A method is described for producing and maintaining Stevia rebaudiana suspensions and regeneration of plants from calli derived from cell suspensions. Suspension cultures composed of isolated cells (ca. 10%) and cellular aggregates (5–100 cells) were obtained in 20–30 days by using friable callus as the initial inoculum in liquid medi with BA (0.5 mg/l)+2,4-D (1.0 mg/l), and periodic filtering (100–500 m sieves) with 6–7 days interval between subcultures. Cultures derived from actively growing calli are mainly diploid (2n=22) whereas those derived from senescent calli showed a wide variation in chromosome number (55–200). Stock cell suspensions which had been maintained for 3 years were plated on basal LS agar medium with BA (0.5 mg/l)+2,4D (0.5 mg/l) to form callus. Calli originating from predominantly 2n cell suspensions when transferred to medium with K (2.0 mg/l)+NAA (0.02 mg/l) were able to form buds. Shoot elongation and further rooting of isolated shoots was better on LS medium devoid of growth regulators. Stevia rebaudiana (Bert.) Bertoni has sporophytic self-incompatibility and this may lead to poor seed yield and quality, which are obstacles to large-scale crop establishment. This research investigated the influence of insect pollinators and the harvesting time on the qualitative and quantitative characteristics of the ripe seeds. A pot trial under open field conditions was established to evaluate the abundance of insect pollinators and three harvesting times on the production of ripe seeds, germination percentage, mean germination time and thousand seed weight (TSW) in 36 F1 open-pollinated genotypes. Large variability in the number of corymbs of capitula, flowering time, reproductive period and seed yield was observed among the genotypes. Genotypes with promising phenological and reproductive properties were identified. The worst germination percentage and TSW were recorded in the latest harvest. Significant linear regressions were found among the investigated parameters (insect number, germination percentage, TSW, seeds yield per plant and cycle length), confirming complex interactions on the yield and quality of stevia seeds. Results indicate that earlier flowering genotypes produced higher seed quantity, due to favourable weather conditions during the reproductive phase. Moreover, cross-pollination and the insect abundance, as Apidae and Syrphidae, play a crucial role for enhancing the stevia seed yield and quality.  相似文献

We have successfully established the co-culture of ginseng (Panax ginseng C.A. Meyer) and echinacea [Echiancea purpurea (L.) Moench.] adventitious roots for the production secondary metabolites. Adventitious roots of ginseng and echinacea were cultured in different proportions (5 g L−1; 4:1, 3:2 and 2:1 ginseng and echinacea, respectively) in 5-L capacity airlift bioreactors containing 4 L Murashige and Skoog medium supplemented with 25 μM indole-3-butyric acid and 50 g sucrose L−1 and maintained at 25°C in the dark for 40 days. Results showed the negative effect of echinacea adventitious roots on the growth of ginseng roots, however, by limiting the inoculum density of echinacea, it was possible to establish the co-cultures. To enhance the accumulation of secondary metabolites, co-cultures were treated with 200 μM methyl jasmonate after 30 days of culture initiation. Methyl jasmonate elicitation promoted the accumulation of ginsenosides in the co-cultures. It was possible to produce ginsenosides and caffeic acid derivatives in higher amounts by establishing co-cultures with higher inoculum proportion of ginseng to echinacea (4:1 and 3:2) followed by elicitation treatment. This work demonstrates the effectiveness of interspecies adventitious root co-cultures for the production of plant secondary metabolites.  相似文献

Hairy root cultures of Scutellaria lateriflora were established using Agrobacterium rhizogenes A4 and produced acteoside (18.5?mg?g(-1) dry wt), baicalin (14.5?mg?g(-1) dry wt) and wogonoside (12?mg?g(-1) dry wt). Yeast extract (50?μg?ml(-1)) increased acteoside production 1.4-fold and flavone production 1.7-fold after 7 and 14?days of elicitation. Addition of Pectobacterium carotovorum lysate in the stationary phase of the hairy root culture stimulated only the accumulation of wogonin to 30?mg?g(-1) dry wt. The production of wogonin in hairy roots could be associated with its role as a phytolaexin.  相似文献

