Although isolated dental remains are a central source of data in mammalian paleontology, the teeth of many taxa remain unidentifiable at low taxonomic levels because of intraspecific variation. Extant New World porcupines, Erethizontinae, illustrate this problem. Since being declared unusually variable over 50 years ago, molar occlusal morphology has been neglected as a potential source of diagnostic traits. Here, we use geometric morphometrics to re-evaluate the occlusal morphology of the third lower molar of erethizontines as the basis for identification of genera while accounting for variation, particularly changes in shape caused by progressive tooth wear. We model the impact and predictability of this source of variation by using CT-based digital sectioning. We use modelled occlusal shapes to analytically search for traits that can be used to improve identification of isolated dental remains, as well as training machine-learning models to assign occlusal surfaces to genus. Nearly half of all shape variation can be explained by variables such as wear and size. Resulting models of shape predict six univariate traits that may diagnose the two extant genera, especially when specimens are compared at the same wear stage. However, high levels of variation previously noted qualitatively also influence quantitative analyses, rendering machine learning models unreliable. Overall, although the identity of some specimens that are highly worn or contain certain combinations of traits will remain ambiguous, large segments of the gnathic erethizontine fossil record have renewed utility when analyzed using these methods.

The recent discovery of unexpectedly ancient human remains has fuelled interest about the first dispersion of Homo outside Africa. The Dmanisi mandible is perhaps one of the most interesting findings, as it supposedly represents one of the oldest hominids outside of Africa. Recently, different interpretations have been published about this specimen. Our comparison of the Dmanisi mandible with a large sample of mandibles and teeth has led us to a new interpretation. In our view, the Dmanisi mandible exhibits a unique combination of traits. Some of its features, taken in isolation, may be attributed to morphological extremes within the genus Homo. The architecture of the mandible as well as the morphology and dimensions of incisors, canines, and P3s are clearly primitive. However, dental traits such as the reduction of the talonid in the P4s and a distally decreasing molar series seems to be derived. Some combinations of these traits are found in specimens of Homo ergaster and differ from those generally present in later hominids. Thus, we propose that the Dmanisi mandible might be taxonomically classified as Homo sp. indet. (aff. ergaster). Furthermore, some aspects of the dentition in Dmanisi display close similarities to Asian Homo erectus. If the 1.8–1.6 Myr dating for the Dmanisi mandible is correct, the differentiation of the Asian branch of the genus Homo could be regarded as a very ancient event. Dental characters are importantly used for reconstructing the evolutionary history of mammals, because teeth represent the most abundant material available for the fossil species. However, the characteristics of dental renewal are presently poorly used, probably because dental formulae are frequently not properly established, whereas they could be of high interest for evolutionary and developmental issues. One of the oldest rodent families, the Ctenodactylidae, is intriguing in having longstanding disputed dental formulae. Here, we investigated 70 skulls among all extant ctenodactylid genera (Ctenodactylus, Felovia, Massoutiera and Pectinator) by using X-ray conventional and synchrotron microtomography in order to solve and discuss these dental issues. Our study clearly indicates that Massoutiera, Felovia and Ctenodactylus differ from Pectinator not only by a more derived dentition, but also by a more derived eruptive sequence. In addition to molars, their dentition only includes the fourth deciduous premolars, and no longer bears permanent premolars, conversely to Pectinator. Moreover, we found that these premolars are lost during adulthood, because of mesial drift of molars. Mesial drift is a striking mechanism involving migration of teeth allowed by both bone remodeling and dental resorption. This dental innovation is to date poorly known in rodents, since it is only the second report described. Interestingly, we noted that dental drift in rodents is always associated with high-crowned teeth favoring molar size enlargement. It can thus represent another adaptation to withstand high wear, inasmuch as these rodents inhabit desert environments where dust is abundant. A more accurate study of mesial drift in rodents would be very promising from evolutionary, biological and orthodontic points of view.

