When N 6 [8–14C] furfuryladenine was applied to the intact root system of Pisum sativum L. cv. Meteor seedlings it was almost completely metabolised to other compounds within 24 h. Of the total activity recovered from the plants 94.5% was retained in the root system itself. 14C was recovered in a number of ethanol-soluble compounds and in ribonucleic acid, deoxyribonucleic acid and protein fractions of roots, stems, leaves and axillary buds. In rapidly growing axillary buds released from apical dominance by removal of the shoot apex the combined nucleic acid fractions accounted for 63.3% of the total 14C recovered from these organs. Xylem exudate collected from decapitated plants 0 to 12 h after supplying N 5[8–14C]furfuryladenine to the roots consistently contained a single major 14C-labelled compound which, in three different solvent systems, had the same Rf values as a major endogenous cytokinin isolated from the xylem of unlabelled plants. The content of N 6 [8–14C] furfuryladenine itself in the xylem exudate was always low and in some experiments it could not be detected.
It is suggested that part of the label from N 6 [8- 14CJfurfuryladenine taken up by the intact root system may have become incorporated in an endogenous cylokinin before export to the shoot.  相似文献

We investigated the interaction of Ca2+ and pH on root elongation in Zea mays L. cv. B73 × Missouri 17 and cv. Merit. Seedlings were raised to contain high levels of Ca2+ (HC, imbibed and raised in 10 m M CaCl2) or low levels of Ca2+ (LC, imbibed and raised in distilled water). In HC roots, lowering the pH (5 m M MES/Tris) from 6.5 to 4.5 resulted in strong, long-lasting growth promotion. Surprisingly, increasing the pH from 6.5 to 8.5 also resulted in strong growth promotion. In LC roots acidification of the medium (pH 6.5 to 4.5) resulted in transient growth stimulation followed by a gradual decline in the growth rate toward zero. Exposure of LC roots to high pH (pH shift from 6.5 to 8.5) also promoted growth. Addition of EGTA resulted in strong growth promotion in both LC and HC roots. The ability of EGTA to stimulate growth appeared not to be related to H+ release from EGTA upon Ca2+ chelation since, 1) LC roots showed a strong and prolonged response to EGTA, but only a transient response to acid pH, and 2) promotion of growth by EGTA was observed in strongly buffered solutions. We also examined the pH dependence of the release of 45Ca2+ from roots of 3-day-old seedlings grown from grains imbibed in 45Ca2+. Release of 45Ca2+ from the root into agar blocks placed on the root surface was greater the more acidic the pH of the blocks. The results indicate that Ca2+ may be necessary for the acid growth response in roots.  相似文献

We investigated the effect of Ca2+ on ethylene production in 2-cm long apical segments from primary roots of corn ( Zea mays L., B73 × Missouri 17) seedlings. The seedlings were raised under different conditions of Ca2+ availability. Low-Ca and high-Ca seedlings were raised by soaking the grains and watering the seedlings with distilled water or 10 m M CaCl2, respectively. Segments from high-Ca roots produced more than twice as much ethylene as segments from low-Ca roots. Indoleacetic acid (IAA; 1 μ M ) enhanced ethylene production in segments from both low-Ca and high-Ca roots but auxin-induced promotion of ethylene production was consistently higher in segments from high-Ca roots. Addition of I-aminocyclopropane-I-carboxylic acid (ACC) to root segments from low-Ca seedlings doubled total ethylene production and the rate of production remained fairly constant during a 24 h period of monitoring. In segments from high-Ca seedlings ACC also increased total ethylene production but most of the ethylene was produced within the first 6 h. The data suggest that Ca2+ enhances the conversion of ACC to ethylene. The terminal 2 mm of the root tip were found to be especially important to ethylene biosynthesis by apical segments and, experiments using 45Ca2+ as tracer indicated that the apical 2 mm of the root is the region of strongest Ca2+ accumulation. Other cations such as Mn2+, Mg2+, and K+ could largely substitute for Ca2+. The significance of these findings is discussed with respect to recent evidence for gravity-induced Ca2+ redistribution and its relationship to the establishment of asymmetric growth during gravitropic curvature.  相似文献

A method is described by which the surface area of a root is estimated from cation uptake data. 24Na+ was supplied to excised roots of tomato ( Lycopersicon esculentum Mill, cv. Tiny Tim) seedlings at 3 μ M in unstirred solution. Coarse roots, for which external surface area and specific gravity could be measured accurately, were used to estimate the thickness (dN) of the Nernst boundary layer at the root surface. 24Na+ uptake (J1) was measured by γ-ray spectroscopy. Jt and dN were used to calculate the total surface area for ion absorption in fine roots, assuming that Na uptake rate was diffusion-limited. The results were compared to data obtained by conventional methods and indicated the usefulness of the cation uptake technique for quantitative estimates of root surfaces.  相似文献