The effects of inoculum density, aeration volume and culture period on accumulation of biomass and secondary metabolites in adventitious roots of Hypericum perforatum in balloon type airlift bioreactors (3 l capacity) were investigated. The greatest increment of biomass as well as metabolite content occurred at an inoculum density of 3 g l−1 and an aeration volume of 0.1 vvm. After 6 weeks of culture, an approximately 50-fold increase in biomass was recorded containing 60.11 mg g−1 dry weight (DW) of phenolics, 42.7 mg g−1 DW of flavonoids and 0.80 mg g−1 DW of chlorogenic acid. Liquid chromatography–mass spectroscopy/mass spectroscopy demonstrated that the presence of quercetin and hyperoside in adventitious roots at a level of 1.33 and 14.01 μg g−1 DW, respectively after 6 weeks of culture. The results suggest scale-up of adventitious root culture of H. perforatum for the production of chlorogenic acid, quercetin and hyperoside is feasible.  相似文献

We investigated the effects of medium salt strength and ammonia/nitrate ratio on biomass production and metabolites accumulation of adventitious root. The medium with full-strength Murashige and Skoog (MS) reached the highest growth rate (16.77), and the contents of saponin and polysaccharide reached the peak (i.e., 0.65 and 24.85 %) at 3/4 MS and 1 MS, respectively. In case of ammonia/nitrate ratio, a NH4 +/NO3 ? ratio of 20:40 was optimal for the production of biomass and polysaccharide (23.27 %). In contrast, the content of saponin achieved the optimum (0.74 %) at a NH4 +/NO3 ? ratio of 30:30. In 5-L balloon-type bubble bioreactor (BTBB) cultures, an approximately 23-fold increase in biomass was recorded. The fresh weight (FW) and dry weight (DW) were 72.78 and 6.79 g per bioreactor with the contents of saponin (0.62 %) and polysaccharide (17.32 %), respectively. It indicated potential application to produce adventitious roots of pseudostellaria heterophylla with bioreactors on large scale in commercial.  相似文献

The establishment of green root cultures of Stevia rebaudiana Bertoni, and the effect of elicitors such as hydrogen peroxide (H2O2) and methyl jasmonate (MeJA), is shown in the present study. Stevioside, rebaudioside A, and the isomers steviol/isosteviol were identified through DFI-ESI-IT-MSn and UPLC-TOFMS spectrometric systems, in combination with solid-phase extraction. The accumulation of steviol glycosides increased by 2.4 times (compared to the control value of 22.35 μgSG per gDW), with the addition of 250 μM H2O2. The non-enzymatic antioxidant response, which resulted from production of phenolic and flavonoid compounds, was modified based on the elicitor and the dose used. The maximum accumulation of flavonoids was induced on the third day with the addition of H2O2 (250 or 500 μM), and with MeJA (250 or 500 μM); the increase was observed on the fifth day. The enzymatic antioxidant response of the catalase and peroxidase from the roots under elicitation confirmed the stress conditions.

To improve root growth and production of bioactive compounds such as anthraquinones (AQ), phenolics, and flavonoids by adventitious root cultures of Morinda citrifolia, the effects of aeration rate, inoculum density, and Murashige and Skoog (MS) medium salt strengths were investigated using a balloon-type bubble bioreactor. The possible mechanisms underlying changes in activities of enzymic (superoxide dismutase, catalase, guaiacol peroxidase, ascorbate peroxidase) and nonenzymic (vitamin E) antioxidants, phenylalanine ammonia lyase, and stress levels (accumulation of hydrogen peroxide and proline, peroxidation of lipids) were also studied. Low aeration rate (0.05 vvm [air volume/culture volume/min]) accelerated accumulation of root fresh weight and dry weight (DW). High aeration rates (0.1 to 0.3 vvm) stimulated accumulation of AQ, phenolics, and flavonoids and reduced root growth. Low inoculum densities (5 and 10 g l–1) increased accumulation of those metabolites but inhibited root growth. Culture of adventitious roots with high concentrations of MS salts (1× and 1.5× MS) resulted in induction of oxidative stress that strongly inhibited root growth. Overall, an aeration rate of 0.05 vvm, 15 g l–1 inoculum density, and half-strength (0.5×) MS medium were optimal for enhancing accumulation of root dry biomass (4.38 g l–1), AQ (103.08 mg g–1 DW), phenolics (54.81 mg g–1 DW), and flavonoids (49.27 mg g–1 DW).  相似文献