The aims of this study were to investigate the effect of allometry on the shape of dm2 and M1 crown outlines and to examine whether the trajectory and magnitude of scaling are shared between species. The sample included 160 recent Homo sapiens, 28 Upper Paleolithic H. sapiens, 10 early H. sapiens, and 33 H. neanderthalensis (Neandertal) individuals. Of these, 97 were dm2/M1 pairs from the same individuals. A two‐block partial least squares analysis of paired individuals revealed a significant correlation in crown shape between dm2 and M1. A principal component analysis confirmed that Neandertal and H. sapiens dm2 and M1 shapes differ significantly and that this difference is primarily related to hypocone size and projection. Allometry accounted for a small but significant proportion of the total morphological variance. We found the magnitude of the allometric effect to be significantly stronger in Neandertals than in H. sapiens. Procrustes distances were significantly different between the two tooth classes in Neandertals, but not among H. sapiens groups. Nevertheless, we could not reject the null hypothesis that the two species share the same allometric trajectory. Although size clearly contributes to the unique shape of the Neandertal dm2 and M1, the largest H. sapiens teeth do not exhibit the most Neandertal‐like morphology. Hence, additional factors must contribute to the differences in dm2 and M1 crown shape between these two species. We suggest an investigation of the role of timing and rate of development on the shapes of the dm2 and M1 may provide further answers.

Construction in the City of Baltimore during 1996 led to the recovery of human skeletal remains dating from 1792 to 1856. Historical research indicates that the skeletal remains come from two adjacent graveyards: Christ's Church Episcopalian Cemetery and the Potters Field East. The different socioeconomic status of the internees in each cemetery suggests the possibility of marked contrasts in lifestyle, health, and diet. To shed further light on these possibilities, analyses of microscopic wear patterns on teeth, or dental microwear analyses, were undertaken. A sample from Spanish Florida was used to help interpret the results. Epoxy casts of incisor and molar teeth were placed in an SEM and photomicrographs of clean wear facets were taken. The photomicrographs were digitized using the software package Microware 4.02. Statistical analyses of rank transformed data consisted of single‐factor ANOVA, followed by post hoc tests. No significant differences were found between Christ's Church and Potters Field East samples for any of the variables examined in either molar or incisor teeth. However, differences between each Baltimore sample and the La Florida samples give suggestions of possible diet differences in antebellum Baltimore. The mosaic of differences between the Baltimore and La Florida samples probably reflects the wide variety of foods available to antebellum Baltimoreans as well as the relative lack of abrasives in their diet. Over 200 hominid specimens were recovered by the International Omo Expedition of 1967–1976. Despite the fragmentary nature of this primarily dental collection, these hominid remains represent a major body of evidence about hominid evolution in eastern Africa during the 2–3 myr time period. Our analysis of the Omo dental collection is based on a large comparative sample of 375 quantifiable mandibular postcanine teeth of A. afarensis, A. africanus, A. aethiopicus, A. boisei, A. robustus, and early Homo. A total of 48 isolated mandibular premolars and molars of the Omo collection spanning the 2–3 myr time period is sufficiently preserved to allow reliable serial allocations and intertaxon comparisons and is the object of study in this paper. We present taxonomic identifications of these teeth and seven other mandibular specimens preserving tooth crowns. Metric analyses of this study include cusp area and crown shape variables taken on occlusal view diagrams. Nonmetric analyses were based on simultaneous observations of all relevant material to ensure accuracy of categorical evaluations. First, a combined metric and morphological evaluation was conducted to allocate each Omo tooth to either robust or nonrobust categories. Further taxonomic affinities were then examined. Our results indicate that nonrobust and robust lineages cooccur by circa 2.7 myr. We consider the Shungura robust specimens from Members C through F to represent A. aethiopicus. A significant phenetic transformation occurs at circa 2.3 myr, with the mosaic emergence of the derived A. boisei morphology across Member G times. Characterization of the East African nonrobust lineage is more difficult because of the comparatively subtle morphological differences seen among the dentitions of A. afarensis, A. africanus, and early Homo. The earlier Members B and C nonrobust specimens are difficult to evaluate and are considered indeterminate to genus or species. Both molars and premolars from Members E through G exhibit phenetic similarities to the early Homo condition and are considered as aff. Homo sp. indet. At present, there is no indication of multiple species in the Omo nonrobust sample at any time horizon. The 2–2.4 myr Omo nonrobust specimens exhibit some similarities to the stated Homo “rudolfensis” condition in size and morphology and are likely to represent the ancestral condition of the genus Homo. The bearing of these results on interpretations of early hominid evolution and diversification is considered.