The K+(86Rb) uptake into the roots and the translocation to the shoots of 11-day-old intact wheat seedlings ( Triticum aestivum L. cv. Martonvásári 8) were investigated using plants grown with different K+ supplies. The effects of environmental conditions (darkness, humidity) and of metabolic and transport inhibitors (oligomycin, disalicylidene-propanediamine, 2,4-dinitriphenol, diethylstilbestrol, colchicine) were also studied. Plants with K content of about 0.2 mmol/g dry weight in the root and 0.5 mmol/g dry weight in the shoot (low K status) showed high K+ uptake into the roots and high translocation rates to the shoots. Both transport processes were very low in plants with K content of more than 1.5 and 2.2 mmol/g dry weight in the root and shoot, respectively (high K status).
Darkness and a relative humidity of the air of 100% did not influence K+ uptake by roots, but did inhibit upward translocation and water transport. Inhibition of photosynthesis and treatments with diethylstilbestrol (10−5 mol/dm3), as well as with colchicine resulted in inhibition of translocation in plants of low K status, but these inhibitors had little effect on K+ uptake by the roots. Oligomycin, 2,4-dinitrophenol and diethylstilbestrol (10−4 mol/dm3), however, inhibited K+ uptake by the roots. In general, K+ transport processes were almost unchanged in plants of high K status. It is concluded that only plants of low K status operating with active K+ transport mechanisms are responsive to environmental factors. In high K+ plants the transport processes are passive and are uncoupled from the metabolic energy flow.  相似文献

Abstract. The effect of fusicoccin (FC) on the K+stimulated Na+ efflux in root cells of Na+ loaded barley roots was studied. FC (0.02 mM) stimulated Na+ efflux in the presence of K+ and its effect was synergistic with that of K+, in a similar way as its effect on proton extrusion. Decreasing the pH of the elution medium promoted Na+ efflux and partially replaced the effect of FC. As FC is known to increase the electrochemical proton gradient at the plasmalemma level, these results are consistent with the hypothesis that Na+ is extruded in exchange for H+. A further support to this view came from the finding that Na+ efflux was also promoted by a lipophilic cation, tributylbenzylammonium (TBBA +), which stimulates H + extrusion and is generally accepted not to enter the cells by means of the same carrier as K +.  相似文献

Six cultivars of spring barley ( Hordeum vulgare L. cvs Salve, Nümberg II, Bomi, Risø 1508, Mona and Sv 73 608) were grown in water culture for three weeks with various combinations of mineral supply and differential roots/shoot temperatures during the growth period. Most important for growth and accumulation of N, K+, Ca2+ and Mg2+ was the mineral supply, followed by the root temperature and the choice of cultivar. Treatments with low mineral supply or low root temperature induced a uniform reduction in growth and accumulation of the ions studied. The effects of low mineral supply and low root temperature on growth and N accumulation was additive, which indicates that these factors exert their influence independently of each other.
Roots grown at 10°C were smaller and Rb+(86Rb) influx was higher than in roots grown at 20°C. It is suggested that the control of Rb+(86Rb) influx is affected by the root temperature and the age of the plants. The higher 86Rb+ (86Rb) influx into the low temperature roots could not compensate for the smaller root size. However, the lower total mineral accumulation made up for the needs of the smaller plants and cannot explain the reduction in growth.  相似文献

The effects of external K+, H+ and Ca2+ concentrations on the intracellular K+ concentration, [K+]i, and the K+-ATPase activity in 2-day-old mung bean roots [ Vigna mungo (L.) Hepper] were investigated. [K+]i, in mung bean roots was markedly decreased by external K+ or H+ stress and did not recover the initial value even after the stress was removed. This decrease in [K+]i, gradually disappeared with the addition of (Ca2+. Ca2+ may offset the harmful effects of ion stress. Ca2+ seems to have two effects on K+ transport; control of K+ permeability and activation of K+ uptake, although K+-ATPase activity was inhibited by Ca2+ concentrations higher than 10–4 M. We suggest that Ca2+ activates K+ uptake indirectly through the acidification of the cytoplasm.  相似文献