Elicitation is a possible aid to overcome various difficulties associated with the large‐scale production of most commercially important bioactive secondary metabolites from wild and cultivated plants, undifferentiated or differentiated cultures. Secondary metabolite accumulation in vitro or their efflux in culture medium has been elicited in the undifferentiated or differentiated tissue cultures of several plant species by the application of a low concentration of biotic and abiotic elicitors in the last three decades. Hairy root cultures are preferred for the application of elicitation due to their genetic and biosynthetic stability, high growth rate in growth regulator‐free media, and production consistence in response to elicitor treatment. Elicitors act as signal, recognized by elicitor‐specific receptors on the plant cell membrane and stimulate defense responses during elicitation resulting in increased synthesis and accumulation of secondary metabolites. Optimization of various parameters, such as elicitor type, concentration, duration of exposure, and treatment schedule is essential for the effectiveness of the elicitation strategies. Combined application of different elicitors, integration of precursor feeding, or replenishment of medium or in situ product recovery from the roots/liquid medium with the elicitor treatment have showed improved accumulation of secondary metabolites due to their synergistic effect. This is a comprehensive review about the progress in the elicitation approach to hairy root cultures from 2010 to 2019 and the information provided is valuable and will be of interest for scientists working in this area of plant biotechnology.  相似文献

Two greenhouse experiments were conducted to study the effect of two plant growth retardants, Chlorocholine chloride (CCC) and Paclobutrazol (PBZ), on growth, Steviol glycosides (SVglys) content and antioxidant capacity in Stevia (Stevia rebaudiana Bertoni). Five concentrations of CCC (0, 250, 500, 750 and 1,000 ppm) and PBZ (0, 6, 12, 18 and 24 ppm) with three replications were applied to Stevia plants as treatments based on completely randomized design. CCC was sprayed on Stevia shoots, but PBZ was applied as a drench. The obtained results showed that CCC reduced plant height but improved leaf and stem dry weight, especially with 750 ppm concentration. Total SVgly content and consequently SVglys yield were significantly reduced by CCC application, and 1,000 ppm of CCC concentration was more effective than other treatments. PBZ had no effect on Stevia height while it significantly enhanced stem and dry weight at 12 ppm. Moreover, PBZ remarkably increased total SVglys contents, SVglys yield, and Rebaudioside A/Stevioside ratio. Total antioxidant capacity significantly varied with CCC and PBZ and the highest activity was obtained with 1,000 and 12 ppm of CCC and PBZ, respectively. The results of these experiments indicated that, although CCC and PBZ are plant growth retardants and act as anti-gibberellins, only CCC reduced plant height and SVglys production in Stevia. On the contrary, PBZ at 12 ppm concentration, improved Stevia growth, SVglys production, and antioxidant capacity.  相似文献

Two-year-old seedlings of licorice plant (Glycyrrhiza uralensis Fisch) were exposed to three degrees of water deficit, namely weak (60–70%), moderate (40–50%), and strong (20–30%) relative water content in soil, whereas control plants were grown in soil with 80–90% water content. Moderate and strong water deficit decreased the net photosynthetic rate, stomatal conductance, and biomass production. Water use efficiency and the root-to-shoot ratio increased significantly in response to water deficit, indicating a high tolerance to drought. Weak water deficit did not decrease root biomass production, but significantly increased the production of glycyrrhizic acid (by 89%) and liquiritin (by 125%) in the roots. Therefore, a weak water deficit can increase the yield of root medical compounds without negative effect on root growth.  相似文献

