrate uptake outweighed the reduction in fixation.  相似文献

Summary The hydraulic resistivity ofVicia faba L. roots grown in soil was estimated from steady state measurements of transpiration rate and leaf and soil water potentials. Root and stem axial resistivities, estimated from xylem vessel radii, were negligible. Root radial resistivity was estimated to be 1.3×1012 sm−1. This root radial resistivity value was used to estimate, root resistance to water uptake for a field crop ofVicia faba. Previously published results were used for root distribution and soil water contents at the drained upper limit (DUL) and the lower limit (LL) of extractable soil water. Soil resistance to water uptake was estimated from single root theory using the steady rate solution. At the DUL, root resistance was about 105 times greater than soil resistance. At the LL, soil resistance exceeded root resistance for depths less than 0.3 m, but for depths greater than this soil resistance was smaller than root resistance. Estimates of possible uptake rates at given leaf water potentials indicated that overall soil resistance had a negligible influence upon uptake, even at the LL. The reliability of this result is examined in detail. It is concluded that over the complete range of extractable soil water contents soil resistanceper se would not have limited water use by this crop. This conclusion may also be valid for a wide range of soil and crop combinations.  相似文献

Before starting a breeding program aimed at improving the nitrogen nutrition ofVicia faba, the authors tried an alternative technique to the acetylene reduction assay, to measure some genetic variability in the plant material. The quantity of dinitrogen fixed by several cultivars ofVicia faba was estimated using a low enrichment15N tracer method and high precision15N mass spectrometry. The fababeans were cultivated for two years in two different soils. The percentage of fixed dinitrogen in the seed varied between genotypes from 40 to 83% of the total nitrogen and was positively correlated with the total seed nitrogen (r=0.64 to 0.86). A highly significant positive correlation was also found between the total seed nitrogen and the quantity of fixed dinitrogen in the seed (r=0.95 to 0.99). The technique used to measure dinitrogen fixation proved to be useful and reliable enough to discriminate between various genotypes, grown over a period of two years in two different soils. However, several non-fixing control plants showed significant differences in their15N enrichment and the problem of choosing a good reference plant was raised and discussed.  相似文献

Summary To increase the level and stability of yield in faba beans (Vicia faba L.), heterosis must be exploited. Hybrids are not available because of the instability of male sterility. Synthetic varieties can and should be bred. Thus, we studied the reproductive behavior of this partially allogamous, insect-pollinated crop. Autofertility (AF) and the rate of cross-fertilization (C) were measured in 36 inbred lines and 28 crosses in F1, F2, and F3 generations for 3 years at Hohenheim, Stuttgart, FRG. Heterozygosity strongly increased AF and decreased C. AF was negatively correlated with C. AF varied from 1% to 98%, and C varied from 7% to 82%. Heritability for both characters was high. For an optimum exploitation of heterosis, breeders should utilize lines with high C as variety components. It is often labor-intensive to multiply such lines, due to low AF. Hence, breeders tend to use autofertile lines with rather limited C. We showed that even in this case about 50% of the total heterosis, which equals a yield increase of at least 25% over the inbred line level, is realized. An increase in yield stability due to heterogeneity will occur simultaneously.  相似文献

Summary The size and symbiotic effectiveness, withVicia faba, ofRhizobium leguminosarum populations from five locations in southern Britain has been estimated. Population numbers varied from 4.54×103 to 1.69×105. Nitrogen fixing potential differed by up to 30%. A long-term comparison between two routine soil nitrogen tests, soil nitrate versus plant indicator method, was performed on the Negev Desert loessial soil in Israel. The Gilat plant indicator method was found to be a better method to reflect the soil nitrogen availability for wheat under field conditions. It was found that 15 to 38 kg ha-1 of NO3-N, measured by nitrate soil test, for each 30 cm soil increment, is not available for plant uptake. This plant unavailable NO3-N background in the soil cannot be leached by repeated irrigation cycles of 100 mm each, or by heavy rains.  相似文献