Form can be viewed as a combination of size and shape. Shape refers to the boundary outline independently from its orientation, relation to reference planes, and dimension (or size). Shape and its changes could be quantified by mathematical methods such as the Fourier series. In this investigation, Fourier analysis has been used to quantify the morphologic characteristics (size and shape) of the outline of the occlusal surface and maximum circumference (equator) in 259 normal, healthy human first permanent maxillary and mandibular molars and to assess the effect of sex. Large within-group variability was found in the Fourier coefficients. Both equatorial and occlusal molar areas were on average larger in male than in female homologous teeth, but the difference was statistically significant only for the equatorial areas. The mean ratios between equatorial and occlusal dental areas were independent from arch (maxillary and mandibular), side, or sex. Both equatorial and occlusal outlines of left and right homologous molars within sex and arch were similar, without size and shape differences. Similarly, no sex differences in shape were found in the comparison of homologous teeth. The method used in the present study could supply information about dental shape in both its entirety and local variations. In particular, the method is extremely sensitive to local variations in dental shape, and it could be usefully employed to compare single teeth to a standard.

HDP1 is an archaeological and faunal site located on the Hoedjiespunt peninsula at Saldanha Bay, South Africa, that has recently yielded fossil human remains. Artefacts from the associated archaeological deposits are identified as being Middle Stone Age. U series analysis of capping calcretes and analysis of the foraminifera and fauna associated with the human fossils indicate an age for the deposit in excess of 74,000 years before present, and it most probably dates to around 300,000 years before present. The fossil human teeth from in situ deposits at Hoedjiespunt are described and found to be large by comparison with modern humans but smaller than the known upper dentitions of southern African “archaic” Homo sapiens. The Hoedjiespunt molars are found to be morphologically within the range of variation observed in the teeth of modern Homo sapiens.

Developing wisdom teeth are easy-accessible source of stem cells during the adulthood which could be obtained by routine orthodontic treatments. Human pulp-derived stem cells (hDPSCs) possess high proliferation potential with multi-lineage differentiation capacity compare to the ordinary source of adult stem cells1-8; therefore, hDPSCs could be the good candidates for autologous transplantation in tissue engineering and regenerative medicine. Along with these benefits, possessing the mesenchymal stem cells (MSC) features, such as immunolodulatory effect, make hDPSCs more valuable, even in the case of allograft transplantation6,9,10. Therefore, the primary step for using this source of stem cells is to select the best protocol for isolating hDPSCs from pulp tissue. In order to achieve this goal, it is crucial to investigate the effect of various isolation conditions on different cellular behaviors, such as their common surface markers & also their differentiation capacity. Thus, here we separate human pulp tissue from impacted third molar teeth, and then used both existing protocols based on literature, for isolating hDPSCs,11-13 i.e. enzymatic dissociation of pulp tissue (DPSC-ED) or outgrowth from tissue explants (DPSC-OG). In this regards, we tried to facilitate the isolation methods by using dental diamond disk. Then, these cells characterized in terms of stromal-associated Markers (CD73, CD90, CD105 & CD44), hematopoietic/endothelial Markers (CD34, CD45 & CD11b), perivascular marker, like CD146 and also STRO-1. Afterwards, these two protocols were compared based on the differentiation potency into odontoblasts by both quantitative polymerase chain reaction (QPCR) & Alizarin Red Staining. QPCR were used for the assessment of the expression of the mineralization-related genes (alkaline phosphatase; ALP, matrix extracellular phosphoglycoprotein; MEPE & dentin sialophosphoprotein; DSPP).14  相似文献