In an attempt to scale-up of adventitious root cultures of Echinacea angustifolia for the production of biomass and caffeic acid derivatives, i.e. echinacoside, chlorogenic acid, cichoric acid, caftaric acid, and cynarin, the effects of Murashige and Skoog (MS) medium dilutions, and initial sucrose concentrations were investigated in a 5-L airlift bioreactor. In addition, the kinetics of adventitious root growth and accumulation of secondary metabolites were also studied. The greatest root dry weight (6.50 g L?l) and accumulation of total phenolics [22.06 mg g?1 DW (dry weight)], total flavonoids (5.77 mg g?1 DW) and total caffeic acid derivatives (10.63 mg g?1 DW) were obtained at quarter-strength MS medium. Of the various gradients of sucrose tested, 5 % sucrose supplementation was regarded as an optimal concentration for enhancing productivity of biomass and bioactive compounds. Neither higher salt strength (3/4–2 MS) nor sucrose concentrations (7 and 9 %) showed promotive effect on root growth and metabolite production. The kinetic studies revealed that 4 weeks of culture period is the optimal time to achieve highest productivity of metabolites. Based on these results, a large-scale (20 L) and a pilot-scale (500 L) adventitious root culture system was established. In the pilot-scale bioreactor, adventitious roots were elicitor-treated with 100 μM methyl jasmonate (MJ) on day 28. After 1 week of elicitation, 1.75 kg dry root biomass was harvested containing 60.41 mg g?1 DW of total phenolics, 16.45 mg g?1 DW of total flavonoids, and 33.44 mg g?1 DW of total caffeic acid derivatives. Among the caffeic acid derivatives, the accumulation of echinacoside (the major bioactive compound) in MJ-treated adventitious roots grown in the 500-L bioreactor was the highest (12.3 mg g?1 DW), which is approximately threefold more than the non-MJ-treated roots cultured in 5- and 20-L bioreactors.  相似文献

Summary Yellowish compact callus, induced from cowpea hypocotyls on Murashige and Skoog(MS) medium (1962) containing 0.2 mg/l(0.93 μM) kinetin and 0.4 mg/l (1.81 μM) 2,4-dichlorophenoxyacetic acid (2,4-D), was subcultured on MS medium containing cytokinin alone, auxin alone, or auxins plus cytokinins in order to determine the effect of cytokinins on root organogenesis in callus cultures. The callus actively proliferated on the same medium but did not show any organogenic activity macroscopically as well as microscopically. On medium with N6-benzyladenine (BA) and 1-naphthaleneacetic acid (NAA), the yellowish compact callus first changed to pale green compact callus and then many green spots appeared on its surface under light culture. But the yellowsih compact callus remained yellowish and white spots appeared on its surface in dark culture. These spots gradually became white nodular structures. Adventitious root formation from the nodular structures occurred not only on the same medium, but also on medium with either auxin or cytokinin but not both. Yellowish compact callus on medium with auxin alone was transformed to yellowish friable callus, which did not develop adventitious roots. The yellowish friable callus could gain rhizogenic activity only after morphological modification to pale green compact callus on medium with auxin plus cytokinin. The modified callus did not form adventitious roots on medium with auxins but only with cytokinins. Therefore, it is suggested that cytokinins have stimulating effects on root formation from callus that previously did not show rhizogenic activity on medium with auxins alone. In addition, the rhizogenic potential of cowpea callus was discriminated from that of leaf explants, which formed adventitious roots directly on medium with auxin alone.  相似文献