Abstract Net nitrate uptake rates were measured and the kinetics calculated in non-nodulated Pisum sativum L. cv. Marma and Lemna gibba L. adapted to constant relative rates of nitrate-N additions (RA), ranging from 0.03 to 0.27 d?1 for Pisum and from 0.05 to 0.40 d?1 for Lemna, Vmax of net nitrate uptake (measured in the range 10 to 100 mmol m?3 nitrate, i.e. ‘system I’) increased with RA in the growth limiting range but decreased when RA exceeded the relative growth rate (RGR), Km was not significantly related to changes in RA. On the basis of previous 13N-flux experiments, it is concluded that the differences in Vmax at growth limiting RA are attributable to differences in influx rates. Linear relationships between Vmax and tissue nitrogen concentrations were obtained in the growth limiting range for both species, and extrapolated intercepts relate well with the previously defined minimal nitrogen concentrations for plant growth (Oscarson, Ingemarsson & Larsson, 1989). Analysis of Vmax for net nitrate uptake on intact plant basis in relation to nitrogen demand during stable, nitrogen limited, growth shows an increased overcapacity at lower RA values in both species, which is largely explained by the increased relative root size at low RA. A balancing nitrate concentration, defined as the steady state concentration needed to sustain the relative rate of increase in plant nitrogen (RN), predicted by RA, was calculated for both species. In the growth limiting range, this value ranges from 3.5 mmol m?3 (RA 0.03 d?1) to 44 mmol m?3 (RA 0.21 d?1) for Pisum and from 0.2 mmol m?3 (RA 0.05 d?1) to 5.4 mmol m?3 (RA 0.03 d?1) for Lemna. It is suggested that this value can be used as a unifying measure of the affinity for nitrate, integrating the performance of the nitrate uptake system with nitrate flux and long term growth and demand for nitrogen.  相似文献

Summary This paper examines evidence which quantifies the relative importance of legume and Rhizobium genotypes as determinants of phenotypic variation in symbiotic nitrogen fixation. It demonstrates potentially large and unpredictable effects of the Rhizobium genotype. The likely importance of such effects on crop yield is considered. The information is then used to assess ways in which legume breeding programmes may be altered to encompass the effects of genetic variation in Rhizobium.  相似文献

Summary The components of variation within each one of two sets of landraces and/or cultivars of Vicia faba, respectively constituted of primitive and advanced morphological types, were studied by means of two sets of 8 × 8 diallel crosses with two repetitions. The results show that primitive and modern forms differ from each other in both the intensity and the kind of selective pressures acting on them, mainly on those characters more modified through the domestication process: i.e., seed morphology and the number of flowers per node. Because of the paramount importance of the additive component in the primitive forms, it is suggested that the most important type of selection on them is the stabilizing one. On the contrary, in the most advanced forms the selection is directional and disruptive : directional towards greater yields, and disruptive separating two morphological types, major and equina. The plant response to these different selective pressures has been to modify the genetic control of different characters: thus the primitive forms generally show only additivity while the most advanced forms show additivity as well as directional and asymmetrical dominance.  相似文献

Nitrate inhibits symbiotic N2 fixation and a number of hypotheses concerned with NO3 assimilation have been suggested to explain this inhibition. These hypotheses were tested using a pea ( Pisum sativum L. cv. Juneau) with normal nitrate reductase NR; (EC 1,6,6,4) activity and two mutants of cv. Juneau, A317 and A334, with impaired NR activity. The plants were inoculated with three strains of Rhizobium leguminosarum and grown for 3 weeks in N-free medium, followed by 1 week in medium supplemented with 0, 5 or 10 m M KNO3 before harvesting. NO3 was taken up at comparable rates by the parent and the mutants and accumulated in leaf and stem tissue of the latter. Acetylene reduction rates were inhibited similarly in both the parent and mutants in the presence of KNO3 but there were differences among rhizobial strains. Starch concentration of the nodules decreased by 46% in the presence of KNO3 and there were differences among rhizobial strains but not among pea genotypes. Malate and succinate accumulated in nodules in the presence of KNO3. These data are not consistent with the photosynthate deprivation hypothesis as a primary mechanism for NO3 inhibition of N2 fixation since NO3 affected the nodule carbohydrate composition of all three pea genotypes in a similar manner. The lack of correlation between NR activity and NO3 inhibition of N2 fixation suggests that NO3 assimilation may be only indirectly involved in the inhibition phenomenon.  相似文献

Summary Homozygous mutant lines of field bean selected for (a) improved yielding potential and (b) altered plant architecture and/or physiological response were tested for symbiotic nitrogen fixing ability under field conditions in comparison with their parent cultivar.15N-tracer techniques were applied to determine %N derived from atmosphere. Data were collected on assimilate and nitrogen accumulation and distribution among various plant parts during two stages of reproductive growth.Symbiotic nitrogen fixation was closely correlated with total plant top biomass and nitrogen yield. A similar close association was found between crop yield and nitrogen harvest index. Both harvest indices tended to be negatively correlated with stage of maturity and with the amount of N derived from air per unit of area. The generally high %N derived from symbiotic N2 fixation and its comparatively small variability implies that this parameter may be difficult to improve inVicia faba under field conditions.It is concluded, that the main genetic potential for improving the amount of biological nitrogen fixation in this crop depends upon factors that promote high photosynthetic productivity and efficient N-use under appropriate agronomic conditions and with effective rhizobial associations. The establishment of rational ideotypes with a possitive impact on yield appears to be of practical significance for increasing the amount of symbiotically fixed nitrogen.  相似文献