Influx, efflux and translocation of K+(86Rb) were studied in the roots of sunflower seedlings ( Helianthus annuus L. cv. Uniflorus) treated with 0–4.0 m M NO3 during a 9 day growth period or a 24 h pretreatment period. Roots treated with high levels of NO3 absorbed and translocated more K+(86Rb) than seedlings treated with low levels of NO3. The content of K+ in the shoots was, however, higher in seedlings treated with low levels of NO3, indicating a low rate of retranslocation of K+ in those plants. K+(86Rb) efflux was highest into the low-NO3 solutions. All effects on K+(86Rb)-fluxes were more obvious in high-K plants than in low-K plants. The results are discussed in relation to the Dijkshoorn-Ben Zioni hypothesis for K++ NO3-uptake and translocation in plants.  相似文献

Effects of nitrate, chloride and chlorate ions upon nitrate and chlorate uptake by roots of maize ( Zea mays L., cv. B73) seedlings were examined. Net nitrate uptake, 36ClO3 influx and 36Cl influx (the latter two in a background of 0.5 m M KNO3) displayed similar pH profiles with optima at pH 5.5 and below. External, non-labeled chloride had little effect on the accumulation of 36ClO3 (both in 5 h and 20 min uptake assays), while nitrate and chlorate had almost identical, marked inhibitory effects. Nitrate pretreatment caused an apparent induction of both 36ClO3 and 15NO3 uptake activities. After 5 h of treatment in nitrate, the uptake activities of chloride- and chlorate-pretreated plants increased to that of nitrate-pretreated plants. During 6 h exposure to chlorate, 36ClO3 uptake activity of nitrate-pretreated plants decreased to that of chlorate- and chloride-pretreated plants. The results support the existence of a shared nitrate/chlorate transport system in maize roots which is not inhibited by external chloride, and which is induced by nitrate, but not by chlorate or chloride. The suggestion is made that selection of chlorate-resistant mutants of maize can identify nitrate uptake as well as nitrate reductase mutants.  相似文献

Root morphology and Zn^2+ uptake kinetics of the hyperaccumulating ecotype (HE) and nonhyperaccumulating ecotype (NHE) of Sedum alfredii Hance were investigated using hydroponic methods and the radiotracer flux technique. The results indicate that root length, root surface area, and root volume of NHE decreased significantly with increasing Zn^2+ concentration in growth media, whereas the root growth of HE was not adversely affected, and was even promoted, by 500μmol/L Zn^2+. The concentrations of Zn^2+ in both ecotypes of S. alfredii were positively correlated with root length, root surface area and root volumes, but no such correlation was found for root diameter. The uptake kinetics for ^65Zn^2+ in roots of both ecotypes of S. alfredii were characterized by a rapid linear phase during the first 6 h and a slower linear phase during the subsequent period of investigation. The concentration-dependent uptake kinetics of the two ecotypes of S. alfredii could be characterized by the Michaelis-Menten equation, with the Vmax for ^65Zn^2+ influx being threefold greater in HE compared with NHE, indicating that enhanced absorption into the root was one of the mechanisms involved in Zn hyperaccumulation. A significantly larger Vmax value suggested that there was a higher density of Zn transporters per unit membrane area in HE roots.  相似文献

The large plaquing (24 mm2) soilborne bacteriophage, Fo-l, did not affect the colonization ability on sugarbeet roots of its host, fluorescent Pseudomonas sp. strain B26. Phage Fo-l did not increase in numbers on sugarbeet roots when seeds were coated with less than 106 CFU (colony forming units) of B26 and when less than 300 PFU (plaque forming units) of phage Fo-l was added per g of soil (dry weight). Above these threshold values, phage replication occurred and up to 2 × 106 PFU per root system could be recovered.  相似文献

The influence of light intensity and phytochrome on the uptake of 14C-kinetin (6-furfurylamino-[8- 14C]-purine) by the plant and the translocation of the phytochrome between the roots, the hypocotyl and the cotyledons were investigated with radish seedlings ( Raphanus sativus L. cv. Saxa Treib) grown in the dark or under white light of high (20,000 lux, 90 W m−2) or low intensity (2,000 lux, 14 W m−2). The highest uptake of labelled kinetin was found in plants grown in continuous darkness. The total uptake of kinetin was decreased by strong light and to a finally higher extent by weak light. Under white light most of the kinetin accumulated in the root, whereas in the dark an enhanced translocation of the phytohormone into the cotyledons was observed. In etiolated radish seedlings, light acting on phytochrome (daily 5 min red or far red light pulses) decreased the translocation of 14C-kinetin into the cotyledons. Under far red light a pronounced uptake of the phytohormone into the roots was found. The data are discussed with regard to the interaction of light and phytohormones on plant development.  相似文献