Despite the general increase in digital techniques for dental morphometric analyses, only a few methods are available to study worn teeth. Moreover, permanent dentitions are studied much more frequently than deciduous teeth. In this study, we address both issues by providing a taxonomic classification of Neanderthal and modern human (MH) lower second deciduous molars (dm2s) through the analysis of crown and cervical outlines. Crown and cervical outlines were obtained from a three‐dimensional (3D) digital sample of uniformly oriented dm2s. Both outlines were centered on the centroid of their area and represented by 16 pseudolandmarks obtained by equiangularly spaced radial vectors out of the centroid. We removed size information from the oriented and centered outlines with a uniform scaling of the pseudolandmark configurations to unit Centroid Size. Group shape variation was evaluated separately for the dm2 crown and cervical outlines through a shape–space principal component (PC) analysis. Finally, quadratic discriminant analysis of a subset of PCs was used to classify the specimens. Our results demonstrate that both outlines successfully separate the two groups. Neanderthals showed a buccodistal expansion and convex lingual outline shape, whilst MHs have buccodistal reduction and straight lingual outline shape. Therefore, we confirmed that the cervical outline represents an effective parameter for distinguishing between the two taxa when dealing with worn or damaged dm2s. Oral biofilms play a crucial role in the development of dental caries and other periodontal diseases. Streptococcus mutans is one of the primary etiological agents in dental caries. Implant systems are regularly employed to replace missing teeth. Oral biofilms accumulate on these implants and are the chief cause of dental implant failure. In the present study, the potential of graphene/zinc oxide nanocomposite (GZNC) against the cariogenic properties of Streptococcus mutans was explored and the anti-biofilm behaviour of artificial acrylic teeth surfaces coated with GZNC was examined. Acrylic teeth are a good choice for implants as they are low cost, have low density and can resist fracture. Microscopic studies and anti-biofilm assays showed a significant reduction in biofilm in the presence GZNC. GZNC was also found to be nontoxic against HEK-293 (human embryonic kidney cell line). The results indicate the potential of GZNC as an effective coating agent for dental implants by efficiently inhibiting S. mutans biofilms.

Dental variation in the Chinese golden monkey (Rhinopithecus roxellana) is here evaluated by univariate, bivariate, and multivariate analyses. Allometric analyses indicate that canines and P3s are positively, but other dimensions negatively scaled to mandible and maxilla, and to body size. With the exception of the mesiodistal dimensions of I1 and M3, and the buccolingual dimension of P4, mandibular dental variables show similar scaling relative to body size. Analysis of residuals shows that males have significantly larger canine, P3 and buccolingual dimensions of the postcanine teeth (M2 and M3) than females. A significant difference in shape between the sexes is found in the buccolingual dimension of the upper teeth, but not in the mandible. Unlike the situation in some other species, female golden monkeys do not exhibit relatively larger postcanine teeth than males. In fact, the reverse is true, expecially for M2s and M3s. The fact that most of the dental variables show low negative allometry to body size might be related a cold environment that has led to the development of larger body size with reduced energy loss. When the raw data are examined by Discriminant Function Analysis the sexes are clearly distinguishable.

The purpose of this article is to examine the patterns of evolutionary relationships between human populations from the later Late Holocene (1,500–100 years BP) of southern South America on the basis of dental morphometric data. We tested the hypotheses that the variation observed in this region would be explained by the existence of populations with different phylogenetic origin or differential action of gene flow and genetic drift. In this study, we analyzed permanent teeth from 17 samples of male and female adult individuals from throughout southern South America. We measured mesiodistal and buccolingual diameters at the base of the crown, along the cement–enamel junction. The results of multiple regression analysis and a mantel correlogram indicate the existence of spatial structure in dental shape variation, as the D2 Mahalanobis distance between samples increases with increasing geographical distance between them. In addition, the correlation test results show a trend toward reduction of the internal variation of samples with increasing latitude. The detected pattern of dental variation agrees with the one expected as an outcome of founder serial effects related to an expansion of range during the initial occupation of southern South America. Mammalian tooth development relies heavily on the reciprocal and sequential interactions between cranial neural crest-derived mesenchymal cells and stomadial epithelium. During mouse tooth development, odontogenic potential, that is, the capability to direct an adjacent tissue to form a tooth, resides in dental epithelium initially, and shifts subsequently to dental mesenchyme. Recent studies have shown that mouse embryonic dental epithelium possessing odontogenic potential is able to induce the formation of a bioengineered tooth crown when confronted with postnatal mesenchymal stem cells of various sources. Despite many attempts, however, postnatal stem cells have not been used successfully as the epithelial component in the generation of a bioengineered tooth. We show here that epithelial sheets of cultured human keratinocytes, when recombined with mouse embryonic dental mesenchyme, are able to support tooth formation. Most significantly, human keratinocytes, recombined with mouse embryonic dental mesenchyme in the presence of exogenous FGF8, are induced to express the dental epithelial marker PITX2 and differentiate into enamel-secreting ameloblasts that develop a human-mouse chimeric whole tooth crown. We conclude that in the presence of appropriate odontogenic signals, human keratinocytes can be induced to become odontogenic competent; and that these are capable of participating in tooth crown morphogenesis and differentiating into ameloblasts. Our studies identify human keratinocytes as a potential cell source for in vitro generation of bioengineered teeth that may be used in replacement therapy.