Stomatal opening on Vicia faba can be induced by high CO2 partial pressures (10.2%) in dark as well as in light. Stomatal aperture was measured in both cases with a hydrogen porometer. The distribution of 14C among early products of photosynthesis was studied. Comparisons are made with carboxylations occurring when stomata were open in the dark with CO2-free air and in light with 0.034% CO2. Results showed that in high CO2 partial pressure in light, less radioactivity was incorporated in Calvin cycle intermediates and more in sucrose. carboxylations and photorespiration seemed to be inhibited. In the dark in both CO2 conditions, 14C incorporation was found in malate and aspartate but also in serine and glycerate in high CO2 conditions. Effects of N source and media-N and P levels were examined on growth, N uptake, and N2 fixation ofAzolla pinnata withAnabaena azollae association (azolla) at two inoculum-P concentrations. Each expeiment was conducted for 7 days in a growth chamber using azolla at a predetermined inoculum-P concentration and the growth media containing a combination of four levels of P (0, 15, 75, and 200 M) and three levels (0, 1, and 5 mM) of either15N-enriched NH 4 + as ammonium sulfate or15N-enriched NO 3 as potassium nitrate. Nitrogen uptake and N2 fixation were measured by15N isotopic dilution method. Tissue P and N, N uptake, and N2-fixation increased with increasing P concentration in the media regardless of the inoculum-P level of azolla. Increasing P concentration in the media increased growth of azolla at low inoculum P, but the effect on high inoculum-P azolla was either small or absen. High inoculum-P concentration resulted in increased growth, tissue-N and P concentrations, N uptake, and N2 fixation by azolla. Ammonium in the growth media caused larger increase in tissue-N and greater repression of N2 fixation than equimolar concentration of NO 3 . In the presence of NH 4 + or NO 3 , in the growth media, N uptake by azolla exceeded the corresponding decrease in N2 fixation, resulting in an overall increase in tissue-N concentration. Phosphorus in the media tended to negate the inhibitory effect of NH 4 + or NO 3 on N2 fixation. A multiple regression model showed that the effect of tissue-N on N2 fixation was negative while that of tissue-P was positive. Therefore, a relative change in tissue-N and P appeared to regulate N2 fixation. Tissue-N and P had similar effects on relative growth rate of azolla also. Summary Lateral roots ofVicia faba were treated with a solution of 5-aminouracil (3.93×10–3M) for 6 hours. After 15 hours roots were recovering from the temporary inhibition of mitosis induced by 5-AU and were approaching peak mitotic indices; they were then treated with 0.1% caffeine or 0.1% isobutylmethylxanthine (IBMX) for 1 hour. Treatment with methylxanthines when the mitotic index was high gave relatively high yields of binucleate cells, 3.8 to 7.5%. DNA synthesis, cell cycle duration and nuclear growth were determined for binucleate cells. Caffeine induced binucleate cells underwent a marked reduction in nuclear volume, from 1,074 m3 at 1+1 hours to 534 m3 at 1+14 hours. Only 15% of these binucleates entered S phase; those that did so were in mitosis or had divided by 1+14 hours. We conclude that 85% of the binucleate cells are so inhibited by caffeine that their G1 is extended to>14 hours or that they are no longer proliferating cells. IBMX-induced binucleate cells, by contrast, did enter S phase and many of them also divided. Though in IBMX-induced binucleate cells there was also a decrease in nuclear volume up to 1+10 hours, subsequently mean nuclear volume increased e.g. at 1+16 and 1+18 hours. Both caffeine and IBMX treatments resulted in decreases in mean volume of prophase nuclei of mononucleate cells; this is further evidence that both methylxanthines inhibit the macromolecular synthesis required to sustain nuclear growth. It also suggests that nuclear division can be initiated at considerably lower nuclear volumes than those of untreated cells. We suggest that caffeine may act as a mimic of the normal mechanism that regulates the switch from a proliferating to a non-proliferative condition.  相似文献