Vitamin D3 at low concentration (10−9 M) inhibited the growth of Phaseolus vulgaris L. (cv. Contrancha) roots in vitro as measured by elongation (14 h) and [3H]-leucine incorporation into protein (2 h), and increased their labelling with 45Ca2+ (2 h). Cycloheximide and puromycin (50 u.M) blocked vitamin D3 stimulation of root 45Ca2+ labelling, indicating that it is mediated by de novo protein synthesis. The calcium ionophore X-537A (10−5JW) induced similar changes both in root elongation and 45Ca2+ uptake (14 h). This may indicate that the inhibitory effects of the sterol on root growth are mediated by changes in Ca2+ fluxes. However, this interpretation should be further strengthened by additional studies as the ionophore may have acted on root growth, affecting physiological processes other than Ca2+ transport.  相似文献

Interactive effects of elevated atmospheric CO2 and arbuscular mycorrhizal (AM) fungi on biomass production and N2 fixation were investigated using black locust ( Robinia pseudoacacia ). Seedlings were grown in growth chambers maintained at either 350 μmol mol−1 or 710 μmol mol−1 CO2. Seedlings were inoculated with Rhizobium spp. and were grown with or without AM fungi. The 15N isotope dilution method was used to determine N source partitioning between N2 fixation and inorganic fertilizer uptake. Elevated atmospheric CO2 significantly increased the percentage of fine roots that were colonized by AM fungi. Mycorrhizal seedlings grown under elevated CO2 had the greatest overall plant biomass production, nodulation, N and P content, and root N absorption. Additionally, elevated CO2 levels enhanced nodule and root mass production, as well as N2 fixation rates, of non- mycorrhizal seedlings. However, the relative response of biomass production to CO2 enrichment was greater in non-mycorrhizal seedlings than in mycorrhizal seedlings. This study provides strong evidence that arbuscular mycorrhizal fungi play an important role in the extent to which plant nutrition of symbiotic N2-fixing tree species is affected by enriched atmospheric CO2.  相似文献

The roots of a mature, field-grown maize plant are dimorphic: the primary root and those from the oldest nodes are bare with a heavily lignified cortex arid sloughed epidermis; those from younger nodes, except for a bare elongation zone, have an intact epidermis surrounded by a persistent soil sheath. Sheathed roots consistently have more layers of cortical cells, but the ratio of volumes of cortex to stele (ca 4) and the cross-sectional area of phloem (ca3× 10−2 mm2) are similar in each type. Assimilated carbon (from 14 C applied to a small area of one leaf) was translocated to all roots and actively metabolized in cortex and stele of both types. After 1 to 2 days the proportion of 14C exuded from a given length of mature root into its soil sheath, or into the adjacent unattached soil in the case of bare roots, was the same (5%) in both root types when compared with the ethanol-soluble 14C in the tissues of this length. Up to 75% of the ethanol-soluble label in the roots was in a cationic fraction (amino acids and unidentified compounds), ca 1% was in an anionic fraction and the remainder was in a neutral fraction (sugars). Approximately equal amounts of soluble 14C were found in the stele, cortex and laterals.  相似文献

Abstract CS fimbriae-associated plasmids of two enterotoxigenic Escherichia coli strains of serotype O6: K15: H16 or H- (biotypes A and F) with M r values of 51 × 106 and 72 × 106, respectively, were mobilized into various alternative host bacteria. Expression of CS1 or CS2 fimbriae was obtained when either of the CS fimbriae-associated plasmids was introduced into CS Fim, O6: K15: H16 or H- recipients with rhamnose-negative and rhamnose-positive fermentation phenotypes, respectively, whereas CS3 fimbriae were expressed irrespective of the biotype of the recipient. On transfer into a CS Fim variant of an enterotoxigenic O8: H9 strain and into two K-12 strains, a CS3-fimbriae-only phenotype was conferred by the presence of either of the plasmids. When a CS Fim variant of a Rha+ CS2-fimbriae-only strain of serotype O6: K15: H16 harboured either of the plasmids, both CS2 and CS3 fimbriae were expressed, indicating that the rare CS2-fimbriae-only wild-type phenotype is probably due to the presence of a defective plasmid in such strains. Mobilization of the 51 MDa CS fimbriae-associated plasmid into five non-enterotoxigenic Rha+ porcine isolates of E. coli with O6 serotypes other than O6: K15: H16 or H- yielded CS3-fimbriae-only transconjugants. Thus the correlation between a Rha+ fermentation phenotype and expression of CS2 fimbriae does not hold in general for O-group 6 strains.  相似文献