Pyrroline-5-carboxylate reductase (P5CR) lies at the converging point of the glutamate and ornithine pathways and is the last and critical enzyme in proline biosynthesis. In the present study, a P5CR gene, named IbP5CR, was isolated from salt-tolerant sweetpotato line ND98. Expression of IbP5CR was up-regulated in sweetpotato under salt stress. The IbP5CR-overexpressing sweetpotato (cv. Kokei No. 14) plants exhibited significantly higher salt tolerance compared with the wild-type. Proline content and superoxide dismutase and photosynthetic activities were significantly increased, whereas malonaldehyde content was significantly decreased in the transgenic plants. H2O2 was also found to be significantly less accumulated in the transgenic plants than in the wild-type. Overexpression of IbP5CR up-regulated pyrroline-5-carboxylate synthase gene and down-regulated proline dehydrogenase and P5C dehydrogenase genes under salt stress. The systemic up-regulation of reactive oxygen species (ROS) scavenging genes was found in the transgenic plants under salt stress. These findings suggest that overexpression of IbP5CR increases proline accumulation, which enhances salt tolerance of the transgenic sweetpotato plants by regulating osmotic balance, protecting membrane integrity and photosynthesis and activating ROS scavenging system. This study indicates that IbP5CR gene has the potential to be used for improving salt tolerance of plants.  相似文献

Nitric oxide (NO) affects the growth and development of plants and also affects plant responses to various stresses. Because NO induces root differentiation, we examined whether or not it is involved in increased ROS generation. Treatments with sodium nitroprusside (SNP), an NO donor, 2-phenyl-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide (PTIO), a specific NO scavenger, and Nω-nitro-l-arginine methyl ester hydrochloride (l-NAME), an NO synthase (NOS) inhibitor, revealed that NO is involved in the adventitious root growth of mountain ginseng. Supply of an NO donor, SNP, activates NADPH oxidase activity, resulting in increased generation of O2 ·−, which subsequently induces growth of adventitious roots. Moreover, treatment with diphenyliodonium chloride (DPI), an NADPH oxidase inhibitor, individually or with SNP, inhibited root growth, NADPH oxidase activity, and O2 ·− anion generation. Supply of the NO donor, SNP, did not induce any notable isoforms of enzymes; it did, however, increase the activity of pre-existing bands of NADPH oxidase, superoxide dismutase, catalase, peroxidase, ascorbate peroxidase, and glutathione reductase. Enhanced activity of antioxidant enzymes induced by SNP supply seems to be responsible for a low level of H2O2 in the adventitious roots of mountain ginseng. It was therefore concluded that NO-induced generation of O2 ·− by NADPH oxidase seems to have a role in adventitious root growth of mountain ginseng. The possible mechanism of NO involvement in O2 ·− generation through NADPH oxidase and subsequent root growth is discussed.  相似文献

Salt-induced changes in the activities of antioxidant enzymes such as superoxide dismutase (SOD), catalase (CAT), peroxidase (POX), and lipid peroxidation in terms of malondialdehyde (MDA), level of H2O2, and some key metabolites such as soluble proteins, free proline and phenolics in the leaves of six radish cultivars (Radish Red Neck, Radish Lal Pari, Radish Mino Japani, Radish 40 Days, Mannu Early and Desi) were investigated. Varying levels of NaCl (0, 80 and 160 mM) applied for 40 days adversely affected the shoot fresh weight, chlorophyll contents and soluble proteins, while increased the levels of proline, and the activities of SOD, POD and CAT. However, leaf H2O2 and total phenolic contents were not affected by salt stress. Cultivars Mannu Early, Radish 40 Days and Desi were relatively higher in shoot fresh weight (percent of control) while cvs. Radish Mino Japani and Mannu Early in proline, and cvs. Radish 40 Days and Desi in total soluble proteins at 160 mM of NaCl. However, levels of H2O2 and phenolics were higher in cvs. Desi, Radish Lal Pari and Mannu Early and SOD, POD and CAT activities only in Radish Lal Pari and Mannu Early than the other cultivars under saline conditions. Overall, the differential salt tolerance of radish cultivars observed in the present study was not found to be associated with higher antioxidant enzyme activities and other key metabolites analyzed, so these attributes cannot be considered as selection criteria for salt tolerance in radish.  相似文献