Polymorphism in ten enzyme systems (ACO, ACP, AAT, EST, FK, ME, NAG, PRX, 6PGD, and SOD) in Vicia faba L. was analyzed, revealing 13 loci, six of which have not been reported before. Inheritance, genetics, possible location, and linkage analysis were studied in 13 different F2 populations trisomic for four of the six chromosomes (nos. 3, 4, 5, and 6) of the species. Each of these loci exhibited typical Mendelian inheritance except for those involved in the trisomic chromosome. Five loci have been assigned to a specific chromosome: Est-2 to chromosome 3, Fk-2 to chromosome 4, Prx-1 to chromosome 5, and Sod-1 and Pgd-p to chromosome 6. Nag-1 and Pgd-c displayed a linkage of 22.8 cM indicating a clear homology with chromosome 5 of garden pea on which both markers are syntenic.  相似文献

The influence of soil nitrate availability, crop growth rate and phenology on the activity of symbiotic nitrogen fixation (SNF) during the growth cycle of pea (Pisum sativum cv. Baccara) was investigated in the field under adequate water availability, applying various levels of fertiliser N at the time of sowing. Nitrate availability in the ploughed layer of the soil was shown to inhibit both SNF initiation and activity. Contribution of SNF to total nitrogen uptake (%Ndfa) over the growth cycle could be predicted as a linear function of mineral N content of the ploughed layer at sowing. Nitrate inhibition of SNF was absolute when mineral N at sowing was over 380 kg N ha–1. Symbiotic nitrogen fixation was not initiated unless nitrate availability in the soil dropped below 56 kg N ha–1. However, SNF could no longer be initiated after the beginning of seed filling (BSF). Other linear relationships were established between instantaneous %Ndfa and instantaneous nitrate availability in the ploughed layer of the soil until BSF. Instantaneous %Ndfa decreased linearly with soil nitrate availability and was nil above 48 and 34 kg N ha–1 for the vegetative and reproductive stages, respectively, levels after which no SNF occurred. Moreover, SNF rate was shown to be closely related to the crop growth rate until BSF. The ratio of SNF rate over crop growth rate decreased linearly with thermal time. Maximum SNF rate was about 40 mg N m–2 degree-day–1, equivalent to 7 kg N ha–1, regardless of the N treatment. From BSF to the end of the growth cycle, the high N requirements of the crop were supported by both SNF and nitrate root absorption but, of the two sources, nitrate root absorption seemed to be less affected by the presence of reproductive organs. However, since soil nitrate availability was low at the end of the growth cycle, SNF was the main source of nitrogen acquisition. The onset of SNF decrease at the end of the growth cycle seemed to be first due to nodule age and then associated to the slowing of the crop growth rate.  相似文献

A trypsin inhibitor from seeds of faba bean (Vicia faba L.) was purified to near homogeneity as judged by native-PAGE with about 11 % recovery using ammonium sulphate fractionation, ion-exchange chromatography on DEAE-cellulose and gel filtration through Sephadex G-100. The inhibitor had a molecular weight of 18 kD as determined by SDS-PAGE and Sephadex G-100. The inhibitor inhibited trypsin and chymotrypsin to the extent of 48 and 12 %, respectively. The inhibtion was of non-competitive type with dissociation constant for the enzyme inhibitor complex in the region of 0.07 mg·ml−1. The inhibtor was stable between pH 4 and 5. It completely lost its activity when heated at 125 °C for 1 h or at 100 °C for 2 h. The inhibitor also lost its activity on exposure to 2-mercaptoethanol. Based on these properties, it could be concluded that Vicia faba trypsin inhibitor belongs to Bowman-Birk type of inhibitors, as it has molecular weight lower than generally observed for Kunitz type inhibitors.  相似文献

Influx isotherms were obtained for nitrate and ammonium from three legumes, Cajanus cajan (L.) Millsp., Cicer arietinum L. and Arachis hypogaea L. and three cereals, Sorghum bicolor (L.) Moench., Pennisetum glaucum L. and Zea mays L. The transition in influx isotherms for both nitrogen sources was found to be within the concentration range (0.05–2.5 mM) tested. There were significant differences in Km and Vmax for ammonium between legumes and cereals. The difference in the kinetic properties for nitrate uptake between the two groups of plants only became apparent at the higher concentration tested. Legumes translocated absorbed nitrate and ammonium to shoots more rapidly than cereals. Results show that there are significant differences in uptake and translocation of ammonium and nitrate between legumes and cereals.  相似文献
